Background: Clostridium autoethanogenum is an acetogenic bacterium that autotrophically converts carbon monoxide (CO) and carbon dioxide (CO2) gases into bioproducts and fuels via the Wood–Ljungdahl pathway (WLP). To facilitate overall carbon capture efficiency, the reaction stoichiometry requires supplementation of hydrogen at an increased ratio of H2:CO to maximize CO2 utilization; however, the molecular details and thus the ability to understand the mechanism of this supplementation are largely unknown. Results: In order to elucidate the microbial physiology and fermentation where at least 75% of the carbon in ethanol comes from CO2, we established controlled chemostats that facilitated a novel and high (11:1) H2:CO uptake ratio. We compared and contrasted proteomic and metabolomics profiles to replicate continuous stirred tank reactors (CSTRs) at the same growth rate from a lower (5:1) H2:CO condition where ~ 50% of the carbon in ethanol is derived from CO2. Our hypothesis was that major changes would be observed in the hydrogenases and/or redox-related proteins and the WLP to compensate for the elevated hydrogen feed gas. Our analyses did reveal protein abundance differences between the two conditions largely related to reduction–oxidation (redox) pathways and cofactor biosynthesis, but the changes were more minor than we would have expected. While the Wood–Ljungdahl pathway proteins remained consistent across the conditions, other post-translational regulatory processes, such as lysine-acetylation, were observed and appeared to be more important for fine-tuning this carbon metabolism pathway. Metabolomic analyses showed that the increase in H2:CO ratio drives the organism to higher carbon dioxide utilization resulting in lower carbon storages and accumulated fatty acid metabolite levels. Conclusions: This research delves into the intricate dynamics of carbon fixation in C. autoethanogenum, examining the influence of highly elevated H2:CO ratios on metabolic processes and product outcomes. The study underscores the significance of optimizing gas feed composition for enhanced industrial efficiency, shedding light on potential mechanisms, such as post-translational modifications (PTMs), to fine-tune enzymatic activities and improve desired product yields. Carbonization is a promising method of carbon fixation, but there has been little research on municipal solid waste (MSW) carbonization as a carbon capture, use, and storage technology. We focused on biomass- and fossil-derived carbon in samples produced at the Saikai City Carbonization Center and evaluated carbon fixation through the MSW carbonization process. We found that the total C content of carbonized fuels is approximately 50%, with fossil C accounting for 9.1–14.4%. Fossil carbon in MSW was considered to be derived mainly from polyethylene and polyamide (nylon 6–6). In terms of the carbon mass balance, negative emissions of 71,300–108,000 kg-CO2/month can be achieved from carbonized fuels that are not burned, such as soil amendments, compared to the case wherein all carbon in MSW is released into the atmosphere. The fossil C content of carbonized fuels is affected by the composition of the input MSW, while the influences of waste quality, desalination time, and pyrolysis temperature changes of several tens of degrees Celsius are small. Landfill disposal of carbonized fuels is limited by the capacity of the final disposal sites. The consumption of this capacity can be reduced through the use of carbonized fuels as a soil amendment. Halophyte shrubs, prevalent in arid regions globally, create saline fertile islands under their canopy. This study investigates the soil microbial communities and their energy utilization strategies associated with tamarisk shrubs in arid ecosystems. Shotgun sequencing revealed that high salinity in tamarisk islands reduces functional gene alpha-diversity and relative abundance compared to bare soils. However, organic matter accumulation within islands fosters key halophilic archaea taxa such as Halalkalicoccus, Halogeometricum, and Natronorubrum, linked to processes like organic carbon oxidation, nitrous oxide reduction, and sulfur oxidation, potentially strengthening the coupling of nutrient cycles. In contrast, bare soils harbor salt-tolerant microbes with genes for autotrophic energy acquisition, including carbon fixation, H2 or CH4 consumption, and anammox. Additionally, isotope analysis shows higher microbial carbon use efficiency, N mineralization, and denitrification activity in tamarisk islands. Light and nitrogen availability are basic requirements for photosynthesis. Changing in light intensity and nitrogen concentration may require adaptive physiological and life process changes in phytoplankton cells. Our previous study demonstrated that two Thalassiosira species exhibited, respectively, distinctive physiological responses to light and nitrogen stresses. Transcriptomic analyses were employed to investigate the mechanisms behind the different physiological responses observed in two diatom species of the genus Thalassiosira. The results indicate that the congeneric species are different in their cellular responses to the same shifting light and nitrogen conditions. When conditions changed to high light with low nitrate (HLLN), the large-celled T. punctigera was photodamaged. Thus, the photosynthesis pathway and carbon fixation related genes were significantly down-regulated. In contrast, the small-celled T. pseudonana sacrificed cellular processes, especially amino acid metabolisms, to overcome the photodamage. When changing to high light with high nitrate (HLHN) conditions, the additional nitrogen appeared to compensate for the photodamage in the large-celled T. punctigera, with the tricarboxylic acid cycle (TCA cycle) and carbon fixation significantly boosted. Consequently, the growth rate of T. punctigera increased, which suggest that the larger-celled species is adapted for forming post-storm algal blooms. Polypropylene carbonate (PPC) is a biodegradable polyester with excellent toughness and ductility that may be employed as carbon fixation materials in biomedicine, agriculture, and environmental engineering. However, relatively lower strength and poor dimensional stability hinder its extensive application. In this study, nanofibrillated cellulose (NFC) and chitosan (CS) were introduced as biobased reinforcements into PPC without compromising its biodegradability. Notably, the composites not only had improved strength but also enhanced toughness after the addition of NFC and CS. The tensile strength and Young's modulus were increased by 25% and 58% in the composites with 3 wt% NFC and 2 wt% CS additions compared to pure PPC. Interestingly, the PPC/NFC/CS composites exhibited superior dimensional stability and antibacterial property. Upon the addition of 3 wt% NFC and 2 wt% CS, the composites took 3 s recovering from 0° to 137°. Additionally, NFC and CS contribute to the hydrophilicity of PPC composites. These findings offer valuable insights into experimental data and preparation techniques to the utilization of PPC in engineering fields. Coccolithophores fix organic carbon and produce calcite plates (coccoliths) that ballast organic matter and facilitate carbon export. Photosynthesis consumes carbon dioxide, while calcification produces it, raising questions about whether coccolithophores are a net sink or source of carbon. We characterized the physiology of calcified and noncalcified (“naked”) phenotypes of Emiliania huxleyi (CCMP374) and investigated the relationship between calcification and photosynthesis across a gradient of light (25–2000 μmol photons m−2 s−1) spanning the euphotic zone. Growth and photophysiological parameters increased with light until reaching a mid‐light (150 μmol photons m−2 s−1) maximum for both phenotypes. Calcified cells were characterized by enhanced photophysiology and less photoinhibition. Further, enhanced bicarbonate transport in calcified cells led to higher rates of particulate organic carbon fixation and growth compared to naked cells at mid‐light to high light (150–2000 μmol photons m−2 s−1). Coccolith production was similarly high at mid and high light, but the rate of coccolith shedding was >3‐fold lower at high‐light (1.2 vs. 0.35 coccoliths cell−1 h−1). The cellular mechanims(s) of this differential shedding remain unknown and underly light‐related controls on coccosphere maintenance. Our data suggest coccoliths shade cells at high light and that enhanced bicarbonate transport associated with calcification increases internal carbon supplies available for organic carbon fixation. Terpenes serve important functions in enhancing plant thermotolerance. Cinnamomum camphora mainly has eucalyptol (EuL), camphor (CmR), linalool (LnL) and borneol (BeL) chemotypes basing on the uppermost monoterpenes. To reveal the thermotolerance mechanisms of these uppermost monoterpenes (eucalyptol, camphor, linalool, and borneol) in C. camphora, we surveyed the ROS metabolism and photosynthesis in the 4 chemotypes fumigated with the corresponding uppermost monoterpene after fosmidomycin (Fos) inhibiting monoterpene synthesis under high temperature at 38°C (Fos+38°C+monoterpene), and investigated the related gene expression in EuL and CmR. Meanwhile, the thermotolerance differences among the 4 uppermost monoterpenes were analyzed. In contrast to normal temperature (28°C), ROS levels and antioxidant enzyme activities in the 4 chemotypes increased under 38°C, and further increased in the treatment with Fos inhibiting monoterpene synthesis at 38°C (Fos+38°C), which may be caused by the alterations in expression of the genes related with nonenzymatic and enzymatic antioxidant formation according to the analyses in EuL and CmR. Compared with Fos+38°C treatment, Fos+38°C+monoterpene treatments lowered ROS levels and antioxidant enzyme activities for the increased non-enzymatic antioxidant gene expression and decreased enzymatic antioxidant gene expression, respectively. High temperature at 38° C reduced the chlorophyll and carotenoid content as well as photosynthetic abilities, which may result from the declined expression of the genes associated with photosynthetic pigment biosynthesis, light reaction, and carbon fixation. Fos+38°C treatment aggravated the reduction. In contrast to Fos+38°C treatment, Fos+38°C+monoterpene treatments increased photosynthetic pigment content and improved photosynthetic abilities by up-regulating related gene expression. Among the 4 uppermost monoterpenes, camphor showed strong abilities in lowering ROS and maintaining photosynthesis, while eucalyptol showed weak abilities. This was consistent with the recovery effects of the gene expression in the treatments with camphor and eucalyptol fumigation. Hydrothermal vents host a diverse community of microorganisms that utilize chemical gradients from the venting fluid for their metabolisms. The venting fluid can solidify to form chimney structures that these microbes adhere to and colonize. These chimney structures are found throughout many different locations in the world's oceans. In this study, comparative metagenomic analyses of microbial communities on five chimney structures from around the Pacific Ocean were elucidated focusing on the core taxa and genes that are characteristic of each of these hydrothermal vent chimneys. The differences among the taxa and genes found at each chimney due to parameters such as physical characteristics, chemistry, and activity of the vents were highlighted. DNA from the chimneys was sequenced, assembled into contigs, and annotated for gene function. Genes used for carbon, oxygen, sulfur, nitrogen, iron, and arsenic metabolisms were found at varying abundances at each of the chimneys, largely from either Gammaproteobacteria or Campylobacteria. Many taxa shared an overlap of these functional metabolic genes, indicating that functional redundancy is critical for life at these hydrothermal vents. A high relative abundance of oxygen metabolism genes coupled with a low abundance of carbon fixation genes could be used as a unique identifier for inactive chimneys. Genes used for DNA repair, chemotaxis, and transposases were found at high abundances at each of these hydrothermal chimneys allowing for enhanced adaptations to the ever-changing chemical and physical conditions encountered. Cyanobacteria are important primary producers, contributing to 25% of the global carbon fixation through photosynthesis. They serve as model organisms to study the photosynthesis, and are important cell factories for synthetic biology. To enable efficient genetic dissection and metabolic engineering in cyanobacteria, effective and accurate genetic manipulation tools are required. However, genetic manipulation in cyanobacteria by the conventional homologous recombination-based method and the recently developed CRISPR-Cas gene editing system require complicated cloning steps, especially during multi-site editing and single base mutation. This restricts the extensive research on cyanobacteria and reduces its application potential. In this study, a highly efficient and convenient cytosine base editing system was developed which allows rapid and precise C → T point mutation and gene inactivation in the genomes of Synechocystis and Anabaena. This base editing system also enables efficient multiplex editing and can be easily cured after editing by sucrose counter-selection. This work will expand the knowledge base regarding the engineering of cyanobacteria. The findings of this study will encourage the biotechnological applications of cyanobacteria. An efficient cytosine base editor was developed in cyanobacteria, which allows rapid and precise C → T point mutation and gene inactivation. Enhancing crop photosynthesis through genetic engineering technologies offers numerous opportunities to increase plant productivity. Key approaches include optimizing light utilization, increasing cytochrome b6f complex levels, and improving carbon fixation. Modifications to Rubisco and the photosynthetic electron transport chain are central to these strategies. Introducing alternative photorespiratory pathways and enhancing carbonic anhydrase activity can further increase the internal CO2 concentration, thereby improving photosynthetic efficiency. The efficient translocation of photosynthetically produced sugars, which are managed by sucrose transporters, is also critical for plant growth. Additionally, incorporating genes from C4 plants, such as phosphoenolpyruvate carboxylase and NADP-malic enzymes, enhances the CO2 concentration around Rubisco, reducing photorespiration. Targeting microRNAs and transcription factors is vital for increasing photosynthesis and plant productivity, especially under stress conditions. Organic management enhances the formation of distinct and stable soil microbial communities, however, its influence on the temporal recovery of microbiome and multifunctionality of sterilized soil remains poorly understood. We used amplicon sequencing and metagenomic sequencing to investigate the effects of microbial communities in long‐term organic and conventional managed soils on restoring soil microbiome and functionality. We calculated multifunctionality of soils at days 30 and 90 of recolonization using the averaging approach. Results showed that organic management (O) significantly increased alpha diversity, niche width and network complexity of microbial community compared to conventional management (C). The alpha diversity, niche width and network complexity of microbial community in soils with organic soil suspension were significantly increased compared to conventional management at days 30 and 90 of recolonization. Soil multifunctionality of sterilized organic managed soil inoculated with organic soil suspension (OO) was 14.6% to 70.6% higher than that of the rest treatments. Macrogenomic analysis revealed that O significantly enriched functional pathways of ABC transporters, carbon metabolism, biosynthesis of amino acids, two‐component and nitrogen metabolism as well as most of the functional genes for carbon degradation, carbon fixation, nitrogen cycling and phosphorus cycles compared to C. These functional pathways and genes were also significantly enriched in soils with organic soil suspension at day 30 and 90 of recolonization. Furthermore, alpha diversity, niche width, network complexity, functional pathways and functional genes of microbiome correlated positively with soil multifunctionality. Synthesis and applications. We estimated hourly air–water gas transfer velocities (k600) for carbon dioxide in the Rhode River, a mesohaline sub-estuary of the Chesapeake Bay. Gas transfer velocities were calculated from estuary-specific parameterizations developed explicitly for shallow microtidal estuaries in the mid-Atlantic region of the United States, using standardized wind speed measurements. Combining the gas transfer velocity with continuous measurements of pCO2 in the water and in the overlying atmosphere, we determined the direction and magnitude of CO2 flux at hourly intervals across a 3-year record (1 July 2018 to 1 July 2021). Continuous year-round measurements enabled us to document strong seasonal cycling, whereby the Rhode River is primarily autotrophic during cold-water months (December–May) and largely net heterotrophic in warm-water months (June–November). Although there is inter-annual variability in CO2 flux in the Rhode River, the annual mean condition is near carbon neutral. Measurement at high temporal resolution across multiple years revealed that CO2 flux and apparent trophic status can reverse during a single 24 h period. pCO2 and CO2 flux are mediated by temperature effects on biological activity and are inverse to temperature-dependent physical solubility of CO2 in water. Biological/biogeochemical carbon fixation and mineralization are rapid and extensive, so sufficient sampling frequency is crucial to capture unbiased extremes and central tendencies of these estuarine ecosystems. Introduction: Salt stress is a major abiotic stress that affects crop growth and productivity. Choline Chloride (CC) has been shown to enhance salt tolerance in various crops, but the underlying molecularmechanisms in rice remain unclear. Methods: To investigate the regulatory mechanism of CC-mediated salt tolerance in rice, we conducted morpho-physiological, metabolomic, and transcriptomic analyses on two rice varieties (WSY, salt-tolerant, and HHZ, saltsensitive) treated with 500 mg·L-1 CC under 0.3% NaCl stress. Results: Our results showed that foliar application of CC improved morphophysiological parameters such as root traits, seedling height, seedling strength index, seedling fullness, leaf area, photosynthetic parameters, photosynthetic pigments, starch, and fructose content under salt stress, while decreasing soluble sugar, sucrose, and sucrose phosphate synthase levels. Transcriptomic analysis revealed that CC regulation combined with salt treatment induced changes in the expression of genes related to starch and sucrose metabolism, the citric acid cycle, carbon sequestration in photosynthetic organs, carbon metabolism, and photosynthetic antenna proteins in both rice varieties. Metabolomic analysis further supported these findings, indicating that photosynthesis, carbon metabolism, and carbon fixation pathways were crucial in CC-mediated salt tolerance. Discussion: The combined transcriptomic and metabolomic data suggest that CC treatment enhances rice salt tolerance by activating distinct transcriptional cascades and phytohormone signaling, alongwithmultiple antioxidants and uniquemetabolic pathways. These findings provide a basis for further understanding the mechanisms of metabolite synthesis and gene regulation induced by CC in rice in response to salt stress, and may inform strategies for improving crop resilience to salt stress. Sugarcane/soybean intercropping and reduced nitrogen (N) application as an important sustainable agricultural pattern can increase crop primary productivity and improve soil ecological functions, thereby affecting soil organic carbon (SOC) input and turnover. To explore the potential mechanism of sugarcane/soybean intercropping affecting SOC sequestration, a two-factor long-term field experiment was carried out, which included planting pattern (sugarcane monocropping (MS), sugarcane/soybean 1:1 intercropping (SB1), and sugarcane/soybean 1:2 intercropping (SB2)) and nitrogen addition levels (reduced N application (N1: 300 kg·hm−2) and conventional N application (N2: 525 kg·hm−2)). The results showed that the shoot and root C fixation in the sugarcane/soybean intercropping system were significantly higher than those in the sugarcane monocropping system during the whole growth period of sugarcane, and the N application level had no significant effect on the C fixation of plants in the intercropping system. Sugarcane/soybean intercropping also increased the contents of total organic C (TOC), labile organic C fraction [microbial biomass C (MBC) and dissolved organic C (DOC)] in the soil during the growth period of sugarcane, and this effect was more obvious at the N1 level. We further analyzed the relationship between plant C sequestration and SOC fraction content using regression equations and found that both plant shoot and root C sequestration were significantly correlated with TOC, MBC, and DOC content. This suggests that sugarcane/soybean intercropping increases the amount of C input to the soil by improving crop shoot and root C sequestration, which then promotes the content of each SOC fraction. The results of this study indicate that sugarcane/soybean intercropping and reduced N application patterns can synergistically improve plant and soil C fixation, which is of great significance for improving crop yields, increasing soil fertility, and reducing greenhouse gas emissions from agricultural fields. A principal goal in ecology is to identify the determinants of species abundances in nature. Body size has emerged as a fundamental and repeatable predictor of abundance, with smaller organisms occurring in greater numbers than larger ones. A biogeographic component, known as Bergmann's rule, describes the preponderance, across taxonomic groups, of larger-bodied organisms in colder areas. Although undeniably important, the extent to which body size is the key trait underlying these patterns is unclear. We explored these questions in diatoms, unicellular algae of global importance for their roles in carbon fixation and energy flow through marine food webs. Using a phylogenomic dataset from a single lineage with worldwide distribution, we found that body size (cell volume) was strongly correlated with genome size, which varied by 50-fold across species and was driven by differences in the amount of repetitive DNA. However, directional models identified temperature and genome size, not cell size, as having the greatest influence on maximum population growth rate. A global metabarcoding dataset further identified genome size as a strong predictor of species abundance in the ocean, but only in colder regions at high and low latitudes where diatoms with large genomes dominated, a pattern consistent with Bergmann's rule. Although species abundances are shaped by myriad interacting abiotic and biotic factors, genome size alone was a remarkably strong predictor of abundance. Taken together, these results highlight the cascading cellular and ecological consequences of macroevolutionary changes in an emergent trait, genome size, one of the most fundamental and irreducible properties of an organism. Groundwater aquifers are ecological hotspots with diverse microbes essential for biogeochemical cycles. Their ecophysiology has seldom been studied on a basin scale. In particular, our knowledge of chemosynthesis in the deep aquifers where temperatures reach 60 °C, is limited. Here, we investigated the diversity, activity, and metabolic potential of microbial communities from nine wells reaching ancient groundwater beneath Israel's Negev Desert, spanning two significant, deep (up to 1.5 km) aquifers, the Judea Group carbonate and Kurnub Group Nubian sandstone that contain fresh to brackish, hypoxic to anoxic water. We estimated chemosynthetic productivity rates ranging from 0.55 ± 0.06 to 0.82 ± 0.07 µg C L−1 d−1 (mean ± SD), suggesting that aquifer productivity may be underestimated. We showed that 60% of MAGs harbored genes for autotrophic pathways, mainly the Calvin–Benson–Bassham cycle and the Wood–Ljungdahl pathway, indicating a substantial chemosynthetic capacity within these microbial communities. We emphasize the potential metabolic versatility in the deep subsurface, enabling efficient carbon and energy use. This study set a precedent for global aquifer exploration, like the Nubian Sandstone Aquifer System in the Arabian and Western Deserts, and reconsiders their role as carbon sinks. All cyanobacteria and some chemoautotrophic bacteria fix CO2 into sugars using specialized proteinaceous compartments called carboxysomes. Carboxysomes enclose the enzymes Rubisco and carbonic anhydrase inside a layer of shell proteins to increase the CO2 concentration for efficient carbon fixation by Rubisco. In the ⍺-carboxysome lineage, a disordered and highly repetitive protein named CsoS2 is essential for carboxysome formation and function. Without it, the bacteria require high CO2 to grow. How does a protein predicted to be lacking structure serve as the architectural scaffold for such a vital cellular compartment? In this study, we identify key residues present in the repeats of CsoS2, VTG and Y, which are necessary for building functional ⍺-carboxysomes in vivo. One of the seven natural CO2 fixation pathways, the anaerobic Wood-Ljungdahl pathway (WLP) is unique in generating CO as a metabolic intermediate, operating through organometallic intermediates, and in conserving (versus utilizing) net ATP. The key enzyme in the WLP is acetyl-CoA synthase (ACS), which uses an active site [2Ni-4Fe-4S] cluster (A-cluster), a CO tunnel, and an organometallic (Ni-CO, Ni-methyl, and Ni-acetyl) reaction sequence to generate acetyl-CoA. Here, we reveal that an alcove, which interfaces the tunnel and the A-cluster, is essential for CO2 fixation and autotrophic growth by the WLP. In vitro spectroscopy, kinetics, binding, and in vivo growth experiments reveal that a Phe229A substitution at one wall of the alcove decreases CO affinity thirty-fold and abolishes autotrophic growth; however, a F229W substitution enhances CO binding 80-fold. Our results indicate that the structure of the alcove is exquisitely tuned to concentrate CO near the A-cluster; protect ACS from CO loss during catalysis, provide a haven for inhibitory CO, and stabilize the tetrahedral coordination at the Nip site where CO binds. The directing, concentrating, and protective effects of the alcove explain the inability of F209A to grow autotrophically. The alcove also could help explain current controversies over whether ACS binds CO and methyl through a random or ordered mechanism. Our work redefines what we historically refer to as the metallocenter "active site". The alcove is so crucial for enzymatic function that we propose it is part of the active site. Biological nitrogen fixation provides fixed nitrogen for microbes living in the oligotrophic open ocean. UCYN‐A2, the previously known symbiont of Braarudosphaera bigelowii, now believed to be an early‐stage B. bigelowii organelle that exchanges fixed nitrogen for fixed carbon, is globally distributed. Indirect evidence suggested that B. bigelowii might be a mixotrophic (phagotrophic) phototrophic flagellate. The goal of this study was to determine if B. bigelowii can graze on bacteria using several independent approaches. The results showed that B. bigelowii grazed on co‐occurring bacteria at a rate of 5–7 cells/h/B. bigelowii and that the overall grazing rate was significantly higher at nighttime than at daytime. Bacterial abundance changes, assessed with 16S rRNA gene amplicon sequencing analysis, may have indicated preferential grazing by B. bigelowii on specific bacterial genotypes. In addition, Lysotracker™ staining of B. bigelowii suggested digestive activity inside B. bigelowii. Carbon and nitrogen fixation measurements revealed that the carbon demand of B. bigelowii could not be fulfilled by photosynthesis alone, implying supplementation by heterotrophy. These independent lines of evidence together revealed that B. bigelowii engages in phagotrophy, which, beyond serving as a supplementary source of carbon and energy, may also facilitate the indirect assimilation of inorganic nutrients. Little is known of primary production in dark hypersaline ecosystems despite the prevalence of such environments on Earth today and throughout its geologic history. Here, we generated and analyzed metagenome-assembled genomes (MAGs) organized as operational taxonomic units (OTUs) from three depth intervals along a 30-cm sediment core from the north arm of Great Salt Lake, Utah. The sediments and associated porewaters were saturated with NaCl, exhibited redox gradients with depth, and harbored nitrogen-depleted organic carbon. Metabolic predictions of MAGs representing 36 total OTUs recovered from the core indicated that communities transitioned from aerobic and heterotrophic at the surface to anaerobic and autotrophic at depth. Dark CO2 fixation was detected in sediments and the primary mode of autotrophy was predicted to be via the Wood–Ljungdahl pathway. This included novel hydrogenotrophic acetogens affiliated with the bacterial class Candidatus Bipolaricaulia. Minor populations were dependent on the Calvin cycle and the reverse tricarboxylic acid cycle, including in a novel Thermoplasmatota MAG. These results are interpreted to reflect the favorability of and selectability for populations that operate the lowest energy requiring CO2-fixation pathway known, the Wood–Ljungdahl pathway, in anoxic and hypersaline conditions that together impart a higher energy demand on cells. SUMMARY: WHIRLY1 is a chloroplast‐nucleus located DNA/RNA‐binding protein with functions in development and stress tolerance. By overexpression of HvWHIRLY1 in barley, one line with a 10‐fold and two lines with a 50‐fold accumulation of the protein were obtained. In these lines, the relative abundance of the nuclear form exceeded that of the chloroplast form. Growth of the plants was shown to be compromised in a WHIRLY1 abundance‐dependent manner. Over‐accumulation of WHIRLY1 in chloroplasts had neither an evident impact on nucleoid morphology nor on the composition of the photosynthetic apparatus. Nevertheless, oeW1 plants were found to be compromised in the light reactions of photosynthesis as well as in carbon fixation. The reduction in growth and photosynthesis was shown to be accompanied by a decrease in the levels of cytokinins and an increase in the level of jasmonic acid. Gene expression analyses revealed that in nonstress conditions the oeW1 plants had enhanced levels of pathogen response (PR) gene expression indicating activation of constitutive defense. During growth in continuous light of high irradiance PR gene expression increased indicating that under stress conditions oeW1 are capable to further enhance defense. Significance Statement: With regard to its dual localization in chloroplasts and nucleus WHIRLY1 is an excellent communicator in retrograde signaling. Previous studies with barley showed that a knockdown of WHIRLY1 compromises acclimation to environment. Here we show that overexpression of WHIRLY1 in barley, leading to an over‐accumulation of the protein in both chloroplasts and nucleus, improves stress resistance whereas it has a negative impact on photosynthesis and growth. Plant physiologists set about comprehending the genesis of the C4 photosynthetic pathway after its discovery by Hatch and Slack. They discovered that a sophisticated combination of morphological and biochemical adaptations allowed the plant to concentrate CO2 around RuBisCO to achieve maximum efficiency. We categorize the evolutionary events leading to C4 photosynthesis, beginning with anoxygenic photosynthesis and the evolution of RuBisCO to the cooling of Earth by the Great Oxygenation Event that led to the oxygenic photosynthesis. The evolutionary descent of the C4 plants is a phenomenon that occurred around 30 million years ago. Due to industrialization and population growth, improved photosynthetic efficiency and carbon fixation of C4 plants could contest the current global scenario of rising CO2 concentration. Petroleum hydrocarbons (PHCs) can be biodegraded into CO2, and PHC‐contaminated aquifers are always deemed as carbon sources. Fortunately, some carbon fixation microorganisms have been found in PHC‐contaminated sites. However, most of the studies are related to volatile short‐chain PHC, and few studies focus on long‐chain PHC‐contaminated sites. To reveal the carbon fixation microorganisms in these sites, in the study, a long‐chain PHC polluted site in North China was selected. Through hydrochemical and metagenomics analysis, the structure and capacity of carbon fixing microorganisms in the site were revealed. Results showed that there were many kinds of carbon fixed microorganisms that were identified such as Flavobacterium, Pseudomonas. HP/4HB, rTCA, and DC/4HB cycles were dominated carbon fixation pathways. The long‐chain PHC were weakly correlated with carbon fixation microorganisms, but it may stimulate the growth of some carbon fixation microorganisms, such as microorganisms involved in rTCA cycle. Shading or low light conditions are essential cultivation techniques for cigar wrapper tobacco leaves production, yet their impact on protein and metabolic regulatory networks is not well understood. In this study, we integrated proteomic and metabolomic analyses to uncover the potential molecular mechanisms affecting cigar tobacco leaves under shading treatment. Our findings include: (1) Identification of 780 significantly differentially expressed proteins (DEPs) in the cigar wrapper tobacco leaves, comprising 560 upregulated and 220 down-regulated proteins, predominantly located in the chloroplast, cytoplasm, and nucleus, collectively accounting for 50.01%. (2) Discovery of 254 significantly differentially expressed metabolites (DEMs), including 148 up-regulated and 106 down-regulated metabolites. (3) KEGG pathway enrichment analysis revealed that the mevalonate (MVA) pathway within 'Terpenoid backbone biosynthesis' was inhibited, leading to a downregulation of 'Sesquiterpenoid and triterpenoid biosynthesis'. Conversely, the 2-C-methyl-D-erythritol 4-phosphate (MEP) pathway was enhanced, resulting in an up-regulation of 'Monoterpenoid biosynthesis', 'Diterpenoid biosynthesis', and 'Carotenoid biosynthesis', thereby promoting the synthesis of terpenoids such as carotenoids and chlorophylls. Simultaneously, the Calvin cycle in 'Carbon fixation in photosynthetic organisms' was amplified, increasing photosynthetic efficiency. These results suggest that under low light conditions, cigar tobacco optimizes photosynthetic efficiency by reconfiguring its energy metabolism and terpenoid biosynthesis. Subalpine forests are a crucial component of the carbon cycling system in the Qinghai–Tibet Plateau (QTP). However, there are currently significant data gaps in the QTP, and it is essential to enhance continuous monitoring of forest carbon absorption processes in the future. This study investigates 2 years' carbon exchange dynamics of a subalpine forest on the QTP using an eddy covariance method. We first characterized the seasonal carbon dynamics of the subalpine forest, revealing the higher carbon dioxide (CO 2) exchange rates in summer and autumn and lower rates in winter and spring, and found that autumn is the peak period for carbon sequestration in this subalpine forest, with the maximum measured value of CO 2 absorption reaching 10.70 µ mol m -2 s -1. Subsequently, we examined the environmental factors influencing the carbon sequestration function. Principal component analysis (PCA) shows that photosynthetically active radiation (PAR) was the major environmental factor driving the net ecosystem exchange (NEE) of CO 2 , significantly influencing forest carbon absorption, and the increase in relative humidity decreases the rate of carbon fixation. In addition, we explored NEE and its influencing factors at the regional scale and found that air temperature promotes carbon dioxide absorption (negative NEE values), while the average annual precipitation shows a minor effect on NEE. At the annual scale, the subalpine forest functions as a strong carbon sink, with an average NEE of -332 to -351 g C m -2 (from November 2020 to October 2022). Despite the challenges of climate change, forests remain robust carbon sinks with the highest carbon sequestration capacity in the QTP, with an average annual CO 2 absorption rate of 368 g C m -2. The use of industrial solid waste to capture and fix CO2 is a promising technology for CO2 sequestration. A thermogravimetric analyzer and CO2 cement hydration mixing device were used to study the effects of mixing method, curing system, temperature, CO2 concentration and other factors on the carbon fixation amount and performance of the circulating fluidized bed fly ash cement-based material system. The results showed that the carbon fixation and early strength of the cementitious materials could be improved by adding CO2 in the stirring process and making CO2 directly participate in the process reaction. The cementing materials samples prepared with CO2 were cured in a standard curing box for 2 days and a carbon atmosphere for 1 day, the carbon fixation amount of the cementing material was increased by 33% and the compressive strength of the cementing material was also improved. This is because under the combined action of carbon mixing and carbon curing, the prepared binding materials produced more Ca(OH)2 in the early stage, and it reacts with the introduced CO2 to form CaCO3. The strength of the calcium carbonate crystals is higher than the strength of the earlier stage of cement, and at the same time, the samples would solidify more CO2. Considering the carbon fixation amount, sample performance and solid waste utilization rate, the best conditions for the cementing materials are as follows: the content of the circulating fluidized bed fly ash (CFA) was 35%, the concentration of carbon curing was 30%, the curing temperature was 40 ℃, the water-binder ratio was 0.4, and the carbon fixation amount of the cementing material could reach about 20%. The use of CFBFA to solidify and storge CO2 is not only a new way to utilize high value-added fly ash resources, but also beneficial for reducing industrial carbon dioxide emissions. Understanding the soil bacterial communities involved in carbon (C) and nitrogen (N) cycling can inform beneficial tillage and crop rotation practices for sustainability and crop production. This study evaluated soil bacterial diversity, compositional structure, and functions associated with C-N cycling at two soil depths (0–15 cm and 15–30 cm) under long-term tillage (conventional tillage [CT] and no-till [NT]) and crop rotation (monocultures of corn, soybean, and wheat and corn–soybean–wheat rotation) systems. The soil microbial communities were characterized by metabarcoding the 16S rRNA gene V4–V5 regions using Illumina MiSeq. The results showed that long-term NT reduced the soil bacterial diversity at 15–30 cm compared to CT, while no significant differences were found at 0–15 cm. The bacterial communities differed significantly at the two soil depths under NT but not under CT. Notably, over 70% of the tillage-responding KEGG orthologs (KOs) associated with C fixation (primarily in the reductive citric acid cycle) were more abundant under NT than under CT at both depths. The tillage practices significantly affected bacteria involved in biological nitrogen (N2) fixation at the 0–15 cm soil depth, as well as bacteria involved in denitrification at both soil depths. The crop type and rotation regimes had limited effects on bacterial diversity and structure but significantly affected specific C-N-cycling genes. For instance, three KOs associated with the Calvin–Benson cycle for C fixation and four KOs related to various N-cycling processes were more abundant in the soil of wheat than in that of corn or soybean. These findings indicate that the long-term tillage practices had a greater influence than crop rotation on the soil bacterial communities, particularly in the C- and N-cycling processes. Integrated management practices that consider the combined effects of tillage, crop rotation, and crop types on soil bacterial functional groups are essential for sustainable agriculture. A novel Gram-stain-negative, facultatively anaerobic, and mixotrophic bacterium, designated as strain LZ166T, was isolated from the bathypelagic seawater in the western Pacific Ocean. The cells were short rod-shaped, oxidase- and catalase-positive, and motile by means of lateral flagella. The growth of strain LZ166T was observed at 10–45 °C (optimum 34–37 °C), at pH 5–10 (optimum 6–8), and in the presence of 0–5% NaCl (optimum 1–3%). A phylogenetic analysis based on the 16S rRNA gene showed that strain LZ166T shared the highest similarity (98.58%) with Aquibium oceanicum B7T and formed a distinct branch within the Aquibium genus. The genomic characterization, including average nucleotide identity (ANI, 90.73–76.79%), average amino identity (AAI, 88.50–79.03%), and digital DNA–DNA hybridization (dDDH, 36.1–22.2%) values between LZ166T and other species within the Aquibium genus, further substantiated its novelty. The genome of strain LZ166T was 6,119,659 bp in size with a 64.7 mol% DNA G+C content. The predominant fatty acid was summed feature 8 (C18:1ω7c and/or C18:1ω6c). The major polar lipids identified were diphosphatidylglycerol (DPG), phosphatidylethanolamine (PE), glycolipid (GL), and phosphatidylglycerol (PG), with ubiquinone-10 (Q-10) as the predominant respiratory quinone. The genomic annotation indicated the presence of genes for a diverse metabolic profile, including pathways for carbon fixation via the Calvin–Benson–Bassham cycle and inorganic sulfur oxidation. Based on the polyphasic taxonomic results, strain LZ166T represented a novel species of the genus Aquibium, for which the name Aquibium pacificus sp. nov. is proposed, with the type strain LZ166T (=MCCC M28807T = KACC 23148T = KCTC 82889T). The roles of Asgard archaea in eukaryogenesis and marine biogeochemical cycles are well studied, yet their contributions in soil ecosystems remain unknown. Of particular interest are Asgard archaeal contributions to methane cycling in wetland soils. To investigate this, we reconstructed two complete genomes for soil-associated Atabeyarchaeia, a new Asgard lineage, and a complete genome of Freyarchaeia, and predicted their metabolism in situ. Metatranscriptomics reveals expression of genes for [NiFe]-hydrogenases, pyruvate oxidation and carbon fixation via the Wood-Ljungdahl pathway. Also expressed are genes encoding enzymes for amino acid metabolism, anaerobic aldehyde oxidation, hydrogen peroxide detoxification and carbohydrate breakdown to acetate and formate. Overall, soil-associated Asgard archaea are predicted to include non-methanogenic acetogens, highlighting their potential role in carbon cycling in terrestrial environments. The roles of Asgard archaea in soil ecosystems are unclear. SUMMARY Photosynthetic and chemosynthetic extremophiles have evolved adaptations to thrive in challenging environments by finely adjusting their metabolic pathways through evolutionary processes. A prime adaptation target to allow autotrophy in extreme conditions is the enzyme Rubisco, which plays a central role in the conversion of inorganic to organic carbon. Here, we present an extensive compilation of Rubisco kinetic traits from a wide range of species of bacteria, archaea, algae, and plants, sorted by phylogenetic group, Rubisco type, and extremophile type. Our results show that Rubisco kinetics for the few extremophile organisms reported up to date are placed at the margins of the enzyme's natural variability. Form ID Rubisco from thermoacidophile rhodophytes and form IB Rubisco from halophile terrestrial plants exhibit higher specificity and affinity for CO2 than their non‐extremophilic counterparts, as well as higher carboxylation efficiency, whereas form ID Rubisco from psychrophile organisms possess lower affinity for O2. Additionally, form IB Rubisco from thermophile cyanobacteria shows enhanced CO2 specificity when compared to form IB non‐extremophilic cyanobacteria. Overall, these findings highlight the unique characteristics of extremophile Rubisco enzymes and provide useful clues to guide next explorations aimed at finding more efficient Rubiscos. The sustainable development of novel photo/electrocatalytic CO2/N2 fixation for chemical synthesis is predicated on reliable, accurate, and rapid detection methods. The directness, specificity, and simultaneity in detecting multiple molecules of the 1H nuclear magnetic resonance (1H‐NMR) technique have aroused increasing interest in photo/electrocatalysis. Thus far, two data processing means have been developed for the precise 1H‐NMR data processing: peak integral and peak height methods. However, few detailed studies offer a clear steer for choosing data processing methods for the concerned products in photo/electrocatalysis, especially the guidelines under the conditions of baseline distortions induced by water suppression. Herein, the feasible practices are restudied to quantify classical products in photo/electrocatalytic CO2/N2 fixation. On the basis of experimental results, 1H‐NMR data processing methods (peak integral and peak height methods) are found to convey different applicability for the different low‐concentration products, while introducing water suppression in the 1H‐NMR tests. The essential reason can be attributed to the baseline distortions caused by the water suppression, which arouses differentiated deviation of peaks with different peak widths. Picocyanobacteria are widespread and globally significant primary producers. In brackish waters, picocyanobacterial populations are composed of diverse species with both single‐cell and colony‐forming lifestyles. Compared to their marine counterparts, brackish picocyanobacteria are less well characterized and the focus of research has been weighted toward single‐cell picocyanobacteria. Here, we investigate the uptake dynamics of single and colony‐forming picocyanobacteria using incubations with dual carbon‐13 and inorganic (ammonium and nitrate) or organic (urea and amino acids) nitrogen‐15 sources during August and September 2020 in the central Baltic Sea. Phytoplankton community and group‐specific uptake rates were obtained using an elemental analyzer isotope ratio mass spectrometer (EA‐IRMS) and nano secondary‐ion mass spectrometry (NanoSIMS). Picocyanobacteria contributed greater than one third of the ammonium, urea, amino acids, and inorganic carbon community uptake/fixation in September but < 10% in August when phytoplankton biomass was higher. Overall, single‐cell ammonium and urea uptake rates were significantly higher for single‐celled compared to colonial picocyanobacteria. In a 6‐yr offshore central Baltic Sea time series (2015–2020), summer abundances of colonial picocyanobacteria reached up to 105 cells mL−1 and represented > 5% of the average phytoplankton biomass, suggesting that they are periodically important for the ecosystem. Colonial strain identification was not distinguishable using 16S rRNA gene amplicon data, highlighting a need for refined tools for identification of colonial forms. This study shows the significance of single‐celled brackish picocyanobacteria to nutrient cycling and the importance of considering uptake and lifestyle strategies when assessing the role of picocyanobacteria in aquatic ecosystems. Elevated concentrations of CO2 in the atmosphere pose significant challenges, notably contributing to the greenhouse effect. CO2 as an abundant C1 resource converting into high-value oxazolidinones is a promising avenue for effective carbon fixation. Herein, a highly stable binuclear cerium–organic framework, [Ce2(DCTP)2(DMA)2(OAc)2]n (1), has been successfully designed and synthesized, which maintains the intact three-dimensional framework after immersing in solutions with pH ranging from 1 to 12 and various common solvents for 24 hours, respectively. Explorations on CO2 conversion indicate that compound 1 exhibits remarkable catalytic efficiency in the cycloaddition reaction of CO2 with aziridines to form oxazolidinones under relatively low CO2 pressure. 1 can be easily regenerated five times without a significant decrease in yield. Mechanistic experiments show that the high catalytic activity of 1 is attributed to the stable three-dimensional framework, high Lewis acidity, and ample cerium active sites. La Muerte lagoon is an ephemeral endorheic water body located in the Monegros desert, Zaragoza, Spain. Amplicon sequencing of the 16S rRNA gene was performed to analyze the bacterial and archaeal communities in biofilmsediment samples over three years, to understand the dynamic changes in the microbial community. PICRUSt and shotgun metagenomics were used to examine energy production and carbohydrate metabolism pathways. The dominant bacterial phyla were Actinobacteriota, Bacteroidota, Cyanobacteriota, and Pseudomonadota, while Halobacteriota was the predominant archaeal phylum. Despite seasonal environmental fluctuations, the biofilm community remained stable over time, suggesting resilience. The Calvin-Benson cycle was the main carbon fixation pathway, carried out by Cyanobacteria and purple non-sulfur bacteria. Nitrogen fixation by diazotrophs supplied an important nitrogen source. Organic carbon was derived primarily from autotrophs, with little use of allochthonous plant material. The comparison of biofilm-sediment and water column biotopes showed distinct but related prokaryote communities. Biofilm-sediments showed higher taxonomic diversity and different proportions of microbial phyla compared to the water column. This study provides initial insights into the complex microbial life in endorheic lagoons and underscores the importance of protecting these globally threatened habitats. The limited sample size in this study warrants further investigation with a more comprehensive sampling strategy to fully characterize the microbial communities and their functional roles in the different biotopes of La Muerte lagoon. Echinochloa glabrescens Munro ex Hook. f. is a weed of the genus Echinocloa (Echinocloa spp.) that occurs frequently in paddy fields, causing serious harm to rice production. Florpyrauxifen-benzyl (FPB) is a foliar-applied herbicide used to control Echinocloa spp. in paddy fields. However, in recent years, with the widespread use of FPB in rice production, FPB-resistant barnyard grasses have been reported. Here, we identified an FPB-resistant E. glabrescens population with a resistance index (RI) of 10.65 and conducted a comparative analysis using untargeted metabolomics and transcriptomics to investigate the differences between an FPB-resistant E. glabrescens population and a susceptible E. glabrescens population after treatment with the recommended field dose of FPB. Our results showed that the FPB-resistant E. glabrescens had 115 differentially accumulated metabolites (DAMs; 65 up-regulated and 50 downregulated) and 6397 differentially expressed genes (DEGs; 65 up-regulated and 50 down-regulated) compared to the susceptible E. glabrescens. The analysis of DAMs and DEGs revealed that DAMs were significantly enriched in Glutathione metabolism, Arginine and proline metabolism, and Zeatin biosynthesis pathways, while DEGs were mainly enriched in carbon fixation in photosynthetic organisms, photosynthesis, cyanoamino acid metabolism and glutathione metabolism, etc. The glutathione metabolism pathway was found to be significantly enriched for both DEGs and DAMs. Within this pathway, the metabolites (spermine) and genes (GSTU8, GSTU18, GSTF1) may play a pivotal role in the resistance mechanism of FPB-resistant E. glabrescens. Furthermore, we demonstrated the presence of GST-mediated metabolic resistance in an FPB-resistant E. glabrescens population by using NBD-Cl. Overall, our study provides new insights into the underlying mechanisms of E. glabrescens resistance to FPB through a comparative analysis of untargeted metabolomics and transcriptomics. Additionally, we identified the GST-mediated metabolic resistance in an FPBresistant E. glabrescens population, and screened for three candidate genes (GSTU8, GSTU18, GSTF1), which has significant implications for improving the weed management efficacy of FPB in rice production and guiding judicious herbicide usage. Ecofuels and their feedstock come in three main product classes: electrofuels (e-Fuels), biofuels, and non-biowaste-derived fuels. Ecofuels originate from non-fossil sources, derived from circular raw materials such as non-food organic waste, renewable hydrogen, and captured CO2 through a rapid process of carbon fixation. Proposed regulation drafts under discussion indicate that new fuels would need to reach a substantial degree of climate neutrality. The manufacture of all ecofuels, however, requires energy input to accomplish the conversion of the initial feedstock; their climate neutrality claims stem from the use of renewable electric energy and/or biomasses in the production process, but fossil fuels are still the main primary sources of global (and the EU's) electric power, and most biofuels consumed in the EU transport mix are still crop-based, with potential conflicts with food and land use. Furthermore, entirely neglecting GHG emissions from renewable energy generation is scientifically debatable, as the impact of the energy (and the related GHG emissions) embedded in the materials used to build renewable energy facilities is small, but not nil. Using carbon-based materials (CBMs) to facilitate phytoremediation shows great promise for simultaneously enhancing the restoration efficiency of contaminated soil and reducing carbon dioxide in the context of global warming, which is still in the exploring and attempting phase. In addition to direct degradation or alteration of pollutant bioavailability, CBMs can enhance phytoremediation by alleviating plant nutrient deprivation and oxidative stress, as well as by modulating soil microbial communities and root secretions. Photosynthetic carbon fixation predominantly affects both phytoremediation efficiency and carbon cycle turnover in terrestrial ecosystems. In this regard, CBMs have extremely positive properties in facilitating plant carbon capture, with intrinsic mechanisms including (1) promoting photosynthetic pigment synthesis and acting as artificial built-in antennae to improve photon capture efficiency, (2) accelerating the photosynthetic electron transport rate in photosystems, (3) improving the Calvin cycle, and (4) maintaining the structural integrity of chloroplasts. Besides, as an ultra-stable type of CBM derived from waste biomass, biochar can preserve the native biomass carbon in the soil environment for decades and attenuate the rhizosphere priming effect by influencing the structure of rhizosphere soil aggregates and microbial communities, thus retarding the native soil organic carbon efflux. This review critically elaborates on the mechanisms by which CBMs assist in improving the efficiency of phytoremediation and their positive effects on the plant-root soil carbon balance. Additionally, a simple full-life cycle analysis encompassing cost analysis as well as ecological, economic, and social benefits is concluded to evaluate the feasibility and sustainability of CBMs-phytoremediation systems. CO2 chemical fixation offers a feasible approach for carbon mitigation and high‐value utilization, but it is still challenging to develop an efficient catalyst for CO2 conversion into cyclic carbonate. Herein, the triethylenediamine‐derived dicationic ionic liquids (DIL‐X, X = Cl, Br, and I) were grafted into UiO‐66 linkers through self‐assembly of Zr4+ ions and the mixed ligands of terephthalic acid and DIL‐bearing dicarboxylic acid, resulting in UiO‐66‐DIL‐Xn, (n designated as molar amount of the feeding DIL‐X). Their catalytic performance was evaluated by the epoxide cycloaddition reaction in the absence of solvent and cocatalyst. Among them, the UiO‐66‐DIL‐Cl0.4 catalyst exhibited outstanding performance, with a chloropropene carbonate yield of 92% and a high selectivity of 99% under 0.1 MPa CO2 at 110 °C for 16 h. Its high activity could be ascribed to the cooperativity among Lewis acidity of MOF nodes, the enhanced CO2 absorption, and the strong nucleophilicity offered by halogen ions of ionic liquid‐modified MOF. Moreover, UiO‐66‐DIL‐Cl0.4 presented excellent recyclability and substrate extension. A potential catalytic mechanism for the epoxide‐CO2 cycloaddition into cyclic carbonate has been proposed. Autotrophy is the basis for complex life on Earth. Central to this process is rubisco—the enzyme that catalyzes almost all carbon fixation on the planet. Yet, with only a small fraction of rubisco diversity kinetically characterized so far, the underlying biological factors driving the evolution of fast rubiscos in nature remain unclear. We conducted a high-throughput kinetic characterization of over 100 bacterial form I rubiscos, the most ubiquitous group of rubisco sequences in nature, to uncover the determinants of rubisco's carboxylation velocity. We show that the presence of a carboxysome CO2 concentrating mechanism correlates with faster rubiscos with a median fivefold higher rate. In contrast to prior studies, we find that rubiscos originating from α-cyanobacteria exhibit the highest carboxylation rates among form I enzymes (≈10 s−1 median versus <7 s−1 in other groups). Our study systematically reveals biological and environmental properties associated with kinetic variation across rubiscos from nature. Synopsis: Form I rubisco, known for its slow kinetics in plants and algae, exhibits a great unexplored diversity in autotrophic bacteria. This article represents the first large-scale survey of bacterial form I rubisco kinetics and reveals unifying features of fast carboxylating rubiscos. Over 100 homologs were systematically screened, spanning the wide genetic diversity of form I rubisco enzymes across a variety of ecological niches and metabolic profiles. Background: Biotransformation of CO2 into high-value-added carbon-based products is a promising process for reducing greenhouse gas emissions. To realize the green transformation of CO2, we use fatty acids as carbon source to drive CO2 fixation to produce succinate through a portion of the 3-hydroxypropionate (3HP) cycle in Cupriavidus necator H16. Results: This work can achieve the production of a single succinate molecule from one acetyl-CoA molecule and two CO2 molecules. It was verified using an isotope labeling experiment utilizing NaH13CO3. This implies that 50% of the carbon atoms present in succinate are derived from CO2, resulting in a twofold increase in efficiency compared to prior methods of succinate biosynthesis that relied on the carboxylation of phosphoenolpyruvate or pyruvate. Meanwhile, using fatty acid as a carbon source has a higher theoretical yield than other feedstocks and also avoids carbon loss during acetyl-CoA and succinate production. To further optimize succinate production, different approaches including the optimization of ATP and NADPH supply, optimization of metabolic burden, and optimization of carbon sources were used. The resulting strain was capable of producing succinate to a level of 3.6 g/L, an increase of 159% from the starting strain. Conclusions: This investigation established a new method for the production of succinate by the implementation of two CO2 fixation reactions and demonstrated the feasibility of ATP, NADPH, and metabolic burden regulation strategies in biological carbon fixation. The behavior of many plant enzymes depends on the metals and other ligands to which they are bound. A previous study demonstrated that tobacco Rubisco binds almost equally to magnesium and manganese and rapidly exchanges one metal for the other. The present study characterizes the kinetics of Rubisco and the plastidial malic enzyme when bound to either metal. When Rubisco purified from five C3 species was bound to magnesium rather than manganese, the specificity for CO2 over O2, (Sc/o) increased by 25% and the ratio of the maximum velocities of carboxylation / oxygenation (Vcmax/Vomax) increased by 39%. For the recombinant plastidial malic enzyme, the forward reaction (malate decarboxylation) was 30% slower and the reverse reaction (pyruvate carboxylation) was three times faster when bound to manganese rather than magnesium. Adding 6‐phosphoglycerate and NADP+ inhibited carboxylation and oxygenation when Rubisco was bound to magnesium and stimulated oxygenation when it was bound to manganese. Conditions that favored RuBP oxygenation stimulated Rubisco to convert as much as 15% of the total RuBP consumed into pyruvate. These results are consistent with a stromal biochemical pathway in which (1) Rubisco when associated with manganese converts a substantial amount of RuBP into pyruvate, (2) malic enzyme when associated with manganese carboxylates a substantial portion of this pyruvate into malate, and (3) chloroplasts export additional malate into the cytoplasm where it generates NADH for assimilating nitrate into amino acids. Background: Advancing the engineering of photosynthesis-based prokaryotic cell factories is important for sustainable chemical production and requires a deep understanding of the interplay between bioenergetic and metabolic pathways. Rearrangements in photosynthetic electron flow to increase the efficient use of the light energy for carbon fixation must be balanced with a strong carbon sink to avoid photoinhibition. In the cyanobacterium Synechocystis sp. PCC 6803, the flavodiiron protein Flv3 functions as an alternative electron acceptor of photosystem I and represents an interesting engineering target for reorganizing electron flow in attempts to enhance photosynthetic CO2 fixation and increase production yield. Results: We have shown that inactivation of Flv3 in engineered sucrose-excreting Synechocystis (S02:Δflv3) induces a transition from photoautotrophic sucrose production to mixotrophic growth sustained by sucrose re-uptake and the formation of intracellular carbon sinks such as glycogen and polyhydroxybutyrate. The growth of S02:Δflv3 exceeds that of the sucrose-producing strain (S02) and demonstrates unforeseen proteomic and metabolomic changes over the course of the nine-day cultivation. In the absence of Flv3, a down-regulation of proteins related to photosynthetic light reactions and CO2 assimilation occurred concomitantly with up-regulation of those related to glycolytic pathways, before any differences in sucrose production between S02 and S02:Δflv3 strains were observed. Over time, increased sucrose degradation in S02:Δflv3 led to the upregulation of respiratory pathway components, such as the plastoquinone reductase complexes NDH-11 and NDH-2 and the terminal respiratory oxidases Cyd and Cox, which transfer electrons to O2. While glycolytic metabolism is significantly up-regulated in S02:Δflv3 to provide energy for the cell, the accumulation of intracellular storage compounds and the increase in respiration serve as indirect sinks for photosynthetic electrons. Conclusions: Our results show that the presence of strong carbon sink in the engineered sucrose-producing Synechocystis S02 strain, operating under high light, high CO2 and salt stress, cannot compensate for the lack of Flv3 by directly balancing the light transducing source and carbon fixing sink reactions. Instead, the cells immediately sense the imbalance, leading to extensive reprogramming of cellular bioenergetic, metabolic and ion transport pathways that favor mixotrophic growth rather than enhancing photoautotrophic sucrose production. The key role of soil quality improvement in achieving sustainable agricultural development based on highly intensive use of farmland is increasingly being recognized, as is the ponderance of suitable evaluation of the soil quality. The overarching goal of this study was to determine an accurate assessment framework by the comparison of the scoring function (linear and non-linear) and integration method (area and weighted additive), which integrally evaluates the soil quality of an eleven-year field fertilization experiment (including CK, no fertilizer; CF, conventional fertilization; SF, formulated fertilization; SFO, SF with organic fertilizer). Thirty-three properties, including eighteen physiochemical-related and fifteen biological-related properties, associated with soil functions were measured as potential soil quality indicators, and the soil multifunctionality (SMF) was applied to validate the soil quality indices (SQIs). Principal component analysis and relationship analysis were used with indicators sensitive to management to determine a minimum data set (MDS). The results showed that the electrical conductivity, large macroaggregate-associated total nitrogen, small macroaggregate-associated organic carbon, carbon fixation, and enzyme activities of phenol oxidase and cellulase were chosen as the MDS. All the SQIs were significantly correlated with the SMF (p < 0.05). The fertilization strategies affected most indicators in different ways, and the index developed using the non-linear function and weighted additive integration method (SQI-NL) had the best sensibility and discriminability. The SQI value with the SQI-NL-MDS method was higher following the fertilization treatments than that of no fertilizer (p < 0.05), and the treatment of the organic fertilizer had the highest SQI value (0.66). Soil quality evaluation in long-term fertilized farmland suggested that the soil quality constraints between treatments of synthetic and organic fertilizer are related to the soil functions of nutrient cycling and sustain biological activity due to their higher contribution rates to the SQI in the organic fertilizer treatment, which provides insights into ways to reduce the gap in soil quality. The framework method can provide an accurate quantitative tool for the evaluation of soil quality from the target indicators by bridging management objectives and field-level actions. Summary: The production of alkaliphilic microalgae can contribute to address the challenging cost of using pure carbon dioxide in large reactors. At high pH values, carbon dioxide is rapidly scavenged and the supply rates of dissolved inorganic carbon from the atmosphere to alkaline media are high. The present study aimed to identify microalgal strains that can cope with high alkalinity and aeration rates. Eight strains were studied and the microalgae Chlorella vulgaris, Scenedesmus almeriensis and Nannochloropsis gaditana were the only ones that were able to grow under these conditions. Their biomass productivities using laboratory‐scale bubble columns with no pH control and high aeration flow were 0.20 ± 0.03, 0.24 ± 0.03 and 0.08 ± 0.01 g·L−1·day−1, respectively. The production of the two former was scaled up to pilot‐scale bubble columns. Overall, the amount of atmospheric carbon dioxide that was transformed into biomass was in the range of 10%–30%, depending on the strain used and the photobioreactor setup. The biomass was rich in proteins and β‐carotene, both valuable products, highlighting the potential production of food ingredients while capturing carbon dioxide from the atmosphere. The presence of antibiotics in irrigation water is an emerging problem. This study aimed to evaluate the effect of irrigation using water-containing ciprofloxacin on alfalfa (Medicago sativa) nitrogen and carbon fixation, number of root nodules, root and stem length, and root and stem weight. A surface layer of soil from Piracicaba, Brazil, was used in controlled conditions. The soil pH was adjusted to neutral levels, and alfalfa (M. sativa) was grown. Seeds inoculated with Rhizobium sp. were distributed in 12 pots comprising three treatments employing three repetitions and three controls. The experiment was conducted in a plant growth chamber, where six irrigations were carried out with contaminated water with ciprofloxacin at 0, 1, 10, and 100 μg L−1 for 40 days. The presence of ciprofloxacin in irrigation water decreased carbon fixation in alfalfa leaves by up to 8.9%, reducing the number of root nodules by 96% at 100 μg L−1 and leading to their elongation. However, it had no significant effect on soil nitrogen and carbon, similarly in stem length and, finally, in root and stem biomass. The decline in global plant diversity has raised concerns about its implications for carbon fixation and global greenhouse gas emissions (GGE), including carbon dioxide (CO2), nitrous oxide (N2O) and methane (CH4). Therefore, we conducted a comprehensive meta‐analysis of 2103 paired observations, examining GGE, soil organic carbon (SOC) and plant carbon in plant mixtures and monocultures. Our findings indicate that plant mixtures decrease soil N2O emissions by 21.4% compared to monocultures. No significant differences occurred between mixtures and monocultures for soil CO2 emissions, CH4 emissions or CH4 uptake. Plant mixtures exhibit higher SOC and plant carbon storage than monocultures. After 10 years of vegetation development, a 40% reduction in species richness decreases SOC content and plant carbon storage by 12.3% and 58.7% respectively. These findings offer insights into the intricate connections between plant diversity, soil and plant carbon storage and GGE—a critical but previously unexamined aspect of biodiversity–ecosystem functioning. Chemosynthesis fixes carbon using energy acquired from reduced chemical compounds and is a primordial process that occurs ubiquitously throughout contemporary marine ecosystems. Diverse and abundant marine bacteria and archaea gain energy from inorganic compounds such as ammonia, nitrite, sulfide, hydrogen, carbon monoxide, and iron using specific enzymes. Chemosynthesis is enriched relative to photosynthesis in aphotic waters, including at increased ocean depths, and may show diel, seasonal, and latitudinal variations. Some regions of the ocean are primarily driven by chemosynthesis, ranging from highly productive geologically fuelled hydrothermal vents to oligotrophic below-shelf waters. Dark carbon fixation through chemosynthetic processes may play a quantitatively significant role in oceanic primary production, though carbon budgets remain poorly resolved. Chemosynthesis is a metabolic process that transfers carbon to the biosphere using reduced compounds. It is well recognised that chemosynthesis occurs in much of the ocean, but it is often thought to be a negligible process compared to photosynthesis. Here we propose that chemosynthesis is the underlying process governing primary production in much of the ocean and suggest that it extends to a much wider range of compounds, microorganisms, and ecosystems than previously thought. In turn, this process has had a central role in controlling marine biogeochemistry, ecology, and carbon budgets across the vast realms of the ocean, from the dawn of life to contemporary times. Decreasing antenna size is considered a potential option for improving photosynthesis and increasing yield potential. Reducing chlorophyll content has been employed as a strategy to decrease antenna size. One of the commonly mentioned advantages of this approach is its ability to enhance crop nitrogen use efficiency (NUE); however, there is limited field evidence supporting this claim. In this study, we utilized a rice mutant called p35s‐Ami‐YGL1, which exhibits lower chlorophyll content and smaller antenna size, to investigate the effects of modifying leaf chlorophyll content on tissue nitrogen content and NUE. Our results demonstrate that the nitrogen contents in various tissues, including seed tissue, increased on a weight basis in p35s‐Ami‐YGL1 mutants while exhibiting a decrease in C:N ratio. Simultaneously, we observed a reduction in tissue carbon content along with an increase in the levels of chlorophyll precursors such as Proto IX. Specifically, we observed an upregulation in the expression of genes associated with photosynthetic light reactions and chlorophyll metabolism, while there was no increase in the expression of genes involved in the CBB cycle and nitrogen metabolism. In addition, p35s‐Ami‐YGL1 experienced increased photodamage. These findings suggest that the alterations in the C:N ratio and nitrogen content in plants may be attributed to Proto IX‐mediated photodamage and chloroplast reverse transduction signaling. Besides, these results suggest that the observed increase in tissue nitrogen content in p35s‐Ami‐YGL1 does not reflect an increase in plant nitrogen absorption or use efficiency, rather it is a result of stunted carbon fixation capacity. High‐latitude oceans experience strong seasonality where low light limits photosynthetic activity most of the year. This limitation is pronounced for algae within and underlying sea ice, and these algae are uniquely acclimated to low light levels. During spring melt, however, light intensity and daylength increase drastically, triggering blooms of ice algae that play important roles in carbon cycling and ecosystem productivity. How the algae acclimate to this dynamic and heterogeneous environment is poorly understood. Here, we measured 14C‐carbon fixation rates, photophysiology, and ribulose 1,5‐bisphosphate carboxylase oxygenase (Rubisco) content of sea‐ice algae in coastal waters near the western Antarctic Peninsula during spring, ranging from a low‐light‐acclimated, bottom community to a light‐saturated bloom. Carbon fixation rates by sea‐ice algae were similar to other Antarctic sea‐ice measurements (2–49 mg C m−2 d−1), and there was little phytoplankton biomass in the underlying water at the time of sampling. Net‐to‐gross ratios of carbon fixation were generally high and showed no relationship with ice type. We found algal photophysiology and Rubisco concentrations varied in relation to the different types of ice, altering the balance between the photochemical and biochemical processes that constrain carbon fixation rates. For algae inhabiting the bottom layers of sea ice, rates of carbon fixation were largely constrained by light availability whereas in surface seawater, interior and rotten/brash ice, carbon fixation rates could be calculated with reasonable accuracy from measurements of Rubisco concentrations.
