3. Protection of Penaeus monodon against white spot syndrome by continuous oral administration of a low concentration of Bacillus subtilis spores expressing the VP28 antigen

In this study, Bacillus subtilis spores expressing a chimeric protein, CotB-VP28, were used as a probiotic vaccine to protect black tiger shrimps (Penaeus monodon) against white spot syndrome virus (WSSV) infection. Oral administration of pellets coated with CotB-VP28 spores (at ≥1 × 109 CFU per g pellet) to shrimps induced immune-relating phenoloxydase activity (PO) in shrimps after 14 days of feeding (prior challenge) and at day 3 post challenge (1·26 and 1·70 fold increase respectively). A 75% protection rate was obtained by continuous feeding of the spore-coated pellets at ≥1 × 109 CFU per g for 14 days prior to WSSV challenge and during all the postchallenge period. Even when the amount of CotB-VP28 spores in feed pellets was reduced down to ≥5 × 107 CFU per g and ≥1 × 106 CFU per g, relatively high protection rates of 70 and 67·5%, respectively, were still obtained. By contrast, feeding pellets without spores (untreated group) and with naked spores (PY79 group) at ≥1 × 109 CFU per g could not protect shrimps against WSSV. These data suggest that supplementation of CotB-VP28 spores at low dose of ≥1 × 106 CFU per g could be effective as a prophylactic treatment of WSS for black tiger shrimps. Significance and Impact of the Study This study reports the protective efficacy of Bacillus subtilis CotB-VP28 spores on black tiger shrimps (Penaeus monodon) against white spot syndrome virus infection. Oral administration of pellets coated with CotB-VP28 spores (≥1 × 109 CFU per g) conferred 75% protection after white spot syndrome virus challenge. Even after reducing CotB-VP28 spores in feed pellets to ≥1 × 106 CFU per g, 67·5% protections was still obtained. These data indicate that supplementation of CotB-VP28 spores at a low dose of ≥1 × 106 CFU per g could be effective in prophylaxis against white spot syndrome in black tiger shrimps.

A search for dark matter is performed looking for events with large missing transverse momentum and a Higgs boson decaying either to a pair of bottom quarks or to a pair of photons. The data from proton-proton collisions at a center-of-mass energy of 13TeV, collected in 2015 with the CMS detector at the LHC, correspond to an integrated luminosity of 2.3 fb(-1). Results are interpreted in the context of a Z'-two-Higgs-doublet model, where the gauge symmetry of the standard model is extended by a U(1) Z' group, with a new massive Z' gauge boson, and the Higgs sector is extended with four additional Higgs bosons. In this model, a high-mass resonance Z' decays into a pseudoscalar boson A and a light SM-like scalar Higgs boson, and the A decays to a pair of dark matter particles. No signi fi cant excesses are observed over the background prediction. Combining results from the two decay channels yields exclusion limits in the signal cross section in the m(Z')-m(A) phase space. For example, the observed data exclude the Z' mass range from 600 to 1860 GeV, for Z' coupling strength gZ' = 0: 8, the coupling of A with dark matter particles g(X) = 1, the ratio of the vacuum expectation values tan beta = 1, and m(A) = 300GeV. The results of this analysis are valid for any dark matter particle mass below 100 GeV.

Automated identification and characterization of clustered weld defects, which comprise a plural of closely distributed individual member defects, is challenging. In this paper, time, Cartesian and Polar domain features in phased array ultrasonic sectorial scanning data are studied to segment clustered weld defect echoes by density based and K-means clustering. Through multiple domain segmentation, all individual member defect echoes are successfully identified. Then size and depth estimation is performed on each individual member defect to describe its patterns and to locate its depth. The developed technique is applied to experimental data collected from scanning on a tubular T weld test sample and is shown to discriminate, locate, and measure all the individual defects inside clustered defects. Based on the geometrical features of each individual member defect, they are classified as either point, linear or planar defects.

1. The vital importance of the cellular defense forces in the resistance of the monkey to combined streptococcus and influenza virus infections has been demonstrated. 2. Some of the conditions prejudicial to the maintenance of an optimum cellular reserve in the infected animal have been revealed; viz., undernutrition, physical cold, intratracheal route of infection. 3. The potential threat exerted by latent foci of streptococci, and the importance, in relation to the combined infection with virus, of cellular and humoral immunity, together or separately, have been demonstrated. The essential rôle of optimum nutrition (notably as concerns the vitamin B complex, and folic acid specifically) in the prevention of disastrous illness from these infectious agents, individually or in combination, would seem to have been proven. 4. Signs of glomerular nephritis appeared in a significant number of monkeys receiving Streptococcus hemolyticus and influenza virus in sequence, followed by reinoculation or spontaneous reappearance of the streptococci. 5. Reinoculation of Streptococcus hemolyticus, group C, resulted in a prompt "booster" increase in the opsonic index. Virus instillation was followed by just as sudden a depression in this index. 6. Reinoculation failed to evoke either the granulocytosis or the leucopenia in monkeys which are characteristic effects of the streptococcus and the virus respectively when these agents are introduced for the first time by way of the nasal mucous membrane. 7. Simultaneous intranasal inoculation of influenza virus, type A, and Streptococcus hemolyticus, group C, in nutritionally normal Macaca mulatta failed to produce obvious signs of disease. In most of the animals, however, a streptococcus-induced leucocytosis followed by a delayed virus-induced granulopenia developed. 8. Inoculation of influenza virus followed in 4 to 17 days by streptococci produced obvious signs of disease in five of eleven animals which had become leucopenic as result of the action of the virus, and fatal streptococcal septicemia in two monkeys. 9. The development of signs of infection in previously healthy monkeys exposed to virus followed by streptococci confirms both the clinical and laboratory experience of other observers, that virus infection may predispose to secondary bacterial invasion, and, that at times, under unfavorable circumstances, the infection may become overwhelming. Although the complete mechanism of resistance is as yet not wholly clear, the depressant or inhibitory effect of the virus on both its cellular and humoral elements has been established.

1. Myeloblasts can be discriminated in the supravital technique by the great numbers of tiny mitochondria in the cytoplasm and the absence of any other vitally stainable substance. 2. There are three stages in the maturation of myelocytes. 3. These three phases can be correlated with three types of the oxidase reaction. 4. One case of myeloblastic leucemia showed such an amount of an abnormal type of amitosis as to suggest the disordered cell division of neoplasms. 5. In this case transfusions were correlated with a maturation of myeloblasts into myelocytes, with an increase of the oxidase reaction, and with an increase in amitosis.

The observations reported in the preceding communication1 led us to consider the possibility that the phagocytic cells occurring in peritoneal exudates might be separated into the same two groups that were found in the spleen. It has been possible to demonstrate that this is true. A normal rabbit was given intraperitoneally 25 cc. of whole blood, which had been removed by a sterile puncture from the heart of another rabbit, and, after 24 hours, samples of the exudate were removed and studied. Another injection was then given and the examination repeated at the end of the second 24 hours. This procedure was repeated at 24 hour intervals over as long a time as was desired. Some of the experiments were carried out on rabbits whose blood-cells had been carefully matched, while in other instances animals were selected in which the serum of one agglutinated the red blood-cells of the other and vice versa. The exhibition of purpura as a clinical sign of disease was first ascribed solely to the effects of pestilential fevers. With the demonstration, however, by Denys 1 in 1887 that the purpura of Werlhof is characterized by a lowered level of circulating blood platelets, the clinical purpuric states were definitely shown to consist of thrombocytopenic and nonthrombocytopenic groups. Further differentiation within the thrombocytopenic group has been accomplished slowly since Denys's observations and has depended largely on cumulative experience with known etiologic states in which both purpura and marked thrombocytopenia have been observed to occur. There has thus arisen the practical and important subclassification of thrombocytopenic purpura into two groups—the idiopathic and the symptomatic. This is justified because the successful management of the former often requires life-saving surgery, whereas the latter should be managed strictly by medical means. The reversal of these therapeutic principles may promote disaster to the patient. The

Progress in the solution of the problems of disease in man must depend to a considerable degree on fundamental progress in the recognition and interpretation of alterations in disturbed physiologic equilibriums. It may be true that physiology "has its own problems, great in number and enormous in complexity, and those of disease are not among them."1Nevertheless, experience has established that the origin and explanation of many abnormal states may be traced to deranged physiologic processes, and more particularly to disturbed physiologic equilibriums. Granted that this is beyond the scope of pure physiologic research, the recognition of states of physiologic imbalance and of pathologic alterations produced in the tissues by this imbalance then becomes an important concern of the clinical investigator. That this "borderline" territory between physiology and medicine has proved a very fruitful field for clinical investigation, in one direction at least, is quite apparent when one reviews
