In this study we present data collected between August 2017 and December 2018 from submarine sediment trap M1 located in the eastern tropical North Atlantic and determine lithogenic and biogenic fluxes, and grain-size distributions of aerosol dust. An unprecedented high sampling resolution of four days in combination with satellite imagery allowed the identification of thirteen major dust events of two types within the 468-day series. Seven dust events were classified as high-flux events identified by a deposition of >=25 mg m-2 d-1. The average dust deposition rate increased by 461.3% during these types of events. The remaining six events were characterized by a high composition of giant particles (>7.3 vol%). Seasonal variations of dust flux were recorded, with highest fluxes observed in spring at an average deposition rate of 14.8 mg m-2 d-1, and lowest fluxes in fall with an average rate of 8.6 mg m-2 d-1. The estimated total dust flux in the tropical North Atlantic was 4040.02 mg m-2 y-1. We suggest that most of the summer dust was likely transported over the sampling site at high altitudes while winter transport occurred closer to the sea surface, resulting in generally higher background fluxes. Grain-size distributions exhibited seasonal variations with increased occurrences of giant particles (>62.5 μm) in fall 2017 and spring, and less occurrences in winter and fall 2018. Grain-size sorting was high in spring and most variable in summer. Precipitation did not affect depositional fluxes or grain-size distributions of the aeolian dust significantly. Organic matter was deposited continuously at the sampling site with a deposition rate ranging from 14.4–862.25 mg m-2 d-1. Organic matter fluxes increased by 199.5% during high-flux dust events but decreased by 13.6% during anomalous grain-size events. Spring experienced the highest number of dust events (n=5). This unprecedented high-resolution dataset of dust deposition in marine sediment-traps enabled the distinction of dust events and their relation to organic matter flux. This suggests the possibility of a dust fertilization or mineral ballasting effect.

This article describes the author's experience designing and implementing an inquiry-based learning (IBL) pedagogical approach to an upper-division undergraduate cryptology course. The author shares the course goals and how the IBL style supports their achievement. The article concludes with sample activities -- in-class exercises that touch on one-time pad, the Birthday Problem, the Meet-in-the-Middle attack, and Diffie-Hellman key exchange protocol, and a programing assignment on the Vigenère cipher.

Bats, as members of the order Chiroptera, are vital to ecosystems and serve as reservoirs for numerous microorganisms, some of which can cause zoonotic diseases. Human interactions with bats are increasing due to habitat alterations, making it essential to understand their microbiota, particularly potential pathogens. This study aimed to evaluate the excretion of zoonotic bacteria and protozoa in insectivorous bats from four caves in the provinces of Ragusa, Catania, and Syracuse (Sicily, Southern Italy) using molecular biology tests for zoonotic agents, including Bartonella henselae, Borrelia, Coxiella burnetii, Leptospira, Chlamydia, Rickettsia, Anaplasma, and Piroplasmids. From December 2020 to April 2023, urine, fecal swabs, ocular conjunctival swabs, and oropharyngeal swabs were collected from 149 bats of six species, along with guano samples from the caves. Bartonella henselae DNA was detected in 3 of the 149 tested bats, one ocular conjunctival swab and two oropharyngeal swabs. Chlamydia spp. DNA was detected in a sample of guano, in feces, ocular conjunctival and oropharyngeal swabs of a bat, and in four urine samples. Piroplasmid DNA was detected in 10 of 149 fecal swabs and in 5 of 16 bat ectoparasites. No samples were positive for Leptospira spp., Borrelia spp., Coxiella burnetii, Rickettsia spp., or Anaplasma spp. These findings underscore the importance of multiple sample types in assessing bats as reservoirs for zoonotic pathogens, particularly highlighting their role in transmitting pathogens through various body habitats, including saliva, urine, and ocular excretions. This study highlights the relevance of monitoring bat populations and studying their microbiota to enhance protections for both human and animal health.

Vector-borne diseases represent a serious threat to human and animal health, especially where environmental conditions favor pathogen-carrying vectors. Dogs serve as natural hosts for two tick-borne pathogens: Ehrlichia canis, which causes canine monocytic ehrlichiosis, and spotted fever group (SFG) Rickettsia spp., a zoonotic threat in the Mediterranean region. Rhipicephalus sanguineus is the primary vector for these pathogens. Shelter dogs, due to increased exposure to ticks and confined living conditions, facilitate the spread of vector-borne pathogens, raising the risk of zoonotic transmission. This study conducted a serological survey of 1287 dogs from two shelters, assessing exposure to Rickettsia spp. and E. canis and examining the influence of demographic and environmental factors. Seroprevalence rates were 41.8% for Rickettsia spp. and 24.5% for E. canis, with 14% of dogs positive for both pathogens. No significant association was found with sex or breed. A higher seroprevalence was observed in dogs older than 12 months and in those from the shelter on the Mediterranean coast compared to those from the Tyrrhenian coast, likely due to climatic differences. The study highlights the role of climate in disease spread and the need for public health interventions, supporting One Health initiatives to prevent zoonotic disease transmission.

This study focused on ticks found in a nature reserve in southern Italy, highly frequented for recreational activities and inhabited by wild boars. Using molecular techniques, 214 ticks, including questing ticks and those removed from wild boars, were examined for tick-borne pathogens (TBPs), with a focus on zoonotic pathogens. Six tick species were identified: Hyalomma lusitanicum, Rhipicephalus pusillus, Rh. sanguineus s.l., Rh. bursa, Rh. turanicus, and Dermacentor marginatus, several of which are known vectors of zoonotic pathogens. Overall, 14% of ticks were positive for TBPs, mainly bacteria of Rickettsia genus. Molecular analyses detected Rickettsia slovaca, R. massiliae, Candidatus R. shennongii, R. conorii, R. felis, and R. barbariae. Additionally, single detections of Coxiella burnetii and Theileria annulata were recorded. Phylogenetic analyses were conducted on Rickettsia sequences. The range of ticks and TBPs present in this area highlights potential public health concerns. New associations between tick species and TBPs were documented, though vector roles need further investigation. The study highlights the importance of monitoring tick populations in both rural and urban environments to protect public health and prevent tick-borne disease spreading.

Q fever is a widespread zoonotic disease caused by the obligate intracellular bacterium Coxiella burnetii, primarily transmitted through the inhalation of contaminated aerosols. This study aimed to detect C. burnetii in two Sicilian sheep flocks, with no better defined reproductive disorders reported by the farmers. Blood, individual and bulk milk, ticks, and conjunctival swabs were collected from both flocks (A and B). Real-time and traditional PCRs were carried out to detect C. burnetii DNA and anti-C. burnetii antibodies were searched using an ELISA. In terms of Farm A, C. burnetii DNA was detected in 7.1% of blood samples, 20% of individual milk samples, bulk milk, 66.6% of conjunctival swabs, and in all the examined tick pools. Anti-C. burnetii antibodies were found in 77.0% of sera, 92.5% of individual milk samples, and bulk milk. In terms of Farm B, C. burnetii DNA was detected in 3.8% of blood samples, 39.4% of individual milk samples, bulk milk, 100% of conjunctival swabs, and in all tick pools; anti-C. burnetii antibodies were present in 53.6% of sera, 73.2% of milk samples, and in bulk milk. Our results highlight the high diffusion of C. burnetii in the two outbreaks, with widespread pathogen circulation, significant shedding in dairy products, and high environmental contamination, highlighting the need for enhanced surveillance and control measures in dairy sheep farms.

Foodborne diseases result from the consumption of foods contaminated with pathogens or their toxins and represent a serious public health problem worldwide. This study aimed to assess the presence of Rotavirus (RoV), Adenovirus (AdV), Norovirus (NoV), Hepatitis A and Hepatitis E viruses (HAV and HEV, respectively), Toxoplasma gondii, Coxiella burnetii and Leptospira spp. across various food matrices in Sicily. The analysis concerned 504 samples, including mussels, farmed meat, game meat, vegetables and bulk milk. Following appropriate pre-treatment, acid nucleic extraction was carried out and amplification of pathogen nucleic acids was carried out by molecular methods. The mussels tested positive for NoVs (3/51, 5.9%) and farm meat resulted positive for T. gondii (1/34, 2.9%). The game offal samples tested positive for HEV, which was detected in 17 out of 222 samples (7.7%), and T. gondii (18/318, 5.7%) and Leptospira spp. (2/318, 0.6%). The milk samples tested positive for C. burnetii (15/85, 17.6%), T. gondii (2/85, 2.4%) and Leptospira spp. (1/85, 1.2%). This study highlights the variability in the risk of contamination of different food matrices, confirming the importance of vigilance in the consumption of potentially contaminated food products.

Abstract The gut microbiota exerts a variety of positive effects on the intestinal homeostasis, including the production of beneficial molecules, control of the epithelial barrier integrity and the regulation of the balance between host’s cell death and proliferation. The interactions between commensal bacteria and intestinal cells are still under-investigated and is then of paramount importance to address such interactions at the molecular and cellular levels. We report an in vitro analysis of the effects of molecules secreted by Lactobacillus gasseri SF1183 on HCT116 cells, selected as a model of intestinal epithelial cells. SF1183 is a L. gasseri strain isolated from an ileal biopsy of a human healthy volunteer, able to prevent colitis symptoms in vivo. Expanding previous findings, we show that bioactive molecules secreted by SF1183 reduce the proliferation of HCT116 cells in a reversible manner determining a variation in cell cycle markers (p21WAF, p53, cyclin D1) and resulting in the protection of HCT116 cells from TNF-alfa induced apoptosis, an effect potentially relevant for the protection of the epithelial barrier integrity and reconstitution of tissue homeostasis. Consistently, SF1183 secreted molecules increase the recruitment of occludin, a major component of TJ, at the cell–cell contacts, suggesting a reinforcement of the barrier function.

Simple Summary: Bats play a vital role in our ecosystems and can carry a variety of microorganisms, some of which may cause diseases in humans. As human activities alter their natural habitats, interactions between bats and people are increasing, making it crucial to understand the pathogens they harbor. This study focused on identifying potentially harmful bacteria and piroplasmids in bats from four caves in southern Italy. Over a span of more than two years, we collected samples from 149 bats representing different species. Our findings revealed the presence of Bartonella henselae, Chlamydia spp., and Piroplasmids in these bats and their ectoparasites, indicating that they could serve as sources of infection for both animals and humans. Bats were negative for the other investigated pathogens, including Coxiella burnetii, Leptospira spp., Rickettsia spp., Anaplasma spp., and Borrelia spp. Understanding the specific pathogens present in bats and their modes of transmission is essential for public health. Background : The microbiome regulates the respiratory epithelium's immunomodulatory functions. To explore how the microbiome's biodiversity affects microbe-epithelial interactions, we screened 58 phylogenetically diverse microbes for their transcriptomic effect on human primary bronchial air–liquid interface (ALI) cell cultures. Results: We found distinct species- and strain-level differences in host innate immunity and epithelial barrier response. Strikingly, we found that host interferon, an antiviral response, was one of the most variable host processes. This variability was not driven by microbial phylogenetic diversity, bioburden, nor by the microbe's ability to stimulate other innate immunity pathways. Conclusions: Microbial colonization differentially stimulates host gene expression with variations observed across phylogenetically diverse microbes and across different strains of the same species. Our study provides a foundation for understanding how the respiratory microbiome's biodiversity affects epithelial, and particularly antiviral, innate immunity. [ABSTRACT FROM AUTHOR]

Background: The Canadian Nosocomial Infection Surveillance Program (CNISP) observed increased mortality among neonatal intensive care unit (NICU) patients with central-line–associated bloodstream infection (CLABSI) starting in 2017. In this study, we compared NICU patients with CLABSIs before and after 2017, and quantified the impact of epidemiological factors on 30-day survival. Methods: We included 1,276 NICU patients from 8–16 participating CNISP hospitals from the pre-2017 period (2009–2016) and the post-2017 period (2017–2022) using standardized definitions and questionnaires. We used Cox regression modeling to assess the impact of age at date of positive culture, sex, birthweight, CLABSI microorganism, region of the country, and surveillance period (before 2017 vs after 2017) on time to 30-day all-cause mortality from date of positive culture. Gestational age was not available for this analysis. We reported model outputs as hazard ratios with 95% CIs. Results: In total, 769 (60%) NICU CLABSIs were reported in the pre-2017 period and 507 (40%) in the post-2017 period. The 30-day all-cause mortality rate was 8% (n = 100 of 1,276) overall, and significantly higher after 2017 (12%, n = 61 of 507) than before 2017 (5%, n = 39 of 769) (P < .001).

BackgroundSince the outbreak, in 2019, of COVID-19, the world has experienced marked changes in daily habits, partly reflecting the exceptional social restrictions and health measures adopted to contain the disease. All these measures significantly affected not only peoples’s daily lives and psychological well-being but also the possibility for the healthcare system to function properly. In this setting, brain tumour patients were at risk due to their higher physical and mental fragility and their need for regular care. The aim of the present study was to assess, using a self-reported online questionnaire, the patients’s perceptions regarding their disease experience.Materials and methodsWe developed an online anonymous self-report survey to assess patients’s disease experience during the pandemic. We investigated the impact of the COVID-19 pandemic on patients’s cancer care schedules, their psychological distress and emotions felt during the pandemic, their levels of worry about COVID-19, and their oncological conditions.Results107 patients answered our survey, most of them suffering from a glioma. Less than one-third of the sample had their appointments cancelled, delayed or converted into online visits due to the pandemic. Of the patients who answered the survey, 95% declared they were satisfied with their Institute’s oncological management. The feelings reported most often were peacefulness or anxiety/worry; the majority of the sample reported high levels of loneliness, which tended to increase with age, whilst the psychological distress was correlated with age and with having a recurrence of the disease. Half of the sample declared severe worry about their oncological condition, in particular subjects with a recurrence or who were receiving adjuvant therapies. Patients with recurrence tended to worry more about the possibility of contracting COVID-19, and its effects.ConclusionOur findings illustrate how fragile and in need of care patients with a brain tumour may be, especially those with more severe clinical conditions. These data may help boost healthcare professionals’s knowledge about brain tumour patients’s needs and fears, so as to be able to offer them a better hospital experience and improve their clinical management, while possibly also reducing the psychological burden on patients and their families.

Abstract Background West Nile Disease (WND) is a zoonotic mosquito‐borne infection involving viral pathogens, human and animal hosts, vectors and environment. Cooperation among medical, veterinary and entomological fields has been promoted by the Italian Public Health Authorities, and an integrated West Nile Virus (WNV) Surveillance Plan has been in force in Italy since 2016 to prevent the transmission risk of WND to humans through an early detection of viral circulation by animal and entomological surveillance. This managing model is unique in Europe. Objectives This survey aimed at presenting the ‘One Health’ approach applied in 2016 to the first autochthonous human case of West Nile Neuroinvasive Disease (WNND) in Sicily (Southern Italy). Methods Serological (anti‐WNV IgM and IgG ELISA, anti‐WNV neutralizing antibodies) and molecular tests were conducted on blood, liquor and urine of a 38‐year‐old man with encephalitis and meningitis. Overall, 2704 adult culicides from 160 mosquito catches were morphologically identified. Female mosquitoes were analysed in pools for WNV RNA detection. Serological (anti‐WNV IgM and IgG ELISA) and molecular analyses for WNV were carried out in 11 horses, 271 chickens and two dogs sampled in farms around the man's residence. Results and conclusions WNND was confirmed by serological analysis on patient's liquor and serum. Collected mosquito species included Culex pipiens (93.56%, CI95% 92.64%–94.49%), Aedes albopictus (5.25%, CI95% 4.41%–6.09%), Culex hortensis (0.59%, CI95% 0.30%–0.88%), Culiseta longiareolata (0.55%, CI95% 0.27%–0.83%) and Anopheles maculipennis s.l. (0.04%, CI95% –0.04% to 0.11%). Mosquito pools were negative for WNV RNA. Two dogs (100%) and two horses (18.18%, CI95% –4.61 to 40.97%) resulted positive for anti‐WNV specific antibodies. The ‘One Health’ approach allowed to report the first human neuroinvasive WND in Sicily and to confirm the local circulation of WNV in animals of the same area where the clinical case occurred, defining the autochthonous origin of the infection.

Abstract Background Cardiovascular complications of severe acute respiratory distress syndrome coronavirus 2 (SARS-CoV2) are known to be associated with poor outcome. A small number of case series and reports have described cases of myocarditis and ischaemic events, however, knowledge on the aetiology of acute cardiac failure in SARS-CoV2 remains limited. We describe the occurrence and risk stratification imaging correlates of ‘takotsubo’ stress cardiomyopathy presenting in a patient with Coronavirus Disease 2019 (COVID-19) in the intensive care unit. Case summary An intubated 53-year old patient with COVID19 suffered acute haemodynamic collapse in the intensive care unit, and was thus investigated with transthoracic echocardiography (TTE), 12-lead electrocardiograms (ECG) and serial troponins and blood tests, and eventually coronary angiography due to clinical suspicion of ischaemic aetiology. Echocardiography revealed a reduced ejection fraction, with evident extensive apical akinesia spanning multiple coronary territories. Troponins and NT-proBNP were elevated, and ECG revealed ST elevation: coronary angiography was thus performed. This revealed no significant coronary stenosis. Repeat echocardiography performed within the following week revealed a substantial recovery of ejection fraction and wall motion abnormalities. Despite requirement of a prolonged ICU stay, the patient now remains clinically stable, and is on spontaneous breathing. Conclusion This case report presents a case of takotsubo stress cardiomyopathy occurring in a critically unwell patient with COVID19 in the intensive care setting. Stress cardiomyopathy may be an acute cardiovascular complication of COVID-19 infection. In the COVID19 critical care setting, urgent bedside echocardiography is an important tool for initial clinical assessment of patients suffering haemodynamic compromise.

In terms of Farm A, C. burnetii DNA was detected in 7.1% of blood samples, 20% of individual milk samples, bulk milk, 66.6% of conjunctival swabs, and in all the examined tick pools. Anti-C. burnetii antibodies were found in 77.0% of sera, 92.5% of individual milk samples, and bulk milk. In terms of Farm B, C. burnetii DNA was detected in 3.8% of blood samples, 39.4% of individual milk samples, bulk milk, 100% of conjunctival swabs, and in all tick pools; anti-C. burnetii antibodies were present in 53.6% of sera, 73.2% of milk samples, and in bulk milk. Our results highlight the high diffusion of C. burnetii in the two outbreaks, with widespread pathogen circulation, significant shedding in dairy products, and high environmental contamination, highlighting the need for enhanced surveillance and control measures in dairy sheep farms. Foodborne diseases result from the consumption of foods contaminated with pathogens or their toxins and represent a serious public health problem worldwide. This study aimed to assess the presence of Rotavirus (RoV), Adenovirus (AdV), Norovirus (NoV), Hepatitis A and Hepatitis E viruses (HAV and HEV, respectively), Toxoplasma gondii, Coxiella burnetii and Leptospira spp. across various food matrices in Sicily. The analysis concerned 504 samples, including mussels, farmed meat, game meat, vegetables and bulk milk. Following appropriate pre-treatment, acid nucleic extraction was carried out and amplification of pathogen nucleic acids was carried out by molecular methods. The mussels tested positive for NoVs (3/51, 5.9%) and farm meat resulted positive for T. gondii (1/34, 2.9%). The game offal samples tested positive for HEV, which was detected in 17 out of 222 samples (7.7%), and T. gondii (18/318, 5.7%) and Leptospira spp. (2/318, 0.6%). The milk samples tested positive for C. burnetii (15/85, 17.6%), T. gondii (2/85, 2.4%) and Leptospira spp. (1/85, 1.2%). This study highlights the variability in the risk of contamination of different food matrices, confirming the importance of vigilance in the consumption of potentially contaminated food products. Simple Summary: Cardiovascular diseases are among the most frequent, although rare, long-term sequalae in adolescents and young adult survivors of breast cancer. However, no dedicated tool exists to help clinicians with planning personalized follow-up strategies for these patients. To make up for this lack, in this work, we developed a Bayesian network, an artificial intelligence model, to predict the 5-year risk for cardiovascular diseases in these patients, leveraging real-world data from two different cohorts. The model showed a very good ability to identify patients at risk and select those that should be prioritized because they are at higher risk, making it useful for guiding clinicians in everyday practice. Finally, the methodological approach proposed in this work is particularly interesting for all researchers who aim at developing causally interpretable tools, also dealing with real-world data and their biases. Background: In the last decades, the increasing number of adolescent and young adult (AYA) survivors of breast cancer (BC) has highlighted the cardiotoxic role of cancer therapies, making cardiovascular diseases (CVDs) among the most frequent, although rare, long-term sequalae. Leveraging innovative artificial intelligence (AI) tools and real-world data (RWD), we aimed to develop a causally interpretable model to identify young BC survivors at risk of developing CVDs. Methods: We designed and trained a Bayesian network (BN), an AI model, making use of expert knowledge and data from population-based (1036 patients) and clinical (339 patient) cohorts of female AYA (i.e., aged 18 to 39 years) 1-year survivors of BC, diagnosed in 2009–2019. The performance achieved by the BN model was validated against standard classification metrics, and two clinical applications were proposed. Results: The model showed a very good classification performance and a clear causal semantic. According to the predictions made by the model, focusing on the 25% of AYA BC survivors at higher risk of developing CVDs, we could identify 81% of the patients who would actually develop it. Moreover, a desktop-based app was implemented to calculate the individual patient's risk. Conclusions: In this study, we developed the first causal model for predicting the CVD risk in AYA survivors of BC, also proposing an innovative AI approach that could be useful for all researchers dealing with RWD. Abstract Background Tick-borne diseases are common throughout Europe. Ticks transmit pathogens to the host while feeding and together with mosquitoes, they are major vectors of infectious agents worldwide. In recent years, there has been a marked increase in the incidence of tick-bite events and tick-borne disease in northwest Italy, but information on the prevalence of tick-borne pathogens in ticks removed from humans remains scarce. To fill this gap, we report here the prevalence of tick bites and tick-borne pathogens documented for humans in Piedmont, northwest Italy, in the 3-year period 2017–2019. Methods Ticks attached to humans during 2017–2019 were collected from residents of urban and rural area by physicians and veterinarians working with local veterinary agencies. All ticks (n = 1290) were morphologically identified to the species level. A subset of ticks removed from children (age 0–18 years) and the elderly (> 70 years), both age groups considered to be at-risk populations, was screened by biomolecular analysis to detect pathogens (e.g. Rickettsia spp., Borrelia spp., Anaplasma spp.). Pathogen identity was confirmed by Sanger sequencing. Results Ticks were taxonomically assigned to ten species of six genera (Amblyomma, Dermacentor, Haemaphysalis, Hyalomma, Ixodes and Rhipicephalus). Most belonged to the genus Ixodes: 1009 ticks (78.22%) were classified as Ixodes ricinus. A subset of 500 ticks collected from the two at-risk populations were subjected to PCR assay to determine the presence of Rickettsia spp., Borrelia spp., and Anaplasma spp. The overall prevalence of infection was 22.8% (n = 114; 95% confidence interval [CI]: 19.19–26.73%), meaning that at least one pathogen was detected: Rickettsia spp. (prevalence 15%, n = 76; 95% CI 12.17–18.65%); Borrelia spp. (prevalence 6.4%, n = 32; 95% CI 4.42–8.92%); and Anaplasma spp. (prevalence 1.2%, n = 6; 95% CI 0.44–2.6%). Conclusions Our data underline the importance of surveillance in the epidemiology of tick-borne diseases and the implementation of strategies to control tick infestation and associated pathogens.
