Introduction: Postpartum period of dairy cattle is an important phase of their life mainly associated with the changes in physiology, rumen function, and energy metabolism. Studies have shown that gut microbial composition undergoes drastic changes during the postpartum period. However, little is known about the temporal variations in digestive tract microbiota in postpartum Tibetan cattle. The aim of this study was to investigate the temporal variations in blood metabolites, ruminal fermentation, and microbial community of oral, rumen, and gut in lactating Tibetan cattle during postpartum. Methods: We collected blood, saliva, rumen fluid, and fecal samples from lactating Tibetan cattle during 1st week (1 W), the 2nd week (2 W), the 1st month (1 M), and the 2nd month (2 M) of the postpartum period. The microbiota of saliva, rumen fluid, and fecal samples were assessed using 16S rRNA sequencing. The rumen volatile fatty acid and blood parameters were also quantified. Results: The content of volatile fatty acids (VFAs) and blood parameters showed opposite tendency to each other and reached to stability at 2 M. Rumen microbiota showed the highest alpha diversity compared to other two sites. At phylum level, the oral cavity was dominated by Proteobacteria, while most dominant phylum in rumen and feces were Firmicutes and Bacteroidetes, respectively. The dominant genera in oral cavity were Moraxella and Bibersteinia, while genera Prevotella 1 and Ruminococcaceae UCG-005 were dominant in rumen and fecal samples, respectively. Discussion: Microbial network analysis revealed that most of the active genera in all networks belonged to phylum Firmicutes, indicating the importance of this phyla during postpartum period of lactating cattle. The functional analysis revealed distinct division of labor among three gastrointestinal sites associated with defense, fatty acid synthesis, and maintaining health of host. Immunoglobulin A vasculitis, historically known as Henoch-Schönlein purpura, is a rare form of autoimmune-induced vasculitis most common in children. This disease is characterized by a purpuric rash, arthritis, digestive tract complication, and renal inflammation (Hopkins). We present the case of a 78-year-old man in the emergency department with findings of weakness, abdominal pain, and bloody diarrhea for 3 days and a new-onset bilateral lower extremity rash. Diagnostic imaging and labs diagnosed this patient with immunoglobulin A vasculitis (IgAV) with associated acute kidney injury and abdominal mesenteric edema. Why Should an Emergency Physician be Aware of This? Recognition of IgAV by emergency physicians and assessment of multiple organ involvement is critical to expedite treatment and minimize complications. Particularly, physicians should consider and recognize the increased severity and different presentation of IgAV in adults in comparison with the more widely known manifestation in children. Eimeria bovis is a pathogenic protozoan that causes cattle digestive tract infections, which can cause economic losses to farmers. It is necessary to develop specific and accurate detection methods to conserve livestock and prevent coccidiosis in Indonesia. This study aims to detect E. bovis by nested PCR and determine the relationship with reference sequences. A total of 167 samples of beef cattle feces were taken randomly from community farms spread across 18 provinces in Indonesia. The feces were examined natively, and then the oocysts were purified by the sugar flotation method, extracted by KIT extraction, and amplified by the nPCR technique. Positive samples were followed by sequencing and phylogenetic analysis using MEGA 11 software. This study used two pairs of primers (outer and inner) taken from ITS-1 molecular markers. As many as 96 out of 167 samples (57.5%) were positive for Eimeria spp., and 48 of the 96 samples were positive for Eimeria spp. (50%) were detected to be positive for E. bovis based on the presence of a 238 bp DNA fragment. The results of the phylogenetic analysis showed that the study sample formed a separate cluster from the E. bovis cluster from abroad. In conclusion, E. bovis was detected in 16 out of 18 provinces in this study, and the nPCR technique proved to have better sensitivity and specificity. [ABSTRACT FROM AUTHOR]

Alpha‐Gal/α‐Gal is an oligosaccharide produced by non‐primate mammals. Humans have developed an immune response mediated by anti‐α‐Gal antibodies that can trigger an allergic reaction and cause anaphylaxis. In recent years, cases of patients with delayed allergic reaction to mammalian meat have been reported worldwide. In Spain, these cases have been related to the species Ixodes ricinus L. (Ixodida: Ixodidae), whose distribution is located in the north of the country. In this work, the presence of α‐Gal in water‐soluble extracts from samples of salivary glands and digestive tracts of Hyalomma lusitanicum Koch (Ixodida: Ixodidae) both engorged and collected from vegetation were studied. The presence of that epitope was confirmed by the presence of reactive proteins of >250 kDa in both samples. The highest concentrations of α‐Gal were detected in salivary glands. Neither sex nor diet influenced the concentration of α‐Gal, which seems to indicate its endogenous production and its possible inoculation to the host during tick feeding. [ABSTRACT FROM AUTHOR]

Importance: Obesity affects approximately 19% of women and 14% of men worldwide and is associated with increased morbidity. Antiobesity medications (AOMs) modify biological processes that affect appetite and significantly improve outcomes, such as type 2 diabetes, hypertension, and dyslipidemia. Observations: AOMs should be administered in combination with lifestyle interventions and can be classified according to their mechanisms of action. Orlistat modifies digestive tract absorption and causes gastrointestinal adverse effects, such as oily fecal spotting and urgency, in more than 25% of patients. Centrally acting drugs, such as phentermine-topiramate and naltrexone-bupropion, regulate appetite in the brain and are associated with constipation in approximately 20% of patients, although the incidence of other adverse effects (eg, paresthesia, nausea) varies by medication. Nutrient-stimulated hormone-based medications, such as liraglutide, semaglutide, and tirzepatide, mimic the actions of enteropancreatic hormones that modify central appetite regulation and provide multiple cardiometabolic weight-loss benefits. Adverse effects of these drugs include nausea (28%-44%), diarrhea (21%-30%), and constipation (11%-24%). The relative potency of adult obesity medications has been studied in meta-analyses. Compared with placebo, orlistat was associated with 3.1% greater weight loss (52 randomized clinical trials [RCTs]; 16 964 participants), phentermine-topiramate was associated with 8.0% greater weight loss (5 RCTs; 3407 participants), naltrexone-bupropion was associated with 4.1% greater weight loss (6 RCTs; 9949 participants), liraglutide was associated with 4.7% greater weight loss (18 RCTs; 6321 participants), semaglutide was associated with 11.4% greater weight loss (5 RCTs; 4421 participants), and tirzepatide 15 mg was associated with 12.4% greater weight loss (6 RCTs; 1972 participants). Conclusion and Relevance: Obesity is associated with increased morbidity. Antiobesity medications are effective adjunctive therapy to lifestyle changes for improved weight loss and health outcomes. This review examines the safety and efficacy of various antiobesity medications in conjunction with lifestyle modification according to their mechanism of action, including intragastrointestinal, centrally acting, and nutrient-stimulated hormone-based medications. Tumors present a formidable health risk with limited curability and high mortality; existing treatments face challenges in addressing the unique tumor microenvironment (hypoxia, low pH, and high permeability), necessitating the development of new therapeutic approaches. Under certain circumstances, certain bacteria, especially anaerobes or parthenogenetic anaerobes, accumulate and proliferate in the tumor environment. This phenomenon activates a series of responses in the body that ultimately produce anti-tumor effects. These bacteria can target and colonize the tumor microenvironment, promoting responses aimed at targeting and fighting tumor cells. Understanding and exploiting such interactions holds promise for innovative therapeutic strategies, potentially augmenting existing treatments and contributing to the development of more effective and targeted approaches to fighting tumors. This paper reviews the tumor-promoting mechanisms and anti-tumor effects of the digestive tract microbiome and describes bacterial therapeutic strategies for tumors, including natural and engineered anti-tumor strategies. Background: Ingestion of foreign bodies may be seen unconsciously or intentionally in patients with mental health problems. Most cases pass through the esophagus slowly; however, in some cases, the tumor may be located in narrower areas of the digestive tract that require endoscopic or surgical intervention. This study describes a rare case of successful removal of more than 450 pieces of metal objects from the stomach of a 36-year-old man via ingestion of foreign bodies at Imam Khomeini Hospital in Ahvaz. Case presentation: A 36-year-old male patient (Aryan race) presented with complaints of chronic abdominal pain, frequent vomiting, and intolerance to liquids and food. The patient's companions mentioned a history of gradual ingestion of small metal objects 3 months prior. The patient was conscious and had stable vital signs. In the patient's X-ray and endoscopy, multiple metal objects inside the patient's stomach were observed, causing gastric outlet obstruction. The patient underwent gastrostomy surgery, and 452 screws, nuts, keys, stones, and other metal parts weighing 2900 g were removed from the stomach. Five days after the operation, the patient was transferred to the psychiatric service in good general condition and was diagnosed with psychosis, and her condition returned to normal at follow-up. Conclusion: Successful removal of this foreign body is rare. In chronic abdominal pain, especially in the context of psychiatric disorders, attention should be given to the ingestion of foreign bodies. In swallowing large amounts of sharp and metallic foreign objects, surgical intervention is necessary, especially in cases of obstruction, and saves the patient's life. [ABSTRACT FROM AUTHOR]

Alterations in seawater chemistry posed by acidification may lead to immunological and antioxidant defence impairment in sea urchins, with differences among local populations. Here, we analyzed the effects of reduced pH on Paracentrotus lividus, with a multibiomarker approach, and the possible intraspecific variations in sea urchin responses. Two groups of animals with different ecological histories (i.e., the pattern of environmental characteristics and pressures experienced throughout the organism's lifetime) were maintained at ambient pH and pH reduced of 0.4 units for 8 months. Changes in gonadosomatic index (GSI), immunological, and oxidative stress biomarkers were assessed in coelomic fluid, gonads, and digestive tract. Animals maintained at reduced pH showed limited impact of seawater acidification compared to the ambient pH condition. However, sea urchins from the two sites were differently influenced by the seawater pH (as shown by multivariate analyses). GSI and immunological and antioxidant status were differentially modulated between the two sexes, with generally higher values in females, but differences between sexes in relation to the pH of exposure were limited. Overall, our findings highlight that the impact of environmental stressors may differ in sea urchins from different locations. Cholangiocarcinoma (CCA) is a rare type of digestive tract cancer originating from the epithelial cells of the liver and biliary tract. Current treatment modalities for CCA, such as chemotherapy and radiation therapy, have demonstrated limited efficacy in enhancing survival rates. Despite the revolutionary potential of immunotherapy in cancer management, its application in CCA remains restricted due to the minimal infiltration of immune cells in these tumors, rendering them cold and unresponsive to immune checkpoint inhibitors (ICIs). Cancer cells within cold tumors deploy various mechanisms for evading immune attack, thus impeding clinical management. Recently, combination immunotherapy has become increasingly essential to comprehend the mechanisms underlying cold tumors to enhance a deficient antitumor immune response. Therefore, a thorough understanding of the knowledge on the combination immunotherapy of cold CCA is imperative to leverage the benefits of immunotherapy in treating patients. Moreover, gut microbiota plays an essential role in the immunotherapeutic responses in CCA. In this review, we summarize the current concepts of immunotherapy in CCA and clarify the intricate dynamics within the tumor immune microenvironment (TIME) of CCA. We also delve into the evasion mechanisms employed by CCA tumors against the anti-tumor immune responses. The context of combination immunotherapies in igniting cold tumors of CCA and the critical function of gut microbiota in prompting immune responses have also been annotated. Three hyperimmune egg-based formulations rich in immunoglobulin Y (IgY) were orally administered (daily, for up to 90 days) to C57BL/6 mice that were not microbially challenged. The serum levels of 32 cytokines were quantified every 30 days. Histopathology, hematology, and serum biochemistry investigations were also performed. As a sign of increased immune activity, lymphohistiocytic infiltrates were detected in the digestive tract and the liver after 30, 60, and 90 days of treatment. These infiltrates were also present in the lungs after 30 and 60 days, but not at 90 days. Blood analysis indicated systemic inflammation after 30 days of treatment: increases in pro-inflammatory cytokines, glycemia, total serum proteins, ALT, and ALP. After 60 and 90 days of treatment, the analyzed blood parameters showed mixed signs of both increased and decreased inflammation. The increased cytokines, which varied with formulation and time of exposure, indicated a combination of mostly Th17- and Th2-type immune responses. As the mice were healthy and housed in standardized sanitary conditions, and were not microbially challenged, the data were consistent with an interaction of IgY with the gut-associated lymphoid tissue as the main mechanism of action. Botulinum neurotoxins (BoNTs) are the most potent toxins known and are used to treat an increasing number of medical disorders. All BoNTs are naturally co-expressed with a protective partner protein (NTNH) with which they form a 300 kDa complex, to resist acidic and proteolytic attack from the digestive tract. We have previously identified a new botulinum neurotoxin serotype, BoNT/X, that has unique and therapeutically attractive properties. We present the cryo-EM structure of the BoNT/X-NTNH/X complex and the crystal structure of the isolated NTNH protein. Unexpectedly, the BoNT/X complex is stable and protease-resistant at both neutral and acidic pH and disassembles only in alkaline conditions. Using the stabilizing effect of NTNH, we isolated BoNT/X and showed that it has very low potency both in vitro and in vivo. Given the high catalytic activity and translocation efficacy of BoNT/X, low activity of the full toxin is likely due to the receptor-binding domain, which presents very weak ganglioside binding and exposed hydrophobic surfaces. Programmed cell death (PCD) plays a crucial role in maintaining the normal structure and function of the digestive tract in the body. Infection with Helicobacter pylori (H. pylori) is an important factor leading to gastric damage, promoting the Correa cascade and accelerating the transition from gastritis to gastric cancer. Recent research has shown that several PCD signaling pathways are abnormally activated during H. pylori infection, and the dysfunction of PCD is thought to contribute to the development of gastric cancer and interfere with treatment. With the deepening of studies on H. pylori infection in terms of PCD, exploring the interaction mechanisms between H. pylori and the body in different PCD pathways may become an important research direction for the future treatment of H. pylori infection and H. pylori-related gastric cancer. In addition, biologically active compounds that can inhibit or induce PCD may serve as key elements for the treatment of this disease. In this review, we briefly describe the process of PCD, discuss the interaction between different PCD signaling pathways and the mechanisms of H. pylori infection or H. pylori-related gastric cancer, and summarize the active molecules that may play a therapeutic role in each PCD pathway during this process, with the expectation of providing a more comprehensive understanding of the role of PCD in H. pylori infection. Adult female and male Gongylonema nematodes were found in the oesophagus of a free-living roe deer (Capreolus capreolus) in Slovenia during passive health surveillance of wildlife. The genus Gongylonema was determined by light microscopy based on the genus-specific cuticular bosses in the anterior part of the parasite. Molecular methods were used to confirm the species Gongylonema pulchrum, which has zoonotic potential. Although Gongylonema species are considered common and distributed worldwide, this is the first report of G. pulchrum in an animal on the territory of Slovenia and the first molecular report in a roe deer worldwide. The parasite is likely to be underdiagnosed, misdiagnosed or goes unnoticed as the animals show little or no clinical signs and minor pathological lesions. Slaughterhouse workers, hunters and veterinarians should be aware of this elusive parasite. This review critically evaluates current in vitro gastrointestinal models for assessing infant formulas from a perspective focusing on a process along digestion, absorption and microbial fermentation. Despite their potential, none of the current in vitro gastrointestinal models fully considers the dynamics and continuous upstream‐downstream process of the human digestive tract. Considering the substantial differences between infants and adults, there is a pressing need for whole‐process gastrointestinal models tailored to infants. Developing such models is crucial for accurately assessing infant formula nutritional value and physiological function, enhancing the formulation of infant formulas to better meet the needs of infants. Roux‐en‐Y gastric bypass (RYGB) involves creating a small stomach pouch, bypassing part of the small intestine, and rerouting the digestive tract. These alterations can potentially change the drug exposure and response. Our primary aim was to assess the impact of RYGB on the pharmacokinetics of simvastatin lactone (SV) and its active metabolite, simvastatin hydroxy acid (SVA). Ultimately, we aimed to optimize dosing for this understudied population by employing a population pharmacokinetic–pharmacodynamic link approach. The study comprised patients who had undergone RYGB surgery and individuals without a previous history of RYGB. All participants received a single oral dose of simvastatin. Plasma concentration data were analyzed with a nonlinear mixed‐effect modeling approach. A parent–metabolite model with first‐order absorption, 2‐compartments for SV and 1‐compartment for SVA, linear elimination, and enterohepatic circulation best described the data. The model was linked to the turnover pharmacodynamic model to describe the SVA inhibition on LDL‐cholesterol production. Our simulations indicated that following RYGB surgery, the exposure to SV and SVA decreased by 40%. Consequently, for low‐intensity statin patients, we recommend increasing the dose from 10 to 20 mg in post‐RYGB patients to maintain a comparable response to that of non‐operated subjects. Moderate‐intensity statin patients should require increasing doses to 40 or 60 mg or the addition of a non‐statin medication to achieve similar therapeutic outcomes. In conclusion, individuals post‐RYGB exhibit diminished exposure to SV and may benefit from increasing the dose or adjunctive therapy with non‐statin drugs to attain equivalent responses and mitigate potential adverse events. Characteristics of the digestive enzymes of the fifth instar larvae of the beet moth were determined to achieve a superior understanding of the digestive physiology of the pest. After dissecting the larval digestive tube, optimum acidity for the activity of α-amylase and α-glucosidase was obtained 8–9, and 8 for β-glucosidase, respectively. The optimum temperatures were 35 °C and 35–40 °C for α-amylase activity and for α- and β-glucosidase, respectively. The zymogram displayed the existence of three α-amylase isoforms in the digestive tract of this insect. The maximum activity of general protease was determined in the digestive tract of this pest using azocasein substrate in the pH range of 10–11. Furthermore, the optimum temperature was 35 °C for general protease. The metal ions including iron, magnesium, manganese, zinc, and copper at 5 and 10mM concentrations had a significant influence on the activity of α-amylase, α- and β-glucosidase, trypsin, chymotrypsin, and elastase enzymes. Zymogram confirmed the significant inhibitory effect of PMSF and TLCK on the activity of serine proteases, especially trypsin. The results of this research have determined to some extent the digestive system physiology and the digestive enzymes activity range in S. ocellatella. Certainly, Study on the characteristics of digestive enzymes of beet moth can be the starting point for further studies on the use of enzyme inhibitors and the possibility of preparing new methods to control this key sugar beet pest. Scalloped hammerhead sharks (Sphyrna lewini) are critically endangered, according to the International Union for Conservation of Nature Red List, likely due to anthropogenic activities such as intense fishing and pollution. Nowadays, plastic debris contamination is a subject of concern due to its extensive presence in the sea and the digestive tracts of many fish species. The possible effects of plastic debris as a vector of other pollutants are still unknown. We analyzed the digestive tract of 58 hammerhead sharks to investigate the correlation between plastic and other anthropogenic microparticle contamination and their feeding habits in the eastern region of the Gulf of California, revealing a debris contamination occurrence of 79.3%. Out of these, 91.4% corresponded to fibers, and the remaining 8.6% to fragments. The main component of the debris was cellulose (64.4%). According to their diet, these organisms exhibit benthopelagic habits, feeding both in the water column and on the seabed. These results indicate a high level of contamination of anthropogenic cellulosic microfibers in the area. Although cellulosic microfibers are recognized as a biomaterial, they can be harmful to marine species, posing an additional threat to this iconic shark. This changed according to the year, indicating that the anthropogenic microparticle ingestion is related to the discharges of human activities and their seasonality rather than to a selection process by the sharks. The passage of food through the digestive tract in an herbivorous rodent with a single-chamber semiglandular stomach, the fat sand rat Psammomys obesus, was studied by multiple marking of the food with inert plastic markers. The total mean retention time of markers in the digestive tract (DT), as well as in the stomach and caecum separately, was determined. The peculiarities of the digesta kinetics depending on the morphology of the digestive tract, diet, and the time of marker intake were clarified. The total time for removal of markers from the DT in the fat sand rat reaches 36 h. The mean retention time of markers in the DT was determined as a whole (17–18 h), as well as separately in the forestomach (7–9 h) and caecum (7–12 h). The uneven passage of feed has been revealed, which is considered as an important mechanism for maximizing the extraction of nutrients from plants, including microbial fermentation of structural carbohydrates of the food. Trichomycterus bogotensis is one of the three species of fish that inhabit the Colombian Andes. However, several aspects of its basic biology, including its digestive system, are unknown. The morphology and histology of the digestive system are of great importance for understanding a species' feeding habits and food preferences. Thus, this study aimed to perform a morphological description of the digestive tract of T. bogotensis. The gastrointestinal tract of seven specimens was processed for paraffin histology, high-resolution optical microscopy, and transmission electron microscopy. Anatomically, T. bogotensis exhibits a terminal mouth with villiform teeth, a narrow esophagus, a large and muscular stomach, and a short intestine. Histology and ultrastructure revealed a tissue organization similar to that reported for other Silurids, composed of four layers: mucosal, submucosal, muscular, and serosa. α‐Melanocyte stimulating hormone (α‐MSH) is a peptide hormone released from the intermediate lobe of the pituitary which regulates body pigmentation. In addition to the pituitary, α‐MSH is also produced in the midbrain, and exerts both anorexigenic and an anxiogenic actions. Acyl ghrelin and cholecystokinin are peripheral hormones derived from the digestive tract which affect the brain to control food intake and feeding behavior in vertebrates. In the present study, hypothesizing that plasma α‐MSH may also stimulate the brain and exert central effects, we examined whether peripherally administered α‐MSH affects food intake and psychomotor activity using a goldfish model. Intraperitoneal (IP) administration of α‐MSH at 100 pmol g−1 body weight (BW) reduced food consumption and enhanced thigmotaxis. These α‐MSH‐induced actions were blocked by intracerebroventricular administration of HS024, an antagonist of the melanocortin 4 receptor (MC4R), at 50 pmol g−1 BW, whereas these actions were not attenuated by pretreatment with an IP‐injected excess amount of capsaicin, a neurotoxin that destroys primary sensory (vagal and splanchnic) afferents, at 160 nmol g−1 BW. Transcripts for the MC4R showed higher expression in the diencephalon in other regions of the brain. These results suggest that, in goldfish, IP administered α‐MSH is taken up by the brain, and also acts as anorexigenic and anxiogenic factor via the MC4R signaling pathway. [ABSTRACT FROM AUTHOR]

Background: Colorectal cancer (CRC) is a malignant tumor within the digestive tract with both a high incidence rate and mortality. Early detection and intervention could improve patient clinical outcomes and survival. Methods: This study computationally investigates a set of prognostic tissue and cell features from diagnostic tissue slides. With the combination of clinical prognostic variables, the pathological image features could predict the prognosis in CRC patients. Our CRC prognosis prediction pipeline sequentially consisted of three modules: (1) A MultiTissue Net to delineate outlines of different tissue types within the WSI of CRC for further ROI selection by pathologists. Simple Summary: In symbiotic interactions, microorganisms associated with the intestinal tract, constituting the gut microbiome, are especially important, because they influence the life history and fitness of their host organisms. The gut microbiome of ground beetles, one of the most species-rich animal families, is poorly known, and research on sex-specific differences is almost completely missing. Investigating the gut bacterial microbiome of a widespread European carnivorous species, Carabus convexus, using next-generation high-throughput sequencing, we found 1138 different operational taxonomic units belonging to 21 bacterial phyla, 90 families, and 197 genera. One of the most abundant bacterial families and genera was previously not known from the digestive tracts of other ground beetle species. We also detected carbohydrate-degrading gut bacterial symbionts, which indicated possible plant feeding in this predatory species. Although the composition of the gut bacterial microbiome was not significantly different between the sexes, the gut symbionts showed more between-individual variability in females. This difference may result from the different mobility of sexes and the difference in reproductive investment. Biological interactions, including symbiotic ones, have vital roles in ecological and evolutionary processes. Microbial symbionts in the intestinal tracts, known as the gut microbiome, are especially important because they can fundamentally influence the life history, fitness, and competitiveness of their hosts. However, their effects on the ruminal and intestinal digestive tract have not yet been studied. Our study represents the first investigation of the effects of polyethylene terephthalate microplastics on the ability of the ruminal-gastro-intestinal system to degrade and digest mixed hay. Our findings reveal that polyethylene terephthalate significantly reduce the digestibility of crude protein in mixed hay. Notably, low levels of PET MPs negatively impacted the intestinal phase, while medium and high levels disrupted the ruminal phase. Moreover, medium and high concentrations of polyethylene terephthalate hindered the degradation of fiber fractions, specifically of neutral detergent fiber. This study aims to investigate the effects of polyethylene terephthalate (PET) MPs on the ability of the ruminal–gastro-intestinal system to degrade and digest mixed hay. Using a three-step in vitro ruminal–gastro-intestinal incubation system, PET MPs were introduced at concentrations of 0, 5, 10, and 15 g/L in ruminal and gastro-intestinal solutions. Ruminal fluid was collected from three 16-month-old Piedmontese bulls. The experiment was conducted on three mixed hays and was repeated three times, with triplicate incubations in each run. The results reveal that PET MPs reduced the degradability and digestibility of crude protein. Specifically, crude protein degradation was reduced by 9% at medium and 16% at high PET MP concentrations in the ruminal phase, while the crude protein digestibility of undegraded crude protein was reduced by 8% at the lowest PET MPs concentration in the gastro-intestinal tract. Additionally, PET MPs reduced the degradation of neutral detergent fiber at medium and high PET MP concentrations in the ruminal phase by 9% and 13%, respectively. These results highlight the risks of PET MPs contamination on ruminal–gastro-intestinal functions and underscore the urgent need to mitigate MPs contamination in the livestock sector. Bladder leiomyoma is benign neoplasm of smooth muscle and rarely reported in domestic animals. This tumor is most commonly diagnosed in the alimentary and urogenital tracts. This report describes the pathological characteristics of leiomyoma in the urinary bladder of a 4-year-old female sheep during routine necropsy. Urinary bladder showed that two sessile masses firm in consistency and well demarcated were located within the muscle layers and protruded from the mucosal layer. Bladder mucosa was intact. In histopathologic investigation, the masses were composed of densely interwoven bundles of smooth muscle with spindle cells. No malignancy changes including mitotic index, hemorrhage, or necrosis foci were detected. Based on the gross and microscopic findings, leiomyoma was diagnosed. Background Complete digestive disunion due to anastomotic necrosis is considered a contraindication to endoscopic repair. However, recent publications have suggested that endoscopic treatment by insertion of a self-expandable metal stent (SEMS) is possible. The report of this patient series aims to demonstrate the use of endoscopic management in selected cases with complete digestive disunion. Methods Seven consecutive patients with complete and circumferential upper gastrointestinal anastomotic disunion were treated at two European tertiary care centers between 2009 and 2021 by endoscopic insertion of an SEMS. Treatment was performed with a therapeutic gastroscope under general anesthesia, carbon dioxide insufflation, and fluoroscopic guidance, after surgical or percutaneous drainage. Results All patients were successfully treated by endoscopic insertion of fully or partially covered SEMS left in place for a median of 8 weeks, with a median of 3 endoscopic sessions. Digestive neo-epithelialization was associated with a restored circumferential gut lumen in all cases. The rate of stent migration was 23% and three patients (43%) experienced symptomatic strictures, which were successfully treated by endoscopic dilation. Conclusion Complete digestive rupture could be successfully treated by endoscopy in selected cases, adding proof-of-concept data regarding guided tissue regeneration alongside SEMS placement. Obesity is primarily caused by excessive intake as well as absorption of sugar and lipid. Postprandial surge in distention pressure and intestinal motility accelerates the absorption of nutrients. The response of intestinal epithelial cells to mechanical stimulation is not fully understood. Piezo1, a mechanosensitive ion channel, is widely expressed throughout the digestive tract. However, its function in intestinal nutrient absorption is not yet clear. In our study, excessive lipid deposition was observed in the duodenum of obese patients, while duodenal Piezo1–CaMKK2–AMPK α was decreased when compared to normal-weight individuals. Under high-fat diet condition, the Piezo1 iKO mice exhibited abnormally elevated sugar and lipid absorption as well as severe lipid deposition in the duodenum and liver. These phenotypes were mainly caused by the inhibition of duodenal CaMKK2–AMPK α and the upregulation of SGLT1 and DGAT2. In contrast, Yoda1, a Piezo1 agonist, was found to reduce intestinal lipid absorption in diet induced obese mice. Overexpression of Piezo1, stretch and Yoda1 inhibited lipid accumulation and the expression of DGAT2 and SGLT1, whereas knockdown of Piezo1 stimulated lipid accumulation and DGAT2 in Caco-2 cells. Our study reveals a previously unexplored mechanical regulation of nutrient absorption in intestinal epithelial cells, which may shed new light on the therapy of obesity. The immunotherapy for gastrointestinal tumors, as a significant research direction in the field of oncology treatment in recent years, has garnered extensive attention due to its potential therapeutic efficacy and promising clinical application prospects. Recent advances in immunotherapy notwithstanding, challenges persist, such as side effects, the complexity of the tumor immune microenvironment, variable patient responses, and drug resistance. Consequently, there is a pressing need to explore novel adjunctive therapeutic modalities. b-glucan, an immunomodulatory agent, has exhibited promising anti-tumor efficacy in preclinical studies involving colorectal cancer, pancreatic cancer, and gastric cancer, while also mitigating the adverse reactions associated with chemotherapy and enhancing patients' quality of life. Linezolid is a critically important antimicrobial used in human medicine. While linezolid is not licensed for food-producing animals, the veterinary use of other antimicrobials, such as phenicols (e.g., florfenicol), could cross/co-select for linezolid-resistant (LR) bacteria. Such LR strains pose a great concern for public health due to their potential transfer between animals and humans. This study explored possible associations between epidemiological risk factors, including phenicol use, and the occurrence of LR bacteria, such as enterococci and staphylococci, in poultry, pigs, and veal calves in Belgium. Florfenicol use significantly increased the likelihood of harboring LR bacteria in veal calves, sows, and fattening pigs, particularly for the digestive tract (odds ratio (OR): [3.19–5.29]) and the respiratory tract (OR: [6.11–9.09]). LR strains from feces from fattening pigs were significantly associated with production type (OR: [3.31–44.14]) and the presence of other animal species (OR: 0.41). The occurrence of LR strains in the respiratory tract from sows was also significantly associated with using antimicrobials other than florfenicol (OR: 10.07) and purchasing animals (OR: 7.28). Our study highlights the potential risks of using certain veterinary antimicrobials, such as florfenicol, in food-producing animals and emphasizes the need for responsible antimicrobial use to safeguard both animal and public health. The Florida pompano, Trachinotus carolinus, is an ideal species for commercial aquaculture because of its rapid growth, tolerance to a range of environmental parameters, and high demand in the seafood market. This study detailed embryonic and early larval development of this species. Florida pompano followed similar developmental sequences to those observed in other marine teleosts, and embryos hatched between 22 and 28 h post‐fertilization. Significant brain lobe differentiation occurred prior to hatching. Pectoral fins were functional within 1‐day post‐hatch (dph). Substantial eye development occurred within 3 dph, indicating visual acuity prior to the onset of exogenous live feeding. Intestinal segments were differentiated by 3 dph, and rotifers were seen in the digestive tract by 4 dph. Digestive functions were limited through the first 7 dph because of the absence of functional gastric glands and gastric mucous cells in the rudimentary stomach, lipid vacuoles in the anterior intestines, and the persistence of acidophilic supranuclear vesicles in the posterior intestines. Within 7 dph, the pharyngeal arches differentiated, and hematopoietic tissue and mesonephric tubules were visible in the larval kidney. Their rapid larval development and organogenesis are additional factors that make Florida pompano an ideal species for aquaculture production and should encourage continued efforts toward commercialization. Simple Summary: Plant-derived compounds, also known as phytogenics, are gaining popularity in the poultry industry as an effective replacement for antibiotic growth promoters, whose wide and irrational use damages the ecosystem and health of animals and humans. Phytogenics are known for improving poultry's health and production performance. These effects are closely related to the cecal microbiota of chickens, which highlights the importance of studying the effects of any feed additive related to gut microbial communities. In this study, we assessed the effects of a phytogenic blend with and without casein in high amounts on cecal microbiota composition and diversity using an artificial gastrointestinal system (CALIMERO-2) that mimics the physiology of a chicken's cecum. The phytogenic blend promoted the abundance of bacteria associated with energy metabolism and production performance in poultry and decreased the presence of opportunistic pathogens. This study showed promising feed additives that can be used as growth promoters for poultry; however, testing in living broiler chickens to prove these data is needed, as artificial GI systems cannot fully reproduce the intestinal physiology of animals. Phytogenic blends (PBs) consist of various bioactive plant-derived compounds that are used as growth promoters for farm animals. Feed additives based on PBs have beneficial effects on farm animals' production performance, health, and overall well-being, as well as positive modulating effects on gut microbiota. In this study, we used a validated in vitro cecal chicken alimentary tract model (CALIMERO-2) to evaluate the effects of a PB (a mix of components found in rosemary, cinnamon, curcuma, oregano oil, and red pepper), alone or in combination with casein (control), on poultry cecal microbiota. Supplementation with the PB significantly increased the abundance of bacteria associated with energy metabolism (Monoglobus) and growth performance in poultry (Lachnospiraceae UCG-010). The PB also decreased the abundance of opportunistic pathogens (Escherichia-Shigella) and, most importantly, did not promote other opportunistic pathogens, which indicates the safety of this blend for poultry. Pancreatic cancer (PC) is one of the most common malignant tumors of the digestive tract and has a very high mortality rate worldwide. Different PC patients may respond differently to therapy and develop therapeutic resistance due to the complexity and variety of the tumor microenvironment. The Eph/ephrin signaling pathway is extensively involved in tumor-related biological functions. However, the key function of the Eph/ephrin signaling pathway in PC has not been fully elucidated. We first explored a pan-cancer overview of Eph/ephrin signaling pathway genes (EPGs). Then we grouped the PC patients into 3 subgroups based on EPG expression levels. Significantly different prognoses and tumor immune microenvironments between different subtypes further validate Eph/ephrin's important role in the pathophysiology of PC. Additionally, we estimated the IC50 values for several commonly used molecularly targeted drugs used to treat PC in the three clusters, which could help patients receive a more personalized treatment plan. Following a progressive screening of optimal genes, we established a prognostic signature and validated it in internal and external test sets. The receiver operating characteristic (ROC) curves of our model exhibited great predictive performance. Meanwhile, we further validated the results through qRT-PCR and immunohistochemistry. AbstractCurrent treatments for osteoporosis include a calcium-rich diet, adequate exercise, and medication. Many synthetic drugs, although fast-acting, can cause a range of side effects for patients when taken over a long period, such as irritation of the digestive tract and a burden on the kidneys. As the world’s population ages, the prevalence of osteoporosis is increasing, and the development of safe and effective treatments is urgently needed. Active compounds in edible and medicinal homologous plants have been used for centuries to improve bone quality. It is possible to employ them as dietary supplements to prevent osteoporosis. In this review, we analyze the influencing factors of osteoporosis and systematically summarize the research progress on the anti-osteoporosis effects of active compounds in edible and medicinal homologous plants. The literature suggests that some naturally occurring active compounds in edible and medicinal homologous plants can inhibit bone loss, prevent the degeneration of bone cell microstructure, and reduce bone fragility through alleviating oxidative stress, regulating autophagy, anti-inflammation, improving gut flora, and regulating estrogen level with little side effects. Extranodal marginal zone lymphoma of mucosa-associated lymphoid tissue (MALT lymphoma) are a group of diseases with marked heterogeneity, including clinical, immunohistochemical, and molecular heterogeneity. The disease remains unspecified in the genetic landscape with only a few sequencing studies to date; however, systematic studies of alimentary canal MALT lymphoma have not been reported. To better understand the genetics of this tumor, targeted sequencing in a group of 31 cases (including 2 esophageal, 2 colonic, 4 small intestinal, and 23 gastric cases) and two cases of lymph node hyperplasiawere performed. We found epigenetic regulation (DNMT3A, KMT2D, KMT2A, EP300, TET2, etc.), signaling pathways (APC, CHD8, TNFAIP3, TNFRSF14, ZAP70, NF1,), and tumor suppressor genes (TP53, BCORL1, FOXO1, ATM, etc.) involved. Moreover, we found MTOR gene mutations in 16% of the cases that made these patients more prone to recurrence and metastasis than those with MTOR wild type genes. More interestingly, ARID2 mutations were detected in 32% of all the cases, and the mutation rate was higher and statistically significant in Helicobacter pylori (Hp)-negative patients in the gastric group. Introduction: Mixed neuroendocrine-nonendocrine neoplasms (MiNENs) of the digestive tract are rare. They contain both neuroendocrine and nonendocrine components. We here report a rare case of a metastatic MiNEN originating in the ascending colon. Case Presentation: An 80-year-old man presented with abdominal pain and vomiting. Colonoscopy showed an obstructing mass in the ascending colon, and a biopsy resulted in diagnosis of an adenocarcinoma. A CT scan revealed multiple liver metastases. Thus, our diagnosis was stage IV colon adenocarcinoma (cT4aN1bM1a according to TNM Classification of Malignant Tumors). The primary lesion was resected to alleviate symptoms. Histopathological and immunohistochemical examination revealed a well-differentiated adenocarcinoma and a small-cell neuroendocrine carcinoma, leading to the diagnosis of a MiNEN. The patient declined postoperative treatment and died approximately 2 months after the surgery. Conclusion: The incidence of MiNENs may be underestimated because they lack typical symptoms or imaging features and are therefore often only diagnosed after resection. A novel medical system (MoPill™) consisting of a capsule that wirelessly transmits radiofrequency signals to assess motility via 3D location, was used to conduct this study. The objectives were to: (1) confirm the safety of the MoPill™ system; (2) compare the 3D location transmitted by the capsule to its location captured by abdominal x‐rays; 3 determine gastric emptying (GE), whole gut transit time (WGTT) and segmental transit times.The MoPill™ system consists of an electronic capsule (2 × 1.2 cm), eight color‐coded adhesive sensors (6 × 5.5 cm), a recorder (15 × 11 × 2 cm), and software on a laptop. Four sensors were applied to the abdomen and four to the back. Healthy subjects who had fasted overnight ingested a 250‐calorie protein bar, 17 oz. of water, followed by an activated capsule. No further caloric contents were permitted for the next 5 h. At 1, 5, and 24 h (if the capsule had not been expelled), upright abdominal X‐rays (AP and lateral) were obtained to assess the location of the capsule, which was compared to the gastrointestinal positioning system (GPS) location determined by the MoPill™ system. Identification of the capsule's anatomical location by the MoPill™ system was based on (1) the 3D (x, y, z) location; (2) time; (3) trajectory (e.g., going up the right side of the body signified ascending colon); (4) frequency of contractions (e.g., 3 cycles/min for the stomach); and (5) milestone relationship (e.g., pyloric passage must follow the end of gastric contractions). GE was determined first by the end of the 3 cycles/min rhythmic movement of the stomach and then again by pyloric expulsion on 3D location. Small intestine transit was taken as the duration from pyloric expulsion to arrival in the cecum. Colon transit time was determined by calculating the duration from 3D arrival in the cecum to passage of the capsule out of the body (i.e., loss of signal accompanying a bowel movement).Ten healthy subjects (five women; mean age 34; mean BMI 24) were enrolled, and nine provided reliable data. The variation between the x‐ray and the estimated (i.e., identified by the MoPill™ system) location of the capsule was within an average of 3.5 cm (range 0.9–9.4 cm). The mean GE was 3.1 h. The small intestine's mean transit time was 4.3 h. The mean colonic transit time was 17.6 h. There were no adverse events recorded during the study.MoPill™ is a novel gastrointestinal positional system that accurately identifies the location of a capsule compared to an X‐ray. Associations between psychiatric disorders and digestive tract cancers have been proposed. However, the causal link between these factors remains unclear. This study pioneers Mendelian randomization (MR) analysis to explore the genetic link between psychiatric disorders and digestive tract cancers risk. We analysed data on six psychiatric disorders [schizophrenia, bipolar disorder, major depressive disorder (MDD), attention deficit hyperactivity disorder, autism spectrum disorder, and panic disorder (PD)] and digestive tract cancers [esophagus cancer (EC), gastric cancer (GC), and colorectal cancer (CRC)] from genome-wide association studies databases. Using instrumental variables identified from significant single nucleotide polymorphism associations, we employed the inverse variance weighted (IVW) method alongside the weighted median (WM) method and MR-Egger regression. The results revealed no causal link between psychiatric disorders and the risk of EC or GC. Psychiatric disorders were not identified as risk factors for CRC. Notably, PD demonstrated a lower CRC risk (OR = 0.79, 95% CI 0.66–0.93, P = 0.01). This MR analysis underscores the lack of a causal association between psychiatric disorders and digestive tract cancers risk while suggesting a potential protective effect of PD against CRC. In this study, for enhancing the resistance of probiotics to environmental factors, we designed a microgel beads delivery system loaded with synbiotics. Multiple droplets of W1/O/W2 emulsions stabilized with zein–apple pectin hybrid nanoparticles (ZAHPs) acted as the inner "egg," whereas a three-dimensional network of poly-L-lysine (PLL)-alginate-CaCl2 (Ca) crosslinked gel layers served as the outermost "box." ZAHPs with a mass ratio of 2:1 zein-to-apple pectin showed excellent wettability (three-phase contact angle = 89.88°). The results of the ζ-potentials and Fourier transform infrared spectroscopy demonstrate that electrostatic interaction forces and hydrogen bonding were the main forces involved in the formation of ZAHPs. On this basis, we prepared W1/O/W2 emulsions with other preparation parameters and observed their microstructures by optical microscopy and confocal laser scanning microscope. The multi-chambered structures of W1/O/W2 emulsions were successfully visualized. Finally, the W1/O/W2 emulsions were coated with PLL-alginate-Ca using the solution extrusion method. The results of the in vitro colonic digestion stage reveal that the survival rate of probiotics in the microgel beads was about 75.11%, which was significantly higher than that of the free. Moreover, probiotics encapsulated in microgel beads also showed positive storage stability. Apple pectin would serve as both an emulsifier and a prebiotic. Thus, the results indicate that the "egg-box" shaped microgel beads, designed on the basis of pH-sensitive and enzyme-triggered mechanisms, can enhance the efficiency of probiotics translocation in the digestive tract and mediate spatiotemporal controlled release. During the past 30 years, several developments have occurred in the antiplatelet field, including the role of aspirin in primary prevention of atherosclerotic cardiovascular disease. There have been several attempts to develop antiplatelet drugs more effective and safer than aspirin and a shift in emphasis from efficacy to safety, advocating aspirin-free antiplatelet regimens after percutaneous coronary intervention. Evidence supporting a chemopreventive effect of low-dose aspirin against colorectal (and other digestive tract) cancer has also strengthened. The aim of this article is to revisit the role of aspirin in the prevention of atherothrombosis across the cardiovascular risk continuum, in view of developments in the antiplatelet field. The review will offer a clinical perspective on aspirin's mechanism of action, pharmacokinetics, and pharmacodynamics. Among parasites of the digestive tract of the black-headed gull (Chroicocephalus ridibundus) in Poland, the best known are species of digenetic trematodes and cestodes. Nematodes of this bird species are not well known. Black-headed gulls, due to their varied diet, migration, life in a flock, and changes of habitat, can become infected with various species of helminths, and like synanthropic birds, they can spread the dispersal stages of parasites across urban and recreational areas. In the present study, an attempt was made to identify the helminth fauna of C. ridibundus from breeding colonies in north-central Poland. The aim of the study was to describe the taxonomic structure of parasites of the digestive tract of the black-headed gull and determine the quantitative parameters of their occurrence. A total of 43 black-headed gulls were examined post-mortem for gastrointestinal helminths, resulting in the identification of four cestodes (Paricterotaenia porosa, Lateriporus clerci, Anomotaenia micracantha, and Wardium fusum), three trematodes (Diplostomum pseudospathaceum, Plagiorchis laricola, and Apophallus muehlingi), and three nematodes (Eucoleus contortus, Cosmocephalus obvelatus, and Porrocaecum ensicaudatum). Colorectal cancer (CRC) is one of the most common digestive tract malignant tumors, which has a high mortality rate especially for patients with CRC recurrence. However, the pathological mechanism of recurrence of CRC is unclear. In this study, we integrated multiple cohort datasets and databases to clarify and verify potential key candidate biomarkers and signal transduction pathways in recurrence of CRC. As results, 628 DEGs were identified from GSE33113 and GSE2630 datasets and their function and pathway were analyzed. 14 hub genes related to CRC recurrence were screened from and their influence on survival were analyzed. Two key genes (IL1B and DDAH1) regarded as prognostic factors were further screened. Relapse-free survival results indicated the interaction between IL1B and DDAH1 genes and B cells was the most obvious and correlated with survival, with statistical significance (P < 0.05). Specially, cox regression analysis suggested that patients with T1 and N0 stages had a higher risk of recurrence than patients with T2 and N1. Fish possess lipases from embryonic development to adulthood. Lipase activity methods vary and significantly differ in terms of the concentration of the substrate used, bile salt, Ca2+, temperature, pH, and type of lipase units, which limits comparative studies. The three most-used substrates are p-nitrophenyl (p-NP), β-naphthyl (β-N) derivates, and emulsified natural oils. These were selected to be redesigned in this study to measure lipase activity under temperature, pH, ion, and bile salt conditions closer to fish physiology, using the appropriate molar absorption coefficient to calculate the lipase units. Cynoscion parvipinnis (CP), Seriola rivoliana (SR), Centropomus viridis (CV), Elop affinis (EA), and Canthidermis maculate (CM) pyloric caeca-intestine extracts were studied. Sodium taurocholate showed the highest activity for intestinal lipases, and the fatty acid length in the substrates changed the lipase hydrolysis rate. The highest lipase activity was obtained with p-NP butyrate and p-NP caprylate in four fish species. Lipase activity was highly activated with Ca2+ (4–7 mM). The β-N absorption spectrum indicates a plateau between 534 and 554 nm for fish lipases. Salmon oil was identified as the most digestible lipid in the four fish species using the in vitro digestibility assay. The lipase zymogram showed an apparent size of 46.3 kDa for CP, 40.2 kDa for SR, 46.2 kDa for CM, 106.6 kDa for EA, and 58.3, 84.6, and 162.1 kDa for CV. [ABSTRACT FROM AUTHOR]

Transient receptor potential (TRP) melastatin member 8 (TRPM8) is a non-selective cation channel that can be activated by low temperatures (8–26 °C), cooling agents (including menthol analogs such as menthol, icilin, and WS-12), voltage, and extracellular osmotic pressure changes. TRPM8 expression has been identified in the digestive system by several research teams, demonstrating its significant involvement in tissue function and pathologies of the digestive system. Specifically, studies have implicated TRPM8 in various physiological and pathological processes of the esophagus, stomach, colorectal region, liver, and pancreas. This paper aims to comprehensively outline the distinct role of TRPM8 in different organs of the digestive system, offering insights for future mechanistic investigations of TRPM8. Additionally, it presents potential therapeutic targets for treating conditions such as digestive tract inflammation, tumors, sensory and functional disorders, and other related diseases. The structural patterns of pyloric tonsil in turkey (Meleagris gallopavo) were investigated in this study. Turkeys, less resistant to disease than chickens, are a crucial source of protein with low cholesterol levels and a high protein/calorie ratio. Pyloric tonsil, involved in the chicken defense barrier, is a component of GALT. To evaluate the specific region, adult turkeys were taken from slaughterhouses. Crossman's modified triple staining demonstrated the general histological structure, PAS revealed the density of goblet cells synthesizing mucus, and Safranin O showed the capsule around the follicles. The width of lymphoid follicles in the pyloric tonsil (204.0±22.33) differed significantly from that of the middle part of the duodenum (111.7±4.741). It was noticed that the specific area harbors T cells, B cells, and follicular dendritic cells. Interfollicular regions were infiltrated with CD3+T cells, but the CD268+B cells occupied primarily germinal centers. However, it was noticed that T cells were present in germinal centers and some B cells in the interfollicular region. Anti-Vimentin antibody revealed follicular dendritic cells in the pyloric zone. It was noted that the epithelial layer of the pyloric tonsil is devoid of CK18-positive cells. Findings of the semi-thin section indicated that some columnar epithelial cells were stained differently in the transition zone (lymphoepithelium). To sum up, a pyloric region in turkeys is structurally dissimilar to pyloric tonsils detected in chicken, chukar partridge, and duck. More research is needed to reveal the effect of pyloric tonsil on the alimentary tract in poultry in terms of defense mechanisms. [ABSTRACT FROM AUTHOR]

Integrative studies are important for a better understanding of the functional development of fish digestive systems and the consequent design of suitable feeding regimes and ad-hoc diets. To characterize the ontogeny of digestive function in larvae and juveniles of two atherinopsids from South America, Odontesthes bonariensis and O. hatcheri, morphologic, histologic, and intestinal enzyme biochemical analyses were performed. Both species showed a functional digestive system from one week after hatching (wah) with a sustained high-level activity of the cytosolic enzyme leucine alanine peptidase (leu-ala) until 9 and 13 wah in O. hatcheri and O. bonariensis, respectively. Maximum specific leu-ala activities in both species were higher compared to other agastric and most gastric fish. These results and the abundant pinocytic vacuoles in the posterior intestine suggest that cytosolic digestion is crucial for these species. It seems that their apparent anatomically simple digestive system is compensated by an uncommonly early development and function of the digestive tract and accessory organs accompanied by high intestinal cytosolic activity. The above findings contribute to the understanding of the Atherinopsidae digestive model and, for the first time, suggest that under farming conditions, these species could be weaned at an early developmental stage with an appropriate balanced diet. Anorectal malformation (ARM), a common congenital anomaly of the digestive tract, is a result of insufficient elongation of the urorectal septum. The cytoplasmic protein Receptor of Activated C‐Kinase 1 (Rack1) is involved in embryonic neural development; however, its role in embryonic digestive tract development and ARM formation is unexplored. Our study explored the hindgut development and cell death mechanisms in ARM‐affected rats using spatial transcriptome analysis. We induced ARM in rats by administering ethylenethiourea via gavage on gestational day (GD) 10. On GDs 14–16, embryos from both normal and ARM groups underwent spatial transcriptome sequencing, which identified key genes and signalling pathways. Rack1 exhibited significant interactions among differentially expressed genes on GDs 15 and 16. Reduced Rack1 expression in the ARM‐affected hindgut, verified by Rack1 silencing in intestinal epithelial cells, led to increased P38 phosphorylation and activation of the MAPK signalling pathway. The suppression of this pathway downregulated Nqo1 and Gpx4 expression, resulting in elevated intracellular levels of ferrous ions, reactive oxygen species (ROS) and lipid peroxides. Downregulation of Gpx4 expression in the ARM hindgut, coupled with Rack1 co‐localisation and consistent mitochondrial morphology, indicated ferroptosis. In summary, Rack1, acting as a hub gene, modulates ferrous ions, lipid peroxides, and ROS via the P38‐MAPK/Nqo1/Gpx4 axis. This study was conducted from August to September 2021 and aimed to detect parasites and to determine their prevalence in the digestive tract of mackerel (Scomberomorus commerson) in Belawan, North Sumatra Province. Fish samples were randomly taken from the study site and classified according to length and weight before being dissected at the Laboratory of the Technical Implementation Unit for the Application of Quality of Fishery Products (UPTPMPH), Medan. The mackerel’s gastrointestinal system was collected and observed under a microscope to detect the presence of parasites. The result of this study showed that four genera of parasites were identified, which infected the organs of mackerel in the intestines and stomach. The prevalence of Rhadinorhynchus sp. was found to be 40% in the intestine and 6.66% in the stomach. In contrast, Anisakis sp. and Camallanus sp. were only found in the intestine with prevalence rates of 6.66 and 36.66%, respectively. Lecithocladium sp. was found in the stomach with a prevalence rate of 16.66%. Moreover, the highest prevalence was observed in the intestine and stomach for Rhadinorhynchus sp. This parasite often attacks most fish, while Anisakis sp., Camallanus sp., and Lecithocladium sp. were found exclusively in different organs. [ABSTRACT FROM AUTHOR]

This study investigated the Anisakis larvae infection in the scads (Decapterus spp.) in the Indian Ocean off the southern coast of East Java. A fish sample of approximately 450 individuals, consisting of three scad species, i.e., the Indian scad (D. russelli), the shortfin scad (D. macrosoma), and the redtail scad (D. kurroides), were obtained from Prigi and Muncar fishing port, East Java. Fish samples were measured individually for length and weight and subsequently dissected and examined in detail to detect the presence of Anisakis larvae in the abdominal cavity, intestinal tract, gonad, liver, and muscle. The collected larvae were preserved in 100% ethanol for the further identification process. The study indicates that the Decapterus spp. were receptive to the Anisakis larvae infection, with varying prevalence and mean intensity across species and locations. The redtail scad (D. kurroides) exhibited the highest prevalence and mean intensity; all of the sample were infected with Anisakis larvae (P= 100%), with a mean intensity of 22.54 larvae/individual.The shortfin scad (D. macrosoma), on the other hand, showed the lowest prevalence (P= 30.61%) and a mean intensity of MI = 1.29 larvae/individual. Most Anisakis larvae (52.30–76.61%) were located within the body cavity, followed by the digestive tract (16.78– 18.40%), and only a small number of the larvae were detected in the liver, gonads, and muscles. The morphological identification indicated that the larva infecting the scad was Anisakis Type I, and it was confirmed as Anisakis typica throughout molecular identification. Anisakis typica parasitizes the redtail and shortfin scads in the Indian Ocean off the southern coast of East Java, indicating genetic variation, which is shown by the differences in nucleotide composition.
