Central conducting lymphatic anomaly (CCLA) due to congenital maldevelopment of the lymphatics can result in debilitating and life-threatening disease with limited treatment options. We identified 4 individuals with CCLA, lymphedema, and microcystic lymphatic malformation due to pathogenic, mosaic variants in KRAS. To determine the functional impact of these variants and identify a targeted therapy for these individuals, we used primary human dermal lymphatic endothelial cells (HDLECs) and zebrafish larvae to model the lymphatic dysplasia. Expression of the p.Gly12Asp and p.Gly13Asp variants in HDLECs in a 2‑dimensional (2D) model and 3D organoid model led to increased ERK phosphorylation, demonstrating these variants activate the RAS/MAPK pathway. Expression of activating KRAS variants in the venous and lymphatic endothelium in zebrafish resulted in lymphatic dysplasia and edema similar to the individuals in the study. Treatment with MEK inhibition significantly reduced the phenotypes in both the organoid and the zebrafish model systems. In conclusion, we present the molecular characterization of the observed lymphatic anomalies due to pathogenic, somatic, activating KRAS variants in humans. Our preclinical studies suggest that MEK inhibition should be studied in future clinical trials for CCLA due to activating KRAS pathogenic variants.

Influenza is a costly disease for the population. It is a cause of seasonal morbidity and mortality, epidemics and pandemics or syndemics. Given the variability of the virus, surveillance systems are implemented in order to update the strains and include them in the annual influenza vaccine. This vaccine is currently recommended in some high-risk groups. However, universal vaccination remains controversial. To evaluate the evidence and describe the position of a panel of experts on the relevance of universal vaccination against influenza virus.Five clinical questions were asked, whereby a systematic search of the literature in electronic sources and a Delphi panel were carried out. The evidence was analyzed, and recommendations were issued by the experts.The group of experts recommends vaccinating the population starting at six months of age and include people who live with egg protein allergy, with comorbidities (diabetes, obesity, cancer), health workers and pregnant women.Vaccination, starting with vulnerable groups, is a necessary, ethical and cost-effective strategy. However, expanding the coverage to achieve universal vaccination could reduce the transmission of the disease and its consequences in the population.La influenza es una enfermedad costosa para la población. Es causa de morbimortalidad estacional, epidemias y pandemias o sindemias. Debido a la variabilidad del virus, se implementan sistemas de vigilancia para actualizar las cepas e incluirlas en la vacuna antiinfluenza anual. Actualmente se recomienda esta vacuna en algunos grupos de alto riesgo. Sin embargo, la vacunación universal es aún controvertida. Evaluar la evidencia y describir la posición de un panel de expertos sobre la pertinencia de la vacunación universal contra el virus de influenza.Se realizaron cinco preguntas clínicas, con las que se realizó una búsqueda sistemática de la literatura en fuentes electrónicas y un panel Delphi. Se analizó la evidencia y se emitieron recomendaciones por los expertos.El grupo de expertos recomienda vacunar a la población desde los seis meses de edad e incluir a personas que viven con alergia a la proteína del huevo, con comorbilidades (diabetes, obesidad, cáncer), trabajadores de la salud y embarazadas.La vacunación, iniciando con los grupos vulnerables, es una estrategia necesaria, ética y costo-efectiva. Sin embargo, extender la cobertura para lograr la vacunación universal podría disminuir la transmisión de la enfermedad y sus consecuencias en la población.

Objectives. This study aimed to develop and validate a simple questionnaire for chronic neuropathic pain that can be administered as a screening tool by general practitioners and internists in order to help them identify patients with probable neuropathic pain. Methods. Following a development phase and a pilot study, the revised version of the screening tool which included eleven descriptors associated with neuropathic pain both in English and Filipino languages was validated on 120 consecutive patients with any type of pain except psychogenic pain, recruited in the out-patient clinics of six hospitals. The questionnaire was validated by assessing the sensitivity, specificity, positive and negative predictive value of each item and the overall questionnaire. The internal consistency of the questionnaire items was assessed using the Kuder-Richardson formula 20. Results. Overall, the internal consistency of the SigN-PQ using the Kuder-Richardson formula 20 was 0.7837; the sensitivity was 91.89% with specificity of 80.22%, PV (+) was 65.38% and PV(-) was 96.0%. For the English version, the descriptors with the highest scores were burning (Sensitivity: 100%, Specificity: 93%) and electricity-like (Sensitivity: 100%, Specificity: 93%). For the Filipino version, mainit (burning) has the highest sensitivity of 88% with specificity of 82.6%, followed by gumagapang (tingling) with sensitivity of 86.96% and specificity of 85.42%. The sensation of saksak (stabbing) and hiwa (lancinating) have the lowest sensitivity, 60% and 54% respectively, although their specificity scores are high. Since this study is a validation of a screening tool for neuropathic pain, the investigators decided to choose descriptors with higher sensitivity. Thus, in the final version of the SignN-PQ, the descriptors saksak and hiwa were removed. Conclusion. The SigN-PQ Neuropathic Pain Questionnaire has a high overall sensitivity of 91.89% and specificity of 80.22%. The pain descriptors in the questionnaire are consistent with the descriptors cited in the literature. It is a valid screening instrument for neuropathic pain that can be easily incorporated in the daily practice of general practitioners and internists.

Retrotransposons and retroviruses shape genome evolution and can negatively impact genome function.Saccharomyces cerevisiaeand its close relatives harbor several families of LTR-retrotransposons, the most abundant being Ty1 in several laboratory strains. The cytosolic foci that nucleate Ty1 virus-like particle (VLP) assembly are not well-understood. These foci, termed retrosomes or T-bodies, contain Ty1 Gag and likely Gag-Pol and the Ty1 mRNA destined for reverse transcription. Here, we report a novel intrinsically disordered N-terminalprion-likedomain (PrLD) within Gag that is required for transposition. This domain contains amino-acid composition similar to known yeast prions and is sufficient to nucleate prionogenesis in an established cell-based prion reporter system. Deleting the Ty1 PrLD results in dramatic VLP assembly and retrotransposition defects but does not affect Gag protein level. Ty1 Gag chimeras in which the PrLD is replaced with other sequences, including yeast and mammalian prionogenic domains, display a range of retrotransposition phenotypes from wildtype to null. We examine these chimeras throughout the Ty1 replication cycle and find that some support retrosome formation, VLP assembly, and retrotransposition, including the yeast Sup35 prion and the mouse PrP prion. Our interchangeable Ty1 system provides a useful, genetically tractablein vivoplatform for studying PrLDs, complete with a suite of robust and sensitive assays, and host modulators developed to study Ty1 retromobility. Our work invites study into the prevalence of PrLDs in additional mobile elements.SignificanceRetrovirus-like retrotransposons help shape the genome evolution of their hosts and replicate within cytoplasmic particles. How their building blocks associate and assemble within the cell is poorly understood. Here, we report a novelprion-likedomain (PrLD) in the budding yeast retrotransposon Ty1 Gag protein that builds virus-like particles. The PrLD has similar sequence properties to prions and disordered protein domains that can drive the formation of assemblies that range from liquid to solid. We demonstrate that the Ty1 PrLD can function as a prion and that certain prion sequences can replace the PrLD and support Ty1 transposition. This interchangeable system is an effective platform to study additional disordered sequences in living cells.

The Dominican Republic has more than 1.200.000 Micro, Small and Medium Enterprises (MSMEs) in different Economic Sectors. Although the State recognizes the value that these have for the economic development of the country, there is no systematized and automated characterization that allows for an analysis that helps decisionmaking. This article shows the results when applying Business Intelligence Techniques for the characterization of Enterprises, taking into account the Systematization of Information through a web application. In this sense, 179 Micro, Small and Mediumsize d Enterprises were analyzed, using an instrument that allowed knowing different variables associated with the economic development of each of the Enterprises. Results show the incidence of different factors in the generation of income for the Enterprise.

Neutron-induced cross sections represent the main nuclear input to models of stellar and Big-Bang nucleosynthesis. While (n,γ) reactions are relevant for the formation of elements heavier than iron, (n,p) and (n,α) reactions can play an important role in specific cases. The time-of-flight method is routinely used at n_TOF to experimentally determine the cross section data. In addition, recent upgrades of the facility will allow the use of activation techniques as well, possibly opening the way to a systematic study of neutron interaction with radioactive isotopes. In the last 20 years n_TOF has provided a large amount of experimental data for Nuclear Astrophysics. Our plan is to carry on challenging measurements and produce nuclear data in the next decades as well.

Extracellular inulinase and invertase production by Aspergillus brasiliensis was evaluated in solid-state fermentation (SSF) and submerged fermentation (SmF) with a high sucrose concentration. The parameters estimated from online CO2 analysis revealed a lower Lag phase in SSF compared to SmF; the specific CO2 production rate (μCO2) decreased in both systems as the sucrose concentration increased. The highest maximum CO2 production rate (10.45 ± 0.72 mg/h mL) and total CO2 production (138.53 ± 6.48 mg/mL) were obtained in SSF with a sucrose concentration of 210 g/L. Inulinase and invertase production (1.13 ± 0.02 and 5.84 ± 0.12 U/mL, respectively) in SmF was independent of the initial sucrose concentration; meanwhile, both activities increased in SSF as the sucrose concentration increased, reaching maximum values of 5.88 ± 0.78 and 41.69 ± 3.60 U/mL, respectively, at the highest sucrose concentration (210 g/L). The zymographic analysis showed one band of inulinase and two bands of invertases involved in sucrose hydrolysis.

The Atacama Desert, one of the driest places on earth, holds a rich biodiversity that becomes most appreciable in years when unusual rainfall accumulation triggers a phenomenon of explosive development of ephemeral herbaceous and woody desert species known as “desierto florido” or “blooming desert.” Despite the scientific importance of this unique phenomenon only few studies have addressed the mechanisms of flower phenotypic divergence under the fluctuating environment provided by this recurrent event. We investigated the mechanisms of floral color diversity in Cistanthe longiscapa (Montiaceae), a dominant species across the ephemeral blooming landscape of Atacama Desert. Our analyses show that the variation in colors of C. longiscapa flowers result from petals containing betalain pigments with different absorption spectra. The different pigment composition of petals causes flower color differences in the visible and ultraviolet (UV) range of the spectrum. Through color vision models we show that C. longiscapa flowers are highly polymorphic in their color appearance for insect pollinators. Our results highlight the variable nature in flower color of C. longiscapa varieties blooming simultaneously in a geographical restricted area. Given the importance of color in attracting floral visitors, the observed color variability could contribute to increased cross pollination in extreme desert conditions, while accounting for complex and fluctuating histories of plant-pollinator interactions.

The SARS-CoV-2 pandemic has challenged blood banks. In Mexico, donors decreased 22% between April and May 2020 compared to the same months in 2019. This study analyzed the effect of the strategies to recover donors (altruistic and family) in a tertiary pediatric care center during the pandemic.The Blood Bank of the Hospital Infantil de México Federico Gómez implemented strategies to obtain blood components to ensure self-sufficiency. The effect of these strategies on donor recovery was analyzed.There were 7,315 eligible donors in 2019 and 5,070 in 2020. Blood component requirements decreased from 10,037 units in 2019 to 8,619 in 2020. The strategies aimed at attracting altruistic donors managed to increase the percentage of this type of donor when comparing the months in which these strategies were applied with the same months in 2019. In addition, it was observed that the greater the number of methods used simultaneously, the higher the percentage of altruistic donors (rho = 0.846, p = 0.002). In contrast, strategies aimed at attracting family donors did not increase the number of this type of donor.Actions to recruit altruistic donors increased the number of this type of donor to meet the hospital's needs.La pandemia por SARS-CoV-2 ha representado un reto en los bancos de sangre. En México, los donadores disminuyeron el 22% entre abril y mayo del 2020 en comparación con los mismos meses del 2019. Este estudio analizó el efecto de las estrategias realizadas para recuperar donantes, altruistas y familiares, en un centro de atención pediátrica de tercer nivel durante la pandemia.El Banco de Sangre del Hospital Infantil de México Federico Gómez implementó estrategias encaminadas a la obtención de componentes sanguíneos para asegurar la autosuficiencia. Se analizó el efecto de dichas estrategias en la recuperación de donantes.Se registraron 7,315 donadores aptos en el año 2019 y 5,070 en el 2020. Los requerimientos de componentes sanguíneos disminuyeron de 10,037 unidades en 2019 a 8,619 en 2020. Las estrategias que estaban destinadas a captar donadores altruistas lograron aumentar el porcentaje de este tipo de donadores al comparar los meses en que se aplicaron dichas estrategias con los mismos meses en el 2019. Además, se observó que, a mayor número de estrategias aplicadas de manera simultánea, mayor porcentaje de donadores altruistas (rho = 0.846, p = 0.002). Por el contrario, las estrategias con la finalidad de atraer donadores familiares no lograron aumentar la cantidad de este tipo de donadores.Las acciones para recabar donadores altruistas aumentaron la cantidad de este tipo de donadores para satisfacer las necesidades del hospital.

Nuclear level densities (NLDs) and photon strength functions (PSFs), also called ¿ -ray or radiation strength functions, represent average properties of the nucleus in the regime of excitation where individual levels and transition probabilities by ¿ decay are not readily accessible by experimental or theoretical means. 3 páginas In this study, we sequenced the first complete chloroplast (cp) genome of a Peruvian chili pepper landrace, “arnacucho” (Capsicum chinense). This cp genome has a 156,931 bp in length with typical quadripartite structure, containing a large single copy (LSC) region (87,325 bp) and a 17,912 bp small single-copy (SSC) region, separated by two inverted repeat (IR) regions (25,847 bp); and the percentage of GC content was 37.71%. Arnaucho chili pepper chloroplast genome possesses 133 genes that consists of 86 protein-coding genes, 37 tRNA, eight rRNA, and two pseudogenes. Phylogenetic analysis revealed that this Peruvian chili pepper landrace is closely related to the undomesticated species C. galapagoense; all belong to the Capsiceae tribe.

Sweet cucumber (Solanum muricatum) sect. Basarthrum, is a neglected horticultural crop native of the Andean region. It is naturally distributed very close to potatoes (Solanum sect. Petota) and tomatoes (Solanum sect. Lycopersicon), two groups of high economic importance. To date, molecular tools for this crop are still undetermined. We here obtained the first complete chloroplast (cp) genome of sweet cucumber and compared with seven Solanaceae species. Pair-end clean reads were obtained by PE 150 library and the Illumina HiSeq 2500 platform. The complete cp genome of S. muricatum had a 155,681 bp with typical quadripartite structure, containing a large single copy (LSC) region (86,182 bp) and a small single-copy (SSC) region (18,360 bp), separated by two inverted repeat (IR) regions (25,568 bp). The annotation of chloroplast genome predicted 88 protein-coding genes (CDS), 8 ribosomal RNA (rRNA) genes, 37 transfer RNA (tRNA) genes, and one pseudogene. A total of 48 perfect microsatellites were identified, divided in mononucleotide repeats (32), followed by tetranucleotide (6) and dinucleotides (5). SSRs with trinucleotides repeats (3), pentanucleotide (1) and hexanucleotide (1) repeats motifs in these genomes were identified in lower quantity. Most of these repeats were distributed in the noncoding regions. Whole chloroplast genome comparison with the other seven Solanaceae species revealed that the small and large single copy regions showed more divergence than inverted regions. Finally, phylogenetic analysis resolved that S. muricatum is a sister species to members of sections Petota + Lycopersicum + Etuberosum. This study reports for the first time the genome organization, gene content, and structural features of the cp genome of S. muricatum. Also, this study may provide the basis for evaluating genetic diversity within Solanum, and will be useful to examine the evolutionary processes in sweet cucumber landraces.

Native to the southeastern United States, the honey locust (Gleditsia triacanthos) is an invasive tree in several South American countries. Since eradication of invasive species is often costly, prevention is a better strategy. The relationship between invasive species and their habitats can be analyzed using species distribution models to produce maps of areas prone to the invasions. These maps can be used to develop efficient early detection plans of exotic species colonization. Here, we employed the Favorability Function model to assess the effects of environment and human activities on the invasive process of the honey locust in Uruguay. By integrating environmental and anthropic factors in our models, we obtained the best fitted prediction and classification indices. We showed that the southwestern region of the country concentrates the largest proportion of areas prone to the invasions. Environment was the main factor explaining the invasion of G. triacanthos, but the effect of human-related factors had a greatest effect in combination with environmental variables than on its own. We generated favorable risk maps and explanatory variables that can be used to more efficiently plan efforts to control the spread of this invasive.

IntroductionAfter knowing the safety of the sentinel lymph node and the omission of axillary dissection inpatients with clinically negative and / or positive (1-2 lymph nodes) breast cancer withconservative surgery and the evidence from the ACOSOG Z011 randomized clinical trial andother evidence caution against proceeding with the use of axillary lymph node dissection inbreast cancer patients; a standardization of this practice is established globally, however indeveloping countries where treatments are affected by their availability, little is known aboutthe frequency of acquisition of these practices.MethodsAn online survey was carried out in different Latin American countries, which was distributedvia electronic link and QR code, with an open invitation to congresses and medical associations.The study lasted from August 16th, 2019 to April 30th, 2020. Surgeons with low, intermediateand high clinical practice in breast cancer were categorized, their type of clinical practice(private or public) and the period of clinical practice after discharge of the specialty.ResultsIn total, 260 replies were collected from surgeons from 10 different countries: Bolivia (1),Colombia (15), Ecuador (6), El Salvador (19), Guatemala (12), Nicaragua (12), Mexico (182),Paraguay (1), Peru (1) and Venezuela (11).Of these, the group that participated the most was the Surgical Oncologists (surgeons andgynecologists with a subspecialty in oncology), representing 82% of the participants, the rest ofthe participants being: Mastologists, Gynecologists and General Surgeons.The age group with the highest participation was 30-40 years old with 51.92% of the total(135), followed by the 41-50 year old group with 24% (63). 60% of the participants reported aprofessional practice of 10 years or less. In addition to referring to a mixed but mostly publicpractice in 48%, however, 56.93% of the participants reported a low practice (10 or less casesof breast cancer per month) of which between 60-100% of the occasions they offer earlysurgical treatment to their patients; responding that they always perform Sentinel Ganglion incN0 only 50% of the total, however it was mentioned that on average 85% of the occasions it isobtained between 2-3 lymph nodes number. The question was asked about completing theaxillary dissection when having 1-2 positive nodes with the scenarios of "always complete""almost always" "only in mastectomy" "only in breast conservation" and 63% answered thatalways or almost always will perform axillary dissection, 38.83% even referred the probabilityof doing it with micrometastasis in the setting of conservative surgery. In an exploratory manner, a question was added about the possibility of sentinel node after neoadjuvanttherapy, for which 52% answered that they would not perform such a procedure.ConclusionThe result of this survey shows a trend of resistance of less extensive surgical treatments in theaxilla for patients with breast cancer. Thus, reflecting that Latino populations are perhapsoverexposed to locoregional treatment; similar findings were reported by Dr. Morrow in astudy published in 2018 which showed the substantial variation in surgeon acceptance formore limited procedures. Nanomedicine and the pharmaceutical industry demand the investigation of new biomaterials to improve drug therapies. Combinations of lipids, proteins, and polymers represent innovative platforms for drug delivery. However, little is known about the interactions between such compounds and this knowledge is key to prepare successful drug delivery systems.Biophysical properties of biohybrid vesicles (BhVs) composed of phospholipids, proteins, and amphiphilic block copolymers, assembled without using organic solvents, were investigated by differential scanning calorimetry and dynamic light scattering. We studied four biohybrid systems; two of them included the effect of incorporating tetracaine. Thermal changes of phospholipids and proteins when interacting with the amphiphilic block copolymers and tetracaine were analyzed.Lysozyme and the copolymers adsorb onto the lipid bilayer modifying the phase transition temperature, enthalpy change, and cooperativity. Dynamic light scattering investigations revealed relevant changes in the size and zeta potential of the BhVs. Interestingly, tetracaine, a membrane-active drug, can fluidize or rigidize BhVs.We conclude that positively charged regions of lysozyme are necessary to incorporate the block copolymer chains into the lipid membrane, turning the bilayer into a more rigid system. Electrostatic properties and the hydrophilic-lipophilic balance are determinant for the stability of biohybrid membranes.This investigation provides fundamental information associated with the performance of biohybrid drug delivery systems and can be of practical significance for designing more efficient drug nanocarriers.

The 235U(n,f) cross section was measured in a wide energy range (18 meV–170 keV) at the n_TOF facility at CERN, relative to 6Li(n,t) and 10B(n,α) standard reactions, with high resolution and accuracy, with a setup based on a stack of six samples and six silicon detectors placed in the neutron beam. In this paper we report on the results in the region between 18 meV and 10 keV neutron energy. A resonance analysis has been performed up to 200 eV, with the code SAMMY. The resulting fission kernels are compared with the ones extracted on the basis of the resonance parameters of the most recent major evaluated data libraries.Acomparison of the n_TOF datawith the evaluated cross sections is also performed from thermal to 10 keV neutron energy for the energy-averaged cross section in energy groups of suitably chosen width. A good agreement, within 0.5%, is found on average between the new results and the latest evaluated data files ENDF/B-VIII.0 and JEFF-3.3, as well as with respect to the broad group average fission cross section established in the framework of the standard working group of IAEA (the so-called reference file).
