Due to the great resistance to first-line drugs, XDR-TB and MDR-TB are often difficult to cure completely. Pretomanid, PA-824 is one of the preferred drugs evaluated in clinical trials for the treatment of drug-resistant Mycobacterium tuberculosis. By inhibiting cell wall formation and respiratory toxicity, Pretomanid shows significant bactericidal activity against both replicative and stationary Mycobacterium tuberculosis. This article explores the effects of PA-824 on releasing active nitrogen substances and producing strong oxidative and cytotoxicity to Mycobacterium tuberculosis, prolonging early bactericidal activity, reducing the speed of Mycobacterium tuberculosis load in patients' sputum, and causing controllable adverse reactions, and comprehensively understand the anti-tuberculosis mechanism of pretomanid, including inhibiting the development of drug resistance of Mycobacterium tuberculosis strains, shortening the duration of tuberculosis treatment, and so on. The combination of pretomanid PA-824, bedaquinoline and linezolid, which has synergistic or additive potential in killing bacteria and inhibiting drug resistance, is also described, and is expected to further improve current TB treatment outcomes. Objective To study the relationship between the allele polymorphisms SNP-1098T > G and -590C >T in interleukin-4 gene promoter and HCV chronic infection in Han population in Yunnan province . Methods Two single nucleotide polymorphisms (SNPs) in the IL-4 promoter region, -1098T>G and-590C>T in 192 HCV chronic infected patients and 170 healthy person were determined by real-time TaqMan polymerase chain reaction. We calculated the linkage disequilibrium of the two SNPs, and then constructed haplotypes, obtained correlative data between SNP-1098T >G, -590C >T and HCV chronic infection, and the correlation between haplotype frequency and HCV chronic infection. Results IL4-1098T >G and -590C >T genotype and haplotypes frequency showed no significant association with HCV infected patients and healthy person (P > 0.05). There was linkage disequilibrium of IL-4 gene between SNP-1098T>G and -590C>T. The frequency of haplotypes constructed by SNP-1098T>G and -590C>T showed no significant difference between HCV infected patients and healthy person (P > 0.05) . Conclu sion SNPs-1098T >G and -590C >T in IL-4 gene promoter has no association with chronic HCV infection in Yunnan Han population. [ABSTRACT FROM AUTHOR]

Objective To investigate the chemical constituents from Ligusticum daucoides Franch. and detect their antibacterial activities in vitro. Methods The roots of L. daucoides were collected in Dali, Yunnan. The methanol extract of this plant was subjected to column chromatography over silica gel, Sephadex LH-20 and RP-18, further isolated by various chromatography methods. The antibacterial activities were assayed in vitro by broth dilution method. Results Nine compounds, including 8 coumarins, were isolated from the roots of this plant. Conclusions L. daucoides is rich in coumarins. The compound ferullic acid exhibits notable antibacterial activity against Staphylococcus aureus, with MIC and MBC values of 2.5 and 5.0 mg/ml, respectively. [ABSTRACT FROM AUTHOR]

Objective To investigate the change of IL-6 in Lyme arthritis. Methods Twenty female Kunming mice were selected and randomly divided into two groups. The recombinant BmpA of Borrelia burgdorferi was injected into the tibiotarsal joint of experimental group mice to induce the arthritis. ELISA experiment was used to detect the concentration of IL-6 in murine joints. Results The average concentration of IL-6 in the joint fluid of experimental mice and normal control group was 72.26 pg/mL and 31.32 pg/mL, respectively. The difference was statistically significant between two groups (P < 0.01). Conclusion IL-6 is a important inflammatory cytokine, it has obvious correlation with Lyme arthritis. Objective To study the seroprevalence of Lyme disease and scrub typhus among college freshmen in Yunnan province. Methods ELISA method was used to detect IgG antibody to Lyme disease and scrub typhus, and the antibody positive rate was calculated. Results Totally 42 college freshmen were enrolled in this study. The positive rates of IgG antibody to Lyme disease and scrub typhus were 2.4% and 4.8%,respectively. Conclusion There are Lyme disease and scrub typhus infections in Yunnan province. The surveillance,prevention and control for these insect-borne infectious diseases should be strengthened. [ABSTRACT FROM AUTHOR]

Aim: To explore the clinical significance of cytometric bead array(CBA) in detecting partial cytokines(IL-2, IL-4, IL-6, IL-10, TNF-α and IFN-γ)in the serum from patients with different types of pulmonary tuberculosis., Methods: The levels of six cytokines (IL-2, IL-4, IL-6, IL-10, TNF-α and IFN-γ) in the serum from 84 cases of patients with pulmonary tuberculosis(46 cases of active pulmonary tuberculosis patients, 38 cases of inactive pulmonary tuberculosis patients) and 30 cases of normal person, were detected by BD CBA Flex Set kit., Results: The levels of IL-2, IL-4, IL-6, IL-10, TNF-α and IFN-γ in pulmonary tuberculosis group were significantly higher than those in control group (P<0.01 or P<0.05.Compared with the inactive pulmonary tuberculosis group, the levels of serum IL-2, IL-4, IL-6, IL-10, TNF-α and IFN-γ in the active pulmonary tuberculosis group were significantly higher (P<0.01 or P<0.05).Serum IL-4 and TNF-α levels in active pulmonary tuberculosis group, inactive pulmonary tuberculosis group and the control group had not significantly different., Conclusion: Six cytokines in serum can be detected quickly and sensitively in batches at one time by CBA. IL-2, IL-4, IL-6, IL-10, TNF-α and IFN-γ play an important role in the immune pathogenesis of pulmonary tuberculosis.
