Abstract Seed quality is the set of physical, genetic, and physiological characteristics, reflecting the overall germination potential. Maintaining an optimal seed quality is essential for agriculture and seed banks to preserve genetic diversity. Compared to conventional methods (e.g., germination tests), non-invasive approaches allow a more sustainable and rapid evaluation of seed quality but this is limited by high costs. The measurement of ultra-weak photon emission (UPE) and delayed fluorescence (DL), defined as biological phenomena potentially related to the physiological status of living systems, may represent a suitable approach to estimate seed quality. To test this hypothesis, seeds of five agriculturally relevant legume species (Phaseolus vulgaris L., Lathyrus sativus L., Cicer arietinum L., Pisum sativum L., and Vicia faba L.), stored at different conditions (room temperature or -18 °C) for several years, were analysed using a LIANA© prototype to collect data regarding DL and UPE occurring after UV excitation. The obtained data were integrated with germination parameters which underline species-specific behaviours in response to storage conditions. The prediction models show variable efficiency in classifying seeds based on germination which underline species-dependent links between photon emission and seed quality. Therefore, these measurements represent novel, non-invasive, and rapid approaches to evaluate seed quality.

Abstract Simple physico-chemical techniques can be used to evaluate the composition, structure, and characteristics of plant seeds to determine their viability, quality, and possible uses in agriculture. Advanced analytical techniques, including thermo gravimetric analysis (TGA), electron paramagnetic resonance (EPR), and high-pressure liquid chromatography (HPLC), provide completely new insights and more precise information. They can be integrated to build up seed quality profiles, with great advantage to assess water content, organic compounds, and inorganic metals without the need to carry out many extraction procedures, as requested by more conventional methods. In this study, seed lots from three different plant species such as Triticum turgidum L. subsp. durum (wheat), Trigonella foenum graecum L. (trigonella or fenugreek), and Atriplex halimus L. (saltbush or sea orach) have been used to test the potential of TGA, EPR, and HPLC to discriminate between seed-specific features. A key finding of this study is that HPLC is essential in Principal Component Analysis (PCA) because various seeds (from the same species or other species) may contain compounds with varying polarity groups. The reported data confirm the efficacy of this approach. These data, fully available for other users, are coherently constructed and provide a proof of concept for future seed quality control studies.

In developing countries, orphan legumes stand at the forefront in the struggle against climate change. Their high nutrient value is crucial in malnutrition and chronic diseases prevention. However, as the ‘orphan’ definition suggests, their seed systems are still underestimated and seed production is scanty. Seed priming is an effective, sustainable strategy to boost seed quality in orphan legumes for which up-to-date guidelines are required to guarantee reliable and reproducible results. How far are we along this path? What do we expect from seed priming? This brings to other relevant questions. What is the socio-economic relevance of orphan legumes in the Mediterranean Basin? How to potentiate a broader cultivation in specific regions? The case study of the BENEFIT-Med (Boosting technologies of orphan legumes towards resilient farming systems) project, developed by multidisciplinary research networks, envisions a roadmap for producing new knowledge and innovative technologies to improve seed productivity through priming, with the long-term objective of promoting sustainability and food security for/in the climate-sensitive regions. This review highlights the existing drawbacks that must be overcome before orphan legumes could reach the state of ‘climate-ready crops’. Only by the integration of knowledge in seed biology, technology and agronomy, the barrier existing between research bench and local agricultural fields may be overcome, generating high-impact technical innovations for orphan legumes. We intend to provide a powerful message to encourage future research in line with the United Nations Agenda 2030 for Sustainable Development.

The purpose of this work was to investigate the production of reactive oxygen species (ROS) in Allium fistulosum seeds stored under different conditions. Optimized seed storage conditions are essential to maintain seed viability, otherwise accumulation of ROS-induced oxidative damage can lead to seed aging. The A. fistulosum seed lots used in this study have been selected based on their breeding background and reproduction site. Seed samples were stored up to 22 months under six different conditions of temperature (25, 10, and 7.5 °C) and relative humidity (RH) (25% and 45% RH). A germination test and ROS quantification assay were performed on the samples collected after 12 and 22 months of storage, respectively. Within a time-window of 10 months, the tested seed lots evidenced a decrease in the germination rate associated with increased ROS levels. Correlation analysis also showed that ROS production was influenced by genotype. The reported data showed that ROS accumulation was dependent on the storage condition and genotype. Some of the tested seed lots appeared to be prone to ROS accumulation, independent of storage conditions. On the other hand, specific condition storages (25 °C, 25% RH; 25 °C, 45% RH; 10 °C, 25% RH; 7.5 °C, 25% RH) resulted in a lower impact on seed aging.

The adaptability of seed metabolism to different environmental conditions represents a crucial aspect to understand the effects of climate change on plant populations in wild environments. Among the indicators of stress and repair in seeds, tocopherols and malondialdehyde have been related to membrane stability in seed deterioration. Alpine plants constitute an interesting system to understand stress response dynamics because of the relevant climate variations challenging seed viability in alpine environments. This study considered five accessions of Viscaria alpina seeds collected over five years, highlighting significant correlations between environmental parameters such as precipitations and temperature, and several indicators of the oxidative stress response. These provide new insights on how changes in indicators of the seed stress response can reflect annual variations in temperature and precipitations affecting their parental plants, with possible implications on the current understanding of seed persistence in alpine environments threatened by climate change and on the effects of seed storage.

Water stress brought about by climate change is among the major global concerns threatening food security. Rice is an important staple food which requires high water resources. Being a semi-aquatic plant, rice is particularly susceptible to drought. The aim of this work was to develop techniques directed to promote rice resilience to water deprivation stress during germination by implementing specific seed priming treatments. Five popular Italian rice varieties were subjected to priming treatments using novel, sustainable solutions, like poly-gamma-glutamic acid (γ-PGA), denatured γ-PGA (dPGA), and iron (Fe) pulsing, alone or in combination. The effect of the developed priming methods was tested under optimal conditions as well as under water deprivation stress imposed by polyethylene glycol (PEG) treatments. The priming efficacy was phenotypically determined in terms of germination behavior by measuring a series of parameters (germinability, germination index, mean germination time, seed vigor index, root and shoot length, germination stress tolerance index). Biochemical analyses were carried out to measure the levels of iron uptake and accumulation of reactive oxygen species (ROS). Integrative data analyses revealed that the rice varieties exhibited a strong genotype- and treatment-specific germination behavior. PEG strongly inhibited germination while most of the priming treatments were able to rescue it in all varieties tested except for Unico, which can be defined as highly stress sensitive. Molecular events (DNA repair, antioxidant response, iron homeostasis) associated with the transition from seed to seedling were monitored in terms of changes in gene expression profiles in two varieties sensitive to water deprivation stress with different responses to priming. The investigated genes appeared to be differentially expressed in a genotype-, priming treatment-, stress- and stage-dependent manner. The proposed seed priming treatments can be envisioned as sustainable and versatile agricultural practices that could help in addressing the impact of climate challenges on the agri-food system.

Seed priming is routinely applied to improve germination rates and seedling establishment, but the decrease in longevity observed in primed seeds constitutes a major drawback that compromises long-term storability. The optimization of priming protocols able to preserve primed seeds from aging processes represents a promising route to expand the scope of seed priming. The present work explores this possibility in the model legume Medicago truncatula by testing the effectiveness of quercetin- and rutin-supplemented seed priming at improving the response to subsequent artificial aging. In comparison with a non-supplemented hydropriming protocol, supplementation with quercetin or rutin was able to mitigate the effects of post-priming aging by increasing germination percentage and speed, improving seed viability and seedling phenotype, with consistent correlations with a decrease in the levels of reactive oxygen species and an increase in antioxidant potential. The results suggest that quercetin and rutin can reduce the effects of post-priming aging by improving the seed antioxidant profiles. The present work provides novel information to explore the physiological changes associated with seed priming and aging, with possible outcomes for the development of tailored vigorization protocols able to overcome the storability constrains associated with post-priming aging processes.

Plant-based biostimulants (PBs), agents rich in bioactive compounds, are emerging as key players able to sustainably improve plant growth and crop productivity to address food security. PBs are generally applied as foliar spray or soil irrigation, while more recently, the application as seed priming treatments is being envisaged as a highly sustainable method to also improve seed quality and germination. Therefore, this review proposes to explore the use of PBs for the seeds industry, specifically discussing about the relevance of product market values, sustainable methods for their production, why and how PBs are used for seed priming, and pinpointing specific strengths and challenges. The collected research studies indicate that PBs applied to seeds result in improved germination, seedling growth, and stress tolerance, although the molecular mechanisms at work are still largely overlooked. The high variability of bioactive molecules and used sources point towards a huge reservoir of nature-based solutions in support of sustainable agriculture practices.

IntroductionSeveral molecular aspects underlying the seed response to priming and the resulting vigor profile are still poorly understood. Mechanisms involved in genome maintenance deserve attention since the balance between stimulation of germination and DNA damage accumulation versus active repair is a key determinant for designing successful seed priming protocols.MethodsChanges in the Medicago truncatula seed proteome were investigated in this study, using discovery mass spectrometry and label-free quantification, along the rehydration-dehydration cycle of a standard vigorization treatment (hydropriming plus dry-back), and during post-priming imbibition.Resuts and discussionFrom 2056 to 2190 proteins were detected in each pairwise comparison, among which six were differentially accumulated and 36 were detected only in one condition. The following proteins were selected for further investigation: MtDRP2B (DYNAMIN-RELATED PROTEIN), MtTRXm4 (THIOREDOXIN m4), and MtASPG1 (ASPARTIC PROTEASE IN GUARD CELL 1) showing changes in seeds under dehydration stress; MtITPA (INOSINE TRIPHOSPHATE PYROPHOSPHORYLASE), MtABA2 (ABSCISIC ACID DEFICIENT 2), MtRS2Z32 (SERINE/ARGININE-RICH SPLICING FACTOR RS2Z32), and MtAQR (RNA HELICASE AQUARIUS) that were differentially regulated during post-priming imbibition. Changes in the corresponding transcript levels were assessed by qRT-PCR. In animal cells, ITPA hydrolyses 2’-deoxyinosine triphosphate and other inosine nucleotides, preventing genotoxic damage. A proof of concept was performed by imbibing primed and control M. truncatula seeds in presence/absence of 20 mM 2’-deoxyinosine (dI). Results from comet assay highlighted the ability of primed seeds to cope with dI-induced genotoxic damage. The seed repair response was assessed by monitoring the expression profiles of MtAAG (ALKYL-ADENINE DNA GLYCOSILASE) and MtEndoV (ENDONUCLEASE V) genes that participate in the repair of the mismatched I:T pair in BER (base excision repair) and AER (alternative excision repair) pathways, respectively.

Little progress has been made in translating the increased awareness of neglected and underutilized crops into a viable tool for advancing not just sustainable development in rural areas but also sustainable agricultural and secured food systems. This is caused, in part, by a lack of understanding the importance of several underutilized crops that are currently in cultivation. Among underutilized crops, grasspea (Lathyrus sativus) is an orphan legume crop with great agronomic potential due to its suitability for a variety of sustainable food and feed production systems throughout the world’s semi-arid and arid regions and its high protein content. The current study presents a SWOT (strengths, weaknesses, opportunities, and threats) analysis to unravel the following: What are the strengths and weaknesses of Lathyrus spp. cultivation, what factors are opportunities or initiatives that encourage the adoption of the crop by growers, and what factors are disincentives or threats that prevent its adoption? What strategies could help expand cultivation? We use data from a questionnaire survey of farmers in Central Greece (Thessaly) to examine their attitudes and perceptions with regards to the above questions. The outcome of this study strongly suggests that farmers are willing to adopt the new Lathyrus spp. cultivation if they are assisted via contractualization, training, and new financial tools.

ROS homeostasis is crucial to maintain radical levels in a dynamic equilibrium within physiological ranges. Therefore, ROS quantification in seeds with different germination performance may represent a useful tool to predict the efficiency of common methods to enhance seed vigor, such as priming treatments, which are still largely empirical. In the present study, ROS levels were investigated in an experimental system composed of hydroprimed and heat-shocked seeds, thus comparing materials with improved or damaged germination potential. A preliminary phenotypic analysis of germination parameters and seedling growth allowed the selection of the best-per-forming priming protocols for species like soybean, tomato, and wheat, having relevant agroeconomic value. ROS levels were quantified by using two noninvasive assays, namely dichloro-dihydro-fluorescein diacetate (DCFH-DA) and ferrous oxidation-xylenol orange (FOX-1). qRT-PCR was used to assess the expression of genes encoding enzymes involved in ROS production (respiratory burst oxidase homolog family, RBOH) and scavenging (catalase, superoxide dismutase, and peroxidases). The correlation analyses between ROS levels and gene expression data suggest a possible use of these indicators as noninvasive approaches to evaluate seed quality. These findings are relevant given the centrality of seed quality for crop production and the potential of seed priming in sustainable agricultural practices.

Cultivation of onion and onion-like crops has become a challenge in the context of climate change and innovative solutions are needed to support breeders as well as farmers, starting from the crucial issues of seed quality. Seed priming is routinely used to enhance germination performance and seedling resilience. Although common for radish, tomato, carrot or head cabbage seeds, the technique has been less popular for onion-like crops. This review provides a state-of-the-art picture of the scientific and technological advances that have so far contributed to enhance seed germination and vigour in onion-like crops. A brief description of the different types of priming approaches is provided whereas attention is focused on the following: (i) the impact of seed priming in terms of improved germination, seedling development and resilience to environmental stresses; (ii) the concept of seed quality hallmarks translated from model plants to the genus Allium; (iii) the drawbacks that currently impair the fully exploitation of seed priming in this specific sector of the agrifood chain.

One of the challenges that living organisms face is to promptly respond to genotoxic stress to avoid DNA damage. To this purpose, all organisms, including plants, developed complex DNA damage response (DDR) mechanisms. These mechanisms are highly conserved among organisms and need to be finely regulated. In this scenario, microRNAs (miRNAs) are emerging as active players, thus attracting the attention of the research community. The involvement of miRNAs in DDR has been investigated prominently in human cells whereas studies in plants are still scarce. To experimentally investigate the involvement of plant miRNAs in the regulation of DDR-associated pathways, an ad hoc system was developed, using the model legume Medicago truncatula. Specific treatments with camptothecin (CPT) and/or NSC120686 (NSC), targeting distinct components of DDR, namely topoisomerase I (TopI) and tyrosyl-DNA phosphodiesterase 1 (TDP1), were used. Phenotypic (germination percentage and speed, seedling growth) and molecular (cell death, DNA damage, and gene expression profiles) analyses demonstrated that the imposed treatments impact DDR. Our results show that these treatments do not influence the germination process but rather inhibit seedling development, causing an increase in cell death and accumulation of DNA damage. Moreover, treatment-specific changes in the expression of suppressor of gamma response 1 (SOG1), master-regulator of plant DDR, were observed. Additionally, the expression of multiple genes playing important roles in different DNA repair pathways and cell cycle regulation were differentially expressed in a treatment-specific manner. Subsequently, specific miRNAs identified from our previous bioinformatics approaches as putatively targeting genes involved in DDR processes were investigated alongside their targets. The obtained results indicate that under most conditions when a miRNA is upregulated the corresponding candidate target gene is downregulated, providing an indirect evidence of miRNAs action over these targets. Hence, the present study extends the present knowledge on the information available regarding the roles played by miRNAs in the post-transcriptional regulation of DDR in plants.

Seed priming can circumvent poor germination rate and uniformity, frequently reported in eggplant (Solanum melongena L.) and its crop wild relatives (CWRs). However, there is still a gap of knowledge on how these treatments impact the pre-germinative metabolism in a genotype- and/or species-dependent manner. The CWR Solanum villosum Miller (hairy nightshade) investigated in this study showed a quite unique profile of fast germination. Although this accelerated germination profile would not apparently require further improvement, we wanted to test whether priming would still be able to impact the pre-germinative metabolism, eventually disclosing the predominant contribution of specific antioxidant components. Hydropriming followed by dry-back resulted in synchronized germination, as revealed by the lowest MGR (Mean Germination Rate) and U (Uncertainty) values, compared to unprimed seeds. No significant changes in ROS (reactive oxygen species) were observed throughout the treatment. Increased tocopherols levels were detected at 2 h of hydropriming whereas, overall, a low lipid peroxidation was evidenced by the malondialdehyde (MDA) assay. Hydropriming resulted in enhanced accumulation of the naturally occurring antioxidant phenolic compounds chlorogenic acid and iso-orientin, found in the dry seeds and ex novo accumulation of rutin. The dynamic changes of the pre-germinative metabolism induced by hydropriming are discussed in view of future applications that might boost the use of eggplant CWRs for breeding, upon upgrade mediated by seed technology.

Seed priming protocols implement incomplete imbibition phases, as well as physical, chemical or biological treatments, to activate pre-germinative metabolism and stress response, thus improving germination performances, seedling establishment and stress tolerance according to agricultural productivity requirements. The dehydration phase following priming treatments represents a critical variable, since an excessively prolonged imbibition (overpriming) impairs desiccation tolerance, compromising seed viability and seedling establishment. Priming protocols generally optimize imbibition-dehydration timing empirically to avoid overpriming. Hence, a better understanding of the dynamics underlying the loss of desiccation tolerance represents a promising route to test and develop efficient and cost-effective priming techniques. In the present work, priming and overpriming conditions were defined to explore the role of desiccation tolerance in seed priming efficiency in the model legume Medicago truncatula. The positive effects of hydropriming and kinetin-mediated hormopriming on germination parameters were screened in combination with conditions of short/prolonged priming and mild/severe overpriming. Biometric analyses highlighted contrasting responses in terms of germination performances and seedling development, while ROS (reactive oxygen species) levels measured during dehydration positively correlate with the loss of desiccation tolerance in early seedlings, suggesting possible applications to monitor priming progression and predict overpriming occurrence.

Trehalose, a non-reducing disaccharide with multiple functions, among which source of energy and carbon, stress protectant, and signaling molecule, has been mainly studied in relation to plant development and response to stress. The trehalose pathway is conserved among different organisms and is composed of three enzymes: trehalose-6-phosphate synthase (TPS), which converts uridine diphosphate (UDP)-glucose and glucose-6-phosphate to trehalose-6-phosphate (T6P), trehalose-6-phosphatase (TPP), which dephosphorylates T6P to produce trehalose, and trehalase (TRE), responsible for trehalose catabolism. In plants, the trehalose pathway has been mostly studied in resurrection plants and the model plant Arabidopsis thaliana, where 11 AtTPS, 10 AtTPP, and 1 AtTRE genes are present. Here, we aim to investigate the involvement of the trehalose pathway in the early stages of seed germination (specifically, seed imbibition) using the model legume Medicago truncatula as a working system. Since not all the genes belonging to the trehalose pathway had been identified in M. truncatula, we first conducted an in silico analysis using the orthologous gene sequences from A. thaliana. Nine MtTPSs, eight MtTPPs, and a single MtTRE gene were hereby identified. Subsequently, the expression profiles of all the genes (together with the sucrose master-regulator SnRK1) were investigated during seed imbibition with water or stress agents (polyethylene glycol and sodium chloride). The reported data show a temporal distribution and preferential expression of specific TPS and TPP isoforms during seed imbibition with water. Moreover, it was possible to distinguish a small set of genes (e.g., MtTPS1, MtTPS7, MtTPS10, MtTPPA, MtTPPI, MtTRE) having a potential impact as precocious hallmarks of the seed response to stress. When the trehalose levels were measured by high-performance liquid chromatography, a significant decrease was observed during seed imbibition, suggesting that trehalose may act as an energy source rather than osmoprotectant. This is the first report investigating the expression profiles of genes belonging to the trehalose pathway during seed imbibition, thus ascertaining their involvement in the pre-germinative metabolism and their potential as tools to improve seed germination efficiency.

During seed imbibition, lipids are engaged in membrane reorganization while facing free radical-mediated oxidative injury. In the present work, we explored changes in lipid components at different timepoints of imbibition (0.5, 2, 4, 6, and 8 h) in the legume Medicago truncatula, by combining biochemical approaches with targeted lipidomics and untargeted metabolomics. ROS and RNS (reactive oxygen and nitrogen species) accumulation was observed throughout the tested timepoints whereas lipid peroxidation increased at 4 h of imbibition. The seed response to oxidative damage was evidenced by a significant increase in tocopherols starting from 0.5 h of imbibition as well as by the reduction in total thiol content occurring at 2 h of imbibition. Since under physiological conditions, the proper functions of the cell membranes are strongly dependent on the qualitative and quantitative balance of fatty acid residues in phospholipids, the investigation was expanded to the fatty acid cohort of M. truncatula seeds. Total saturated fatty acids (SFAs), monounsaturated fatty acids (MUFAs), polyunsaturated fatty acids (PUFAs), omega(ω)-3 and omega(ω)-6 fatty acids showed fluctuations during seed imbibition. The most remarkable finding was the profile of the ω-3 PUFA docosopentaenoic acid (DPA, 7 cis, 10 cis, 13 cis, 16 cis, and 19 cis-22:5) that showed a peak (up to 1.0% of the total fatty acid content) at 0.5 and 8 h of imbibition, concomitant with the peaks observed in tocopherol levels. It is possible that the observed changes in DPA alter the physical properties of membranes, as reported in animal cells, triggering signaling pathways relevant for the cell defense against oxidative injury. Furthermore, the content and balance between tocopherols and PUFAs is regarded as a determinant of storage stability. No enhancement in trans-fatty acids occurred throughout imbibition, suggesting for a proper antioxidant response carried by the seed. Fatty acids profiles were integrated with data from untargeted metabolomics showing changes in lipid sub-pathways, among which fatty acid amide, lyso-phospholipids, and phospholipid metabolism. The emerging lipid profiles and dynamics are discussed in view of the overall imbibition damage generated during M. truncatula seed imbibition.

Plants are continuously faced with complex environmental conditions which can affect the oxidative metabolism and photosynthetic efficiency, thus leading to the over-production of reactive oxygen species (ROS). Over a certain threshold, ROS can damage DNA. DNA damage, unless repaired, can affect genome stability, thus interfering with cell survival and severely reducing crop productivity. A complex network of pathways involved in DNA damage response (DDR) needs to be activated in order to maintain genome integrity. The expression of specific genes belonging to these pathways can be used as indicators of oxidative DNA damage and effective DNA repair in plants subjected to stress conditions. Managing ROS levels by modulating their production and scavenging systems shifts the role of these compounds from toxic molecules to key messengers involved in plant tolerance acquisition. Oxidative and anti-oxidative signals normally move among the different cell compartments, including the nucleus, cytosol, and organelles. Nuclei are dynamically equipped with different redox systems, such as glutathione (GSH), thiol reductases, and redox regulated transcription factors (TFs). The nuclear redox network participates in the regulation of the DNA metabolism, in terms of transcriptional events, replication, and repair mechanisms. This mainly occurs through redox-dependent regulatory mechanisms comprising redox buffering and post-translational modifications, such as the thiol-disulphide switch, glutathionylation, and S-nitrosylation. The regulatory role of microRNAs (miRNAs) is also emerging for the maintenance of genome stability and the modulation of antioxidative machinery under adverse environmental conditions. In fact, redox systems and DDR pathways can be controlled at a post-transcriptional level by miRNAs. This review reports on the interconnections between the DDR pathways and redox balancing systems. It presents a new dynamic picture by taking into account the shared regulatory mechanism mediated by miRNAs in plant defense responses to stress.

Roses (Rosa hybrida) are the most important ornamental cut-flowers and breeders’ main focus is to develop new desirable modern cultivars. Rose breeding programs center on the introduction of new flower colors, thornless stems, higher production and good post-harvest performance. The study of the main pollen traits, such as pollen quantity and quality, viability, longevity, morphological homogeneity, germination and tube growth, is important for building suitable breeding programs. Recently, a number of studies have shown that reactive oxygen species, like hydrogen peroxide, and nitric oxide, are involved in a wide range of signaling processes including pollen tube growth and pollen-pistil recognition. Pollen viability after anther dehiscence is crucial for successful crossbreeding. In the present work, pollen grains from 5 hybrid tea rose cultivars were stored at -20 °C up to 12 months. Pollen viability and germination rate was monitored in order to provide useful information about pollen storage length. Additionally, pollen grains were tested for their content in hydrogen peroxide and nitric oxide by using a novel approach where the fluorescence is read in a quantitative RealTime PCR (qRT-PCR) machine. Pollen viability and in vitro pollen germination capacity varied among the rose genotypes, while a progressive decrease was evidenced during 12 months of storage at low temperature. Both hydrogen peroxide and nitric oxide production were found to be genotypedependent, whilst accumulation of the two molecules was observed during the storage period. A putative detrimental effect of these molecules during pollen conservation is hypothesized.

The role of tyrosyl-DNA phosphodiesterase 2 (Tdp2) involved in the repair of 5′-end-blocking DNA lesions is still poorly explored in plants. To gain novel insights, Medicago truncatula suspension cultures overexpressing the MtTdp2α gene (Tdp2α-13C and Tdp2α-28 lines, respectively) and a control (CTRL) line carrying the empty vector were investigated. Transmission electron microscopy (TEM) revealed enlarged nucleoli (up to 44% expansion of the area, compared to CTRL), the presence of nucleolar vacuoles, increased frequency of multinucleolate cells (up to 4.3-fold compared to CTRL) and reduced number of ring-shaped nucleoli in Tdp2α-13C and Tdp2α-28 lines. Ultrastructural data suggesting for enhanced nucleolar activity in MtTdp2α-overexpressing lines were integrated with results from bromouridine incorporation. The latter revealed an increase of labeled transcripts in both Tdp2α-13C and Tdp2α-28 cells, within the nucleolus and in the extra-nucleolar region. MtTdp2α-overexpressing cells showed tolerance to etoposide, a selective inhibitor of DNA topoisomerase II, as evidenced by DNA diffusion assay. TEM analysis revealed etoposide-induced rearrangements within the nucleolus, resembling the nucleolar caps observed in animal cells under transcription impairment. Based on these findings it is evident that MtTdp2α-overexpression enhances nucleolar activity in plant cells.

This work provides novel insights into the effects caused by the histone deacetylase inhibitor trichostatin A (TSA) during Medicago truncatula seed germination, with emphasis on the seed repair response. Seeds treated with H2O and TSA (10 and 20 μM) were collected during imbibition (8 h) and at the radicle protrusion phase. Biometric data showed delayed germination and impaired seedling growth in TSA-treated samples. Comet assay, performed on radicles at the protrusion phase and 4-days old M. truncatula seedlings, revealed accumulation of DNA strand breaks upon exposure to TSA. Activation of DNA repair toward TSA-mediated genotoxic damage was evidenced by the up-regulation of MtOGG1(8-OXOGUANINE GLYCOSYLASE/LYASE) gene involved in the removal of oxidative DNA lesions, MtLIGIV(LIGASE IV) gene, a key determinant of seed quality, required for the rejoining of DNA double strand breaks and TDP(TYROSYL-DNA PHOSPHODIESTERASE) genes encoding the multipurpose DNA repair enzymes tyrosyl-DNA phosphodiesterases. Since radical scavenging can prevent DNA damage, the specific antioxidant activity (SAA) was measured by DPPH (1,1-diphenyl-2-picrylhydrazyl) and Folin-Ciocalteu reagent assays. Fluctuations of SAA were observed in TSA-treated seeds/seedlings concomitant with the up-regulation of antioxidant genes MtSOD(SUPEROXIDE DISMUTASE, MtAPX(ASCORBATE PEROXIDASE) and MtMT2(TYPE 2 METALLOTHIONEIN). Chromatin remodeling, required to facilitate the access of DNA repair enzymes at the damaged sites, is also part of the multifaceted seed repair response. To address this aspect, still poorly explored in plants, the MtTRRAP(TRANSFORMATION/TRANSACTIVATION DOMAIN-ASSOCIATED PROTEIN) gene was analyzed. TRRAP is a transcriptional adaptor, so far characterized only in human cells where it is needed for the recruitment of histone acetyltransferase complexes to chromatin during DNA repair. The MtTRRAP gene and the predicted interacting partners MtHAM2 (HISTONE ACETYLTRANSFERASE OF THE MYST FAMILY) and MtADA2A (TRANSCRIPTIONAL ADAPTOR) showed tissue- and dose-dependent fluctuations in transcript levels. PCA (Principal Component Analysis) and correlation analyses suggest for a new putative link between DNA repair and chromatin remodeling that involves MtOGG1 and MtTRRAP genes, in the context of seed germination. Interesting correlations also connect DNA repair and chromatin remodeling with antioxidant players and proliferation markers.

In the cut flower market, traditional breeding is still the best way to achieve new rose cultivars. The geographical delocalization of cultivar constitution (generally made in Europe and North America) and plant cultivation (large areas in Africa and South America) represents a limit point for crossing and selection. Rose breeders often need to overcome geographical distances, resulting in asynchrony in flowering among crossing parents, by storing and sending pollen. Hence, a key aspect in breeding programs is linked to pollen availability and conservation, jointly with the identification of parameters related to pollen fertility. In this study we present the results of three different trials. In the first, pollen diameter and pollen viability were chosen as fertility predictors of 10 Rosa hybrida commercial cultivars. In the second trial, aliquots of dried pollen grains of six R. hybrida cultivar were stored under two different temperatures (freezer at T = −20 °C and deep freezer at T = −80 °C) and after a wide range of conservation period, their viability was measured. In the third trial, the effective fertilization capacity of frozen pollen of 19 pollen donor cultivars was evaluated during 2015 crossing breeding plan, performing 44 hybridizations and correlating the number of seeds and the ratio seeds/crossing, obtained by each cultivar, with in vitro pollen germination ability.
