353. Clinical Significance of Methicillin-Resistant Staphylococcus aureus Colonization on Hospital Admission: One-Year Infection Risk.

Background: Methicillin-resistant Staphylococcus aureus (MRSA) nasal colonization among inpatients is a well-established risk factor for MRSA infection during the same hospitalization, but the long-term risk of MRSA infection is uncertain. We performed a retrospective cohort study to determine the one-year risk of MRSA infection among inpatients with MRSA-positive nasal polymerase chain reaction (PCR) tests confirmed by positive nasal culture (Group 1), patients with positive nasal PCR but negative nasal culture (Group 2), and patients with negative nasal PCR (Group 3). Methodology/Principal Findings: Subjects were adults admitted to a four-hospital system between November 1, 2006 and March 31, 2011, comprising 195,255 admissions. Patients underwent nasal swab for MRSA PCR upon admission; if positive, nasal culture for MRSA was performed; if recovered, MRSA was tested for Panton-Valentine Leukocidin (PVL). Outcomes included MRSA-positive clinical culture and skin and soft tissue infection (SSTI). Group 1 patients had a one-year risk of MRSA-positive clinical culture of 8.0% compared with 3.0% for Group 2 patients, and 0.6% for Group 3 patients (p<0.001). In a multivariable model, the hazard ratios for future MRSA-positive clinical culture were 6.52 (95% CI, 5.57 to 7.64) for Group 1 and 3.40 (95% CI, 2.70 to 4.27) for Group 2, compared with Group 3 (p<0.0001). History of MRSA and concurrent MRSA-positive clinical culture were significant risk factors for future MRSA-positive clinical culture. Group 1 patients colonized with PVL-positive MRSA had a one-year risk of MRSA-positive clinical culture of 10.1%, and a one-year risk of MRSA-positive clinical culture or SSTI diagnosis of 21.7%, compared with risks of 7.1% and 12.5%, respectively, for patients colonized with PVL-negative MRSA (p = 0.04, p = 0.005, respectively). Conclusions/Significance: MRSA nasal colonization is a significant risk factor for future MRSA infection; more so if detected by culture than PCR. In this study, a TaqMan real-time polymerase chain reaction (PCR) assay was developed to detect and quantify orf virus (ORFV) DNA in infected cell culture and clinical samples. Primers and probes were designed to amplify an 87 bp fragment DNA based on the sequence of ORFV024 gene encoding an NF-κB inhibitor of orf virus. The assay was highly specific and sensitive for ORFV DNA and no cross-reactions were detected with any other poxviruses; the sensitivity was 5 fg or 15 copies of ORFV genomic DNA. Both intra- (1.490 ± 1.261%) and inter-assay (1.958 ± 0.568%) variabilities were within the acceptable range, indicating the high efficiency and reproducibility of the assay. Further, the assay has shown a relative diagnostic sensitivity and specificity of 100%, when compared to B2L gene-based semi-nested PCR. The assay is simple, rapid, specific and sensitive with a wide potential for rapid field diagnosis of orf in sheep and goats. [ABSTRACT FROM AUTHOR]

ABSTRACT We developed a luminescent terbium sensor (LTS) based on energy resonance transfer for homogeneous bioassays. The effect of temperature on photoluminescence and time-resolved fluorescence of the LTS was investigated. When the temperature was increased from 277 K to 369 K, the photoluminescence quantum yield decreased by up to 25 %, time-resolved fluorescence decreased by up to 54 %, and the lifetime shortened dramatically. Studies showed that both photoluminescence and time-resolved fluorescence quantum yields were largely recovered after samples were heated from 298 to 310, 333 or 369 K and subsequently cooled to 298 K. These results indicate that the homogeneous bioassay with LTS is sensitive to temperature and should be conducted at a constant temperature to ensure the temperature effect does not influence data and to increase the accuracy of the results. lthough the effects of male mating history on female reproductive output and longevity have been studied in insects, few such studies have been carried out in spiders. In a mating system in which females are monandrous while males are polygynous, females may incur the risk by mating with successful males that have experienced consecutive matings and suffer from the possible depletion of sperm and/or associated ejaculates. Here, we examine the effects of male mating history on male courtship and copulation duration, female reproductive fitness, and female adult longevity of the wolf spider, Pardosa astrigera. Results indicated that male mating frequency had little effect on their subsequent copulation success, and of 35 males tested, about half of the males were able to copulate with five virgin females successively at an interval of 24 h. Male mating history had little effect on their courtship duration. However, male mating history significantly affected male copulation duration, female adult longevity, and reproductive output. Males that mated more frequently copulated longer and more likely failed to cause their mates to produce a clutch, although there was no significant difference in the number of eggs laid and the number of eggs hatched regardless of the first clutch or the second one. Multiple mating of male P. astrigera resulted in significant reduction in female adult longevity. Our results indicate that monandrous females mating with multiple-mated males may incur substantial fitness costs. [ABSTRACT FROM AUTHOR]

Xanthine oxidase (XO) is a purine catabolic enzyme related to hyperuricemia and gout. Porphyra polysaccharide (PP) is a kind of sulfated polysaccharide with potent biological activity. Herein, the interaction mechanism between PP and XO was studied by enzyme kinetics and multi-spectroscopy methods for the first time. Inhibition kinetics assay showed that PP reversibly inhibited XO activity in a mixed competitive manner with an IC 50 of 10.53 ± 0.69 mg/ml. Fluorescence titration studies and thermodynamic parameter calculations revealed that PP could spontaneously bind to XO through hydrophobic interactions, with a class of binding site. Circular dichroism analysis demonstrated that PP induced secondary structure rearrangement and conformational change of XO. Molecular docking further revealed that PP inserted into the hydrophobic cavity of XO, occupying the catalytic center, leading to the inhibition of XO activity. - Abstract
Purpose To evaluate if the difference between pre- and post-Gd-DTPA
2- relaxation rate (ΔR1 ) provides better differentiation of osteoarthritic patients (OA) from healthy subjects (HS) with dGEMRIC, as compared to post-Gd-DTPA2- spin-lattice relaxation time (T1Gd ). Materials and Methods Seventeen OA and 14 HS underwent pre- and 90 minutes postcontrast (Gd-DTPA2- ) magnetic resonance imaging (MRI) of the knee, using inversion recovery fast spin-echo and/or Lock-Locker sequences for T1 mapping. Effect sizes for T1pre , T1Gd , and ΔR1 were calculated, and receiver operating characteristic (ROC) curve and regression analysis were also performed to assess the effectiveness of each parameter in the separation of OA and HS. Results T1Gd and ΔR1 were almost identical in terms of areas under ROC curves (0.903 and 0.914, respectively), and effect sizes (1.34 and 1.31, respectively). These were significantly higher than T1pre . In addition, a high inverse correlation was observed between ΔR1 vs. T1Gd (R = 0.96). Conclusion Either T1Gd or ΔR1 could be used as an index in the evaluation of native cartilage. However, considering the practical logistical cost involved in terms of time and effort to acquire precontrast T1 measurements, our data further support the continued use of T1Gd as the dGEMRIC index in the evaluation of native cartilage. Abstract: The action mechanism of vanadocene dichloride, Cp2VCl2 (Cp= η 5–C5H5), has been investigated by interaction with human serum transferrin for its promising antitumor activities. Our results have shown that Cp2VCl2 binds to transferrin and form a new complex, and the calculated apparent association constant is 1.37×105 M−1 from the fluorescence quenching. Simultaneously, the variation of the secondary structure of transferrin occurs, most probably due to the coordination of the amino residues of protein with VIV. It was evidenced that Cp is released free in solution after VIV binding to transferrin by 1H NMR measurements. These results have shown that Cp2VCl2 forms a complex with transferrin, which may provide a possible pathway in the transport and targeted delivery of the antitumor agent. [Copyright &y& Elsevier]

The compositional effects on the crystal structure, phase transition, dielectric, ferroelectric and piezoelectric properties of the (1- x)BaTiO
3 - x(Bi0.5 Na0.5 )TiO3 solid solution ceramics were investigated. After sintering at 1200 °C for 2 h, the ceramics with different content of (Bi0.5 Na0.5 )TiO3 (BNT) formed single-phase solid solutions with perovskite structure. The lattice constant c/ a ratio of the solid solutions decreased as BNT content increased except that with 20 mol% BNT, which had the largest value of c/ a ratio for all samples. The second phase transition corresponding to BaTiO3 (BT) at about 5 °C shifted to low temperature and gradually disappeared as BNT increased. A new secondary phase transition appeared at 32 °C and 64 °C, respectively, when BNT content was 5 and 14 mol%. The Curie temperature, Tc , shifted in the temperature range between 112 °C and 166 °C, and the remanent polarization, Pr , decreased whereas the coercive field, Ec , increased as the BNT content increased. The relatively high value of piezoelectric coefficient d33 together with a relatively high Curie temperature and low loss tangent and stable dielectric properties were obtained when the addition of BNT was 20 mol%. Background: Limited evidence exists regarding the relationship between literacy and health-related quality of life (HRQL). Research is needed to develop measurement techniques for low literacy populations and to evaluate potential literacy-related measurement bias.Methods: A Talking Touchscreen (TT) was developed for an HRQL study. Low (n = 214) and high literacy (n = 201) adult cancer outpatients participated, 70% of whom were from racial/ethnic minorities. Patients completed three questionnaires by TT: FACT-G (cancer-specific), SF-36 (generic health status), and a standard gamble utility questionnaire. Measurement bias was evaluated using item response theory (IRT). Effects of literacy on HRQL were evaluated using regression models.Results: Most (97%) patients rated the TT easy to use. In IRT analysis, 6/27 FACT-G and 12/31 SF-36 items demonstrated literacy bias; this was relatively balanced (10 items 'biased against' low literacy; 8 'biased against' high literacy). Mean literacy group differences were statistically and clinically non-significant for 9/14 HRQL outcomes. Adjustment for bias and/or covariates eliminated most remaining differences.Conclusions: The TT is valid and useful for HRQL assessment in low literacy populations. There appears to be no systematic literacy bias in reporting HRQL, and low literacy is not an independent risk factor for poorer HRQL. [ABSTRACT FROM AUTHOR]- Published

Objective.To identify predictors of community-onset extended-spectrum ß-lactamase (ESBL)-producing Escherichia coliinfection.Design.Prospective case-control study.Setting.Acute care hospitals and ambulatory clinics in the Chicago, Illinois, region.Patients.Adults with E. coliclinical isolates cultured in ambulatory settings or within 48 hours of hospital admission.Methods.Cases were patients with ESBL-producing E. coliclinical isolates cultured in ambulatory settings or within 48 hours of admission, and controls were patients with non-ESBL-producing E. coliisolates, matched to cases by specimen, location, and date. Clinical variables were ascertained through interviews and medical record review. Molecular methods were used to identify ESBL types, sequence type ST131, and aac(6')-Ib-cr.Results.We enrolled 94 cases and 158 controls. Multivariate risk factors for ESBL-producing E. coliinfection included travel to India in the past year (odds ratio [OR], 14.40 [95% confidence interval (CI), 2.92-70.95]), ciprofloxacin use (OR, 3.92 [95% CI, 1.90-8.1]), and age (OR, 1.04 [95% CI, 1.02-1.06]). Case isolates exhibited high prevalence of CTX-M-15 (78%), ST131 (50%), and aac(6')-Ib-cr(66% of isolates with CTX-M-15).Conclusions.Providers should be aware of the increased risk of ESBL-producing E. coliinfection among returned travelers, especially those from India.

364. Electronic Health Record–Based Detection of Risk Factors for Clostridium difficileInfection Relapse

Objective.A major challenge in treating Clostridium difficileinfection (CDI) is relapse. Many new therapies are being developed to help prevent this outcome. We sought to establish risk factors for relapse and determine whether fields available in an electronic health record (EHR) could be used to identify high-risk patients for targeted relapse prevention strategies.Design.Retrospective cohort study.Setting.Large clinical data warehouse at a 4-hospital healthcare organization.Participants.Data were gathered from January 2006 through October 2010. Subjects were all inpatient episodes of a positive C. difficiletest where patients were available for 56 days of follow-up.Methods.Relapse was defined as another positive test between 15 and 56 days after the initial test. Multivariable regression was performed to identify factors independently associated with CDI relapse.Results.Eight hundred twenty-nine episodes met eligibility criteria, and 198 resulted in relapse (23.9%). In the final multivariable analysis, risk of relapse was associated with age (odds ratio [OR], 1.02 per year [95% confidence interval (CI), 1.01–1.03]), fluoroquinolone exposure in the 90 days before diagnosis (OR, 1.58 [95% CI, 1.11–2.26]), intensive care unit stay in the 30 days before diagnosis (OR, 0.47 [95% CI, 0.30–0.75]), cephalosporin (OR, 1.80 [95% CI, 1.19–2.71]), proton pump inhibitor (PPI; OR, 1.55 [95% CI, 1.05–2.29]), and metronidazole exposure after diagnosis (OR, 2.74 [95% CI, 1.64–4.60]). A prediction model tuned to ensure a 50% probability of relapse would flag 14.6% of CDI episodes.Conclusions.Data from a comprehensive EHR can be used to identify patients at high risk for CDI relapse. Major risk factors include antibiotic and PPI exposure.

Asthma is a chronic airway inflammation that caused by many factors. The voltage-gated proton channel Hv1 has been proposed to extrude excessive protons produced by NADPH oxidase (NOX) from cytosol to maintain its activity during respiratory bursts. Here, we showed that loss of Hv1 aggravates ovalbumin (OVA)-induced allergic lung asthma in mice. The numbers of total cells, eosinophils and neutrophils in bronchoalveolar lavage fluid (BALF) of Hv1-deficiency (KO) mice are obviously increased after OVA challenge compared with that of wild-type (WT) mice. Histopathological staining reveals that Hv1-deficiency aggravates OVA-induced inflammatory cell infiltration and goblet cell hyperplasia in lung tissues. The expression of IL-4, IL-5 and IL-13 are markedly increased in lung tissues of OVA-challenged KO mice compared with that of WT mice. Furthermore, the expression levels of NOX2, NOX4 and DUOX1 are dramatically increased, while the expression levels of SOD2 and catalase are significantly reduced in lung tissues of OVA-challenged KO mice compared with that of WT mice. The production of ROS in lung tissues of KO mice is significantly higher than that of WT mice after OVA challenge. As an important member of ion channels family, the voltage-gated sodium channel (VGSC/Nav) is associated with a variety of diseases, including epilepsy, migraine, ataxia, etc., and has always been a hot target for drug design and discovery. Many subtype-selective modulators targeting VGSCs have been reported, and some of them have been approved for clinical applications. However, the drug design resources related to VGSCs are insufficient, especially the lack of accurate and extensive compound data toward VGSCs. To fulfill this demand, we develop the Voltage-gated Sodium Channels Database (VGSC-DB). VGSC-DB is the first open-source database for VGSCs, which provides open access to 6055 data records, including 3396 compounds from 173 references toward nine subtypes of Navs (Nav1.1 ~ Nav1.9). A total of 28 items of information is included in each data record, including the chemical structure, biological activity (IC50/EC50), target, binding site, organism, chemical and physical properties, etc. VGSC-DB collects the data from small-molecule compounds, toxins and various derivatives. Users can search the information of compounds by text or structure, and the advanced search function is also supported to realize batch query. ABSTRACTScreening assays for Neisseria gonorrhoeaeexhibit low positive predictive values, particularly in low-prevalence populations. A new real-time PCR assay that detects and identifies individual Neisseriaspp. using melt curve analysis was compared to two previously published supplementary assays. NsppID, a 16S rRNA real-time PCR/melt curve assay developed to distinguish N. gonorrhoeaefrom other Neisseriaspp., was compared to real-time PCR assays targeting genes reportedly specific for N. gonorrhoeae, the cppBgene and the porApseudogene. A total of 408 clinical specimens (324 female endocervical swabs and 84 male urine or urogenital swab specimens) were screened using the COBAS Amplicor assay for Chlamydia trachomatisand N. gonorrhoeae(CT/NG) (Roche Diagnostics, Indianapolis, IN) followed by confirmatory testing via real-time PCR. The NsppID assay detected Neisseriaspp. in 150/181 COBAS-positive specimens (82%), including six dual infections, and identified N. gonorrhoeaein 102 (56%) specimens. Sixty-nine of 181 (38%) specimens were positive for N. gonorrhoeaeby porApseudogene, and 115/181 (64%) were positive for cppB. However, cppBwas also positive in 15% of COBAS-negative specimens, more than either NsppID (4%) or porApseudogene (2%) assays. The porApseudogene assay had the highest specificity for both genders but the lowest sensitivity, especially in female specimens. NsppID had a slightly lower specificity but greater sensitivity and overall accuracy. The least desirable confirmatory assay was cppB, due to poor specificity. The NsppID assay is an accurate confirmatory assay for N. gonorrhoeaedetection. In addition, the NsppID assay can identify the non-N. gonorrhoeaespecies responsible for the majority of false-positive results from the COBAS Amplicor CT/NG assay.

Objective Develop and validate a health-related quality-of-life (measure for patients with acute and chronic leukemia. Methods The study consisted of two phases: scale construction and scale validation. For the item-generation phase, a summary of the literature combined with qualitative results from item-generation interviews with 29 acute or chronic leukemia patients and 16 health care providers yielded an initial item pool reflecting leukemia-specific concerns and symptoms. Items underwent iterations of review and reduction according to defined retention criteria to support content validity, as defined by priority concerns of patients. Seventeen final leukemia-specific items were combined with the Functional Assessment of Cancer Therapy–General to create the FACT-Leukemia (FACT-Leu) scale. For the validation phase, 79 individuals with acute or chronic leukemia completed questionnaires at three time points. Results All FACT-Leu subscale and aggregated scores showed high internal consistency ( α s ranging from 0.75 to 0.96). Test-retest reliability was adequate for all subscales (intraclass correlation range 0.765–0.890). The FACT-Leu scale demonstrated good convergent validity, with significant correlations with quality-of-life criteria and performance status, in the expected direction. FACT-Leu subscale scores were significantly different among the three performance status change groups, suggesting good responsiveness to change. Conclusions The FACT-Leu scale is a valid, reliable, and efficient measure of leukemia-specific health-related quality of life for acute and chronic disease.

This paper explores the differences between public perceptions and existing monitoring indicators in environmental quality in Beijing. The results reveal, existing indicators do not accurately reflect public perceptions of the environment. Some environmental problems, such as fluffy catkins, are not reflected in the existing indicators, yet have a relatively large influence on public perception. The policymakers and public's variegated understanding of the environment, the lag period of public perceptions of environmental quality change, and lack of standards and monitoring for emerging problems are the three main reasons that resulted in a deviation of monitoring indicators from the public perceptions. Resultantly, to improve the level of environmental governance, it is necessary to take relevant measures that reduce the difference between public perceptions and monitoring indicators. Residents should be surveyed regularly to obtain their public perceptions, and the selection of indicators should not be limited to environmental pollution. In particular, some indicators based on public perception should be developed to complement the existing environmental monitoring protocol. Shale hydration dispersion and swelling are primary causes of wellbore instability in oil and gas reservoir exploration. In this study, inulin, a fructo-oligosaccharide extracted from Jerusalem artichoke roots, was modified by acylation with three acyl chlorides, and the products (C10-, C12-, and C14-inulin) were investigated for their use as novel shale hydration inhibitors. The inhibition properties were evaluated through the shale cuttings hot-rolling dispersion test, the sodium-based bentonite hydration test, and capillary suction. The three acylated inulins exhibited superb hydration-inhibiting performance at low concentrations, compared to the commonly used inhibitors of KCl and poly (ester amine). An inhibition mechanism was proposed based on surface tension measurements, contact angle measurements, Fourier-transform infrared analysis, and scanning electron microscopy. The acylated inulin reduced the water surface tension significantly, thus, retarding the invasion of water into the shale formation. Then, the acylated inulin was adsorbed onto the shale surface by hydrogen bonding to form a compact, sealed, hydrophobic membrane. Furthermore, the acylated inulins are non-toxic and biodegradable, which meet the increasingly stringent environmental regulations in this field. Digitalisation exerts a large influence on enterprise value creation. We identify four dimensions for measuring digitalisation at the enterprise level, i.e. value-chain digitalisation, business-process digitalisation, product-service digitalisation, and application of digital technology. Through the survey of 262 Chinese enterprises, we empirically find that there is an inverted U-shaped relationship between enterprise digitalisation and financial performance. And this relation is positively moderated by dynamic capability. Fusarium oxysporum causes vascular wilt in more than 100 plant species, resulting in massive economic losses. A deep understanding of the mechanisms of pathogenicity and symptom induction by this fungus is necessary to control crop wilt. The YjeF protein has been proven to function in cellular metabolism damage-repair in Escherichia coli and to play an important role in Edc3 (enhancer of the mRNA decapping 3) function in Candida albicans, but no studies have been reported on related functions in plant pathogenic fungi. In this work, we report how the FomYjeF gene in F. oxysporum f. sp. momordicae contributes to conidia production and virulence. The deletion of the FomYjeF gene displayed a highly improved capacity for macroconidia production, and it was shown to be involved in carbendazim's associated stress pathway. Meanwhile, this gene caused a significant increase in virulence in bitter gourd plants with a higher disease severity index and enhanced the accumulation of glutathione peroxidase and the ability to degrade hydrogen peroxide in F. oxysporum. These findings reveal that FomYjeF affects virulence by influencing the amount of spore formation and the ROS (reactive oxygen species) pathway of F. oxysporum f. sp. momordicae. Taken together, our study shows that the FomYjeF gene affects sporulation, mycelial growth, pathogenicity, and ROS accumulation in F. oxysporum. Developing aqueous electrolyte with wide electrochemical stability window (ESW) is one of the key technologies to boosting the energy density of aqueous electrochemical double-layer capacitors (EDLCs). However, expanding the ESW using super-concentrated electrolytes comes at the expense of cost, ionic conductivity and the mass density of the device. Herein, we selected a cost-effective and environmentally friendly molecular crowding agent, polyethylene glycol dimethyl ether (PEGDME-250), as the electrolyte additive designed to achieve high ESW at low salt concentrations. First, PEGDME can reduces the activity of water and increases the ESW of the electrolyte by forming hydrogen bonds with free water molecule. In addition, PEGDME does not have the interaction between terminal groups compared to conventional polyethylene glycols (PEG), which helps to reduce the viscosity and improve the ionic conductivity of the electrolyte. By adjusting the composition of the electrolyte, the optimal electrolyte with a high ESW of 2.96 V was achieved with the formula of 3 m NaClO 4 -25 % H 2 O-75 % PEGDME. The EDLCs assembled with this diluted aqueous electrolyte and YP-50 F electrodes presents a high operational voltage window of 2.4 V and outstanding cycle stability (>10000 cycles), and maintains excellent rate performance in a temperature range of −10–35 ℃. • The addition of PEGDME reduces the activity of free water molecules. • Low concentration aqueous electrolyte realized electrochemical stabilization window of 2.96 V. • The EDLCs with optimized electrolyte and YP-50 F activated carbon were assembled. • The EDLCs were able to cycle 10,000 cycles at an operating voltage of 2.4 V. • The EDLCs were capable to maintain 72.3 % capacity when the current density increasing from 1 A g−1 to 10 A g−1. Studies in multiple species have shown that phospholipid:diacylglycerol acyltransferase (PDAT) and oil bodies are important factors affecting plant oil accumulation. However, little information is available about the PDAT genes and oil bodies in Eucommia ulmoides Oliv. a high-quality oil tree. In this study, the oil bodies in Eucommia ulmoides seeds at different developmental stages were observed by frozen section technology combined with Nile red staining. It was found that there was a significant positive correlation between oil content and oil body number. In addition, seven Eucommia ulmoides PDAT (EuPDAT) genes were cloned from Eucommia ulmoides seeds based on genome data. They were assembled into three subgroups according to their classifications in Arabidopsis. EuPDAT genes in the same subgroup had similar gene structures and conserved motifs. Putative cis-element analysis of EuPDAT gene promoter regions uncovered numerous elements related to stress and plant hormones response. The expression patterns showed that EuPDAT2 and EuPDAT7 expression levels were positively related to oil content in different periods, and the correlation coefficients were 0.78 and 0.91, respectively. Our results suggest that oil body number and EuPDAT genes are important factors affecting oil accumulation in E. ulmoides seeds, and this work provides a theoretical reference for the directional cultivation of new high-oil-content E. ulmoides varieties. Endometrial hyperplasia (EH), a uterine pathology characterized by an increased gland-to-stroma ratio compared to normal endometrium (NE), may precede the development of endometrial cancer (EC). Particularly, atypical EH also known as endometrial intraepithelial neoplasia (EIN), has been proven to be a precursor of EC. Thus, diagnosing different EH (EIN, hyperplasia without atypia (HwA) and NE) and screening EIN from non-EIN are crucial for the health of female reproductive system. Computer-aided-diagnosis (CAD) was used to diagnose endometrial histological images based on machine learning and deep learning. However, these studies perform single-scale image analysis and thus can only characterize partial endometrial features. Empirically, both global (cytological changes relative to background) and local features (gland-to-stromal ratio and lesion dimension) are helpful in identifying endometrial lesions. We proposed a global-to-local multi-scale convolutional neural network (G2LNet) to diagnose different EH and to screen EIN in endometrial histological images stained by hematoxylin and eosin (H&E). The G2LNet first used a supervised model in the global part to extract contextual features of endometrial lesions, and simultaneously deployed multi-instance learning in the local part to obtain textural features from multiple image patches. The contextual and textural features were used together to diagnose different endometrial lesions after fusion by a convolutional block attention module. In addition, we visualized the salient regions on both the global image and local images to investigate the interpretability of the model in endometrial diagnosis. In the five-fold cross validation on 7812 H&E images from 467 endometrial specimens, G2LNet achieved an accuracy of 97.01% for EH diagnosis and an area-under-the-curve (AUC) of 0.9902 for EIN screening, significantly higher than state-of-the-arts. In external validation on 1631 H&E images from 135 specimens, G2LNet achieved an accuracy of 95.34% for EH diagnosis, which was comparable to that of a mid-level pathologist (95.71%). Specifically, G2LNet had advantages in diagnosing EIN, while humans performed better in identifying NE and HwA. The developed G2LNet that integrated both the global (contextual) and local (textural) features may help pathologists diagnose endometrial lesions in clinical practices, especially to improve the accuracy and efficiency of screening for precancerous lesions. Molecular property prediction models based on machine learning algorithms have become important tools to triage unpromising lead molecules in the early stages of drug discovery. Compared with the mainstream descriptor- and graph-based methods for molecular property predictions, SMILES-based methods can directly extract molecular features from SMILES without human expert knowledge, but they require more powerful algorithms for feature extraction and a larger amount of data for training, which makes SMILES-based methods less popular. Here, we show the great potential of pre-training in promoting the predictions of important pharmaceutical properties. By utilizing three pre-training tasks based on atom feature prediction, molecular feature prediction and contrastive learning, a new pre-training method K-BERT, which can extract chemical information from SMILES like chemists, was developed. The calculation results on 15 pharmaceutical datasets show that K-BERT outperforms well-established descriptor-based (XGBoost) and graph-based (Attentive FP and HRGCN+) models. In addition, we found that the contrastive learning pre-training task enables K-BERT to 'understand' SMILES not limited to canonical SMILES. Moreover, the general fingerprints K-BERT-FP generated by K-BERT exhibit comparative predictive power to MACCS on 15 pharmaceutical datasets and can also capture molecular size and chirality information that traditional binary fingerprints cannot capture. Abstract: Eucommia ulmoides Oliver is one of the few woody plants producing trans-polyisoprene (TPI or Eucommia rubber). Eucommia fruits contain average 12–17% of TPI, much higher than in leaf or bark. However, commercialization of TPI production from Eucommia fruits is confronted with two major problems: the low yield and high cost of fruit harvest. To develop an economically viable solution to those two problems, we conducted a 9-year study from 1998 to 2006 to compare the fruit or leaf production among different training systems (orchard-like model vs conventional model), cultivars (conventional vs new cultivars) and tree densities. Our results showed that the trees grafted with the new cultivar commenced fruiting about 3years earlier and produced significantly higher fruit yield over the study period than the non-grafted Eucommia trees. This study also demonstrated for the first time that the orchard system with a tree planting density of 3m×3m and a female proportion of up to 90% yielded 40 times as much fruit as the conventional model without using the new cultivars or 5 times as much as the conventional model using the new cultivar grafted trees. Eucommia orchard in spacing 3m×3m yielded more than 4000kgha−1 fruit in 2006, equivalent to 520kgha−1 of TPI, similar to low yield of cis-1,4-polyisoprene (CPI) from Hevea brasiliensis plantations. The relatively low tree height in the orchard system also significantly reduced the cost of fruit harvest, as compared to the conventional model. We estimated that the input–output ratio of managing one hectare Eucommia orchard was 646% and 642% with planting densities of 1665treesha−1 and 1112treesha−1, respectively. Introduction and hypothesis: The purpose of this study was to investigate change in bladder capacity as a measure of response to combined intravesical dimethyl sulfoxide (DMSO) and triamcinolone instillations for the treatment of newly diagnosed bladder pain syndrome/interstitial cystitis (BPS/IC). Methods: 141 newly diagnosed women were identified retrospectively. 79 were treated with weekly DMSO/triamcinolone instillations. Change in bladder capacity with bladder retrofill, daytime urinary frequency, nocturia episodes per night, and Likert scale symptom scores were reviewed. Wilcoxon signed-rank tests, Wilcoxon rank-sum tests, Spearman's rank correlations, COX regression analysis, and a Kaplan-Meier survival curve were performed. Results: Significant changes (median (25-percentile to 75-percentile) were noted for bladder capacity (75 mL (25 to 130 mL), p < 0.0001), inter-void interval (0 hrs (0 to 1 hour), p < 0.0001), nocturia episodes per night (−1 (−2 to 0), p < 0.0001), and aggregate Likert symptom scores (−2 points (−5 to 0), p < 0.0001). Percent change in bladder capacity correlated positively with percent change in inter-void interval (p = 0.03) and negatively with percent changes in nocturia (p = 0.17) and symptom scores (p = 0.01). Women without detrusor overactivity (DO) had greater percent changes in capacity than women with DO (62.5 % vs. 16.5 %, p = 0.02). 61.3 % of patients were retreated with a 36 weeks median time to retreatment and no difference in time to retreatment based upon DO. Greater capacity was protective against retreatment (hazard ratio = 0.997 [95 % CI 0.994,0.999], p = 0.02). Conclusions: Percent change in bladder capacity is a useful objective measure of response to intravesical DMSO/triamcinolone for newly diagnosed BPS/IC. Reducing health care-associated urinary tract infection (UTI) is a National Patient Safety Goal. The purpose of this investigation was to establish a colony count threshold to predict clinically significant UTIs that develop in hospitalized patients. A total of 185 cases were reviewed sequentially by 2 physicians. The information extracted included subjective complaints, presence of an indwelling urinary catheter, clinical signs and symptoms, WBC count, urinalysis, and urine culture results. The first reviewer recorded whether the patient was diagnosed and treated for a UTI by the clinician. The second reviewer determined if the patient met National Healthcare Safety Network guidelines for nosocomial UTI. Compared with patients with colony counts less than 100,000 colony-forming units per milliliter (CFU/mL), patients with colony counts 100,000 CFU/mL or more were 73.86 times more likely to have a clinically significant UTI (odds ratio, 73.86; 95% confidence interval, 24.23 ~ 225.15; P < .0001; c-statistic, 0.859). Reporting positive results only for patients with 100,000 CFU/mL or more would have reduced the number of positive cultures by 38%. These data suggest that reporting colony counts less than 100,000 CFU/mL encourages treatment of non-clinically significant UTIs in hospitalized patients, causing inappropriate antibiotic use. Meningiomas are dural-based brain tumors that are typically histologically benign. Some meningiomas grow slowly or seemingly not at all with planimetric measurement. Volumetric measurement may be more accurate because tumors may grow in different directions than the planimetric axes. Twenty-one patients (with 22 tumors) had serial MRI brain scans available for review. We reviewed the charts and measured tumor dimensions on the MRI scans. Relative growth rates were calculated for volume and maximum initial diameter using published formulas. Patient demographics, tumor location, and special radiologic characteristics (calcification, T2 hypointensity, dural tail, mass effect, and midline shift) were compared to the volumetric growth rate. Patients included 17 females and 4 males; age at diagnosis 36 to 74 years (mean 61). Follow-up was 2.08 to 10.83 years (mean 3.64). Most tumors were located in the convexity (27.27 %), sphenoid wing (27.27 %), or cerebellopontine angle (13.04 %). Two meningiomas (9.09 %) demonstrated no growth. The mean relative volumetric growth rate was 5.82 %/year, and planimetric was 2.00 %/year (difference 3.82 %/year, p-value < 0.0001). Convexity location had near significant association with slower relative volumetric growth. There were no significant associations between other tumor locations, age, gender, or radiologic characteristics and volumetric growth. The mean volumetric growth rate was significantly greater than the planimetric growth rate, suggesting that volumetric measurement conveys more information and is superior in assessing tumor growth. Abstract: ZnO/amorphous-BaTiO3 thin-films were prepared on glass substrates by a sol–gel process. DSC/TGA, XRD and AFM were used to analyze the thermolysis of the precursor and the crystal growth characteristics of ZnO, and IR spectroscopy was used to determine the presence of OH groups and Cies containing in the films. Violet emission centering at 418 and 438 nm was observed in room temperature PL spectra of the films annealed at 400–500 °C. The strongest violet emission was observed in the film annealed at 400 °C. The emission intensity reduces as the annealing temperature increases, and vanishes in the film annealed at 550 °C. All of the films contain OH and C and the films annealed at the temperature over 450 °C contain CO2 impurities, indicating the enhancing oxidation of the films. The vanishing of the violet emission is probably due to the improvement of the oxygen deficient of ZnO crystals. [Copyright &y& Elsevier]

Summary The p21-activated kinase, Shk1, is required for the proper establishment of cell polarity in the fission yeast, Schizosaccharomyces pombe . We showed recently that loss of the essential Shk1 inhibitor, Skb15, causes significant spindle defects in fission yeast, thus implicating Shk1 as a potential regulator of microtubule dynamics. Here, we show that cells deficient in Shk1 function have malformed interphase microtubules and mitotic microtubule spindles, are hypersensitive to the microtubule-destabilizing drug thiabendazole (TBZ) and cold sensitive for growth. TBZ treatment causes a downregulation of Shk1 kinase activity, which increases rapidly after release of cells from the drug, thus providing a correlation between Shk1 kinase function and active microtubule polymerization. Consistent with a role for Shk1 as a regulator of microtubule dynamics, green fluorescent protein (GFP)–Shk1 fusion proteins localize to interphase microtubules and mitotic microtubule spindles, as well as to cell ends and septum-forming regions of fission yeast cells. We show that loss of Tea1, a cell end- and microtubule-localized protein previously implicated as a regulator of microtubule dynamics in fission yeast, exacerbates the growth and microtubule defects resulting from partial loss of Shk1 and that Shk1 localizes to illicit growth tips produced by tea1 mutant cells. Our results demonstrate that Shk1 is required for the proper regulation of microtubule dynamics in fission yeast and implicate Tea1 as a potential Shk1 regulator. [ABSTRACT FROM AUTHOR]

Objective. To assess association between congenital cytomegalovirus (CMV) infection and brain injury in neonates. Methods. The literatures from inception to November 4, 2020, were searched through PubMed, Embase, Cochrane Library, and Web of Science. Heterogeneity test was conducted for each indicator and measured by I 2 statistics. If I 2 ≥ 50 % , the random effects model was applied; otherwise, the fixed effects model was used. Sensitivity analysis was performed for all models. Weighed mean difference (WMD) was used as the effect size for measurement data, and risk ratio (RR) was as the effect indicator. Results. A total of 13 studies, including 4,262 congenital CMV infection neonates, were enrolled in this study. Our results showed that the rate of hearing impairment (RR: 2.105, 95% CI: (1.115, 3.971), P = 0.002), sensorineural hearing loss (SNHL) (RR: 17.051, 95% CI: (6.201, 46.886), P < 0.001), and microcephaly (RR: 2.283, 95% CI: (1.325, 3.935), P =0.003) in neonates infected congenital CMV was higher than that in control group. Conclusion. The risks of hearing impairment, SNHL, and microcephaly in neonates during childhood may be associated with congenital CMV infection. It is necessary to establish neonatal screening programs and comprehensive diagnostic tests for patients to reduce the risk of adverse brain damage to the congenital CMV infection as early as possible and to improve the prognosis of the newborn. Zinc-ion batteries, known for safety and efficiency, are a hotspot in electrochemical energy storage field. Nonetheless, the emergence of dendrites on zinc anodes during electrochemical cycles can lead to capacity attenuation and even failure, particularly under low temperatures. In this study, we develop a novel dendrite-free and low-temperature resistant Zn(ClO 4) 2 /Li 2 SO 4 /Dimethyl sulfoxide (DMSO) hybrid electrolyte by using DMSO as a hydrogen bond acceptor and solvation regulator. Our research demonstrates that, by reducing the desolvation energy barrier of the Zn2+ solvation sheath and the nucleation overpotential of Zn deposition, DMSO effectively inhibits zinc dendrites and side reactions. Furthermore, DMSO disrupts hydrogen bonding between water molecules, preventing electrolyte freezing at low temperatures. As a result, at 0.5 mA cm−2, the Zn||Zn symmetric device using this hybrid electrolyte exhibits consistent operation for over 4000 h even at -25 °C, featuring a minimal deposition/dissolution overpotential of approximately 0.045 mV. Additionally, at -20 °C, the Zn||Zn(ClO 4) 2 -Li 2 SO 4 -DMSO||LiFePO 4 battery demonstrates stable cycling for over 600 cycles at 0.6 C (1 C = 170 mA g − 1), delivering a capacity of 103 mA h g−1. Aplosporella javeedii is a pathogenic fungus that causes canker and dieback of jujube in China. In this study, we report a new mycovirus, Aplosporella javeedii partitivirus 1 (AjPV1), isolated from A. javeedii strain NX55-3. The AjPV1 genome contains two double-stranded RNA elements (dsRNA1 and dsRNA2). The size of dsRNA1 is 2,360 bp, and it encodes a putative RNA-dependent RNA polymerase (RdRp), while dsRNA2 is 2,301 bp in length and encodes a putative capsid protein (CP). The sequences of RdRp and CP have significant similarity to those of members of the family Partitiviridae. Sequence alignment and phylogenetic analysis showed that AjPV1 is a new member of the family Partitiviridae that is related to members of the genus Betapartitivirus. The 2′,3′-cycling ribonuclease (RNase) genes are catalysts of RNA cleavage and include the RNase T2 gene family. RNase T2 genes perform important roles in plants and have been conserved in the genome of eukaryotic organisms. In this study we identified 21 EURNS genes in Eucommia ulmoides Oliver (E. ulmoides) and analyzed their structure, chromosomal location, phylogenetic tree, gene duplication, stress-related cis-elements, and expression patterns in different tissues. The length of 21 predicted EURNS proteins ranged from 143 to 374 amino acids (aa), their molecular weight (MW) ranged from 16.21 to 42.38 kDa, and their isoelectric point (PI) value ranged from 5.08 to 9.09. Two classifications (class I and class III) were obtained from the conserved domains analysis and phylogenetic tree. EURNS proteins contained a total of 15 motifs. Motif 1, motif 2, motif 3, and motif 7 were distributed in multiple sequences and were similar to the conserved domain of RNase T2. EURNS genes with similar structure and the predicted EURNS proteins with conserved motif compositions are in the same group in the phylogenetic tree. The results of RT-PCR and transcription data showed that EURNS genes have tissue-specific expression and exhibited obvious trends in different developmental stages. Gene duplication analysis results indicated that segment duplication may be the dominant duplication mode in this gene family. This study provides a theoretical basis for research on the RNase T2 gene family and lays a foundation for the further study of EURNS genes. [ABSTRACT FROM AUTHOR]

It is greatly meaningful but remains very challenging to obtain high-performance microwave absorber with thin thickness, lightweight, wide bandwidth, and strong absorption. Here, sulfur-enriched nitrogen-doped carbon nanofibers (S-NCFs) with excellent microwave absorption performance have been constructed through electrospinning technology and heat treatment with sublimed sulfur. Experimental results and the theoretical calculation evidenced that nitrogenous carbon nanofibers doping with sulfur could not only generate a large number of electric dipoles but also favor the promotion of conductivity. Benefiting from the unique architecture and heteroatoms doping that contributed to the well-matched impedance and dielectric enhancement, the S-NCFs exhibited the minimum reflection loss (RL) of − 48.67 dB at 16.1 GHz with a matching thickness of 1.39 mm at a filler loading of as low as 15%. In addition, the samples can obtain the broadest effective absorption bandwidth (EAB, RL < − 10 dB) of 4.90 GHz with the thickness of 1.53 mm. High glucose (HG)-induced oxidative stress contributes to the dysfunction of pancreatic β cells in diabetes. The voltage-gated proton channel Hv1 has been proposed to support reactive oxygen species (ROS) production during respiratory bursts. However, the effect of Hv1 on glucotoxicity in pancreatic β cells is not clear yet. In this study, we examined the protective effects of Hv1-deficiency in HG cultured β cells. Following 48 h of treatment with 30 mM high glucose, Hv1 KO β cells showed higher cell viability, lower cell apoptosis and a more stable insulin gene expression level compared to WT β cells. In both control and HG cultured β cells, deficiency of Hv1 decreased the glucose- and PMA-induced ROS production. Finally, HG incubation led to NOX4 upregulation in WT β cells, which could be inhibited by HV1 deficiency. In conclusion, Hv1-deficiency prevents the HG treatment-induced NOX4 upregulation and protects β cells from glucotoxicity. • Hv1-deficiency protects β cells from high glucose(HG)treatment-induced damage. • Hv1-deficiency protects β cells by reducing HG-induced ROS production. • The high ROS level in HG-treatment β cells may owe to NOX4 upregulation. • HV1-deficiency inhibits the HG-treatment induced NOX4 upregulation in β cells. Eucommia ulmoides Oliver produces trans -1,4-polyisoprene (TPI or Eucommia rubber), with leaves and bark as the main sources. Raw materials from short rotation coppice (SRC) cultures for TPI industrialization are a valuable option. Currently, there is a shortage of field yield data beginning from early establishment of Eucommia SRC to support TPI manufacturers. We investigated an Eucommia SRC plantation established in 2015 with application of four plant growth regulators in Mengzhou Forest Farm, China, during its first two annual rotations. By using an orthogonal experimental design, productivity related data were derived after each growing season. We observed significant variations in biomass yield and TPI productivity among the 12 dosages of 4 regulator types and control, with plants treated with brassinolide at 5 mg L −1 , DCPTA at 500 mg L −1 and DTA-6 at 20 mg L −1 reaching the highest TPI productivity among treatments of 406 kg ha −1 , 391 kg ha −1 and 382 kg ha −1 in the second rotation, respectively, all of which were more than 1.4 times that of the control. Foliage spray of regulators led to more and larger leaves, more bark yield, higher tree height, and higher leaf TPI content, which reflected the enhancing effects of regulators on leaf and bark biomass as well as TPI productivities in both the first and second rotations. Meanwhile, the harvest of Eucommia leaf and bark from the coppice plantation was less labor intensive than that from tall mature trees. Our study suggested that the annual rotation period and increase of TPI from Eucommia SRC by application of regulators could reduce the payback period. Therefore, we recommend the application of growth regulators to coppice Eucommia plantations beginning from the first rotation as an economic means to increase TPI production. [ABSTRACT FROM AUTHOR]

Prunus mira Koehne, an important economic fruit crop with high breeding and medicinal values, and an ancestral species of many cultivated peach species, has recently been declared an endangered species. However, basic information about genetic diversity, population structure, and morphological variation is still limited for this species. In this study, we sampled 420 P. mira individuals from 21 wild populations in the Tibet plateau to conduct a comprehensive analysis of genetic and morphological characteristics. The results of molecular analyses based on simple sequence repeat (SSR) markers indicated moderate genetic diversity and inbreeding (A = 3.8, Ae = 2.5, He = 0.52, Ho = 0.44, I = 0.95, FIS = 0.17) within P. mira populations. STRUCTURE, GENELAND, and phylogenetic analyses assigned the 21 populations to three genetic clusters that were moderately correlated with geographic altitudes, and this may have resulted from significantly different climatic and environmental factors at different altitudinal ranges. Significant isolation-by-distance was detected across the entire distribution of P. mira populations, but geographic altitude might have more significant effects on genetic structure than geographic distance in partial small-scale areas. Furthermore, clear genetic structure, high genetic differentiation, and restricted gene flow were detected between pairwise populations from different geographic groups, indicating that geographic barriers and genetic drift have significant effects on P. mira populations. Analyses of molecular variance based on the SSR markers indicated high variation (83.7% and 81.7%), whereas morphological analyses revealed low variation (1.30%–36.17%) within the populations. Large and heavy fruits were better adapted than light fruits and nutlets to poor climate and environmental conditions at high altitudes. Based on the results of molecular and morphological analyses, we classified the area into three conservation units and proposed several conservation strategies for wild P. mira populations in the Tibet plateau. Increased expression of angiotensin II type 2 receptor (AT2R) induces apoptosis in numerous tumor cell lines, with either Angiotensin II-dependent or Angiotensin II-independent regulation, but its molecular mechanism remains poorly understood. Here, we used PCR Array analysis to determine the gene and microRNA expression profiles in human prostate cancer cell lines transduced with AT2R recombinant adenovirus. Our results demonstrated that AT2R over expression leads to up-regulation of 6 apoptosis-related genes (TRAIL-R2, BAG3, BNIPI, HRK, Gadd45a, TP53BP2), 2 cytokine genes (IL6 and IL8) and 1 microRNA, and down-regulation of 1 apoptosis-related gene TNFSF10 and 2 cytokine genes (BMP6, BMP7) in transduced DU145 cells. HRK was identified as an up-regulated gene in AT2R-transduced PC-3 cells by real-time RT-PCR. Next, we utilized siRNAs to silence the up-regulated genes to further determine their roles on AT2R overexpression mediated apoptosis. The results showed downregulation of Gadd45a reduced the apoptotic effect by ∼30% in DU145 cells, downregulation of HRK reduced AT2R-mediated apoptosis by more than 50% in PC-3 cells, while downregulation of TRAIL-R2 enhanced AT2R-mediated apoptosis more than 4 times in DU145 cells. We also found that the effects on AT2R-mediated apoptosis caused by downregulation of Gadd45a, TRAIL-R2 and HRK were independent in activation of p38 MAPK, p44/42 MAPK and p53. Taken together, our results demonstrated that TRAIL-R2, Gadd45a and HRK may be novel target genes for further study of the mechanism of AT2R-mediated apoptosis in prostate cancer cells.
