Forty castrated Holstein calves underwent an adrenocorticotropic hormone (ACTH) challenge to assess the effects of premortem stress on the longissimus lumborum (LL) following harvest. LL biopsies were collected before the challenge, at different harvest times (2, 12, 24, and 48 h; n = 10), and after 14 d aging. The expression of small heat shock proteins (SHSPs), deglycase 1 (DJ-1), and troponin were analyzed. Blood was analyzed throughout the ACTH challenge and at harvest for cortisol, oxidative stress, and complete blood count (CBC). Color and myofibrillar fragmentation index (MFI) were measured in aged samples. Unexpectedly, calves from different harvest times differed ( p = 0.05) in cortisol response. Calves were divided into two different cortisol response groups (high or low; n = 20). Statistical analysis assessed the effects of cortisol response ( n = 20), harvest time ( n = 10), and their interaction. Harvest time altered SHSPs ( p = 0.03), DJ-1 ( p = 0.002), and troponin ( p = 0.02) expression. Harvest time and cortisol response impacted steak color ( p < 0.05), and harvest time altered steak pH ( p < 0.0001). Additionally, various CBCs were changed ( p < 0.05) by harvest time. Harvest time changed ( p = 0.02) MFI. These data demonstrate that the protein expression, color, and MFI of the LL may be influenced by premortem stress.

Holstein steers (n = 40; initial BW = 84.9 ± 7.1 kg) were used to study the genesis of liver abscesses (LA) using an acidotic diet challenge with or without intraruminal bacterial inoculation. Steers were housed in individual pens inside a barn and randomly assigned to one of three treatments: (1) low-starch control diet comprised primarily of dry-rolled corn and wet corn gluten feed (CON); (2) high-starch acidotic diet with steam-flaked corn (AD); or (3) acidotic diet plus intraruminal inoculation with Fusobacterium necrophorum subsp. necrophorum (9.8 × 108 colony forming units [CFU]/mL), Trueperella pyogenes (3.91 × 109 CFU/mL), and Salmonella enterica serovar Lubbock (3.07 × 108 CFU/mL), previously isolated from LA (ADB). Steers in AD and ADB were fed the acidotic diet for 3 d followed by 2 d of the CON diet, and this cycle was repeated four times. On day 23, ADB steers were intraruminally inoculated with the bacteria. At necropsy, gross pathology of livers, lungs, rumens, and colons was noted. Continuous data were analyzed via mixed models as repeated measures over time with individual steer as the experimental unit. Mixed models were also used to determine the difference in prevalence of necropsy scores among treatments. Ruminal pH decreased in AD and ADB steers during each acidotic diet cycle (P ≤ 0.05). LA prevalence was 42.9% (6 of 14) in ADB vs. 0% in AD or CON treatments (P < 0.01). Ruminal damage was 51.1% greater in ADB than in AD (P ≤ 0.04). Culture of LA determined that 100% of the abscesses contained F. necrophorum subsp. necrophorum, 0% contained T. pyogenes, 50% contained Salmonella, and 50% contained a combination of F. necrophorum subsp. necrophorum and Salmonella. The F. necrophorum subsp. necrophorum was clonally identical to the strain used for the bacterial inoculation based on phylogenetic analysis of the whole genome. This experimental model successfully induced rumenitis and LA in Holstein steers and confirms the central dogma of LA pathogenesis that acidosis and rumenitis lead to the entry of F. necrophorum into the liver to cause abscesses. Our findings suggest that an acidotic diet, in conjunction with intraruminal bacterial inoculation, is a viable model to induce LA. The objective was to investigate the influence of cattle origin and region of finishing on the prevalence of Salmonella , Escherichia coli O157:H7, and select antimicrobial resistance in E. coli populations. Yearling heifers ( n  = 190) were utilized in a 2 × 2 factorial arrangement. After determining fecal Salmonella prevalence, heifers were sorted into one of four treatments: heifers originating from South Dakota (SD) and finished in SD (SD-SD); heifers originating from SD and finished in Texas (SD-TX); heifers originating from TX and finished in SD (TX-SD); and heifers originating from TX and finished in TX (TX-TX). Fecal, pen, and water scum line samples were collected longitudinally throughout the study; hide swab and subiliac lymph node (SLN) samples were collected at study end. A treatment × time interaction was observed ( p  ≤ 0.01) for fecal Salmonella prevalence, with prevalence being greatest for TX-TX and TX-SD heifers before transport. From day (d) 14 through study end, prevalence was greatest for TX-TX and SD-TX heifers compared with SD-SD and TX-SD heifers. Salmonella prevalence on hides were greater ( p  ≤ 0.01) for heifers finished in TX compared with SD. Salmonella prevalence in SLN tended ( p  = 0.06) to be greater in TX-TX and SD-TX heifers compared with TX-SD and SD-SD. Fecal E. coli O157:H7 prevalence had a treatment × time interaction ( p  = 0.04), with SD-TX prevalence being greater than TX-SD on d 56 and SD-SD and TX-TX being intermediate. A treatment × time interaction was observed for fecal trimethoprim-sulfamethoxazole-resistant and cefotaxime-resistant E. coli O157:H7 prevalence ( p  ≤ 0.01). Overall, these data suggest that the region of finishing influences pathogenic bacterial shedding patterns, with the initial 14 d after feedlot arrival being critical for pathogen carriage.

Multidrug resistant (MDR) Escherichia coli threaten the preservation of antimicrobials to treat infections in humans and livestock. Thus, it is important to understand where antimicrobial-resistant E. coli persist and factors that contribute to its their development. Crossbred cattle ( n  = 249; body weight = 244 kg ±25 kg standard deviation) were blocked by arrival date and assigned metaphylactic antimicrobial treatments of sterile saline control, tulathromycin (TUL), ceftiofur, or florfenicol at random. Trimethoprim-sulfamethoxazole (COT R ) and third-generation cephalosporin (CTX R )-resistant E. coli were isolated from fecal samples on days 0, 28, 56, 112, 182, and study END (day 252 for block 1 and day 242 for block 2). Then, susceptibility testing was conducted on all confirmed isolates. MDR was detected in both COT R and CTX R E. coli isolates. In COT R isolates, the number of antimicrobials each isolate was resistant to and the minimum inhibitory concentration (MIC) for amoxicillin-clavulanic acid, ceftriaxone, and gentamicin was greatest on day 28 compared with all other days ( p  ≤ 0.04). Similarly, chloramphenicol MIC was greater on day 28 than on day 0 ( p  < 0.01). Overall, sulfisoxazole MIC was less for TUL than all other treatments ( p  ≤ 0.02), and trimethoprim-sulfamethoxazole MIC was greater for TUL than all other treatments ( p  ≤ 0.03). Finally, there was no effect of treatment, day, or treatment × day for tetracycline or meropenem MIC ( p  ≥ 0.07). In CTX R isolates, there was an effect of day for all antimicrobials tested except ampicillin and meropenem ( p  ≤ 0.06). In conclusion, administering a metaphylactic antimicrobial at feedlot arrival did influence the susceptibility of COT R and CTX R E. coli . However, MDR E. coli are widely distributed, and the MIC for most antimicrobials was not different from the initial value upon completion of the feeding period.

The objective was to provide an updated profile of the bovine acute-phase response to include recent advancements in technologies and expanded hematological, cytokine, and serum chemistry variables. Beef steers (n = 32; body weight [BW] = 251 ± 19.5 kg) were fitted with indwelling jugular catheters 1 d before lipopolysaccharide (LPS; 0.25 µg LPS/kg BW from Escherichia coli O111:B4) administration to facilitate serial blood collection. Rectal temperature was measured using indwelling probes, and ocular temperature was measured using infrared thermal imaging. Blood samples were collected for subsequent analysis of serum chemistry, hematology, and cytokine concentrations. Pearson correlation of rectal temperature and ocular infrared temperature was 0.61 (P < 0.01) and the Spearman correlation coefficient was 0.56 (P < 0.01). Interactions of hour × method were observed for ocular and rectal measurements of body temperature in response to endotoxin exposure. Maximum observed temperature was 39.6 °C at 2.5 h for both rectal and ocular measurements. Body temperature differed by method at hours 0.5, 2.5, 4.5, 7.5, 12.5, 36.5, and 47.5 (P < 0.01), but were not different otherwise. All variables of serum chemistry and complete blood count were influenced by LPS administration, except creatinine, serum glucose, and percent basophils (P ≤ 0.02). Alanine aminotransferase and alkaline phosphatase peaked at hour 2 relative to LPS administration, returned to baseline at hour 12 and continued to decrease below the baseline value at hour 48 (P < 0.01). Total protein concentration decreased 3% in response to LPS (P = 0.01). Total white blood cell count decreased 75% after LPS administration at hour 1 (P < 0.01). Lymphocyte count recovered to baseline at hour 6; sooner than neutrophil count at hour 36. Serum cortisol concentration increased 294% relative to baseline at hour 1 followed by a sustained decrease and return to normal concentration at hour 4 (P < 0.01). Additionally, circulating cytokine concentrations changed with time in response to the LPS challenge, excluding aFGF, bFGF, IGF-1, IL-2, IL-4, MCP-1, and ANG-1 (P ≤ 0.08). Maximum observed concentration of TNF-α at hour 1 was 117% greater than the pre-challenge value (P < 0.01). Data presented herein add to existing works to understand the endocrine and immune responses of beef steers administered exogenous LPS, and incorporate recent technologies, additional biomarkers, and an expanded cytokine profile that can be used as referential data in future research., (Published by Oxford University Press on behalf of the American Society of Animal Science 2023. Holstein steers (n = 40; initial body weight [BW] = 96.0 ± 10.5 kg) were individually housed in a climate-controlled barn to evaluate potential models for the genesis of liver abscesses (LA). In this 2 × 2 factorial, steers were balanced by BW and randomly assigned to one of two treatments: 1) intravenous saline injection followed by intraruminal bacterial inoculation with Fusobacterium necrophorum subsp. necrophorum (1 × 109 colony forming unit [CFU]/mL) and Salmonella enterica serovar Lubbock (1 × 106 CFU/mL; CON; n = 20 steers); or 2) intravenous injection with 0.25 µg/kg BW of lipopolysaccharide (LPS; Escherichia coli O111:B4) followed by intraruminal bacterial inoculation of F. necrophorum subsp. necrophorum (1 × 109 CFU/mL) and S. enterica serovar Lubbock (1 × 106 CFU/mL; LBI; n = 20 steers) and 1 of 2 harvest dates (3 or 10 d post LPS infusion). Body weights were recorded on days -4, -1, 3, and 10, and blood was collected for hematology on days -4, 3, and 10, relative to LPS infusion on day 0. Intraruminal bacterial inoculation occurred on day 1. Steers from each treatment group were harvested at two different time points on day 3 or 10 to perform gross pathological examination of the lung, rumen, liver, LA (if present), and colon. Feed disappearance was less for LBI than CON (P < 0.01); however, BW did not differ (P = 0.33) between treatments. Neither treatment nor time differed for hematology (P ≥ 0.13), and no gross pathological differences were noted in the lung, liver, LA, or colon (P ≥ 0.25). A treatment × harvest date interaction was noted for ruminal pathology in which LBI had an increased percentage of abnormal rumen scores on day 3 (P < 0.01). These results suggest that an LPS challenge in combination with intraruminal bacterial inoculation of pathogens commonly isolated from LA was not sufficient to induce LA in steers within 3 or 10 d (P = 0.95) when compared to CON. This study evaluated the growth and immune response of beef calves born from Bos indicus-influenced beef heifers provided pre- and postpartum heat abatement on pasture. On 83 ± 4 d prepartum (day 0), 64 Brangus crossbred beef heifers (~¼ B. indicus) were stratified by body weight (BW; 454 ± 37 kg) and body condition score (BCS; 6.3 ± 0.28; scale 1 to 9), and then allocated into 1 of 16 bahiagrass pastures (1 ha and 4 heifers per pasture). Treatments were randomly assigned to pastures (8 pastures per treatment) and consisted of heifers provided (SH) or not (NSH) access to artificial shade (4.5 m2 of shade area per heifer) from 83 d prepartum to 50 d postpartum (days 0 to 133). Heifers and calves were managed similarly from day 133 until the start of the breeding season (day 203). Calves were weaned on day 203 (at 119 ± 19 d of age), limit-fed the same drylot diet at 3.5% of BW (DM basis) days 209 to 268 (3 to 4 calves per pen; 8 pens per treatment) and vaccinated against respiratory disease pathogens on days 222 and 236. Heifer intravaginal temperatures from days 35 to 42 were lower (P ≤ 0.03) for NSH vs. SH heifers from 0000 to 0800 hours but greater (P ≤ 0.05) for NSH vs. SH heifers from 1100 to 1800 hours. Heifer intravaginal temperature from days 126 to 132 did not differ (P = 0.99) between NSH and SH heifers. Heifers assigned to NSH had greater respiration rates from days 20 to 96 (P ≤ 0.0007), greater plasma concentration of cortisol on days 35 (P = 0.07) and 55 (P = 0.02), less plasma concentration of insulin-like growth factor 1 (IGF-1) on days 35 (P = 0.10), 55, and 133 (P ≤ 0.05), and less BCS from days 55 to 203 (P ≤ 0.01) compared to SH heifers. Calves born from NSH heifers had less birth BW (P = 0.05), greater overall plasma haptoglobin concentrations (P = 0.05), greater seroconversion against bovine respiratory syncytial virus on day 222 (P = 0.02), tended to have greater ADG from days 209 to 268 (P = 0.07), and had greater BW on day 268 (P = 0.05) compared to SH offspring. Plasma concentrations of cortisol and serum titers against other respiratory disease pathogens did not differ (P ≥ 0.15) between NSH and SH offspring. Hence, removing maternal access to artificial shade: (1) increased prepartum intravaginal temperature and plasma concentrations of cortisol but reduced prepartum BCS and plasma concentrations of IGF-1 in grazing B. indicus-influenced beef heifers; and (2) increased post-weaning BW gain and had positive effects on humoral immune response of their offspring., (© The Author(s) 2023. The potential for sexually dimorphic innate immune responses to respiratory disease was evaluated, where eight steers and seven heifers (280 ± 4 kg) were subjected to a viral−bacterial respiratory disease challenge utilizing bovine herpesvirus-1 (BoHV-1; intranasal; 1 × 108 PFU/nostril) and Mannheimia haemolytica (MH; intratracheal; 1.3 × 107 CFU/head) administered 72 h later. Body temperature was lesser in heifers than steers (p < 0.01). There was a sex × time interaction (p = 0.05) for white blood cells where heifers had reduced concentrations compared with steers at −72 and 0 h but greater concentrations from 36 to 60 h post-MH. Concentrations of neutrophils were lesser in heifers compared to steers from 0 to 4 h, and from 8 to 12 h (p = 0.03). Lymphocytes were greater in heifers compared to steers at 12 h and from 36 to 60 h post-MH (p < 0.01). The neutrophil−lymphocyte ratio was lesser in heifers compared to steers from 2 to 24 h and at 48 h post-MH (p < 0.01). Monocytes were greater in heifers compared to steers from 24 to 60 h post-MH (p < 0.01), while eosinophils were greater in heifers compared to steers at 48 and 60 h (p < 0.01). Serum IL-4 was lesser in heifers compared to steers at 0 h and from 2 to 72 h post-MH challenge (p = 0.02). Non-esterified fatty acid concentrations were lesser (p < 0.01) in heifers compared to steers from 2 to 4 h post-MH challenge. Urea nitrogen concentrations were greater (p < 0.01) in heifers than steers at 36 h post-MH challenge. Data from this study reveal distinct differences in the acute phase response following a respiratory disease challenge where steers produced an early response, while the response in heifers appeared to be delayed.

Bovine respiratory disease (BRD) is the primary animal health concern facing feedlot producers. Many antimicrobial mitigation strategies are available, but few studies have compared feedlot performance during both the receiving and finishing periods following application of different antimicrobials used as metaphylaxis at arrival. The objective of this study was to compare antimicrobial metaphylaxis methods on clinical health and growth performance across both the receiving and finishing periods. A total of 238 multiple-sourced steers in two source blocks were used in a generalized complete block design. The four treatments included: 1) a negative control, 5 mL of sterile saline injected subcutaneously (CON); 2) subcutaneous administration of florfenicol at 40 mg/kg of BW (NUF); 3) subcutaneous administration of ceftiofur in the posterior aspect of the ear at 6.6 mg/kg of BW (EXC); and 4) subcutaneous administration of tulathromycin at 2.5 mg/kg of BW (DRA). The morbidity rate for the first treatment of BRD was decreased for the DRA and EXC treatments compared to CON and NUF ( P <  0.01). Additionally, average daily gain (ADG), dry matter intake (DMI), and gain-to-feed (G:F) were greater ( P  ≤ 0.02) in the DRA treatment during the receiving period compared to all other treatments. The ADG was also greater ( P  < 0.05) for EXC than the CON treatment throughout the finishing period. Nonetheless, other growth performance variables did not differ among metaphylactic treatments during the finishing period ( P ≥  0.14). Likewise, no differences in carcass characteristics or liver abscess score were observed ( P ≥  0.18). All complete blood count (CBC) variables were affected by day ( P ≤  0.01) except mean corpuscular hemoglobin concentration ( P  = 0.29). Treatment × time interactions were observed for platelet count, white blood cell (WBC) count, monocyte count and percentage, and lymphocyte percentage ( P ≤  0.03). However, there were no observed hematological variables that differed among treatment ( P ≥  0.10). On day 0 of years 1 and 2, 64 Brangus crossbred heifers per year were stratified by initial body weight (BW) and age (mean = 257 ± 20 kg and 271 ± 22 d) and allocated into 16 bahiagrass (Paspalum notatum) pastures (4 heifers per pasture per yr). Treatments were randomly allotted to pastures in a 2 × 2 factorial arrangement of treatments (4 pastures per treatment per yr). Treatments consisted of concentrate dry matter (DM) supplementation at 1.50% of BW from days 0 to 100 (CON) or concentrate DM supplementation at 1.05% of BW from days 0 to 49 and 1.95% of BW from days 50 to 100 (SST). Then, each respective supplementation strategy was added or not with immunomodulatory feed ingredients from days 0 to 100 (OMN; 4 g/45 kg of BW). Heifers were assigned to an estrus synchronization protocol from days 100 to 114. Heifers detected in estrus from days 111 to 114 were inseminated (AI) 12 h after estrus detection. Heifers not detected in estrus were timed AI on day 114. All heifers were exposed to Angus bulls from days 120 to 210 (1 bull per pasture). Effects of supplementation strategy × OMN inclusion × hour were detected (P < 0.0001) only for intravaginal temperature from days 26 to 30, which were the least (P ≤ 0.03) for SST heifers offered OMN supplementation and did not differ (P ≥ 0.17) among all remaining treatments from 0830 to 1600 hours. Effects of supplementation strategy × OMN inclusion and OMN inclusion were not detected (P ≥ 0.12) for any variable, except for the percentage of heifers detected in estrus, which was greater (P = 0.01) for heifers supplemented with vs. without OMN. Total concentrate DM offered from days 0 to 100 and heifer BW on days 0 and 56 did not differ (P ≥ 0.49) between CON and SST heifers, but SST heifers were heavier (P ≤ 0.01) on days 100 and 210 compared with CON heifers. Body surface temperature on day 25 and plasma IGF-1 concentrations on day 75 were greater (P ≤ 0.04) for SST vs. CON heifers. Percentage of pubertal heifers, heifers detected in estrus, and pregnancy to AI did not differ (P = 0.36) between SST and CON heifers but the final pregnancy percentage was greater (P = 0.04) for SST vs. CON heifers. Thus, OMN supplementation decreased the intravaginal temperature of SST heifers but failed to improve their growth and reproduction, whereas the SST strategy improved body thermoregulation, growth, and final pregnancy percentage of heat-stressed Bos indicus-influenced beef heifers compared with a constant concentrate supplementation strategy., (© The Author(s) 2022. This study evaluated whether administration of lipopolysaccharide (LPS) at each trimester of gestation would alter the acute phase (APR) and metabolic responses to a postnatal LPS challenge in weaned heifers. Pregnant crossbred multiparous cows ( n = 50) were randomized into prenatal immune stimulation (PIS; n = 24; administered 0.1 µg/kg BW LPS subcutaneously at 71 ± 2, 170 ± 2 and 234 ± 2 d of gestation) and saline (CON; n = 26) groups. From these treatment groups, heifer calves ( n = 12 PIS and 11 CON) were identified at weaning (244 ± 3 d of age) to receive an LPS challenge. On d 0, heifers were fitted with vaginal temperature (VT) devices, jugular catheters, and moved into individual stalls. On d 1, heifers were challenged i.v. with LPS (0.5 µg/kg BW) at 0 h. Blood samples were collected and sickness behavior scores (SBS) recorded at 0.5 h intervals from -2 to 8 h and at 24 h relative to LPS challenge. Serum was analyzed for cortisol, cytokines, glucose, non-esterified fatty acids (NEFA), and serum urea nitrogen (SUN) concentrations. Baseline VT was lesser in PIS heifers from -11 to -5 h pre-LPS (treatment × time: P < 0.01) compared to the CON; however, the post-LPS VT response did not differ between treatments ( P = 0.89). There was a treatment × time interaction ( P < 0.01) for SBS with PIS heifers having lesser SBS from 0.5 to 2 h post-LPS compared to CON. There was a treatment × time interaction ( P = 0.03) for cortisol with PIS heifers having greater cortisol at 0.5, 3, 3.5, 5.5 and 6.5 h post-LPS compared to CON. There were treatment × time interactions for the post-LPS cytokine responses ( P ≤ 0.05). Specifically, PIS heifers had greater TNF-α from 1.5 to 2 h, yet less TNF-α at 3 h than CON ( P < 0.01), and PIS heifers had greater IFN-γ from 3.5 to 5.5 h post-LPS than CON ( P < 0.01). In contrast, IL-6 was less in PIS than CON heifers from 1.5 to 8 h post-LPS ( P < 0.001). Glucose concentrations were greater in PIS heifers at -1 h, but less at 2, 3 and 5.5 h compared to CON (treatment × time: P < 0.01). Serum NEFA concentrations were greater ( P = 0.04) in PIS than CON heifers. There was a treatment × time interaction ( P < 0.01) for SUN with PIS heifers having greater SUN concentrations at -2, -1.5, 2, 3, 6.5 and 24 h than CON. These data demonstrate that in utero exposure to multiple low doses of endotoxin has lasting physiological and immunological effects when the offspring encounter a similar postnatal immunological insult., (Published by Oxford University Press on behalf of the American Society of Animal Science 2021.)

Nutritional supplementation has been used by livestock producers for many years in order to increase animal performance, improve animal health, and reduce negative effects associated with enteric and/or respiratory pathogens. Supplements such as yeast and yeast-based products have broad applications across many livestock production systems, including poultry, aquaculture, cattle, and swine and have been shown to benefit animal production at various stages. These benefits include improvement in milk production, weight gain and feed conversion, as well as immune function. Initial research into the mode of action for these effects has focused on stimulation of the immune system by the β-glucan fractions of yeast. However, emerging studies have revealed that some of the beneficial effects of yeast products may stem from altering metabolism, including the availability of glucose and fatty acids. These changes in metabolism, and potentially energy availability, may partially explain differences in immune function observed in yeast-supplemented livestock, as the energy demands of an activated immune system are extremely high. Thus, this paper explores the influence of yeast products on metabolism in cattle and swine, and how changes in metabolism and energy availability may contribute to improvements in immune function in supplemented animals.

Yearling crossbred beef steers [ N = 238; initial shrunk body weight (BW) = 402 ± 31.2 kg] were used to investigate the influence of a Bacillus subtilis probiotic on animal growth performance, efficiency of dietary net energy (NE) utilization, carcass characteristics, and fecal and subiliac lymph node Salmonella prevalence during a 140-d finishing period at the Southeast Research Farm in Beresford, SD. Steers were allotted to 1 of 24 pens ( N = 9-10 steers/pen) and assigned to 1 of 2 dietary treatments (12 pens/treatment): no probiotic (CON) or 0.5 g/steer/d of a B. subtilis PB6 probiotic (CLOSTAT500, Kemin Industries, Des Moines, IA; CLO). Bunks were managed according to a slick bunk management approach. Fecal samples were collected on study days 1, 28, 56, 112, and 140 from a subsample of steers from each pen ( N = 5 steers/pen) via rectal palpation and composited by pen for the determination of Salmonella prevalence using selective enrichment and culture media. Upon harvest, subiliac lymph nodes were obtained from an equal number of steers from each treatment (collected from every other steer) following evisceration and hide removal. Data were analyzed as a randomized complete block design and pen served as the experimental unit; an α of 0.05 determined significance. Live-basis final BW and average daily gain tended ( P ≤ 0.06) to be reduced for CLO. No differences were detected ( P ≥ 0.11) between treatments for dry matter intake or gain efficiency. Treatment neither altered the efficiency of dietary NE utilization nor calculated dietary NE content based upon observed performance ( P ≥ 0.46). No differences were detected between treatments for any carcass traits ( P ≥ 0.15). No Salmonella was recovered in any fecal samples collected on study days 1, 28, or 56. On day 112, steers from CLO had a numerically lower ( P = 0.17; 25.0 vs. 8.3%) incidence of fecal Salmonella compared to CON. On study day 140, fecal Salmonella incidence did not differ between treatments ( P = 0.34; 0.0 vs. 8.3%) for CON and CLO, respectively. Upon harvest, no Salmonella was recovered in any subiliac lymph nodes. These data indicate that B. subtilis PB6 did not influence feedlot phase growth performance or fecal Salmonella prevalence. The objective of this study was to determine if immunosuppression through daily dexamethasone (DEX) infusion altered Salmonella translocation from the gastrointestinal tract. Weaned Holstein steers ( n  = 20; body weight [BW] = 102 ± 2.7 kg) received DEX ( n  = 10; 0.5 mg/kg BW) or saline (control [CON]; n  = 10;) for 4 days (from day -1 to 2) before oral inoculation of naldixic acid-resistant Salmonella enterica Typhimurium (SAL; 3.4 × 10 6 colony-forming units [CFU]/animal) on day 0. Fecal swabs were obtained daily, and blood was collected daily for hematology. At harvest (day 5), ileum, cecal fluid, lymph nodes (ileocecal, mandibular, popliteal, and subscapular), and synovial (stifle, coxofemoral, and shoulder) samples were collected for isolation of the inoculated strain of SAL. White blood cell (WBC) and neutrophil concentrations were elevated ( p  < 0.01) in DEX calves following each administration event. Following inoculation, 100% of DEX calves shed the experimental strain of SAL for all 5 days, 90% of CON calves shed from day 1 to 3, and 100% of CON calves shed from day 4 to 5. Greater ( p  < 0.01) concentrations of SAL were quantified from the cecum of DEX calves (3.86 ± 0.37 log CFU/g) compared with CON calves (1.37 ± 0.37 log CFU/g). There was no difference in SAL concentrations between DEX and CON calves in ileal tissue ( p  = 0.07) or ileocecal ( p  = 0.57), mandibular ( p  = 0.12), popliteal ( p  = 0.99), or subscapular ( p  = 0.83) lymph nodes. Of the stifle samples collected, 3.3% were positive for SAL, highlighting a contamination opportunity during hindquarter breakdown. While more research is needed to elucidate the interactions of immunosuppression and pathogen migration patterns, these data confirm that orally inoculated SAL can translocate from the gastrointestinal tract and be harbored in atypical locations representing a food safety risk.

This study was conducted to determine if feeding a Saccharomyces cerevisiae fermentation product (SCFP) to calves would alter the acute phase response to a lipopolysaccharide (LPS) challenge. Crossbred steer calves [ n = 32; 274 ± 1.9 kg body weight (BW)] were randomly allotted to two treatment diets for 21 d: 1) control, fed RAMP (Cargill, Dalhart, TX) and 2) SCFP, fed the control ration supplemented with NaturSafe at 12 g/hd/d mixed into the TMR (NaturSafe, Diamond V, Cedar Rapids, IA). On day 22, steers were fitted with indwelling jugular catheters and rectal temperature monitoring devices and placed in individual bleeding stalls. On day 23, steers were challenged i.v. with 0.25 µg/kg BW LPS. Blood samples were collected at 0.5-h (serum) or 2-h (complete blood counts) intervals from -2 to 8 h and again at 24 h relative to the LPS challenge at 0 h. Sickness behavior scores (SBS) were recorded after the collection of each blood sample. Rectal temperatures were greater in SCFP steers from 6 to 11 h, at 13 h, from 15 to 20 h, and from 22 to 24 h following the LPS challenge compared to Control steers (treatment × time: P = 0.01). Additionally, SCFP-supplemented steers had reduced ( P < 0.01) SBS compared to Control steers. Platelet concentrations remained greater in SCFP-supplemented steers compared to Control steers throughout the study ( P = 0.05), while there was a tendency ( P = 0.09) for SCFP steers to have greater white blood cells and eosinophils concentrations than Control steers. There was a treatment × time interaction for serum cortisol and glucose ( P < 0.01). Specifically, cortisol was greater at 0.5 and 2 h postchallenge but was reduced at 3 h for SCFP steers compared to Control steers. Glucose was greater in SCFP steers at -0.5, 2, and 7.5 h compared to Control steers. Serum amyloid A was reduced in SCFP steers at 0.5 h, yet greater at 1 and 7.5 h postchallenge compared to Control steers (treatment × time: P < 0.01). Fibrinogen concentrations were greater ( P < 0.01) in SCFP compared to Control steers. There was a treatment × time interaction ( P < 0.01) for tumor necrosis factor-α (TNF-α) such that concentrations were reduced in SCFP steers from 1 to 2 h postchallenge compared to Control steers. Overall, these data suggest that supplementing calves with SCFP may have primed the innate immune response prior to the challenge, particularly platelets, which resulted in an attenuated sickness behavior and TNF-α response to LPS., (© The Author(s) 2020. Anecdotal data would suggest that weather patterns influence beef cattle health in feedyards, and cattle producers often associate the seasonality of some illnesses with changes in environmental temperatures. However, to our knowledge, there is little information from large-scale feeding operations and precision weather stations that establishes a link or lack thereof between weather patterns and cattle health. Additionally, we are unaware of any studies correlating other weather parameters with animal health data. Therefore, the objective of this study was to test for associations between monthly temperature variation and animal morbidity/mortality in feedlots in the Texas Panhandle. Weather data was collected from a Texas Tech University Mesonet weather station in close proximity to 19 beef cattle feedyards in the Texas Panhandle. Additionally, near real-time morbidity and mortality data was collected from those yards from 2015 to 2018. These data document a seasonal pattern relative to cattle morbidity and mortality with most health events occurring from November to January. This pattern is differentiated when comparing morbidity and mortality by listed causation (e.g., respiratory, digestive, other), and the majority of deaths over the entire time course were attributed to respiratory disease. Most cattle morbidity was documented in the winter months, most of which were classified as respiratory disorders. Additionally, an increase in health events was observed as the population of the feedyard increased. However, the overall effects of ambient temperature on cattle health were minimal and the two may not be causally linked. To evaluate the effects of a patented Bacillus subtilis probiotic, weaned Holstein steers, not shedding Salmonella ( n  = 40; ∼90 kg), were supplemented (CLO) or not (CON) with CLOSTAT ® (13 g/hd per day; Kemin Industries, Des Moines, IA) in a starter ration for 35 d. The calves were assigned to one of four treatments in a 2 × 2 factorial design with CLO and CON calves that were orally administered Salmonella (STM) or not (NoSTM). Calves were challenged with 1.6 × 10 6 colony-forming unit (CFU) Salmonella Typhimurium (resistant to 50 μg/mL nalidixic acid) in 1 L of milk replacer on day 0. Blood samples were collected through jugular catheters every 6 h for 96 h, and body temperature was measured every 5 min through indwelling rectal temperature recording devices. Five calves from each treatment were harvested 48 h postchallenge, and the remaining calves were harvested 96 h postchallenge. During necropsy, tissues were collected for the isolation and quantification of the inoculated STM from various tissues. The CLOSTM group had reduced STM concentrations in the jejunum, ileum, and transverse colon 48 h after the challenge ( p  ≤ 0.03), but were not different 96 h postchallenge ( p  > 0.05). Decreased ( p  < 0.01) pyrexia was observed after the challenge in CLOSTM calves when compared with CONSTM calves. White blood cells and lymphocyte counts were increased ( p  ≤ 0.05) in CLOSTM calves after the challenge in comparison with other treatments. In calves given STM, the CLO group had greater feed intake before and after the challenge ( p  < 0.01) compared with the CON group. Increased serum IL-6 and IFN-γ concentrations were observed in the CONSTM group compared with other treatments. Overall, CLO reduced Salmonella presence and concentrations in gastrointestinal tissues while simultaneously reducing the severity of the challenge as indicated by blood parameters and the reduced febrile response.

Adrenal responsiveness was tested in nonpregnant, lactating Holstein dairy cows fed diets supplemented with OmniGen-AF (OG; Phibro Animal Health Corp., Teaneck, NJ), an immune modulator, and in nonsupplemented control (CON) cows following bolus infusions of a combination of corticotropin-releasing hormone (CRH; 0.3 µg/kg of BW) and arginine vasopressin (VP; 1.0 µg/kg of BW) or ACTH (0.1 IU/kg of BW) in 2 environments: thermoneutral [TN; temperature-humidity index (THI) <60] for 24 h/d and heat stress (HS; THI >68 for 17 h/d). Cows (506) were initially fed OG (n = 254) or CON (n = 252) diets for 44 d before selection of a subgroup of cows (n = 12; 6 OG, 6 CON) for the study. The 2 subgroups were balanced for parity, milk yield, and days in milk. All cows were transported to and housed in 2 environmentally controlled rooms at the University of Arizona Agricultural Research Complex (Tucson). Cows were given 3 d to acclimate to the rooms and then underwent 12 d of TN conditions and then 8 d of HS conditions for a total of 24 d on experiment. Cows were infused with CRH-VP on d 9 of TN and on d 1 of HS and with ACTH on d 10 of TN and on d 2 of HS. Hormone infusions took place at 1000 h (0 h) on each infusion day. Blood samples, taken in 30-min intervals, were first collected at 0800 h (-2 h) and were drawn until 1800 h (8 h). Before infusion, serum progesterone was elevated in OG cows compared with CON cows. Infusion of releasing factors (CRH-VP or ACTH) caused increases in serum cortisol and progesterone, but cortisol release was greater in CON cows than in OG cows during HS, whereas progesterone did not differ between the 2 treatments. Serum ACTH increased following infusion of releasing factors, but this increase was greater following CRH-VP infusion than ACTH infusion. Serum bovine corticosteroid-binding globulin also increased following infusion of releasing factors in both treatment groups, but this increase was greater during HS in cows fed OG. The free cortisol index (FCI) increased following CRH-VP and ACTH and was higher in HS than in TN for both OG and CON cows. However, the FCI response was blunted in OG cows compared with CON cows during HS. Heat stress enhanced the adrenal response to releasing factors. Additionally, the adrenal cortisol and FCI response to releasing factors was reduced during acute heat stress in cows fed OG. Objective: To quantify acute immunologic and metabolic responses of beef heifers following topical administration of transdermal flunixin meglumine (TDFM) at various times relative to bovine herpesvirus 1 (BHV1) and Mannheimia haemolytica challenges., Animals: 32 beef heifers (mean body weight, 170 kg)., Procedures: Heifers were assigned to 1 of 4 groups. Heifers in the control group did not receive TDFM, whereas 1 dose of TDFM (3.3 mg/kg) was topically applied to heifers of groups A, V, and B at -144, -72, and 0 hours. All heifers were inoculated with 1 × 10 8 plaque-forming units of BHV1 in each nostril at -72 hours and with 1.18 × 10 6 CFUs of M haemolytica intratracheally at 0 hours. Vaginal temperature was recorded and blood samples were collected for quantification of select immunologic and metabolic biomarkers at predetermined times from -144 to 360 hours., Results: Mean vaginal temperature was similar between group A and the control group. Mean vaginal temperatures for groups V and B were generally lower than that for the control group following BHV1 and M haemolytica challenges, respectively. Mean neutrophil oxidative burst capacity and L-selectin expression at 0 hours were significantly decreased for group V relative to the other groups. Other biomarkers did not differ among the groups at any time., Conclusions and Clinical Relevance: Results suggested that topical administration of TDFM to beef cattle effectively alleviated pyrexia without adverse effects on acute immunologic or metabolic responses when TDFM was administered at the same time as, but not before, respiratory pathogen challenge.

This study was designed to determine if supplementation of a combination live yeast and yeast cell wall product in feed could mitigate the negative impacts associated with heat stress (HS). Crossbred, phenotypically similar beef heifers ( n = 32; BW = 385 ± 43 kg) were fed a standard finishing ration without (CON) or with a combination of a live yeast (1.5 g/hd/d) and yeast cell wall product (2.5 g/hd/d; YEAST; Phileo Lesaffre Animal Care, Milwaukee, WI). After 50 d of supplementation, heifers were transported to an environmentally-controlled facility and placed in individual bleeding stalls after indwelling jugular catheters and vaginal temperature (VT) loggers were inserted. Heifers were kept in thermoneutral (TN) conditions for 48 h [temperature-humidity index (THI) ~67; d 1-2] then were subjected to HS for 4 d (peak THI ~80; d 3-6). From d 2-6, hourly blood samples were collected for serum isolation from 1400 to 1800 h and again from 2200 to 0200 h which represented the daily targeted peak and nadir of THIs. A whole blood sample was collected twice daily at 1400 and 2200 h for complete blood counts (CBC). There was no difference in BW ( P = 0.14) or ADG ( P = 0.53) between the treatments during HS. Yeast-supplemented heifers exhibited reduced VT during HS compared to CON heifers ( P < 0.01). There was no difference in water intake during the TN phase ( P = 0.25); however, YEAST heifers consumed more water/h ( P < 0.01) and had increased drinking bouts ( P < 0.01) during HS compared to CON heifers. Respiration rates (RR) did not differ ( P = 0.21) during TN, but YEAST heifers tended ( P = 0.09) to have decreased RR during HS compared to CON heifers. There were no differences between treatments when evaluating CBC parameters ( P ≥ 0.10). There was a tendency ( P = 0.08) for greater cortisol in the CON than YEAST heifers during HS; however, glucose ( P = 0.38) and NEFA ( P = 0.70) concentrations did not differ. In summary, supplementation of live yeast and yeast cell wall products to feedlot heifers may mitigate some of the negative effects associated with HS in feedlot cattle as observed in decreased RR and VT and increased water intake., (Copyright © 2020 Broadway, Carroll, Burdick Sanchez, Cravey and Corley.)

Dietary omega-3 polyunsaturated fatty acids (n-3 PUFA) are precursors for lipid metabolites that reduce inflammation. Two experiments were conducted to test the hypothesis that enriching the sow diet in n-3 PUFA during late gestation and throughout lactation reduces stress and inflammation and promotes growth in weaned pigs. A protected fish oil product (PFO; Gromega) was used to enrich the diet in n-3 PUFA. In the initial experiment, time-bred gilts were fed a gestation and lactation diet supplemented with 0% (control; n = 5), 0.25% (n = 4), 0.5% (n = 4), or 1% (n = 5) PFO from 101 ± 2 d of gestation to day 16 of lactation. Adding 1% PFO to the diet increased the n-3:n-6 PUFA ratio in colostrum and milk compared with controls (P = 0.05). A subsequent experiment was performed to determine whether supplementing the sow diet with 1% PFO improved growth and reduced circulating markers of acute inflammation and stress in the offspring. Plasma was harvested from piglets (16 per treatment group) on day 0 (d of weaning) and days 1 and 3 postweaning. Pigs from the 1% PFO treatment group weighed more (P = 0.03) on day 3 postweaning and had a greater (P ˂ 0.05) n-3:n-6 PUFA ratio in plasma on each day sampled compared with 0% PFO controls. There was an overall treatment effect on plasma total cortisol (P = 0.03) and haptoglobin (P = 0.04), with lesser concentrations in pigs on the 1% PFO diet. Plasma corticosteroid-binding globulin (CBG) concentrations were not different between treatment groups but were less (P ˂ 0.001) on days 1 and 3 when compared with day 0. The resultant free cortisol index [FCI (cortisol/CBG)] was less (P = 0.02) on days 1 and 3 for pigs from the 1% treatment group compared with the controls. An ex vivo lipopolysaccharide (LPS) challenge of whole blood collected on days 0 and 1 was used to determine whether 1% PFO attenuated release of inflammatory cytokines (IL-1β, IL-6, and TNF-α). Blood from pigs within the 1% PFO treatment group tended (P = 0.098) to have a lesser mean concentration of TNF-α in response to LPS compared with blood from controls. These results suggest that providing a PFO supplement as 1% of the diet to sows beginning in late gestation and during lactation can increase the n-3:n-6 PUFA ratio in their offspring, which may improve growth and reduce the acute physiological stress response in the pigs postweaning., (Published by Oxford University Press on behalf of the American Society of Animal Science 2019.)

Available energy plays a critical role in the initiation and maintenance of an immune response to a pathogen, a process that is further altered by activation of the stress system. This study was designed to determine the effect of an acute vs chronic stress model on the metabolic response to vaccination in naïve beef steers. Steers ( n = 32; 209 ± 8 kg) were blocked by body weight (BW) and randomly assigned to one of three treatments: 1) Chronic stress (CHR), 0.5 mg/kg BW dexamethasone (DEX) administered i.v. at 1000 h on day 3 to day 0; 2) Acute stress (ACU), 0.5 mg/kg BW DEX administered i.v. at 1000 h on day 0 only; or 3) Control (CON), no DEX. On day -4, steers were fitted with jugular vein catheters and moved into individual bleeding stalls in an environmentally-controlled facility. Blood samples were collected at -74, -50, and -26 h, at 0.5-h intervals from -4 to 6 h, and at 12, 24, 36, 48, and 72 h relative to vaccination with a combination vaccine (Pyramid 5 + Presponse SQ, Boehringer Ingelheim Animal Health USA, Duluth, GA) at 1200 h on day 0. Data were analyzed by the MIXED procedure of SAS specific for repeated measures. There was a treatment × time interaction ( P < 0.001) for serum glucose concentrations. Specifically, glucose concentrations increased at -50 h in CHR steers and at 1200 h in ACU steers and remained elevated through 72 h postvaccination period in these two treatments compared to CON steers. The change in nonesterified fatty acid ( NEFA ) concentrations relative to baseline values was affected by treatment and time ( P < 0.001) such that the change in NEFA was greater in CHR (0.06 ± 0.01 mmol/L), followed by CON (-0.01 ± 0.01 mmol/L) and ACU steers (-0.04 ± 0.01 mmol/L). There was a tendency ( P = 0.08) for a treatment × time interaction for change in serum NEFA concentrations. Serum urea nitrogen (SUN) was affected by treatment and time ( P < 0.001) such that SUN concentrations were greatest in CHR (12.0 ± 0.1 mg/dL) followed by ACU (10.4 ± 0.1 mg/dL) and CON steers (9.6 ± 0.1 mg/dL); however, the treatment × time interaction was not significant ( P = 0.12). These data demonstrate that activation of the stress and immune axes using an acute or chronic stress model can increase energy mobilization prior to and following vaccination in naïve steers, potentially affecting available energy needed to mount an adequate antibody response to vaccination., (Published by Oxford University Press on behalf of the American Society of Animal Science 2019.)

This study determined whether feeding the immunomodulating supplement, OmniGen-AF, to feedlot heifers would alter metabolic profiles to a glucose tolerance test. Heifer calves ( n = 32; 217 ± 2 kg) were allocated into two treatment diets: 1) Control, fed a standard receiving ration, and 2) OmniGen, fed the Control diet supplemented with OmniGen at 4.54 g/45 kg BW/d. Heifers were fed for 42 d. On d 42, Heifers were processed through a working facility for placement of indwelling jugular catheters. After these procedures, heifers were moved into individual stanchions in an enclosed barn and all heifers were fed their treatment diets at 1400 h. All orts were removed at 2000 h to allow for a 12-h fast prior to first blood collection. The following day, heifers were administered 0.5 mL/kg BW of a 50% dextrose solution at 0900 h (0 min). Blood samples were collected for serum isolation at -60, -45, -30, -15, 0, 10, 20, 30, 45, 60, 90, 120, and 150 min relative to bolus dextrose infusion. Serum was stored at -80 o C until analyzed for cortisol, glucose, insulin, non-esterified fatty acid (NEFA) and urea N concentrations. There was a treatment × time interaction for post-challenge cortisol ( P = 0.004) such that cortisol was greater in OmniGen heifers than Control heifers from 10- to 45- min post-infusion. Glucose concentrations increased post-infusion ( P < 0.01) and were reduced in OmniGen compared to Control heifers at 10-, 45-, and 90-min after challenge (treatment × time P < 0.001). Similarly, there was a treatment × time interaction for post-challenge insulin concentrations ( P = 0.04) such that insulin was greater in OmniGen-fed heifers than Control heifers from 10 to 30 min. In addition, there was a treatment × time interaction ( P = 0.01) for NEFA concentrations such that concentrations were reduced in OmniGen-supplemented heifers from 10 to 30 min following administration of the dextrose bolus. Serum urea N concentrations were greater in Control heifers at 150 min compared to OmniGen-fed heifers (post-challenge treatment × time interaction: P < 0.001). These data suggest that OmniGen-fed heifers were more responsive to changes in glucose, perhaps affecting the storage and/or redistribution of energy deposits and provide further evidence for altered metabolism in OmniGen-supplemented cattle. Stressors experienced by pigs at weaning may negatively impact health and productivity. Thus, supplements that enhance pig immunity during the early post-weaned period are of great interest to the swine industry. The objective of this experiment was to evaluate the performance and hematological responses of weaned pigs supplemented with yeast cell wall (YCW) when challenged orally with Salmonella Typhimurium . Weaned pigs were assigned to one of three treatments for 22d ( n = 13/treatment): Control diet, which was a non-medicated starter diet (Control); Control diet supplemented with YCW at 250 mg/kg BW (YCW250; Phileo Lesaffre Animal Care, Milwaukee, Wisconsin, USA); and Control diet supplemented with YCW at 500 mg/kg BW (YCW500). On d19 blood samples were collected from -6 to 72 h relative to oral Salmonella Typhimurium (1 × 10 6 cfu/pig) challenge. Gain:feed was greater ( P = 0.01) in YCW250 treatment compared to both Control and YCW500 pigs. Baseline intraperitoneal temperature was greater ( P < 0.001) in YCW250 pigs than Control or YCW500 pigs. There was a treatment x time interaction for the change in intraperitoneal temperature ( P < 0.01), post-challenge cortisol, white blood cell counts (WBC), neutrophils, and neutrophil:lymphocyte ratio ( P ≤ 0.03). Control pigs had greater ( P < 0.05) cortisol concentrations than both YCW-supplemented groups at 0 h, but Control pigs had reduced ( P < 0.05) cortisol compared to YCW500 pigs at 24 and 30 h post-challenge. Control pigs had greater ( P < 0.05) WBC counts than both YCW-supplemented groups 6 and 12 h post-challenge, and YCW250 pigs had reduced ( P < 0.01) WBC counts than Control and YCW500 pigs 18 h post-challenge. Neutrophil counts were greater ( P < 0.05) in Control pigs than both YCW-supplemented groups at 6 and 12 h post-challenge and were greater ( P = 0.02) than YCW250 pigs at 18 h post-challenge. Lymphocytes were greater ( P < 0.001) in Control and YCW500 pigs pre- and post-challenge compared to YCW250 pigs. Control pigs had the greatest ( P < 0.001) monocyte counts compared to YCW treatments. There was no effect of yeast supplementation on fecal shedding or Salmonella counts in the rectum, colon or cecum ( P ≥ 0.05). While some differences were observed in intraperitoneal temperature and some hematological variables, data suggests there were minimal effects of yeast supplementation on the acute immune response to Salmonella challenge.

The objective of this study was to assess the influence of prenatal stress (PNS) on innate immune responses to an endotoxin challenge in weaned bull calves. Altered innate immune response to lipopolysaccharide (LPS), as characterized by changes in a range of variables was hypothesized in PNS bull calves. Brahman cows (n = 96; 48 stressed by transportation at five stages of gestation and 48 Controls) produced 85 calves, from which 16 uncastrated male (bull) calves from each PNS and Control treatment were selected for an LPS challenge period. Rectal temperature (RT), sickness behavior score (SBS), serum concentrations of cortisol, interferon-gamma (IFN-γ), tumor necrosis factor-alpha (TNF-α), interleukin-6 (IL-6), and complete blood count (CBC) variables were assessed in response to intravenous LPS (0.25 μg/kg body weight) administration. Each reported variable increased or decreased following LPS administration. Prior to LPS, PNS bull calves exhibited increased TNF-α, IL-6, and monocyte counts, but decreased IFN-γ, eosinophils, and basophils (p < .05). Compared with Control, in response to LPS, PNS bull calves exhibited greater circulating concentrations of cortisol. PNS bull calves exhibited lower (p < .05) eosinophil and basophil counts at time 0 (time of LPS administration) but similar counts to Control bull calves 2 h after LPS. PNS bull calves exhibited a greater change from baseline for IFN-γ and monocytes in response to LPS administration. No other variables were influenced by prenatal treatment (p > .05). These findings suggest that PNS did not adversely affect basal or induced components of the innate immune response to an immunological challenge. Lay summary Our laboratory studied the influence of prenatal stress (i.e., transportation of pregnant cows) on immune function of bull calves at 8 months of age. This was accomplished by studying aspects of their innate immune response to an immunological challenge. Prenatal stress did not adversely affect basal or induced components of the innate immune response to an immunological challenge.

Lactobacillus acidophilus fermentation products have been used to improve the performance of nursery pigs. However, research on the influence of this supplement on health is lacking. This study was designed to determine if feeding a Lactobacillus acidophilus fermentation product to weaned pigs would reduce stress and acute phase responses (APR) following a lipopolysaccharide (LPS) challenge. Pigs (n=30; 6.4±0.1 kg) were individually housed in stainless steel pens with ad libitum access to feed and water. Pigs were weighed upon arrival, assigned to one of three groups (n=10/treatment), and fed for 18 days: (1) Control, fed a non-medicated starter diet; (2) Control diet with the inclusion of a Lactobacillus acidophilus fermentation product at 1 kg/metric ton (SGX1) and (3) Control diet with the inclusion of a Lactobacillus acidophilus fermentation product at 2 kg/metric ton (SGX2). On day 7 pigs were anesthetized for insertion of an i.p. temperature device, and similarly on day 14 for insertion of a jugular catheter. Pigs were challenged i.v. with LPS (25 µg/kg BW) on day 15. Blood samples were collected at 0.5 h (serum) and 1 h (complete blood cell counts) intervals from -2 to 8 h and at 24 h relative to LPS administration at 0 h. Pigs and feeders were weighed on days 7, 14 and 18. The supplemented pigs had increased BW and average daily gain before the challenge. In response to LPS, there was a greater increase in i.p. temperature in Control pigs compared with supplemented pigs. In addition, cortisol was reduced in SGX2 pigs while cortisol was elevated in SGX1 pigs at several time points post-challenge. White blood cells, neutrophils and lymphocytes were decreased in SGX1 and SGX2 compared with Control pigs. Furthermore, the pro-inflammatory cytokine response varied by treatment and dose of treatment. Specifically, serum TNF-α was greatest in SGX2, intermediate in Control, and least in SGX1 pigs, while the magnitude and temporal pattern of IFN-γ in SGX2 pigs was delayed and reduced. In contrast, IL-6 concentrations were reduced in both SGX treatment groups compared with Control pigs. These data demonstrate that different supplementation feed inclusion rates produced differential responses, and that feeding SynGenX to weaned pigs attenuated the APR to an LPS challenge.

Two treatments were evaluated in heifers to determine the effects of a yeast supplement on immune and metabolic responses to a combined (tandem viral-bacterial) respiratory disease challenge. Thirty-two beef heifers (325 ± 20.1 kg BW) were selected from a larger population previously assigned to one of two treatments: Control (CON), receiving no yeast supplement in the diet, or yeast (YST), CON diet plus a combination live yeast (2.5 g·heifer -1 ·d -1 ) and yeast cell wall (2.5 g·heifer -1 ·d -1 ) supplement (Phileo Lesaffre Animal Care, Milwaukee, WI). Heifers were maintained on treatments for 31 d prior to the challenge. On day -3 all heifers were fitted with an indwelling vaginal temperature recording device, received an intranasal challenge with 2 × 10 8 plaque-forming units of bovine herpesvirus-1 (BHV-1), and placed in outdoor pens. On day 0, all heifers were fitted with an indwelling jugular catheter, challenged intratracheally with an average dose of 3.0 × 10 7 colony-forming units (cfu) of Mannheimia haemolytica in 100 mL media, and were transferred into individual stanchions in an enclosed, environmentally controlled barn. Whole blood samples were collected at -72 h and at 0, 2, 4, 6, 8, 12, 24, 36, 48, 60, and 72 h (hematology) and at 0, 1, 2, 3, 4, 5, 6, 7, 8, 12, 24, 36, 48, 60, and 72 h (serum isolation) relative to M. haemolytica challenge (0 h). Data were analyzed using the MIXED procedure of SAS specific for repeated measures with fixed effects of treatment, time, and the treatment × time interaction. Vaginal temperature and cortisol concentrations were similar between treatments ( P ≥ 0.39). Although total leukocyte count following BHV-1 challenge was similar between treatments ( P = 0.21), there was a tendency ( P = 0.07) for CON heifers to have greater neutrophil counts than YST heifers. Serum haptoglobin concentration was similar between treatments ( P = 0.13). Heifers in the YST treatment had similar serum glucose concentrations ( P = 0.25) and decreased serum concentrations of urea nitrogen compared to CON ( P = 0.03). Dietary treatment did not affect serum nonesterified fatty acid concentrations ( P = 0.37). Nasal lesion score severity (0-4) tended ( P = 0.07) to be decreased in YST compared to CON (2.5 vs. 3.19 ± 0.26), while water intake tended to be increased in YST ( P = 0.06). Feeding a yeast supplement had little effect on the acute-phase response but improved metabolic outcomes in heifers during a respiratory disease challenge., (Published by Oxford University Press on behalf of the American Society of Animal Science 2018.)

This study was designed to determine the effect of prenatal lipopolysaccharide (LPS) exposure on the postnatal metabolic response to an LPS challenge in beef heifers. Pregnant, crossbred cows ( = 50) were assigned to a prenatal immune stimulation (PIS; = 25; administered 0.1 µg/kg BW LPS subcutaneously 233 ± 15d of gestation) or saline treatment group (Control; = 25). Birth and weaning BW of calves were collected. There was not (> 0.05) a treatment × gender interaction for birth weight or 205-d adjusted weaning BW. Treatment did not affect (> 0.05) birth BW, but steers and heifers of PIS cows had greater ( < 0.02) 205-d adjusted weaning BW than offspring from Control cows. From the 2 prenatal treatment groups, heifer calves ( = 12 PIS, 11 Control) were identified at weaning (238 ± 15 d of age) to subsequently receive an LPS challenge. On d 0, heifers were fitted with indwelling jugular catheters and were moved into individual pens. On d 1, heifers (fed at 0600 h) were challenged i.v. with LPS (0.5 µg/kg BW) at 0 h (1000 h). Blood samples were collected at 30-min intervals from -2 to 8 h and again at 24 h relative to the LPS challenge. There was a treatment × time interaction ( < 0.01) for cortisol; PIS heifers had greater cortisol from 4 to 6.5 h post-LPS challenge ( < 0.001). There was a treatment × time interaction ( = 0.04) for serum glucose such that glucose was greater ( = 0.01) in PIS than Control heifers at 0.5 h, but was greater in Control than PIS heifers at 2, 4.5, and 7 h post-LPS challenge. This resulted in overall time ( < 0.01) and treatment ( < 0.01) effects such that Control heifers had greater glucose concentrations than PIS heifers. There was a tendency ( = 0.10) for a treatment × time interaction for serum NEFA, such that NEFA was greater in Control than PIS heifers at -2, -1.5, and 7 h relative to the LPS challenge ( ≤ 0.02). Also, there were time ( < 0.01) and treatment effects ( < 0.01) for NEFA with Control heifers having greater NEFA than PIS heifers. Serum BUN was affected by a treatment × time interaction ( < 0.01). Concentrations of BUN were greater in PIS heifers from -1.5 to -1 h, 1 to 2 h, at 4 h, and from 5 to 24 h relative to the LPS challenge. These results demonstrate postnatal growth and the metabolic responses of weaned beef calves can be significantly altered with a single exposure to LPS in utero.

The objective of this research was to examine the effects of dexamethasone (DEX) treatment on various aspects of immunity following administration of a multivalent respiratory vaccine, using a model intended to mimic acute versus chronic stress. Angus × Hereford steers ( = 32; 209 ± 8 kg) were stratified by BW and randomly assigned to 1 of 3 treatments: 1) acute stress (ACU), in which 0.5 mg/kg BW DEX was intravenously administered at 1000 h only on d 0; 2) chronic stress (CHR), in which 0.5 mg/kg BW DEX was intravenously administered at 1000 h on d -3 to 0; or 3) control (CON), in which no DEX was administered. Steers were fitted with indwelling jugular catheters and rectal temperature (RT) recording devices on d -4 relative to vaccination and placed in individual stanchions in an environmentally controlled facility. Blood samples were collected and serum was isolated at -74, -50, and -26 h; at 0.5-h intervals from -4 to 6 h; and at 12, 24, 36, 48, and 72 h relative to multivalent respiratory vaccination at 1200 h on d 0. Additional blood samples were used to analyze complete blood cell count (CBC) and functional capacities of neutrophils. There was a treatment × time interaction ( < 0.01) for RT such that DEX treatment in CHR and ACU steers decreased RT on d -3 and 0, respectively. A treatment × time interaction ( < 0.01) was observed for total white blood cells (WBC), neutrophils, lymphocytes, and monocytes. Specifically, DEX increased WBC and neutrophils in CHR and ACU steers ( < 0.001) yet decreased lymphocytes in CHR steers ( = 0.02) compared with CON steers. Neutrophil concentration increased rapidly, within 2 h of the DEX infusion, in ACU steers. Monocytes transiently increased ( < 0.001) in response to DEX treatment in CHR and ACU steers. In contrast, eosinophils were greater ( < 0.01) in CON steers than in ACU and CHR steers. A treatment × time interaction ( = 0.004) was observed for interferon-γ, with CON cattle exhibiting greater concentrations than the ACU and CHR cattle at 5 h after vaccination, through d 3. Treatment also influenced ( ≤ 0.001) the expression of L-selectin on the surface of neutrophils. The percentage of neutrophils engaging in phagocytosis and the oxidative burst were suppressed ( ≤ 0.001) among only the CHR steers, whereas the intensity of the oxidative burst was suppressed ( ≤ 0.001) for both ACU and CHR steers. These data suggest that our model induced acute and chronic immunosuppression and defined the acute response to a multivalent vaccine in CON steers.

Sexually dimorphic innate immune responses have been observed in several species, but have not been studied in response to a live pathogen challenge in pigs. This study aimed to elucidate sexually dimorphic innate immune responses along with Salmonella translocation patterns in newly weaned pigs orally inoculated with Salmonella . Newly weaned pigs ( n = 8 gilts and 12 barrows; 6.2 ± 0.2 kg BW) were obtained from a commercial swine facility and were maintained in an environmentally-controlled facility in individual pens equipped with feeders and nipple waterers. Pigs were allowed ad libitum access to a commercial non-medicated starter ration and water throughout the study. On d 12 post-weaning, pigs were anesthetized to allow placement of a temperature measuring device in the abdominal cavity for measurement of intraperitoneal temperature (TEMP). On d 17, pigs were anesthetized and fitted with indwelling jugular vein catheters. On the following day (d 18), pigs were orally inoculated with 4.7×10 9 Salmonella typhimurium . Blood samples were collected at 0.5-h intervals from -2 to 8 h, and at 8-h intervals from 8 to 72 h post-challenge. Whole blood was analyzed for complete blood cell counts. Serum was isolated for measurement of cortisol. Following collection of the 72 h sample, pigs were humanely euthanized and tissues were collected for Salmonella isolation. There was a sex × time interaction ( P < 0.001) for TEMP such that gilts had a greater TEMP response to the Salmonella challenge compared to barrows. There was also a sex × time interaction ( P = 0.03) for serum cortisol with gilts having decreased cortisol at 16 h yet greater cortisol at 32 h than barrows. Barrows had greater total white blood cells (17.8 vs. 16.2 ± 0.4 10 3 cells/μL; P < 0.01; respectively) and neutrophils (7.8 vs. 6.1 ± 0.4 10 3 cells/μL; P < 0.01; respectively) than gilts. However, gilts had greater lymphocytes (9.6 vs. 9.0 ± 0.2 10 3 cells/μL; P = 0.05; respectively) than barrows. While immune parameters were influenced by sex, there was no effect of sex ( P > 0.05) on Salmonella concentrations from fecal shedding 3 d post-inoculation in the cecum, mesenteric and subiliac lymph nodes, liver, spleen, gallbladder, or kidney tissues. These data demonstrate that weaned gilts appear to produce a stronger acute phase response to a Salmonella challenge compared to barrows, without affecting the tissue translocation or shedding of Salmonella .

The potential effect of prenatal LPS exposure on the postnatal acute phase response (APR) to an LPS challenge in heifers was determined. Pregnant crossbred cows were separated into prenatal immune stimulation (PIS) and saline groups (Control). From these treatments, heifer calves were identified at weaning to subsequently receive an exogenous LPS challenge. Sickness behavior scores (SBS) were recorded and blood samples were collected at 30-min intervals from -2 to 8 h and again at 24 h relative to the LPS challenge. There was a treatment × time interaction for the change in vaginal temperature (VT) such that the change in VT was greater in Control than PIS from 150 to 250 min, yet it was greater in PIS than Control from 355 to 440 min and from 570 to 1145 min. There was also a treatment × time interaction for SBS such that scores were greater in Control than PIS at 0.5 h, yet were greater in PIS than Control from 2.5 to 4 h post-LPS. There was a tendency for a treatment × time interaction for serum concentrations of IL-6, which were greater in PIS than Control heifers from 5.5 to 6 h and from 7 to 8 h post-challenge. Thus, a single exposure to LPS during gestation can alter the postnatal APR to LPS in heifer calves.

Our objective was to examine immunosuppression induced by dexamethasone (DEX) administration in cattle on immunological responses to a multivalent respiratory vaccine containing replicating and nonreplicating agents. Steers ( = 32; 209 ± 8 kg) seronegative to infectious bovine rhinotracheitis virus (IBRV), bovine viral diarrhea virus (BVDV), bovine respiratory syncytial virus (BRSV), and parainfluenza-3 virus (PI3V) were stratified by BW and randomly assigned to 1 of 3 treatments: 1) acute immunosuppression (ACU; 0.5 mg/kg BW DEX intravenously at 1000 h only on d 0), 2) chronic immunosuppression (CHR; 0.5 mg/kg BW DEX intravenously at 1000 h on d -3 to 0), or 3) a control (CON; no DEX). On d -4, steers were fitted with intravenous catheters in the jugular vein and placed into individual stanchions. At 1200 h on d 0, steers were administered a respiratory vaccine containing modified-live virus (MLV) isolates of IBRV, BVDV, BRSV, and PI3V and a (MH) toxoid. On d 4, cattle were transported (177 km) and housed in an isolated outdoor pen. Serum was harvested on d 0, 7, 14, 21, 28, 35, 42, and 56 to determine IBRV-, BVDV-, BRSV-, and PI3V-specific antibody titers and MH whole cell and leukotoxin antibody concentrations. Sera from d -2, 0, 1, 3, 7, and 14 were used to quantify haptoglobin (Hp) concentration and ceruloplasmin (Cp) activity. Nasal swab specimens were collected on d 0, 3, and 14 to determine the presence of IBRV, BVDV, BRSV, and PI3V via PCR analysis. There was a treatment × day interaction ( < 0.01) such that CHR steers had a greater ( ≤ 0.07) BVDV antibody titer on d 14, 21, and 28. Moreover, IBRV-specific antibodies increased beginning on d 14 for CHR and on d 28 for ACU and remained greater through d 56 compared with CON ( ≤ 0.03). Conversely, serum MH whole cell antibody concentration was least ( ≤ 0.06) for CHR from d 7 to 28 and greatest for CON ( ≤ 0.04) on d 56. Treatment altered Hp such that CON exhibited a greater ( < 0.01) Hp concentration than CHR but was not different from ACU ( = 0.16). On d 3, Cp was greatest for CON, intermediate for ACU, and least for CHR (treatment × day; ≤ 0.01). The prevalence of IBRV and BVDV in nasal swabs on d 14 was 67 and 56%, respectively, for CHR; 10 and 10%, respectively, for CON; and 9 and 0%, respectively, for ACU ( ≤ 0.006). Results suggest that CHR allowed increased replication of MLV vaccine agents. Conversely, DEX-induced immunosuppression blunted the acute phase protein and antibody response against the nonreplicating MH toxoid.

Cattle temperament, defined as the reactivity of cattle to humans or novel environments, can greatly influence several physiological systems in the body, including immunity, stress, and most recently discovered, metabolism. Greater circulating concentrations of nonesterified fatty acids (NEFAs) found in temperamental cattle suggest that temperamental cattle are metabolically different than calm cattle. Further, elevated NEFA concentrations have been reported to influence insulin sensitivity. Therefore, the objective of this study was to determine whether cattle temperament would influence the metabolic response to a glucose tolerance test (GTT) and insulin sensitivity test (IST). Angus-cross steers (16 calm and 15 temperamental; 216 ± 6 kg BW) were selected based on temperament score measured at weaning. On day 1, steers were moved into indoor stanchions to allow measurement of individual ad libitum feed intake. On day 6, steers were fitted with indwelling rectal temperature probes and jugular catheters. At 9 AM on day 7, steers received the GTT (0.5-mL/kg BW of a 50% dextrose solution), and at 2 PM on day 7, steers received the IST (2.5 IU bovine insulin/kg BW). Blood samples were collected and serum isolated at -60, -45, -30, -15, 0, 10, 20, 30, 45, 60, 90, 120, and 150 min relative to each challenge. Serum was stored at -80°C until analyzed for cortisol, glucose, NEFA, and blood urea nitrogen concentrations. All variables changed over time (P < 0.01). For the duration of the study, temperamental steers maintained greater (P < 0.01) serum NEFA and less (P ≤ 0.01) serum blood urea nitrogen and insulin sensitivity (calculated using Revised Quantitative Insulin Sensitivity Check Index) compared with calm steers. During the GTT, temperamental steers had greater (P < 0.01) serum glucose, yet decreased (P = 0.03) serum insulin and (P < 0.01) serum insulin: serum glucose compared to calm cattle. During the IST, temperamental steers had greater (P < 0.01) serum insulin and a greater (P < 0.01) serum insulin: serum glucose as compared with calm steers. These data demonstrate that differences exist in the manner in which temperamental steers respond to glucose and insulin, potentially a result of elevated serum NEFA concentrations, which may result in changes in utilization and redistribution of energy in temperamental vs calm cattle., (Published by Elsevier Inc.)

The objective of this study was to determine the metabolic, stress, and hematology response of beef heifers supplemented with zilpaterol hydrochloride (ZH) when exposed to an endocrine stress challenge. Heifers ( = 20; 556 ± 7 kg BW) were randomized into 2 treatment groups: 1) control (CON), no ZH supplementation, and 2) zilpaterol (ZIL), supplemented with ZH at 8.33 mg/kg (DM basis). The ZIL group was supplemented ZH for 20 d, with a 3-d withdrawal period. On d 24, heifers received an intravenous bolus of corticotropin-releasing hormone (CRH; 0.3 µg/kg BW) and arginine vasopressin (VP; 1.0 µg/kg BW) to activate the stress axis. Blood samples were collected at 30-min intervals for serum and 60-min intervals for plasma and whole blood, from -2 to 8 h relative to the challenge at 0 h (1000 h). Samples were analyzed for glucose, insulin, NEFA, blood urea nitrogen (BUN), cortisol, epinephrine, norepinephrine, and complete blood cell counts. Following the challenge, cattle were harvested over a 3-d period. Liver, LM, and biceps femoris (BF) samples were collected and analyzed for glucose, lactate, and glycolytic potential (GP). There was a treatment ( ≤ 0.001) effect for vaginal temperature (VT), with ZIL having a 0.1°C decrease in VT when compared with CON. A treatment × time effect ( = 0.002) was observed for NEFA. A treatment effect was observed for BUN; ZIL had decreased BUN concentrations compared with CON ( < 0.001) prior to the challenge; however, no treatment × time effect was observed. There was also a treatment effect for cortisol ( ≤ 0.01) and epinephrine ( = 0.003); ZIL had decreased cortisol and epinephrine during the CRH/VP challenge when compared with CON. There was a time effect for total white blood cells, lymphocytes, and monocytes; each variable increased ( ≤ 0.01) 2 h postchallenge. Additionally, neutrophil counts decreased ( ≤ 0.01) in response to CRH/VP challenge in both treatment groups. Glucose concentrations within the LM were greater ( = 0.03) in CON when compared with ZIL. Lactate concentrations and GP within the BF were greater in CON ( = 0.05) when compared with ZIL. These data suggest there are some variations observed between treatments in terms of response to the CRH/VP challenge; however, in the environmental conditions of this trial, none of the variations observed suggest that the supplementation of ZH detrimentally alters the ability of cattle to effectively respond to stressful stimuli.

English × Continental heifers ( = 180) were sourced in 2 loads (219.3 ± 16.0 and 221.4 ± 16.4 kg, respectively) from commercial auction barns to study the effects of feeding dehydrated citrus pulp (DCP) on feedlot performance of newly received heifers. A completely randomized block design was used with BW nested within arrival load and blocked by BW into 3 dietary treatments (36 pens, 5 heifers/pen, 12 blocks, 3 pens/block, and 12 pens/treatment). Treatment diets contained 1) 0% DCP (control diet [CON]), 2) 10% DCP, or 3) 20% DCP on a DM basis. Diets containing DCP were exchanged with steam-flaked corn on a 1:1 basis. Cattle were fed a 63, 73, and 83% concentrate diet from d 0 to 28, d 28 to 42, and d 42 to 56, respectively. Over the 56-d trial period, as the amount of dietary DCP increased, DMI decreased ( = 0.01), ADG decreased ( < 0.01), and G:F decreased ( = 0.02). From d 0 to 28, there was no difference in the observed minus the predicted NEg of the diet ( = 0.73); from d 28 to 42, there was a linear increase in NEg favoring DCP treatments ( < 0.01); and from d 42 to 56, there was a linear decrease in NEg against the DCP treatments ( < 0.01). At the conclusion of the trial, a subset of heifers ( = 22; 307.89 ± 3.32 kg on d 63) were used to evaluate blood metabolite concentrations before and after a lipopolysaccharide (LPS) challenge. On d 63, heifers were fitted with jugular catheters and moved into individual stalls. On d 64, heifers were intravenously challenged with LPS (0.5 μg/kg BW), and blood samples were collected every 0.5 h from -2 to 8 h and at 24 h relative to the LPS challenge (0 h). Serum glucose, serum urea nitrogen (SUN), and NEFA concentrations were determined. Cattle lost less weight at both 24 and 72 h after the LPS challenge with increasing DCP percentage ( < 0.01). Glucose ( = 0.12) and NEFA ( = 0.13) concentrations did not differ before the LPS challenge; however, there was a treatment effect for SUN, with elevated concentrations of SUN in CON cattle ( < 0.01). After the LPS challenge, DCP-fed cattle had reduced glucose, elevated NEFA, and reduced SUN concentrations ( ≤ 0.01). Results indicate that dietary DCP modulated metabolite concentrations in heifers following an endotoxin challenge and affected feedlot performance when incorporated in receiving diets in replacement of corn. Future studies will need to address strategies to increase DMI or explore levels of DCP less than 10% in the diet of newly received heifer calves.

This study was designed to characterize potential sexually dimorphic immunological responses following a lipopolysaccharide (LPS) challenge in beef cattle. Six female (heifers) and five male (bulls) Brahman calves (average age=253 ± 19.9 and 288 ± 47.9 days; average body weight=194 ± 11 kg and 247 ± 19 kg for heifers and bulls, respectively) were challenged with LPS (0.25 μg LPS/kg body weight). Following administration of LPS, all cattle displayed increased sickness behavior beginning at 0.5h, with heifers on average displaying less sickness behavior than bulls. A febrile response was observed in all animals following LPS administration, with a maximum response observed from 4 to 5.5h. The average rectal temperature response was greater in heifers than bulls. In all cattle there were elevated serum concentrations of cortisol from 0.5 to 8h, TNF-α from 1 to 2.5h, IL-6 from 2 to 8h, and IFN-γ from 2.5 to 7h after LPS challenge. Additionally, serum concentrations of TNF-α were greater in heifers than bulls from 1.5 to 2h after the LPS challenge. Concentrations of IFN-γ were also greater on average in bulls than heifers. Leukopenia occurred from 1 to 8h, with a decreased neutrophil to lymphocyte ratio for the first 5h among all calves. These data demonstrate the existence of a sexually dimorphic acute-phase response in pre-pubertal Brahman calves. Pain management and welfare are increasingly prevalent concerns within animal agriculture. Analgesics may alleviate pain and inflammation associated with castration of beef cattle. This study was conducted to elucidate the effects of surgical castration on the acute inflammatory response and immunomodulation and whether concurrent oral administration of meloxicam (1 mg/kg BW) would alter these responses. On d -1, crossbred bull calves ( = 30; initial BW = 227.4 ± 10.3 kg) were fitted with indwelling jugular catheters and rectal temperature (RT) recording devices, placed into individual stanchions, and randomly assigned to 1 of 3 treatments. Treatment application occurred at h 0 and consisted of 1) intact bull calves treated with sham castration (CON), 2) bulls surgically castrated without meloxicam administration (CAS), and 3) bulls surgically castrated with oral meloxicam (1 mg/kg BW) administration (MEL). Blood samples were collected at 0.5-h intervals from h -2 to 4, 1.0-h intervals from h 4 to 8, and 12-h intervals from h 12 to 72. Serum was analyzed for cortisol and haptoglobin (Hp) concentrations using ELISA. Whole blood was analyzed for complete blood counts at -2, 0, 2, 4, 6, 8, 12, 24, 36, 48, 60, and 72 h, and RT was recorded in 5-min intervals. Postcastration RT was greatest for MEL (39.04), intermediate for CAS (38.99), and least for CON (38.93°C; ≤ 0.01). Serum cortisol was increased ( < 0.001) for CAS (12.3) and MEL (11.3) compared with CON (6.7 ng/mL) during the postcastration period. At 0.5 and 1.5 h, cortisol concentration was greater in CAS and MEL than CON, whereas at 2 and 2.5 h, cortisol concentration was greatest for CAS, intermediate for MEL, and least for CON (treatment × time, < 0.001). Total white blood cell ( ≤ 0.04), lymphocyte ( ≤ 0.02), and monocyte ( ≤ 0.002) counts were greatest for CAS, intermediate for MEL, and least for CON. Administration of MEL reduced ( ≤ 0.002) eosinophil counts during the postcastration period when compared with CON and CAS. The change in serum Hp, relative to baseline values, was reduced for MEL at 36 ( < 0.01) and 60 h ( ≤ 0.03), and the overall Hp concentration was least for MEL ( < 0.001). Oral administration of meloxicam at the time of castration reduced the acute inflammatory response in castrates, as evidenced by a reduction in Hp and certain leukocyte concentrations; it also caused a delayed increase in RT. Further research is needed to determine if this reduced acute inflammatory response would equate to improved health and/or performance after castration.

Beef bull calves (n = 62) were assigned randomly, within sire breed, to 1 of 4 treatments at birth. Treatments were 1) surgical castration near birth, 2) surgical castration near birth with oral administration of meloxicam (1 mg/kg BW), 3) surgical castration at weaning (WNG), or 4) surgical castration at weaning with oral administration of meloxicam (1 mg/kg BW; WMX). A subset of calves (n = 7/treatment group) were selected randomly near birth for blood collection, behavioral analyses, and rectal temperature (RT) records for a 7-d postcastration period on d 0 (birth), 1, 3, and 7, and on d 214 (weaning), 214 + 6 h, 215, 217, 221, and 228. Calf standing and lying activity were monitored from the same subsets by recording x- and y-axis positions of an accelerometer attached to the right metatarsus for 7 d postcastration. Calf BW was recorded throughout the entire production cycle, and carcass data were collected at slaughter. For statistical analyses, bulls left intact at birth were considered a positive control (BUL) for observations that occurred before their treatment application at weaning; likewise, bulls castrated at birth were considered a negative control (STR) during postweaning observations. No difference (P > 0.88) occurred in ADG between treatments throughout the preweaning period (d 0 to 214); however, 56-d postweaning ADG was greatest ( P= 0.02) in STR, intermediate in WMX, and least in WNG. At weaning, haptoglobin (Hp) was greater (P ≤ 0.005) for WNG and WMX compared to STR on d 214+6 h, 215, and 217, and Hp was greater (P = 0.05) in WNG compared to WMX on d 217. Neutrophils increased (P < 0.001) and red blood cells decreased (P ≤ 0.03) for WNG and WMX on d 214+6 h and 217, respectively. Postweaning behavior observations indicated that STR calves spent the least proportion of time standing (P = 0.002) when compared to WNG and WMX. Furthermore, WMX calves exhibited a greater proportion of time spent standing (P = 0.03) compared to WNG. Grazing and finishing phase ADG and carcass measurements did not differ (P ≥ 0.24) across treatments. In this study, surgical castration at weaning, but not near birth, altered the acute phase response, behavior, and growth performance. Oral meloxicam reduced serum Hp and improved ADG briefly when administered to calves castrated at weaning. Oral administration of meloxicam may be efficacious for mitigating some of the stress and inflammation associated with castration of weaning-age bull calves.

This study characterized physiological responses to repeated transportation (TRANS) of gestating cows of differing temperaments. Cows were classified as Calm (C; = 10), Intermediate (I; = 28), or Temperamental (T; = 10). Based on artificial insemination date and pregnancy confirmation, cows were TRANS for 2 h on d 60 (TRANS1), 80 (TRANS2), 100 (TRANS3), 120 (TRANS4), and 140 (TRANS5) ± 5 d of gestation. Indwelling vaginal temperature (VT) monitoring devices were inserted 24 h before each TRANS with VT recorded from 2 h before TRANS and averaged into 5-min intervals through 30 min after TRANS. Serum samples were collected before loading and on unloading from the trailer to determine concentrations of cortisol, glucose, and nonesterified fatty acids (NEFA). Data were analyzed by repeated measures analysis in SAS. Serum cortisol concentrations were affected by temperament ( < 0.001), with T cows having the greater concentrations of cortisol before each TRANS event. All cows (100%) regardless of temperament exhibited elevations in cortisol following each TRANS event. Peak VT was greater ( < 0.001) at TRANS1 relative to all other TRANS events regardless of cow temperament. During TRANS, the T cows tended ( < 0.09) to have greater peak VT (39.86 ± 0.15°C) compared to C (39.41 ± 0.16°C) and I cows (39.55 ± 0.08°C). Area under the VT curve decreased ( = 0.002) from TRANS1 through TRANS5. Pre-TRANS serum glucose concentration at TRANS1 was greater ( < 0.03) for T (68.13 ± 4.31mg/dL) compared to I (53.42 ± 2.78 mg/dL) and C cows (52.76 ± 4.60 mg/dL). The C and I cows had greater changes in NEFA concentration between pre- and post-transport, and T cows showed the least change ( < 0.001). Cow VT and serum glucose concentration decreased in all temperaments ( < 0.01) with repeated TRANS; however, serum NEFA concentration post-TRANS did not vary ( > 0.10) with repeated TRANS events. Serum glucose concentrations were affected ( < 0.02) by a TRANS event by temperament interaction with T cows taking more TRANS events to decrease their change in glucose concentration compared to C and I cows. These results demonstrate that temperament influences physiological responses to stress in gestating Brahman cows. Although repeated transport in our study is confounded with day of gestation, seasonal changes, and learning from repeated handling and transport, repeated transport is a useful model of repeated stress in studying the effects of temperament.

Acknowledgment that modern livestock production systems impose stress on animals has been accepted by the scientific community and producers. As the economic burden has increased for livestock producers, expectations for animal performance have increased, thus placing more strain on the entire production system. Whether or not periodic exposure to stress within the production system jeopardizes the well-being of animals continues to be an area of debate largely because of the inability to accurately quantify the magnitude and severity of the stress response on other biological systems. Adding to the confusion is the fact that activation of the stress axis can be both beneficial and detrimental to the body depending on the duration of the stress response and the frequency at which an animal is exposed to stressful stimuli. Few would argue that continuous long-term stress inhibits livestock productivity and overall well-being. Less clear is whether or not occasional exposure to acute stress jeopardizes the productivity and well-being of livestock. To fully appreciate the complexity associated with activation of the stress axis and the overall biological impact on the body, one must delve deep into the scientific literature and examine the science in an unbiased manner. It is imperative to appreciate and understand that activation of the stress axis is an essential survival mechanism necessary to maintain homeostasis during biologically challenging times. Acute activation of the stress axis leads to repartitioning of energy to organs and tissues essential for coping with stress, redirection of blood flow from the peripheral to large muscle groups, decreased digestive function, and priming of the immune system to prepare for subsequent infections. Conversely, chronic activation of the stress axis disrupts digestive function, causes catabolism of muscle and adipose tissue, and suppresses overall immune function, thus making an animal more susceptible to disease. But what parameters are needed to distinguish periods of acute stress from those of chronic stress, and what biological markers are the best indicators of "stress" in an animal? Although there are a plethora of physiological responses and endocrine biomarkers that can be quantified, an integrative tool that has been readily embraced by scientists and producers as an effective and efficient indicator of the duration and magnitude of stress that an animal is experiencing has yet to be identified.

Activation of the innate immune system and acute phase response (APR) results in several responses that include fever, metabolic adaptations and changes in behavior. The APR can be modulated by many factors, with stress being the most common. An elevation of stress hormones for a short duration of time can be beneficial. However, elevation of stress hormones repeatedly or for an extended duration of time can be detrimental to the overall health and well-being of animals. The stress and APR responses can also be modulated by naturally-occurring variations, such as breed, gender, and temperament. These three natural variations modulate both of these responses, and can therefore modulate the ability of an animal to recover from a stressor or infection. Understanding that cattle have different immunological responses, based on naturally occurring variations such as these, may be the foundation of new studies on how to effectively manage cattle so that health is optimized and production is benefited., (© The Author(s) 2013 Reprints and permissions: sagepub.co.uk/journalsPermissions.nav.)

The influence of temperament on the alteration of metabolic parameters in response to a lipopolysaccharide(LPS) challenge was investigated. Brahman bulls were selected based on temperament score. Bulls (10 months; 211±5kg BW; n = 6, 8 and 7 for Calm, Intermediate and Temperamental groups, respectively) were fitted with indwelling jugular catheters to evaluate peripheral blood concentrations of glucose, blood urea nitrogen (BUN),non-esterified fatty acids (NEFA), insulin, epinephrine and cortisol before and after LPS administration (0.5 μg/kg BW LPS). Feed intake was also recorded. Intermediate bulls consumed more feed than the Temperamental bulls during the challenge (p = 0.046). Pre-LPS glucose (p = 0.401) and BUN (p = 0.222) did not differ among the temperament groups. However, pre-LPS insulin (p = 0.023) was lower, whereas pre-LPS NEFA (p < 0.001),cortisol (p < 0.001) and epinephrine (p < 0.001) were greater in Temperamental than in Calm and Intermediate bulls. Post-LPS glucose was increased in Calm and Intermediate bulls but not in Temperamental bulls(p < 0.001). Insulin concentrations post-LPS were greater in Calm than in Intermediate and Temperamental bulls (p < 0.001). Concentrations of NEFA post-LPS were greater in Temperamental than in Calm and Intermediate bulls (p < 0.001). Serum BUN concentration increased post-LPS, with values being greater in Calm and Intermediate than in Temperamental bulls (p = 0.012). Collectively, these data demonstrate that animal temperament is related to the metabolic responses of Brahman bulls following a provocative endotoxin challenge.Specifically, Temperamental bulls may preferentially utilize an alternate energy source (i.e. NEFA) to a greater degree than do bulls of Calm and Intermediate temperaments. The use of circulating NEFA from lipolysis may reduce the negative metabolic consequences of an immune response by allowing for a prompt answer to increasing energy demands required during immunological challenge, compared with the time required for glycogenolysis and gluconeogenesis.

The response of the immune and stress systems have been assessed in response to a lipopolysaccharide (LPS) challenge, yet the role of metabolism in mediating energy requirements during the acute phase response has not been sufficiently studied. This study tested heat-tolerant (Romosinuano [RO]) and heat-sensitive (Angus [ANG]) Bos taurus breeds at different ambient temperatures (Ta) to determine differential metabolic responses to LPS challenge. Twenty-one heifers (ANG: n = 11, 306 ± 26 kg BW; RO: n = 10, 313 ± 32 kg BW) were housed in stanchions in 4 temperature-controlled chambers. Initially, Ta in all 4 chambers was cycling at thermoneutrality (TN; 18.5°C-23.5°C) for a 1-wk adjustment period, followed by an increase in 2 chambers to cycling heat stress (HS; 24°C-38°C) for 2 wk. Five ANG and 5 RO heifers were housed at TN, whereas 6 ANG and 5 RO heifers were housed at HS. On day 19, heifers were fitted with jugular catheters. On day 20, heifers were challenged with LPS (0.5 μg/kg BW; 0 h), and blood samples were collected from -2 to 8 h and at 24 h relative to LPS challenge. Serum was analyzed for glucose, insulin, and NEFA concentrations. In addition, feed intake was measured 3 d before and on the day of the challenge. Feed intake decreased over time (P < 0.001) and was decreased in heifers housed at HS compared with heifers housed at TN (P = 0.013). Glucose concentrations before LPS challenge were greater in RO (P = 0.01) than in ANG heifers and greater in TN-housed heifers (P = 0.02) than in HS heifers. Glucose after LPS challenge initially increased before decreasing below baseline concentrations (P < 0.01) in all heifers. In addition, there was a breed by Ta interaction (P < 0.004), such that HS decreased glucose concentrations in ANG heifers compared with ANG heifers housed at TN (P < 0.001), whereas HS did not affect glucose concentrations after LPS challenge in RO heifers (P = 0.941). Nonesterified fatty acid concentrations before LPS challenge were not affected by breed (P = 0.37) or Ta (P = 0.60). Although NEFA concentration after LPS challenge was unaffected by Ta (P = 0.78), there tended to be a breed by Ta interaction (P = 0.07) such that, when housed at HS, RO heifers had greater serum NEFA concentrations after LPS challenge than ANG heifers (P = 0.009). Insulin concentration before LPS challenge was greater in RO heifers than in ANG heifers (P < 0.01). Insulin after LPS challenge increased (P < 0.01), with RO heifers producing a greater insulin response than ANG heifers (P < 0.01). These data suggest that HS decreases the metabolic response of heat-sensitive ANG heifers in response to LPS challenge, thus providing physiological evidence that may explain differences observed in the acute phase response between heat-sensitive ANG and heat-tolerant RO cattle breeds., (Published by Elsevier Inc.)

The difference in the acute phase response of a heat-tolerant and a heat-sensitive Bos taurus breed to a lipopolysaccharide (LPS) challenge when housed at different air temperatures (Ta) was studied. Angus (ANG; heat-sensitive; n = 11; 306 ± 26 kg BW) and Romosinuano (RO; heat-tolerant; n = 10; 313 ± 32 kg BW) heifers were transported from the USDA Agricultural Research Service SubTropical Agricultural Research Station in Florida to the Brody Environmental Chambers at the University of Missouri, Columbia. Heifers were housed in stanchions in 4 temperature-controlled environmental chambers. Initially, Ta in the 4 chambers was cycling at thermoneutrality (TN; 18.5°C-23.5°C) for a 1-wk adjustment period, followed by an increase in 2 of the 4 chambers to cycling heat stress (HS; 24°C-38°C) for 2 wk. On day 19, heifers were fitted with jugular catheters and rectal temperature (RT) recording devices. On day 20, heifers were challenged with LPS (0.5 μg/kg BW; 0 h), sickness behavior scores (SBSs) were recorded, and blood samples were collected at 0.5-h intervals from -2 to 8 h and again at 24 h relative to LPS challenge at 0 h. Serum was isolated and stored at -80°C until analyzed for cortisol and cytokine concentrations. A breed by Ta interaction (P < 0.001) was observed for RT such that the post-LPS average RT in RO heifers housed at TN was lower than the RT of all other treatment groups (P < 0.001), whereas ANG heifers housed at HS had greater post-LPS average RT than all other treatment groups (P < 0.001). In response to LPS, HS increased SBS after LPS in RO heifers compared to RO heifers housed at TN (P < 0.001), whereas HS decreased SBS after LPS in ANG heifers compared to ANG heifers housed at TN (P = 0.014). The cortisol response to LPS was greater in TN than in HS heifers (P < 0.01) and was also greater in RO than in ANG heifers (P = 0.03). A breed by Ta interaction (P < 0.01) was observed for tumor necrosis factor-α (TNF-α) concentration such that HS increased post-LPS serum concentrations of TNF-α in ANG heifers compared to ANG heifers housed at TN (P = 0.041), whereas HS decreased post-LPS concentrations of TNF-α in RO heifers compared to RO heifers housed at TN (P = 0.008). A tendency (P < 0.06) was observed for a breed by Ta interaction for IL-6 concentrations such that RO heifers had greater post-LPS concentrations of IL-6 than ANG heifers when housed at HS (P = 0.020). A breed by Ta interaction was observed for interferon-γ (IFN-γ; P < 0.01) concentrations such that HS decreased post-LPS concentrations of IFN-γ in ANG heifers compared to ANG heifers housed at TN (P < 0.001), and HS increased post-LPS concentrations of IFN-γ in RO heifers compared to RO heifers housed at TN (P = 0.017). These data indicate differences in the acute phase response between the heat-tolerant RO and heat-sensitive ANG heifers under different Ta which may aid in elucidating differences in productivity, disease resistance, and longevity among cattle breeds., (Published by Elsevier Inc.)

A study was conducted to determine the effect of feeding yeast cell wall (YCW) products on the physiological and acute phase responses of crossbred, newly-received feedlot heifers to an endotoxin challenge. Heifers (n = 24; 219 ± 2.4 kg) were separated into treatment groups receiving either a control diet (n = 8), YCW-A (2.5 g/heifer/d; n = 8) or YCW-C (2.5 g/heifer/d; n = 8) and were fed for 52 d. On d 37 heifers were challenged i.v. with LPS (0.5 µg/kg body mass) and blood samples were collected from -2 h to 8 h and again at 24 h relative to LPS challenge. There was an increase in vaginal temperature in all heifers post-LPS, with YCW-C maintaining a lower vaginal temperature post-LPS than control and YCW-A heifers. Sickness behavior scores increased post-LPS in all heifers, but were not affected by treatment. Cortisol concentrations were greatest in control heifers post-LPS compared with YCW-A or YCW-C heifers. Concentrations of IFN-γ and TNF-α increased post-LPS, but were not affected by treatment. Serum IL-6 concentrations increased post-LPS and were greater in control heifers than YCW-A and YCW-C heifers. These data indicate that YCW supplementation can decrease the physiological and acute phase responses of newly-received heifers following an endotoxin challenge.
