Background: The chemistry and applications of 2-pyridone structures have recently attracted a lot of attention due to their uses as synthetic intermediates and their biological importance. Furthermore, 2-pyridones are a different category of pharmacophores that exhibit several biological activities. Results: The 2-pyridones substrates are synthetically multilateral, where they can be used to synthesize a wide range of heterocyclic compounds and as a drug synthesis raw material. Therefore, it was the purpose of this review to evaluate the pharmacological properties of various substituted 2-pyridone systematically and collates these findings to be used as a guide for future structure–activity relationship and mode of action studies. Conclusion: Taken together, we demonstrate that the fused 2-pyridone is a versatile scaffold for biological applications.
