Objective To investigate the effects of different abstinence time on the parameters of sperm DNA damage, morphology and routine semen analysis. Methods A total of 1 128 semen samples were collected from male patients in the Center for Reproductive Medicine, Zhongda Hospital, Southeast University during September 2018 and February 2021. Routine semen analysis parameters, including sperm concentration, sperm motility, percentage of progressively motile sperm (PR), etc., were detected by the CFT-9201 computer-aided sperm analysis (CASA) system. Sperm morphology was observed after Diff-Quik staining, and sperm DNA fragmentation index (DFI) was detected by sperm chromatin structure analysis (SCSA). Results Abstinence time of 1 128 patients ranged from 0 to 30 days, with a median of 3. 0 [3.0, 5.0]. In addition to sperm motility (P = 0. 117) showed normal distribution, other parameters all showed non normal distribution (P <0. 01). Abstinence time was positively correlated with semen volume (r = 0. 21, P <0. 01), sperm concentration (r = 0. 098, P < 0. 01) and sperm DFI (r = 0. 068, P < 0. 05). Although abstinence time was negatively correlated with PR, sperm motility and percentage of normal morphology sperm, there was no significant correlation (P >0. 05). Sperm DFI was negatively correlated with PR, sperm motility and percentage of normal sperm morphology (P <0. 01), but not with sperm concentration (P > 0. 05). Conclusion With the extension of abstinence time, semen volume and sperm concentration increase, and sperm DFI also increases. Therefore, appropriately shortening abstinence time can reduce the production of sperm DNA fragments. Clinicians should consider the influence of abstinence time when explaining semen analysis results to patients. [ABSTRACT FROM AUTHOR]
