Abstract This study investigated the anti-inflammatory compounds in Ogaza, Acanthopanax sessiliflorus fruit, and their extracts using metabolomic screening. Ogaza extracts were obtained in various solvents, such as 70% ethanol, 70% methanol, and water. The anti-inflammatory activity was estimated by evaluating nitric oxide production in lipopolysaccharide (LPS)-induced RAW 264.7 cells treated with the extracts. The 70% ethanol extract (EO) showed the most effective anti-inflammatory activity, inhibiting nitric oxide production by approximately 50% and downregulating iNOS expression. The 70% ethanol extract was further fractionated into three partial subfractions by preparative LC to identify the anti-inflammatory compounds. Assessment of the anti-inflammatory activity of each subfraction revealed that the third subfraction (E-F3) showed the highest inhibitory activity against nitric oxide. E-F3 effectively suppressed iNOS expression. Subsequently, liquid chromatography-tandem mass spectrometry (LC-MS/MS) and multivariate statistical analyses were performed to identify the compounds that significantly contributed to the anti-inflammatory activity of the Ogaza extract. Fourteen and 16 compounds in the negative- and positive-ion modes, respectively, were identified as significant constituents of Ogaza. Compounds like quercetin, hyperoside, acanthoside D, oleanolic acid, and scopoletin were identified as potential anti-inflammatory components in Ogaza extract. This study characterized the functional properties of Acanthopanax sessiliflorus fruit and indicated the possibility that other compartments of Acanthopanax sessiliflorus may also serve as natural sources of nutraceuticals.

Acanthopanax senticosus (ASC) contains a variety of bioactive compounds and serves as an important traditional Chinese medicinal resource. However, its prolonged growth cycle and reliance on wild populations limit its practical use. To explore the potential of Acanthopanax sessiliflorus (ASF) as an alternative, this study focused on optimizing the extraction process and assessing the bioactivity of stem extracts. The Analytic Hierarchy Process-Entropy Weight Method (AHP-EWM) was applied to comprehensively score five key active compounds in ASF stems, establishing a benchmark for evaluating extraction efficiency. Ultrasound-assisted enzymatic extraction (UAEE) was employed, and orthogonal and response surface experiments were conducted to refine the extraction parameters. The optimal conditions identified were an ultrasound temperature of 59 °C, a duration of 57 min, an ethanol concentration of 61%, and a liquid-to-material ratio of 39:1, resulting in an AHP-EWM composite score of 94.76. Comparative analysis of antibacterial and antioxidant activities revealed that ASC extracts exhibited superior antibacterial effects, while ASF extracts demonstrated enhanced antioxidant properties. These findings support the partial replacement of ASC with ASF, contributing to the conservation of wild resources and expanding the application of ASF in medicinal development.

The processing of Acanthopanax sessiliflorus has attracted interest due to its health benefits. In this work, an emerging blanching technology, called hot-air flow rolling dry-blanching (HMRDB), was employed to treat A. sessiliflorus before drying. The effects of varied blanching times (2–8 min) on enzyme inactivation, drying characteristics, bioactive compound retention, and microstructure were examined. The results demonstrated that blanching for 8 min rendered polyphenol oxidase and peroxidase nearly inactive. The blanching process reduced the drying time of samples by up to 57.89% compared to an unblanched sample. The Logarithmic model showed good fitting performance for the drying curves. The total phenolic and flavonoid content of the dried product increased as blanching time increased. The total anthocyanin content of the samples blanched for 6 min was 3.9 times higher than that of the unblanched samples, and 8 min of blanching produced the greatest DPPH• and ABTS• scavenging capabilities. The retention of active compounds in a dried product is a result of the inactivation of enzymes and a reduced drying period. Changes in the porous structure of the blanched samples would be responsible for the accelerated drying rate, according to microstructural analysis. Key Message: Nitrogen addition leads to large increases in shoot growth but limited increases in root growth and reductions in mycorrhizal colonization of Sorbus pohuashanensis and Acanthopanax sessiliflorus. Soil in the cultivated fields of Changbai Mountain region of China is rich in phosphorus (P) and deficient in nitrogen (N) for most woody plants. However, currently N deposition is increasing and reducing its limitation on plant growth. How N addition shifts carbon investment among shoots, roots and arbuscular mycorrhizal (AM) fungi is not well understood, especially in woody plants growing in the field. We examine the responses of the growth, biomass partitioning and AM colonization of Sorbus pohuashanensis Hedl. and Acanthopanax sessiliflorus Seem. to low and high N fertilization in northeastern China on high-P soil over 3 years. With N addition, both plants increased shoot biomass by 20–45%, and N and P content by 13–30%, while root biomass increased only by 2.1–5.4%. The slower increase in root growth relative to shoot growth resulted in lower root mass fraction. After plant size (ontogeny) was accounted for, root mass fraction still decreased significantly with high N fertilization in both species. Mycorrhizal colonization intensity and AM-colonized root length decreased with an increase in N addition. In this P-rich site, the limited increase in root biomass and large decrease in AM colonization with N addition presumably promoted plant growth and nutrient uptake. Our results imply that the growth of these two species may be improved by increased carbon allocation to shoots, as N addition permitted sufficient nutrient uptake by roots and AM fungi to meet shoot nutrient demand without additional belowground carbon expenditure. Objective: The demands for measures to improve disease resistance and productivity of livestock are increasing, as most countries prohibit the addition of antibiotics to feed. This study therefore aimed to uncover functional feed additives to help enhance livestock immunity and disease resistance, using Acanthopanax sessiliflorus fruit extract (ASF). Methods: ASF was extracted with 70% EtOH, and total polyphenolic and catechin contents were measured by the Folin-Ciocalteu and vanillin assay, respectively. The 3D4/31 porcine macrophage cells (MΦ) were activated by phorbol 12-myristate 13-acetate (PMA), and cell survival and growth rate were measured with or without ASF treatment. Flow-cytometric analysis determined the lysosomal activity, reactive oxygen species levels (ROS), and cell cycle distribution. Nuclear factor kappa B (NF-κB) and superoxide dismutase (SOD) protein expression levels were quantified by western blotting and densitometry analysis. Quantitative polymerase chain reaction was applied to measure the lipid metabolism-related genes expression level. Lastly, the antibacterial activity of 3D4/31 MΦ cells was evaluated by the colony forming unit assay. Results: ASF upregulated the cell viability and growth rate of 3D4/31 MΦ, with or without PMA activation. Moreover, lysosomal activity and intracellular ROS levels were increased after ASF exposure. In addition, the antioxidant enzyme SOD2 expression levels were proportionately increased with ROS levels. Both ASF and PMA treatment resulted in upregulation of NF-κB protein, tumor necrosis factor (TNF)α mRNA expression levels, lipid synthesis, and fatty acid oxidation metabolism. Interestingly, co-treatment of ASF with PMA resulted in recovery of NF-κB, TNFα, and lipid metabolism levels. Finally, ASF pretreatment enhanced the in vitro bactericidal activity of 3D4/31 MΦ against Escherichia coli. Conclusion: This study provides a novel insight into the regulation of NF-κB activity and lipid metabolism in MΦ, and we anticipate that ASF has the potential to be effective as a feed additive to enhance livestock immunity. Eco-friendly synthesis of metal nanoparticles using plant extracts plays a vital role in green nanotechnology. In this paper, a green method for synthesizing silver nanoparticles (AgNPs) is described by treating silver ions with the aqueous bark extract of Acanthopanax sessiliflorus at ambient temperature. The synthesized AgNPs were characterized by UV-Vis spectrophotometry, X-ray diffractometry (XRD), Fourier transform infrared spectroscopy and transmission electron microscopy (TEM). The formation of AgNPs was monitored using a UV-Vis spectrophotometer, which exhibited a maximum absorption peak ( λ ) at 435 nm, and also a color change from light yellowish to yellowish-brown was observed. Powder XRD analysis revealed the formation of fcc crystals, and TEM analysis revealed formation of roughly-spherical shaped AgNPs with an average diameter of ~20 nm. Thermogravimetric analysis showed a two-step thermal decomposition of AgNPs, which majorly started at 150 °C and continued gradually until 700 °C. These AgNPs were found to catalyze the reduction of p-nitroaniline to p-phenylenediamine in the presence of sodium borohydride (NaBH). The catalytic reduction reaction followed pseudo-first order kinetics with a reaction rate constant ( k) of 8.7 × 10 min. In vitro cytotoxic assay revealed dose-dependent toxicity of these AgNPs in human keratinocytes at higher concentrations. A new lignan, named acanthosessilin A (1), as well as eight known lignan and lignan glycosides 2-9 were isolated from an ethanolic extract of Acanthopanax sessiliflorus fruits. The chemical structures were determined by spectroscopic methods, including HR-EIMS, 1D NMR (1H, 13C, DEPT), 2D NMR (gCOSY, gHSQC, gHMBC, NOESY), and IR spectroscopy. All isolated compounds were tested for the ability to inhibit LPS-induced nitric oxide production in RAW264.7 macrophages.

A specific, simple, and sensitive ultra high performance liquid chromatography with tandem mass spectrometry method utilizing the Triple Quad system has been developed and validated for the simultaneous determination and pharmacokinetic study of four triterpenoid components of Acanthopanax sessiliflorus in rat plasma. The components are 22-α-hydroxychiisanogenin, chiisanogenin, (1R,11α)1,4-epoxy-11-hrdroxy-3,4-secolupane-20(30)-ene-3,28-dioic acid, and 22-α-hydroxychiisanoside. Sample preparation involved a liquid-liquid extraction of the analytes with ethyl acetate. Chromatographic separation was accomplished using an Agilent SB-C18 column (1.8 μm, 2.1 mm × 50 mm) with 2.0 min isocratic elution. The compounds were detected with a triple quadrupole tandem mass spectrometer in multiple reaction monitoring mode and an ESI source in negative mode. The method was linear for all analytes over the investigated range, with all determined correlation coefficients exceeding 0.9906. The limit of quantification of each analyte was lower than 1 ng/mL. The intraday and interday precisions were less than 14.9%, and the accuracy ranged from -10.2 to 11.8%. The mean recoveries of the analytes were higher than 80.0%, and the matrix effects were between 100.4 and 107.1%. Acanthopanax sessiliflorus , a small woody shrub has traditionally been referred to have anticancer activity, but it has not been scientifically explored so far . Therefore, to investigate the anticancer effects of A. sessiliflorus stem bark extracts (ASSBE), MDA-MB-231 and MCF-7 human breast cancer cells were treated with one of its bioactive fractions, n-hexane (ASSBE-nHF). Cytotoxicity (24 h) was determined by MTT assay and antiproliferative effect was assessed by counting cell numbers after 72 h treatment using hemocytometer. The role of ASSBE-nHF on apoptosis was analysed by annexin V-FITC/PI, Hoechst 33342 staining, DNA fragmentation pattern and immunoblotting of apoptosis markers. For the assay of enhanced production of ROS and mitochondrial membrane depolarization, specific stains such as DCFH-DA and JC-1 were used, respectively. To understand the mode of action of ASSBE-nHF on MCF-7 cells, cells were pre-treated with antioxidant, n-acetylcysteine. The hexane fraction of ASSBE showed maximum activity towards human breast cancer cells compared to other two fractions at a minimal concentration of 50 μg/ml. The annexin V-FITC/PI, Hoechst 33342 staining, DNA fragmentation and immunoblotting assays showed that ASSBE-nHF induces non-apoptotic cell death in MCF-7 and MDA-MB-231 cells. ASSBE-nHF significantly increased the production of ROS and decreased the mitochondrial membrane potential (MMP) in MCF-7 cells. Similarly, it decreased the MMP in MDA-MB-231 cells, but had no effect on ROS production. Further, the cytotoxic effect of ASSBE-nHF in MCF-7 cells was not significantly reversed even in the presence of n-acetylcysteine, an antioxidant. These findings revealed that ASSBE-nHF induces non-apoptotic cell death via mitochondria associated with both ROS dependent and independent pathways in human breast cancer cells. Acanthopanax sessiliflorus, a plant mainly found in Korea, Japan, and China, has been used as a traditional medicine due to the bioactivities of some plant compounds. The antioxidant activity of A. sessiliflorus root in vivo and the anti-aging effect using Caenorhabditis elegans were studied. Pre-treatment with an A. sessiliflorus root extract significantly ( p<0.05) increased resistance to oxidative stress and induced increased survival after heat shock and ultraviolet irradiation treatment, compared with controls. A lifespan assay revealed that both the mean and maximum lifespans were significantly ( p<0.05) extended due to A. sessiliflorus root extracts, compared with controls. The lifespan extension was not accompanied by reduced reproduction, unlike other long-lived mutants. A. sessiliflorus root extracts also protected worms against human β-amyloid-induced toxicity. Acanthopanax sessiliflorus has been used as traditional medicine in many diseases. The extracts from Acanthopanax sessiliflorus leaves were examined for their anti-oxidant activity in vivo. Resistance to oxidative stress was significantly increased by Acanthopanax sessiliflorus extracts. Since increased resistance to oxidative stress is commonly positively correlated with lifespan extension, we determined the effect of Acanthopanax sessiliflorus on lifespan of C. elegans. Both mean and maximum lifespan were markedly increased by Acanthopanax sessiliflorus extracts. In addition, the treatment of Acanthopanax sessiliflorus enhanced the survival of worms under heat stress. However, the reduced fertility or delayed reproduction period frequently accompanied with increased lifespan were not observed in Acanthopanax sessiliflorus-treated animals. These findings suggest that Acanthopanax sessiliflorus leaves can confer longevity phenotype without reduced fertility, possibly through their anti-oxidant activity. [ABSTRACT FROM AUTHOR]

Abstract: The Nafion multi-walled carbon nanotube composite modified graphite paste electrode (Nafion/MWCNT-GPE) was investigated for electrocatalytic oxidation of quercetin in 0.1molL−1 citric–NaOH buffer (pH=3.7) and in 20% methanol solution. This was performed by cyclic voltammetry, electrochemical impedance spectrometry (EIS) and square-wave anodic stripping voltammetry (SWASV) techniques. Quercetin shows a reversible electron transfer at the modified electrode in potential range of −0.1V to 0.6V. Results indicated that the electrode process was diffusion limiting. The optimum parameters for SWASV deposition condition were purging time 4min, deposition time 150s, deposition potential 0.0V, pulse amplitude 40mV, step potential 6mV and frequency 50Hz. At optimal conditions, the content of quercetin was found to be 2.89mgg−1 and 4.21mgg−1 in dry and frozen fruits of Acanthopanax sessiliflorus (A. sessiliflorus) respectively with recoveries on the quercetin in dry 99.4% and frozen 99.2% (n =5). The SWASV method with the Nafion/MWCNT-GPE is proven to be a viable technique for determining quercetin in fruits of A. sessiliflorus samples. [Copyright &y& Elsevier]

Abstract: Consumption of fruits and vegetables is correlated with a lower incidence of cancer. Here, we identified hyperoside as an active compound from Acanthopanax sessiliflorus, and investigated the effect of hyperoside on UVB-induced transactivation of activator protein 1 (AP-1) and on the mitogen-activated protein kinase signalling pathway in JB6 P+cells. Hyperoside inhibited UVB-induced AP-1 transactivation. It inhibited the UVB-induced phosphorylation of p90RSK. Kinase assays revealed that hyperoside significantly inhibited ERK1/2 activity. Furthermore, hyperoside bound to ERK1/2 to suppress its activity. In addition, phosphorylation of cAMP response element binding protein (CREB) and signal transducers and activators of transcription (STAT) 3 were suppressed by hyperoside. Overall, these results indicate that hyperoside may be a promising chemopreventive agent that acts by suppressing the transactivation of AP-1 and the phosphorylation of p90RSK, CREB, and STAT3 through the binding and inhibition of ERK1/2. The article discusses a study which investigated the function of chiisanoside in the small intestines of mice. Chiisanoside is the main element of Acanthopanax sessiliflorus leaves. It is revealed that simulataneous administration of chiisanoside resulted in a decrease in the plasma triacylglycerol level and increase of undigested triacylglycerol in the intestinal lumen. The study suggests that chiisanoside has the potential to prevent obesity as a lipase inhibitor.

Six compounds were isolated from the fruits of Acanthopanax sessiliflorus. Their structures cwere elucidated as (-)-sesamin, scoparone, protocatechuic acid, ursolic acid, hyperin and 5-chydroxymethylfurfural by physicochemical and spectroscopic analysis. A simple and accurate liquid chromatographic method was developed for the simultaneous determination of six constituents in the fruit of Acanthopanax sessiliflorus. The conditions of sample extraction were optimised by using orthogonal design. The method provided good accuracy with recoveries in the range of 95.6–101.6% and good precision with RSD values less than 3.0%, which has been successfully applied to the quantitative determination of the six compounds in the fruit of A. sessiliflorus from two maturation periods.
