Purpose: The original aim of the study was to determine, in a double-blind 3-arm crossover human trial (n = 7), the effect of supplemental levels of iron (25 mg) and zinc (30 mg) on β-carotene (synthetic) bioavailability (10 h postprandial). However, despite the high dose of supplemental β-carotene (15 mg) consumed with the high fat (18 g), dairy-based breakfast test meal, there was a negligible postprandial response in plasma and triglyceride rich fraction β-carotene concentrations. We then systematically investigated the possible reasons for this low bioavailability of β-carotene. Methods: We determined (1) if the supplemental β-carotene could be micellised and absorbed by epithelial cells, using a Caco-2 cell model, (2) if the fat from the test meal was sufficiently bioavailable to facilitate β-carotene bioavailability, (3) the extent to which the β-carotene could have been metabolised and converted to retinoic acid/retinol and (4) the effect of the test meal matrix on the β-carotene bioaccessibility (in vitro digestion) and Caco-2 cellular uptake. Results: We found that (1) The supplemental β-carotene could be micellised and absorbed by epithelial cells, (2) the postprandial plasma triacylglycerol response was substantial (approximately 75–100 mg dL−1 over 10 h), indicating sufficient lipid bioavailability to ensure β-carotene absorption, (3) the high fat content of the meal (approximately 18 g) could have resulted in increased β-carotene metabolism, (4) β-carotene bioaccessibility from the dairy-based test meal was sixfold lower (p < 0.05) than when digested with olive oil. Conclusion: The low β-carotene bioavailability is probably due to a combination of the metabolism of β-carotene to retinol by BCMO1 and interactions of β-carotene with the food matrix, decreasing the bioaccessibility. The human body can easily convert β‐carotene into retinol (a precursor of vitamin A), a significant ingredient in dietary supplements. The need for commercial natural pigment production, including β‐carotene, has led to intensive research in this area. Processing after biosynthesis is a vital stage that involves the use of nontoxic solvents, efficient mechanical disruption of cells, and the isolation of the required compounds. With the growing popularity of green extraction methods and the increasing market demand for carotenoids due to their health benefits, the use of clean, nontoxic, flash‐hydrolyzed biomass for carotenoid production presents a significant advantage. Furthermore, the remaining biomass can serve as a protein source for animal feed, thereby aligning with the principles of the biorefinery concept. In this study, our aim was to optimize the disruption of cells in two microalgal species using flash hydrolysis (FH). We applied two distinct levels of pressure and four separate temperature conditions over a brief residence time. Following this, we employed an ethanol extraction method to assess the efficiency of carotenoid pigment extraction. Flash hydrolysis is a chemical‐free subcritical water‐based continuous‐flow process, injected with wet biomass slurry at a high temperature (150–250 °C) for a very short time (8–12 s) to obtain bioproducts. In this study, we discovered that the pigment β‐carotene constituted 0.04 to 0.3% of the dry biomass in the case of Chlorella vulgaris. Bands of lutein and zeaxanthin were also observed in the thin layer chromatography (TLC) analysis of the treated slurry. This was achieved under conditions of an effective temperature of 200 °C and a pressure of 1700 psi. The Arthrospira platensis (1Tex), β‐carotene yield was 0.014 to 0.021% of dry biomass. Some lutein and zeaxanthin bands were also found in the treated slurry with an effective temperature of 150 °C and 1000 psi pressure. This study reports pigment extraction by FH for the first time. It is a viable process to scale up high‐value natural products like carotenoids from microalgae at the industrial level without adverse environmental impacts. Due to low residence time, the temperature does not have a negative effect on pigments; cell rupture was accomplished effectively using water as a solvent. A considerable amount of pigment could be recovered from microalgae at specific combinations of temperature and pressure depending on the type of cell wall that microalgae possess. Plastic packaging materials have caused significant environmental issues because they are nonrenewable and nonbiodegradable. Wax‐emulsified films from biomaterials, on the other hand, are renewable and biodegradable, making them promising packaging alternatives. The development of bioactive wax‐emulsified films (BWEFs) with antimicrobial and antioxidant properties has been a topic of extensive research. In this review, we provide an overview of wax emulsification, BWEF processing, and applications. BWEF contains several bioactive agents such as tannins, flavonoids, phenolic acids, and carotenes. The addition of emulsified wax and active agents to films not only enhances their antimicrobial and antioxidant properties but also reduces their hydrophilicity. Other reagents have been added to BWEF partially to increase hydrophobicity and mainly to improve mechanical properties. BWEFs prolong food shelf life, monitor wound healing, and control drug delivery. We have identified some research challenges that need to be solved to make BWEFs industrially attractive. The purpose of this study was to investigate the effects of different chromatographic column absorbents (activated clay, silica gel, and activated aluminum oxide) on the adsorption and removal of minor components (sterols, tocopherols, β‐carotene, free fatty acids (FFAs), secondary oxidation products, and peroxides) in vegetable oils. For single absorbents, activated aluminum oxide was the most effective material for removing tocopherols and FFAs from oil. All the adsorbents used have the capacity to remove β‐carotene from edible oils, and activated clay has the strongest ability, with removal rates reaching 99.8%. The brassicasterol, campesterol, β‐sitosterol, γ‐tocopherol, and δ‐tocopherol in vegetable oil can be completely removed by silica gel. Using a combination of these absorbents, a multistage optimized method was developed to maximize the removal of various minor components from vegetable oils. After multistage purification, free sterols, tocopherols, FFAs, β‐carotene, and secondary oxidation products were completely removed, whereas peroxides were also removed at concentrations less than 0.71 mmol/kg. The purified oils could be used as standard samples for in‐depth study of the effects of minor components on formation of association colloids and their influence on oil oxidation, as well as for evaluating the antioxidant potential of novel antioxidants or extracts. Neoporphyra haitanensis, a red alga harvested for food, thrives in the intertidal zone amid dynamic and harsh environments. High irradiance represents a major stressor in this habitat, posing a threat to the alga's photosynthetic apparatus. Interestingly, N. haitanensis has adapted to excessive light despite the absence of a crucial xanthophyll cycle‐dependent photoprotection pathway. Thus, it is valuable to investigate the mechanisms by which N. haitanensis copes with excessive light and to understand the photoprotective roles of carotenoids. Under high light intensities and prolonged irradiation time, N. haitanensis displayed reduction in photosynthetic efficiency and phycobiliproteins levels, as well as different responses in carotenoids. The decreased carotene contents suggested their involvement in the synthesis of xanthophylls, as evidenced by the up‐regulation of lycopene‐β‐cyclase (lcyb) and zeaxanthin epoxidase (zep) genes. Downstream xanthophylls such as lutein, zeaxanthin, and antheraxanthin increased proportionally to light stress, potentially participating in scavenging reactive oxygen species (ROS). When accompanied by the enhanced activity of ascorbate peroxidase (APX), these factors resulted in a reduction in ROS production. The responses of intermediates α‐cryptoxanthin and β‐cryptoxanthin were felt somewhere between carotenes and zeaxanthin/lutein. Furthermore, these changes were ameliorated when the organism was placed in darkness. In summary, down‐regulation of the organism's photosynthetic capacity, coupled with heightened xanthophylls and APX activity, activates photoinhibition quenching (qI) and antioxidant activity, helping N. haitanensis to protect the organism from the damaging effects of excessive light exposure. Asphodelus-based CQDs were successfully synthesized in a single step by the hydrothermal synthesis method for detection of β-carotene. TEM, XRD, FTIR, XPS, Zeta potential, UV–Vis absorption and fluorescence measurements were taken to structurally and optically characterize the newly synthesized CQDs. It was determined that the characterized CQDs show high selectivity and sensitivity only to β-carotene among 11 different biomolecules. The detection limit of CQDs for β-carotene was calculated as 8.28 µM. It was determined that the new CQDs respond very quickly and stably without interfering with any other biomolecules. The studies in different pH environments exhibited that CQDs can detect β-carotene at all pH studied. Real sample experiments show that β-carotene can be determined accurately and reliably in practical applications with the novel CQDs. Tomato (Solanum lycopersicum L.), as one of the most valuable horticulture crops, was chosen to investigate the effect of nanoparticles (NPs) in the form of nano-ZnO combined with conventional fertilizer on the quality of tomato fruits, including their antioxidant potential (total antioxidant activity, lycopene and β-carotene content), sugars content and allergenic potential (profilin and Bet v 1 content). Nano-ZnO was implemented during plant cultivation, applied by foliar spraying or directly via soil, at three different concentrations (50, 150 and 250 mg/L). The obtained results suggest that the usage of NPs during tomato plant cultivation had minor impacts on parameters such as total antioxidant activity or the content of selected allergens. Even though the total antioxidant activity was not affected by nano-ZnO, the malondialdehyde activity (MDA) content was notably decreased in fruits under nano-ZnO treatment. The content of lycopene and β-carotene was significantly affected by the use of nano-ZnO. Moreover, the usage of nano-ZnO significantly increased the total sugar content in fruits treated with nanoparticles via foliar spraying. Based on the obtained results, it can be stated that nano-ZnO, regardless of the method of application, significantly affected tomato fruits which can be beneficial for fruit production. Objective: Oxidative stress is strongly associated with atopic dermatitis (AD), and increased antioxidant intake could potentially reduce the risk of or alleviate its symptoms. However, the argument is disputed. Therefore, we conducted a Mendelian randomization (MR) analysis to explore the causal relationship between dietary antioxidant vitamin intake and AD. Methods: We applied MR analysis to examine the causative association between dietary antioxidant vitamin intake (vitamin C, vitamin E, carotene, and retinol) and AD. The genome‐wide association study (GWAS) summary data for antioxidant vitamins intake and AD were obtained from the IEU OpenGWAS database and the UK biobank. Our study consisted of two major parts, MR analysis to detect the causal relationship between exposure and outcome, and sensitivity analysis as supplemental evidence to verify the robustness of the results. Result: The results revealed a suggestive causal relationship between vitamin E intake and AD (p = 0.038, OR 95% CI = 0.745–0.992). However, there was no causal relationship between the other three vitamins (vitamin C, carotene, and retinol) and AD (p = 0.507, OR 95% CI = 0.826–1.099) (p = 0.890, OR 95% CI = 0.864–1.184) (p = 0.492, OR 95% CI = 0.893–1.264). None of the single nucleotide polymorphisms (SNPs) were detected as heterogeneous and pleiotropy in the sensitivity analysis (p > 0.05). Conclusion: The analysis suggested that dietary intake of vitamin E may potentially lower the risk of AD. Conversely, intake of vitamin C, retinol, and carotene is not causally related to AD. The study aims at investigating the impact of ultrasound enhancement on the water change and quality characteristics of dried carrots by heat pump drying (HPD). The results showed that ultrasound had obvious strengthening effect on the drying and dehydration process of HPD, but there was an attenuation effect of ultrasound in the propagation process of materials, and the magnetic resonance imaging results could visually demonstrate the change and migration of moisture inside carrot slices. Higher drying temperature and ultrasonic power could cause more micropores and higher content of polyphenols, flavonoids and niacin of carrot slices. Conversely, the elevated drying temperature reduced rehydration ratio. β-carotene content showed a trend of increasing first and then decreasing due to excessive temperature and ultrasonic power. Based on AHP-CRTITC method, the highest comprehensive score was attained at drying temperature of 60 °C and ultrasonic power of 80 W. Dietary supplementation with synthetic β‐carotene was investigated in this study to assess its impact on gonad development, antioxidant status, color parameters, and carotenoid composition in adult female swimming crab (Portunus trituberculatus). Four diets, designated as Diet 1 to 4, were formulated to contain 3.07, 53.78, 103.23, and 205.22 mg/kg dry diets of synthetic β‐carotene. The findings revealed the following: (1) No significant differences in body weights or feed utilization were observed, indicating that β‐carotene did not significantly enhance the growth of adult P. trituberculatus. (2) The inclusion of 50–100 mg/kg β‐carotene in the diets led to improvements in antioxidant capacity indicators (T‐AOC, MDA) and immune‐related parameters (AKP, ACP) in both hepatopancreas and hemolymph. (3) Notably, the redness of ovaries and hepatopancreas increased significantly with higher dietary β‐carotene supplementation for color enhancement. In conclusion, incorporating β‐carotene into the diet enhanced the antioxidative capability, nonspecific immunity, and tissue carotenoid contents in adult female P. trituberculatus, and the optimal level of dietary synthetic β‐carotene supplementation for adult female P. trituberculatus was suggested to be around 100 mg/kg. Cold stress causes considerable damage to tender tea seedlings. Previous studies have explored changes in the physiological and biochemical factors of tea in response to cold stress; however, the mechanisms of cold resistance in ancient tea tree plants are unclear. The aim of this study was to analyze the effects of 0 °C cold stress for 15 days and 24 °C ambient temperature recovery for 5 days on the physiological and biochemical characteristics of two representative old tea varieties: Dali tea and Siqiu tea. The results revealed significant changes in antioxidant, photosynthetic efficiency, and physiological and biochemical indicators in response to cold stress, with the two species exhibiting different patterns. Cold stress decreased chlorophyll and carotene content, Fv/Fm, Y(II), non-photochemical quenching coefficient, photochemical quenching, and superoxide dismutase (SOD) activity, and increased intercellular CO2 concentration and ascorbate peroxidase activity. Siqiu tea showed a higher increase in soluble sugar content and antioxidant enzyme activity and a lower accumulation of malondialdehyde and minimal fluorescence (F0) than Dali, indicating a greater tolerance to cold stress. Based on partial least-squares discriminant analysis, six key differential physiological indicators of cold resistance—water-soluble sugar, F0, peroxidase, catalase, SOD, and gas conductance—were identified. Introduction: The African oil palm (Elaeis guineensis Jacq.) is the predominant oil crop in the world. In addition to triacylglycerols, crude palm oil (CPO) extracted from the mesocarp of the fruits, contains high amounts of provitamin A (carotenes) and vitamin E (tocochromanols). Because of their unsaturated nature, the carotenes are prone to oxidation and therefore are in part limiting for the shelf life of CPO. Results: In the present study, a wild Oil Palm tree (C59) from Cameroon was identified that lacks α-tocopherol and α-tocotrienol and instead accumulates the respective γ forms, suggesting that the activity of γ-tocopherol methyltransferase (VTE4) was affected. Sequencing of the VTE4 locus in the genome of plant C59 identified a G/C polymorphism that causes the exchange of a highly conserved tryptophan at position 290 with serine. The W290S exchange renders the VTE4 enzyme inactive, as shown after expression in Escherichia coli and Arabidopsis thaliana. The oxidative stability of carotenes in the mesocarp of the wild palm C59 was enhanced compared with control accessions. Furthermore, supplementation of commercial palm oil with different tocochromanols showed that γ-tocotrienol exerts a stronger effect during the protection of carotenes against oxidation than α-tocotrienol. Methods: A tree with unusual toochromanol composition was identified by HPLC screening of the mesocarp of wild trees. Polymorphisms in a candidate gene were identified by DNA sequencing. The candidate protein was heterologously expressed in Escherichia coli coli and Arabidopsis thaliana to test for enzyme activity. Oxidative stability of the CPO was studied by following carotene degradation over time. The liver, being the most metabolically active organ, is highly vulnerable to damage caused by oxidative stress. Rosa davurica Pall. seed oil (RDPO), a novel vegetable oil, and its bioactive components have been extensively researched in the field of antioxidants. In this research, the antioxidant properties and hepatoprotection by RDPO were evaluated. A series of antioxidant evaluation systems and a CCl4-induced acute liver injury model in mice were used to investigate the antioxidant activity and hepatoprotective efficacy of RDPO. The results showed that the extraction rate of RDPO was 11.12% using the optimal extraction process. Three major unsaturated fatty acids of the oil were α-linolenic acid (11.89 ± 0.017%), linoleic acid (18.52 ± 0.072%), and oleic acid (11.54 ± 0.425%). Furthermore, its antioxidant small-molecule compounds were β-sitosterol (1.429 ± 0.002 μg/g), α-tocopherol (1.273 ± 0.079 μg/g), β-carotene (0.012 ± 0.001 μg/g), lycopene (0.108 ± 0.002 μg/g), squalene (178.950 ± 0.794 μg/g), total polyphenols (1.114 ± 0.032 μg GAE/mg), and total flavonoids (0.504 ± 0.009 mg RU/g), respectively. In vitro, RDPO significantly inhibited the production of ABTS+•, DPPH•, O2•−, and hydroxyl radicals, as well as Fe3+. In vivo, RDPO significantly reversed the activity of total superoxide-dismutase, catalase, L-glutathione, and the level of malondialdehyde (MDA) in liver tissue. It also obviously inhibited the activity of aspartate transaminase (AST) and the level of MDA in the serum. Therefore, RDPO has demonstrated excellent antioxidant activity and a potential liver protective effect. This effect may be ascribed to its capacity for decreasing AST activity, inhibiting lipid peroxidation, and boosting endogenous antioxidant enzyme activity. • Among Korean adults ≥65y, 25% of men and 36% of women had low muscle strength. • Only 16% of Korean adults met the recommended nutrient intake of dietary vitamin C. • Dietary vitamin C intake was correlated with handgrip strength in Korean women. • Korean women ≥65y with low dietary vitamin C had a higher risk of possible sarcopenia. Although vitamin C is one of the most important antioxidants, its effect on muscle quality is not fully understood. Therefore, we hypothesized that low dietary vitamin C intake is associated with low muscle strength. To test the hypothesis, a single 24-h dietary recall and handgrip strength test of 10,883 younger adults 19-64 y and 3,961 older adults ≥65 y from the seventh Korea National Health and Examination Survey (KNHANES VII 2016-2018) was analyzed by multivariable linear and logistic regression models, and low muscle strength was defined as handgrip strength <28 kg for men and <18 kg for women. Approximately 15.5% of Korean adults met the recommended intake of dietary vitamin C, and those with higher dietary vitamin C intake had higher total energy and protein intake. After adjusting for confounding variables, including age, body mass index, total energy intake, household income, alcohol consumption, smoking, resistance exercise, medical condition, and dietary intake of protein, vitamin E, and β-carotene, dietary vitamin C was correlated with maximal handgrip strength in younger women 19-64 y (β = 0.002; SE = 0.001; P -value =.026) and older women ≥65 y (β = 0.005; SE = 0.002; P -value =.013). Among older women ≥65 y, those in the lowest quartile of dietary vitamin C intake had a higher risk of low muscle strength compared to those in the highest quartile after adjustment of confounding factors (odds ratio, 2.16; 95% confidence interval, 1.49-3.15). These results imply that adequate dietary vitamin C intake may reduce the risk of sarcopenia in older Korean women. Dietary vitamin C was shown to be inversely associated with the risk of low muscle strength in older Korean women. Although no association between the two variables among younger men 19-64 y, older men ≥65 y, and women 19-64 y was observed, older Korean women ≥65 y in the lowest quartile of vitamin C intake had significantly higher odds of low muscle strength compared to those in the highest quartile with adjustment for confounding variables (OR, 2.16; 95% CI, 1.49-3.15). A high mortality rate in the gold rabbitfish larvae of Siganus guttatus has been found during the initial developmental phase. This condition is thought to be due to unprepared larvae obtaining exogenous feed. Various efforts have been conducted to prevent high mortality rates, such as nutrient enrichment. This study was performed to evaluate the combined effect of βcarotene and taurine on the growth performance and survival rate of the gold rabbitfish larvae of S. guttatus. This study had four treatments for rotifer enrichment, namely BT0 without β-carotene and taurine enrichment, B7.5 (7.5mg L -1 β-carotene), T50 (50mg L -1 taurine), and a combination of 7.5mg L -1 β-carotene and 50mg L -1 taurine (Com-BT). The S. guttatus larvae were reared for 10 days after hatching (DAH). The parameter observed was growth performance, which contained absolute growth (body length and weight), fin development (dorsal, pectoral, and caudal), visual development (eye diameter and retina), total prey intake, and survival rate. The results indicated that enrichment of the rotifers with β-carotene and taurine can improve the absolute growth rate, fin development, visual development, and total prey intake. These parameters obtained better values as both ingredients were combined together (P< 0.05) Com-BT. Nutrient enrichment with β-carotene and taurine had no significant difference (P< 0.05) on the survival rate of S. guttatus larvae, but significantly showed a better survival rate in the combination of β-carotene and taurine treatment (Com-BT). Effective utilization of glucose, xylose, and acetate, common carbon sources in lignocellulose hydrolysate, can boost biomanufacturing economics. However, carbon leaks into biomass biosynthesis pathways instead of the intended target product remain to be optimized. This study aimed to enhance α-carotene production by optimizing glucose, xylose, and acetate utilization in a high-efficiency Corynebacterium glutamicum cell factory. Heterologous xylose pathway expression in C. glutamicum resulted in strain m4, exhibiting a two-fold increase in α-carotene production from xylose compared to glucose. Xylose utilization was found to boost the biosynthesis of pyruvate and acetyl-CoA, essential precursors for carotenoid biosynthesis. Additionally, metabolic engineering including pck, pyc, ppc , and aceE deletion, completely disrupted the metabolic connection between glycolysis and the TCA cycle, further enhancing α-carotene production. This strategic intervention directed glucose and xylose primarily towards target chemical production, while acetate supplied essential metabolites for cell growth recovery. The engineered strain C. glutamicum m8 achieved 30 mg/g α-carotene, 67% higher than strain m4. In fed-batch fermentation, strain m8 produced 1802 mg/L of α-carotene, marking the highest titer reported to date in microbial fermentation. Moreover, it exhibited excellent performance in authentic lignocellulosic hydrolysate, producing 216 mg/L α-carotene, 1.45 times higher than the initial strain (m4). Some physicochemical characteristics and elemental composition of refined sunflower oil, as well as linseed oil added to it, were investigated; linseed oil and olive oil; truffle oil and rosemary oil. Fatty acid analysis shows substantial increases in monounsaturated fatty acids with the addition of truffle and rosemary oils (up to about 78 %). With the same supplements, a significant oxidative stability over 20 hours was also observed. High concentrations of chlorophyll were found with the addition of rosemary oils and oils of linseed oil and olive oil. ß-carotene was affected three to six times in all supplements compared to the commonly refined oil. Eight elements (Mg, Cr, Mn, Zn, Ni, As, Pd and Cd) were analyzed in the studied oils, no presence of toxic elements As and Cd (< 0.02 mg kg-1), lead was up to 0.04 mg kg-1. The remaining elements vary in different concentrations depending on the additive oils used. The fluorescence spectra of the tested samples were obtained for excitation wavelengths of 380 nm, and the fluorescence maxima allowed to determine the relationship between the optical and chemical properties of the samples. Moderate non‐covalent interaction of protein and polyphenols can improve the emulsifying property of protein itself. The corn protein hydrolysate (CPH) and tannic acid (TA) complex was successfully used to construct nanoemulsion for algal oil delivery. There has been no study on the feasibility of this nanoemulsion delivery system for other food functional components, for example, β‐carotene (β‐CE). CPH/TA complex‐based nanoemulsion system for β‐CE delivery was studied, focusing on the effect of β‐CE content on the physicochemical stability of the nanoemulsions. The nanoemulsion delivery systems (dia. 150 nm) with low viscosity and good liquidity were easily fabricated by two‐step emulsification. The nanoemulsions with high β‐CE content (>71.5 μg/mL) significantly increased (p <.05) the emulsion droplet size. However, there was no significant (p >.05) effect of β‐CE content on polydispersity index (PDI) and zeta potential of the nanoemulsions. The storage (30 days) experiment results demonstrated that the droplet size of the nanoemulsions with varying β‐CE content increased slightly during storage. However, the PDI values showed a slightly decreasing trend. Zeta potentials of the nanoemulsions showed no noticeable change during storage. Moreover, after storage of 30 days, the retention ratios of β‐CE were found to be up to 90%, which suggests an excellent protective effect for β‐CE by the nanoemulsion systems. The CPH/TA complex stabilized nanoemulsions could aggregate in gastric condition, but the β‐CE content did not have obvious effect on the digestive stability of the nanoemulsions. β-Carotene is an attractive compound and that its biotechnological production can be achieved by using engineered Saccharomyces cerevisiae. In a previous study, we developed a technique for the efficient establishment of diverse mutants through the introduction of point and structural mutations into the yeast genome. In this study, we aimed to improve β-carotene production by applying this mutagenesis technique to S. cerevisiae strain that had been genetically engineered for β-carotene production. Point and structural mutations were introduced into β-carotene-producing engineered yeast. The resulting mutants showed higher β-carotene production capacity than the parental strain. The top-performing mutant, HP100_74, produced 37.6 mg/L of β-carotene, a value 1.9 times higher than that of the parental strain (20.1 mg/L). Gene expression analysis confirmed an increased expression of multiple genes in the glycolysis, mevalonate, and β-carotene synthesis pathways. In contrast, expression of ERG9, which functions in the ergosterol pathway competing with β-carotene production, was decreased in the mutant strain. The introduction of point and structural mutations represents a simple yet effective method for achieving mutagenesis in yeasts. In yeast metabolic engineering, there is a need for technologies that simultaneously suppress and regulate the expression of multiple genes and improve the production of target chemicals. In this study, we aimed to develop a novel technology that simultaneously suppresses the expression of multiple genes by combining RNA interference with global metabolic engineering strategy. Furthermore, using β-carotene as the target chemical, we attempted to improve its production by using the technology. First, we developed a technology to suppress the expression of the target genes with various strengths using RNA interference. Using this technology, total carotenoid production was successfully improved by suppressing the expression of a single gene out of 10 candidate genes. Then, using this technology, RNA interference strain targeting 10 candidate genes for simultaneous suppression was constructed. The total carotenoid production of the constructed RNA interference strain was 1.7 times compared with the parental strain. In the constructed strain, the expression of eight out of the 10 candidate genes was suppressed. We developed a novel technology that can simultaneously suppress the expression of multiple genes at various intensities and succeeded in improving carotenoid production in yeast. Because this technology can suppress the expression of any gene, even essential genes, using only gene sequence information, it is considered a useful technology that can suppress the formation of by-products during the production of various target chemicals by yeast. The integration of β-carotene into functional foods is limited due to its limited hydrophilicity and chemical stability. To address this issue, emulsion was encouraged to be utilized. Monolayer, composite, and bilayer emulsions were prepared using pea protein isolate (PPI) alone or in combination with poly-γ-glutamic acid (γ-PGA). Our findings indicate that the bilayer emulsion utilizing γ-PGA as an external emulsifier exhibited the smallest droplet size (737.61 nm) and highest zeta potential (−35.23 mV). Under various environmental stresses, the chemical stability of the composite emulsion surpassed that of the monolayer emulsion but was inferior to that of the bilayer emulsion. Moreover, following simulated gastrointestinal digestion, the β-carotene bioaccessibility in the composite emulsion (12.76 %) exceeded that in the monolayer emulsion (6.57 %) but was less than that in the bilayer emulsion (20.01 %). Therefore, the emulsion preparation method or the sequence of γ-PGA addition significantly affects emulsion stability. By making use of the outstanding osteoinductive effects of β‐carotene, in this innovative research, we investigate the potential for application of β‐carotene‐reinforced PMMA resins. Different amounts of β‐carotene, from 0% to 5%, have been mixed with standard bone cements and characterized by various spectroscopic and microscopic techniques before testing with KUSA‐A1 murine mesenchymal cells. In vitro results showed that not only the amount of bone produced by the cells on the composite is comparable if not superior to modern bioglasses but also both adhesion and cellular proliferation are strongly promoted by the presence of β‐carotene. The increased biological properties came at the price of a small loss in elastic modulus, but it was observed that the presence of β‐carotene leads to an increase of ultimate strength, reaching an increase of about 30% at a concentration of about 2.5%. Sweet cherry grown under sustainable management produces highly valuable fruits, whose quality shows important biochemical and morphological changes during ripening. Research was carried out in Iasi (Romania), with the aim to assess the quality characteristics of the sweet cherry fruits of three cultivars (Van, Andreiaș, Margonia), grown in an inner or outer position inside the tree crown, at the pre-ripening or full ripeness phase. In 2022, the colour component a* showed higher values in cv. Van and Andreias red fruits and in an inner position, whereas the components L* and b* at the full ripeness phase were highest in cv. Margonia. The dry matter and total soluble solids contents increased from the pre-ripening to the full ripeness phase and were highest in cv. Van sweet cherry fruits; the DM of fruit from the outer part of crown was higher than that of fruit from the inner part at the pre-ripening phase. The content of phenolics was the highest in cv. Margonia fruits at the pre-ripening stage and in cv. Van at the full ripeness phase and higher in the inner tree crown zones. The cultivar Margonia generally showed the highest vitamin C content in both years and development phases. The yellow fruit cv. Margonia mostly showed the highest values of chlorophyll a and b. The fruit's content of carotene, lycopene, and anthocyanins was generally the highest in the red fruits of cv. Andreias. Protein-polysaccharide emulsion hydrogel, as an encapsulation system of active substances, has the advantages of high loading capacity, biocompatibility and stability. In this work, Spirulina protein isolate (SPI) and hyaluronic acid (HA) were used as emulsifier and gel matrix to prepare SPI emulsions, SPI-HA emulsions, and emulsion hydrogels with various oil phase volume fractions. The microstructure and droplet sizes of the emulsions were characterized. The effect of oil phase volume fractions on the formation of emulsion hydrogel was analyzed, and the protective effect of emulsion hydrogel on β-carotene under high temperature was studied. The results showed that the droplet sizes of SPI emulsion decreased obviously with the introduction of HA. The structure of emulsion hydrogel became much denser with the increase of oil phase volume fraction. When the oil phase volume fraction was 60%, the yield stress, storage modulus and apparent viscosity of emulsion hydrogel were higher than those of SPI emulsion and SPI-HA emulsion. BACKGROUND: Little information is available on α‐ and β‐amylase activity and their associations with starch, sugars and other culinary qualities in sweetpotato. The present study aimed to assess sweetpotato storage root α‐ and β‐amylase activity in relation to starch, sugars, β‐carotene content and storage root flesh color. RESULTS: α‐ and β‐amylase activity (α‐AA and β‐AA) were assayed from a Tanzania (T) × Beauregard (B) genetic mapping population in their uncured (raw), cured and stored (approximately 11 weeks) forms during 2016 and 2017. Ceralpha and Betamyl methods, with modifications to suit a high‐throughput microplate assay format, were used to quantify α‐AA and β‐AA, respectively. Storage root dry matter, starch, glucose, fructose, sucrose and β‐carotene content were predicted using near infrared reflectance spectroscopy. There was little relationship (r2 = 0.02–0.08, P ≤ 0.05 in 2016 and r2 = 0.05–0.11, P ≤ 0.05 in 2017) between α‐AA and β‐AA. We observed negative linear associations between α‐AA and dry matter content and generally no correlations between β‐AA and dry matter content. β‐AA and sugars were weakly positively correlated. β‐AA and β‐carotene content were positively correlated (r = 0.3–0.4 in 2016 and 0.3–0.5 in 2017). CONCLUSION: Generally, the correlation coefficient for amylase enzyme activity and sugar components of storage roots at harvest increased after curing and during post‐harvest storage. The present study aimed to prepare a multilayer complex of polysaccharides for encapsulation of β-carotene for delivery into aqueous media. β-carotene, a highly beneficial carotenoid for human health, can be effectively combined with three polysaccharides that serve as prebiotics in the human gut microbiota. The process of preparing the β-carotene-polysaccharides complex involved a three-step method. Firstly, β-carotene was encapsulated using xylan. Subsequently, the encapsulated β-carotene was complexed with chitooligosaccharides. Finally, the resulting double-layer complex was mixed with fucoidan to form a three-layer complex that readily dissolves in aqueous media. The formation of the complex was verified through SEM and spectroscopic analyses. The complex exhibited a favourable loading capacity ranging from 0.144 to 0.293 mg/mL, with sizes varying between 205 and 297 nm. The solubility of the complex ranged from 26.84 to 29.04 mg/mL. Notably, the complex demonstrated antioxidant activity as observed by DPPH and FRAP methods. These remarkable properties make the new complex a potential candidate for incorporation into the food and beverage industries, thanks to its antioxidant capabilities and advantageous features. Algae constitute a significant part of marine biodiversity. They represent a renewable source of bioactive metabolites from drug development and therapeutic fields. Fucoxanthin and β-carotene from the brown macroalgae Halopteris scoparia, were extracted using conventional organic solvent extraction, then purified, to homogeneity, based on various chromatographic principles. Their effects on digestive enzymes and harmful bacteria were investigated. The capacities of both purified pigments to inhibit α-amylase and trypsin enzymes were evaluated. Purified fucoxanthin and β-carotene exhibited interesting α-amylase inhibition activities, with IC50 of 300 and 500 µg/mL, respectively. Moreover, trypsin inhibition activities were detected using purified these two pigments. The antibacterial potential of the purified pigments was evaluated. β-carotene showed to be a great antibacterial natural compound against gram-positive and gram-negative bacteria such as Listeria monocytogenes, Staphylococcus aureus and Salmonella enterica with Minimal Inhibitory Concentration (MIC) of about 0.225, 0.1125, 0.225 µg/mL, respectively. Those findings are in favor of the exploitation of H. scoparia pigments in therapeutic fields as an antidiabetic source directly by the inhibition of α-amylase and trypsin as well as antibacterial agents against gastrointestinal infections. Production of carotenoids by yeast fermentation is an advantaged technology due to its easy scaling and safety. Nevertheless, carotenoid production needs an economic culture medium and other efficient yeast stains. The study aims to isolate and identify a yeast strain capable of producing carotenoids using a cost-effective substrate. A new strain was identified as Rhodotorula toruloides L/24-26-1, which can produce carotenoids at different pretreated and unpretreated sugarcane molasses concentrations (40 and 80 g/L). The highest biomass concentration (18.6 ± 0.6 g/L) was reached in the culture using 80 g/L of hydrolyzed molasses. On the other hand, the carotenoid accumulation reached the maximum value using pretreated molasses at 40 g/L (715.4 ± 15.1 µg/g d.w). In this case, the β-carotene was 1.5 times higher than that on the control medium. The yeast growth in molasses was not correlated with carotenoid production. The most outstanding production of The DPPH, ABTS, and FRAP tests demonstrated the antioxidant activity of the obtained carotenogenic extracts. This research demonstrated the R. toruloides L/24-26-1 strain biotechnological potential for carotenoid compounds. Carotenoid cleavage oxygenases can cleave carotenoids into a range of biologically important products. Carotenoid isomerooxygenase (NinaB) and β, β-carotene 15, 15′-monooxygenase (BCO1) are two important oxygenases. In order to understand the roles that both oxygenases exert in crustaceans, we first investigated NinaB-like (EsNinaBl) and BCO1-like (EsBCO1l) within the genome of Chinese mitten crab (Eriocheir sinensis). Their functions were then deciphered through an analysis of their expression patterns, an in vitro β-carotene degradation assay, and RNA interference. The results showed that both EsNinaBl and EsBCO1l contain an RPE65 domain and exhibit high levels of expression in the hepatopancreas. During the molting stage, EsNinaBl exhibited significant upregulation in stage C, whereas EsBCO1l showed significantly higher expression levels at stage AB. Moreover, dietary supplementation with β-carotene resulted in a notable increase in the expression of EsNinaBl and EsBCO1l in the hepatopancreas. Further functional assays showed that the EsNinaBl expressed in E. coli underwent significant changes in its color, from orange to light; in addition, its β-carotene cleavage was higher than that of EsBCO1l. After the knockdown of EsNinaBl or EsBCO1l in juvenile E. sinensis, the expression levels of both genes were significantly decreased in the hepatopancreas, accompanied by a notable increase in the redness (a*) values. Furthermore, a significant increase in the β-carotene content was observed in the hepatopancreas when EsNinaBl-mRNA was suppressed, which suggests that EsNinaBl plays an important role in carotenoid cleavage, specifically β-carotene. The present investigation was performed to evaluate the effects of various synthetic antioxidants (vitamin A, vitamin E, β-carotene, and BHT) on the oxidation of sunflower oil subjected to accelerated thermal storage at 60 °C for three months (12 weeks). The performance of the antioxidants studied was evaluated using several quality parameters: the free fatty acid value (FFA), primary oxidation (via the peroxide value (PV) and K232 value), secondary oxidation products (via the anisidine value (p-AV) and K270 value), and the total oxidation value (TOTOX). The fatty acid composition (FAC), oxidizability value (COX), iodine value (IV), and pigment content (chlorophyll and carotenoid) were also evaluated. The results revealed that the control sample of sunflower oil exhibited higher susceptibility to oxidative deterioration. Antioxidants at 200 ppm were more effective in preserving the oxidative stability of sunflower oil subjected to accelerated storage compared to the control oil. The smallest increases in all stability parameter indexes were recorded for antioxidant-supplemented sunflower oil. However, the IV and chlorophyll and carotenoid contents were reduced. At 200 ppm, vitamin E and β-carotene showed the greatest stability in sunflower oil, while their combination with vitamin A at 100 ppm of each showed the lowest stability. In addition, synthetic antioxidants provided greater protection against the degradation of polyunsaturated fatty acids (PUFAs). The highest level of PUFA degradation was recorded in the control oil, followed by the oil containing vitamin A. In conclusion, adding synthetic antioxidants to sunflower oil improves its stability during storage. A better understanding of how emulsifier type could differently influence the behavior of nanostructured lipid carriers (NLC) under the gastrointestinal digestion process, as well as at the cellular level, is of utmost importance for the NLC‐based formulations' optimization and risk assessment in the food field. In this study, NLC composed by fully hydrogenated soybean and high‐oleic sunflower oils were prepared using soy lecithin (NLC Lβ) or Tween 80 (NLC Tβ) as an emulsifier. β‐Carotene was entrapped within NLC developed as a promising strategy to overcome β‐carotene's low bioavailability and stability. The effect of emulsifier type on the digestibility of β‐carotene‐loaded NLC was evaluated using an in vitro dynamic digestion model mimicking peristalsis motion. The influence of β‐carotene‐loaded NLC on cell viability was assessed using Caco‐2 cells in vitro. NLC Tβ remained stable in the gastric compartment, presenting particle size (PS) similar to the initial NLC (PS: 245.68 and 218.18 nm, respectively), while NLC Lβ showed lower stability (PS > 1000 nm) in stomach and duodenum phases. NLC Tβ also provided high β‐carotene protection and delivery capacity (i.e., β‐carotene bioaccessibility increased 10‐fold). Based on the results of digestion studies, NLC Tβ has shown better physical stability during the passage through the in vitro dynamic gastrointestinal system than NLC Lβ. Moreover, the developed NLC did not compromise cell viability up to 25 µg/mL of β‐carotene. Thus, the NLC developed proved to be a biocompatible structure and able to incorporate and protect β‐carotene for further food applications. Practical Application: The findings of this study hold significant implications for industrial applications in terms of developing nanostructured lipid carriers from natural raw materials widely available and used to produce other lipid‐based products in the food industry, as an alternative to synthetic ones. It was largely observed that the increased MFE had a significant impact on the overall growth and to a lesser extent on the protein and beta-carotene production. For TR1 in general, the samples treated with the increased MFE demonstrated a higher growth rate than the control. Specifically, for the specimen treated with 40 min of the increased MFE, the growth on the 21st day was measured to be 38% higher than the control. For the specimen treated with 120 min of the increased MFE, the protein content on the 15th day was measured to be 15.6% higher than the control. For the specimen treated with 40 min of the increased MFE, the beta-carotene content on the 15th day was measured to be 20.4% higher than the control. For TR2 in general, the results were inferior compared to TR1 but showed higher production than the control specimen. Specifically, for the specimen treated with 80 min of the increased MFE, the protein content on the 21st day was measured to be 4.3% higher than the control. For the specimen treated with 100 min of the increased MFE, the beta-carotene content on the 15th day was measured to be 17.1% higher than the control. For the specimen treated with 100 min of the increased MFE, the growth on the 21st day was measured to be 5% higher than the control. Optimized sugar-added passion fruit juice (OPFJ) was obtained after optimization of thermosonication treatments of passion fruit juice using hybrid Artificial neural network/Genetic algorithm. The quality parameters and shelf life of OPFJ stored at various temperatures were assessed. Baranyi and Ratkowsky's square-root models proved the prediction accuracy of the theoretical model concerning the microbial population during storage. The lag phase value decreased with increasing storage temperature. Temperature rise from 8 to 25 °C decreased the shelf life from 17 to 8 days for OPFJ and 3.5 to 1.5 days for untreated juice. The activation energy and increase in the reaction rate on increase in temperature by 10 °C (Q10 value) were also evaluated. Thermosonication increased the phenolic and β-carotene contents and maintained the pH, titratable acidity, ascorbic acid content, and antioxidant activity during storage. Background: Purple sweet potato productivity in Indonesia is notably high, particularly in Bondowoso at 213.84 quintals/ha. This variant of sweet potato is rich in betacarotene, with a content of 4237 µg/100 g, known for its ability to protect cells from free radicals and reduce the risk of diabetes mellitus. Notably, Indonesia ranks third globally in the prevalence of prediabetes, affecting 27.7 million people. In 2022, in Tapen District, Bondowoso, the prevalence of prediabetes, based on Tapen Health Center data, was recorded at 12.36%. Objectives: Analyzing betacarotene, total sugar, and organoleptic propeties of purple sweet potato jam as a prediabetes distraction food. Methods: A quasi-experimental design with a post-test-only control group was employed. The research involved producing purple sweet potato jam with varying ratios of cassava sugar and purple sweet potato (X0: 0% cassava sugar; 100% purple sweet potato; X1: 15% cassava sugar; 85% purple sweet potato; X2: 30% cassava sugar; 70% purple sweet potato; X3: 45% cassava sugar; 55% purple sweet potato). Analysis included determining betacarotene content using the UV-Visible Spectrophotometry method, total sugar using the Luff Schoorl method, and organoleptic evaluation through the hedonic test method. Data were subjected to statistical analysis using the One-Way ANOVA test for betacarotene and total sugar levels, followed by Duncan's post-hoc test. Organoleptic test data were analyzed using the Kruskall Wallis test and Dunn's post-hoc test. Results: The average betacarotene content in treatments X0, X1, X2, and X3 were 4230.3 µg/100 g; 3464 µg/100 g; 2955.6 µg/100 g; and 2257.3 µg/100 g, respectively. Total sugar content averages for treatments X0, X1, X2, and X3 were 2.2%; 13.7%; 27.5%; and 39.2%, respectively. The panelists’ preference indicated that sample X3 (45% cassava sugar and 55% purple sweet potato) was the most favored jam. Conclusions: Significant differences were observed in beta-carotene, total sugar, and organoleptic levels of purple sweet potato jam, suggesting that the variation in purple sweet potato and cassava sugar ratios influenced the beta-carotene and total sugar levels in the jam. The nutritional composition of bovine liver, particularly in terms of vitamins E, β-carotene, and cholesterol concentration, is significantly influenced by the cattle-rearing system and diet. This study aimed to elucidate the impact of four predominant cattle-rearing systems in the Eastern Amazon region on the vitamin E, β-carotene content, and cholesterol levels in bovine liver during the rainy season. Liver-tissue samples were collected from 48 cattle, with twelve specimens representing each rearing system. The systems encompassed two native pastures located in flood-prone areas, cultivated pastures on upland terrain and a confinement-rearing system. Our findings indicate that, when considering all rearing systems collectively, there were no significant differences in the levels of vitamins and cholesterol in the liver tissues (p > 0.05). However, a marked disparity in cholesterol levels emerged when comparing intensive rearing systems to extensive ones, with the former demonstrating notably higher concentrations (p = 0.01). Additionally, the intensive rearing system was associated with elevated levels of β-carotene (p < 0.01), α-tocopherol (p = 0.01), and β-tocopherol (p = 0.01) relative to the extensive systems. No significant variations were observed amongst the different extensive systems (p > 0.05). These results suggest that, while all rearing systems yielded liver tissues with typical concentrations of vitamins and cholesterol, the intensive rearing system led to a higher accumulation of certain vitamin compounds. This study highlights the substantial nutritional implications of different cattle-rearing systems in the Eastern Amazon and provides valuable insights for developing dietary strategies to optimize the nutritional quality of bovine liver. The high rate of metabolic activity during breeding, pregnancy, parturition and lactation may lead to oxidative stress in sheep and goats. Antioxidant supplementation is proposed as a therapy for encountering resultant stress. In animals, β-carotene functions mainly as pro-vitamin A and as a scavenger of free radicals, especially against singlet-state oxygen. This raises the question of whether supplementing β-carotene as an antioxidant above the vitamin A requirement is beneficial. While a number of studies have been conducted on the effect of β-carotene supplementation as a tool to improve the production and reproduction performance of female goats and sheep, the findings are inconsistent. Some of the main reasons for this inconsistency could be attributed to the variation in the amount of β-carotene supplemented, the amount of β-carotene in the animal diet and the difference in the animal requirement for β-carotene. The objective of this review was to determine the effects of β-carotene supplementation on the production and reproductive performance of female sheep and goats fed on a diet deficient or not deficient in β-carotene. Data were extracted from peer-reviewed articles that were searched in the Web of Science, Google Scholar and Scopus databases. Thirteen peer-reviewed published articles were found on the subject under review. We estimated the animal requirement for β-carotene, the amount of β-carotene provided to the animal in the diet and the amount of β-carotene supplementation used in each study. These estimations were made to indicate whether the diet fulfilled the animal's requirement for β-carotene or not. The review revealed that β-carotene supplementation enhances the production and reproduction performance of the sheep when fed on a carotene-deficient diet. Additionally, β-carotene supplementation may increase the levels of some hormones such as progesterone in sheep and goats when fed on a diet that fulfils their requirements of β-carotene. However, the supplementation of β-carotene above the requirements seems to not affect the production and reproduction parameters such as body weight, milk production, estrus parameters, conception rate, pregnancy rate, and litter size. In conclusion, while β-carotene supplementation is essential for female sheep and goats fed β-carotene-deficient diets to attain optimal production and reproductive performance, supplementing β-carotene above the requirements does not appear to provide any further benefits to their overall performance. The biological activities of hesperidin-related compounds, such as hesperetin laurate (HTL), hesperetin (HT), hesperidin (HD), and hesperidin glucoside (HDG), were investigated in vitro. The compounds showed different hydrophobicities, and the octanol–water partition coefficient log P were 7.28 ± 0.06 for HTL, 2.59 ± 0.04 for HT, 2.13 ± 0.03 for HD, and −3.45 ± 0.06 for HDG, respectively. In the DPPH assay and β-carotene bleaching assay to determine antioxidant capacity, all compounds tested showed antioxidant activity in a concentration-dependent manner, although to varying degrees. HTL and HT showed similarly high activities compared to HD or HDG. HD and HDG did not show a significant difference despite the difference in solubility between the two. Cytotoxicity was high; in the order of hydrophobicity—HTL > HT > HD > HDL in keratinocyte HaCaT cells. All compounds tested showed reducing effects on cellular inflammatory mediators and cytokines induced by UV irradiation. However, HTL and HT effectively reduced nitric oxide (NO), tumor necrosis factor α (TNF-α), and interleukin-6 (IL-6) levels compared to HD and HDG. The inhibitory effects of hesperidin-related compounds on skin-resident microorganisms were evaluated by measuring minimum inhibitory concentration (MIC). HTL showed the highest inhibitory effects against Staphylococcus aureus, Cutibacterium acnes, Candida albicans, and Malassezia furfur, followed by HT, while HD and HDF showed little effect. In conclusion, the hydrophobicity of hesperidin-related compounds was estimated to be important for biological activity in vitro, as was the presence or absence of the sugar moiety. Dunaliella salina is by far the most salt-tolerant organism and contains many active substances, including β-carotene, glycerol, proteins, and vitamins, using in the production of dried biomass or cell extracts for the biofuels, pharmaceutical formulations, food additives, and fine chemicals, especially β-carotene. We report a high-quality genome sequence of D. Salina FACHB435, which has a 472 Mb genome size, with a contig N50 of 458 Kb. A total of 30,752 proteincoding genes were predicted. The annotation results evaluated by BUSCO was shown that completeness was 91.0% and replication was 53.1%. The fragments were 6.3% and the deletions were 2.6%. Phylogenomic and comparative genomic analyses revealed that A. thaliana diverged from Volvocales about 448 million years ago, then Volvocales C. eustigma, D. salina, and other species diverged about 250 million years ago. High light could promote the accumulation of β-carotene in D. salina at a 13 d stage of culture. The enrichment of DEGs in KEGG, it notes that the predicted up-regulated genes of carotenoid metabolic pathway include DsCrtB, DsPDS, DsZ-ISO, DsZDS, DsCRTISO, DsLUT5, DsCrtL-B, and DsCCD8, while the predicted down-regulated genes include DsCrtF, and DsLUT1. The four genes that were both up-regulated and down-regulated were DsZEP, DsCrtR-b, DsCruA/P and DsCrtZ 4. This study represents a pioneering investigation and comparative analysis of lipid extracts from four different colors of peach palm (Bactris gasipaes Kunt) fruits—red, yellow, green, and white—by employing a green method based on ethanolic ultrasound-assisted extraction. This study examined the extraction yield, physico-chemical-quality attributes, chromatographic profiles (GC), color measurements, total carotenoid content, differential thermogravimetry (TG/DTA), and infrared spectroscopy (FTIR). The obtained lipid extracts displayed a high quality, considering the physico-chemical parameters of the Codex Alimentarius, and a fatty acids profile characterized by unsaturated fatty acids, notably omegas (ω-3, ω-6, and ω-9). The indices of atherogenicity (A.I.), thrombogenicity (I.T.), and hypocholesterolemic and hypercholesterolemic ratios revealed superior outcomes for the red peach palm lipid extract (approximately 0.35, 0.52, and 2.75, respectively), along with higher levels of β-carotene (748.36 µg of β-carotene per 100 g−1 of lipid extract) compared to the yellow, green, and white counterparts. Consequently, this research successfully demonstrates the efficacy of using a green extraction method in preserving the lipid's quality, which can display cardiovascular functionality and thermal stability. These findings underscore the considerable potential of peach palm lipid extract as a valuable raw material for diverse industrial applications across various sectors. Carotenoids and chlorophylls are among the most widely distributed pigments in nature that play essential roles in the photosynthetic apparatus and confer diverse colours in plants. Among all vegetables, cauliflower (Brassica oleracea L. ssp. var. botrytis) is rich in phytochemicals and is an important crop grown all over the world. This study investigates carotenoid and chlorophyll concentrations in differently pigmented cultivars and elucidates the role of transcriptional regulation of carotenoid accumulation including lutein and β-carotene. Here, we characterised changes in pigments by UHPLC-DAD-ToF-MS and changes in transcript levels of carotenoid metabolic genes by qRT-PCR in florets and leaves of orange ('Jaffa' and 'Sunset'), purple ('Di Sicilia Violetto' and 'Graffiti'), green ('Trevi') and white ('Clapton') cultivars. Transcript levels of all carotenoid metabolic genes showed different transcript level patterns in the leaves and florets. Compared to the other cultivars, the orange cultivars had the highest levels of b-carotene in the florets and lutein in the leaves resulting in changes lutein/β-carotene ratios. In the green cultivar, higher transcript levels were also found, especially for phytoene synthase and phytoene desaturase genes of the core biosynthesis pathway. However, no increased carotenoid concentrations were observed, possibly due to a higher carotenoid turnover induced by the carotenoid cleavage dioxygenase 4 in the green cultivar. In the white ('Clapton') and purple ('Di Sicilia Violetto' and 'Graffiti') cultivars the phytoene desaturase transcript levels as well as carotenoid concentrations were low. Chlorophyll concentrations changed in trend comparable to the carotenoid concentrations and were only significantly lower in the leaves of the orange cultivar 'Jaffa'. Also, the chlorophyll a/b ratio changed in 'Jaffa'. In florets the highest chlorophylls concentrations were observed for the green cultivar ('Trevi') and the purple cultivar ('Di Sicilia Violetto'). β-Carotene is an orange fat-soluble compound, which has been widely used in fields such as food, medicine and cosmetics owing to its anticancer, antioxidant and cardiovascular disease prevention properties. Currently, natural β-carotene is mainly extracted from plants and algae, which cannot meet the growing market demand, while chemical synthesis of β-carotene cannot satisfy the pursuit for natural products of consumers. The β-carotene production through microbial fermentation has become a promising alternative owing to its high efficiency and environmental friendliness. With the rapid development of synthetic biology and in-depth study on the synthesis pathway of β-carotene, microbial fermentation has shown promising applications in the β-carotene synthesis. Accordingly, this review aims to summarize the research progress and strategies of natural carotenoid producing strain and metabolic engineering strategies in the heterologous synthesis of β-carotene by engineered microorganisms. The aim of this study was to develop a stable palm carotene-based emulsion using the rice protein isolate-flaxseed gum complex (RPI-FG) with different ratios (3:1, 5:1, 7:1, 9:1, 11:1). RPI achieved highest solubility at pH 12. Meanwhile, RPI-FG complex shows low turbidity value from pH 6 to 12 for all ratios. Emulsion with ratio 7:1 exhibited 33 % droplet size increment throughout 7 days storage and evenly distributed based on the microstructure images. For creaming index (CI), all emulsion shows 0 % separation on day zero except emulsion with ratio 11:1 (1.90 %) and lowest CI on day 7 was 9.05 % in ratio 3:1. Furthermore, emulsion activity index and emulsion stability index of the emulsions decreased significantly with the reduced of FG in the formulation. These results suggested that carotene-based emulsion fabricated using RPI-FG complex with ratio of 7:1 exhibited the highest emulsion stability and could be further applied in aqueous food systems. β-Carotene is one kind of the most important carotenoids. The major functions of β-carotene include the antioxidant and anti-cardiovascular properties, which make it a growing market. Recently, the use of metabolic engineering to construct microbial cell factories to synthesize β-carotene has become the latest model for its industrial production. Among these cell factories, yeasts including Saccharomyces cerevisiae and Yarrowia lipolytica have attracted the most attention because of the: security, mature genetic manipulation tools, high flux toward carotenoids using the native mevalonate pathway and robustness for large-scale fermentation. In this review, the latest strategies for β-carotene biosynthesis, including protein engineering, promoters engineering and morphological engineering are summarized in detail. Processing cricket-based products is essential to improve consumer acceptability. The effect of species and drying methods on the nutritional compositions of cricket powder (CP) obtained from Gryllus bimaculatus and Acheta domesticus was investigated. Drying methods included tray drying, oven drying, solar drying, and freeze drying, with drying time dependent on drying method to reach aw <0.6. Freeze-dried CP required the longest drying time, followed by solar-dried CP and tray-dried CP, whereas the shortest drying time was achieved by oven-dried CP. Nutritional compositions of CP after tray drying and freeze drying were comparable. CP obtained from the two cricket species showed high protein (59–70%) and fat (9.71–25.35%) content. All CPs had histamine content lower than 4 µg/g. The contents of two essential fatty acids, and eight essential six non-essential amino acids of both cricket species were comparable. CP from G. bimaculatus had noticeably higher levels of B-group vitamins (B1, B2, B5, B6, B12), vitamin A, and β-carotene and twice as much as cis-9-oleic acid compared to A. domesticus. Results suggested that tray drying was the most suitable to prepare CP from both cricket species, with potential as an insect-based product, rich in proteins, lipids, and vitamins. In the production of high-quality processed foods, it is essential to satisfy consumer preferences, which determine the purchase of products. However, although there are many previous studies on quality evaluation for processed foods, there are few studies on quality evaluation methods with consideration of consumer preference characteristics. In this study, we measured the sugar content, β-carotene content, and color of carrot juices produced by different blanching methods as model samples. In addition, we analyzed the preference characteristics by conjoint analysis, and utilized the results of that analysis as measured values of quality to construct a quality evaluation model considering consumer preference characteristics. Purpose: Vitamin A deficiency (VAD) remains a significant contributor to childhood morbidity and mortality in developing countries; therefore, the implementation of sustainable and cost-effective approaches to control VAD is of utmost pertinence. This study aims to investigate the efficacy of red palm olein (RPO)-enriched biscuit supplementation in improving vitamin A, haematological, iron, and inflammatory status among vitamin A-deficient schoolchildren. Methods: We conducted a double-blinded, randomised controlled trial involving 651 rural primary schoolchildren (8–12 years) with VAD in Malaysia. The schoolchildren were randomised to receive either RPO-enriched biscuits (experimental group, n = 334) or palm olein-enriched biscuits (control group, n = 317) for 6-month duration. Results: Significant improvements in retinol and retinol-binding protein 4 levels were observed in both groups after supplementation (P < 0.001). The improvement in retinol levels were similar across groups among subjects with confirmed VAD (P = 0.40). Among those with marginal VAD, greater improvement in retinol levels was recorded in the control group (P < 0.001) but lacked clinical significance. The levels of α- and β-carotenes, haematological parameters (haemoglobin, packed cell volume, mean corpuscular volume and mean corpuscular haemoglobin) and iron enhanced more significantly in the experimental group (P < 0.05). The significant reduction in the prevalence of microcytic anaemia (− 21.8%) and high inflammation (− 8.1%) was only observed in the experimental group. Conclusion: The supplementation of RPO-enriched biscuits enhanced levels of provitamin A carotenes, iron, and erythropoiesis, and exhibited anti-inflammatory effects. Therefore, the incorporation of RPO into National Nutritional Intervention Programs may be a potential measure to improve the health status of vitamin A-deficient children, among various other interventions. Summary: β‐carotene is a natural carotenoid pigment that widely exists in various fruits and vegetables, possessing antioxidant, anti‐inflammatory, and other biological activities. This makes it highly suitable for the development of nutritional functional foods and cosmetics. However, its poor water solubility and physical and chemical instability hinder its actual bioavailability. Therefore, adopting a rational stabilisation strategy to improve the stability and bioavailability of β‐carotene is a feasible approach. This review provides a comprehensive overview of the properties, biological activities, extraction methods, and synthesis methods of β‐carotene.
