Lake snow, caused by the freshwater centric diatom Lindavia intermedia, has become problematic in several large, oligotrophic New Zealand lakes over the past decade. Macroaggregates produced by L. intermedia foul fishing lines, intake screens, and water filters, and have a negative impact on recreational values. It was confirmed that the fibers constituting lake snow are composed of chitin, two chitin synthase genes (chs1 and 2) from L. intermedia were characterized, new qPCR-based tools to quantify the abundance of the species and measure expression of chs2 relative to the reference gene act1 (the product of which has cytoskeletal functions) were developed. The strong heterogeneity and mucilaginous nature of lake snow samples create particular difficulties for calibrations of gene or transcript copy numbers with cell densities and obtaining high yields of mRNA. However, data collected from four lakes during November 2018 and February and May 2019 show that abundance of L. intermedia is always high when lake snow is also abundant, but that a full range of L. intermedia abundance can occur when lake snow is absent, suggesting that chitin production is not obligate in L. intermedia. This result is consistent with the available data for chs2 expression, which suggest higher transcription when lake snow is abundant. Lake snow production by L. intermedia therefore requires an as yet undetermined stimulus independent of cell abundance.

Reduced body size is among the universal ecological responses to global warming. Our knowledge on how altered body size affects ecosystem functioning in ectothermic aquatic organisms is still limited. We analysed trends in the cell size structure of phytoplankton in the middle Danube River over a 34-year period at multiple levels: (1) average cell size of assemblages (ACS), (2) within the centric diatom community and (3) in the dominant centric diatom taxon: Stephanodiscus. We asked whether global warming and human impacts affected the average cell size of phytoplankton. Also, whether the altered size structure affected how chlorophyll-a, as an ecosystem functioning measure, relates to the ACS of phytoplankton. The cell size of phytoplankton decreased significantly at all organisation levels, and the assemblages became more dispersed in cell size over time. Environmental variables related to global warming and human impacts affected the ACS of phytoplankton significantly. The relationship between chlorophyll-a and the ACS of phytoplankton shifted from negative linear to broad and then narrow hump shape over time. Longer water residence time, warming and decline in nutrients and suspended solids decrease the ACS of phytoplankton in the middle Danube and expectedly in other large rivers. Our results suggest that cell size decrease in phytoplankton, especially of centric diatoms, constrains planktic algal biomass production in large rivers, independently of algal density. Such cell size decrease may also affect higher trophic levels and enhance the more frequent occurrence of “clear-water” plankton in large, human-impacted rivers under global change. In the present study 12 water samples of five sampling sites (Tatabánya, Dandár, Szentendre, Szent Flórián and Ciprián groundwaters) known as nutrient-depleted aquatic environments were studied using amplicon sequencing (NGS) and cultivation techniques. Diversity indices and cell counts were determined to assess the species richness in relation to the cell counts within the samples, and the oligocarbophile growth capability of the isolated bacteria was tested in microtiter plates. Altogether, 55 bacterial phyla were identified from the samples by amplicon sequencing. The microbial communities of the different sampling times of the same sites did not differ significantly. Patescibacteria and Proteobacteria were present in all samples. Ciprián sample was dominated by Bacteroidetes, while in Dandár sample a high ratio of Chloroflexi was detected. Rokubacteria and WOR-1 dominated Szent Flórián sample and Tatabánya had a high number of Epsilonbacteraeota. Nine archaeal phyla were also detected, the samples were characterized by the presence of unclassified archaea and Nanoarchaeota, among them Woesearchaeia, as the most dominant. Crenarchaeota and Altiarchaeota were detected in high ratios in Dandár water samples. Among Thaumarchaeota the family Nitrosopumilaceae, and orders of Nitrosotaleales and Nitrososphaerales appeared in Szent Flórián and Tatabánya samples. Key organisms of the different biogeochemical cycles were discovered in these nutrient-depleted environments: methanogenic archaea, methanotrophic bacteria, ammonia oxidizer, nitrate reducers, diazotrophs, sulfate reducers, and sulfur oxidizer. Diversity indices and cell counts of the samples show negative correlation in case of bacteria and positive in case of archaea in Ciprián sample. The high diversity indices in Szentendre samples are connected to low cell counts, most probably due to the vulnerability of the groundwaters to the external environment factors which lead to the infiltration of soil microbes and contaminants to the water. The isolated bacteria were affiliated into four phyla, most of them belonging to Proteobacteria (59%) followed by Actinobacteria (21%), Firmicutes (17%) and Verrucomicrobia (1%). The members of the facultative chemolithotrophic genera of Sphingobium, Sphingomonas, Sphingopyxis were characterizing only Szentendre, Szent Flórián and Tatabánya samples. Only 10% of the isolated species showed an obligate oligocarbophile character. From the samples, a high number of novel bacterial taxa were cultivated. As a conclusion, our results confirmed the predominance of unclassified and unknown taxa in subsurface water, pointing to the importance and necessity of further studies to characterize these microbial populations.

EnglishThe spatial and temporal patterns of phytoplankton in relation to physical and chemical properties were studied in a drinking water reservoir - the Lazberc Reservoir - located in north-east Hungary. The main objectives were to determine the role of the hydrologic regime in structuring algal growth and the effects of physical and chemical variables on the coexistence of a dominant species: cyanobacteria, Microcystis aeruginosa (Kutzing) Kutzing and an eukaryotic dinoflagellates, Ceratium hirundinella (O.F. Muller) Dujardin. The results suggest that nutrients (TP, NH4–N and NO3–N), turbidity, and the hydrologic regime play an important role in regulating the occurrence of investigated species. The rainy summer period resulting in a mixing of the water column, and the low level of nitrogen coupled with the internal release of phosphorus from the lake sediment under brief periods of anoxia promote cyanobacterial bloom. During this period, the water column was characterised by high turbidity, which created favourable conditions for developing a huge C. hirundinella biomass and the simultaneous occurrence of M. aeruginosa. This study also demonstrated - that contrary to what was previously known, deep standing waters in the temperate zone are not only dimictic (mixing twice a year during the spring and the fall), but they can also be mixed during the summer period due to continued rainfall, which equalizes the temperature in the whole water column. O periodo de verao chuvoso que resulta um equilibrio do nivel da agua, o baixo nivel de nitrogenio, juntamente com a liberacao interna de fosforo do sedimento, durante um curto periodo anoxico promove a floracao de cianobacterias. Durante este periodo, a coluna de agua foi caracterizada por alta turbidez, criando condicoes favoraveis para o surgimento de uma enorme expansao da biomassa C. hirundinella e a aparicao simultânea de M. aeruginosa. Este estudo mostrou tambem que as aguas profundas nas areas da zona moderada, nao tem apenas dimictic especialidade (misturando duas vezes por ano, na primavera e no outono), mas tambem durante a estacao do verao pode ser misturada por chuvas continuas que compensam a temperatura em toda a coluna da agua. During the past decade genetic approaches have been developed to monitor biodiversity in aquatic ecosystems. These enable access to taxonomic and genetic information from biological communities using DNA from environmental samples (e.g. water, biofilm, soil) and methods based on high-throughput sequencing technologies, such as DNA metabarcoding. Within the context of the Water Framework Directive (WFD), such approaches could be applied to assess Biological Quality Elements (BQE). These are used as indicators of the ecological status of aquatic ecosystems as part of national monitoring programs of the european network of 110,000 surface water monitoring sites with 79.5% rivers and 11% lake sites (Charles et al. 2020). A high-throughput method has the potential to increase our spatio-temporal monitoring capacity and to accelerate the transfer of information to water managers with the aim to increase protection of aquatic ecosystems. Good progress has been made with developing DNA metabarcoding approaches for benthic diatom assemblages. Technological innovation and protocol optimization have allowed robust taxonomic (species) and genetic (OTU, ESV) information to be obtained from which diatom quality indices can be calculated to infer ecological status to rivers and lakes. Diatom DNA metabarcoding has been successfully applied for biomonitoring at the scale of national river monitoring networks in several countries around the world and can now be considered technically ready for routine application (e.g. Apothéloz-Perret-Gentil et al. 2017, Bailet et al. 2019, Mortágua et al. 2019, Vasselon et al. 2019, Kelly et al. 2020, Pérez-Burillo et al. 2020, Pissaridou et al. 2021). However, protocols and methods used by each laboratory still vary between and within countries, limiting their operational transferability and the ability to compare results. Thus, routine use of DNA metabarcoding for diatom biomonitoring requires standardization of all steps of the metabarcoding procedure, from the sampling to the final ecological status assessment in order to define good practices and standards. Following previous initiatives which resulted in a CEN technical report for biofilm sampling and preservation (CEN 2018), a set of experiments was initiated during the DNAqua-Net WG2 diatom workshop (Cyprus, 2019) to focus on DNA extraction and PCR amplification steps in order to evaluate: i) the transferability and reproducibility of a protocol between different laboratories; ii) the variability introduced by different protocols currently applied by the scientific community. 19 participants from 14 countries performed DNA extraction and PCR amplification in parallel, using i) the same fixed protocol and ii) their own protocol. Experiments were performed by each participant on a set of standardized DNA and biofilm samples (river, lake, mock community). In order to specifically test the variability of DNA extraction and PCR amplification steps, all other steps of the metabarcoding process were fixed and the preparation of the Miseq sequencing was performed by only one laboratory. The variability within and between participants will be evaluated on DNA extracts quantity, taxonomic (genus, species) and genetic richness, community structure comparison and diatom quality index scores (IPS). We will also evaluate the variability introduced by different DNA extraction and PCR amplification protocols on diatom quality index scores and the final ecological status assessment. The results from this collaborative work will not serve to define “one protocol to rule them all”, but will provide valuable information to define guidelines and minimum requirements that should be considered when performing diatom metabarcoding for biomonitoring.

The chemical ecology of rotifers has been little studied. A yet unknown property is presented within some monogonant rotifers, namely the ability to produce an exogenic filamentous biopolymer, named ‘Rotimer’. This rotifer-specific viscoelastic fiber was observed in six different freshwater monogonants (Euchlanis dilatata, Lecane bulla, Lepadella patella, Itura aurita, Colurella adriatica and Trichocerca iernis) in exception of four species. Induction of Rotimer secretion can only be achieved by mechanically irritating rotifer ciliate with administering different types (yeast cell skeleton, denatured BSA, epoxy, Carmine or urea crystals and micro-cellulose) and sizes (approx. from 2.5 to 50 µm diameter) of inert particles, as inductors or visualization by adhering particles. The thickness of this Rotimer is 33 ± 3 nm, detected by scanning electron microscope. This material has two structural formations (fiber or gluelike) in nano dimension. The existence of the novel adherent natural product becomes visible by forming a ‘Rotimer-Inductor Conglomerate’ (RIC) web structure within a few minutes. The RIC-producing capacity of animals, depends on viability, is significantly modified according to physiological- (depletion), drug- (toxin or stimulator) and environmental (temperature, salt content and pH) effects. The E. dilatata-produced RIC is affected by protein disruptors but is resistant to several chemical influences and its Rotimer component has an overwhelming cell (algae, yeast and human neuroblastoma) motility inhibitory effect, associated with low toxicity. This biopolymer-secretion-capacity is protective of rotifers against human-type beta-amyloid aggregates.

Diatoms, eukaryotic algae with silica cell wall have been proved to be reliable bioindicators and are applied in ecological status assessment of aquatic ecosystems using indices calculated from pollution sensitivity and indicator values of species. Traditional method of identification of diatom species is based on morphological features of frustule that requires in-depth taxonomical knowledge and expertise. Metabarcoding that combines barcodes and high-throughput sequencing offers a promising alternative. In this pilot study we tested the applicability of metabarcoding of benthic diatom assemblages for ecological status assessment of water bodies in Hungary comparing its performance to that of morphology-based identification of species. Samples from 78 rivers and streams arranged along a trophic gradient and 14 soda pans representing unique types of habitats were investigated. Sequences of the barcode region of rbcL gene suggested by Vasselon et al. (2017) were acquired revealing 1135 diatom amplicon sequence variants (ASVs) of which 54% could be assigned at species level using Diat.barcode (Rimet et al. 2019) as reference database. Morphological investigation detected more species and intraspecific taxa in the lotic and lentic samples (413 and 78, respectively) than metabarcoding (190 and 75). Correspondence between taxa detected based on morphology and DNA sequences was relatively low (on average 24% in lotic and 26% in lentic samples) but considerably increased (on average 66% and 56%, respectively) when taking into account only the taxa reaching higher than 5% relative abundance in microscopy counting. The differences mainly derived from that a considerable portion (59% and 32%, respectively) of the morphologically identified taxa were not recorded in the reference database. Community structure of samples from both running and standing waters based on microscopy and DNA sequence analysis showed significant correlation as revealed using Mantel test. For rivers and streams diatom index IPS (Specific Pollution Sensitivity Index, (Coste in Cemagref, 1982) for soda pans indices H (Ziemann et al., 2001) and IBD (Indice Biologique Diatomées, Lenoir and Coste, 1996) were calculated from the results of the two aspects. Strong correlation was found between morphology- and sequence-based indices and in about half of the samples, the ecological status class obtained with the two methods coincided. Our results suggest that metabarcoding inheres a great opportunity and could be successfully applied in benthic diatom-based ecological status assessment of Hungarian waters after the reference database is supplemented with taxa frequently occurring in these habitats. Although metacommunity dynamics of lentic phytoplankton are relatively well-documented, studies on the role of environmental and spatial processes in shaping phytoplankton communities of large rivers are still scarce. Here, we examined six phytoplankton data sets, which were collected in 1978-2017 from large river-scale segments (mean spatial extent 1117 km) in the Danube River. Our aim was to elucidate role of climatic, spatial and temporal predictors in variation of phytoplankton beta diversity using variance partitioning for compositions of species and functional groups sensu Reynolds. We hypothesised that phytoplankton beta diversity (measured as average distance to group centroid) would be positively related to both climatic heterogeneity and spatial extent used as a proxy for dispersal limitation. Additionally, we tested alternative dispersal models to evaluate different spatial processes structuring phytoplankton community. Our results revealed that spatial variables were more important than climatic factors in controlling both species and functional group composition. Climatic heterogeneity showed significant positive relationship with beta diversity. In contrast, there was no significant relationship between beta diversity and spatial extent, suggesting that spatial effect on beta-diversity was attenuated by anthropogenic disturbance. The better performance of non-directional model compared to model of water directionality suggested that spatial dynamics of phytoplankton metacommunity was in large part regulated by differences in the regional species pools. Spatial and temporal variables outperformed environmental (including climatic) factors in explaining phytoplankton metacommunity structure, indicating that phytoplankton exhibited strong biogeographical patterns. Thus, dispersal limitation interfered with species-sorting processes in determining phytoplankton community structure. In conclusion, our findings revealed that the development of a more reliable bioassessment program of the Danube River should be based on separation into basin regions.

Attachment of microorganisms to natural or artificial surfaces and the development of biofilms are complex processes which can be influenced by several factors. Nevertheless, our knowledge on biofilm formation in karstic environment is quite incomplete. The present study aimed to examine biofilm development for a year under controlled conditions in quasi-stagnant water of a hydrothermal spring cave located in the Buda Thermal Karst System (Hungary). Using a model system, we investigated how the structure of the biofilm is formed from the water and also how the growth rate of biofilm development takes place in this environment. Besides scanning electron microscopy, next-generation DNA sequencing was used to reveal the characteristic taxa and major shifts in the composition of the bacterial communities. Dynamic temporal changes were observed in the structure of bacterial communities. Bacterial richness and diversity increased during the biofilm formation, and 9–12 weeks were needed for the maturation. Increasing EPS production was also observed from the 9–12 weeks. The biofilm was different from the water that filled the cave pool, in terms of the taxonomic composition and metabolic potential of microorganisms. In these karstic environments, the formation of mature biofilm appears to take place relatively quickly, in a few months.

In the present study, the prokaryotic community structure of the water of Lake Balaton was investigated at the littoral region of three different points (Tihany, Balatonmáriafürdő and Keszthely) by cultivation independent methods [next-generation sequencing (NGS), specific PCRs and microscopy cell counting] to check the hidden microbial diversity of the lake. The taxon-specific PCRs did not show pathogenic bacteria but at Keszthely and Máriafürdő sites extended spectrum beta-lactamase-producing microorganisms could be detected. The bacterial as well as archaeal diversity of the water was high even when many taxa are still uncultivable. Based on NGS, the bacterial communities were dominated by Proteobacteria, Bacteroidetes and Actinobacteria, while the most frequent Archaea belonged to Woesearchaeia (Nanoarchaeota). The ratio of the detected taxa differed among the samples. Three different types of phototrophic groups appeared: Cyanobacteria (oxygenic phototrophic organisms), Chloroflexi (anaerobic, organotrophic bacteria) and the aerobic, anoxic photoheterotrophic group (AAPs). Members of Firmicutes appeared only with low abundance, and Enterobacteriales (order within Proteobacteria) were present also only in low numbers in all samples.

Recent decades have been marked by unprecendented environmental changes which threaten the integrity of freshwater systems and their ecological value. Although most of these changes can be attributed to human activities, disentagling natural and anthropogenic drivers remains a challenge. In this study, surface sediments from Lake Ighiel, a mid-altitude site in the Carpathian Mts (Romania) were investigated following high-resolution sedimentological, geochemical, environmental magnetic and diatom analyses supported by historical cartographic and documentary evidence. Our results suggest that between 1920 and 1960 the study area experienced no significant anthropogenic impact. An excellent correspondence is observed between lake proxy responses (e.g., growth of submerged macrophytes, high detrital input, shifts in diatom assemblages) and parameters tracking natural hydroclimate variability (e.g., temperature, NAO). This highlights a dominant natural hydroclimatic control on the lacustrine system. From 1960 however, the depositional regime shifted markedly from laminated to homogenous clays; since then geochemical and magnetic data document a trend of significant (and on-going) subsurface erosion across the catchment. This is paralleled by a shift in lake ecosystem conditions denoting a strong response to an intensified anthropogenic impact, mainly through forestry. An increase in detrital input and marked changes in the diatom community are observed over the last three decades, alongside accelerated sedimentation rates following enhanced grazing and deforestation in the catchment. Recent shifts in diatom assemblages may also reflect forcing from atmospheric nitrogen (N) deposition, a key recent drive of diatom community turnover in mountain lakes. In general, enhanced human pressure alongside intermittent hydroclimate forcing drastically altered the landscape around Lake Ighiel and thus, the sedimentation regime and the ecosystem's health. However, paleoenvironmental signals tracking natural hydroclimate variability are also clearly discernible in the proxy data. Our work illustrates the complex link between the drivers of catchment-scale impacts on one hand, and lake proxy responses on the other, highlighting the importance of an integrated historical and palaeolimnological approach to better assess lake system changes.

Here we report the results of our decades-long study on epiphytic communities from two tributary systems of the Szigetk&ouml, z section of the Danube River. The main goal of the investigation was to detect changes in the epiphytic communities at structural (core species, changes in the relative abundance of common species) and functional (trait changes) levels as a result of the most important anthropogenic effects on Szigetk&ouml, z, i.e., hydro-morphological modifications. We also examined the impact of rehabilitation on the tributary systems in terms of ecological potential. We discovered that mainly motile diatom species characterized the epiphyton due to reduced water volume were introduced into the tributary system after the diversion of the Danube. The ecosystem stabilized in the rehabilitated section, while the non-rehabilitated section showed a worsening tendency, mainly in the parapotamic branches. Our long-term data sets may provide a good basis for comparisons of different aquatic ecosystems, to define changes in the abundance of core species and in the structure of community in response to different anthropogenic pressures. It is fundamental to determine adaptive traits in assessing the impact of global warming stressors on biodiversity.

The Sakarya River basin is one of the largest basins in Turkey, and encompasses the Kocaeli, D&uuml, zce, Sakarya, Bursa, Bilecik, Bolu, K&uuml, tahya, Eskişehir, Ankara, Afyon, and Konya provinces. In this study, the water quality status of the basin was investigated using 18 diatom indices, calculated in Omnidia software. For this purpose, a total of 46 stations were surveyed in the rivers and streams of the basin in May 2018. As a result, 41 of 195 diatom taxa were found to be the most frequent (&gt, 10% share in assemblage). According to Detrented Correspondence Analysis (DCA), three subgroups were described as the spring section, Ankara and Polatlı section, and lowland section. The river basin quality was evaluated as moderate or lower quality status, while only a few sites had good status. The diatom index scores showed that the Descy&rsquo, s Index (DES), Pampean Diatom Index (IDP), Artois-Picardie Diatom Index (IDAP), and Specific Pollution Sensitivity Index (IPS) appear best suited to water quality assessment in this area, showing the largest number of significantly important correlation with environmental variables.

The morphology and ontogenetic allometric trends of a rare diatom Fragilaria heidenii Ostrup and the morphologically related Staurosira tabellaria (W. Smith) Leuduger-Fortmorel were compared using conventional and semilandmark-based geometric morphometric analyses. Fragilaria heidenii was studied in detail by light and electron microscopy using type material and recent samples from Lake Ladoga and Lake Ilmen (north-western Russia). The taxon is transferred into the genus Staurosira Ehrenberg as Staurosira inflata comb. nov. on the basis of its valve morphology. This taxon is characterized by the absence of rimoportulae, lack of perforated copulae, spines located on the interstriae, internal vola occlusion in the areolae and the features of the areolae and apical pore fields. Here we report the first occurrence of the diatom Gomphonema sancti-naumii D. Metzeltin & Z. Levkov beside the type locality, during the Last Glacial Maximum. The species presented here with its morphological characteristics demonstrated on detailed scanning electron microscope (SEM) pictures and its palaeolimnological significance in diatom-based pH reconstruction is proclaimed. The species was observed in the material collected from a borecore obtained from a lake sediment core of Lake St. Anne (Romania) between 16 and 15 kyr cal BP. Gomphonema sanctinaumii occurred in low abundance with other narrow valved (< 5 μm) gomphonemoid taxa.

Neurodegenerative diseases are linked to a systemic enzyme resistance of toxic aggregated molecules and their pathological consequences. This paper presents a unique phenomenon that Philodina acuticornis, a bdelloid rotifer, is able to catabolize different types of neurotoxic peptide and protein aggregates (such as beta-amyloids /Aβ/, alpha-synuclein, and prion) without suffering any damage. P. acuticornis is capable of using these aggregates as an exclusive energy source (i.e., as ‘food’, identified in the digestive system and body) in a hermetically isolated microdrop environment, increasing their survival. As regards Aβ1–42, five other bdelloid rotifer species were also found to be able to perform this phenomenon. Based on our experiments, the Aβ1–42-treated bdelloid rotifers demonstrate significantly increased survival (e.g. mean lifespan = 51 ± 2.71 days) compared to their untreated controls (e.g. mean lifespan = 14 ± 2.29 days), with similar improvements in a variety of phenotypic characteristics. To our knowledge, no other animal species have so far been reported to have a similar capability. For all other microscopic species tested, including monogonant rotifers and non-rotifers, the treatment with Aβ1–42 aggregates proved to be either toxic or simply ineffective. This paper describes and proves the existence of an unprecedented in vivo catabolic capability of neurotoxic aggregates by bdelloid rotifers, with special focus on P. acuticornis. Our results may provide the basis for a new preclinical perspective on therapeutic research in human neurodegenerative diseases.

In our work, we sought to answer whether we find differences among the various zones of an oxbow lake with different land uses based on physico-chemical variables and dominant algal plankton species. The two ends of the oxbow lake are bordered by settlements, and near them there are open water areas where fishing is the major utilization form. Between the two open water areas we find a protected area with a large aquatic plant coverage and two transition zones towards the open water areas. The oxbow lake receives periodic water replenishment only at one end from one of the open water areas. During summer—due to the lack of rain—the water of the oxbow lake is used for irrigation in the surrounding arable land, so the water level fluctuation can be significant in the riverbed. Our study was performed within a vegetation period of spring, early summer, mid-summer, and fall. In connection with the ecological classification of a smaller water body, studies on the physical and chemical properties of the water and the composition of the algal plankton are usually carried out in few places and relatively infrequently. The characteristics of a water body are also influenced by seasonal changes, which can be the changes in the extent of vegetation coverage, the way land is used and the possibility of water replenishment, to which the algal community usually responds with changes. Based on our study, it can be said that even in a relatively small water body, we found a large differences based on the chemical and physical properties of the water and the characteristic algal species. Open water zones, areas with large macrovegetation coverage, and the transition zones were separated from each other.

While phytoplankton studies on large potamal rivers have increased in number in recent years, upper river sections have received considerably less attention. However, in order to better understand processes that govern the development of dominance of euplanktonic elements in the lower river sections, detailed studies of the upstream areas are necessary. We studied the composition, diversity and recruitment of the planktonic algal communities in the River Sajo and in its main tributary River Hernad, two characteristic rhithral river systems in Central Europe. Results revealed that diatoms are dominant elements of the phytoplankton in the upper river segments both in terms of taxa richness, and relative abundance. We found that composition of the phytoplankton showed the closest resemblance to that of benthic communities of soft substrates, highlighting the key role of riverbed characteristics in river phytoplankton recruitment. The occurrence of euplanktonic elements was not restricted to human impacted river segments, these taxa also occurred in pristine headwaters. Accordingly, planktonic algae potentially colonise small headwater streams naturally, although their dominance is expected to occur only at downstream reaches following impounded river segments. Diatom metrics used in ecological state assessment were calculated for the rithroplankton. These metrics fell in the range of values calculated for diatom flora on the hard substrates. However, the potential use of rithroplankton for quality assessment might be limited, because the high variability of index values at site level would result in misclassification of the ecological status.

Dissipation of the herbicide active ingredient glyphosate was investigated in natural waters. To assess combined effects, glyphosate was applied in its pure form (glyphosate isopropylammonium salt) and in preparation Roundup Classic® formulated with polyethoxylated tallowamines (POEA). Standing and running surface water samples originated from Lake Balaton and River Danube between early May and mid-June of 2015. The kinetics of dissipation of glyphosate, measured by high-performance liquid chromatography combined with UV-VIS absorbance detection or tandem mass spectrometry, was investigated under laboratory conditions in aquaria with or without the presence of biofilms. Considering the ongoing loss of aquatic habitats, anthropogenic ponds are gaining importance as substitute habitats. It is therefore important to assess their functioning in comparison to their natural precursors. Here we assess the biodiversity value of sodic bomb crater ponds by comparing their gamma diversity to that of natural reference habitats, astatic soda pans, and assess their importance on the landscape level by studying alpha and beta diversity. We studied aquatic organisms ranging from algae to vertebrates in a dense cluster of 54 sodic bomb crater ponds in Central Europe. Despite the overall small area of the pond cluster, gamma diversity was comparable to that found in surveys of natural habitats that encompassed much wider spatial and temporal scales. We also found a considerable number of species shared with reference habitats, indicating that these anthropogenic habitats function as important refuge sites for several species that are associated with the endangered soda pans. Moreover, we found a number of regionally or worldwide rare species. Among the components of beta diversity, species replacement dominated community assembly. Individual ponds contributed similarly to beta diversity in terms of replacement, being equally important for maintaining high gamma diversity and emphasising the role of the pond network rather than individual ponds. This pattern was seen in all studied groups. Bomb crater ponds therefore acted as important contributors to aquatic biodiversity. Considering the tremendous losses of ponds throughout Europe, anthropogenic ponds should be taken into consideration in nature conservation, especially when occurring in pond networks.

Specimens of a very small diatom were found in a dead oxbow of Tisza River (Hungary), in Eula River (Germany) and Parroy Pond (France). After SEM studies we identified all of them as Eolimna archibaldii J.C. Taylor et Lange-Bert., which is currently regarded as endemic species to South Africa. Furthermore, the results of Principal Components Analysis of morphometric characters clearly demonstrated an overlap between the type material and European populations. Therefore this diatom cannot be considered endemic to South Africa. Moreover, E. archibaldii and a very similar species, E. becaresii are transferred to the genus Sellaphora. Our results confirm the benefit of SEM for verifying the identity of small-sized diatoms, especially for routine monitoring, and allow more precise ecological assessment.

During our work, we investigated the physical and chemical variables of a small surface watercourse to investigate how different anthropogenic effects affect its water quality. Along this small watercourse, there are well-separated areas that are affected by various anthropogenic effects. In addition to its origin and branches, in many places it is surrounded by agricultural land with insufficient buffer zones, which burdens the small watercourse with nitrogen and phosphorus forms. In the lower stages, artificial damming inhibits the natural flow of the T&oacute, c&oacute, Canal, thereby creating eutrophicated stagnant water areas. This is further strengthened by, in many cases, illegal communal and used water intake that further burdens the small watercourse. Considering the experience of our investigation, it can be stated that the examined small watercourse could barely suffer human impacts, and it could be described with great heterogeneity using physical and chemical variables. We experienced that this heterogeneity caused by anthropogenic effects appeared in all hydrologic states and seasons. Furthermore, our research showed that these small watercourses had such high heterogeneity that their monitoring and examination should be taken just as seriously as when it comes to larger watercourses.

The stress dominance hypothesis (SDH) postulates that strong environmental gradients drive trait convergence in communities over limiting similarity. Previous studies, conducted mostly with terrestrial plant communities, found controversial evidence for this prediction. We provide here the first test for SDH for epiphytic diatoms. We studied community assembly in diatom communities of astatic ponds. These water bodies serve as a good model system for testing SDH because they exhibit stress gradients of various environmental factors. Functional diversity of diatom communities was assessed based on four traits: (1) combined trait reflecting the trade-off between stress tolerance and competitive dominance, (2) cell size, (3) oxygen requirement and (4) N-uptake strategy. According to our results, salinity, pH and the width of the macrophyte belt appeared as significant predictors of the trait convergence/divergence patterns presumably acting through influencing the availability of carbon dioxide and turbidity. Lower trait diversity was found in turbid, more saline and more alkaline ponds and functional diversity was higher in transparent, less saline and less alkaline ponds. Overall, our results supported the stress dominance hypothesis. In habitats representing increased environmental stress, environmental filtering was the most important community assembly rule, while limiting similarity became dominant under more favourable conditions.

Using this method, we produced viable larvae via in vitro fertilisation. The aim of this study was to investigate the time-dependent fertilizing capacity of sperm which were introduced into ovary. (1) Urbanization significantly influences the ecosystems of rivers in various ways, including the so-called loading effect of wastewater production. Benthic diatoms are used in ecological status assessments of waters. Beside species composition, traits can be used as indicators. We aimed to evaluate how the loading of the large city of Budapest manifests in the physico-chemical variables of the River Danube and what species composition and trait response this loading results in for the benthic diatom communities. (2) Weekly samplings were performed at points upstream and downstream of Budapest on both riverbanks. Samples were compared, based on general physical-chemical variables and the concentration of thirty-four elements, as well as species composition and seven traits of species of diatom communities. Ecological status was assessed using the Specific Pollution Sensitivity Index (IPS). (3) Only a few measured environmental variables showed differences between the sampling points, suggesting that the nutrient loading has significantly decreased due to the installation of several efficiently working wastewater treatment plants since the introduction of the European Union Water Framework Directive. In contrast, the species composition and traits of species showed the effect of land use. Benthic diatoms indicate the environmental changes caused by land use in the longer-term, while chemical measurements reflect instantaneous status.
