Abstract

Aim: Cardiac hepatopathy arises due to heart failure and influences has effects on heart recovery after myocardial infarction (MI).The aim of this study was to investigate the protective effect of Erythropoietin (EPO) on liver tissue exposed to ischemia due to MI.Material and Methods: Experimental MI was established by left anterior descending coronary artery ligation (CAL) and EPO or saline was injected immediately after CAL to five groups of rats, which groups are Control, Saline, EPO 5000, EPO 10000, CAL+1h. CAL+1h group was sacrificed one hour after CAL without any treatment. Other groups were sacrificed six hours after the operation. Liver tissues were examined histopathologically by Hematoxylin Eosin (HE) staining and electron microscopy. Results: Degenerative changes in liver tissue such as vacuolization, sinusoidal dilatation, hepatocyte pyknosis, Kuppfer cell activation were observed. Vacuolization, and sinusoidal dilatation increased in the Saline group compared to the control group (p=0.010 for both). Degenerated hepatocytes with pyknotic nuclei as well as activated Kuppfer cells were decreased in the EPO 10000 group compared to the Saline group (p=0.009), and activated Kupfer cells were decreased compared to the Saline and CAL+1h groups (p=0.035 and p=0.019, respectively).Conclusion: EPO protected liver tissue from histopathological damages regardless of dose, when given at the time of MI. EPO, when given immediately after MI, protected liver tissue from histopathological damage regardless of dose. Considering the mutual interaction of liver and heart, applying EPO to MI patients at first sight may prevent post-MI liver damage and contribute to the recovery of the heart., Amaç: Kardiyak hepatopati, kalp yetmezliğine bağlı olarak ortaya çıkar ve miyokard infarktüsü (MI) sonrası kalbin iyileşmesini etkiler. kalp dokusu iyileşmesi üzerinde etkileri bulunmaktadır. Bu çalışmanın amacı, MI nedeniyle iskemiye maruz kalan karaciğer dokusu üzerinde Eritropoetinin (EPO) koruyucu etkisinin araştırılmasıdır. Gereç ve Yöntemler: Sol ön inen koroner arter ligasyonu (KAL) ile deneysel MI oluşturuldu ve Kontrol, SF (serum fizyolojik), EPO 5000, EPO 10000, KAL+1s. olmak üzere beş grup sıçana KAL'dan hemen sonra EPO veya SF enjekte edildi: KAL+1s grubu KAL'dan bir saat sonra herhangi bir tedavi uygulanmadan sakrifiye edildi. Diğer gruplar, operasyondan altı saat sonra sakrifiye edildi. Karaciğer dokuları Hematoksilen Eozin (HE) boyama ve elektron mikroskobu ile histopatolojik olarak incelendi.Bulgular: Karaciğer dokusunda vakuolizasyon, sinüzoidal dilatasyon, hepatosit piknozu, Kuppfer hücre aktivasyonu gibi dejeneratif değişiklikler gözlendi. SF grubunda vakuolizasyon ve sinüzoidal dilatasyon Kontrol grubuna göre arttı (her ikisi için p=0,010). EPO 10000 grubunda piknotik çekirdekli dejenere hepatositler SF grubuna göre azalırken (p=0,009), ve aktive Kuppfer hücreleri SF ve KAL+1s gruplarına göre azaldı (sırasıyla p=0,035 ve p=0,019).Sonuç: EPO, MI sırasında meydana gelmesinden hemen sonra verildiğinde, dozdan bağımsız olarak karaciğer dokusunu histopatolojik hasarlanmadan korumuştur. Karaciğer ve kalbin karşılıklı etkileşimi göz önüne alındığında, MI hastalarına ilk görüşte EPO uygulanması, MI sonrası karaciğer hasarını önleyebilir ve kalbin iyileşmesine katkıda bulunabilir.

Abstract

Aim: Studies on drugs or alternative therapies are still the main treatment options for PNI. In this study, we aimed to research the effects of PBM and OT on nerve repair in a rat sciatic injury model. Material and Methods: 29 Wistar albino rats were divided into four groups: control (n = 2), sham (n = 9), OT (n = 9) and PBM (n = 9). After 30 days of surgery and treatments, tissue specimens and blood samples were taken for histological and biochemical processing. Histological evaluations were performed at light and electron microscopy levels. Myelin basic protein (MBP) and S100 from the rat serum were analysed also. Results: The OT and PBM groups had a significant increase in regeneration of the sciatic nerve in light microscopic evaluation. In the PBM and OT groups, Schwann cells (SC) around the axons and also axons with a thin myelin sheath were seen, regarded as signs of the myelination process in transmission electron microscopy (TEM) examinations. Discussion: OT and PBM both resulted in a good healing pattern for sciatic nerve injury in the rat model. Therefore, OT and PBM are considered to be simple and reliable alternative treatment methods for PNI.

© 2021 European Journal of Biology. All rights reserved.Objective: This study aims to investigate the effect of Momordica charantia L. (MoC) ethanolic extract on ulcerative colitis (UC) and was explored in vitro and in vivo. Materials and Methods: The rats were divided into control (C), saline-treated colitis (AA), MoC extract-treated colitis (AA+MoC), and sulfasalazine (SS)-treated colitis (AA+SS) groups. Colitis was induced by acetic acid. MoC extract, SS or saline were given to the related groups for 3 days. Interleukine-1β, malondialdehyde, glutathione levels, myeloperoxidase activity, bax/bcl-2 ratio, caspase-9 and caspase-3 levels were measured in colon and macroscopic and histopathologic examinations were done. Total phenolic/flavonoid content and biological activity of MoC were evaluated by in vitro analysis. Results: Compared to the control group, with acetic acid application interleukin-1β levels, myeloperoxidase activity, malondialdehyde levels, bax/bcl-2 ratio, caspase-9 and caspase-3 levels were significantly upregulated, while glutathione levels were significantly decreased in the AA group. In contrast, MoC and SS treatments reduced interleukin-1β, malondialdehyde levels, myeloperoxidase activity, bax/bcl-2 ratio, and caspase-9 and caspase-3 levels. Glutathione levels increased upon MoC or SS treatment. Increased macroscopic and microscopic scoring with AA improved with MoC or SS treatment, but the MoC or SS treated groups had higher score values than the control. Also, in vitro results showed that MoC exhibited 2,2-diphenyl-1- picrylhydrazyl and 2,2\"-azino-bis-3-ethylbenzothiazoline-6-sulfonic acid radical scavenging activity as well as significant antilipoxygenase activity. In addition, MoC extract showed a potent anti-inflammatory activity compared to standard indomethacin. Conclusion: Our biochemical, in vitro and histopathologic analysis indicate that MoC is likely to prove beneficial in UC therapy.

14. Effect of astaxanthin on uterus tissues in ovariectomized rats: a histopathological study

Abstract

Öz Amaç: Güçlü bir antioksidan olarak bilinen astaksantin (AST), inflamasyona karşı yararlı etkilere sahiptir. Bu çalışmada, overektomili sıçanlarda astaksantinin uterus dokuları üzerindeki etkisi araştırılmıştır. Gereç ve Yöntem: Sprague-Dawley sıçanlar (n = 18); rastgele üç gruba ayrıldı (n = 6): sağlıklı kontrol, overektomi (OVT) ve overektomi+ astaksantin (OVT + AST). AST (1 mg / ml), overektomiden dört hafta önce başlanarak günlük 50 ml su ile deney grubuna oral yoldan verildi. AST tedavisi overektomi sonrası dört hafta daha sürdürüldü. Sekizinci haftanın sonunda, hayvanlar servikal dislokasyon ile ötenazi edildi. Ötenazi sonrası uterus formalin ile sabitlendi ve Hematoksilin-Eozin boyaması ile histolojik açıdan incelendi. Bulgular: OVT ile ortaya çıkan uterus epitelinde apiko-bazal yükseklikteki azalma ve hücresel bozulma, uterus bezlerindeki dejenerasyon ve stromadaki inflamasyon belirtileri, AST uygulanan sıçanlarda kontrole yakın düzelme göstermiştir. Sonuç: Tuba uterina, uterus ve vajinada menopoz sonrası ortaya çıkan değişiklikler menopozun klinik bulgularıyla yakından ilişkilidir. Astaksantinin bu dokular üzerindeki olumlu etkisinin mekanizmasının araştırılması gerekmektedir

15. Therapeutic Potential of Essential Oil of Melaleuca Quinquenervia (Myrtaceae) in a Rat Model of Ethanol-Induced Peptic Ulcer

Abstract

Purpose: To evaluate the effects of M. quinquenervia extract on ethanol-induced peptic ulcer in rats. Methods: The following three groups of (n = 6) Sprague Dawley rats were included in this study: vehicle (C), ethanol-administered (E) and ethanol + M. quinquenervia-treated (MQ). MQ group rats received 100 μg/mL M. quinquenervia essential oil just before 96 % ethanol induction (1 ml/kg). One hour after ulcer induction, the animals were euthanized, and gastric and duodenal tissues were removed. Tissue samples were analysed for myeloperoxidase (MPO) activity, malondialdehyde (MDA), glutathione (GSH), myeloperoxidase (MPO) activity, malondialdehyde (MDA) and glutathione (GSH) levels, and histopathological examinations were performed by light microscopy. Results: Gastric and duodenal GSH levels that decreased in the ethanol-administered ulcer groups (p < 0.001), rose following MQ treatment (p < 0.5). Moreover, elevated MPO and MDA levels (p < 0.5) in gastric tissues decreased after MQ-treatment. Similarly, the MQ-treated group showed recovery and control-like morphology compared to the ethanol group in both gastric and duodenal tissues when examined by microscopy. Conclusion: The results indicate that M. quinquenervia extract has a positive impact on gastric injury in rats due to its antioxidant activity. Thus, the plant has a potential for the clinical management of gastric ulcer.

16. The Efficacy of Hydrodilatation for the Prevention of Vasospasm following Microsurgical Anastomosis.

Abstract

Background  Vasospasm is a major problem following microsurgical reconstruction which can result in the partial or complete loss of the flap tissue. The aim of this study was to investigate the efficiency of hydrodilatation for the prevention of vasospasm. Methods  Thirty male Wistar rats were used for this experimental study. Femoral arteries of were exposed, photographed, and transected. In group 1, group 2, and group 3 papaverine solution, hydrodilatation, and minimal mechanical dilatation (control group) was performed, respectively. The anastomosis was completed and the arteries were photographed again 10 minutes after completion of the anastomosis. Following 7-day period samples for transmission electron microscopy (TEM) and light microscopy were obtained. Results  The mean vessel diameters prior to transection were 0.43, 0.45, and 0.52 mm in the papaverine, hydrodilatation, and control groups, respectively. The mean vessel diameter 10 minutes following the completion of anastomosis was 0.76, 0.75, and 0.51 mm in the papaverine, hydrodilatation, and control groups, respectively. Median score for papaverine group regarding histological parameters of regular endothelial lining and lumen, neutrophil infiltration, vascular congestion, and edema in tunica adventitia was 2, 3, 2, and 3 positive, respectively. Median score for the papaverine group regarding histological parameters of regular endothelial lining and lumen, neutrophil infiltration, vascular congestion, and edema in tunica adventitia was 3, 3, 3, and 3 positive, respectively. All the histological scores were negative in the control group. The difference between the control group and the experiment groups 1 and 2 was significant regarding all four histological parameters (p  < 0.05). Conclusion  Hydrodilatation and papaverine application were both effective in preventing vasospasm following microsurgical intervention but papaverine caused slightly less damage to the endothelial lining and less edema in the tunica adventitia when compared with the hydrodilatation. Hydrodilatation group showed a vasodilatory effect that was statistically similar to that of papaverine, which has a proven efficacy. [ABSTRACT FROM AUTHOR]

22. The Effects of Memantine on Cisplatin-Induced Ototoxicity.

Abstract

Introduction: We aimed to investigate electrophysiologically and histopathologically, the protective effects of intratympanic memantine, an N-methyl-D-aspartate receptor antagonist, on ototoxicity caused by cisplatin, an anti-neoplastic agent used in many types of cancer. Methods: Thirty-seven guinea pigs with a normal auditory function were randomly allocated to group 1 (cisplatin; n = 8), group 2 (memantine; n = 8), group 3 (cisplatin + memantine; n = 8), group 4 (cisplatin + physiological serum [PS]; n = 8), and group 5 (control; n = 5). Auditory assessments were conducted using distortion product otoacoustic emissions (DPOAE) within a frequency range of 1–32 kHz and auditory brainstem responses (ABRs) within 8–32 kHz. A single dose of cisplatin (12 mg/kg) was administered intraperitoneally, followed by intratympanic administration of 0.2 mL of either memantine or PS to both ears at least half an hour before cisplatin administration. Subsequent auditory evaluations were conducted 72 h after cisplatin administration. Histopathological analyses were performed using light microscopy of the right ear and scanning electron microscopy (SEM) of the left ear. Results: Auditory evaluations conducted before and after treatment revealed significant findings. Specifically, within groups 3 and 4, ABR thresholds were elevated at all frequencies (p = 0.00), whereas the DPOAE signal-to-noise ratios were reduced at frequencies of 8, 12, 16, and 24 kHz (p = 0.001, p = 0.01, p = 0.01, and p = 0.00, respectively). Histopathologically, both light microscopy and SEM revealed that the cisplatin + memantine group exhibited fewer hair cells and nuclear degeneration in the spiral ganglion than the cisplatin and cisplatin + PS groups. Additionally, the stria vascularis thickness was greater in the cisplatin + memantine group than in cisplatin and cisplatin + PS groups. Conclusion: Despite the negative electrophysiological findings, the histopathological outcomes suggest that intratympanic memantine may have a potential protective effect against cisplatin-induced ototoxicity. However, further investigations are warranted to corroborate these findings and elucidate the underlying mechanisms of action of memantine. [ABSTRACT FROM AUTHOR]

Abstract

Objective: The aim of the present study was to determine the effect of topical and systemic tranexamic acid (TXA) on fracture healing in a rat surgical model., Methods: We created standard, right-sided, open, diaphyseal femoral fractures with intramedullary Kirschner wire fixation in 48 male rats and divided them into three groups: a topical TXA (10 mg/kg) group, a systemic TXA (10 mg/kg) group, and a control group. Fracture healing was evaluated radiographically and histologically after early (week 2) and late (week 4) postoperative sacrifice., Results: The radiological scores differed significantly among the all groups (p=0.001), as did the week 2 and 4 scores (p=0.003 and p=0.010, respectively). Radiologically, the topical TXA group exhibited better bone healing at both 2 (p=0.001) and 4 (p=0.007) weeks than the control group, and the systemic group showed better healing at both 2 (p=0.027) and 4 (p=0.023) weeks than the control TXA group. Moreover, bone healing was better in the group treated with topical rather than systemic TXA on radiological examinations performed at 2 (p=0.001) and 4 (p=0.007) weeks postoperatively (p=0.001 and p=0.007, respectively). Histologically, the groups differed significantly (p=0.001). The histological scores differed significantly among the all groups (p=0.001). At 2 weeks, the topical TXA group exhibited significantly better bone healing than the control group (p=0.001)., Conclusion: Our results suggested that topical application of TXA in fracture patients may accelerate healing, whereas systemic administration may adversely affect healing.
