Background In the present study, we aim to provide an inventory of digenetic trematodes (Platyhelminthes, Trematoda, Digenea) from Georgia including records from the freshwater, marine and terrestrial realms. The checklist is based on a critical review of data from 109 papers, 11 monographs and four Ph.D. theses published between 1935 and 2019 and our new records. The checklist includes information on synonymy, the host species, site of infection, geographical distribution and bibliographical references. The present data will serve as a baseline for further studies on trematodes from Georgia focused on integrative taxonomy, life-cycle elucidation, parasite ecology and epidemiology. New information We compiled data on the digenean trematode fauna of Georgia, which is represented by 186 species (of these 173 identified to species level) belonging to 108 genera, 47 families and 17 superfamilies. This is the first checklist of the digeneans of Georgia. The majority of digenean species were recorded as adults (160 species), only a small fraction being found as cercariae (33 species) or metacercariae (24 species), in their first or second intermediate hosts, respectively. Predominantly, records of trematodes (62 species) from birds were found, followed by those parasitising fish (50 species, i.e. 32 species as adults and 18 as metacercariae), mammals (33 species) and amphibians (25 species, i.e. 23 species as adults and 2 as metacercariae), with the least number of species reported from reptiles (12 species, i.e. 9 species as adults and 3 as metacercariae). Adult digeneans recorded together with another life-cycle stage (metacercariae and/or cercariae) comprised 28 species, i.e. for 15% of the total trematode species number, a part of their life-cycle is known. More than 30 species of exotic ungulates are currently kept in the Askania-Nova Biosphere Reserve (Kherson Region, Ukraine). During the years 1978–2014, 146 ungulates of 24 species (16 species of Bovidae, 4 — Cervidae, 4 — Camelidae) were examined by partial helminthological dissection; more than 402,700 specimens of helminths were collected and identified. The purpose of the present study was to summarize these data and analyze the species diversity in exotic ungulates. Totally, 38 species of helminths: 3 species of Trematoda, 6 — Cestoda, 29 — Nematoda were found. Ungulates from the family Bovidae were the most infected; they harbored 36 species of helminths, 1–18 species per host. In Camelidae, 15 species were found; 3–10 species per host. Cervidae harbored 9 species; 1–6 species per host. The highest species diversity was detected in the ungulates introduced from regions with climatic conditions similar to those in southern Ukraine: saiga antelope (19 species), European mouflon (18), Barbary sheep (18), and markhor (17). Ten species typical for domestic ruminants (Haemonchus contortus, Nematodirus sp., Aonchotheca bovis, Moniezia expansa, Oesophagostomum venulosum, Ostertagia circumcincta, Trichostrongylus axei, T. colubriformis, T. probolurus and Trichuris ovis) dominated in the parasite communities. Our results indicated reduction in the species diversity and alteration of the parasite community structures in these exotic ungulates kept in the Askania-Nova Reserve compared to their natural habitats. Abstract A rich fauna of digenetic trematodes has been documented from the Great Barrier Reef (GBR), yet little is known of the complex life-cycles of these parasites which occur in this diverse marine ecosystem. At Heron Island, a small coral cay at the southern end of the GBR, the intertidal marine gastropod Clypeomorus batillariaeformis Habe & Kusage (Cerithiidae) is especially abundant. This gastropod serves as an intermediate host for 12 trematode species utilising both fish and avian definitive hosts. However, 11 of these species have been characterised solely with morphological data. Between 2015 and 2018 we collected 4870 C. batillariaeformis from Heron Island to recollect these species with the goal of using molecular data to resolve their phylogenetic placement. We found eight of the 12 previously known species and two new forms, bringing the total number of digenean species known to parasitise C. batillariaeformis to 14. The families of this trematode community now include the Atractotrematidae Yamaguti, 1939, Bivesiculidae Yamaguti, 1934, Cyathocotylidae Mühling, 1898, Hemiuridae Looss, 1899, Heterophyidae Leiper, 1909, Himasthlidae Odhner, 1910, Microphallidae Ward, 1901, and Renicolidae Dollfus, 1939. Molecular data (ITS and 28S rDNA) were generated for all trematode species, and the phylogenetic position of each species was determined. The digenean community parasitising C. batillariaeformis includes several common species, as well as multiple species which are uncommon to rare. Although most of those trematodes in the community which exploit fishes as definitive hosts have remained common, the composition of those which utilise birds appears to have shifted over time. Plagioporus ictaluri n. sp. and Plagioporus carolini n. sp. are described from the intestines of the Ouachita Madtom, Noturus lachneri, and the Banded Sculpin, Cottus carolinae, respectively, from adjacent drainages in Arkansas. The new species are morphologically most similar to one another and in turn similar to Plagioporus sinitsini, Plagioporus chiliticorum, Plagioporus serratus, and Plagioporus hypentelii, but they can be distinguished from these congeners in possession of an excretory vesicle that extends anteriorly to the level of the anterior testis as opposed to 1 reaching only the posterior testis (P. hypentelii) or 1 confined to the posttesticular space (P. sinitsini, P. serratus, and P. chiliticorum), a feature that necessitates altering the generic diagnosis for the genus. Plagioporus ictaluri n. sp. is distinguished from Plagioporus carolini n. sp. in having tandem vs. oblique testes, a submedian to median ovary as opposed to 1 that is dextral, a ventral sucker occupying 53-71% of the body width (BW) vs. 80-92% of the BW, an oral sucker occupying 36-47% of the BW as opposed to 49-58% of the BW, and a pharynx occupying 21-26% of the BW compared to 28-36% of the BW. A Bayesian inference analysis of partial 28S rDNA sequences of the 2 new species and those of 34 opecoelids obtained from GenBank found that P. ictaluri and P. carolini formed a highly supported clade that was sister to P. chiliticorum and in turn to P. sinitsini. These 4 species are notably the only Nearctic plagioporids included in the analysis without a uterus extending to the posterior end that lack a confluent vitelline field in the posttesticular space. This study includes the first species of Plagioporus to be described from an ictalurid host and the first species in the genus to be described from a cottid east of the Rocky Mountains. Allacanthochasmus texanus n. sp. is described from white bass, Morone chrysops, collected from the Trinity River system in southeastern Texas, U.S.A. Specimens of the new species can be distinguished from Allacanthochasmus varius Van Cleave, 1922 by the former's elongate body shape, prominent prepharynx, and tandem testes. The new species is most similar to Allacanthochasmus artus Mueller and Van Cleave, 1932, with which it shares a common body shape, structure of the gonotyl, and distribution and arrangement of most internal structures; the new species differs in having vitelline follicles confined more posteriorly, a gonotyl with more root-like processes, and a smaller number of circumoral spines on the oral sucker. The shape of the circumoral spines in A. texanus n. sp. is highly variable, and the type series includes specimens with spines similar to what described for both A. artus and A. varius. Allacanthochasmus texanus is the third species in the genus from North America, and at this time, all 3 species that are considered valid members of this genus have been described from white bass. Caecincola aubergina n. sp. is described from white crappie ( Pomoxis annularis) from B. A. Steinhagen Reservoir in southeastern Texas, U.S.A. Members of C. aubergina n. sp. have an elongate body (5.5-8.8:1 length:width ratio), a multilobed ovary, large vitelline follicles primarily in the hindbody but extending into the forebody, and an oral sucker that is wider than long. The elongate form of the new specimens distinguishes them from all other species in the genus except Caecincola longiscens and Caecincola cookorum. In the former species, vitelline follicles are confined to the hindbody; in the latter species, the body is peanut-shaped and the vitelline follicles are almost entirely in the forebody. Background: An opisthorchiid liver fluke was recently reported from ducks (Anas platyrhynchos) in Binh Dinh Province of Central Vietnam, and referred to as "Opisthorchis viverrini-like". This species uses common cyprinoid fishes as second intermediate hosts as does Opisthorchis viverrini, with which it is sympatric in this province. In this study, we refer to the liver fluke from ducks as "Opisthorchis sp. BD2013", and provide new sequence data from the mitochondrial (mt) genome and the nuclear ribosomal transcription unit. A phylogenetic analysis was conducted to clarify the basal taxonomic position of this species from ducks within the genus Opisthorchis (Digenea: Opisthorchiidae). Methods: Adults and eggs of liver flukes were collected from ducks, metacercariae from fishes (Puntius brevis, Rasbora aurotaenia, Esomus metallicus) and cercariae from snails (Bithynia funiculata) in different localities in Binh Dinh Province. From four developmental life stage samples (adults, eggs, metacercariae and cercariae), the complete cytochrome b (cob), nicotinamide dehydrogenase subunit 1 (nad1) and cytochrome c oxidase subunit 1 (cox1) genes, and near-complete 18S and partial 28S ribosomal DNA (rDNA) sequences were obtained by PCR-coupled sequencing. The alignments of nucleotide sequences of concatenated cob + nad1 + cox1, and of concatenated 18S + 28S were separately subjected to phylogenetic analyses. Homologous sequences from other trematode species were included in each alignment. Results: Phylogenetic trees were inferred from concatenated (cob + nad1 + cox1) nucleotide sequences and combined 18S + 28S nucleotide sequences of five Opisthorchis sp. BD2013 samples and additional reference taxa. Both trees demonstrated the anticipated clustering of taxa within the superfamily Opisthorchioidea, the paraphyly of the genus Opisthorchis and the sister-species relationship of Opisthorchis sp. BD2013 with O. viverrini. Conclusions: While it is likely that Opisthorchis sp. BD2013 is distinct from O. viverrini, it is clearly a sister taxon of O. viverrini within the limited number of Opisthorchis species for which appropriate sequence data are available. We describe Schikhobalotrema huffmani n. sp. from Tylosurus crocodilus (Péron and Leseur) (Belonidae) collected off Lizard Island, Great Barrier Reef, Queensland, Australia and Tylosurus gavialoides (Castelnau) collected from Moreton Bay, Queensland. Schikhobalotrema huffmani n. sp., along with Schikhobalotrema ablennis (Abdul-Salam and Khalil, 1987) Madhavi, 2005, Schikhobalotrema acutum (Linton, 1910) Skrjabin and Guschanskaja, 1955 and Schikhobalotrema adacutum (Manter, 1937) Skrjabin and Guschanskaja, 1955 are distinguished from all other species of Schikhobalotrema Skrjabin and Guschanskaja, 1955 in having ventral suckers which bear lateral lobes and have longitudinal apertures. Schikhobalotrema huffmani n. sp. differs from S. ablennis in having an obvious post-vitelline region and a longer forebody. From S. acutum, S. huffmani n. sp. differs in having a prostatic bulb smaller than the pharynx and more anterior testis. From S. adacutum, S. huffmani n. sp. differs in having more prominent ventral sucker lobes, a conspicuous prostatic bulb and a longer forebody. We also report the first Australian record of Haplosplanchnus pachysomus (Eysenhardt, 1829) Looss, 1902, from Mugil cephalus Linnaeus (Mugilidae) collected in Moreton Bay. Molecular sequence data (ITS2, 18S and 28S rDNA) were generated for Schikhobalotrema huffmani n. sp., H. pachysomus and archived specimens of Hymenocotta mulli Manter, 1961. The new 18S and 28S molecular data were combined with published data of five other haplosplanchnid taxa to expand the phylogeny for the Haplosplanchnata. Bayesian inference and Maximum Likelihood analyses recovered identical tree topology and demonstrated the Haplosplanchnata as a well-supported monophyletic group. The two ruminant parasites, Paramphistomum cervi and Carmyerius gregarius, were collected from fresh-slaughtered native cattle at local abattoirs in Sadat district, Menoufia province and identified morphologically, then molecularly by sequencing the nucleotides of 18S ribosomal RNA gene (18S rRNA). The nucleotide sequences of the two isolates were 456 ( P. cervi) and 401 bases for ( C. gregarius). The data were used along with those of several other helminth species from the GenBank to identify these two species genetically. The nucleotide sequences were aligned using multiple sequence alignments of nucleotides by Clustal W 12.1 V and construct their relationship. Neighbor-joining analytical method was used showing sister relationship between C. gregarius from Sadat district and Gastrodiscoides hominis (EF027096) with relative identity of (98%) due to the presence of single nucleotides polymorphisms (SNPs) in the form of indels as nine nucleotides positions. But when clustering of P. cervi Sadat isolate with Paramphistomoidea sp. S4 isolate P5 (GU735643), this relationship shows complete identity (99%) between them. The homology and diversity was done using Bayesian analyses in MrBayes v3.1. This work will give a useful guide for other researchers for the molecular taxonomic position of Paramphistomatidae spp. in Sadat district among the different species around the world. Two species of Thaparocleidus Jain (1952a) were found harboring W. attu from the Ganga River at two localities, Meerut and Farrukhabad, Uttar Pradesh, India, during the period of 2013-2015. Morphology and morphometric study of specimens identified as Thaparocleidus gomtius (Jain, 1952a) Lim, 1996 and T. sudhakari (Gusev, 1976) Lim, 1996. Molecular analyses using the 18S rRNA gene confirmed the validity of T. gomtius and T. sudhakari and demonstrated that both the species clustered with other Thaparocleidus species from different geographical regions. We aim at reassessing the taxonomy and establishing the phylogenetic relationships among these two redescribed species with other representatives of the genus Thaparocleidus. [ABSTRACT FROM AUTHOR]

In the Ross Sea and Amundsen Sea, four representatives of the genus Paralepidapedon-Paralepidapedon cf. dubium Prudhoe et Bray 1973 sensu Sokolov et Gordeev 2013, P. lepidum (Gaevskaya et Rodyuk 1988), Paralepidapedon sp., and P. variabile sp. n.-were found in demersal fishes Muraenolepis marmorata and Macrourus whitsoni. Paralepidapedon variabile sp. n. is described from Muraenolepis marmorata in the Amundsen Sea. Paralepidapedon variabile sp. n. differs from other species of the genus Paralepidapedon by the position of the anterior border of the vitellarium at the level of the anterior edge of the ventral sucker or genital pore and by the highly variable shape of the testes: from roundish with a smooth edge to sinuate-lobate. Trematodes are recognized as a group of emerging parasites in tropical countries. We identified a trematode as a cause of ocular granulomas that developed in children who bathed in ponds or rivers in South India. DNA was isolated from patients' surgically excised granulomas and from the trematode cercariae (larvae) released by the snail Melanoides tuberculata in water in which the children bathed. Real-time and conventional PCRs were performed that targeted ribosomal DNA regions spanning the internal transcribed spacer 2 and 28S sequences of this trematode. The PCR-amplified products were subjected to bidirectional sequencing. Analysis of sequences for the granuloma samples and the trematode cercariae showed maximum sequence similarity with Procerovum varium (family Heterophyidae). Our results confirmed the etiology of the ocular infection, implicating snail vectors as environmental risk factors for ocular parasitosis. [ABSTRACT FROM AUTHOR]

Polychaete worms are hosts to a wide range of marine parasites; yet, studies on trematodes using these ecologically important species as intermediate hosts are lacking. During examination of the spionid polychaete Marenzelleria viridis collected on the north shore of Long Island, New York, putative trematode cysts were discovered in the body cavity of these polychaetes. In order to verify these cysts as metacercariae of trematodes, specimens of the eastern mudsnail Ilyanassa obsoleta (a very common first intermediate host of trematodes in the region) were collected for molecular comparison. DNA barcoding using cytochrome C oxidase I regions confirmed the presence of three species of trematodes ( Himasthla quissetensis, Lepocreadium setiferoides, and Zoogonus lasius) in both M. viridis and I. obsoleta hosts. Brown bodies were also recovered from polychaetes, and molecular testing confirmed the presence of L. setiferoides and Z. lasius, indicating an immune response of the polychaete leading to encapsulation of the cysts. From the 125 specimens of M. viridis collected in 2014, 95 (76.8 %) were infected with trematodes; of these 95 infected polychaetes, 86 (90.5 %) contained brown bodies. This is the first confirmation that trematodes use M. viridis as a second intermediate host and that this intermediate host demonstrates a clear immune response to metacercarial infection. Future research should explore the role of these polychaetes in trematode life cycles, the effectiveness of the immune response, and transmission pathways to vertebrate definitive hosts. Paradiplozoon homoion is a representative of blood-feeding ectoparasites from the family Diplozoidae (Polyopisthocotylea, Monogenea). Although these worms have been the subject of numerous taxonomical, phylogenetic and ecological studies, the ultrastructure of the alimentary system and related structures, as well as the mechanisms of essential processes like fish blood digestion, remain mostly unknown. Our observation of P. homoion using a transmission electron microscopy (TEM) revealed two main types of digestive cells-U-shaped haematin cells and connecting syncytium. Particular structures such as mouth cavity with specialised receptors, two oval-shaped muscular buccal suckers, pharynx surrounded with the glandular cells, oesophagus, the intestinal caeca with intact erythrocytes in the lumen, the apical pinocytotic fibrous surface complex and haematin vesicles of U-shaped cells have been shown in detail. The systematic position of the Collyricloides massanae, a rare cyst-dwelling parasite, located on intestinal wall of European birds and rodents, have always been controversial. Based on newly obtained sequences of the 28 sDNA of C. massanae from avian and rodent host from Central Europe, and on the previously published sequences of several genera and families among Microphalloidea, we evaluate its taxonomic position and the phylogenetic relationships within the genera Collyriclum Kossack, 1911 and Collyricloides Vaucher, which form the family Collyriclidae Ward, . In the cladogram, C. massanae appears among the Pleurogenidae, forming a clade with Gyrabascus amphoraeformis (Modlinger, 1930) and Cortrema magnicaudata (Bykhovskaya-Pavlovskaya, 1950). We reject the commonly accepted placement of Collyricloides as the sister genus to Collyriclum within the Collyriclidae. Background Small liver and minute intestinal flukes are highly prevalent in Southeast Asia. Definitive diagnosis of parasite infection is usually achieved parasitologically by finding the fluke eggs in feces. However, their eggs are difficult to differentiate morphologically in fecal samples, even for experienced technicians. The present study developed a PCR assay coupled with DNA pyrosequencing for identification of the fish-borne trematodes (FBT), Opisthorchis viverrini, Clonorchis sinensis, Haplorchis taichui, H. pumilio and Stellantchasmus falcatus, and to evaluate potential detection in fecal specimens, and identification and differentiation of cercarial and metacercarial stages. Methods Primers targeting the partial 28S large subunit ribosomal RNA gene were designed and about 46-47 nucleotides were selected as the target region for species identification by a PCR assay coupled with a pyrosequencing technique. Results The nucleotide variations at 24 positions, which is sufficient for the identification of the five species of FBT were selected. The method could identify O. viverrini and C. sinensis eggs in feces, cercarial and metacercarial stages of O. viverrini, and metacercarial stage of H. pumilio and H. taichui. The detection limit was as little as a single O. viverrini or C. sinensis egg artificially inoculated in 100 mg of non-infected fecal sample (equivalent to 10 eggs per gram), indicating highly sensitivity. The method was found to be superior to the traditional microscopy method and was more rapid than Sanger DNA sequencing. Conclusions DNA pyrosequencing-based identification is a valuable tool for differentiating O. viverrini and other Opisthorchis-like eggs, and can be applied to epidemiological studies and for molecular taxonomic investigation of FBT in endemic areas. Abstract: Two schistosome species—Schistosoma haematobium and S. mansoni—with two very different pathological profiles (urogenital versus intestinal), are responsible for the majority of human schistosomiasis infections across sub-Saharan Africa. The aim of this study was to determine whether coinfections have an impact on species-specific morbidity measures when compared to single species infections. Children from two neighbouring schools in Taveta, Kenya were grouped by infection status, i.e. uninfected, single species infections or coinfected. Clinical examination of the liver and spleen by palpation was performed and urinary albumin levels were recorded at baseline and at 12 months after praziquantel administration. Additional ultrasonographic profiles of the children's liver, spleen and bladder were incorporated at follow-up. It was found that S. haematobium-associated urogenital morbidity was lower in the coinfected group relative to single S. haematobium infections, even when infection intensities were taken into account. We also observed an association between S. haematobium infection and liver (intestinal-associated) morbidity regardless of coinfections. The findings reported here suggest that further research should be performed on the impact of S. haematobium infections on liver morbidity as well as to determine the impact of mixed schistosome species infections on human morbidity outcomes across different endemic settings. Given the importance in tropical diseases caused by food-borne trematodes infecting humans, it is essential to understand the taxonomy and distribution of its first intermediate hosts, which are in Southeast Asia freshwater Cerithioidean gastropods such as the widely distributed Melanoides tuberculata. Next to the occurrence of this type species in Thailand there is a second, congeneric thiarid neglected and, therefore, remained enigmatic to date. Described as ' Melania ' jugicostis by Hanley & Theobald (1876) it was originally reported and only known from the type locality, 'Tenasserim River' (i.e. Tanintharyi River) of Southeast Burma (today Myanmar), but later recorded also for Thailand based on shell morphology. In evaluating the status of this taxon, we here report on its geographical occurrence ranging from the Kanchanaburi Province to the southern most provinces in Thailand (anticipating that it might extend further south on the Malay Peninsula and even into Sumatra). Based on the more detailed study of material from five locations we provide conchological and anatomical evidence for the comparison with other thiarids, in particular the closely related Melanoides tuberculata and Pseudoplotia scabra. Interestingly, with respect to its reproductive mode we found in one population of M. jugicostis from Erawan waterfall, Kanchanaburi Province, the lack of any sign of a subhaemocoelic brood pouch otherwise typical for viviparous thiarids. This is discussed in context of climatic factors (e.g. seasonal changing precipitation), but anticipated to be caused by an unusually high infection rate of human intestinal trematodes. These parasites having infected 98 of 222 snails studied, resulting in an infection rate of 44.14 %, are assumed to be responsible for the sterility of the females in this particular population, in comparison with considerable lower infection rates in two other populations, viz. Although many studies on the taxonomy of digenean trematodes of marine fishes have been completed in the Eastern Indo-Pacific (EIP) marine ecoregion, only a few have considered metacercarial stages. Here, the results are presented of a taxonomic survey of the digenean metacercariae of fishes from Palmyra Atoll, a remote and relatively pristine US National Wildlife Refuge located 1680 km SSW of Hawaii. Up to 425 individual fish were collected, comprising 42 fish species, from the sand flats bordering the lagoon of the atoll. Quantitative parasitological examinations of each fish were performed. Morphological descriptions of the encountered digenean metacercariae are provided, together with their prevalence, mean intensities, host and tissue-use. Up to 33,964 individuals were recovered representing 19 digenean metacercaria species from eight families. The species composition of digeneans in lagoon fishes at Palmyra Atoll is a subset of what has previously been reported for the EIP. Further, the large diversity and abundance of metacercariae reported in this study highlight the utility of including this group in future ecological research in the EIP marine ecoregion. The article presents a case study of a 10-year-old girl in Kokourou, Niger who was infected by livestock-specific schistosomes through hybridization and introgression of Schistosoma (S.) bovis with S. curassoni. It states that Schistosoma eggs were recovered from the urine of the child, and mitochondrial and nuclear DNA regions of larvae were analysed. It discusses how hybrid livestock:livestock schistosomes can infect humans, and public health concerns due to hybridization of parasites.

The complete mitogenome sequences of the intestinal flukeFasciolopsis buskiare presented for the first time in this study. It is 14,119 bp long and is thus the shortest trematode mitochondrial genome sequenced to date. TheF. buskimtDNA genome has a close resemblance withF. hepaticaand has a similar gene order tallying with that of other trematodes. The overall base composition ofF. buskimitogenome is 17.89% for A, 9.16% for C, 27.59% for G and 45.36% for T, and has a GC content of 36.75%. The assembled mitogenome (GenBank accession number KX449331) consists of 12 protein-coding genes (PCGs), 22 transfer RNAs and two ribosomal RNA genes. The mtDNA for the intestinal fluke reported herein would help investigate Fasciolidae taxonomy and systematics with the aid of mtDNA NGS data. [ABSTRACT FROM PUBLISHER]
