Background and Objectives: Atovaquone is a hydroxynaphthoquinone with selective action in the mitochondrial respiratory chain of malaria parasite. It is employed for both the treatment and prevention of malaria, in a combination with proguanil. The aim of this study was to elucidate the in vitro metabolites from atovaquone and to evaluate their cytotoxic activities. Methods: The biotransformation of atovaquone was performed using Mucor rouxii NRRL 1894, Cunninghamella echinulata var. elegans ATCC 8688a and C. elegans ATCC 10028b, which have been reported as microbial models of mammalian drug metabolism. Experiments were also carried out with two probiotic strains from the human intestinal tract: Bifidobacterium sp. and Lactobacillus acidophilus. The phase I metabolite was isolated, its chemical structure was elucidated and its toxicity was evaluated using the neoplastic cell line SKBR-3 derived from human breast cancer and normal human fibroblast cell line GM07492-A. Cell cytotoxicity assays were also carried out with atovaquone. Result: Only the fungi were able to convert atovaquone to metabolite trans-3-[4′-(4″-chlorophenyl)cyclohexyl)-1,2-dioxo-dihydro-1H-indene-3-carboxylic acid. The metabolite displayed 50 % inhibitory concentration (IC) values of 110.20 ± 2.2 and 108.80 ± 1.5 µmol/L against breast cancer cell line SKBR-3 and fibroblasts cell line GM07492-A, respectively. The IC values of atovaquone were 282.30 ± 1.8 and 340.50 ± 1.4 µmol/L against breast cancer and normal fibroblasts cell lines, respectively. Conclusions: The produced metabolite was more toxic than atovaquone and was not selective to normal or cancer cell lines. Chlorella vulgaris cultivation in two-stage process was applied to increase the lipid productivity without compromising the biomass productivity. At the first stage, microalgae was cultivated under nutrient sufficient conditions to obtain a maximized cell density; at the second stage, nitrate conditions are changed to trigger the accumulation of TAG. During first stage, the maximum biomass productivity (32 mg L −1 d −1 ) was observed after 13 days under nutrient sufficient conditions with 1.21 g L −1 NaNO 3 and 0.00449 g L −1 K 2 HPO 4 . Maximum lipid content (25.4% DW), lipid productivity (7.5 mg L −1 d −1 ) and TAG content (41.3% in total lipids) were favored by the nitrogen starvation conditions for more 4 days, at the second stage. Oil extracted at the second stage contained lower percentage of PUFAs being more suitable for the biodiesel production when compared with the oil extracted at the first stage. This two-stage phototrophic process is promising to provide a more efficient way for on a large-scale production of algal biomass and biodiesel production. [ABSTRACT FROM AUTHOR]

Currently most economical and technological bottlenecks in protein production are placed in the downstream processes. With the aim of increasing the efficiency and reducing the associated costs, various affinity ligands have been developed. Affitins are small, yet robust and easy to produce, proteins derived from the archaeal extremophilic “7 kDa DNA-binding” protein family. By means of combinatorial protein engineering and ribosome display selection techniques, Affitins have shown to bind a diversity of targets. In this work, two previously developed Affitins (anti-lysozyme and anti-IgG) were immobilized onto magnetic particles to assess their potential for protein purification by magnetic fishing. The optimal lysozyme and human IgG binding conditions yielded 58 mg lysozyme/g support and 165 mg IgG/g support, respectively. The recovery of proteins was possible in high yield (≥95%) and with high purity, namely ≥95% and 81%, when recovering lysozyme from Escherichia coli supernatant and IgG from human plasma, respectively. Static binding studies indicated affinity constants of 5.0 × 10 4 M −1 and 9.3 × 10 5 M −1 for the anti-lysozyme and anti-IgG magnetic supports. This work demonstrated that Affitins, which can be virtually evolved for any protein of interest, can be coupled onto magnetic particles creating novel affinity adsorbents for purification by magnetic fishing. Trypanosomiasis is an anthropozoonosis caused by a flagellated protozoan of the Trypanosoma genus. Dogs play a significant role in the transmission chain of these diseases, serving as hosts and reservoirs while developing symptoms similar to humans. The main representatives of this genus are T. cruzi and T. evansi, responsible for American trypanosomiasis, also known as Chagas disease, and "surra," recognized in equines as "mal das cadeiras." Chagas disease is transmitted through biological vectoring by hematophagous insects of the Triatoma genus, from the Triatominae subfamily, commonly known as kissing bugs. This work aimed to report a case of trypanosomiasis in a dog in São Luís, MA. A canine was attended to at a veterinary clinic in São Luís, showing a sudden onset of dyspnea, anorexia, adipsia, lethargy, and progressive weakness. The physical examination revealed pale mucous membranes, a low body condition score, and abdominal distension with tenderness on palpation. Hematological analysis identified normocytic and normochromic anemia with signs of regeneration, lymphocytosis, and left shift, accompanied by the presence of neutrophil rods. Blood smear examination revealed structures compatible with flagellated protozoa of the Trypanosoma genus. Despite a prompt diagnosis and initiation of therapeutic protocols, the animal succumbed on the same day as admission. [ABSTRACT FROM AUTHOR]

The over-production of reactive oxygen species (ROS) can cause oxidative damage to a large number of molecules, including DNA, and has been associated with the pathogenesis of several disorders, such as diabetes mellitus (DM), dyslipidemia and periodontitis (PD). We hypothesise that the presence of these diseases could proportionally increase the DNA damage. The aim of this study was to assess the micronucleus frequency (MNF), as a biomarker for DNA damage, in individuals with type 2 DM, dyslipidemia and PD. One hundred and fifty patients were divided into five groups based upon diabetic, dyslipidemic and periodontal status (Group 1 – poor controlled DM with dyslipidemia and PD; Group 2 – well-controlled DM with dyslipidemia and PD; Group 3 – without DM with dyslipidemia and PD; Group 4 – without DM, without dyslipidemia and with PD; and Group 5 – without DM, dyslipidemia and PD). Blood analyses were carried out for fasting plasma glucose, HbA1c and lipid profile. Periodontal examinations were performed, and venous blood was collected and processed for micronucleus (MN) assay. The frequency of micronuclei was evaluated by cell culture cytokinesis-block MN assay. The general characteristics of each group were described by the mean and standard deviation and the data were submitted to the Mann–Whitney, Kruskal–Wallis, Multiple Logistic Regression and Spearman tests. The Groups 1, 2 and 3 were similarly dyslipidemic presenting increased levels of total cholesterol, low density lipoprotein cholesterol and triglycerides. Periodontal tissue destruction and local inflammation were significantly more severe in diabetics, particularly in Group 1. Frequency of bi-nucleated cells with MN and MNF, as well as nucleoplasmic bridges, were significantly higher for poor controlled diabetics with dyslipidemia and PD in comparison with those systemically healthy, even after adjusting for age, and considering Bonferroni’s correction. Elevated frequency of micronuclei was found in patients affected by type 2 diabetes, dyslipidemia and PD. This result suggests that these three pathologies occurring simultaneously promote an additional role to produce DNA impairment. Metallic radionuclides possess a variety of half-lives, emission types, energies, and branching ratios, making it possible to apply them as diagnostic and/or therapeutic agents. Mono (thiosemicarbazones) and bis (thiosemicarbazones) show wide pharmacological profile and ability to chelate metals forming stable complexes. In the present work the behaviors of different radiometal complexes with mono (thiosemicarbazones) and bis (thiosemicarbazones) as diagnostic and therapeutic radiopharmaceutical drug candidates are reported. In particular, the applications of metal complexes with bis (thiosemicarbazones) as diagnostic radiopharmaceuticals have mostly been focused on bis (thiosemicarbazonato)copper(II) complexes, due to the success of the hypoxia-selective PET imaging agent 64Cu(ATSM), H 2 ATSM = diacetyl- bis (N 4-methylthiosemicarbazone). However, metal complexes with mono - and bis (thiosemicarbazones) have also been obtained with other radioisotopes, mainly 67/68Ga, 111/114mIn and 99mTc. We evaluated the antibacterial activity of three diterpenes isolated from natural sources against a panel of microorganisms responsible for bovine mastitis. ent-Copalic acid (CA) was the most active metabolite, with promising MIC values (from 1.56 to 6.25 μg mL-1) against Staphylococcus aureus (ATCC and clinical isolate), Staphylococcus epidermidis, Streptococcus agalactiae, and Streptococcus dysgalactiae. We conducted time-kill assays of CA against S. aureus, a commensal organism considered to be a ubiquitous etiological agent of bovine mastitis in dairy farms worldwide. In the first 12 h, CA only inhibited the growth of the inoculums (bacteriostatic effect), but its bactericidal effect was clearly noted thereafter (between 12 and 24 h). Background and Aims There is a great need to search for natural compounds with superior prebiotic, antioxidant and immunostimulatory properties for use in (food) applications. Raffinose family oligosaccharides (RFOs) show such properties. Moreover, they contribute to stress tolerance in plants, acting as putative membrane stabilizers, antioxidants and signalling agents. Methods A large-scale soluble carbohydrate screening was performed within the plant kingdom. An unknown compound accumulated to a high extent in early-spring red deadnettle (Lamium purpureum) but not in other RFO plants. The compound was purified and its structure was unravelled with NMR. Organs and organ parts of red deadnettle were carefully dissected and analysed for soluble sugars. Phloem sap content was analysed by a common EDTA-based method. Key Results Early-spring red deadnettle stems and roots accumulate high concentrations of the reducing trisaccharide manninotriose (Galα1,6Galα1,6Glc), a derivative of the non-reducing RFO stachyose (Galα1,6Galα1,6Glcα1,2βFru). Detailed soluble carbohydrate analyses on dissected stem and leaf sections, together with phloem sap analyses, strongly suggest that stachyose is the main transport compound, but extensive hydrolysis of stachyose to manninotriose seems to occur along the transport path. Based on the specificities of the observed carbohydrate dynamics, the putative physiological roles of manninotriose in red deadnettle are discussed. Conclusions It is demonstrated for the first time that manninotriose is a novel and important player in the RFO metabolism of red dead deadnettle. It is proposed that manninotriose represents a temporary storage carbohydrate in early-spring deadnettle, at the same time perhaps functioning as a membrane protector and/or as an antioxidant in the vicinity of membranes, as recently suggested for other RFOs and fructans. This study describes important components of the reproductive biology of the Amazonian amphibian fish Copella arnoldi. The species sex ratio biased to female 1.8 to 1 male and the length‐weight relationship also shown differences between sexes (b value = 1.854 for females and 2.235 for males), although both presented allometric growth (b < 3), "being lighter for its length". The results also show the importance of the histological analysis to describe the gametogenesis and, consequently, define the reproductive cycle of a fish species. It showed both, males and females, spawning capable during a whole year, whereas the gonadosomatic index pointed spawning capable females during the winter and summer, and spawning capable males only in the autumn. The species presents the rare type of semicystic spermatogenesis and the spermatids complete their differentiation into the tubular lumen, forming a Type I spermatozoa. Both the characteristics are described for the first time for the Lebiasinidae family. Such data can contribute to the discussions about the position of the Lebiasinidae family within the Characiform order, which still does not have a conclusion. The proposal of this work was to investigate the effect of the radioactive (NH)PtCl, cis-diamminedichloroplatinum (II) or CDDP* on malignant glioma cells and verify if the low-dose continuous internal radio-chemotherapy would be able to produce additive effects. The antitumoral activity of CDDP* and the non labeled cisplatin, CDDP, were evaluated in glioblastoma. Cisplatin was cytotoxic for glioblastoma cells in a dose dependent manner. Treatment with CDDP*, (IC = 1.75 ± 0.07 μM), proved to be more potent than using just CDDP, (IC = 4.96 ± 0.40 μM). These results suggest that cisplatin is a very potent radiosensitizer evoking a supra additive effect. Internal radio-chemotherapy treatment based on CDDP* may be useful alternative to reduce the drug concentration required for effective inhibition of glioblastoma growth. [ABSTRACT FROM AUTHOR]

The antimicrobial activity of four labdane-type diterpenes isolated from the oleoresin of Copaifera langsdorffii as well as of two commercially available diterpenes (sclareol and manool) was investigated against a representative panel of microorganisms responsible for periodontitis. Among all the evaluated compounds, (-)-copalic acid (CA) was the most active, displaying a very promising MIC value (3.1 μg mL-1; 10.2 μM) against the key pathogen (Porphyromonas gingivalis) involved in this infectious disease. Moreover, CA did not exhibit cytotoxicity when tested in human fibroblasts. Time-kill curve assays performed with CA against P. gingivalis revealed that this compound only inhibited the growth of the inoculums in the first 12 h (bacteriostatic effect). However, its bactericidal effect was clearly noted thereafter (between 12 and 24 h). It was also possible to verify an additive effect when CA and chlorhexidine dihydrochloride (CHD, positive control) were associated at their MBC values. The time curve profile resulting from this combination showed that this association needed only six hours for the bactericidal effect to be noted. In summary, CA has shown to be an important metabolite for the control of periodontal diseases. Moreover, the use of standardized extracts based on copaiba oleoresin with high CA contents can be an important strategy in the development of novel oral care products. [ABSTRACT FROM AUTHOR]

A very appropriate method for antigenotoxicity evaluation of antioxidants is the comet assay, since this analytical method detects initial DNA lesions that are still subject to repair; in other words, lesions that are very associated to damages resulting from the generation and subsequent action of reactive species. However, a solid evaluation should be developed in order to avoid inexact interpretations. In our study, besides the association of curcumin with cisplatin, curcumin and cisplatin agents were also tested separately. Classical genotoxic compounds, when tested by the comet assay, present an increase in the nucleoid tail; however, the cisplatin treatment has resulted in a decrease of DNA migration. This was an expected effect, as the cross-links between cisplatin and DNA decrease the DNA electrophoretic mobility. A similar effect was observed with the curcumin treatment, which decreased the nucleoid tail. Such effect was not expected and reinforced the necessity of including in the study, separate treatment groups with potentially antigenotoxic substances. The comet assay results have been analyzed using specific software for image analysis, as well as the classical visual analysis, and we have observed that the effect of decrease in DNA electrophoretic mobility was more easily observed when the data were analyzed by the software. The article presents the author's views concerning a better approach and understanding between Brazil and Argentina, having their cultural production and background as linking element. The author offers an analysis of cultural relationships between these two countries from 1930 to 1954. New guidelines for historical interpretation of these relationships, supported by recent theoretical reflections connected to constructivism and international cultural relations are offered.

Sedimentary organic pollution in the urban reaches of the Thames estuary is changing from fossil fuel hydrocarbons to emerging synthetic chemicals. De-industrialisation of London was assessed in three cores from Chiswick (Ait/Eyot) mud island using pharmaceuticals, faecal sterols, hydrocarbons (TPH, PAH), Black Carbon (BC) and organotins (TBT). These ranked in the order; BC 7590-30219 mg/kg, mean 16,000 mg/kg > TPH 770–4301, mean 1316 mg/kg > Σ16PAH 6.93–107.64, mean 36.46 mg/kg > coprostanol 0.0091–0.42 mg/kg, mean of 0.146 mg/kg > pharmaceuticals 2.4–84.8 μg/kg, mean 25 μg/kg. Hydrocarbons co-varied down-profile revealing rise (1940s), peak (1950s -1960s) and fall (1980s) and an overall 3 to 25-fold decrease. In contrast, antibiotics, anti-inflammatory (ibuprofen, paracetamol) and hormone (17β-estradiol) increased 3 to 50-fold toward surface paralleling increasing use (1970s-2018). The anti-epileptics, carbamazepine and epoxcarbamazepine showed appreciable down-core mobility. Faecal sterols confirmed non-systematic incorporation of treated sewage. Comparison to UK sediment quality guidelines indicate exceedance of AL2 for PAH whereas TBT was below AL1.
