Less is known about the relationship between conduct disorder (CD), callous-unemotional (CU) traits, and positive and negative parenting in youth compared to early childhood. We combined traditional univariate analyses with a novel machine learning classifier (Angle-based Generalized Matrix Learning Vector Quantization) to classify youth (N = 756; 9-18 years) into typically developing (TD) or CD groups with or without elevated CU traits (CD/HCU, CD/LCU, respectively) using youth- and parent-reports of parenting behavior. At the group level, both CD/HCU and CD/LCU were associated with high negative and low positive parenting relative to TD. However, only positive parenting differed between the CD/HCU and CD/LCU groups. In classification analyses, performance was best when distinguishing CD/HCU from TD groups and poorest when distinguishing CD/HCU from CD/LCU groups. Positive and negative parenting were both relevant when distinguishing CD/HCU from TD, negative parenting was most relevant when distinguishing between CD/LCU and TD, and positive parenting was most relevant when distinguishing CD/HCU from CD/LCU groups. These findings suggest that while positive parenting distinguishes between CD/HCU and CD/LCU, negative parenting is associated with both CD subtypes. These results highlight the importance of considering multiple parenting behaviors in CD with varying levels of CU traits in late childhood/adolescence. Copyright © Cambridge University Press 2020.

To understand trends in individual responses to medication, one can take a purely data-driven machine learning approach, or alternatively apply pharmacokinetics combined with mixed-effects statistical modelling. To take advantage of the predictive power of machine learning and the explanatory power of pharmacokinetics, we propose a latent variable mixture model for learning clusters of pharmacokinetic models demonstrated on a clinical data set investigating 11 beta-hydroxysteroid dehydrogenase enzymes (11 beta -HSD) activity in healthy adults. The proposed strategy automatically constructs different population models that are not based on prior knowledge or experimental design, but result naturally as mixture component models of the global latent variable mixture model. We study the parameter of the underlying multi-compartment ordinary differential equation model via identifiability analysis on the observable measurements, which reveals the model is structurally locally identifiable. Further approximation with a perturbation technique enables efficient training of the proposed probabilistic latent variable mixture clustering technique using Estimation Maximization. The training on the clinical data results in 4 clusters reflecting the prednisone conversion rate over a period of 4 h based on venous blood samples taken at 20-min intervals. The learned clusters differ in prednisone absorption as well as prednisone/prednisolone conversion. In the discussion section we include a detailed investigation of the relationship of the pharmacokinetic parameters of the trained cluster models for possible or plausible physiological explanation and correlations analysis using additional phenotypic participant measurements. Astroinformatics is a new discipline at the cross-road of astronomy, advanced statistics and computer science. With next generation sky surveys, space missions and modern instrumentation astronomy will enter the Petascale regime raising the demand for advanced computer science techniques with hard- and software solutions for data management, analysis, efficient automation and knowledge discovery. This tutorial reviews important developments in astroinformatics over the past years and discusses some relevant research questions and concrete problems. The contribution ends with a short review of the special session papers in these proceedings, as well as perspectives and challenges for the near future.

With electronic data increasing dramatically in almost all areas of research, a plethora of new techniques for automatic dimensionality reduction and data visualization has become available in recent years. These offer an interface which allows humans to rapidly scan through large volumes of data. With data sets becoming larger and larger, however, the standard methods can no longer be applied directly. Random subsampling or prior clustering still being one of the most popular solutions in this case, we discuss a principled alternative and formalize the approaches under a general perspectives of dimensionality reduction as cost optimization. We have a first look at the question whether these techniques can be accompanied by theoretical guarantees.

Recently, several groups have developed computational methods for identifying cell populations in multidimensional FCM data. The Flow Cytometry: Critical Assessment of Population Identification Methods (FlowCAP) challenges were established to compare the performance of these methods on two tasks: (i) mammalian cell population identification, to determine whether automated algorithms can reproduce expert manual gating and (ii) sample classification, to determine whether analysis pipelines can identify characteristics that correlate with external variables (such as clinical outcome). This analysis presents the results of the first FlowCAP challenges. Several methods performed well as compared to manual gating or external variables using statistical performance measures, which suggests that automated methods have reached a sufficient level of maturity and accuracy for reliable use in FCM data analysis.

The amount of electronic information as well as the size and dimensionality of data sets have increased tremendously. Consequently, dimension reduction and visualization techniques have become increasingly popular in recent years. Dimension reduction is typically connected with loss of information. In supervised classification problems, class labels (:an I)e used to minimize the loss of information concerning the specific task. The aim is to preserve and potentially enhance tire discrimination of classes in lower dimensions. Here we propose a prototype-based local relevance learning scheme, that results in an efficient nonlinear discriminative dimension reduction of labeled data sets. The method is introduced and discussed in terms of artificial and real world data sets.
