Abstract Plants possess the ability to induce programmed cell death (PCD) in response to abiotic and biotic stresses; nevertheless, the evidence on PCD initiation during pear scald development and the involvement of the scald trigger 6-methyl-5-hepten-2-one (MHO) in this process is rudimentary. Pyrus bretschneideri Rehd. cv. ‘Dangshansuli’ pear was used to validate such hypothesis. The results showed that superficial scald occurred after 120-d chilling exposure, which accompanied by typical PCD-associated morphological alterations, such as plasmolysis, cell shrinkage, cytosolic and nuclear condensation, vacuolar collapse, tonoplast disruption, subcellular organelle swelling, and DNA fragmentation. These symptoms were aggravated after MHO fumigation but alleviated by diphenylamine (DPA) dipping. Through transcriptome assay, 24 out of 146 PCD-related genes, which were transcribed during cold storage, were identified as the key candidate members responsible for these cellular biological alternations upon scald development. Among these, PbrCNGC1, PbrGnai1, PbrACD6, and PbrSOBIR1 were implicated in the MHO signaling pathway. Additionally, PbrWRKY2, 34 and 39 could bind to the W-box element in the promoter of PbrGnai1 or PbrSOBIR1 and activate their transcription, as confirmed by dual-luciferase, yeast one-hybrid, and transient overexpression assays. Hence, our study confirms the PCD initiation during scald development and explores the critical role of MHO in this process.

Abstract Sepsis is a critical global health concern linked to high mortality rates, often due to acute lung injury (ALI)/acute respiratory distress syndrome (ARDS). While the gut-lung axis involvement in ALI is recognized, direct migration of gut immune cells to the lung remains unclear. Our study reveals sepsis-induced migration of γδ T17 cells from the small intestine to the lung, triggering an IL-17A-dominated inflammatory response in mice. Wnt signaling activation in alveolar macrophages drives CCL1 upregulation, facilitating γδ T17 cell migration. CD44+ Ly6C– IL-7Rhigh CD8low cells are the primary migratory subtype exacerbating ALI. Esketamine attenuates ALI by inhibiting pulmonary Wnt/β-catenin signaling-mediated migration. This work underscores the pivotal role of direct gut-to-lung memory γδ T17 cell migration in septic ALI and clarifies the importance of localized IL-17A elevation in the lung.

Bismuth sodium titanate (BNT) ceramics exhibit outstanding strain responses but are unfavorable for application in high-sensitivity displacement actuators due to the large negative strain resulting from irreversible changes in their phase transition and domain structure. Here, (1−x)Bi0.50Na0.41K0.09TiO3–xNaNbO3 (BNKT−xNN) solid solutions were prepared to improve the strain properties through the strategy of modulating the phase boundary and domain structures. The introduction of sodium niobate could effectively regulate the relative content of the tetragonal (P4bm) and rhombohedral (R3c) phases in the phase boundary region. The ferroelectric-to-relaxor phase transition (TF−R) was reduced, and the ergodic relaxor (ER) state was nurtured at room temperature. Excellent zero-negative strain properties of S = 0.41% and d33* = 742 pm/V were achieved from the reversible transition between the ER and ferroelectric states under an applied electric field (x = 0.04). Additionally, understanding the domain states via piezoelectric force microscopy (PFM) and first-order reversal curve (FORC) revealed that the superior strain responses originated from the reversible inter-transformation of substable macrodomains and polar nanoregions (PNRs) in the phase boundary. This study provides new insight into the interplay between the evolution of phase boundaries and domain structures and the strain properties of BNT-based ceramics.

Nitrogen deposition and drought significantly influence plant growth and soil physicochemical properties. This study investigates the effects of nitrogen deposition and water stress on the growth and physiological responses of Quercus dentata, and how these factors interact to influence the overall productivity. Two-year-old potted seedlings were selected to simulate nitrogen deposition and water stress. Nitrogen was applied at rates of 0 kg·ha-1·year-1 (N0) and 150 kg·ha-1·year-1 (N150). The levels of water stress corresponded to 80% (W80), 50% (W50), and 20% (W20) of soil saturation moisture content. High nitrogen (N150) significantly increased stem elongation and stem diameter by enhancing photosynthetic parameters, including Pn (W80) and Gs (W50), and maintained higher water use efficiency. Under drought conditions, nitrogen enhanced leaf water content, stabilized electrical conductivity, regulated antioxidant enzyme activity, and increased the accumulation of proline. However, under severe drought, nitrogen did not significantly improve biomass, highlighting the critical role of water availability. Additionally, increased nitrogen levels enhanced soil enzyme activity, facilitated the uptake of crucial nutrients like K and Zn. Mantel tests indicated significant correlations between soil enzyme activity, water use efficiency, and leaf Fe content, suggesting that nitrogen deposition altered nutrient uptake strategies in Q. dentata to sustain normal photosynthetic capacity under water stress. This study demonstrates that nitrogen deposition substantially enhances the growth and physiological resilience of Q. dentata under W50 by optimizing photosynthetic efficiency, water use efficiency, and nutrient uptake. However, the efficacy of nitrogen is highly dependent on water availability, highlighting the necessity of integrated nutrient and water management for plant growth.

In order to study the durability of molybdenum tailings concrete after a specific period of wet-dry cycle tests and exposure to high temperatures, this paper investigates the rule of change of the mechanical properties of molybdenum tailings concrete under the coupled influence of sulfate corrosion and high-temperature environmental conditions. A total of five C30 concrete prisms with molybdenum tailings content (0 %, 25 %, 50 %, 75 %, and 100 %) were considered in the tests, with sulfate solution concentrations of 0 g/L, 20 g/L, and 50 g/L, and exposure temperatures of 25° at room temperature and 400° at high temperature, respectively. The test results showed that the mass loss rate of concrete prisms with different molybdenum tailings substitution rates under sulfate corrosion and high temperature environment showed an increasing and then decreasing trend. The specimens with 25 % molybdenum tailings content still maintain high mechanical properties after wet-dry cycle tests of sulfate solution and high temperature, and the reduction of peak stress in this group is only 11 %, and the reduction of elastic modulus is 1 %; the rest of the molybdenum tailings content has a reduction of peak stress of about 25 %, and the reduction of modulus of elasticity is more than 10 %. The plastic deformation capacity of molybdenum tailings concrete decreases under the single-factor effects of wet-dry cycle tests of sulfate solution and high temperature effects, but the plastic deformability capacity of the specimens is enhanced under the combined effects of multiple factors. The stress-strain curve formulas for molybdenum tailings concrete under high temperature, wet-dry cycle tests of sulfate solution, and coupled wet-dry cycle tests of sulfate solution and high temperature were fitted. The results of this study can provide experimental data and valuable references for the engineering application of molybdenum tailings concrete under wet-dry cycle tests of sulfate solution and high temperature conditions, laying a foundation for further research.

Abstract Three mediators for the laccase mediator system, 3-hydroxyanthranilic acid (3-HAA), 4-hydroxybenzoic acid (HBA), and phenol red (PR) were investigated as mediators in an electrolytic mediator system (EMS) for the degradation of guaiacyl synthetic lignin (G-DHP). All the electron-oxidations of G-DHP with 3-HAA, HBA and PR in the absence of 2,6-lutidine proceeded to give the electrolysis residues in moderate yields. The significant β-β and β-5 linkage loss was found in all the electrolysis residues, especially the residue in the electro-oxidation with PR was significant. The addition of 2,6-lutidine as a base increased the yields of the electrolysis residues and influenced the relative ratio of β-O-4, β-5 and β-β linkages to some extent, that is, increase of β-O-4 linkage loss and decrease of β-β linkages loss (in the electro-oxidation with 3-HAA), increase of β-O-4 linkage loss (in that with 3-HBA), increase of β-5 linkages loss (in that with PR at 0.35 V) and decrease of β-O-4 and β-β linkages loss (in that with PR at 0.70 V). Thus, the base such as 2,6-lutidine was also one of the critical factors for reaction efficiency and reaction selectivity in the EMS. Consequently,3-HAA, HBA, and PR could be used as mediators in EMS for lignin degradation, especially 3-HAA is the most preferable because of the low applied potential.

Abstract This paper proposes a comprehensive control strategy for a battery energy storage system (BESS) participating in primary frequency modulation (FM) while considering the state of charge (SOC) recovery. On the one hand, when the frequency fluctuations are outside the dead zone of the FM, variable coefficient virtual sag control and variable coefficient virtual inertia control are coordinated for more accurate primary FM. Also, an assisted FM strategy is proposed, where the temporal alignment of assisted FM is determined by the regional segmentation of the frequency deviation and SOC, and it enables the BESS to recover SOC concurrently during its involvement in primary FM. On the other hand, when the frequency fluctuations are inside the dead zone of the FM, typical S‐type functions are deployed to dynamically establish the mutual constraints relationships of SOC recovery demand coefficient and frequency deviation. Subsequently, the recovery control coefficients are derived, and accordingly, a SOC recovery strategy is proposed. Finally, simulation results verify the efficacy of the proposed control strategy in the grid primary FM system and demonstrate that the strategy expedites the SOC recovery and concurrently diminishes the conventional unit output.

While municipal solid waste (MSW) provides an ideal habitat for pathogen propagation, the dynamics and assembly of airborne pathogen communities in these environments remain largely unknown. Here, we combined amplicon and metagenomics with spatiotemporal sampling to study inhalable particulate matter-carried potential pathogenic bacteria at full-scale food waste treatment plants (FWTPs), alongside comparisons to urban air in the area. The results showed that pathogenic bacteria constituted a notable portion (64.5 % ± 20.6 %, n = 75) of the total bacterial communities in FWTPs-impacted air, with species and relative abundance 2–4 times higher than that of urban air, and contributed over 50 % of pathogens to the outdoor air. Airborne pathogen community structures were highly shaped by sampling sites (i.e. treatment units), but conserved across seasons (summer vs. winter) and particle sizes (PM2.5 vs. PM10). Notably, Acinetobacter johnsonii-dominated pathogens (i.e. biofilm-related species) presented high levels of aerosolization and consistently occupied the upper-representative niches in all neutral models, highlighting their persistent exposure risk. Furthermore, pathogen community assembly was strongly driven by stochastic processes (58.8 %–96.8 %), while environmental variables explained only limited variations (3.4 %–28.7 %). In particular, the relative importance of stochastic processes clearly increased along an outdoor-to-indoor gradient (84.9 %–96.5 % vs. 71.3 %–76 %), which might be related to indoor anthropogenic activities that weaken microbial network stability and environmental filtering effects. This work enhances our knowledge of the dynamic behaviors and risk of airborne pathogen communities in MSW disposal and underscores the role of FWTPs in disseminating airborne pathogens.

Acoustic signals are vital in animal communication, and quantifying them is fundamental for understanding animal behaviour and ecology. Vocalizations can be classified into acoustically and functionally or contextually distinct categories, but establishing these categories can be challenging. Newly developed methods, such as machine learning, can provide solutions for classification tasks. The plains zebra is known for its loud and specific vocalizations, yet limited knowledge exists on the structure and information content of its vocalzations. In this study, we employed both feature-based and spectrogram-based algorithms, incorporating supervised and unsupervised machine learning methods to enhance robustness in categorizing zebra vocalization types. Additionally, we implemented a permuted discriminant function analysis to examine the individual identity information contained in the identified vocalization types. The findings revealed at least four distinct vocalization types—the ‘snort’, the ‘soft snort’, the ‘squeal’ and the ‘quagga quagga’—with individual differences observed mostly in snorts, and to a lesser extent in squeals. Analyses based on acoustic features outperformed those based on spectrograms, but each excelled in characterizing different vocalization types. We thus recommend the combined use of these two approaches. This study offers valuable insights into plains zebra vocalization, with implications for future comprehensive explorations in animal communication.

【Objective】The study aims to explore the effects of low-light stress on the main quality characteristics of fruits of different muskmelon varieties.【Method】The content changes of carbohydrates, carotenoids and chlorophyll in different fruit positions (pedicle, areola and pulp) of three varieties (Xizhoumi No. 1, Zaohuanghou and Jiashi) of muskmelon under different light intensities (100%, 42% and 23% transmittance) at different development stages (0, 3, 7, 14, 21, 35, 40, 60 d after blooming) were investigated.【Result】Results showed that contents of fruit starch, sucrose and reducing sugar all increased continuously during fruit development and the highest contents exhibited at mature stage (35 d), and Xizhoumi No. 1 had the highest starch content (10.835%), while Jiashi had the lowest starch content (8.342%). The pulp and areola of the fruits of three varieties showed higher sucrose content than that in pedicle, but no difference in reducing sugar content. The contents of carotenoid and chlorophyll increased during fruit development but decreasedat full ripe stage (60 d after blooming). Low-light stress significantly reduced the carbohydrate content, increased carotenoid and chlorophyll contents, and most significant effect was found under transmittance 23% treatment, under which the contents of starch, sucrose and reducing sugar in Xizhoumi No. 1 muskmelon reduced by 32.3%, 29.87% and 28.02%, while contents of carotene and chlorophyll increased by 19.47% and 13.98%, respectively. The contents of starch, sucrose and reducing sugar in Zaohuanghou muskmelon reduced by 32.63%, 25.09% and 20.24%, and the contents of carotene and chlorophyll increased by 54.74% and 21.06%, respectively. The contents of starch, sucrose and reducing sugar in Jiashi muskmelon reduced by 41.08%, 32.39% and 39.1%, and the contents of carotene chlorophyll increased by 55.20% and 31.79%, respectively. Compared with Xizhoumi No. 1 and Zaohuanghou muskmelons, the Jiashi muskmelon showed the lowest starch and chlorophyll contents and the highest sucrose and reducing sugar contents, and exhibited the most significant decline of starch, sucrose and reducing sugar contents, and increase of carotenoid and chlorophyll contents, indicating that the Jiashi muskmelon was affected most significantly by low-light stress and showed poorest low-light tolerance.【Conclusion】The low-light stress reduces the commercial quality of muskmelon fruits, and the quality is worse when the light transmittance is lower. Jiashi muskmelon is most sensitive to low-light stress.

It is a challenge to obtain highly tunable multifunctional performances in one ferroelectric system by a simple approach to meet the miniaturization, integration, and functionalization requirements of advanced electronic components. Herein, rare earth erbium (Er) modulated 0.9K0.5Na0.5NbO3-0.1Sr(1-x)ErxTi(1-x/4)O3, (0.9KNN-0.1ST: xEr) transparent-photoluminescent-ferroelectric energy storage multifunctional ceramics are prepared to solve this problem. The effect of lattice distortion and oxygen vacancies by Er doping on the optical and electrical properties is systematically investigated. The Er3+ ions can introduce a large distortion of the NbO6 octahedron by replacing the A-site in KNN-based ceramics. Thanks to the higher c/a ratio and lower oxygen vacancy content are simultaneously obtained in 0.9KNN-0.1ST: 0.1Er ceramics. The effective energy storage density (Wrec) of 0.86 J/cm3, excellent near-infrared transmittance of 51.7% (1 100 nm) and strong green upconversion photoluminescence are achieved in this multifunctional ceramic. This study provides a solid basis for rare earth ions doped ferroelectric ceramics with tunable multifunctional properties and has significant potential for applications in optoelectronic devices.

Logs are valuable information for oil and gas fields as they help to determine the lithology of the formations surrounding the borehole and the location and reserves of subsurface oil and gas reservoirs. However, important logs are often missing in horizontal or old wells, which poses a challenge in field applications. To address this issue, conventional methods involve supplementing the missing logs by either combining geological experience and referring data from nearby boreholes or reconstructing them directly using the remaining logs in the same borehole. Nevertheless, there is currently no quantitative evaluation for the quality and rationality of the constructed log. In this paper, we utilize data from the 2020 machine learning competition of the Society of Petrophysicists and Logging Analysts (SPWLA), which aims to predict the missing compressional wave slowness (DTC) and shear wave slowness (DTS) logs using other logs in the same borehole. We employ the natural gradient boosting (NGBoost) algorithm to construct an Ensemble Learning model that can predicate the results as well as their uncertainty. Furthermore, we combine the SHAP (SHapley Additive exPlanations) method to investigate the interpretability of the machine learning model. We compare the performance of the NGBosst model with four other commonly used Ensemble Learning methods, including Random Forest, GBDT, XGBoost, LightGBM. The results show that the NGBoost model performs well in the testing set and can provide a probability distribution for the prediction results. This distribution allows petrophysicists to quantitatively analyze the confidence interval of the constructed log. In addition, the variance of the probability distribution of the predicted log can be used to justify the quality of the constructed log. Using the SHAP explainable machine learning model, we calculate the importance of each input log to the predicted results as well as the coupling relationship among input logs. Our findings reveal that the NGBoost model tends to provide greater slowness prediction results when the neutron porosity (CNC) and gamma ray (GR) are large, which is consistent with the cognition of petrophysical models. Furthermore, the machine learning model can capture the influence of the changing borehole caliper on slowness, where the influence of borehole caliper on slowness is complex and not easy to establish a direct relationship. These findings are in line with the physical principle of borehole acoustics. Finally, by using the explainable machine learning model, we observe that although we did not correct the effect of borehole caliper on the neutron porosity log through preprocessing, the machine learning model assigned a greater importance to the influence of the caliper, achieving the same effect as caliper correction.

Abstract Background Sutureless scleral fixed intraocular lens implantation (SF-IOL) has become one of the mainstream schemes in clinical treatment of aphakic eyes because of its advantages, such as avoiding dislocation of intraocular lens or subluxation caused by suture degradation or fracture and significant improvement of postoperative visual acuity. However, a consensus on the relative effectiveness and safety of this operation and other methods is still lacking. This study aimed to compare the efficacy and safety of sutureless SF-IOL with other methods. Aphakia means that the lens leaves the normal position and loses its original function, including absence or complete dislocation and subluxation of the lens which could cause anisometropic amblyopia, strabismus, and loss of binocular function in children and adolescents. For adults, the loss of the lens could lead to high hyperopia and affect vision. Above all this disease can seriously affect the quality of life of patients. Methods Literature about sutureless SF-IOL in PubMed, Cochrane Library, Embase, Web of Science, China National Knowledge Infrastructure, China Technical Journal VIP database, and Wanfang database published from 2000 to 2022 was reviewed. The weighted average difference was calculated by RevMan5.3 software for analysis. Two researchers independently selected the study and used the Cochrane collaboration tool to assess the risk of errors. Cochrane bias risk tool was used to evaluate the quality of evidence. This study is registered on PROSPERO (CRD42022363282). Results The postoperative IOL-related astigmatism of sutureless SF-IOL was lower than that of suture SF-IOL, and there was statistical difference when we compared the absolute postoperative spherical equivalent after sutureless SF-IOL and suture SF-IOL. Indicating that the degree of refractive error after sutureless SF-IOL was lower. Meanwhile, the operation time of sutureless SF-IOL was shorter than that of suture SF-IOL. The subgroup analysis showed that the absolute postoperative spherical equivalent and astigmatism values in Yamane technique were lower than those in suture SF-IOL. Conclusion Sutureless SF-IOL has the advantages of stable refraction, short operation time, and less postoperative complications. However, high-quality literature to compare these technologies is lacking. Some long-term follow-up longitudinal prospective studies are needed to confirm the findings.

During winter of 2021/2022, the temperature in China is characterized by a warm-to-cold transition, and the average temperature anomaly in February 2022 is −1.6 °C, the coldest February in 2013–2022. We revealed the circulation regimes and physical mechanisms associated with this reversal event and demonstrated the advantage of a regional model downscaling over the use of the global model alone in predicting. In early winter, the warm anomalies are mainly related to an anomalous anticyclonic system downstream of a PNA-like (Pacific–North America) Rossby-wave train induced by La Niña. In late winter, due to the circulation response to the central Pacific warming and negative tropical Indian Ocean Dipole (TIOD), two ‘−＋−’ Rossby-wave trains from high latitudes and the tropical Indian Ocean jointly lead to an anomalous cyclonic system in China. Meanwhile, an anticyclonic blocking system on the northern side of Baikal brings strong and cold air to China. These two systems together cause a significant drop in surface air temperature anomaly in China during the late winter. The Beijing Climate Center climate system model (BCC_CSM1.1 m) can essentially predict this temperature reversal in China about five months in advance. However, the reversal amplitude is weaker due to warm deviations over the tropical Pacific Ocean and equatorial Indian Ocean. Using dynamic downscaling, a regional Climate–Weather Research and Forecasting (CWRF) model correctly predicts the cold SAT anomalies in late winter 2021/2022. The regional model depicts more realistic circulation patterns in East Asia; the anomalous cyclonic system in Inner Mongolia accompanied by the northerly anomalies contribute to a lower-than-normal SAT over China. This study reveals the cooperative effect of wave trains from high latitudes and the tropics on the subseasonal temperature reversal and demonstrates a possible solution to improve the forecast skill by dynamic downscaling according to precise characterization of local surface information.

Taking the constituent elements other than the main buildings of local Confucian temples as a research object, including memorial archway, screen wall, Pan-chi (pond), worship altar, sacrificial hall, pavilion and tower, this paper combines textual narrative and historical measurement to sort out the production and development history of these constituent elements. It analyzes the spatial evolution trend of Confucian temples to present the dynamic processes of derivation of constituent elements and spatial evolution of local Confucian temples. On this basis, this paper surveys the causes of the growth of Confucian temples from a macro historical perspective by considering political, social and cultural factors. This dynamic development process, sacred and secular as it is, will grow and last forever under the multiple official implications and folk beliefs.

Abstract Background Surgical treatment of finger nerve injury is common for hand trauma. However, there are various surgical options with different functional outcomes. The aims of this study are to compare the outcomes of various finger nerve surgeries and to identify factors associated with the postsurgical outcomes via a systematic review and meta-analysis. Methods The literature related to digital nerve repairs were retrieved comprehensively by searching the online databases of PubMed from January 1, 1965, to August 31, 2021. Data extraction, assessment of bias risk and the quality evaluation were then performed. Meta-analysis was performed using the postoperative static 2-point discrimination (S2PD) value, moving 2-point discrimination (M2PD) value, and Semmes–Weinstein monofilament testing (SWMF) good rate, modified Highet classification of nerve recovery good rate. Statistical analysis was performed using the R (V.3.6.3) software. The random effects model was used for the analysis. A systematic review was also performed on the other influencing factors especially the type of injury and postoperative complications of digital nerve repair. Results Sixty-six studies with 2446 cases were included in this study. The polyglycolic acid conduit group has the best S2PD value (6.71 mm), while the neurorrhaphy group has the best M2PD value (4.91 mm). End-to-side coaptation has the highest modified Highet’s scoring (98%), and autologous nerve graft has the highest SWMF (91%). Age, the size of the gap, and the type of injury were factors that may affect recovery. The type of injury has an impact on the postoperative outcome of neurorrhaphy. Complications reported in the studies were mainly neuroma, cold sensitivity, paresthesia, postoperative infection, and pain. Conclusion Our study demonstrated that the results of surgical treatment of digital nerve injury are generally satisfactory; however, no nerve repair method has absolute advantages. When choosing a surgical approach to repair finger nerve injury, we must comprehensively consider various factors, especially the gap size of the nerve defect, and postoperative complications. The data from new distribution system is increasingly multi-dimensional and massive. When the distribution network participates in virtual grid division, the deep mining of distribution network data is required. In this study, a feature extraction and source–load collaborative analysis method for distribution network is proposed to master the suitability between renewable energy generation and various loads. Firstly, this method realizes raw data preprocessing through anomaly identification and reconstruction. It then uses information entropy to improve traditional piecewise aggregation approximation to reduce data dimension while considering the fluctuations of data. Finally, the method extracts the features of source–load data through spectral clustering and obtains quantitative analysis results of the matching degree between source and load through collaborative analysis. The effectiveness of the proposed method is verified by load power data and photovoltaic output data.

The exploration and comprehensive assessment of fractured-vuggy reservoir information have perennially constituted focal points and challenges within the domain of oil and gas reservoir evaluation. The verification of geological phenomena, identification of various fracture and hole types, and the quantitative characterization thereof currently present pressing challenges. This study meticulously examines the deep carbonate reservoirs within the Dengying Formation in the Penglai gas region of the Sichuan Basin. The Core Rolling Scan images reveal five discernible features: unfilled holes, filled holes, filled fractures, open fractures, and algae. The analysis pinpoints three primary challenges in semantic segmentation recognition: the amalgamation of feature scales, class imbalance, and the scarcity of datasets with substantial sample sizes. To address these challenges, this paper introduces a Multi-Scale Feature Aggregation Pyramid Network model (MFAPNet), achieving a pixel accuracy of 68.04% in recognizing the aforementioned five types. Lastly, the model is employed in calculating core porosity, exposing a scaling relationship between wellbore image porosity and core porosity ranging from 1.5 to 3 times. To a certain extent, it reveals the correlation between the wellbore image logging data and the actual formation of the Dengying Formation in the Penglai Gas Field of the Sichuan Basin, and also provides a basis for the subsequent logging evaluation of the formation. Coke plays a key role as the skeleton of the charge column in BF. The gas path formed by the coke layer in the BF has a decisive influence on gas permeability. At high temperatures, the interface between coke and ore undergoes a melting reaction of coke and a reduction reaction of ore. The better the reducibility of the ore, the more conducive it is to the coupling reaction of ore and coke. The melting loss reaction of coke becomes more intense, and the corresponding strength of coke will decrease, which will affect the permeability of the blast furnace and is not conducive to the smooth operation of the blast furnace. Especially with a deterioration in iron ore quality, BF operation faces severe challenges, which makes it necessary to find an effective way to strengthen BF operation. In this study, a melting-dropping furnace was used to develop and clarify the high-temperature interaction between coke and iron ores with different layer thicknesses. The influencing factors were studied by establishing a gas permeability mathematical model and observing the metallographic microscope images of samples after the coke solution loss reaction. The relationships between coke layer thickness, distribution of gas flow, and pressure drop were obtained. The results showed that, under certain conditions, the gas permeability property of a furnace burden has been improved after the coke layer thickness increased. Upon observing the size of coke particles at the interface reaction site, the degree of melting loss reaction can be determined. A smaller particle size indicates more melting loss reaction. A dripping eigenvalue for molten metal was introduced to evaluate the dynamic changes in the comprehensive dripping properties of molten metal of furnace burden, which showed that the dripping eigenvalue for the molten metal could deteriorate because of the unruly thickness and the coke layer thickness should be limited through considering the operational indicators of the blast furnace.

Abstract Our study looks at the precipitation responses to two possible future emission‐mitigation pathways, pushed by the Coronavirus Disease 2019 pandemic (COVID‐19) and achieving carbon neutrality in the mid‐21st century. We find that a simultaneous‐reduction in well‐mixed greenhouse gases (WMGHGs) and anthropogenic aerosol emissions results in an enhanced interhemispheric precipitation contrast in the 2040s by amplifying the interhemispheric thermal contrast and strengthening the meridional overturning circulation in the tropics. Reduced aerosol emissions induce generally‐increased precipitation in the Northern Hemisphere (NH) and an amplified intertropical rainfall contrast, while reduced WMGHG emissions dominate decrease in precipitation in the areas away from aerosol emission sources. Further, the above precipitation contrast will be enhanced under stronger emission‐mitigation pathways, mainly attributed to larger precipitation increases in the NH caused by reduced aerosols. More aggressive WMGHGs mitigation policies are necessary to counteract the aerosol‐induced warming in the NH, thereby mitigating the risk of regional drying or wetting due to the asymmetry in interhemispheric energy budgets.

Biodegradable plastics (BDPs) have attracted extensive attention as an alternative to conventional plastics. BDPs could be mineralized by composting, while the quality of compost affected by the presence of BDPs and the residual microplastics (MPs) has not been well evaluated. This study aimed to explore the MPs release potential and environmental implications of commercial BDPs (aromatic−aliphatic copolymer) films in uncontrolled composting. Results showed that the molecular weight of BDPs decreased by >60% within 60 d. However, the non-extracted organic matter and wet-sieving measurements indicated that MPs continuously released and accumulated during regular composting. The average MPs release potential (0.1–5 mm) was 134.6 ± 18.1 particles/mg (BDPs), which resulted in 103–104 particles/g dw in compost. The plastisphere of MPs showed a significantly higher (0.95–16.76 times) abundance of antibiotic resistance genes (ARGs), which resulted in the rising (1.34–2.24 times) of ARGs in compost heaps, in comparison to the control groups. Overall, BDPs promote the spread of ARGs through the selective enrichment of bacteria and horizontal transfer from released MPs. These findings confirmed that BDPs could enhance the release potential of MPs and the dissemination of ARGs, which would promote the holistic understanding and environmental risk of BDPs.

Varicocele surgical repair can improve the function of the testis for patients with varicocele. We carried out a systematic review and meta-analysis to assess the effects of varicocele surgical repair on serum hormones and inhibin B levels in patients with varicocele. A literature search was performed in August 2022, and no language or geographic region restrictions were applied. The search included the following databases: PubMed, Embase, and Medline. A literature review was performed to identify all published clinical trials assessing serum hormone and inhibin B levels before and after varicocele surgical repair. The reference lists of retrieved studies were also investigated. A systematic review and meta-analysis were conducted. Eight articles were selected from 162 articles, including 452 patients. The combined analysis showed that after surgical treatment, mean serum testosterone, inhibin B, and sperm concentration levels increased compared with preoperative levels ( p < .05). After surgical treatment, mean serum follicle-stimulating hormon (FSH), and Luteinizing hormone (LH) levels decreased compared with preoperative levels ( p < .05). This meta-analysis demonstrates that varicocele surgical repair can improve testicular function, increase serum testosterone, and inhibin B levels and decrease serum FSH and LH levels in patients with varicocele. This might be related to the improvement of infertility. A large-scale multicenter randomized controlled study is needed for further confirmation.

Abstract Purpose: To evaluate the characteristics of dome-shaped macula (DSM) in children aged 4–6 years with normal visual acuity using optical coherence tomography angiography. Method: This is a cross-sectional study. A total of 19 children aged 4–6 years were included. The results of optical coherence tomography angiography images were analysed to identify and quantify retinal structural and vascular parameters in DSM children. The dome height, dome base, and sub-dome choroidal thickness were manually measured. Participants with DSM and those without DSM from our previous study were compared on these parameters. Results: Nineteen eyes of the preschool subjects with normal visual acuity showed horizontal DSM on optical coherence tomography (OCT). The DSM was significantly smooth and low in the children, and we did not observe differences between sex and age. Compared to the children without DSM, the average axial length was longer, and the average macular vessel density was lower in the DSM group, especially in the deep retinal vascular density. Additionally, the dome height was positively correlated with the sub-dome choroidal thickness. When the dome base/height was increased, the fovea avascular zone (FAZ) area was larger. Conclusion: Dome-shaped macula was detected in the preschool children in the process of the emmetropization with normal visual acuity. The changes in macular structure and vasculature provide new ideas for further investigation into the characteristics of DSM formation.

AIM: To report on the clinical features of patients with retinal amyloid angiopathy (RAA) who were identified to be caused by the transthyretin (TTR) Gly83Arg variant. METHODS: Case series of five patients diagnosed with RAA was collected at Affiliated Hospital of Zunyi Medical University from January 2010 to December 2021. The clinical features, therapeutic strategies, and prognoses of all patients were reviewed. RESULTS: Five patients with a mean age of 52.00±7.23y were diagnosed as RAA. These patients were previously diagnosed with hereditary transthyretin amyloidosis caused by the TTR Gly83Arg variant. Vitreous opacity was found in all 10 eyes, and 7 eyes developed RAA 2 to 20y after the onset of hereditary transthyretin amyloidosis. The clinical manifestations were recurrent vitreous hemorrhage in 2 eyes (29%), neovascular glaucoma in 2 eyes (29%), and iris neovascularization in 1 eye (14%). Microangioma lesions were found in all affected eyes that underwent fundus fluorescein angiography (FFA) in this group of cases, and the incidence of the retinal non-perfusion area was 67%. Although no cases of retinal neovascularization were found, the prognosis of visual acuity was not ideal. CONCLUSION: This is the first report of RAA in patients with the TTR Gly83Arg variant. Complications such as RAA and glaucoma will seriously affect the visual prognosis of patients. Thereafter, regular ophthalmic follow-up of patients with hereditary transthyretin amyloidosis is essential. And FFA after vitrectomy is very important, which can help ophthalmologists detect RAA earlier and treat it in time.

In order to know how surface air temperature (SAT) changes in East Asia under different emission scenarios after the COVID-19 outbreak, we investigated the impacts of greenhouse gases (GHGs) and anthropogenic aerosols changes on SAT in East Asia by using the aerosol-climate coupled model BCC-AGCM 2.0_CUACE/Aero, combining with the post-pandemic emission scenarios proposed by Covid multi-Earth system model intercomparison project (CovidMIP scenarios for short, including fossil-fueled recovery, moderate green stimulus, strong green stimulus, hereinafter as FFF, MGG, SGG, respectively). We assessed the impacts of changes in GHGs and anthropogenic aerosols together and separately on SAT in East Asia and its typical subregions during 2020–2050. The results show that by mid-21st-century, SAT in East Asia will increase by 0.81 ± 0.083 °C under Baseline (same as SSP2-4.5 scenario), i.e., SAT difference between 2045–2050 and 2020–2025), and there will be more intense warming in all the three scenarios in East Asia, in which the largest SAT difference (SAT-d) compared to Baseline is 0.33 ± 0.11 °C under SGG and the smallest SAT-d is 0.07 ± 0.14 °C under FFF. To further explore the mechanism of these SAT-d, we analyzed the trend of surface longwave and shortwave net radiation flux driven by GHGs and anthropogenic aerosols there. It is found that in early period (2020–2035), the role of aerosol changes is bigger than that of GHG changes in dominating SAT-d, particularly sulfate, whose reduction will become the main contributor to SAT-d by affecting the net solar flux at surface. In later period (2036–2050), because of GHGs’ longer atmospheric lifetime than aerosols, the role of decreasing GHGs concentrations will determine the drop in SAT-d through affecting the net longwave flux at surface.

A proper assessment of annual climatic status (ACS) is conducive to rationally formulating disaster prevention and mitigation measures. The former definition of ACS lacks either information on extreme climate or an intuitive grade feature service to the public. The ACS defined in the National Standard of the People's Republic of China (GBACS) only considers the accumulated climate effects of each weighted 10-d temperature/precipitation anomaly in a year. Under global warming, the losses caused by extreme climate events often have a significant impact on the grades of ACS, but this impact cannot be reasonably reflected by GBACS. This study proposed the assessment of ACS using extreme climate indices (extreme climate-based ACS (ECACS)) and compared it with GBACS. The results indicated that GBACS and ECACS can be used to evaluate the ACS from different angles. The ECACS is an important supplement to the GBACS, especially considering the years with frequently occurring extreme climate events. The sum of GBACS and ECACS is a reasonable and comprehensive way to evaluate the ACS. The empirical orthogonal function (EOF) analysis indicated a uniform mode (EOF1) and a dipole mode (EOF2) in GBACS and ECACS in China. The interannual variation characteristics of ECACS in northern and southern China (EOF2 pattern) are consistent with real climate features, which is conducive to providing better and more detailed regional information in the ACS forecast service. The results have essential instructive and application value for ACS assessment and government decision making.

Abstract Background Central Retinal Vein Occlusion (CRVO) is a rare complication of von Hipple-Lindau (VHL) disease. This report presents the first case of VHL disease complicated with CRVO caused by VHL c.208G > A mutation. Case presentation A 20 s man whose left eye visual acuity gradually declined for half a year. The visual acuity of the left eye is counting fingers. Fundus examination revealed that retinal hemangioblastoma was also found in addition to typical CRVO signs such as tortuous expansion of retinal veins and flame-shaped hemorrhage of the retina. Liver tumor, cerebral infarction and erythrocytosis were found during systemic examination, and the diagnosis of polycythemia was confirmed by bone marrow smear. Furthermore, both family history and genetic analysis indicated that the patient had VHL disease caused by VHL c.208G > A. In this patient, a large number of bone marrow erythrocytes proliferated due to VHL disease, which led to the increase of blood viscosity and erythrocyte vascular adhesion, resulting in the obstruction of central retinal vein blood flow, and finally CRVO. For CRVO and its pathogenic factor polycythemia, patient received laser retinal photocoagulation and phlebotomies. After a 1-year follow-up, the vision in the left eye improved to 0.2 logMAR. Conclusions This is a rare case of polycythemia complicated by CRVO in patient with VHL disease. It reminds us that the systemic disease factors should be fully considered in the diagnosis of young patients with CRVO, and that treatment requires a coordinated effort of physicians.

CaZrO3 (CZO) thin films were deposited on Pt/Ti/SiO2/Si substrates at 450 °C by radio-frequency magnetron sputtering technology. The microstructures and dielectric properties of CZO thin films were investigated. X-ray diffraction analysis reveals that the perovskite orthogonal CZO phase would be promoted by a higher O2 partial pressure in the flow ratio of O2/Ar after thin films were annealed at 700 °C for 3 h in air. The films prepared under the flow ratio of O2/Ar (20:40, 30:40 and 40:40) show the main perovskite crystal phase of CaZrO3 with a small amount of Ca0.2Zr0.8O1.8. The main crystal phase was Ca0.2Zr0.8O1.8 when the film was deposited under an O2/Ar ratio of 40:10. The annealed film with a 40:40 O2/Ar ratio exhibits a dielectric performance with a high dielectric constant (εr) of 25 at 1 MHz, a temperature coefficient of permittivity of not more than 122.7 ppm/°C from 0 °C to 125 °C, and a low leakage current density of about 2 × 10−7 A/cm2 at 30 V with an ohmic conduction mechanism.

BackgroundPelvic floor muscle strength is well-known to be associated with female sexual function. However, there were a few studies that reported on the relationship between pelvic floor muscle strength and female sexual function in pregnant women, and the presented results were inconsistent. Nulliparae represent a specific cohort with simplicity to exclude confounding factors that are caused by parity. The present study aimed to explore the association of pelvic floor muscle strength and sexual function based on the Pelvic Organ Prolapse/Urinary Incontinence Sexual Questionnaire (PISQ-12) of nulliparae during pregnancy.MethodsThis is the second analysis of the baseline data from a randomized controlled trial (RCT), which aimed to study the protective efficacy of pelvic floor muscle training on stress urinary incontinence at 6th week postpartum (registration number: ChiCTR2000029618). Nulliparae aged 20–40 years with singleton pregnancy before 16 weeks of gestation were enrolled in this study, and data, including participants' demographic information, the Modified Oxford Scale (MOS), and PISQ-12, were collected. Eligible nulliparae were divided into two groups: Group MOS > 3 and Group MOS ≤ 3. Demographic information of the two groups was compared. Sexual function based on the PISQ-12 scores of the two groups was compared. A comparison of the PISQ-12 scores between the two groups was calculated by the Mann–Whitney U-test using SPSS version 23.0.ResultsA total of 735 eligible nulliparae were enrolled in this study. Along with MOS grading up, PISQ-12 scores tended to get lower. Of the 735 nulliparae, there were 378 and 357 participants included in Group MOS > 3 and Group MOS ≤ 3, respectively. The PISQ-12 scores of Group MOS > 3 were significantly lower than those of Group MOS ≤ 3 (11 vs. 12, p < 0.001). The scores of the frequency of feeling sexual desire, orgasm achievement, sexual excitement, sexual activity satisfaction, sexual intercourse pain, fear of urinary incontinence, and negative emotion reactions with the sexual intercourse of Group MOS > 3 were lower than those of Group MOS ≤ 3 (p < 0.05).ConclusionPelvic floor muscle strength was positively associated with sexual function based on the questionnaire of young nulliparae during their first trimester. Up to half of the nulliparae during the first trimester were suffering from weak pelvic floor muscle strength and nearly a quarter of the nulliparae were facing this weakness combined with sexual dysfunction.Trial registrationThis study has been registered at http://www.chictr.org.cn (registration number: ChiCTR2000029618).

Abstract Background The explosive radiation and diversification of the advanced snakes (superfamily Colubroidea) was associated with changes in all aspects of the shared venom system. Morphological changes included the partitioning of the mixed ancestral glands into two discrete glands devoted for production of venom or mucous respectively, as well as changes in the location, size and structural elements of the venom-delivering teeth. Evidence also exists for homology among venom gland toxins expressed across the advanced snakes. However, despite the evolutionary novelty of snake venoms, in-depth toxin molecular evolutionary history reconstructions have been mostly limited to those types present in only two front-fanged snake families, Elapidae and Viperidae. To have a broader understanding of toxins shared among extant snakes, here we first sequenced the transcriptomes of eight taxonomically diverse rear-fanged species and four key viperid species and analysed major toxin types shared across the advanced snakes. Results Transcriptomes were constructed for the following families and species: Colubridae - Helicops leopardinus, Heterodon nasicus, Rhabdophis subminiatus; Homalopsidae – Homalopsis buccata; Lamprophiidae - Malpolon monspessulanus, Psammophis schokari, Psammophis subtaeniatus, Rhamphiophis oxyrhynchus; and Viperidae – Bitis atropos, Pseudocerastes urarachnoides, Tropidolaeumus subannulatus, Vipera transcaucasiana. These sequences were combined with those from available databases of other species in order to facilitate a robust reconstruction of the molecular evolutionary history of the key toxin classes present in the venom of the last common ancestor of the advanced snakes, and thus present across the full diversity of colubroid snake venoms. In addition to differential rates of evolution in toxin classes between the snake lineages, these analyses revealed multiple instances of previously unknown instances of structural and functional convergences. Structural convergences included: the evolution of new cysteines to form heteromeric complexes, such as within kunitz peptides (the beta-bungarotoxin trait evolving on at least two occasions) and within SVMP enzymes (the P-IIId trait evolving on at least three occasions); and the C-terminal tail evolving on two separate occasions within the C-type natriuretic peptides, to create structural and functional analogues of the ANP/BNP tailed condition. Also shown was that the de novo evolution of new post-translationally liberated toxin families within the natriuretic peptide gene propeptide region occurred on at least five occasions, with novel functions ranging from induction of hypotension to post-synaptic neurotoxicity. Functional convergences included the following: multiple occasions of SVMP neofunctionalised in procoagulant venoms into activators of the clotting factors prothrombin and Factor X; multiple instances in procoagulant venoms where kunitz peptides were neofunctionalised into inhibitors of the clot destroying enzyme plasmin, thereby prolonging the half-life of the clots formed by the clotting activating enzymatic toxins; and multiple occasions of kunitz peptides neofunctionalised into neurotoxins acting on presynaptic targets, including twice just within Bungarus venoms. Conclusions We found novel convergences in both structural and functional evolution of snake toxins. These results provide a detailed roadmap for future work to elucidate predator–prey evolutionary arms races, ascertain differential clinical pathologies, as well as documenting rich biodiscovery resources for lead compounds in the drug design and discovery pipeline.

Fentanyl and its analogs are selective agonists of the µ-opioid receptor (MOR). Among novel synthetic opioids (NSOs), they dominate the recreational drug market and are the main culprits for the opioid crisis, which has been exacerbated by the COVID-19 pandemic. By taking advantage of the crystal structures of the MOR, several groups have investigated the binding mechanism of fentanyl, but have not reached a consensus, in terms of both the binding orientation and the fentanyl conformation. Thus, the binding mechanism of fentanyl at the MOR remains an unsolved and challenging question. Here, we carried out a systematic computational study to investigate the preferred fentanyl conformations, and how these conformations are being accommodated in the MOR binding pocket. We characterized the free energy landscape of fentanyl conformations with metadynamics simulations, and compared and evaluated several possible fentanyl binding conditions in the MOR with long-timescale molecular dynamics simulations. Our results indicate that the most preferred binding pose in the MOR binding pocket corresponds well with the global minimum on the energy landscape of fentanyl in the absence of the receptor, while the energy landscape can be reconfigured by modifying the fentanyl scaffold. The interactions with the receptor may stabilize a slightly unfavored fentanyl conformation in an alternative binding pose. By extending similar investigations to fentanyl analogs, our findings establish a structure–activity relationship of fentanyl binding at the MOR. In addition to providing a structural basis to understand the potential toxicity of the emerging NSOs, such insights will contribute to developing new, safer analgesics.

Objectives: To explore the differences in the cortical morphometric similarity network (MSN) between COVID-19 survivors and healthy controls, and the correlation between these differences and behavioral features and transcriptional signatures. Materials & methods: 39 COVID-19 survivors and 39 age-, sex- and education years-matched healthy controls (HCs) were included. All participants underwent MRI and behavioral assessments (PCL-17, GAD-7, PHQ-9). MSN analysis was used to compute COVID-19 survivors vs. HCs differences across brain regions. Correlation analysis was used to determine the associations between regional MSN differences and behavioral assessments, and determine the spatial similarities between regional MSN differences and risk genes transcriptional activity. Results: COVID-19 survivors exhibited decreased regional MSN in insula, precuneus, transverse temporal, entorhinal, para-hippocampal, rostral middle frontal and supramarginal cortices, and increased regional MSN in pars triangularis, lateral orbitofrontal, superior frontal, superior parietal, postcentral, and inferior temporal cortices. Regional MSN value of lateral orbitofrontal cortex was positively associated with GAD-7 and PHQ-9 scores, and rostral middle frontal was negatively related to PHQ-9 scores. The analysis of spatial similarities showed that seven risk genes (MFGE8, MOB2, NUP62, PMPCA, SDSL, TMEM178B, and ZBTB11) were related to regional MSN values. Conclusion: The MSN differences were associated with behavioral and transcriptional signatures, early psychological counseling or intervention may be required to COVID-19 survivors. Our study provided a new insight into understanding the altered coordination of structure in COVID-19 and may offer a new endophenotype to further investigate the brain substrate.

Abstract Background Atrial natriuretic peptide (ANP) secreted from atrial myocytes is shown to possess anti-inflammatory, anti-oxidant and immunomodulatory effects. The aim of this study is to assess the effect of ANP on bacterial lipopolysaccharide (LPS)-induced endotoxemia-derived neuroinflammation and cognitive impairment. Methods LPS (5 mg/kg) was given intraperitoneally to mice. Recombinant human ANP (rhANP) (1.0 mg/kg) was injected intravenously 24 h before and/or 10 min after LPS injection. Subdiaphragmatic vagotomy (SDV) was performed 14 days before LPS injection or 28 days before fecal microbiota transplantation (FMT). ANA-12 (0.5 mg/kg) was administrated intraperitoneally 30 min prior to rhANP treatment. Results LPS (5.0 mg/kg) induced remarkable splenomegaly and an increase in the plasma cytokines at 24 h after LPS injection. There were positive correlations between spleen weight and plasma cytokines levels. LPS also led to increased protein levels of ionized calcium-binding adaptor molecule (iba)-1, cytokines and inducible nitric oxide synthase (iNOS) in the hippocampus. LPS impaired the natural and learned behavior, as demonstrated by an increase in the latency to eat the food in the buried food test and a decrease in the number of entries and duration in the novel arm in the Y maze test. Combined prophylactic and therapeutic treatment with rhANP reversed LPS-induced splenomegaly, hippocampal and peripheral inflammation as well as cognitive impairment. However, rhANP could not further enhance the protective effects of SDV on hippocampal and peripheral inflammation. We further found that PGF mice transplanted with fecal bacteria from rhANP-treated endotoxemia mice alleviated the decreased protein levels of hippocampal polyclonal phosphorylated tyrosine kinase receptor B (p-TrkB), brain-derived neurotrophic factor (BDNF) and cognitive impairment, which was abolished by SDV. Moreover, TrkB/BDNF signaling inhibitor ANA-12 abolished the improving effects of rhANP on LPS-induced cognitive impairment. Conclusions Our results suggest that rhANP could mitigate LPS-induced hippocampal inflammation and cognitive dysfunction through subdiaphragmatic vagus nerve-mediated gut microbiota–brain axis.

BackgroundChest computerized tomography (CT) plays an important role in detecting patients with suspected coronavirus disease 2019 (COVID-19), however, there are no systematic summaries on whether the chest CT findings of patients within mainland China are applicable to those found in patients outside.MethodsRelevant studies were retrieved comprehensively by searching PubMed, Embase, and Cochrane Library databases before 15 April 2022. Quality assessment of diagnostic accuracy studies (QUADAS) was used to evaluate the quality of the included studies, which were divided into two groups according to whether they were in mainland China or outside. Data on diagnostic performance, unilateral or bilateral lung involvement, and typical chest CT imaging appearances were extracted, and then, meta-analyses were performed with R software to compare the CT features of COVID-19 pneumonia between patients from within and outside mainland China.ResultsOf the 8,258 studies screened, 19 studies with 3,400 patients in mainland China and 14 studies with 554 outside mainland China were included. Overall, the risk of quality assessment and publication bias was low. The diagnostic value of chest CT is similar between patients from within and outside mainland China (93, 91%). The pooled incidence of unilateral lung involvement (15, 7%), the crazy-paving sign (31, 21%), mixed ground-glass opacities (GGO) and consolidations (51, 35%), air bronchogram (44, 25%), vascular engorgement (59, 33%), bronchial wall thickening (19, 12%), and septal thickening (39, 26%) in patients from mainland China were significantly higher than those from outside; however, the incidence rates of bilateral lung involvement (75, 84%), GGO (78, 87%), consolidations (45, 58%), nodules (12, 17%), and pleural effusion (9, 15%) were significantly lower.ConclusionConsidering that the chest CT features of patients in mainland China may not reflect those of the patients abroad, radiologists and clinicians should be familiar with various CT presentations suggestive of COVID-19 in different regions.

Being an industrially valuable species of bivalve, Mytilus is widespread in the area around the eastern coast of China. Starvation is an environmental stress that mussels often encounter in their growth. However, few report carried out on the physiological and biochemical response and molecular regulation and adaptation mechanism of mussels under starvation. To illustrate the molecular mechanism of hosts of Mytilus to starvation, the gill transcriptome was used for analysis before and after 9 days of starvation in Mytilus using the Illumina/HiSeq-2000 deep sequencing platform, accompanied by the study of differentially expressed genes (DEGs). In total, 42.137 GB of clean data were obtained from six sample libraries, the average amount of clean data of each sample is 7.023 GB, and 58,540 unigenes that average 780 bp in length were assembled. Unigenes were illustrated by commenting them against the NR, Swiss-Prot, Pfam, String, GO and KEGG databases. After 9 days of starvation, 2,188 and 2,672 genes were determined to be significantly up- or down-regulated expression genes, separately. Amongst, 4,860 genes were associated with 260 pathways, contains vital enrichment pathways, such as “Metabolic”, “PI3K-Akt signaling pathway”, “Phagosome”, “Apoptosis” and “Lysosome”. In inclusion, autophagy cells were investigated by an electron microscopy, alongside further observations of the expression of autophagy-related genes in gill tissue before and after starvation. The data indicates that the amount of autophagosomes increased and found that atg2, atg6 and atg13 were significantly up-regulated. These results indicated that Mytilus may use autophagy to cope with their damage after starvation. With our research results, we have contributed to a deeper understanding of the role of the molecular mechanisms of immune defence in Mytilus under environmental stress, which has given insights into mussel breeding and the understanding of the molecular mechanisms of Mytilus immune system.

This study aimed to investigate the association between air pollutants and methylation of peripheral brain-derived neurotrophic factor (BDNF) promoters. A total of 101 individuals were recruited in this panel study. BDNF promoter methylation was detected by bisulfite-PCR amplification and pyrosequencing. Participants’ exposure to air pollutants was estimated using a satellite-based random forests approach. A generalized estimated equation model with natural cubic splines was employed to examine the associations between air pollutants and BDNF promoter methylation levels. The associations between air pollution and BDNF promoter methylation showed nonlinear curves with threshold effects. The threshold concentration for the association of nitrogen dioxide (NO2) with average methylation level was 59.7 μg/m3, and that for the association of particulate matter ≤ 1 µm in diameter (PM1) with CpG2 methylation level was 70.9 μg/m3. The percent change of average methylation level at the 95th percentile of NO2 against the threshold concentration was 43.25% (95%CI: 13.10%, 73.40%), and that of CpG2 methylation at the 95th percentile of PM1 was 128.29% (95%CI: 43.27%, 213.31%). Overall, long-term exposures of PM1, PM2.5, PM10, and NO2 were associated with significant changes in BDNF promoter methylation levels with threshold effects.

In addition to the essential pharmacological effects of opioids, situational cues associated with drug addiction memory are key triggers for drug seeking. CircRNAs, an emerging hotspot regulator in crown genetics, play an important role in central nervous system-related diseases. However, the internal mediating mechanism of circRNAs in the field of drug reward and addiction memory remains unknown. Here, we trained mice on a conditional place preference (CPP) model and collected nucleus accumbens (NAc) tissues from day 1 (T0) and day 8 (T1) for high-throughput RNA sequencing. QRT-PCR analysis revealed that circTmeff-1 was highly expressed in the NAc core but not in the NAc shell, suggesting that it plays a role in addiction memory formation. Meanwhile, the down-regulation of circTmeff-1 by adeno-associated viruses in the NAc core or shell could inhibit the morphine CPP scores. Subsequently, the GO and KEGG analyses indicated that circTmeff-1 might regulate the addiction memory via the MAPK and AMPK pathways. These findings suggest that circTmeff-1 in NAc plays a crucial role in morphine-dependent memory formation.

TC4 alloy is widely used in dental implantation due to its excellent biocompatibility and low density. However, it is necessary to further improve the corrosion resistance and surface hardness of the titanium alloy to prevent surface damage that could result in the release of metal ions into the oral cavity, potentially affecting oral health. In this study, Ti-N-O layers were fabricated on the surface of TC4 alloy using a two-step hollow cathode plasma source oxynitriding technique. This resulted in the formation of TiN, Ti2N, TiO2, and nitrogen-stabilized α(N)-Ti phases on the TC4 alloy, forming a Ti-N-O modified layer. The microhardness of the samples treated with plasma oxynitriding (PNO) was found to be 300–400% higher than that of untreated (UN) samples. The experimental conditions were set at 520 °C, and the corrosion current density of the PNO sample was measured to be 7.65 × 10−8 A/cm2, which is two orders of magnitude lower than that of the UN sample. This indicates that the PNO-treated TC4 alloy exhibited significantly improved corrosion resistance in the artificial saliva solutions.

Choroidal thickening spectrum diseases include pachychoroid pigment epitheliopathy(PPE)central serous chorioretinopathy(CSC), pachychoroid neovascularization(PNV)and polypoidal choroidal-vasculopathy(PCV). PPE refers to permanent abnormal increase in choroidal thickness. It is manifested as dilatation of large choroidal vessels(Haller layer)oppressing the surrounding middle vascular layer(Sattler layer)and capillary layer(Choriocapillaris layer), resulting in insufficient blood supply to the retinal pigment epithelium and causing a series of retinitis pigmentosa. The other three diseases can progress from the pachychoroid pigment epitheliopathy. The study of the pathogenic characteristics and imaging changes of choroidal thickening spectrum diseases will help to explore the diseases' pathogenesis, providing reference for early clinical diagnosis, prevention and treatment.

Background: Poststroke social isolation could worsen poststroke depression and dampen neurogenesis. (R,S)-ketamine has antidepressant and neuroprotective effects; however, its roles and mechanisms in social isolation–mediated depressive-like behaviors and sensorimotor recovery remain unclear. Methods: Mice were subjected to transient middle cerebral artery occlusion, and then were pair-housed with ovariectomized female mice or were housed isolated (ISO) starting at 3 days postischemia. ISO mice received 2 weeks of (R,S)-ketamine treatment starting at 14 days postischemia. Primary ependymal epithelial cells and choroid plexus epithelial cells were cultured and treated with recombinant human atrial natriuretic peptide (ANP) protein. Results: The poststroke social isolation model was successfully established using middle cerebral artery occlusion combined with poststroke isolation, as demonstrated by a more prominent depression-like phenotype in ISO mice compared with pair-housed mice. (R,S)-ketamine reversed ISO-mediated depressive-like behaviors and increased ANP levels in the atrium. The depression-like phenotype was negatively correlated with ANP levels in both the atrium and plasma. Atrial GLP-1 and GLP-1 receptor signaling was essential to the promoting effects of (R,S)-ketamine on the synthesis and secretion of ANP from the atrium in ISO mice. (R,S)-ketamine also increased ANP and TGF-β1 levels in the choroid plexus of ISO mice. Recombinant human ANP increased TGF-β1 levels in both the primarily cultured ependymal epithelial cells and choroid plexus epithelial cells. Furthermore, (R,S)-ketamine increased TGF-β1 levels in the ischemic hemisphere and promoted striatal neurogenesis and sensorimotor recovery via ANP in ISO mice. Conclusions: (R,S)-ketamine alleviated poststroke ISO-mediated depressive-like behaviors and thus promoted striatal neurogenesis and sensorimotor recovery via ANP.

Previous studies mainly focused on the relationship between the size of the prolapse and injury to the supporting tissues, but the strain and stress distributions of the supporting tissues as well as high-risk areas of injury are still unknown. To further investigate the effect of supporting tissues on organs and the interactions between organs, this study focused on the relationship between high intra-abdominal pressure and the compliance of the pelvic floor support system in a normal woman without pelvic organ prolapse (POP), using a finite element model of the whole pelvic support system. A healthy female volunteer (55 years old) was scanned using magnetic resonance imaging (MRI) during rest and Valsalva maneuver. According to the pelvic structure contours traced by a gynecologist and anatomic details measured from dynamic MRI, a finite element model of the whole pelvic support system was established, including the uterus, vagina with cavity, cardinal and uterosacral ligaments, levator ani muscle, rectum, bladder, perineal body, pelvis, and obturator internus and coccygeal muscles. This model was imported into ANSYS software, and an implicit iterative method was employed to simulate the biomechanical response with increasing intra-abdominal pressure. Stress and strain distributions of the vaginal wall showed that the posterior wall was more stable than the anterior wall under high intra-abdominal pressure. Displacement at the top of the vagina was larger than that at the bottom, especially in the anterior–posterior direction. These results imply potential injury areas with high intra-abdominal pressure in non-prolapsed women, and provide insight into clinical managements for the prevention and surgical repair plans of POP.

Recent research suggest that some non-coding RNAs (ncRNAs) are important regulators of chromatin dynamics and gene expression in nervous system development and neurological diseases. Nevertheless, the molecular mechanisms of long non-coding RNAs (lncRNAs), acting as competing endogenous RNAs (ceRNAs), underlying morphine addiction are still unknown. In this research, RNA sequencing (RNA-seq) was used to examine the expression profiles of lncRNAs, miRNAs and mRNAs on the nucleus accumbens (NAc) tissues of mice trained with morphine or saline conditioned place preference (CPP), with differential expression of 31 lncRNAs, 393 miRNAs, and 371 mRNAs found. A ceRNA network was established for reciprocal interactions for 9 differentially expressed lncRNAs (DElncRNAs), 10 differentially expressed miRNAs (DEmiRNAs) and 12 differentially expressed mRNAs (DEmRNAs) based on predicted miRNAs shared by lncRNAs and mRNAs. KEGG pathway enrichment analyses were conducted to explore the potential functions of DEmRNAs interacting with lncRNAs in the ceRNA network. These DEmRNAs were enriched in synaptic plasticity-related pathways, including pyrimidine metabolism, ECM-receptor interaction, and focal adhesion. The correlation between the relative expression of lncRNAs, miRNAs and mRNAs was analyzed to further validate predicted ceRNA networks, and the Lnc15qD3-miR-139-3p-Lrp2 ceRNA regulatory interaction was determined. These results suggest that the comprehensive network represents a new insight into the lncRNA-mediated ceRNA regulatory mechanisms underlying morphine addiction and provide new potential diagnostic and prognostic biomarkers for morphine addiction.

The aim of this study was to investigate the occurrence and molecular characteristics of Bartonella infections in small rodents in the Shangdang Basin, China. Small rodents were captured using snap traps, and their liver, spleen, and kidney tissues were harvested for Bartonella detection and identification using a combination of real-time PCR of the ssrA gene (296 bp) and conventional PCR and sequencing of the gltA gene (379 bp). Results showed that 55 of 147 small rodents to be positive for Bartonella, with a positivity rate of 37.41%, and 95% confidence interval of 29.50%- 45.33%. While the positivity rate across genders (42.62% in males and 33.72% in females, χ2 = 1.208, P = 0.272) and tissues (28.57% in liver, 33.59% in spleen, and 36.76% in kidney, χ2 = 2.197, P = 0.333) of small rodents was not statistically different, that in different habitats (5.13% in villages, 84.44% in forests, and 54.17% in farmlands, χ2 = 80.105, P

Abstract Background Neovascularization is a leading cause of visual loss typically associated with diabetic retinopathy (DR) and retinopathy of prematurity (ROP). Interleukin-17A (IL-17A) and endoplasmic reticulum (ER) stress both have been demonstrated to play a proangiogenic role in ischemic retinopathies. However, the relationship between IL-17A and ER stress in retinal neovascularization (RNV) under hypoxic conditions and its underlying mechanisms remain unclear. Methods In this study, oxygen-induced retinopathy (OIR) mice model was established and intravitreal injections were conducted. Changes of IL-17A and ER stress markers in retinas and cultured primary bone marrow derived macrophage (BMDM) under normoxic or hypoxic conditions were detected. Western blotting, Real-Time RT-PCR, Immunofluorescence assays were conducted to explore the roles and relationship of IL-17A and ER stress in RNV, as well as its underlying mechanisms. Results Compared to that in normal controls, IL-17A and ER stress markers were all remarkably increased under hypoxic conditions both in vivo and in vitro. Neutralization or knock out of IL-17A decreased ER stress. ER stress inhibitor 4-phenylbutyrate (4-PBA), attenuated the production of IL-17A, suggesting a positive feedback loop between IL-17A and ER stress. Inhibition of IL-17A or ER stress decreased areas of nonperfusion and neovascularization in OIR retinas. As TXNIP/NLRP3 pathway activation has been demonstrated to be involved in increased retinal vascular permeability of ischemic retinopathy, we observed that TXNIP/NLRP3 pathway mediated in the interaction between IL-17A and ER stress under hypoxic conditions. Conclusion The interplay between IL-17A and ER stress contributes to RNV in macrophages via modulation of TXNIP/NLRP3 signaling pathway under hypoxic conditions. The feedback loops may become an innovative and multiple pharmacological therapeutic target for ischemic retinopathy.

Fatal intoxication with sedative-hypnotic drugs is increasing yearly. However, the plasma drug concentration data for fatal intoxication involving these substances are not systematic and even overlap with the intoxication group. Therefore, developing a more precise and trustworthy approach to determining the cause of death is necessary. This study analyzed mice plasma and brainstem samples using the liquid chromatography-high resolution tandem mass spectrometry (LC-HR MS/MS)-based metabolomics method to create discriminative classification models for estazolam fatal intoxication (EFI). The most perturbed metabolic pathway between the EFI and EIND (estazolam intoxication non-death) was examined, Both EIND and EFI groups were administered 500 mg of estazolam per 100 g of body weight. Mice that did not die beyond 8 hours were treated with cervical dislocation and were classified into the EIND groups; the lysine degradation pathway was verified by qPCR (Quantitative Polymerase Chain Reaction), metabolite quantitative and TEM (transmission electron microscopy) analysis. Non-targeted metabolomics analysis with EFI were the experimental group and four hypoxia-related non-drug-related deaths (NDRDs) were the control group. Mass spectrometry data were analyzed with Compound Discoverer (CD) 3.1 software and multivariate statistical analyses were performed using the online software MetaboAnalyst 5.0. After a series of analyses, the results showed the discriminative classification model in plasma was composed of three endogenous metabolites: phenylacetylglycine, creatine and indole-3-lactic acid, and in the brainstem was composed of palmitic acid, creatine, and indole-3-lactic acid. The specificity validation results showed that both classification models distinguished between the other four sedatives–hypnotics, with an area under ROC curve (AUC) of 0.991, and the classification models had an extremely high specificity. When comparing different doses of estazolam, the AUC value of each group was larger than 0.80, and the sensitivity was also high. Moreover, the stability results showed that the AUC value was equal to or very close to 1 in plasma samples stored at 4 °C for 0, 1, 5, 10 and 15 days; the predictive power of the classification model was stable within 15 days. The results of lysine degradation pathway validation revealed that the EFI group had the highest lysine and saccharopine concentrations (mean (ng/mg) = 1.089 and 1.2526, respectively) when compared to the EIND and control group, while the relative expression of SDH (saccharopine dehydrogenase) showed significantly lower in the EFI group (mean = 1.206). Both of these results were statistically significant. Furthermore, TEM analysis showed that the EFI group had the more severely damaged mitochondria. This work gives fresh insights into the toxicological processes of estazolam and a new method for identifying EFI-related causes of mortality.

Cloud changes and their attribution under global warming still remains a challenge in climatic change studies, especially in decomposing the fast and slow cloud responses to anthropogenic forcing. In this study, the responses of global cloud cover to the quadrupled CO2 forcing are investigated quantitatively by decomposing the total response into fast and slow ones using the multi-model data from the Coupled Model Intercomparison Project Phase 6 (CMIP6). During the quasi-equilibrium period after the quadrupling of CO2 forcing, the global mean changes of simulated total cloud cover (TCC) in the total, fast, and slow responses are −2.42%, −0.64%, and −1.78%, respectively. Overall, the slow response dominates the total response in most regions over the globe with similar spatial patterns. TCC decreases at middle and low latitudes but increases at high latitudes in the total and slow responses. Whereas, it mainly decreases in the middle and low latitudes of the southern hemisphere as well as in the middle and high latitudes of the northern hemisphere in the fast response. A change in vertical motion is the major contributor to the cloud cover change at middle and low latitudes, while the decrease in upper atmospheric temperature leads to an increase in high cloud cover at high latitudes. In addition, the anomaly in water vapor convergence/diffusion also contributes to the cloud cover increase/decrease at low latitudes.

As a biomarker, alpha-fetoprotein (AFP) is valuable for detecting some tumors in men, non-pregnant women, and children. However, the detection sensitivity in some methods needs to be improved. Therefore, developing a simple, reliable, and sensitive detection method for AFP is important for non-malignant diseases. An aptamer binding was developed based on aggregation-induced emission luminogen (AIEgen) nanosphere labeled with Fe3O4@MPTMS@AuNPs. AFP was detected with a sandwich structure of AuNPs magnetic composite particles. An aggregation-induced emission (AIE) molecule and polystyrene (PS) nanosphere complex were assembled, enhancing the fluorescence and improving the sensitivity of detection. The limit of detection (LOD) was at a given level of 1.429 pg/mL, which can best be achieved in serum samples. Finally, the results obtained showed the complex to be promising in practical applications.

Abstract Tissue factor pathway inhibitors (TFPI), including TFPI-1 and TFPI-2, are Kunitz-type serine protease inhibitors that mainly inhibit the blood coagulation induced by tissue factors. Previous reports on teleost proved TFPI play important roles in innate immunity. In this study, two TFPI (PoTFPI-1 and PoTFPI-2) molecules from Japanese flounder (Paralichthys olivaceus) were analyzed and characterized for their expression patterns, antibacterial and anticancer activities of the C-terminal derived peptides. Quantitative real time RT-PCR analysis shows that constitutive PoTFPI-1 expression occurred, in increasing order, in the brain, muscle, spleen, gills, head kidney, blood, intestine, heart, and liver; PoTFPI-2 was expressed, in increasing order, in the brain, gills, head kidney, muscle, intestine, spleen, liver, heart, and blood. Under the stimulation of fish pathogens, both PoTFPI-1 and PoTFPI-2 expressions increased significantly in a manner that depended on the pathogens, tissue type, and infection stage. Furthermore, C-terminal peptides TP25 and TP26, derived from PoTFPI-1 and PoTFPI-2, respectively, were synthesized and proved to be active against Micrococcus luteus (for TP25 and TP26) and Staphylococcus aureus (for TP25) via retardation effects on bacterial nucleic acids. In addition, TP25 and TP26 also displayed significant inhibitory effects on human colon cancer cell line HT-29. These results reveal that both PoTFPI-1 and PoTFPI-2 play important roles in host innate immunity. The antibacterial activity and anticancer cells function of TP25 and TP26 will add new insights into the roles of teleost TFPI.

Abstract Anthropogenic aerosol (AA) forcing has been shown as a critical driver of climate change over Asia since the mid-20th century. Here we show that almost all Coupled Model Intercomparison Project Phase 6 (CMIP6) models fail to capture the observed dipole pattern of aerosol optical depth (AOD) trends over Asia during 2006–2014, last decade of CMIP6 historical simulation, due to an opposite trend over eastern China compared with observations. The incorrect AOD trend over China is attributed to problematic AA emissions adopted by CMIP6. There are obvious differences in simulated regional aerosol radiative forcing and temperature responses over Asia when using two different emissions inventories (one adopted by CMIP6; the other from Peking university, a more trustworthy inventory) to driving a global aerosol-climate model separately. We further show that some widely adopted CMIP6 pathways (after 2015) also significantly underestimate the more recent decline in AA emissions over China. These flaws may bring about errors to the CMIP6-based regional climate attribution over Asia for the last two decades and projection for the next few decades, previously anticipated to inform a wide range of impact analysis.

Abstract Background Sepsis is a significant cause of mortality in-hospital, especially in ICU patients. Early prediction of sepsis is essential, as prompt and appropriate treatment can improve survival outcomes. Machine learning methods are flexible prediction algorithms with potential advantages over conventional regression and scoring system. The aims of this study were to develop a machine learning approach using XGboost to predict the 30-days mortality for MIMIC-III Patients with sepsis-3 and to determine whether such model performs better than traditional prediction models. Methods Using the MIMIC-III v1.4, we identified patients with sepsis-3. The data was split into two groups based on death or survival within 30 days and variables, selected based on clinical significance and availability by stepwise analysis, were displayed and compared between groups. Three predictive models including conventional logistic regression model, SAPS-II score prediction model and XGBoost algorithm model were constructed by R software. Then, the performances of the three models were tested and compared by AUCs of the receiver operating characteristic curves and decision curve analysis. At last, nomogram and clinical impact curve were used to validate the model. Results A total of 4559 sepsis-3 patients are included in the study, in which, 889 patients were death and 3670 survival within 30 days, respectively. According to the results of AUCs (0.819 [95% CI 0.800–0.838], 0.797 [95% CI 0.781–0.813] and 0.857 [95% CI 0.839–0.876]) and decision curve analysis for the three models, the XGboost model performs best. The risk nomogram and clinical impact curve verify that the XGboost model possesses significant predictive value. Conclusions Using machine learning technique by XGboost, more significant prediction model can be built. This XGboost model may prove clinically useful and assist clinicians in tailoring precise management and therapy for the patients with sepsis-3.
