Females versus males are less frequently diagnosed with autism spectrum disorder (ASD), and while understanding sex differences is critical to delineating the systems biology of the condition, female ASD is understudied. We integrated functional MRI and genetic data in a sex-balanced sample of ASD and typically developing youth (8-17 years old) to characterize female-specific pathways of ASD risk. Our primary objectives were to: (i) characterize female ASD (n = 45) brain response to human motion, relative to matched typically developing female youth (n = 45); and (ii) evaluate whether genetic data could provide further insight into the potential relevance of these brain functional differences. For our first objective we found that ASD females showed markedly reduced response versus typically developing females, particularly in sensorimotor, striatal, and frontal regions. This difference between ASD and typically developing females does not resemble differences between ASD (n = 47) and typically developing males (n = 47), even though neural response did not significantly differ between female and male ASD. For our second objective, we found that ASD females (n = 61), versus males (n = 66), showed larger median size of rare copy number variants containing gene(s) expressed in early life (10 postconceptual weeks to 2 years) in regions implicated by the typically developing female > female functional MRI contrast. Post hoc analyses suggested this difference was primarily driven by copy number variants containing gene(s) expressed in striatum. This striatal finding was reproducible among n = 2075 probands (291 female) from an independent cohort. Together, our findings suggest that striatal impacts may contribute to pathways of risk in female ASD and advocate caution in drawing conclusions regarding female ASD based on male-predominant cohorts. In this paper, the effect of pre-buckling boundary conditions and the type of nonlinearity for stress stiffening used in different linear and nonlinear buckling approaches is studied for laminated composite plates. The study is conducted using a C 0 finite element (FE) plate model, employing a unified C 1 higher-order shear deformation theory (HSDT). The set of governing equations is derived using the principle of virtual displacement and solved using the tangent-based arc-length method in conjunction with a simple branch switching technique. The performance of the present C 0 FE model is assessed through a validation exercise and comparison with results obtained via the use of ANSYS and, for linear analysis, Navier solution, as well as solutions available in the literature. The influence of the different in-plane loads, boundary conditions, side-to-thickness ratio, fiber orientation, types of imperfection and penalty stiffness matrix are also examined. The results show that the same boundary conditions must be utilized in both pre-buckling and linear eigenvalue analyses for accurate and realistic predictions of critical buckling loads, as confirmed from the nonlinear buckling analyses. Furthermore, the critical buckling loads obtained using Green–Lagrange nonlinearity are observed to be more conservative than those obtained using von Kármán nonlinearity. The nonlinear buckling approach is a generalized approach while the nonlinear eigenvalue approach has a limited range of application. • Importance of consistent pre-boundary conditions in pre-buckling analysis is highlighted. • Green–Lagrange strain is better than von Kármán strain for geometric stiffness matrix. • The superiority of the nonlinear buckling approach over the nonlinear eigenvalue approach is shown. • The significance of considering the penalty stiffness matrix for C1 HSDT in C0 FEM formulation is demonstrated. Heat Shock Protein 90 (HSP90) is a potential drug target for cancer therapy as it is often dysregulated in several cancers, including lung, breast, pancreatic, and prostate cancers. In cancer, HSP90 fails to maintain the structural and functional integrity of its several client proteins which are involved in the hallmarks of cancer such as cell proliferation, invasion, migration, angiogenesis, and apoptosis. Several small molecule inhibitors of HSP90 have been shown to exhibit anticancer effects in vitro and in vivo animal models. However, a few of them are currently under clinical studies. The status and potential limitations of these inhibitors are discussed here. Studies demonstrate that several noncoding RNAs (ncRNAs) such as microRNAs (miRNAs) and long noncoding RNAs (lncRNAs) regulate HSP90 and its client proteins to modulate cellular processes to exhibit oncogenic or tumor suppressing properties. Over the last decade, miRNAs and lncRNAs have drawn significant interest from the scientific community as therapeutic agents or targets for clinical applications. Here, we discuss the detailed mechanistic regulation of HSP90 and its client proteins by ncRNAs. Moreover, we highlight the significance of these ncRNAs as potential therapeutic agents/targets, and the challenges associated with ncRNA-based therapies. Describes the training provided to tutors and trainers of adult learning centers in Hampton Roads, Virginia. Results show that trainees, prior to training, had limited access to computer technology and needed training in incorporating computer technology in the curriculum. After training, trainees have shown increased levels of comfort with various software, including word processing programs such as MS Word, Power Point, Netscape, Hyperstudio, and MS Frontpage. Background: The role of denosumab in patients with resectable giant cell tumour of bone remains unclear. We asked the following research question: for patients (aged ≥ 12 years) with resectable giant cell tumour of bone, what are the benefits and harms of denosumab compared with no denosumab in terms of (1) facilitation of surgery (operative time, blood loss), (2) disease recurrence, (3) pain control, (4) disease stability, and (5) adverse effects (e.g., malignant transformation, osteonecrosis of jaw, atypical femur fracture)? One previous systematic review addressed only one outcome—disease recurrence. Therefore, we undertook this new systematic review to address the above five outcomes. Methods: MEDLINE, EMBASE, PubMed, and Cochrane Database of Systematic Reviews databases were searched on June 30, 2020. Results: This systematic review included one previous systematic review and five comparative studies. Due to poor quality, non-randomized studies fraught with selection bias, it is difficult to determine if a significant difference exists in the outcomes for surgical giant cell tumour of bone with perioperative denosumab. There were no reported cases of adverse effects from denosumab. Conclusion: To date, there is insufficient evidence to understand the value of denosumab in the perioperative setting in patients with giant cell tumour of bone. [ABSTRACT FROM AUTHOR]

The aim of this paper is to investigate the clinical utility of the application of deep learning denoise algorithms on standard wide-field Optical Coherence Tomography Angiography (OCT-A) images. This was a retrospective case-series assessing forty-nine 10 x 10 mm OCT-A1 macula scans of 49 consecutive patients attending a medical retina clinic over a 6-month period. Thirty-seven patients had pathology; 13 had none. Retinal vascular layers were categorised into superficial or deep capillary plexus. For each category, the retinal experts compared the original standard image with the same image that had intelligent denoise applied. When analysing the Superficial Capillary Plexus (SCP), the denoised image was selected as "best for clinical assessment" in 98% of comparisons. No difference was established in the remaining 2%. On evaluating the Deep Capillary Plexus (DCP), the denoised image was preferred in 35% of comparisons. No difference was found in 65%. There was no evidence of new artefactual features nor loss of anatomical detail in denoised compared to the standard images. The wide-field denoise feature of the Canon Xephilio OCT-A1 produced scans that were clinically preferable over their original OCT-A images, especially for SCP assessment, without evidence for causing a new artefactual error. Survivors of adolescent and young adult cancers (AYAs) often live 50 to 60 years beyond their diagnosis. This rapidly growing cohort is at increased risk for cancer- and treatment-related 'late effects' that persist for decades into survivorship. Recognition of similar issues in pediatric cancer survivors has prompted the development of evidence-based guidelines for late effects screening and care. However, corresponding evidence-based guidelines for AYAs have not been developed. We hosted an AYA survivorship symposium for a large group of multidisciplinary AYA stakeholders (approximately 200 were in attendance) at Princess Margaret Cancer Centre (Toronto, Ontario, Canada) to begin addressing this disparity. The following overview briefly summarizes and discusses the symposium's stakeholder-identified high-priority targets for late effects screening and care and highlights knowledge gaps to direct future research in the field of AYA survivorship. This overview, although not exhaustive, is intended to stimulate clinicians to consider these high-priority screening and care targets when seeing survivors in clinical settings and, ultimately, to support the development of evidence-based late effects screening and care guidelines for AYAs. Abstract Background Ocular damage from radiation treatment is a well established phenomenon. Many factors are now known to influence the incidence of radiation retinopathy, including total dosage and daily fraction size. Patients who are diabetic, hypertensive or received previous chemotherapy are more susceptible to radiation retinopathy. Case Presentation A 55 year old male was referred from the oncology department with epiphora. His medical history included Type 2 Insulin treated Diabetes Mellitus and hypertension. One year prior to presentation he had undergone a total rhinectomy with a 4 week course of post-operative radiotherapy for an aggressive sqaumous cell carcinoma of the nose. On examination the visual acuity was noted to be 6/36 left eye and 6/9 right eye. Posterior segment examination revealed marked retinal ischaemia present in the posterior pole and macular region of both eyes. The appearance was not thought to be typical of diabetic changes, radiation retinopathy being the more likely diagnosis especially in view of his history. Over the next four months the vision in both eyes rapidly deteriorated to 3/60 left eye and 1/60 right eye. Bilateral pan retinal photocoagulation was thought to be appropriate treatment at this point. Conclusion This case highlights the importance for ophthalmologists and oncologists to be aware of the close relationship between diabetes and radiation treatment and the profound rapid impact this combination of factors may have on visual function. Radiation is being used with increasing frequency for ocular and orbital disease, because of this more cases of radiation retinopathy may become prevalent. Factors which may potentiate radiation retinopathy should be well known including, increased radiation dosage, increased fraction size, concomitant systemic vascular disease and use of chemotherapy. Counselling should be offered in all cases at risk of visual loss. As no effective treatment currently exists to restore visual function, monitoring of visual acuity in all cases and early referral to the ophthalmologist as appropriate is warranted.
