5. 卡格列净对高糖环境下小鼠肾小球系膜细胞的 抗炎、抗凋亡作用及其机制.

- Abstract
Objective To observe the anti-inflammation and anti-apoptosis effects of canagliflozin on high glucoseinduced glomerular mesangial cells, and to explore its possible mechanisms. Methods SV40 MES13 cells were randomly divided into normal control group, high glucose group, HG+canagliflozin 5 µmol/L group (canagliflozin 1 group), and HG+canagliflozin 15 µmol/L group (canagliflozin 2 group). The cells in the normal control group were cultured in 5. 5 mmol/L DMEM medium, and cells in the other groups were cultured in 35 mmol/L DMEM medium; 5 µmol/L cana‐ gliflozin was added to the canagliflozin 1 group, and 15 µmol/L canagliflozin was added to the canagliflozin 2 group. All groups were cultured for 24 h, the levels of monocyte chemoattractant protein (MCP)-1, tumor necrosis factor (TNF)-α, and interleukin (IL)-6 were detected by ELISA; the levels of TNF-α and IL-6 were detected by immunofluorescence assay; the apoptotic rate was measured by flow cytomtry, and the expression levels of transforming growth factor (TGF)-β1, p-Smad3, and Smad3 were detected by Western blotting. SV40 MES13 cells were randomly divided into NC group, HG group, HG+Can group, and HG+Can+Smad3 OE group. The cells in the NC group were cultured in 5. 5 mmol/L DMEM medium, and cells in the other groups were cultured in 35mmol/L DMEM medium; 15 µmol/L canagliflozin was added to the HG+Can group; pcDNA3. 1-Smad3 plasmid was transfected into the HG+Can+Smad3 OE group, and 15 µmol/L canagliflozin was added at 24 h after transfection. All groups were cultured for 24 h, the expression levels of TGF- β1, pSmad3, and Smad3 were detected by Western blotting, the levels of TNF-α and IL-6 were detected by immunofluorescence assay, and the apoptotic rate was measured by flow cytomtry. Results The levels of MCP-1, TNF-α, and IL-6 in supernatant and the expression levels of TNF-α and IL-6 in cells in the high glucose group were higher than those in the normal control group, the levels of the above indexs in the canagliflozin 1 group and canagliflozin 2 group were lower than those in the high glucose group, and the levels of above indexs in the canagliflozin 2 group were lower than those in the canagliflozin 1 group (all P<0. 05). The proportion of early and late apoptotic cells and the expression levels of TGF-β1 and p-Smad3/ Smad3 in the high glucose group were higher than those in the normal control group; the proportion of early apoptotic cells and the expression levels of TGF-β1 and p-Smad3/Smad3 in the canagliflozin 1 group and canagliflozin 2 group were lower than those in the high glucose group; and the proportion of early apoptotic cells in the canagliflozin 2 group was lower than that in the canagliflozin 1 group (all P<0. 05). The expression levels of TGF-β1, p-Smad3/Smad3, TNF-α, IL-6 and the proportion of early and late apoptotic cells in the HG group were higher than those in the NC group; the levels of above indexs in the HG+Can group were lower than those in the HG group; the expression levels of TGF-β1, p-Smad3/Smad3, TNF-α, and the proportion of early and late apoptotic cells in the HG+Can+Smad3 OE group were higher than those in the HG+Can group (all P<0. 05). Conclusion Canagliflozin inhibits high glucose-induced secretion of inflammatory factors and relieve the apoptosis in glomerular mesangial cells by inhibiting the activation of TGF-β/Smad3 signaling pathway. BACKGROUND: MicroRNAs (miRNAs) exert crucial effects on the regulation of cell apoptosis, signal transduction, differentiation and proliferation. Literatures demonstrate that regular exercise regulates biological pathways at the genomic and post-genomic levels. OBJECTIVE: To summarize the effects of exercises on miRNAs and the correlation between miRNAs and exercise performance. METHODS: In this review, relevant studies were searched in PubMed database using “microRNAs, athletes, physical function” for relevant articles published from 2007 to 2019. Inclusion criteria included publications within recent 10 years, impact factor > 3, and sorting according to “best match.” Finally 73 articles were selected. RESULTS AND CONCLUSION: Growing evidence suggests that miRNAs are mainly regulated by human skeletal muscle movement and varies with different factors, including the type, time, frequency and intensity of exercise. MiRNAs have at least three characteristics as biomarkers, which can be obtained by noninvasive methods (found in accessible body fluids), can specifically express specific pathology and physiological state, and have stable biological characteristics. MiRNAs have been considered as potential diagnosis and prognosis biomarkers based on the physiological functions of miRNAs in different exercise states, to evaluate exercise adaptation status and prevent sports injuries. 9. 模块化+ 项目驱动教学法在医学生信息素养教学中的应用.

- Abstract
Objective To explore the feasibility and function in improving the information literacy of medical students by using the modularization and project-driven teaching in the teaching of Medical Information Retrieval. Methods A total number of 197 students in four classes majoring in clinical medicine were randomly allocated into the experimental group (98 students) and the control group (99 students). All classes were taught by one teacher, who applied modularization and project-driven teaching in the experimental group and traditional teaching in the control group. Two groups took the same exam at the same time,and test scores were analyzed. Results The scores of experimental group were significantly higher than those of the control group(P < 0.05). Conclusions Modularization and project-driven teaching can mobilize students' internal cause for learning and cultivate consciousness of information, the ability of autonomous learning,independent thinking and teamwork, which shows the positive effect for improving students' information literacy. 10. 基于混合更新策略的可变形物体碰撞检测算法.

- Abstract
Concerning the real-time and accurate collision detection of deformable objects,this paper proposed a fast parallel collision detection algorithm based on mixed update strategy. It established A ABB bounding box hierarchy tree for the objects, and used octree instead of the usual binary tree,so improved the efficiency of collision detection algorithm. Then it used the hybrid update method combining the top-down and bottom-up update strategy to update the bounding box hierarchy tree at each time step,so further improved the efficiency of algorithm. And finally it used multi-threading technology to achieve parallel collision detection algorithm on multicore devices. Experimental results show that the algorithm has obvious advantages compared with the classic AABB algorithm in the aspects of efficiency and accuracy, and can meet the requirements of deformable object collision detection.
