Silver nanoparticles (SNPs) were biofunctionalized using aqueous leaf extracts of G. sylvestre. The anticancer properties of the bioactive compounds and the biofunctionalized SNPs were compared using the HT29 human adenoma colon cancer cell line.Methods: The preliminary phytochemical screening for bioactive compounds from aqueous extracts revealed the presence of alkaloids, triterpenes, flavonoids, steroids, and saponins. Biofunctionalized SNPs were synthesized using silver nitrate and characterized by ultraviolet–visible spectroscopy, scanning electron microscopy, energy-dispersive X-ray analysis, Fourier transform infrared spectroscopy, and X-ray diffraction for size and shape. The characterized biofunctionalized G. sylvestre were tested for its in vitro anticancer activity against HT29 human colon adenocarcinoma cells.Results: The biofunctionlized G. sylvestre SNPs showed the surface plasmon resonance band at 430 nm. The scanning electron microscopy images showed the presence of spherical nanoparticles of various sizes, which were further determined using the Scherrer equation. In vitro cytotoxic activity of the biofunctionalized green-synthesized SNPs (GSNPs) indicated that the sensitivity of HT29 human colon adenocarcinoma cells for cytotoxic drugs is higher than that of Vero cell line for the same cytotoxic agents and also higher than the bioactive compound of the aqueous extract.Conclusion: Our results show that the anticancer properties of the bioactive compounds of G. sylvestre can be enhanced through biofunctionalizing the SNPs using the bioactive compounds present in the plant extract without compromising their medicinal properties. The aim of this study was to develop a simple biological method for the synthesis of silver and gold nanoparticles using Chrysopogon zizanioides. To exploit various plant materials for the biosynthesis of nanoparticles was considered a green technology. An aqueous leaf extract of C. zizanioides was used to synthesize silver and gold nanoparticles by the bioreduction of silver nitrate (AgNO3) and chloroauric acid (HAuCl4) respectively. Water-soluble organics present in the plant materials were mainly responsible for reducing silver or gold ions to nanosized Ag or Au particles. The synthesized silver and gold nanoparticles were characterized by ultraviolet (UV)-visible spectroscopy, scanning electron microscopy (SEM), energy dispersive X-ray analysis (EDAX), Fourier transform infrared spectroscopy (FTIR), and X-ray diffraction (XRD) analysis. The kinetics decline reactions of aqueous silver/gold ion with the C. zizanioides crude extract were determined by UV-visible spectroscopy. SEM analysis showed that aqueous gold ions, when exposed to the extract were reduced and resulted in the biosynthesis of gold nanoparticles in the size range 20–50 nm. This eco-friendly approach for the synthesis of nanoparticles is simple, can be scaled up for large-scale production with powerful bioactivity as demonstrated by the synthesized silver nanoparticles. The synthesized nanoparticles can have clinical use as antibacterial, antioxidant, as well as cytotoxic agents and can be used for biomedical applications. The reaction process was simple and easy to handle, and was monitored using ultraviolet-visible spectroscopy. The effect of the phytochemicals present in M. umbellatum, including saponins, phenolic compounds, phytosterols, and quinones, on formation of stable silver and gold nanoparticles was investigated by Fourier-transform infrared spectroscopy. The morphology and crystalline phase of the nanoparticles were determined by transmission electron microscopy and energy-dispersive x-ray spectroscopy. The results indicate that the saponins, phytosterols, and phenolic compounds present in the plant extract play a major role in formation of silver and gold nanoparticles in their respective ions in solution. The characteristics of the nanoparticles formed suggest application of silver and gold nanoparticles as chemical sensors in the future. Given the simple and eco-friendly approach for synthesis, these nanoparticles could easily be commercialized for large-scale production.Keywords: green synthesis, phytochemicals, saponins, nanoparticles, transmission electron microscopy

To our knowledge, this is the first report where M. edule leaf broth was found to be a suitable plant source for the green synthesis of silver and gold nanoparticles. On treatment of aqueous solutions of silver nitrate and chloroauric acid with M. edule leaf extract, stable silver and gold nanoparticles were rapidly formed. The gold nanoparticles were characterized by UV-visible spectroscopy, scanning electron microscopy (SEM), transmission electron microscopy (TEM), energy dispersive X-ray analysis (EDAX) and Fourier transform infra-red spectroscopy (FTIR). The kinetics of reduction of aqueous silver and gold ions during reaction with the M. edule leaf broth were easily analyzed by UV-visible spectroscopy. SEM analysis showed that aqueous gold ions, when exposed to M. edule leaf broth, were reduced and resulted in the biosynthesis of gold nanoparticles in the size range 20–50 nm. TEM analysis of gold nanoparticles showed formation of triangular, circular, and hexagonal shapes in the size range 10–45 nm. The resulting silver nanoparticles were predominantly square with uniform size range 50–90 nm. EDAX results confirmed the presence of triangular nanoparticles in the adsorption peak of 2.30 keV. Further FTIR analysis was also done to identify the functional groups in silver and gold nanoparticles. The characterized nanoparticles of M. edule have potential for various medical and industrial applications. Recently, more consumers are interested in purchasing probiotic food and beverage products that may improve their immune health. The market for functional foods and beverages that include Bifidobacterium is expanding because of their potential uses in both food and therapeutic applications. However, maintaining Bifidobacterium's viability during food processing and storage remains a challenge. Microencapsulation technique has been explored to improve the viability of Bifidobacterium . Despite the technical, microbiological, and economic challenges, the market potential for immune-supporting functional foods and beverages is significant. Additionally, there is a shift toward postbiotics as a solution for product innovation, a promising postbiotic product that can be incorporated into various food and beverage formats is also introduced in this review. As consumers become more health-conscious, future developments in the functional food and beverage market discussed in this review could serve as a reference for researchers and industrialist.

Background: Mutations affecting transcription, translation, or beta-globin stability, among other stages of beta-globin production, cause beta-thalassemia., Methods: Beta-thalassemia results in a high red blood cell count with a low mean corpuscular volume (60-70fl) and mean corpuscular hemoglobin (19-23pg), and Naked Eye Single Tube Red Cell Osmotic Fragility Test (NESTROFT) is recommended for mass screening of populations. Among tribals in Dharmapuri district, this cross-sectional analysis evaluated the efficacy of RBC and NESTROFT against HPLC, regarded as the gold standard, in a study involving 484 subjects., Results: Findings indicated that out of the 484 samples, 73 tested positive for the beta-thalassemia trait through HPLC. The NESTROFT test demonstrated 87% sensitivity, 98.5% specificity, a positive predictive value of 99.3%, and a negative predictive value of 73.3%. In the multivariate analysis, NESTROFT and hemoglobin showed high significance with P values of 0.003 and 0.000, respectively., Conclusion: NESTROFT's high sensitivity is particularly noteworthy due to the absence of research among the Malayali tribes in the Dharmapuri district. Microplastics pose a significant environmental threat, with potential implications for toxic chemical release, aquatic life endangerment, and human food chain contamination. In Asia, rapid economic growth coupled with inadequate waste management has escalated plastic pollution in rivers, positioning them as focal points for environmental concern. Despite Asia's rivers being considered the most polluted with plastics globally, scholarly attention to microplastics in the region's freshwater environments is a recent development. This study undertakes a systematic review of 228 scholarly articles to map microplastic hotspots in Asian freshwater systems and synthesize current research trends within the continent. Findings reveal a concentration of research in China and Japan, primarily investigating riverine and surface waters through net-based sampling methods. Polyethylene (PE), polypropylene (PP), and polyethylene terephthalate (PET) emerge as the predominant microplastic types, frequently observed as fibers or fragments. However, the diversity of sampling methodologies and reporting metrics complicates data synthesis, underscoring the need for standardized analytical frameworks to facilitate comparative analysis. Infections caused by antibiotic-resistant pathogens result in a delayed wound-healing process. As an approach to prevent infections, alternatives in the form of natural antimicrobial products have become public interest. Essential oils derived from plants are used as antimicrobials owing to their broad-spectrum activity against pathogenic organisms. In this study, essential oil from seeds of watermelon, jackfruit, and papaya was incorporated into poly-ε-caprolactone/gelatin nanofibers using an electrospinning technique. The synthesized nanofibers were smooth, continuous, and bead-free. The nanofibers were found to be mechanically competent as confirmed by the universal tensile tester. The antibacterial activity of the various essential oil-loaded nanofibrous mats was determined by disc diffusion assay. Furthermore, they were found to be non-toxic and biocompatible by MTT and CMFDA assays on fibroblast cells. The obtained results have demonstrated that essential oil-loaded nanofiber mats are promising alternatives to conventional antibacterial wound dressings., (© 2023. The current study aims to investigate the influence of seasonal changes on the pollution loads of the sediment of a coastal area in terms of its physicochemical features. The research will focus on analyzing the nutrients, organic carbon and particle size of the sediment samples collected from 12 different sampling stations in 3 different seasons along the coastal area. Additionally, the study discusses about the impact of anthropogenic activities such as agriculture and urbanization and natural activities such as monsoon on the sediment quality of the coastal area. The nutrient changes in the sediment were found to be: pH (7.96-9.45), EC (2.89-5.23 dS/m), nitrogen (23.98-57.23 mg/kg), phosphorus (7.75-11.36 mg/kg), potassium (217-398 mg/kg), overall organic carbon (0.35-0.99%), and sediment proportions (8.91-9.3%). Several statistical methods were used to investigate changes in sediment quality. According to the three-way ANOVA test, the mean value of the sediments differs significantly with each season. It correlates significantly with principal factor analysis and cluster analysis across seasons, implying contamination from both natural and man-made sources. The distribution of heavy metals in the seafood intake by various age group representatives around the Kalpakkam coastal region was part of the baseline study. Totally 40 different types of fish species were estimated on heavy metals (Cu, Cr, Co, Cd, Pb, Ni, Zn, and Mn) in the coastal zone; the average concentration of heavy metals were 0.71, 0.06, 0, 0, 0.07, 0.02, 1.06 and 0.36 ppm, respectively. Individual mean bioaccumulation index (IMBI) and Metal pollution index (MPI) with heavy metals distributed around the coastal zone were compared with fish tissue and were found to be higher for Zn and Cu. The human health risk was calculated using uncertainty modeling of risk assessment of Estimated daily intake (EDI), Maximum allowable consumption rate (CRlim), Target hazard quotient (THQ), and Hazard index (HI) were estimated for different age groups. Our present values were suggestively high (>1) for both kids and adults. The cumulative cancer risk assessment based on heavy metals and the Hospital-Based Cancer Registry (HBCR) compared to the region did not exceed the recommended threshold risk limit around the Kalpakkam coastal zone. The current study focused on the monitoring of pollution loads in the Kalpakkam coastal zone of India in terms of physico-chemical characteristics of sediment. The investigation took place at 12 sampling points around the Kalpakkam coastal zone for one year beginning from 2019. The seasonal change of nutrients in the sediment, such as nitrogen, phosphorus, potassium, total organic carbon, and particles size distribution, was calculated. Throughout the study period, the pH (7.55 to 8.99), EC (0.99 to 4.98 dS/m), nitrogen (21.74 to 58.12 kg/ha), phosphorus (7.5 to 12.9 kg/ha), potassium (218 to 399 kg/ha), total organic carbon (0.11 to 0.88%), and particle size cumulative percent of sediments (from 9.01 to 9.39%) was observed. A number of multivariate statistical techniques were used to examine the changes in sediment quality. The population means were substantially different according to the three-way ANOVA test at the 0.05 level. Principal component analysis and cluster analysis showed a substantial association with all indicators throughout all seasons, implying contamination from both natural and anthropogenic causes. Marine fish biodiversity and conservation linked to human livelihoods along the coast were studied in 2019 using a survey and personal interviews to investigate the effects of environmental and anthropogenic issues on changes in marine fish diversity that affect sustainability. The Zoological Survey of India (ZSI) authenticated 42 finfish, 6 crabs, and 1 shrimp from three groups collected along the coastal zone. The estimated fish species diversity index revealed that alpha and beta biodiversity were more prevalent in the study zone than gamma fish diversity. When the collected fish species were compared to the International Union for Conservation of Nature (IUCN) threatened lists, 51% were found to be least concerned. The variation in the livelihood status, age group, religious, education, houses, training, graft, and gear of fishermen was 93.6% and 4.3%, respectively, according to the principle component analysis. The observation of 51% of the least concerned species in the study zone suggests species declination as a result of overexploitation of natural resources. The current study investigated seasonal fluctuations in diversity of fish and heavy metal concentrations in coastal areas, as well as the possible human health risks associated by the heavy metals (Mercury, Lead, Chromium, Cadmium, Copper and Zinc). From five different locations across the coastal area, 44 finfish species from 11 orders and 33 families were collected. Four finfish species such as Mugil cephalus, Lates calcarifer, Etroplus suratensis, and Chanos chanos were used to estimate and assess the heavy metal concentrations based on abundance and distribution across coastal area. Results revealed that the metal concentration in these fish species, water, and sediment were all found to be significantly comparable. During the southwest monsoon season, the highest concentrations of metals were found in Chanos chanos, Mugil cephalus, and Lates calcarifer. A hazard index and a target hazard quotient were calculated to determine the human-related health risk. Radioactivity monitoring post-cold war has become more complex due to the nuclear fallout and the surge in use of radioactive materials. This requires novel methods to detect, trace and distinguish natural and anthropogenic radioactive sources in the environment. We explored the feasibility of using ZrSiO 4 (Zircon), as a reference signature for radioactivity monitoring due to the unique phenomenon of metamictization. We investigated the variations in microstructural properties of Zircon samples collected from a proposed Uranium site to identify these signatures using analytical techniques such as Gamma-ray Spectroscopy, XRD and Raman spectrum analysis. Besides elevated levels of radioactivity, the samples exhibited distinct properties such as increased lattice parameters observed from the XRD analysis and dramatic broadening of A 1g (439 cm -1 ) and B 1g (1008 cm -1 ) vibrational modes in the Raman spectrum. Structural parameters were further analyzed by modeling the crystal from experimentally observed lattice parameters. Ab-initio calculations were then performed on the modeled structure providing more insight into the microstructural variations. Samples collected from proposed Uranium mines indicate an increase of 1.226% and 0.9389% in Si-O and Zr-O bond lengths of the Zircon crystal signifying the ongoing process of metamictization from radiation damage. By correlating radioactivity levels with the lattice parameters variations of the collected samples, the study establishes a linear relation between the degree of damage to a mineral's crystal structure and the amount of radioactivity. We propose to use the variations in damage found in a mineral's structure as a nuclear forensic signature for advanced assessment of accumulated radioactivity in a particular geographical location., (Copyright © 2021 Elsevier Ltd. In this study, Polyphenols functionalized Graphitic Hematite Nanocomposite (PGHN) was used as an adsorbent to remove Caesium (Cs) ions from a simulated solution. The nanocomposite was produced by synthesizing iron oxide nanoparticles using orange peel extract (OPE) as the reducing and capping agent in the presence of graphite produced from sugarcane bagasse. The nanocomposite exhibited a scaly morphology and the mean particle size of rhombohedral structured hematite nanoparticles was found to be 148.9 nm. The simulated solution of Cs ions was treated with PGHN and the treatment conditions were optimized by batch method. The concentration of Cs ion in the treated solution was determined using atomic emission spectroscopy (AES). The maximum Cs adsorption of 97.95% was attained at an optimum condition of pH - 9.0 and adsorbent dose - 70 mg/mL for treatment period of 110 min. The experimental data of adsorption fitted well with pseudo 1st order kinetics and was favorable for both Langmuir and Freundlich isotherm models. The study reports a facile method for the production of nanocomposite using agro-wastes such as sugarcane bagasse and orange peels. Purpose: Freshwater fish Pangasius sutchi was used in this study as a vertebrate model. We evaluated the induction of certain antioxidant enzymes in various vital organs. The radioprotective efficacy of Gymnema sylvestre leaves extract (GS) [25   mg/kg Body Weight (B.W)] and its bioactive compound Gymnemagenin (GG) [0.3   mg/kg B.W] was compared with Amifostine (Ami), the only radioprotector clinically approved by the US-FDA [Ami- 83.3   mg/kg B.W] against different doses of gamma radiation - 60 Co (Lethal Dose: LD 30 -9.2   Gy, LD 50 -10.2   Gy and LD 70 -11.4   Gy)., Materials and Methods: This study was done via stress marker enzymes, cell cycle analysis (CCA) and DNA damage assay prediction with molecular docking, which are reported here for the first time. The results indicate an elevated LPO level and decreased level of CAT, SOD and GSH due to oxidative stress initiation by 60 Co Ionizing Radiation (IR) on 4th day and slightly reduced on 32nd day while the reverse observed when the fishes were pretreated with Ami, GS and GG. Similarly, CCA and dead/live cells counts were conducted with pretreatment of Ami, GS and GG against 60 Co IR dose (LD 50 -10.2   Gy)., Results: In CCA, G0/G1 phase was observed to be the highest in Ami and lowest in GG, against 60 Co IR doses 10.2   Gy which was 51.76   ±   7.55. The dead cells range observed in pretreated group of Ami, GS and GG was lowest in Ami and highest in GG and live cells (highest in Ami and lowest in GG) as compared to 60 Co IR group (86.43   ±   3.42 and 8.77   ±   5.95). Thus, antioxidant profile improvement by oxidative stress reduction and gradual progression of different phases of cell cycle except the apoptotic phase along with the live cells counts indicates that the radio-protective efficacy of GS is similar to Ami., Conclusion: Predictive assessment was carried out by docking of Ami, various components of GS with p53, NF-κβ cells and Rad51 proteins structures responsible for CCA, apoptosis and repair mechanism. These structural proteins were docked with other structural proteins like USP7, TNF-α and partner and localizer of BRCA2 associated (PALB2/BRCA2) complex which made us perform these systemic efforts to find the functional activity of these known radio-protectants.

Karankadu mangrove situated along the Southeast coast of India is known for its unique and extreme species diversity and richness. Ecological functions of this mangrove comprise of nutrient cycling, coastal protection, fish fauna production and carbon sequestration besides providing livelihood to nearby coastal communities. The current study having assessed seasonal fluctuations of various Physico-chemical factors viz., rainfall, pH, surface water temperature, salinity, dissolved nutrients in the water and sediments, primary productivity and plant pigments (chlorophylls a,b,c) during the study period from July 2018 to June 2019, recorded a total of 29 species of zooplankton, 26 species of phytoplankton, and 19 species of fish with observed maximum density at summer and pre-monsoon period of the year. The waste water released from industries which contain pollutants like heavy metals, dyes and other toxic chemicals brings numerous harms to the ecosystem and humans. Nowadays the nanocomposites based technologies are effectively used for environmental remediation. In the present study, hexavalent chromium was removed from the industrial effluent using magnetite carbon nanocomposite. The nanocomposite composed of highly porous carbon and iron oxide nanoparticles prepared by using agrowastes (sugarcane bagasse and orange peel extract). Iron oxide nanoparticles (FeONPs) formation was confirmed by UV-visible spectroscopy; incorporation of magnetite with highly porous carbon was established by Fourier Transforms Infrared Spectroscopy and X-ray Diffraction Spectroscopy. Morphological features of magnetite nanoparticles and highly porous carbon were analyzed using Scanning Electron Microscope and Transmission Electron Microscope. Magnetic properties analyzed by Vibrating Sample Magnetometer revealed magnetite carbon nanocomposite exhibited better Ms value than highly porous carbon. The concentration of Cr 6+ in treated effluent was determined using Atomic Absorption Spectroscopy. Pseudo-second order equation fitted with kinetics and the Langmuir monolayer favors for isotherm. Iron oxide nanoparticles synthesis is an expanding area of research due of their magnetic properties and possible applications in several novel technologies. FeONPs are indispensable in the biomedical field for diagnosis, treatments and drug delivery and in bioremediation applications. The synthesis route of nanoparticles is a major concern because biological methods are eco-friendly, and chemical methods are considered toxic. The objective of this study is to synthesize FeONPs by two different methods and to compare their properties and efficiency in applications. FeONPs were synthesized and characterized by microscopic and various spectroscopic techniques. The synthesized FeONPs were screened for their cytotoxic activity on PBMCs using MTT assay and found to exhibit good biocompatibility. Moreover, the GS FeONPs exhibited potential antibacterial activities and meanwhile showed less toxicity in brine shrimp lethality assay. Radiation damages genetic material, which is reflected in genotoxicity and can cause hereditary damage. In the medical field, it is essential to avoid the harmful effects of radiation. Radiation countermeasures and the need for radioprotective agents have been explored in recent years. Considering plants that evolve in radiative conditions, their ability to protect organisms against radiation has been studied and demonstrated. Crude extracts, fractioned extracts, isolated phytocompounds, and plant polysaccharides from various plants have been used in radioprotection studies, and their efficiency has been proven in various in vitro and in vivo experimental models. It is important to identify the mechanism of action to develop a potent plant-based radioprotective agent. To identify this protective mechanism, it is necessary to understand the damage caused by radiation in biological systems. This review intends to discuss the effects of ionizing radiation on biological systems and evaluate plant-based radioprotectants that have tested thus far as well as their mechanism of action in protecting against the toxic effects of radiation. From the review, the mechanism of radioprotection exhibited by the plant-based products could be understood. Burn wounds are susceptible to microbial invasion from both resident and exogenous bacteria, which becomes a critical public health issue and causes substantial economic burden. There is a perceived demand to produce new antimicrobial wound dressings that hinder bacterial colonization while accelerating the healing process and hence would provide an improved standard of care for patients. Since ancient times, herbal extracts from medicinally important plants have extensively been used for treating burn injuries. This work reports the utility of electrospun nanofibers containing plant extracts and antibiotics combination as a multifunctional scaffold for treating second-degree burns. First, we determined the various components of plant extracts from Gymnema sylvestre by two different processing methods and their synergism with minocycline antibiotics. Then, we prepared core-shell nanofibrous dressings with poly-ε-caprolactone/gelatin laden with minocycline hydrochloride as a shell and gelatin infused with G. sylvestre extracts (ultrasound-assisted extracts and cold macerated extracts) as the core using coaxial electrospinning. The electrospun nanofibers displayed a smooth, continuous, and bead-free morphology with adequate wettability. The presence of extract components in the core-shell nanofibers resulted in enhanced mechanical properties when compared to pristine mats. The core-shell structures resulted in sustained release of the bioactive components when compared to nanofiber blends. Core-shell nanofiber mats containing plant extracts and antibiotic combinations displayed potent antimicrobial and antibiofilm properties while promoting the spread and proliferation of skin cells when compared to pristine mats. In a porcine model of cutaneous second-degree burns, we showed that wounds treated with the antimicrobial dressing improved re-epithelialization and collagen organization in comparison to untreated wounds.

In the situation of radiation triage, accidental exposure to uranium, or uranium contamination in food or water; haematopoietic decline or bone marrow sickness is observed in the aftermath followed by other systemic effects. Most studies done previously have been on cytogenetic analysis in blood lymphocytes of uranium miners wherein causal relationship was difficult to be established. This study provides new insights into the minimum risk level of uranium to human lymphocytes, DNA damage induced and alterations in the cell cycle progression through 96-h acute toxicity study. Cytotoxicity studies by MTT assay and flow cytometry showed that uranyl nitrate concentration of 1280 μM lead to 50% cell death, 640 μM caused 25% death, 250 μM caused 10% cell death and 5 μM was the NOAEL. Uranium caused DNA damages in a dose dependent manner as evident from comet and CBMN assays. A marked increase in G2/M phase cells was observed in the test culture groups. Halting of cell cycle at G2/M checkpoint also signified the extent of double strand breaks and genetic instability with increasing uranium dose in this study. Better cell cycle responses and lower genetic damage index observed in lower dosage of exposure, suggests adaptability and repair responses in human lymphocytes. A study was carried out in the coastal waters of Kalpakkam with the objectives to evaluate the seasonality in hydrobiological parameters in surface and bottom waters, and assess the anthropogenic stress and monsoonal flux on a spatiotemporal scale. The study covered an area of approximately 100 km 2 in the coastal environment. Relatively high values for pH, temperature, and TP were observed during the post-monsoon (POM) season. The monsoon (MON) season was linked with TN, ammonia, and DO concentrations as all these parameters have shown increased values during this season due to freshwater input. The summer (SUM) season was characterized by salinity, turbidity, nitrate, phosphate, and silicate, indicating a true marine environmental condition for plankton production. Principal component analysis (PCA) and cluster analysis (CA) indicated the presence of distinct coastal water masses with respect to seasons and sampling regions. The spatial pattern indicated the distinctness of the coastal nearshore water (CNW) and coastal offshore water (COW) with respect to water quality. The CNW was more dynamic due to direct external influence as compared to the relatively stable COW environment. Similarly, the study region in the northern part, which is continuously exposed to the backwater inputs and tourism activities, was statistically different from the southern part.

The steady increase in the world's population has intensified the need for crop productivity, but the majority of the agricultural practices are associated with adverse effects on the environment. Such undesired environmental outcomes may be mitigated by utilizing biological agents as part of farming practice. The present review article summarizes the analyses of the current status of global agriculture and soil scenarios; a description of the role of earthworms and their products as better biofertilizer; and suggestions for the rejuvenation of such technology despite significant lapses and gaps in research and extension programs. By maintaining a close collaboration with farmers, we have recognized a shift in their attitude and renewed optimism toward nature-based green technology. Based on these relations, it is inferred that the application of earthworm-mediated vermitechnology increases sustainable development by strengthening the underlying economic, social and ecological framework.

Wound care management presents one of the substantial and tenacious challenges to the healthcare systems worldwide. Microbial colonization and subsequent biofilm formation after injury have garnered much attention, as there is an appreciable correlation between biofilms formation and delayed healing in chronic wounds. Nanotechnology has emerged as a potential platform for the management of treating acute and chronic wounds. This study presents the utility of electrospun nanofiber mats containing a natural extract (Gymnema sylvestre) that averts biofilm formation but supports human dermal fibroblasts (hDFs) attachment. The scaffolds exhibited good wettability, enhanced mechanical properties and contact mediated inhibition of Gram-positive and Gram-negative bacteria. MTS viability assay and confocal imaging further confirmed that the natural extract loaded mats remained non-cytotoxic for hDFs. Overall these findings evidenced the suitability of the Gymnema sylvestre (GS) functionalized electrospun poly-ε-caprolactone (PCL) nanofibers, as an effective wound dressing with broad spectrum anti-bacterial properties., (Copyright © 2018. Electrospun fibers have emerged as promising materials in the field of biomedicine, due to their superior physical and cell supportive properties. In particular, electrospun mats are being developed for advanced wound dressing applications. Such applications require the firers to possess excellent antimicrobial properties in order to inhibit potential microbial colonization from resident and non-resident bacteria. In this study, we have developed Poly-ε-Caprolactone /gelatin hybrid composite mats loaded with natural herbal extract ( Gymnema sylvestre ) to prevent bacterial colonization. As-spun scaffolds exhibited good wettability and desirable mechanical properties retaining their fibrous structure after immersing them in phosphate buffered saline (pH 7.2) for up to 30 days. The initial burst release of Gymnema sylvestre prevented the colonization of bacteria as confirmed by the radial disc diffusion assay. Furthermore, the electrospun mats promoted cellular attachment, spreading and proliferation of human primary dermal fibroblasts and cultured keratinocytes, which are crucial parenchymal cell-types involved in the skin recovery process. Overall these results demonstrated the utility of Gymnema sylvestre impregnated electrospun PCL/Gelatin nanofibrous mats as an effective antimicrobial wound dressing.

Exposure to ionizing radiation is harmful to any living organism. It may cause varying levels of genetic mutation or ultimately death. Synthetic compounds have been used to counteract the hazardous effect of radiation on the live cells, but the possibility of these synthetic compounds being harmful to the organism being treated also exists. Herbal formulations are thus being explored as a possible alternative for the synthetic radioprotectant. Induction of DNA damage in fishes caused by ionizing radiation and its protection by phytocompounds is a hardly studied topic. In this study, we analyzed the radioprotective effect of Gymnema sylvestre leaves extract (GS) and its active compound gymnemagenin (GG) against different doses of gamma radiation ( 60 Co) on the freshwater fish Pangasius sutchi. The radioprotective efficacy was assessed by micronuclei and alkaline comet assays. The freshwater fish P. sutchi was pre-treated with intramuscular injection (IM) of amifostine (83.3 mg/kg of B.W.), GS (25 mg/kg of B.W.) and GG (0.3 mg/kg of B.W.), 1 h prior to the gamma radiation. The fishes were exposed to LD 30 , LD 50 and LD 70 of gamma radiation and the protection activities were assessed by analyzing the number of micronuclei (MN) and erythrocytic abnormalities in the blood after 2, 4, 8, 16 and 32 days after exposure. Compared to the irradiated fishes, frequency of erythrocytic abnormalities were decreased in response to the radio-protection in the amifostine treated groups for all three doses of gamma radiation (LD 70 - 77.62%), (LD 50 - 80.11%) and (LD 30 - 82.30%); GS (LD 70 - 62.66%), (LD 50 - 69.74%) and (LD 30 - 70.81%); and GG (LD 70 - 49.42%), (LD 50 - 53.43%) and (LD 30 - 58.42%). Similarly, a significant radio-protective effect in terms of decremented DNA damage was observed using the comet assay after post exposure. The percentage of protection noted for amifostine was (LD 70 - 58.68%), (LD 50 - 64.52%) and (LD 30 - 74.40%); GS (LD 70 - 53.84%), (LD 50 - 59.02%) and (LD 30 - 65.97%); GG (LD 70 - 49.85%), (LD 50 - 52.56%) and (LD 30 - 64.30%). From the current study, we can conclude that the radioprotective efficacy of the GS is similar to the synthetic compound (amifostine) and also greater than the bioactive compound (GG). The synergetic effect of the plant extract which leads to a better protection than the bioactive compound must be further studied. MN and Comet assays can easily identify the damage due to radiation exposure and thus can be used as predictive biomarkers for aquatic organisms exposed to radiation., (Copyright © 2018 Elsevier Inc. Ten sampling locations in Nagarjuna Sagar Dam have been selected to assess the suitability of the reservoir water for human consumption. The sediment, water, and fish samples were collected and analyzed for radionuclide ( 238 U, 232 Th, 210 Po, 226 Ra, 210 Pb) and physicochemical parameters like pH, TOC, total hardness, alkalinity, DO, cation exchange capacity, and particle size. The spatial variations among the radionuclides ( 238 U, 232 Th, 210 Po, 226 Ra, 210 Pb) in water and bottom sediments of Nagarjuna Sagar Dam were determined. The uranium concentration in the sediment and water was in BDL (<0.5 ppb). The maximum permissible limits in water samples of the analyzed radionuclides are 238 U-10 Bq/l, 210 Po-0.1 Bq/l, 226 Ra-1 Bq/l, and 210 Pb-0.1 Bq/l. The radionuclides in our water samples were approximately 50 times far below the recommended limit. The ingestion of water and fish would not pose any significant radiological impact on health or cancer risk to the public, implicating that the fishes from Nagarjuna Sagar Dam reservoir are safe for human consumption except the fisherman community.

The early biomarkers for the hematological, serological and histological alterations due to the effect of ½ and ¼ LC 50 of 238 U in different organs in freshwater fish Pangasius sutchi for water-borne 238 U accumulation was investigated. The toxicological data due to 238 U accumulation on the hematological parameters such as hemoglobin (Hb), red blood cells (RBCs), white blood cells (WBCs) and hematocrit (Hct) to evaluate the oxygen carrying capacity has been indicated as the secondary response of the organisms. The biomarkers of liver damage were determined as by Serum Glutamic Oxaloacetic Transaminase (SGOT), Serum Glutamic Pyruvic Transaminase (SGPT), Alkaline Phosphatase (ALP), γ-Glutamyl Transferase (γ-GT). Similarly, the renal biomarkers of kidney damage were accessed by creatinine, uric acid, triglycerides, and cholesterol. The decrease in hemoglobin in the experimental group due to disturbed synthesis of hemoglobin was directly proportional to the concentration and exposure duration of 238 U. The histological studies proved that liver and gills are the target organ for 238 U toxicity. The extensive histological lesions were observed in various tissues due to oxidative stress by the accumulation of 238 U, and the 238 U toxicity in the organs was in the order of Gills238 U toxicity to assess fish health in Uranium ( 238 U) biomonitoring programs., (Copyright © 2017 Elsevier B.V. The strategic plan of this study is to analyze any possible radiological impact on aquatic organisms from forthcoming uranium mining facilities around the Nagarjuna Sagar Dam in the future. The predominantly consumed and dominant fish species Pangasius sutchi, which is available year-round at Nagarjuna Sagar Dam, was selected for the study. To comprehend the outcome and to understand the mode of action of 238 U, the fish species Pangasius sutchi was exposed to ¼ and ½ of the LC 50 doses of waterborne 238 U in a static system in duplicate for 21 days. Blood and organs, including the gills, liver, brain and muscles, were collected at different time periods-0h, 24h, 48h, 72h, 96h, 7, days 14days and 21 days-using ICP-MS to determine the toxic effects of uranium and the accumulation of 238 U concentrations. The bioaccumulation of 238 U in P. sutchi tissues was dependent on exposure time and concentration. The accumulation of uranium was, in order of magnitude, measured as gills>liver>brain>tissue, with the highest accumulation in the gills. It was observed that exposure to 238 U significantly reduced antioxidant enzymes such as superoxide dismutase, catalase, and lipid peroxidase. The analysis of DNA fragmentation by comet assay and cell viability by flow cytometry was performed at different time intervals. DNA histograms by flow cytometry analysis revealed an increase in the G2/M phase and the S phase. The long-term 238 U exposure studies in fish showed increasing micronucleus frequencies in erythrocytes with greater exposure time. The higher the concentration of 238 U is, the greater is the effect observed, suggesting a close relationship between accumulation and toxicity. Purpose: To study the distribution of Polonium ((210)Po) activity in dietary sources in the high background radiation zone of Puttetti in southern Tamil Nadu., Materials and Methods: (210)Po was analyzed in the food materials consumed by the male and female individual representatives living in the high background areas by 24-h Duplicate Diet Study (DDS) and Market Basket Study (MBS). The MBS was performed by collecting the food samples such as, cereals, fruits, leafy vegetables, roots and tubers, other vegetables, fish, meat and milk grown in the high background radiation zone of southern Tamil Nadu as a part of baseline study in this region. The DDS was done by collecting the food materials consumed including the beverages in 24 h from different age groups of male and female individuals living in the village of Puttetti. The intake and ingestion dose of the radionuclide (210)Po was estimated., Results: The average concentration of (210)Po in DDS (n = 33) was found to be 74 mBq.kg(- 1) of fresh weight. The MBS was collected based on food consumption representing more than 85-95% of annual supply, and were divided into eight food groups. The average concentration of (210)Po in the eight food groups namely leafy vegetables was 2176 mBq.kg(- 1) (n = 3), vegetables 55 mBq.kg(- 1) (n = 10), roots and tubers 251 mBq.kg(- 1) (n = 4), fruits 65 mBq.kg(- 1) (n = 5), fish 345 mBq.kg(- 1) (n = 2), meat food 117 mBq.kg(- 1) (n = 3), milk 20 mBq.kg(- 1) (n = 1) and cereal 290 (n = 1) mBq.kg(- 1) of fresh weight, respectively. The annual intake and ingestion dose due to (210)Po was estimated by DDS and MBS in adults, adolescents and children. The overall results showed that the MBS was moderately higher than the DDS in all age groups. Moreover, a DDS approach may even be more realistic, as cooked foodstuffs are used for dietary exposure assessment., Conclusion: The study confirms that the current levels of (210)Po do not pose a significant radiological risk to the local inhabitants.

Background: Gymnema sylvestre is a highly valued ethno pharmacologically important medicinal plant used currently in many poly-herbal formulations due to its potential antidiabetic activity and other health benefits. The present study was carried out to analyze the anti-stress, anti-allergic, and antiulcer activity of the bioactive compounds present in Gymnema sylvestre leaves., Methods: The preliminary phytochemical screening for bioactive compounds from aqueous extracts revealed the presence of alkaloids, triterpenes, flavonoids, steroids, and saponins. The antioxidant activities were investigated using DPPH radical scavenging method. The characterization of the extract was carried out using standard compound by High Performance Thin Layer Chromatography (HPTLC) and phytochemical analysis in terms of total phenol, total flavonoids, reducing power and antioxidant potentials, etc. The in vivo studies on albino mice proved the purified fraction has anti-stress/anti-allergic activity against milk induced leucocytosis/eosinophilia and able to inhibit the aspirin induced gastric ulcers., Results: The quantitative estimation for aqueous extract exhibited total antioxidant (9.13 ± 0.04 μg/g), flavonoids (125.62 ± 26.84 μg/g), tannin (111.53 ± 15.13 μg/g), total phenol content (285.23 ± 1.11 μg/g) and free radical scavenging (52.14 ± 0.32%). Further the aqueous extract was consecutively purified by TLC and silica column chromatography. The purified fractions were characterized by HPTLC and GC-MS and the component was identified as gymnemic acid. The potency of the antimicrobial activity of the extract was studied with bacteria. Pharmacological experiments clearly demonstrated that the extracts of all plants given orally showed significant gastric protection against the asprin-induced gastric ulcer model in mice. Furthermore, healing effects were also confirmed through histopathological examination., Conclusions: The aqueous extracts of the leaves of Gymnema sylvestre possess anti ulcerogenic, Anti allergic, Anti stress, properties that may be due to cytoprotective mechanism. These results support the ethno medical uses of the plant in the treatment of gastric ulcer.

The genotoxic effect of 1,2 dichlorobenzene (1,2 DCB), a volatile organic compound in the Indian Major Carp, Catla catla L. was assessed using the alkaline comet assay in the gills and blood. Fish were exposed to various sub-lethal concentrations of 1,2 DCB in vivo. At 24 h, DNA damage scores (expressed as arbitrary units) increased at 0.35 and 0.7 mg/L whereas at 28 days, there was a statistically significant increase in the DNA damage score at all the doses tested (0.175, 0.23, 0.35 and 0.7 mg/L). When the DNA damage scores were considered in the blood samples, the trend was similar to that observed in the gills - significant increase at 0.35 and 0.7 mg/L at 24 h and at all doses at 28 days. The results indicate that 1,2 DCB induces genotoxicity in the form of strand breaks in the DNA of fish as evidenced by the alkaline comet assay.

Use of plant extracts for treatment of burns and wound is a common practice followed over the decades and it is an important aspect of health management. Many medicinal plants have a long history of curative properties in wound healing. Electrospun nanofibers provide high porosity with large surface area-to-volume ratio and are more appropriate for cell accommodation, nutrition infiltration, gas exchange and waste excretion. Electrospinning makes it possible to combine the advantages of utilizing these plant extracts in the form of nanofibrous mats to serve as skin graft substitutes. In this study, we investigated the potential of electrospinning four different plant extracts, namely Indigofera aspalathoides, Azadirachta indica, Memecylon edule (ME) and Myristica andamanica along with a biodegradable polymer, polycaprolactone (PCL) for skin tissue engineering. The ability of human dermal fibroblasts (HDF) to proliferate on the electrospun nanofibrous scaffolds was evaluated via cell proliferation assay. HDF proliferation on PCL/ME nanofibers was found the highest among all the other electrospun nanofibrous scaffolds and it was 31% higher than the proliferation on PCL nanofibers after 9 days of cell culture. The interaction of HDF with the electrospun scaffold was studied by F-actin and collagen staining studies. The results confirmed that PCL/ME had the least cytotoxicity among the different plant extract containing scaffolds studied here. Therefore we performed the epidermal differentiation of adipose derived stem cells on PCL/ME scaffolds and obtained early and intermediate stages of epidermal differentiation. Background: Radiation workers are constantly exposed to low background radiation which is their occupational hazard. This continuous and prolonged exposure produces genotoxicity and cancerous condition in many workers. The authors have tested casein phosphopeptides (CPP) as a radioprotectant against low background radiation using animal models., Methods: Fermented milk was produced by addition of a bacterial culture, Lactobacillus acidophilus to a commercially available milk brand. After the fermentation process is completed in the milk, CPP is isolated from fermented milk by enzymatic hydrolysis-based method. The radioprotective role of CPP was proved using albino mice and Catla catla fish., Results: The micronucleus assay showed higher level of cell deformation and micronucleus formation in the control animal cells than the test animal cells. CPP has found to be having radioprotective activity potential., Conclusions: This radioprotective potential of CPP can be harnessed to produce formulations which can be used by radiation workers and personnel exposed to low ionization background as an occupational hazard, thus reducing the risk and preventing any type of cancer.

The purpose of this study was to determine the effect of i.t. St 587 (lumbar injection) on tail-flick (TF) latency and paw pressure (PP) withdrawal threshold in conscious rats, and the effect of i.t. (midthoracic injection) and i.v. St 587 on blood pressure in urethane-anesthetized rats. Unlike i.t. methoxamine (alpha 1 agonist), which produced antinociception, i.t. St 587 (0.5-30 micrograms) decreased TF latency and PP threshold below base line. Hyperalgesia was also produced by i.t. Wy 27127 (alpha 2-selective antagonist). At i.t. doses of St 587 > 3 micrograms, there was an apparent but incomplete return of TF latency and PP threshold toward base line. Pretreatment with i.t. prazosin (2.5 micrograms) enhanced the hyperalgesic effect of 30 micrograms, but not 1 microgram of St 587. Intrathecal St 587 and Wy 27127 each antagonized the antinociceptive effect of i.t. guanfacine (alpha 2 agonist) in the TF and PP tests. Intravenous St 587 produced a dose-dependent pressor effect that was antagonized by pretreatment with i.v. prazosin (0.14 mg/kg; 52-fold increase in the ED50), but weakly antagonized by Wy 27127 (0.5 mg/kg; 1.5-fold increase in the ED50). ST-587 also produced a dose-dependent pressor response after i.t. injection which was antagonized by i.t. prazosin (10 micrograms) or i.v. hexamethonium (10 mg/kg).(ABSTRACT TRUNCATED AT 250 WORDS)
