New tetrahydroisoquinoline-5,7-dicarbonitrile derivatives have been synthesized by piperazine hydrate-catalyzed reaction of acetoacetanilide with 2-(1-arylethylidene)malononitriles (Knoevenagel condensation products of acetophenones and malononitrile) in aqueous ethanol at room temperature. The product structure was confirmed by NMR spectra and X-ray analysis of 1,6-diamino-4a-methyl-3-oxo-2,8-diphenyl-2,3,4,4a-tetrahydroisoquinoline-5,7-dicarbonitrile.

Polysubstituted phthalimide derivatives have been isolated in excellent yield from the addition reaction between acetoacetanilide and dialkyl acetylenedicarboxylates by using DABCO as an efficient basic catalyst in EtOH at room temperature. This procedure led to the formation of new phthalimide derivative in the presence of di-tert-butylacetylendicarboxylate as acetylenic ester. The methodology has also been applied successfully for the synthesis of novel piperidine substitutes via the simple reaction between acetoacetanilide and alkyl propiolates. These reactions have some advantages such as not only the process is eco-friendly but also the yields are good and the work-up is simple.

The kinetics of oxidation of acetoacetanilide by potassium hexacyanoferrate(III) at a constant ionic strength of 0.5 mol dm−3 in aqueous alkaline medium has been studied successfully. The reaction follows less than unit order kinetics with respect to acetoacetanilide and hydroxide ion concentrations. The effect of dielectric constant and ionic strength was investigated. Activation parameters with respect to slow step were determined and discussed. Thermodynamic quantities were also determined. The final product of the reaction has been recognised as a dimer with the evidence of LCMS, UV–Vis, and IR spectra. Based on the experimental results and product analysis a mechanism involving free radicals was proposed.

Chloroacetic acid immobilized on chitosan (CS) coated iron oxide decorated by silver nanoparticles (Fe3O4@CS@Ag@CH2COOH) was synthesized as a biocompatible magnetic material. The Fe3O4@CS@Ag@CH2COOH nanocomposite was characterized using FT-IR, XRD, SEM, EDS, and TGA instruments. The surface morphology and size of Fe3O4@CS@Ag@CH2COOH nanocomposite were determined through SEM micrographs analysis. Moreover, magnetic characterization of the prepared nanocomposite was determined by VSM. The produced Fe3O4@CS@Ag and Fe3O4@CS@Ag@CH2COOH nanocomposites were screened for their antibacterial activity against gram-negative Escherichia coli and gram-positive Staphylococcus aureus. The results showed that the Fe3O4@CS@Ag and Fe3O4@CS@Ag@CH2COOH nanocomposites presented good antibacterial performance toward gram-negative Escherichia coli and gram-positive S. aureus. Furthermore, Fe3O4@CS@Ag@CH2COOH nanoparticles catalyzed one-pot synthesis of hexahydroquinoline-3-carboxamide derivatives by four-component reaction of arylaldehydes, dimedone, acetoacetanilide and ammonium acetate in ethanol at 70 °C.

Michael addition of various substituted benzylidene- and thiophen-2-ylmethylidenemalononitriles with acetoacetanilide gave the corresponding 5-acetyl-2-amino-4-aryl(hetaryl)-6-oxo-1-phenyl-1,4,5,6-tetrahydropyridine-3-carbonitriles. Treatment of the latter with ethylenediamine in boiling methanol resulted in their rearrangement with elimination of the acetyl group and formation of 2-anilino-4-aryl(hetaryl)-6-oxo-1,4,5,6-tetrahydropyridine-3-carbonitriles. A clean, facile and catalyst-free four-component domino reaction of aromatic aldehyde, ethyl cyanoacetate, acetoacetanilide and ammonium acetate in ethanol under microwave irradiation (MWI) conditions is described. A convenient and straightforward acid-promoted synthesis of a new series of polyfunctionalized pyrroles has been developed based on a one-pot three-component reaction of acetoacetanilide, benzoin, and ammonium acetate/diverse anilines in acetic acid as a green solvent at 100 °C. The new nickel coordination polymer [Ni(HL1) (CH3OH)2]n (1) and co-crystal H3L1∙DMF (2) were obtained by reaction of NiCl2·2H2O with 2-(2-(1,3-dioxo-1-(phenylamino)butan-2-ylidene) hydrazineyl)benzoic acid (H3L1) in the presence and absence of triethylamine, respectively, in a mixture of methanol and dimethylformamide (DMF) (20:1, v/v). Reaction of CuCl2·2H2O with H3L1 in the presence of 2,2'-(methylazanediyl)bis (ethan-1-ol) (HR) in methanol yields [Cu(HR)2(H2L1)2] (3), whereas treatment of this copper salt with sodium 2-(2-(1,3-dioxo-1-(phenylamino)butan-2-ylidene)hydrazineyl)benzenesulfonate (NaH2L2) in a mixture of acetone and water (10:1, v/v) produces [Cu(HL2) (H2O)2]∙2(H2O)∙{(CH3)2C = O} (4). Compounds 1−4 were characterized by IR and ESI-MS spectroscopies, elemental and X-ray crystal structural analyses, and tested as catalyst for the Henry reaction of aliphatic and aromatic aldehydes with nitroethane in different solvents such as acetonitrile, methanol or water. Good yields (up to 87%) and diasteroselectivities (syn/anti 77:23) were observed in the reactions catalyzed by 3 in water.

A variety of novel heteroannulated pyrano[3,2-c]quinolines 2–12 was efficiently synthesized via a domino ‘Michael/retro-Michael/nitrile addition/heterocyclization’ reactions between 6-ethyl-5,6-dihydro-4,5-dioxo-4H-pyrano[3,2-c]quinoline-3-carbonitrile (1) and a diversity of carbon nucleophilic reagents. Pyrido[3′,2′:5,6]pyrano[3,2-c]quinolines 2–6 were synthesized from ring opening ring closure reactions of carbonitrile 1 with some methylene active nitrile namely malononitrile, cyanoacetamide, N-phenylcyanoacetamide, (phenylthio)acetonitrile and ethyl cyanoacetate, respectively. Reactions of carbonitrile 1 with dimer malononitrile and cyanoacetohydrazide showed different behavior producing the novel heteroannulated pyranoquinoline derivatives 7 and 8, respectively. Treatment of carbonitrile 1 with some methylene active ketones namely acetylacetone, acetoacetanilide, ethyl acetoacetate and ethyl benzoylacetate afforded pyrido[3′,2′:5,6]pyrano[3,2-c]quinolines 9–12, respectively. Structures of the synthesized products were deduced on the basis of their analytical and spectral data.

An integrated platform consisting of a microfluidic paper-based analytical device (μPAD) and a portable detection system is proposed for low concentration formaldehyde (CH 2 O) detection. In the proposed approach, the reaction zone of the μPAD is coated with ammonium acetate (AA) and acetoacetanilide (AAA) indicator and is dried at 40 °C for 30 min. The formaldehyde sample is then dripped on the reaction zone and the μPAD is transferred to a hot plate in the portable detection system. A Hantzsch reaction is induced by heating the μPAD at 30 °C for 10 min. The resulting fluorescent formaldehyde-Acetoacetanilide complex (dihydropyridine derivative) is observed by a CMOS camera and the color image is transferred to a smartphone by means of a connector. Finally, the CH 2 O concentration is derived using self-written RGB color analysis mobile software implemented on the phone. The experimental results obtained using control samples with known CH 2 O concentrations in the range of 0.2–2.5 ppm show that the B (blue) fluorescence intensity (Y) and formaldehyde concentration (X) are related as Y = 29.455 X + 117.64. Moreover, the correlation coefficient is equal to R 2 = 0.9937. The real-world applicability of the proposed detection platform is demonstrated by measuring the CH 2 O concentration in twelve commercial food samples. It is shown that the concentration measurements obtained using the proposed system deviate by no more than 6.2% from those obtained using a conventional spectrophotometric technique. Overall, the results presented in this study show that the proposed integrated microfluidic paper-based system provides a compact and reliable tool for low concentration CH 2 O measurement purposes.

The new mononuclear nickel(II) complexes [Ni(HL 1 )(H 2 O) 2 (CH 3 OH)] ( 1 , NaH 2 L 1 = sodium ( Z )-2-(2-(1,3-dioxo-1-(phenylamino)butan-2-ylidene)hydrazinyl)benzenesulfonate) and [Ni(H 2 L 2 ) 2 (H 2 O) 4 ] [(CH 3 ) 2 S O) 2 ] ( 2 , H 3 L 2 = (Z)-2-(2-(1,3-dioxo-1-(phenylamino)butan-2-ylidene)hydrazinyl)benzoic acid) were synthesized and characterized by IR and ESI-MS spectroscopies, elemental and X-ray crystal structural analyses. Both compounds 1 and 2 act as homogenous catalysts for the diastereoselective nitroaldol (Henry) reaction of aliphatic and aromatic aldehydes with nitroethane in different solvents such as acetonitrile, methanol or water. Complex 1 was found to be the more efficient catalyst for the Henry reaction in aqueous medium, providing β-nitroalcohols with good yields (67–82%) and diastereoselectivities ( syn/anti 59:41–70:30).

The chemical reactivity of electron deficient chromone–linked acrylonitrile [3-(6-methylchromonyl)acrylonitrile (1)] was studied towards some active methylene nitriles and active methylene ketones. Reaction of compound 1 with malononitrile, cyanoacetamide, ethyl cyanoacetate, malononitrile dimer and acetoacetanilide afforded 5-cyanomethylchromeno[4,3-b]pyridines 2–4 and 9. Compound 1 reacted with 1H-benzimidazol-2-ylacetonitrile producing pyrido[1,2-a]benzimidazole derivative 5. Benzonitrile derivatives 6–8 were efficiently synthesized from the reaction of compound 1 with acetylacetone, ethyl acetoacetate and diethylmalonate. In these reactions a diversity of products has been synthesized through a domino process, including Michael addition, retro-Michael with γ-pyrone ring opening followed by different types of recyclization (RORC). Structures of the new synthesized products were deduced on the basis of their analytical and spectral data, and the reaction mechanisms are discussed.

An integrated platform consisting of a microfluidic paper-based chip and a mini-box detection system is proposed for the concentration detection of formaldehyde. In the proposed approach, the reaction region of the paper-based chip is implanted with Acetoacetanilide reagent, and the formaldehyde concentration is deduced from the UV light-induced fluorescence intensity of the formaldehyde-Acetoacetanilide complex (dihydropyridine) produced in a Hantzsch reaction process at room temperature for 2 min. The experimental results obtained using control samples with formaldehyde concentrations ranging from 0 to 8 ppm show that the formaldehyde concentration (Y) and fluorescence intensity (X) are related as Y = 0.0039 e0.058X. Moreover, the correlation coefficient (R2) is equal to 0.9987. The real-world applicability of the proposed paper-based platform is demonstrated by measuring the formaldehyde concentration in eleven commercial food samples. It is found that the measurement results deviate from those obtained using a standard bench top method by no more than 4.7%. Overall, the results presented in this study show that the proposed system provides a rapid and reliable technique for formaldehyde concentration detection.

A facile synthetic strategy has been reported for the diastereoselective synthesis of N,N′-diaryl-2-aryl-6-hydroxy-6-methyl-4-oxocyclohexane-1,3-dicarboxamide via Et3N-catalyzed. Acetoacetanilide undergoes a diastereoselective condensation reaction with various aromatic and aliphatic aldehydes in EtOH at 50 °C to afford desirable products via one-pot pseudo-three-component reaction. The present protocol provides an inexpensive and efficient route to obtain functionalized cyclohexanones containing four quaternary stereogenic centers with high yields from the simple and readily available starting materials under mild conditions in shorter reaction times. The products have been characterized by IR, 1H NMR, and 13C NMR spectroscopy. The stereoselectivity of compounds was established with crystallography and NMR spectroscopy.

A novel 4 H -chromene-3-carboxamide derivatives were synthesized from a multicomponent reaction between 2-hydroxybenzaldehyde, acetoacetanilide, and 4-hydroxycoumarin under solvent free conditions in the presence of ceric ammonium nitrate (CAN) via Knoevenagel-Michael reaction. In this work, we attempted and developed several amendments under solvent-free conditions to obtain the biologically important 4 H -chromene-3-carboxamides and characterized by NMR, HR-MS and UV–Vis spectroscopy analysis. The solvatochromic properties of compounds 4a – l was studied with solvents of increasing order of polarity. A series of 4 H -chromene-3-carboxamides 4 ( a – l ) and their antibacterial activities against Gram positive and Gram negative organisms was investigated using agar well technique. In the in vitro assay, the compounds 4k and 4l (9.3 μg/mL) showed promising antibacterial activity compared to ampicillin (standard). The compounds 4 ( a – l ) were studied for their in vitro antioxidant (DPPH method) activity, 4d , 4h , 4k , and 4l compounds showed strong antioxidant activity with IC 50 values of 1.39, 1.33, 1.26 and 1.10 μg/mL when compared to ascorbic acid (standard).

The new trinuclear [Co II (H 2 O) 2 (Me 2 NCHO) 2 {Co III (1κ NOO′ :2κ O″ -μ-HL)} 2 (1κ NOO′ )]·2H 2 O ( 1 ) and polymeric [Co II (1κ NOO′ :2κ O″ -μ-HL)(MeOH) 2 ] n ( 2 ) complexes were isolated by the reaction of ( Z )-2-(2-(1,3-dioxo-1-(phenylamino)butan-2-ylidene)hydrazinyl)benzoic acid (H 3 L) with CoCl 2 ·6H 2 O and Co(ClO 4 ) 2 ·6H 2 O, respectively, in a mixture of DMF/MeOH (1/10, v/v), in the presence of Et 3 N at room temperature, and characterized by IR and ESI-MS spectroscopies, elemental and X-ray crystal structural analyses. In both structures the cobalt centres display distorted octahedral geometries. The structural units in 1 and 2 are connected via intermolecular hydrogen bonds involving the coordinated HL 2− and the bound and non-bound water molecules (in 1 ) or the methanol ligands (in 2 ) which provides supramolecular networks. 1 and 2 are catalyst precursors for the cyanosilylation reaction of a variety of both aromatic and aliphatic aldehydes with trimethylsilyl cyanide leading to the corresponding cyanohydrin trimethylsilyl ethers in high yields (up to 89%) in methanol and at room temperature.

The new copper(II) complexes [Cu(HL1)(H2O)(CH3OH)] (1, H3L1 = (Z)-2-(2-(1,3-dioxo-1-(phenylamino)butan-2-ylidene)hydrazinyl)benzoic acid) and [Cu(HL2)(H2O)(CH3OH)] (2, NaH2L2 = sodium (Z)-2-(2-(1,3-dioxo-1-(phenylamino)butan-2-ylidene)hydrazinyl)benzenesulfonate) were synthesized and characterized by IR and ESI-MS spectroscopies, elemental and X-ray crystal structural analyses. The coordination environment of the central copper(II) has a square pyramidal geometry, three sites being occupied by (HL1,2)2−, which chelates in the O,N,O fashion, while the two other sites are filled with the water and methanol ligands. Multiple intra- and intermolecular non-covalent interactions between the (HL1,2)2−, water and methanol ligands lead to supramolecular network. Both compounds 1 and 2 act as homogenous catalysts for the cyanosilylation reaction of a variety of both aromatic and aliphatic aldehydes with trimethylsilyl cyanide affording the corresponding cyanohydrin trimethylsilyl ethers in high yields (up to 90%) and at room temperature.
