In diabetes, increased accumulation of sorbitol has been associated with diabetic complications through polyol pathway. Aldose reductase (AR) is one of the key factors involved in reduction of glucose to sorbitol, thereby its inhibition is important for the management of diabetic complications. In the present study, a series of seven 4-oxo-2-thioxo-1,3-thiazolidin-3-yl acetamide derivatives 3(a–g) were synthesized by the reaction of 5-(4-hydroxy-3-methoxybenzylidene)-4-oxo-2-thioxo-1,3-thiazolidin-3-yl acetic acid (2a) and 5-(4-methoxybenzylidene)-4-oxo-2-thioxo-1,3-thiazolidin-3-yl acetic acid (2b) with different amines. The synthesized compounds 3(a–g) were investigated for their in vitro aldehyde reductase (ALR1) and aldose reductase (ALR2) enzyme inhibitory potential. Compound 3c, 3d, 3e, and 3f showed ALR1 inhibition at lower micromolar concentration whereas all the compounds were more active than the standard inhibitor valproic acid. Most of the compounds were active against ALR2 but compound 3a and 3f showed higher inhibition than the standard drug sulindac. Overall, the most potent compound against aldose reductase was 3f with an inhibitory concentration of 0.12 ± 0.01 µM. In vitro results showed that vanillin derivatives exhibited better activity against both aldehyde reductase and aldose reductase. The molecular docking studies were carried out to investigate the binding affinities of synthesized derivatives with both ALR1 and ALR2. The binding site analysis of potent compounds revealed similar interactions as were found by cognate ligands within the active sites of enzymes.

An efficient, zirconium-catalyzed conversion of unprotected aldose sugars with acetylacetone to polyhydroxyalkyl furans or C-glycosylfurans is reported. The furan products are formed in up to 93% yield using 5-10 mol % ZrCl4. Pentoses are readily converted at room temperature, while hexoses and their oligosaccharides require mild heating (i.e., 50 °C). Efficient conversions of glycolaldehyde, glyceraldehyde, erythrose, a heptose, and glucosamine are also demonstrated. This approach outpaces each of the previous Lewis acid-catalyzed methods in at least one the following ways: (i) lower catalyst loadings; (ii) reduced reaction temperatures; (iii) shorter reaction times; (iv) equimolar substrate stoichiometry; (v) expanded sugar scope; (vi) higher selectivities; and (vii) the use of an Earth-abundant Zr catalyst.

The 1H and 13C NMR spectroscopy was used to study the structure of previously unknown aldose series condensation products (L-fucose, L-rhamnose, D-mannose, D-galactose, D-glucose, N-acetyl-D-glucosamine, N-acetyl-D-mannosamine, D-lactose and D-maltose) with 6-mercapto­hexanoic acid hydrazide—promising glycoligands of noble metal nanoparticles. It was shown that L-fucose, L-rhamnose, D-mannose, D-galactose and N-acetyl-D-mannosamine derivatives exist in solution in DMSO-d6 as a tautomeric mixture of open hydrazone and cyclic pyranose forms. The linear hydrazone form is represented by a set of Z′,E′-conformational isomers, which differ in the arrangement of substituents relative to the C–N amide bond in comparable amounts. The condensation products obtained on the basis of D-glucose, N-acetyl-D-glucosamine, D-lactose and D-maltose in the crystalline state and in solutions in DMSO-d6 have an exclusively cyclic pyranose structure represented by α,β-configurational isomers. A similar transition to the pyranose form is observed in solutions of all the studied compounds in D2O.

Advanced Glycation End Products (AGEs) could be formed with α-dicarbonyl compounds as intermediates; however, simultaneous determination of AGES as well as their precursors and intermediates have rarely been mentioned, so as to investigate the reaction. In this work, lysine was reacted with aldoses (galactose, glucose) and ketoses (fructose, sorbose) to investigate the effects of sugar type and heating temperature. Two high-performance liquid chromatography-tandem mass spectrometry methods were developed to quantify AGEs and the corresponding α-dicarbonyl compounds. The results demonstrated that aldoses were more prone to form intermediates and final products than ketoses from the point of view of absorbance (at 294 and 420 nm). Aldose systems were prone to form glyoxal, whereas ketose systems tended to generate methylglyoxal and 3-deoxyglucosone. The relative reaction activity of sugars in forming Nε-carboxymethyl-lysine depended upon heating temperature for aldose and ketose systems. The aldose was more liable to generate Nε-Carboxyethyl-lysine (CEL) than ketose, with the CEL generation for each sugar occurred in the order galactose > glucose > sorbose > fructose. The obvious differences between different types of sugars suggest that further research is needed on the degradation of sugars to form intermediates and on the reaction of α-dicarbonyl to generate AGEs.

Chemoselective reactions can contribute to streamlining synthesis of pharmaceuticals, agrochemicals, and other functional molecules by avoiding use of protecting groups. In this review, copper catalysts were demonstrated useful for developing two types of chemoselective reactions: C-C bond-forming reactions at an anomeric carbon of unprotected aldoses and difunctionalization reaction of C-C multiple bonds. The "soft" nucleophilic copper species exhibit orthogonal reactivity toward "hard" polar functional groups and preferentially react with "soft" functional groups. The catalysis also controls stereoselectivity and/or regioselectivity to provide value-added products from readily available feedstock compounds.

Unprotected aldoses in water undergo an isomerization reaction via a radical pathway when irradiated with light in the presence of water-soluble benzophenone. Whereas its anomeric carbon (C1) is oxidized to a carboxy group, the hydroxy group on the C2 carbon is replaced by hydrogen. The generated 2-deoxy lactones are readily reduced to the corresponding 2-deoxy aldoses, which are often contained in bioactive compounds.

The acyclic form of monosaccharides exists in a structural equilibrium, with aldose having the aldehyde group and ketose the ketone group (ketose-aldose equilibrium). A basic catalyst facilitates their transformation, which affects the chemical properties of the monosaccharide. In this study, we investigated the ketose-aldose transformation of 1,3-dihydroxyacetone (1,3-DHA), one of the simplest systems of the ketose-aldose equilibrium. We examined the effects of piperidine as the basic catalyst and used IR electroabsorption spectroscopy to study the responses to an external electric field. We analyzed the changes in IR absorption by considering the changes in the molecular orientation and number of molecules in response to the external electric field. Poly(ionic) liquid (PIL) augmented membranes were fabricated through self-polymerization of 2-vinyl pyridine and 4-vinyl pyridine followed by dopamine triggered polymerization and bridging with inert polyamide support. The resulting membranes acquired a positive surface charge with a high degree of hydrophilicity. Fourier transformed Infra-red (FTIR) and Energy dispersive X-ray (EDX) spectroscopic investigation revealed the successful augmentation of PIL surface layer, whereas surface morphology was investigated through scanning electron microscopy (SEM) imaging. This manuscript demonstrates pi electron-induced separation of dyes with the trend in permeability: Coomassie Brilliant Blue G (CBBHG) &gt, Remazol Brilliant Blue R (RBBR) &gt, Eichrome Black T (EBT) &gt, Congo Red (CR). CBBG exhibited extended conjugation over large aromatic domain. RBBR and EBT were associated withtheelectron-donating -NH2 group and electron-withdrawing -NO2 group, respectively, hence pi electron density on aromatic ring varied. The steric repulsion between two pairs of ortho hydrogens (Hs) in biphenyl moieties of CR resulted in deviation of planarity and hence aromaticity leading to the lowest permeability. The sugar fractionation followed the trend: Galactose &gt, Mannose &gt, Fructose &gt, Glucose &gt, Xylose. More hydroxyl (-OH) groups in sugars and their conformational alignment in the same direction, exhibited more lone pair of electrons leading to more interaction with PIL and hence better permeability. Pentose showed poorer permeation than hexose, whereas aldose showed better permeation than ketose.

d -Mannose is the aldose isomer of d -fructose and displays unique physiological functions and health applications. As a result, it has attracted increasing interest from the public. Because of its wide substrate specificity, d -lyxose isomerase (D-LI) has been applied to d -mannose bioproduction. In this article, the Thermoflavimicrobium dichotomicum D-LI encoding gene was cloned and overexpressed in Escherichia coli BL21(DE3). The novel protein T. dichotomicum D-LI was identified and characterized, and its maximum enzyme activity was exhibited at pH 7.5 (phosphate buffer) and 60 °C in the presence of Mn2+. Similar to other D-LIs, T. dichotomicum D-LI formed a homodimeric structure and showed strict conservation of the metal coordination and substrate binding sites. The D-LI half-life (t1/2) was 9.91 h and 3.05 h at 55 and 60 °C, respectively, and its melting temperature (Tm) was 72.85 °C. T. dichotomicum D-LI displayed the highest specific activity towards d -lyxose among the substrates tested, followed by d -mannose. It also efficiently converted d -fructose to d -mannose, and the equilibrium ratio between d -mannose and d -fructose during T. dichotomicum D-LI activity was 25:75. From 500 g/L d -fructose, 110.5 g/L d -mannose was produced after 6 h, suggesting that T. dichotomicum D-LI might provide an alternative for d -mannose production.

New sugar hydrazones incorporating furan and/or 1,3,4-thiadiazole ring systems were synthesized by reaction of the corresponding hydrazide with different aldose sugars. Heterocyclization of the formed hydrazones afforded the derived acyclic nucleoside analogues possessing the 1,3,4-oxadiazoline as modified nucleobase via acetylation followed by the heterocyclization process. The anticancer activity of the synthesized compounds was studied against human liver carcinoma cell (HepG-2) and at human normal retina pigmented epithelium cells (RPE-1). High activities were revealed by compounds 3, 12 and 14 with IC50 values near to that of the reference drug doxorubicin.

Nanomaterials possessing enzyme-like activity have been extensively studied owing to their high stability and tunable catalytic properties. In this work, a simple method has been developed for the synthesis of porous manganese oxide/manganese ferrite (MnOx/MnFe2O4) nanopopcorns (MFNPs) in neutral media. The MFNPs exhibit dual enzymatic activities towards selective oxidation of ketoses followed by H2O2-induced decline of its catalytic activity. MFNPs, with MnFe2O4 as the core material and an outer layer rich in MnOx, were synthesized from ammonium iron(III) citrate and potassium permanganate at 70 °C for 12 h followed by annealing at 300 °C for 6 h. The nanozyme, MFNPs, exhibited oxidase-like activity, which was proved by the oxidation of amplex red (AR) in the presence of dissolved oxygen in the solution, to form fluorescent resorufin. The activity of MFNPs is highly suppressed by H2O2 as a result of its induced dissolution of MnOx. In addition, MFNPs having catalytic activity towards the selective oxidation of ketoses (e.g., fructose) followed by the formation of H2O2. The as-formed H2O2 diminished the catalytic activity of MFNPs for the AR oxidation to form fluorescent resorufin. Upon increasing fructose concentration, the fluorescence of resorufin decreases. Since the MFNPs do not show catalytic activity towards aldose sugars, such as glucose, sucrose, and mannose, the AR/MFNPs probe has high selectivity and sensitivity for detection of fructose with a limit of detection of 32 μM. Our study shows its great potential for quantitation of fructose in honey samples.

A procedure has been developed for the synthesis of previously unknown aldose 4-[(ω-sulfanylalkyl) oxy]benzoylhydrazones (where alkyl is hexyl or decyl and aldoses are D-glucose, D-galactose, D-maltose, and D-lactose) that a repromising glycoligands for noble metal nanoparticles. According to the 1H and 13C NMR data, 4-[(ω-sulfanylalkyl)oxy]benzoylhydrazones derived from D-glucose, D-maltose, and D-lactose in crystal and in DMSO-d6 solution have exclusively the cyclic pyranose structure (α- and β-anomers). D-Galactose 4-[(ω-sulfanylalkyl)oxy]benzoylhydrazones in DMSO-d6 solution exist as tautomeric mixtures of cyclic pyranose and open-chain acylhydrazone structures.

Methanol extract of Indigofera hirsuta, was evaluated for its antiradical potential and capacity in inhibiting lipoxygenase and aldose/aldehyde reductase enzymes. The ethyl acetate fraction derived from the methanol extract partition, showed the greatest antioxidant capacity, while the butanol was the strongest inhibitor of lipoxygenase enzyme. All fractions (diethyl ether, ethyl acetate, butanol and the aqueous residue) exhibited strong inhibition capacity of both aldose/aldehyde reductase enzymes, which comes in agreement with the ethnomedicinal plant utilization as an antidiabetic agent. LC-DAD-MS(ESI+) fraction analysis verified the findings above, leading to a conclusion regarding the biological activities attributed to the main compounds. Phytochemical analysis led to the identification of an indolic dimer, cinnamic acids, phenolics, flavonoid glycosides, a cyclic polyol, the rare sugar 1-methyl-β-D-glucopyranoside and glycerol. Many of these compounds were isolated for the first time in Indigofera species while the indolic dimer was isolated for the first time in the Fabaceae family.

Alcohols have a wide range of applicability, and their functions vary with the carbon numbers. C6 and C4 alditols are alternative of sweetener, as well as significant pharmaceutical and chemical intermediates, which are mainly obtained through the fermentation of microorganism currently. Similarly, as a bulk chemical, C2 alditol plays a decisive role in chemical synthesis. However, among them, few works have been focused on the chemical production of C4 alditol yet due to its difficult accumulation. In this paper, under a static and semi‐flowing procedure, we have achieved the product control during the conversion of C6 aldose toward C6 alditol, C4 alditol and C2 alditol, respectively. About C4 alditol yield of 20 % and C4 plus C6 alditols yield of 60 % are acquired in the one‐pot conversion via a cascade retro‐aldol condensation and hydrogenation process. Furthermore, in the semi‐flowing condition, the yield of ethylene glycol is up to 73 % thanks to its low instantaneous concentration., The product control of C6 aldose toward C6, C4 and C2 alditols have been achieved, which have depended on the well‐matched between retro‐aldol condensation and hydrogenation process driving by temperature, H2 pressure and injection modes.

Several microorganisms can utilize l-rhamnose as a carbon and energy source through the nonphosphorylative metabolic pathway, in which l-rhamnose 1-dehydrogenase (RhaDH) catalyzes the NAD(P)+ -dependent oxidization of l-rhamnose to l-rhamnono-1,4-lactone. We herein investigated the crystal structures of RhaDH from Azotobacter vinelandii in ligand-free, NAD+ -bound, NADP+ -bound, and l-rhamnose- and NAD+ -bound forms at 1.9, 2.1, 2.4, and 1.6 A resolution, respectively. The significant interactions with the 2'-phosphate group of NADP+ , but not the 2'-hydroxyl group of NAD+ , were consistent with a preference for NADP+ over NAD+ . The C5-OH and C6-methyl groups of l-rhamnose were recognized by specific residues of RhaDH through hydrogen bonds and hydrophobic contact, respectively, which contribute to the different substrate specificities from other aldose 1-dehydrogenases in the short-chain dehydrogenase/reductase superfamily.

The transformation of aldose-based carbohydrates into 5-ethoxymethylfurfural (EMF) is very challenging as compared to ketose-based carbohydrates, but the formers are more abundant and cheaper. Here, a series of sulfated complex metal oxides were synthesized for the conversion of aldose-based mono-, di-, and poly-saccharides, as well as starchy food waste into EMF. The catalysts were carefully characterized and the results showed that the type and strength of the acid sites were more important than their concentration. It was also shown that the efficiency of these catalysts was significantly affected by the metal species in the catalyst composition and followed the order tetra- > tri- > bi- > mono-component metal oxides based catalyst. Among the prepared catalysts, Zr-Sn-Fe-Al-O-S exhibited superior catalytic activity, with an EMF yield of 33.1% from glucose, and yields ranging from 4.1−26.3% for di-, poly-saccharides and starchy food waste in ethanol/dimethyl sulfoxide solvent system under glucose/catalyst mass ratio of 4. The role of co-solvent in the reaction pathway was also studied. It was found that the predominant reaction pathway for EMF production was closely related to the co-solvent amount. A kinetic model of glucose conversion to EMF was developed and the thermodynamic analysis was performed, the main features of the experimental observations can be described by the model. Zr-Sn-Fe-Al-O-S was reused for four runs without intermediate regeneration steps, showing a slight decay in activity. After reactivation by calcination before the fifth cycle, the catalyst recovered its activity, indicating good reusability and thermal stability.

Poly-trans-[(2-carboxyethyl)germasesquioxane], Ge-132, is a water-soluble organogermanium compound reported to have physiological effects such as immunostimulatory and antiviral effects. The hydrolysate of Ge-132, 3-(trihydroxygermyl)propanoic acid (THGP), can interact with diols; therefore, it likely can interact with diol-containing sugars in sugar chains, glycoproteins, and glycolipids, which have important physiological functions. In this study, we quantitatively assessed the ability of THGP to interact with saccharides using nuclear magnetic resonance (NMR) spectroscopy and THGP derivatives. THGP was complexed by binding its trihydroxy group with saccharides in aqueous solutions via the cis-diol group rather than the trans-diol group. The spectra of THGP and monosaccharides indicated that THGP has a higher affinity for ketose than aldose. Moreover, the complexation ability between THGP and saccharides was influenced by the number of cis-diol groups on the saccharide structure. Thus, interactions of THGP with important biological sugars might be involved in the physiological functions of Ge-132.

Water enriched with oxygen-18 (H2 18 O) is a potential tracer for evaluating the sources of glucose and glycogen synthesis since it is incorporated into specific sites of glucose-6-phosphate via specific enzyme-mediated exchange/addition mechanisms. Unlike 2 H, 18 O does not experience significant isotope effects for any of these processes. Therefore, H2 18 O might provide more precise estimates of endogenous carbohydrate synthesis compared with deuterated water provided that positional 18 O enrichments of glucose can be measured. As a proof of concept, H2 18 O was incorporated into a well characterized hemolysate model of sugar phosphate metabolism and 13 C NMR was applied to quantify positional 18 O enrichment of glucose-6-phosphate oxygens. Human erythrocyte hemolysate preparations were incubated overnight at 37 °C with a buffer containing sugar phosphate precursors and 20% (n = 5) and 80% (n = 1) H2 18 O. Enrichment of glucose-6-phosphate was analyzed by 13 C NMR analysis of 18 O-shifted versus unshifted signals following derivatization to monoacetone glucose (MAG). 13 C NMR MAG spectra from hemolysate revealed resolved 18 O-shifted signals in Positions 1-5. Mean 18 O enrichments were 16.4 ± 1.6% (Position 1), 13.3 ± 1.3% (Position 2), 4.1 ± 1.1% (Position 3), 12.6 ± 0.8% (Position 4), 10.7 ± 1.4% (Position 5), and no detectable enrichment of Position 6. No 18 O-shifted glucose-6-phosphate signals were detected in preparations containing sugar phosphate precursors only. H2 18 O is incorporated into Positions 1-5 of glucose-6-phosphate in accordance with spontaneous aldose hydration and specific enzymatic reaction mechanisms. This provides a basis for its deployment as a tracer for glucose and glycogen biosynthesis.

Transketolase catalyzes the transfer of a glycolaldehyde residue from ketose (the donor substrate) to aldose (the acceptor substrate). In the absence of aldose, transketolase catalyzes a one-substrate reaction that involves only ketose. The mechanism of this reaction is unknown. Here, we show that hydroxypyruvate serves as a substrate for the one-substrate reaction and, as well as with the xylulose-5-phosphate, the reaction product is erythrulose rather than glycolaldehyde. The amount of erythrulose released into the medium is equimolar to a double amount of the transformed substrate. This could only be the case if the glycol aldehyde formed by conversion of the first ketose molecule (the product of the first half reaction) remains bound to the enzyme, waiting for condensation with the second molecule of glycol aldehyde. Using mass spectrometry of catalytic intermediates and their subsequent fragmentation, we show here that interaction of the holotransketolase with hydroxypyruvate results in the equiprobable binding of the active glycolaldehyde to the thiazole ring of thiamine diphosphate and to the amino group of its aminopyrimidine ring. We also show that these two loci can accommodate simultaneously two glycolaldehyde molecules. It explains well their condensation without release into the medium, which we have shown earlier.

The effects of cell voltage and of concentration of sugars (glucose and xylose) on the performances of their electro-reforming have been evaluated at a Pd3Au7/C anode in 0.10 mol L&minus, 1 NaOH solution. The catalyst synthesized by a wet chemistry route is first comprehensively characterized by physicochemical and electrochemical techniques. The supported catalyst consists in alloyed Pd3Au7 nanoparticles of circa 6 nm mean diameter deposited on a Vulcan XC72 carbon support, with a metal loading close to 40 wt%. Six-hour chronoamperometry measurements are performed at 293 K in a 25 cm2 electrolysis cell for the electro-conversion of 0.10 mol L&minus, 1 and 0.50 mol L&minus, 1 glucose and xylose at cell voltages of +0.4 V, +0.6 V and +0.8 V. Reaction products are analyzed every hour by high performance liquid chromatography. The main products are gluconate and xylonate for glucose and xylose electro-reforming, respectively, but the faradaic yield, the selectivity and the formation rate of gluconate/xylonate decrease with the increase of aldose concentration, whereas lower faradaic yields and higher formation rates of gluconate/xylonate are observed at +0.8 V than at +0.4 V (higher chemical yields).

The conversion of abundant hexoses (e.g. glucose, mannose and galactose) and pentoses (e.g. xylose and arabinose) to 5-hydroxymethylfurfural (5-HMF) and 2-furfural (2-F) is subject to intensive research in the hope of achieving competitive production of diverse materials from renewable resources. However, the abundance of literature on this topic as well as the limited number of studies systematically comparing numerous monosaccharides hinder progress tracking. Herein, we compare and rationalize reactivities of different ketoses and aldoses. Dehydration mechanisms of both monosaccharide types are reviewed regarding the existing experimental evidence. Ketose transformation to furan derivatives likely proceeds through cyclic intermediates and is hindered by side-reactions such as isomerization, retro-aldol reactions and polymerization. Different strategies can improve furan derivative synthesis from ketoses: limiting the presence of water, improving the dehydration rate, protecting 5-HMF and 2-F reactive moieties with derivatization or solvent interactions and extracting 5-HMF and 2-F from the reaction medium. In contrast to ketoses, aldose conversion to furan derivatives is not favored compared to polymerization reactions because it involves their isomerization or a ring contraction. Enhancing aldose isomerization is possible with metal catalysts (e.g. CrCl3) promoting a hydride shift mechanism or with boric/boronic acids promoting an enediol mechanism. This catalysis is however far more challenging than ketose dehydration because catalyst activity depends on numerous factors: Bronsted acidity of the medium, catalyst ligands, catalyst affinity for monosaccharides and their accessibility to several chemical species simultaneously. Those aspects are methodically addressed to support the design of new monosaccharide dehydration systems.

In this work various amount of phosphomolybdic acid (PMo) were immobilized on amine functionalized SBA-15 and used as heterogeneous catalysts in the epimerization of glucose in aqueous solution. 13.3PMo/NH2-SBA-15 exhibited the best catalytic performance with a glucose conversion of 34.8% and mannose selectivity of 85.6% within two hours at 120 &deg, C. The activation energy of 80.1 &plusmn, 0.1 kJ&middot, mol&minus, 1 was lower than that of 96 kJ&middot, 1 over the homogeneous H3PMo12O40 catalyst. The catalytic activities of 13.3PMo/NH2-SBA-15 for the transformation of some other aldoses including mannose, arabinose and xylose were also investigated.

A novel group of carbohydrate derivatives is described that uniquely assign cis/ trans-2,3-aldose stereoisomers at low nanomolar concentrations. Aldopentoses, aldohexoses, or component aldoses from hydrolysis of polysaccharides or oligosaccharides react with cysteamine in pyridine to give quantitative formation of thiazolidines, which are subsequently peracetylated in a one-pot reaction. The nonpolar thiazolidines peracetate (TPA) derivatives are analyzed by gas chromatography and electron impact mass spectrometry (GC/EI-MS), each aldose giving rise to two TPA geometric isomers. The quantitative ratio of these diastereomers is dependent upon whether the parent monosaccharide is cis-2,3-(Rib, Lyx, Man, All, Gul, and Tal), or trans-2,3-aldose (Xyl, Ara, Glc, Gal, Ido, and Alt). TPAs generate observed EI-MS fragment ions characteristic of C1-C2 and C3-C4 bond cleavage of the parent sugars. This has been used to estimate the extent of metabolic labeling of microbial cell-wall carbohydrates, especially into the defining anomeric carbons and during aldolase / ketolase -catalyzed rearrangements.

This study characterizes the saccharide sugar contained in whole sweet corn seeds by extracting using Ultrasound Assisted Solvent Extraction for 60 minutes with 24 kHz frequency using ethanol solvent with concentrations of 70%, 75%, 80%, and 85%. The concentrated extract was then characterized by using Gas Chromatography Mass Spectrometry (GCMS) method. Sample test conducted were tested to odor, color and shelf life of extract and determination of water content, ash content, and characterization of saccharide sugar from GCMS test. The odor and color test results show that the extract had a distinctive smell of sweet corn, yellow, and had the ability to store good extract in the condition of the room. The water content of each sample ranged from 3% - 9% with the lowest water content was in 85% concentration sample which was 3.62%, while the ash content ranged from 1.5% with the highest ash content which was in 85% concentration sample which was 1.59%. The results of characterization were identified by 3 compounds having the highest percentage of 2-Furaldehyde, Hexamethyl Cyclotriloxane, and 1,2,4-Trimethyl Benzene with the percentage of 26,94%, 9,95% and 13,82% respectively. 2-Furaldehyde includes heterocyclic aldehyde group sugars, whereas Hexamethyl Cyclotriloxane and 1,2,4-Trimethyl Benzene belong to the class of organosilicon and aromatic hydrocarbons. 2-Furaldehyde or Furfural serves as a tongue nerve stimulator and has great potential to be developed as an important non-petroleum-based chemical raw material. The GCMS results provide information that the obtained 2-Furaldehyde has a molecular formula C5H4O2 or C4H3OCHO which is a monosaccharide group which is aldose with the number of C atoms classified as pentose. The retention time was 3.062 minutes with a mass peak of 300 m/z with molecular weight was 96 g/mol.

d-Mannose is not only the epimer of d-glucose at the C-2 position, but also the aldose isomer of d-fructose. Because of its physiological properties and health benefits, d-mannose has attracted public interest. It has been confirmed that d-mannose has broad applications in food, cosmetics, and pharmaceutical industries. According to the Izumoring strategy, d-glucose isomerase (d-GI) and d-lyxose isomerase (d-LI) play important roles in the conversions of d-fructose from d-glucose and of d-mannose from d-fructose respectively. In this study, a one-step enzyme process of d-mannose production from d-glucose has been constructed by co-expression of the d-GI from Acidothermus cellulolyticus and d-LI from Thermosediminibacter oceani in Escherichia coli BL21(DE3) cells.; Results: The co-expression system exhibits maximum activity at pH 6.5 and 65 °C with Co2+ supplement. It is relatively thermostable at less than 65 °C. When the reaction reaches equilibrium, the ratio of d-glucose, d-fructose, and d-mannose is approximately 34 : 49.6 : 16.4. By using this co-expression system, about 60.0 g L-1 d-mannose is obtained from 400 g L-1 d-glucose in 8 h.; Conclusion: This co-expression of d-GI and d-LI system provides a novel and efficient approach for d-mannose production. © 2018 Society of Chemical Industry.; © 2018 Society of Chemical Industry.

The MgO/NaY catalysts prepared by impregnation method were used for the conversion of glucose to fructose in water medium. The effects of MgO loading, reaction temperature, glucose concentration and reaction time on the catalytic performance for the reaction were studied. The activity testing results indicated that fructose could be generated effectively by controlling the components of the catalyst and reaction conditions. The maximal fructose yield of 33.8% with the selectivity of 67.3% was achieved over the 10%MgO/NaY catalyst at 100 °C for 2 h. Moreover, the catalysts were characterized by XRD, BET, and CO2-TPD techniques. The structural property of NaY with higher surface area facilitated glucose conversion, and the modulated basicity of the catalyst with MgO addition contributed to the formation of fructose in the tautomerization of aldose to ketose.
