Background White striping (WS) is an emerging quality defect with adverse consequences for the sensorial, technological, and nutritional qualities of breast meat in broiler chickens. The genetic determinism of this defect is little understood and thus the aim of the study presented here was to estimate the genetic parameters of WS in relation to other traits of economic importance such as body weight, carcass composition, and technological meat quality in an experimental population consisting of two divergent lines selected for high (pHu + line) or low (pHu- line) ultimate pH (pHu) of the pectoralis major (p. major) muscle. Dans les troupeaux ovins allaitants, l’allaitement artificiel strict avec un lait reconstitué est fréquemment utilisé pour sauver des agneaux surnuméraires mais peut entrainer des performances décevantes. L’objectif de cette étude est d’évaluer, chez des agneaux de race Romane, l’impact de ce mode d’élevage sur leurs performances et sur leurs réponses immunitaire et comportementale à court et moyen terme. Trois traitements d’allaitement sont comparés : agneaux maternés (un agneau par mère), agneaux mis en allaitement artificiel à 3 jours d’âge et nourris soit avec du lait commercial reconstitué ou avec du lait de brebis Lacaune traites quotidiennement. Au sevrage à 45 jours, les agneaux des différents traitements ont été réallotés par sexe et alimentés avec du foin et du concentré. Les agneaux nourris avec l’aliment d’allaitement ont une croissance avant 22 jours significativement plus faible que les agneaux maternés, ceux nourris au lait de brebis étant intermédiaires. Au-delà, il n’apparait plus de différence et le poids final est équivalent. Ces agneaux allaités artificiellement, quel que soit le lait, apparaissent plus actifs en case d’élevage et moins craintifs vis-à-vis de l’homme en situation de test à J21. Lorsqu’ils sont nourris avec l’aliment d’allaitement, l’aspect de leur arrière train a tendance à être plus sale que celui des agneaux nourris au lait de brebis sans pour autant qu’ils soient plus infectés par des pathogènes intestinaux. Le stress oxydant est plus faible chez les agneaux en allaitement artificiel, malgré un statut antioxydant global moins favorable, probablement en rapport avec leur meilleure familiarisation à l’homme lors des prélèvements. Par contre le lait de brebis Lacaune prépare mieux les agneaux au stress du sevrage. De même, la réponse immunitaire à une vaccination contre la chlamydiose est plus forte pour ce traitement. Dans les conditions de cette étude avec un faible taux de mortalité et un effectif d’agneaux limité, l’allaitement artificiel strict pénalise la croissance et le statut antioxydant et favorise la relation à l’homme uniquement dans la période d’allaitement. Les différences s’estompent rapidement après le sevrage., In sheep production, artificial rearing with commercial milk is frequently used to save lambs born from large litters but is usually connected with poor animal performance. The objective of this study was to estimate the impact of this rearing method on the performance of lambs from the Romane breed, but also their short and medium-term immune and behavioual responses. Lambs were assigned to three rearing treatments: left with the mother (one lamb per dam), or separated at 3 days of age and fed either with a commercial milk substitute or with milk from Lacaune ewes milked daily. When weaned at 45 days, the lambs of the three treatments were mixed, reared in single sex groups and fed with hay and concentrate. Lambs fed commercial milk had significantly lower growth rate before 22 days than mothered lambs, those fed Lacaune milk being intermediate. No difference was detectable later on and the final weight was similar. Artificially reared lambs, regardless of the type of milk, seemed more active in their rearing pens and less afraid of humans in tests performed at 21 days. The fleece around the perianal area of lambs fed commercial milk tended to be dirtier than in lambs fed with ewe’s milk without any increase on the presence of pathogens in the faeces. Consumption of artificial milk was associated with lower antioxidant status and higher level of stress at 30 days of age compared to mothered lambs. The immune response following vaccination against chlamydiosis was not different between lambs fed artificial milk and those nursed by their mother. In this study where mortality rate was low, artificial rearing was seen to impair growth rate and antioxidant status and to promote a lamb human relationship only during the artificial rearing period.

The aim of this study was to evaluate the capacity of chickens to adapt to and compensate for early dietary restriction of non-phytate P ( NPP: ) and/or Ca (10 to 21 d) in a later phase (22 to 35 d), and to determine whether compensatory processes depend on the P and Ca concentrations in the finisher diet. Four diets were formulated and fed to broilers from 10 to 21 d in order to generate birds with different mineral status: L1 (0.6% Ca, 0.30% NPP), L2 (0.6% Ca, 0.45% NPP), H1 (1.0% Ca, 0.30% NPP), and H2 (1.0% Ca, 0.45% NPP). On d 22, each group was divided into three groups which received a low (L, 0.48% Ca, 0.24% NPP), moderate (M, 0.70% Ca, 0.35% NPP), or high (H, 0.90% Ca, 0.35% NPP) finisher diet until 35 d, resulting in a total of 12 treatments. Lowering the Ca level enhanced apparent ileal digestibility of P (P AID) at 21 d especially with the high NPP level (Ca × NPP, P < 0.01). The lower bone mineralization observed at 21 d in broilers fed the L1 diet compared to those fed the H2 diet had disappeared by 35 d with long-term stimulation of the P AID with the low NPP level (P

International audience; Exposure of laying hens to chronic heat stress results in loss of egg production. It should be possible to improve hen resilience to chronic heat stress by genetic selection but measuring their sensitivity through internal temperature is time consuming and is not very precise. In this study we used infrared thermography to measure the hen’s capacity to dissipate heat, in a commercial line of laying hens subjected to cycles of neutral (N, 19.6°C) or high (H, 28.4°C) ambient temperatures. Mean body temperatures (BT) were estimated from 9355 infrared images of wing, comb and shank taken from 1200 hens. Genetic parameters were estimated separately for N and H temperatures. Correlations between BT and plumage condition were also investigated. Wing temperature had low heritability (0.00 to 0.09), consistent with the fact that wing temperature mainly reflects the environmental temperature and is not a zone of heat dissipation. The heritability of comb temperature was higher, from 0.15 to 0.19 in N and H conditions, respectively. Finally, the shank temperature provided the highest heritability estimates, with values of 0.20 to 0.22 in H and N conditions, respectively. Taken together, these results show that heat dissipation is partly under genetic control. Interestingly, the genetic correlation between plumage condition and shank and comb temperatures indicated that birds with poor condition plumage also had the possibility to dissipate heat through featherless areas. Genetic correlations of temperature measurements with egg quality showed that temperatures were correlated with egg width and weight, yolk brightness and yellowness and Haugh units only under H conditions. In contrast, shell colour was correlated with leg temperature only at thermo-neutrality.

International audience; Milk and dairy products are an important source of Ca for humans. Recent studies have shown fluctuations in cow milk Ca content during the year in France, with high values in winter and with corn silage diets, and a decrease during May and June and with grass diets. The aim of this study was to identify the reasons for this seasonal decrease in milk Ca content by testing the effect of 2 levels of dietary cation-anion differences (DCAD; 0 mEq/kg of dry matter for DCAD 0 and 400 mEq/kg for DCAD 400) and 2 day lengths (8 h of light/d for short days: SD; and 16 h/d for long days: LD) on the Ca balances of dairy cows. The DCAD treatments were designed to mimic diets based either on corn silage or on herbage. The cows were only illuminated by solarium lights providing UVA and UVB. The trial was conducted according to 2 simultaneous replicates of a 4 × 4 Latin square design using 8 dairy cows averaging 103 ± 44 d in milk with 4 periods of 14 d. Data were analyzed by ANOVA with a model including treatment, cow, and period effects. No significant interaction was found between day length and DCAD treatments. With DCAD 400 compared with DCAD 0, blood pH increased and plasma ionized Ca content decreased, whereas the plasma total Ca content did not differ between treatments. Milk Ca content, however, increased with DCAD 400 compared with DCAD 0, in relation to a decrease in the amount of Ca excreted in urine. The DCAD had no significant effect on protein and casein contents and DCAD 400 tended to decrease milk yield. This illustrates that the udder did not decrease Ca uptake from the blood at high DCAD even though DCAD 400 decreased the mammary availability of Ca by decreasing the proportion of blood ionized Ca. Milk Ca and casein contents were significantly lower with LD compared with SD, whereas day length had no effect on milk yield after 14 d of treatment. Bone accretion of cows increased when the Ca content of milk increased (i.e., with DCAD 400 compared with DCAD 0 and with SD compared with LD). This work suggests that long and sunny days could explain part of the seasonal decrease in milk Ca content in summer and refutes the hypothesis that low milk Ca contents at grazing could be due to the high DCAD of herbage.

Background Meat type chickens have limited capacities to cope with high environmental temperatures, this sometimes leading to mortality on farms and subsequent economic losses. A strategy to alleviate this problem is to enhance adaptive capacities to face heat exposure using thermal manipulation (TM) during embryogenesis. This strategy was shown to improve thermotolerance during their life span. The aim of this study was to determine the effects of TM (39.5 °C, 12 h/24 vs 37.8 °C from d7 to d16 of embryogenesis) and of a subsequent heat challenge (32 °C for 5 h) applied on d34 on gene expression in the Pectoralis major muscle (PM). A chicken gene expression microarray (8 × 60 K) was used to compare muscle gene expression profiles of Control (C characterized by relatively high body temperatures, Tb) and TM chickens (characterized by a relatively low Tb) reared at 21 °C and at 32 °C (CHC and TMHC, respectively) in a dye-swap design with four comparisons and 8 broilers per treatment. Real-time quantitative PCR (RT-qPCR) was subsequently performed to validate differential expression in each comparison. Gene ontology, clustering and network building strategies were then used to identify pathways affected by TM and heat challenge. Results Among the genes differentially expressed (DE) in the PM (1.5 % of total probes), 28 were found to be differentially expressed between C and TM, 128 between CHC and C, and 759 between TMHC and TM. No DE gene was found between TMHC and CHC broilers. The majority of DE genes analyzed by RT-qPCR were validated. In the TM/C comparison, DE genes were involved in energy metabolism and mitochondrial function, cell proliferation, vascularization and muscle growth; when comparing heat-exposed chickens to their own controls, TM broilers developed more specific pathways than C, especially involving genes related to metabolism, stress response, vascularization, anti-apoptotic and epigenetic processes. Conclusions This study improved the understanding of the long-term effects of TM on PM muscle. TM broilers displaying low Tb may have lower metabolic intensity in the muscle, resulting in decreased metabolic heat production, whereas modifications in vascularization may enhance heat loss. These specific changes could in part explain the better adaptation of TM broilers to heat. Agroecology uses ecological processes and local resources rather than chemical inputs to develop productive and resilient livestock and crop production systems. In this context, breeding innovations are necessary to obtain animals that are both productive and adapted to a broad range of local contexts and diversity of systems. Breeding strategies to promote agroecological systems are similar for different animal species. However, current practices differ regarding the breeding of ruminants, pigs and poultry. Ruminant breeding is still an open system where farmers continue to choose their own breeds and strategies. Conversely, pig and poultry breeding is more or less the exclusive domain of international breeding companies which supply farmers with hybrid animals. Innovations in breeding strategies must therefore be adapted to the different species. In developed countries, reorienting current breeding programmes seems to be more effective than developing programmes dedicated to agroecological systems that will struggle to be really effective because of the small size of the populations currently concerned by such systems. Particular attention needs to be paid to determining the respective usefulness of cross-breeding v. straight breeding strategies of well-adapted local breeds. While cross-breeding may offer some immediate benefits in terms of improving certain traits that enable the animals to adapt well to local environmental conditions, it may be difficult to sustain these benefits in the longer term and could also induce an important loss of genetic diversity if the initial pure-bred populations are no longer produced. As well as supporting the value of within-breed diversity, we must preserve between-breed diversity in order to maintain numerous options for adaptation to a variety of production environments and contexts. This may involve specific public policies to maintain and characterize local breeds (in terms of both phenotypes and genotypes), which could be used more effectively if they benefited from the scientific and technical resources currently available for more common breeds. Last but not least, public policies need to enable improved information concerning the genetic resources and breeding tools available for the agroecological management of livestock production systems, and facilitate its assimilation by farmers and farm technicians.

Is Winter Past? Some plants are adapted to wait through the winter before their genetic programs for flowering are initiated. But the duration of winter varies with location, and thus, to avoid flowering too late into the summer or exposing blossoms to frost, the adaptation needs to hold the flowers off for just the right amount of time. Working with Arabidopsis thaliana , Coustham et al. (p. 584 , published online 12 July) identified a quantitative mechanism that “counts” off the duration of winter. FLC, a repressor of flowering, carries sequence polymorphisms that direct epigenetic events. Plants with fewer of these polymorphisms are adapted to short winters, and plants with all four polymorphisms are adapted to long winters.

Gizzard motility was compared between chickens from D(+) (high digestion efficiency) and D(-) (low digestion efficiency) genetic lines selected for divergent digestion efficiency, using strain gauge transducers. Motility was recorded continuously during 24 h in 6 birds per line. Two stimuli, a meal distribution after a feed-deprivation period and lighting after a dark period, were tested during the recording period. A functional test with intravenous injection of serotonin performed at the end of the recording day often resulted in a sharp reduction in gizzard motility, without a significant difference between lines. Compared with D(+) birds, gizzard activity in D(-) birds remained high during fasting or dark periods (P < 0.0006), which reduced the effects of stimuli in D(-) birds. So, coordination between bird activity and gizzard motility tended to be reduced in D(-) compared with D(+) birds (P = 0.0018). This coordination was observed to be positively correlated (P = 0.011) with the relative weight (g/kg of BW) of the stomach (gizzard + proventriculus). This experiment pointed out differences in gizzard motility between D(+) and D(-) chicken lines in terms of response to environmental stimuli, characterized by a failure in the gizzard relaxation process in D(-) birds during rest periods.

Optimizing phosphorus (P) utilization in pigs requires improving our capacity to predict the amount of P absorbed and retained, with the main modulating factors taken into account, as well as precisely determining the P requirements of the animals. Given the large amount of published data on P utilization in pigs, a meta-analysis was performed to quantify the impact of the different dietary P forms, calcium (Ca) and exogenous phytases on the digestive and metabolic utilization criteria for dietary P in growing pigs. Accordingly, the amount of phytate P (PP) leading to digestible P (g/kg) was estimated to be 21%, compared with 73% for non-phytate P (NPP) from plant ingredients and 80% for NPP from mineral and animal ingredients (P < 0.001). The increase in total digestible dietary P following the addition of microbial phytase (PhytM) from Aspergillus niger (P < 0.001) was curvilinear and about two times higher than the increase following the addition of plant phytase, which leads to a linear response (P < 0.001). The response of digestible P to PhytM also depends on the amount of substrate, PP (PhytM(2) × PP, P < 0.001). The digestibility of dietary P decreased with dietary Ca concentration (P < 0.01) independently of phytase but increased with body weight (BW, P < 0.05). Although total digestible dietary P increased linearly with total NPP concentration (P < 0.001), retained P (g/kg), average daily gain (ADG, g/day) and average daily feed intake (ADFI, g/day) increased curvilinearly (P < 0.001). Interestingly, whereas dietary Ca negatively affected P digestibility, the effect of dietary Ca on retained P, ADG and ADFI depended on total dietary NPP (NPP × Ca, P < 0.01, P < 0.05 and P < 0.01, respectively). Increasing dietary Ca reduced retained P, ADG and ADFI at low NPP levels, but at higher NPP concentrations it had no effect on ADG and ADFI despite a positive effect on retained P. Although the curvilinear effect of PhytM on digestible P increased with PP (P < 0.001), this effect was lessened by total NPP for ADG and ADFI (PhytM × NPP and PhytM(2) × NPP, P < 0.05) and depended on both total NPP and Ca for retained P (PhytM(2) × NPP × Ca, P < 0.01). This meta-analysis improves our understanding of P utilization, with major modulating factors taken into account. The information generated will be useful for the development of robust models to formulate environmentally friendly diets for growing pigs.

Increased stocking densities are frequently reported to depress chicken growth performance, but the mechanisms behind this are not fully understood. This study was conducted to investigate the effects of stocking density on growth performance and digestive microbiota, known to be sensitive to environmental factors. Chickens were reared at 2 stocking densities, 12 or 17 birds/m(2). Growth performance was recorded between d 1 and 39, and litter was scored for quality on d 25, 31, and 37. Digestive microbiota was analyzed along the digestive tract (crop, ileum, ceca) of 3- and 6-wk-old chickens by using 2 molecular approaches: a qualitative method (fingerprinting by temporal temperature gradient gel electrophoresis) and a quantitative method (real-time PCR). An increase in stocking density was found to negatively affect the feed conversion ratio (+3.1%) and depress the daily BW gain of broilers (-5.5%) during the period from d 32 to 39 (P

Toll-like receptor 4 (TLR4), which recognizes lipopolysaccharide from Gram-negative bacteria, plays a major role in resistance of mice and humans to Salmonella infection. In chickens, Salmonella may establish a carrier state whereby bacteria are able to persist in the host organism without triggering clinical signs. Based on cellular morphological parameters, we developed a method, without using antibodies, to separate three cecal cell subpopulations: lymphocytes, enterocytes, and a population encompassing multiple cell types. We analyzed the mRNA expression of TLR4, interleukin-1β (IL-1β), IL-8, IL-12, and lipopolysaccharide-induced tumor necrosis factor alpha factor (LITAF) in cecal subpopulations of chicks from inbred lines resistant or susceptible to the carrier state infected with Salmonella enterica serovar Enteritidis. The results showed that resistance to the carrier state in chicks is associated with a larger percentage of lymphocytes and with higher levels of expression of TLR4 and IL-8 at homeostasis in the three cell subpopulations, as well as with a higher level of expression of LITAF in lymphocytes during the carrier state. In contrast to the early phase of infection, the carrier state is characterized by no major cell recruitment differences between infected and noninfected animals and no significant modification in terms of TLR4, IL-1β, IL-8, IL-12, and LITAF expression in all cell subpopulations measured. However, TLR4 expression increased in the lymphocytes of chicks from the susceptible line, reaching the same level as that in infected chicks from the resistant line. These observations suggest that the carrier state is characterized by a lack of immune activation and highlight the interest of working at the level of the cell population rather than that of the organ.

International audience; Protein metabolism is considered to be regulated by amino acids, with major consequences on tissue development. There is evidence that lysine greatly affects carcass composition and muscle growth. In particular, a drastic effect of dietary provision of lysine has been observed on breast muscle development in chickens. Other essential amino acids, such as threonine and valine, do not have as pronounced an effect as lysine on body composition. Increasing lysine can also improve chicken breast muscle quality by increasing its ultimate pH and water holding capacity, but the underlying mechanisms are still unknown. Studies conducted over the last ten years indicate that, in addition to being substrates for protein synthesis, amino acids act as modulators of signal transduction pathways that control metabolism and cell functions. For instance, certain amino acids can modulate the activity of the intracellular protein kinases involved in the control of mRNA translation. Interestingly, enhanced responses to amino acids have been reported during the neonatal period, suggesting that early protein nutrition impacts on the development of broiler chicks. Methionine and cysteine have a very significant place among amino acids because they have several additional roles: they are precursors of essential molecules, for example cysteine is used for the synthesis of the antioxidant glutathione, and thus participates in the control of oxidative status, methionine is a source of the methyl groups needed for all biological methylation reactions, including methylation of DNA and histones, etc. These findings together indicate the importance of optimizing amino acid nutrition and providing a rationale for nutritional advice.

Evolutionary analyses of biological data are becoming a prerequisite in many fields of biology. At a time of high-throughput data analysis, phylogenetics is often a necessary complementary tool for biologists to understand, compare and identify the functions of sequences. But available bioinformatics tools are frequently not easy for non-specialists to use. We developed PhyleasProg (http://phyleasprog.inra.fr), a user-friendly web server as a turnkey tool dedicated to evolutionary analyses. PhyleasProg can help biologists with little experience in evolutionary methodologies by analysing their data in a simple and robust way, using methods corresponding to robust standards. Via a very intuitive web interface, users only need to enter a list of Ensembl protein IDs and a list of species as inputs. After dynamic computations, users have access to phylogenetic trees, positive/purifying selection data (on site and branch-site models), with a display of these results on the protein sequence and on a 3D structure model, and the synteny environment of related genes. This connection between different domains of phylogenetics opens the way to new biological analyses for the discovery of the function and structure of proteins.

Les poulets à croissance rapide sont sensibles à la chaleur en période de finition. En effet, leur sélection pour la croissance a favorisé la masse musculaire au détriment d’organes impliqués dans la thermorégulation. Ceci peut se traduire par des baisses de performances en cas de chaleur modérée et chronique, mais également des problèmes de bien-être et des mortalités lors d’un coup de chaleur. Durant la dernière décennie les efforts ont porté sur l’acclimatation à la chaleur pendant la période périnatale, notamment pendant l’embryogenèse. Cette stratégie nécessite de mettre au point les conditions d’incubation (température, phase de l’embryogenèse, durée d’exposition…) qui favorisent la thermotolérance jusqu’en finition. Les conséquences sur les performances des poulets (croissance, qualité de viande…) sont évaluées. L’acclimatation périnatale met en jeu des mécanismes de régulation épigénétiques qui modulent à long terme la température interne des volailles. A terme, si cette technique se révélait applicable à grande échelle, elle pourrait favoriser la robustesse et le bien-être des volailles face aux variations de températures, tout en préservant leurs performances zootechniques. Dans cet article, sont rappelés les principales techniques d’exposition thermique embryonnaire et les fondements physiologiques de leur mise en œuvre. Indeed, broiler chickens have limited capacities to sustain high temperatures, with reduced performances during moderate and chronic heat exposure, and depressed welfare, morbidity and mortality during acute heat exposure. During the last decade, a great effort concerned early thermal acclimation. This strategy requires a fine tuning of incubation conditions (temperature, phasis of embryogenesis, duration of exposure) favoring long term thermotolerance. Consequences on growth performance and meat quality were also evaluated. Perinatal heat acclimation is aimed at inducing epigenetic thermoregulatory mechanisms that modulate body temperature on a long term. If it is further proved to be efficient on a large scale, early thermal acclimation could favor poultry sturdiness towards temperature variations without affecting growth performance. In this paper, we will review the main techniques of early thermal exposure and the physiological bases of their use. We will focus on the research axes concerning the comprehension of underlying mechanisms and the interest of this technique for poultry species.

In view of the complexity of meat processes after slaughter and quality trait determination, it is not surprising that the results obtained in different studies and laboratories are not always in agreement. For comparison of results it is therefore necessary to keep strictly to measurable specifications, which is why standardisation is indispensable. The Working Group 5 Poultry Meat Quality group of the WPSA European Federation has been asked to produce a document which would serve as a common base methodology that would permit comparison between research projects carried out by different groups, based on international research programmes. This paper represents the first step of this work including chemical (moisture, total lipids, proteins, ash, fatty acid composition, cholesterol, susceptibility to oxidation, amino acids, collagen and pigments) and physical traits (pH, R-value, colour, water holding capacity, texture and sarcomere length). For the evaluation of chemical composition, there are standard methods available which are largely adopted in the majority of published papers. However, there is still a need to standardise methods for determining the physical traits to facilitate comparisons between studies and to provide reference values.

International audience; Under current practices, broiler parent stock (broiler breeders) encounter several welfare problems, such as feed restriction and injury during mating. Intensive selection for production traits, especially growth rate, is associated with increased nutritious requirement and thus feed consumption, but also reproductive dysfunctions and decreased sexual activity in broiler breeders. A resulting serious welfare problem is the subsequent severe feed restriction which is applied during rearing, in order to prevent health problems and to achieve better egg production. This severe feed restriction has negative effects on bird welfare as it causes chronic stress resulting from hunger. Recent research has focused on management practices to alleviate the negative effect of this feed restriction. Using both a more fibrous feed and an appetite suppressant seems most successful. There are welfare concerns involved in the mating of broiler breeders. In many countries mutilations (beak trimming, despurring and toe clipping) are carried out in male broiler breeders as a standard procedure to prevent excessive damage to the hens during mating. It has been reported that broiler breeder males show low libido but, at the same time, may be aggressive and show rough behaviour during mating, which can cause severe injuries to the hens, although some injuries may be caused by over-mating hens. Current studies aim to develop management practices to reduce hen injury due to over-mating and improve the sexual behaviour in both males and females. In the last part of the review, an examination of how breeding companies are taking welfare into consideration in their commercial selection scheme is made. In practice, although for example reduced mortality and some health criteria are included in selection programs and some alternative breeds as used, other welfare issues remain, such as feed restriction during rearing.

International audience; The avian endogenous virus gene (ev21) and dominant sex-linked late feathering (LF) gene (K) are closely associated on the Z chromosome of LF chickens. The K gene is linked to two large repeats: ev21-unoccupied repeat region (URa) and ev21-occupied repeat region (OR). On the other hand, the recessive allele (early feathering, EF: k(+)) is linked to ev21-unoccupied region (URb). These three regions show high similarity but due to minor sequence differences, restriction enzyme treatment distinguishes URa and URb in White Leghorn. However, the applicability of the PCR-RFLP technique or the association between ev21 and K gene are not well investigated in the other breeds or strains. In this study, ev21 loci were detected in White Leghorn, Nagoya, Silky, Geline but not Gifujidori breeds of chickens, and the URa and URb from the various breeds were sequenced. Four types of sequence (URa-1 and URa-2 from LF or ev21 positive, URb-1 and URb-2 from EF or ev21 negative) were identified. At position 1072-1075, URa-2, URb-1 and URb-2 had "GGCC" whereas URa-1 had "AGCC". The transition at 1072 clearly indicates that the PCR-RFLP technique used for White Leghorn is not applicable to Nagoya. Interestingly, both URa-1 and URa-2 were obtained from the ev21 positive sample of Geline. Presence of different sequence of URa in the Geline may explain that fixation of one of sequences to White Leghorn or Nagoya had occurred during breed establishment.

Selective breeding is an effective tool to improve livestock. Several selection experiments have been conducted to study direct selection responses as well as correlated responses in traits of skeletal muscle growth and function. Moreover, comparisons of domestic with wild-type species and of extreme breeds provide information on the genetic background of the skeletal muscle phenotype. Structural muscular components that differed with increasing distance in lean growth or meat quality in mammals were found to be myofibre number, myofibre size, proportions of fibre types as well as the numbers and proportions of secondary and primary fibres. Furthermore, markers of satellite cell proliferation, metabolic enzyme activities, glycogen and fat contents, the expression of myosin heavy chain isoforms, of activated AMPKα and other proteins in skeletal muscle tissue and circulating IGF1 and IGF-binding proteins have been identified to be involved in selection responses observed in pigs, cattle and/or chicken. The use of molecular methods for selective breeding of fish has only recently been adopted in aquaculture and studies of the genetic basis of growth and flesh quality traits are scarce. Some of the molecular markers of muscle structure/metabolism in livestock have also been identified in fish, but so far no studies have linked them with selection response. Genome scans have been applied to identify genomic regions exhibiting quantitative trait loci that control traits of interest, for example, muscle structure and meat quality in pigs and growth rate in chicken. As another approach, polymorphisms in candidate genes reveal the relationship between genetic variation and target traits. Thus, in large-scale studies with pigs' associations of polymorphisms in the HMGA2, CA3, EPOR, NME1 and TTN genes with traits of carcass and meat quality were detected. Other studies revealed the significance of mutations in the IGF2 and RYR1 genes for carcass lean and muscle fibre traits in pigs. Mutations in the myostatin (MSTN) gene in fish were also examined. Advances in research of the genetic and environmental control of traits related to meat quality and growth have been made by the application of holistic 'omics' techniques that studied the whole muscle-specific genome, transcriptome and proteome in relation to muscle and meat traits, the development of new methods for muscle fibre typing and the adaptation of biophysical measures to develop parameters of muscle fibre traits as well as the application of in vitro studies. Skeletal muscle development in vertebrates - also termed myogenesis - is a highly integrated process. Evidence to date indicates that the processes are very similar across mammals, poultry and fish, although the timings of the various steps differ considerably. Myogenesis is regulated by the myogenic regulatory factors and consists of two to three distinct phases when different fibre populations appear. The critical times when myogenesis is prone to hormonal or environmental influences depend largely on the developmental stage. One of the main mechanisms for both genetic and environmental effects on muscle fibre development is via the direct action of the growth hormone-insulin-like growth factor (GH-IGF) axis. In mammals and poultry, postnatal growth and function of muscles relate mainly to the hypertrophy of the fibres formed during myogenesis and to their fibre-type composition in terms of metabolic and contractile properties, whereas in fish hyperplasia still plays a major role. Candidate genes that are important in skeletal muscle development, for instance, encode for IGFs and IGF-binding proteins, myosin heavy chain isoforms, troponin T, myosin light chain and others have been identified. In mammals, nutritional supply in utero affects myogenesis and the GH-IGF axis may have an indirect action through the partitioning of nutrients towards the gravid uterus. Impaired myogenesis resulting in low skeletal myofibre numbers is considered one of the main reasons for negative long-term consequences of intrauterine growth retardation. Severe undernutrition in utero due to natural variation in litter or twin-bearing species or insufficient maternal nutrient supply may impair myogenesis and adversely affect carcass quality later in terms of reduced lean and increased fat deposition in the progeny. On the other hand, increases in maternal feed intake above standard requirement seem to have no beneficial effects on the growth of the progeny with myogenesis not or only slightly affected. Initial studies on low and high maternal protein feeding are published. Although there are only a few studies, first results also reveal an influence of nutrition on skeletal muscle development in fish and poultry. Finally, environmental temperature has been identified as a critical factor for growth and development of skeletal muscle in both fish and poultry. Copyright © 2010 The Animal Consortium.

International audience; This study was conducted to evaluate the effect of early rearing conditions on physiological, haematological and immunological responses relevant to adaptation and long-term stress in white Leghorn hens with intact beaks housed in furnished cages (FC) or conventional cages (CC) during the laying period. Pullets were cage reared (CR) or litter floor reared (FR). From 16 to 76 weeks of age, hens were housed in FC (eight hens per cage) or in CC (three hens per cage). As measures of long-term stress at the end of the laying period, adrenal reactivity was quantified by assessing corticosterone responses to adrenocorticotropin challenge, and immune response was assessed by measuring antibody responses after immunization with sheep red blood cells (SRBC) and keyhole limpet haemocyanin (KLH). Heterophil to lymphocyte (H/L) ratio was employed as an indicator of stress. Rearing conditions significantly affected anti-SRBC titres (P < 0.0001) and tended to affect H/L ratios (P = 0.07), with the highest values found in FR hens. Layer housing affected H/L ratio (P < 0.01); the highest ratio was found in FR birds housed in FC during the laying period. This study shows that early rearing environment affects immunological indicators that are widely used to assess stress in laying hens. However, while results on H/L ratio indicated that FR birds experienced more stress particularly when they were housed in FC during the laying period, the immune responses to SRBC in FR hens was improved, indicating the opposite. This contradiction suggests that the effects on immune response may have been associated with pathogenic load due to environmental complexity in FR and FC hens rather than stress due to rearing system or housing system per se.

Samples in the calibration and validation databases (DB1 and DB2) corresponded to animals fed with a similar wheat-based diet. A second validation study was performed on excreta samples from animals fed more variable diets, including peas and maize (DB3). Excreta samples were freeze-dried and ground. Near infrared reflectance spectra were taken on a monochromator spectrometer between 400 nm and 2500 nm. Samples were analysed for mineral matter (MM), gross energy (GE), starch, crude fat (CFAT), total nitrogen (NTOT), uric acid nitrogen (NUA) and protein nitrogen estimated directly (PNTERP) or by difference between NTOT and NUA (PNUA). Depending on the parameters studied, 250 to 700 samples were analysed by reference methods. The standard error of cross-validation (SECV) and R² of calibrations were: 0.60% and 0.96 for MM, 166 kJ kg –1 and 0.99 for GE, 0.59% and 1.00 for starch, 0.44% and 0.99 for CFAT, 0.25% and 0.89 for NTOT and 0.22% and 0.97 for NUA, respectively. Calibration for PNTERP (SECV = 0.07%; R²=0.98) was much more precise than PNUA (SECV = 0.21%, R² = 0.85). Validation carried out on databases DB2 and DB3 resulted in standard errors of prediction (on DB2) and extrapolation (on DB3) generally higher than SECV, while remaining relatively precise with prediction r² values from 0.83 to 0.99 and extrapolation r² from 0.86 to 0.98, with the exception of PNUA for which r² was 0.22 and 0.64, respectively. For some parameters, the lower validation performance was due to biases, particularly in the case CFAT and NUA for prediction and MM, GE and NUA for extrapolation. Global calibrations made with DB1+DB2+DB3 were more precise (GE, NTOT) or equally precise (all other parameters) than with DB1 alone. These results confirmed the potential precision of calibrations for the major organic compounds in poultry excreta and suggested that their use could be extended to excreta issued from a wider range of diets without losing precision.

La France est actuellement au premier rang des productions européennes de poulets et d’œufs biologiques. Ces productions ne représentent toutefois au mieux que quelques pourcents de la production nationale. Elles sont en régression pour le poulet ou stable pour l’œuf et ne couvrent pas les besoins du marché intérieur. Dans un contexte à la fois de demande du citoyen, d’une volonté politique nationale affichée et d’évolution du règlement européen, l’aviculture biologique française pourrait reprendre sa croissance en saisissant ces opportunités pour se développer. Aussi nous proposons d’analyser les atouts et les verrous qui caractérisent ce mode de production. Les analyses multicritères de ces productions restent à conduire à l’échelle du territoire. Elles devront prendre en compte les différentes composantes biotechniques, depuis la gestion agronomique des sols jusqu’au produit terminal, en passant par la production territorialisée des matières premières végétales, de même que les composantes économiques, sociétales ou politiques. Au-delà des images positives que les produits biologiques véhiculent en termes de bien-être, de qualité des produits ou de bilan environnemental, certaines contraintes déjà identifiées sont autant d’obstacles à leurs développements. Il convient donc de les analyser finement et de les prendre en compte afin de définir de nouveaux itinéraires de production et de ommercialisation. Ceux-ci devront permettre de concilier un développement cohérent et durable, répondant à des attentes sociétales complexes, de produire des produits de qualité, respectueux de l’environnement, accessibles au plus grand nombre et favorisant l’emploi rural et le développement des territoires. Cette problématique complexe constitue l’objet de deux programmes de recherches pluriannuels complémentaires initiés en 2009, AlterAviBio et AviBio., France currently leads European organic chicken and egg production. Each production, however, represents only a small percentage of national production. Organic production has declined for chicken and remains stable for eggs, but each does not cover the respective domestic demand. The French organic poultry industry could resume growth by taking advantage of new opportunities for development within a context that includes consumer pressures, expressed political will, and evolution of European regulations. We analyse the advantages and bottlenecks characteristic of these types of production. Multicriteria analyses of these production systems must be done at the national scale. They should include the various biotechnical components, from agricultural soil management to the final product, via regional production of raw vegetable matter, along with economic, social, and political components. Despite the positive image that organic products have in terms of animal welfare, product quality, or environmental respect, some previously identified constraints are obstacles to their development. It is therefore necessary to take them into account and carefully analyse them to define new production and commercial paths. These paths should accommodate a consistent and sustainable development, meet society's complex expectations, produce products of quality that respect the environment and are available to many, and favour rural employment and regional development. This complex problem is the object of two complementary pluriannual research programmes beginning in 2009, AlterAviBio and AviBio.

Hypothalamic neuropeptides, neurotrophins, and systemic hormones modulate food intake and body composition. Although advances toward elucidating these interactions have been made, many aspects of the underlying mechanisms remain vague. Hypothalami from fat and lean chicken lines were assessed for differential expression of anabolic/orexigenic and catabolic/anorexigenic genes. Effects of triiodothyronine (T3), corticosterone (Cort), and brain-derived neurotrophic factor (BDNF) on expression of anabolic/orexigenic and catabolic/anorexigenic genes were tested in cultures of hypothalamic neurons. From this, we found that BDNF increased and T3decreased gene expression for BDNF, leptin receptor (LEPR), pro-opiomelanocortin (POMC), thyrotropin releasing hormone (TRH), and agouti-related protein (AGRP). Thyroid hormone levels were manipulated during development to show that T3inhibited BDNF, TRH, and BDNF receptor gene expression. Delivery of T3, Cort, T3plus Cort, or vehicle in vivo continuously for 72 h indicated that Cort and T3have overlapping roles in regulating TRH, LEPR, and POMC gene expression and that Cort and T3regulate BDNF, neuropeptide Y, and AGRP in opposite directions. Collectively, these findings suggest that interactions between the neuropeptide BDNF and the hormones T3and/or Cort may constitute a homeostatic mechanism that links hypothalamic energy regulation controlling body composition.

Chantier qualité GA; International audience; The occurrence of interchromosomal effects (ICE) in reciprocal translocation carriers still remains contradictory in the human literature. We used the pig as an animal model to investigate whether the structure of the reciprocal translocations as well as the size and/or type of the chromosomes not involved in the rearrangement may influence the occurrence and the extent of ICE. Analyses of chromosomal sperm content by fluorescence in situ hybridization (FISH) using whole-chromosome painting probes for 7 chromosomes (1, 10, 11, 13, 18, X and Y) were carried out on sperm samples of 2 boars with normal semen parameters carrying different balanced reciprocal translocations: 38, XY, t(3;15)(q27;q13) or 38, XY, t(12;14)(q13;q21). One fertile boar with normal karyotype was also studied as a control. Aneuploidy rates for the 7 chromosomes were estimated by scoring 10,000 to 20,000 spermatozoa for each probe combination. No significant ICE was found except for chromosome 1 in the case of the t(3;15) translocation. Even if statistically significant, this ICE remained very weak and should have very little impact on the reproductive performance of the carrier boar. The size and/or type of chromosomes not involved in the translocation do not seem to have a major influence on the occurrence of ICE. The structure of the translocation could play a role, but complementary studies should be carried out to confirm this assumption.

Chantier qualité GA; International audience; The aim of this study was to determine the specific characteristics of carcass and meat from an old French chicken breed, the 'Geline de Touraine' (GT), characterised by a very slow-growing rate and usually slaughtered at 120 days of age. For this purpose, we compared the GT with an experimental crossbreed (EC) exhibiting the same growth rate, and with a 'Label rouge' (LR) genotype usually slaughtered at 84 days of age. A total of 250 males and 250 females per genotype were reared by separating sexes and genotypes. The growth performances were recorded. At 84 days of age, 80 birds per sex and per genotype were slaughtered. The frequency of clawing and pecking injuries on the carcass was noted. We also measured the skin colour and the thickness of wing membrane. The relative percentages of carcass, breast, thigh + drumstick, abdominal fat, testis or ovary to body weight were determined. On breast and thigh muscles the ultimate pH (pHu) and colour were measured. The juice loss after 3 days' storage at + 4 degrees C and after cooking at 85 degrees C, and the shear force value of Warner-Bratzler were only measured on breast muscles. At 120 days of age, we repeated the same measurements but only on EC and GT genotypes in order to compare birds at the same age or at the respective slaughter age for each production. Whatever the slaughter age, the body weight of males was always higher than that of the females but the carcass yield was similar for both sexes. The females had higher breast yield and carcass fatness but lower thigh + drumstick yield than the males. The yellowness of skin and meat was higher for the females than for the males while the contrary was observed for the redness of the meat. The breast meat of the females also had higher cooking loss than that of the males. GT and EC birds exhibited a higher occurrence of carcass defects and a higher pHu in meat than LR birds. The GT chickens were characterised by a lower breast yield, a higher fattiness and an earlier sexual maturity than the other genotypes, which could confer typical sensorial attributes to their meat. Finally, the EC chickens exhibited a skin and a meat more coloured than the other genotypes, particularly for yellowness, a character which could be under genetic control.

International audience; Protein synthesis is affected when an insufficient level of sulfur amino acids is available. This defect may originate from dietary amino acid deficiency and/or excessive amino acid utilisation for other purposes such as the synthesis of glutathione and acute-phase proteins during catabolic stress. Sulfur amino acids are recognised to exert other significant functions since they are precursors of essential molecules, are involved in the methylation process, participate in the control of oxidative status, and may act as mediators affecting metabolism and cell functions. Despite this increased understanding of the role of sulfur amino acids, many questions still remain unanswered due to the complexity of the mechanisms involved. Moreover, surprising effects of dietary sulfur amino acids have been reported, with the development of disorders in cases of both deficiency and excess. These findings indicate the importance of defining adequate levels of intake and providing a rationale for nutritional advice. The aim of the present review is to provide an overview on the roles of sulfur amino acids as regulators of nutrient metabolism and cell functions, with emphasis placed on the implications for nutrition.

International audience; Emerging evidence showed that variations in environmental temperature affect both leptin and uncoupling protein (UCP) gene expression in mammals, whereas a little is known about such interactions in birds. Thus, we conducted the present study to investigate the influence of acute (2 hours) cold (4 degrees C) and chronic (10 days) heat (32 degrees C) exposure on hepatic leptin and muscle UCP gene expression in 5-wk-old broiler chickens. Both cold- and heat-exposure significantly (P < 0.05 to P < 0.001) upregulated hepatic leptin (by 35 and 46%, respectively) and muscle UCP mRNA levels (by 71 and 71%, respectively) compared to the thermoneutrality (22 degrees C). This result suggests that leptin and UCP may be involved in the thermoregulation response of chickens to extreme climate (cold and hot temperatures). The upregulation of hepatic leptin gene expression was accompanied by an increase in plasma leptin levels, indicating that leptin may be regulated at transcriptional level. The increase of leptin and UCP mRNA abundance, and leptinemia we report here were not related to plasma glucose or insulin levels. In conclusion, the exposure of broiler chickens to extreme ambient temperatures (cold and heat) increases hepatic leptin and muscle UCP gene expression.

National audience; Important changes in the ways of consumption of poultry (towards more cuts or elaborated products to the detriment of whole carcasses) have highlighted the importance of meat quality. Besides technological adaptations, research has been developed in order to improve the quality of the meat upstream, in particular by genetics. Additional approaches of positional (QTL research) and expressional (microarrays for gene expression profiling) genomics were intended to identify the genes involved in the quality of chicken meat. These studies take advantage of animal models, either experimental or commercial lines, which exhibit great differences in growth and meat quality. First original results were obtained for the chromosomal regions (or QTL) involved in the variation of post mortem pH fall, colour and drip loss in the breast meat of the chicken. Further studies are now aimed at identifying the underlying polymorphisms, or closely linked genetic markers, that could be used for selection. This future research will benefit from the progress made in genotyping technologies (with the access to high-density SNP markers in the chicken) as well as from the information revealed by microarray analyses on the genes and metabolic pathways contributing to chicken meat quality.; Les évolutions importantes des modes de consommation des volailles (vers davantage de produits découpés ou élaborés au détriment des carcasses entières) ont renforcé l’importance de la qualité de la viande. Il existe aujourd’hui une forte variabilité de cette qualité, qu’il convient de mieux maîtriser pour contrôler les caractéristiques finales du produit. A côté des adaptations technologiques développées par les industriels, des recherches ont été initiées pour améliorer en amont la qualité de la viande, notamment par la voie génétique. Des approches complémentaires de génomique positionnelle (recherche de QTL) et expressionnelle (étude du transcriptome musculaire) ont été initiées pour identifier les gènes impliqués dans les variations de la qualité chez le Poulet. Ces études s’appuient sur des modèles animaux, lignées expérimentales ou commerciales, qui présentent de fortes différences de croissance et de qualité. Des premiers résultats originaux ont été obtenus sur les régions chromosomiques (ou QTL) contrôlant la cinétique de chute du pH post mortem, la couleur ou les pertes en eau par exsudation du filet de poulet. Les perspectives visent maintenant à identifier les polymorphismes sous-jacents, ou à défaut des marqueurs génétiques proches, pour une utilisation en sélection. Ces futures recherches bénéficieront des progrès importants réalisés dans les technologies de typage (avec l’accès chez le Poulet aux marqueurs haut débit de type SNP), mais aussi de la mise en évidence par les études du transcriptome des gènes et voies métaboliques contribuant à l’élaboration de la qualité.

The objective of this experiment was to study two feeding methods, which could potentially act on the gut microflora, the structure and/or the function of the digestive tract and thereby improve the performance of broilers. 2. Four dietary treatments were studied: a negative control (wheat based) with no additives (C), a positive control with 0.01 g/kg avilamycin (AV), a treatment with 0.6 g/kg fructo-oligosaccharides (FOS) and a treatment with the same composition as treatment C but in which a part or all (400 g/kg) of the wheat was given as whole wheat and a concentrate supplement (WW). The measurements were: the performance from 0 to 6 weeks, the bacterial counts at 3 and 6 weeks, the digestive tract morphology and the activity of some intestinal enzymes at 3 weeks. 3. The birds given AV had better daily live weight gain (DLWG) and feed conversion ratio (FCR) compared to treatment C. The birds given FOS had a lower feed intake and a lower DLWG than the birds on treatment C but their FCR was significantly improved. WW resulted in a numerically lower feed intake and a significantly lower DLWG than treatment C. With AV, the number of aerobic mesophilic bacteria in the caeca was reduced at 3 weeks. With WW, gizzard and pancreas weights were greater and the surfaces of the ileal crypts were larger. An increased activity of leucine aminopeptidase (LAP) in the duodenum was found for treatments AV, FOS and WW. 4. In conclusion, in this study, treatments WW and FOS reduced DLWG, which may have been due to a lower feed intake during the whole period. With WW, the FCR was not affected, maybe because of both positive and negative effects on digestive tract (higher gizzard and pancreas development and LAP activity; larger crypts). However, FOS improved FCR, which may be partly explained by the higher LAP activity.

International audience; The objective of the current study was to investigate the impact of including whole wheat in broiler diets on the development of the digestive tract. Chickens were fed a standard feed containing 400 g ground wheat/kg or the same diet with a part of the wheat given separately as whole grains that increased progressively from 200 g/kg at 8 d to 400 g/kg at 22 d. Every week, from 16 to 44 d, growth performance, modifications of the size of the digestive tract organs and intestinal enzyme activities were investigated. Morphology of villi and crypts in the small intestinal segments (duodenum, jejunum, ileum) were analyzed at 23 and 44 d. Microbacterial counts were performed in jejunal, Real and caecal contents weekly from 16 to 44 d. During the adaptation period from 8 to 15 d, the birds fed the whole wheat diet had lower feed intake and lower weight gain. Thereafter, they showed improved growth performance so that by the end of the experiment, they had higher body weight compared to the standard-fed birds, 2430 +/- 29 versus 2331 +/- 36 g. Higher relative weights of gizzard (+26%) and pancreas (+12%) were observed from 16 to 44 d for whole wheat-fed birds compared to standard-fed birds. No differences in relative size of the different intestinal segments were observed, except that the jejunum was shorter. Increased villus to crypt length and surface ratios were observed at 23 d in the duodenum of whole wheat-fed birds, with no differences in morphometry between groups thereafter. Alkaline phosphatase activity was higher from 16 to 44 d in the duodenum and jejunum of whole wheat-fed birds. However, the activities of the digestive enzymes, leucine aminopeptidase and maltase, were similar between the two diets in the measured intestinal segments. A lower number of facultative anaerobic bacteria was found in the ileum of the whole wheat-fed birds, with no differences between treatments for Escherichia coli and for Lactobacillus counts. In the jejunum and the caeca, no differences in microflora counts were observed. The present results showed that feeding whole grains to broilers led mainly to modifications in the upper part of the digestive tract (gizzard, pancreas) and had little influence on the small and large intestine. International audience; Amino acids regulate protein synthesis and breakdown (i.e., protein turnover) and consequently protein deposition, which corresponds to the balance between the two processes. Elucidating the mechanisms involved in such regulation is important from fundamental and applied points of view since it can provide a basis to optimize amino acid requirements and to control protein mass, body composition and so forth. Amino acids, which have long been considered simply as precursors of protein synthesis, are now recognized to exert other significant influences; that is, they are precursors of essential molecules, act as mediators or signal molecules and affect numerous functions. For example, amino acids act as mediators of metabolic pathways in the same manner as certain hormones. Thus, they modulate the activity of intracellular protein kinases involved in the regulation of metabolic pathways such as mRNA translation. We provide here an overview of the roles of amino acids as regulators of protein metabolism, by focusing particularly on sulfur amino acids. The potential importance of methionine as a "nutrient signal" is discussed in the light of recent findings. Emphasis is also placed on mechanisms controlling oxidative status since sulfur amino acids are involved in the synthesis of intracellular antioxidants (glutathione, taurine etc.) and in the methionine sulfoxide reductase antioxidant system. International audience; The duck growth hormone encoding gene and its promoter region were amplified by polymerase chain reaction (PCR). A total of 5.25 kb were cloned and sequenced. Duck growth hormone (GH) consists of five exons and four introns and is structurally similar to mammalian and chicken GH gene. Although the distal region of duck GH promoter showed no similarity to chicken and turkey promoters, the proximal region of the promoter contained two putative Pit-1 binding sequences, and showed similarity to chicken and turkey GH promoters. Genetic variation was detected at five positions of the promoter region. The results of this study indicate that the expression of duck GH is likely regulated in a similar manner to that of chicken GH via enhancer-type cis-acting elements and the presence of genetic variation in the duck GH gene may be applicable to marker-assisted selection.

La majorité des cas de salmonellose humaine dans les pays industrialisés est liée à la consommation d’oeufs et de viande de volaille mais aussi de charcuteries contaminées. Pour réduire ce risque alimentaire, la stratégie développée à l’INRA, en collaboration avec l’AFSSA, vise principalement à réduire la contamination des matières premières alimentaires d’origine animale, en particulier celles issues des volailles et du porc. Cela implique d’améliorer l’état sanitaire des animaux et de lutter contre le portage asymptomatique de cette bactérie par des animaux qui abritent, voire excrètent de grande quantité de pathogènes sans signe de maladie. Une telle démarche implique une meilleure compréhension des mécanismes qui permettent à la bactérie de coloniser l’animal, mais aussi des réponses immunitaires mises en place par l’hôte pour résister à ce pathogène. Ces études débouchent sur le développement de vaccins ou de moyens thérapeutiques et sur la sélection d’animaux plus résistants à ce portage asymptomatique. Enfin, une approche intégrative complémentaire des études expérimentales et observationnelles de terrain permet de modéliser la contamination des animaux ou des industries agroalimentaires pour analyser les risques de transmission et l’impact des mesures prophylactiques., Most human cases of salmonellosis in developed countries are related to the consumption of contaminated eggs, poultry and pig meat. To improve food safety, INRA aims, in collaboration with AFSSA, at decreasing the prevalence of contamination of raw animal material, especially those originating from poultry or pigs. This implies improving animal health and reducing asymptomatic carrier-state (Le animals carrying or shedding bacteria without any clinical signs). This involves increasing the knowledge of mechanisms of animal colonisation by bacteria and also the immune response allowing the host to resist the pathogen. These studies will result in the development of vaccines and drugs but also in the selection of animals more resistant to the Salmonella carrier state. Finally, an integrative approach, using results from experiments and field studies, allows modelising the contamination of animals and food industries and in turn evaluating the risk of transmission and the impact of prophylactic means.

International audience; IGF-1 plays a key role in the proliferation and differentiation of granulosa cells. However, the molecular mechanism of IGF-1 action in avian granulosa cells during follicle maturation is unclear. Here, we first studied IGF-1 receptor (IGF-IR) expression, IGF-1-induced progesterone production and some IGF-IR signaling pathways in granulosa cells from different follicles. IGF-IR (mRNA and protein) was higher in fresh or cultured granulosa cells from the largest follicles (171 or 172) than in those from smaller follicles (173 or 174). In vitro, IGF-1 treatment (110(-8) M, 36 h) increased progesterone secretion by four-fold in mixed F3 and F4 (F3/4) granulosa cells and by 1.5-fold in F1 granulosa cells. IGF- 1 (10(-8) M, 30 min)-induced increases in tyrosine phosphorylation of IGF-IR beta subunit and phosphorylation of ERK were higher in 171 than in F3/4 granulosa cells. Interestingly, IGF-1 stimulation (10-8 M, 10 min) decreased the level of AMPK Thr172 phosphorylation in F1 and 173/4 granulosa cells. We have recently showed that AMPK (AMP-activated protein kinase) is a protein kinase involved in the steroidogenesis in chicken granulosa cells. We then studied the effects of AMPK activation by AICAR (5-aminoimidazole-4-caiboxamide ribonucleoside), an activator of AMPK, on IGF-1-induced progesterone secretion by 173/4 and F1 granulosa cells. AICAR treatment (1 mM, 36 h) increased IGF-1-induced progesterone, secretion, StAR protein levels and decreased ERK phosphorylation in F1 granulosa cells. Opposite data were observed in 173/4 granulosa cells. Adenovirus-mediated expression of dominant negative AMPK totally reversed the effects of AICAR on IGF-1-induced progesterone secretion, StAR protein production and ERK phosphorylation in both 173/4 and F1 granulosa cells. Thus, a variation of energy metabolism through AMPK activation could modulate differently IGF-1-induced progesterone production in F1 and 173/4 granulosa cells. (c) 2007 Elsevier Inc. Tonic immobility (TI) is an unlearned fear response induced by a brief physical restraint and characterized by a marked autonomic nervous system involvement. This experiment aimed at studying the relative involvement of both autonomic sub-systems, the sympathetic and parasympathetic nervous systems, during TI, by analyzing Heart Rate Variability. Quail selected genetically for long (LTI) or short (STI) TI duration and quail from a control line (CTI) were used. The animals were surgically fitted with a telemetric device to record electrocardiograms before and during a TI test. Heart rate did not differ between lines at rest. The induction of TI, whether effective or not, induced an increase in HR characterized by a shift of the sympathovagal balance towards a higher sympathetic dominance. Parasympathetic activity was lower during effective than during non-effective inductions in CTI quail. During TI, the increase in sympathetic dominance was initially maintained and then declined, while relative parasympathetic activity remained low, especially in CTI and STI lines. The end of tonic immobility was characterized by a rise in overall autonomic activity in all lines and an increase in parasympathetic influence in CTI and STI quail. To conclude, the susceptibility to TI cannot be explained only by autonomic reflex changes. It is probably strongly related to the perception of the test by the quail. During TI, the differences between lines in autonomic responses probably reflect behavioural differences in the fear response.

International audience; Short-term feed preferences were studied in individually caged chickens fed sequentially in order to understand a previously described imbalance in the intake of diets offered. Sequential feeding (SF) was carried out for four 48 h cycles in male broiler chickens. The diets varied in energy (2800 (E-) and 3200 kcal/kg (E+)) and protein (230 (P+) and 150 g/kg (P-)) contents. SF was compared to standard feeding (C) (3000 kcal/kg ME and CP = 190 g/kg). In experiment 1, three treatments were used., C, SE (E- followed by E+) and SE, (E+ followed by E-). Four treatments were used in experiment 2: C, S-P (P+ followed by P-), S-E and S-EP (P+E- followed by P-E+). Total feed intake was measured during the SF period. After this, short-term preferences were evaluated with a choice test on chickens previously fed with the same feeds during the SF period (experienced birds) and in C chickens (naive birds). In both experiments, total feed intake was similar among treatments and the percentage of each feed consumed was not significantly different from controls (50%). In experiment 1, S-E and S-E' chickens over-consumed E+ and under-consumed E- diets only during the first 15 min of the fourth cycle. The choice test indicated that experienced chickens preferred E+, while naive chickens preferred E-. Similarly, in experiment 2, chickens over-consumed E+ and E+P- during the first 15 min of the fourth cycle, but the intake of diets varying in protein content was not different from controls. During the choice test, as in experiment 1, experienced chickens preferred E+, while naive chickens preferred E-. There was a slight preference for the protein-poor diet in naive birds and there was no preference in the diet varying in both protein and energy contents. Experience modified choice between feeds varying in energy content but not in protein. When feeds were known, preference for energy affected the feed intake immediately after switching from one diet to the other although lower with the diet also varying in protein, it did not influence the total intake of each diet. Interactions between the nutritional properties and sensorial cues of feed could explain these results.

Higher corticosterone (CORT) responses to acute stress have previously been reported in quail selected for short (STI) duration of tonic immobility (TI) than for long TI (LTI), although behavioral studies indicated that LTI quail were more fearful. To investigate adrenal and pituitary function in these quail lines and their possible involvement in the differences in hypothalamic-pituitary-adrenal (HPA) axis reactivity, we measured CORT responses to adrenocorticotropin (1-24 ACTH), corticotropin-releasing factor (CRF), and arginine vasotocin (AVT) after characterizing the nucleotide acid sequences of these peptides in quail. Although maximum adrenal responses, assessed by ACTH challenge, were higher in STI quail, adrenal sensitivity was comparable for the two genotypes. It is therefore unlikely that differences in HPA axis reactivity involved the adrenal level. AVT and ACTH induced comparable CORT responses in both genotypes, whereas those induced by CRF were much lower. AVT is thus more potent than CRF in quail, but the respective maximum pituitary capacity of both genotypes to secrete ACTH was similar, and it is doubtful that the AVT pathway is involved in the difference in HPA axis reactivity between genotypes. On the other hand, the higher CORT responses induced by CRF in STI quail suggest that CRF might be involved in the differences in HPA axis reactivity between LTI and STI genotypes.

Since feed conversion ratio (FCR) is higher in slow-growing "Label Rouge" chickens than in broiler chickens, it is important to work on its improvement in this breed. However, this involves rearing animals in cages (C), an environment very different from that used for selection (in floor pens, S) and production (outdoor, E). The aim of this study was to evaluate the importance of genotype by environment (G × E) interactions between S, C, and E environments, to find the best way to select for FCR, using 2002 related animals. Growth curve parameters were estimated and body composition measured. Individual feed conversion ratios (FCR) were recorded between 8 and 10 weeks in C. The presence of G × E interactions was assessed by the genetic correlations between the same trait recorded in different environments. Moderate but significant G × E interactions were detected for carcass traits, a significant one was observed between E and S or C for growth curve parameters but none between C and S. If G × E interactions are set aside, i.e. selecting on traits recorded in C, abdominal fatness is the best indirect selection criterion for FCR but if they are taken in account then leg yield or growth curve parameters in S and growth curve parameters in E are better.

The avian eggshell is a highly ordered biomineral composed mainly of calcium carbonate associated with an organic matrix composed of proteins, glycoproteins and proteoglycans. This structure provides the developing embryo with protection from physical damage and microbial invasion. Ovocalyxin-32 (OCX-32) is a 32 kDa eggshell-specific matrix protein which has been cloned and demonstrates 30% identity with the mammalian carboxypeptidase inhibitor, latexin. In order to further study its function, recombinant OCX-32 protein was expressed in E. coli. The protein was extracted from inclusion bodies and purified by sequential DEAE Sepharose and Ni2+ metal ion affinity chromatographies as a 58 kDa GST-fusion protein. The refolded GST-OCX-32 significantly inhibited bovine carboxypeptidase and also inhibited the growth of Bacillus subtilis. The results suggest that OCX-32 may show similar activity to the fusion protein and reinforce the antimicrobial properties of the eggshell by providing protection to the developing avian embryo. OCX-32 is the first example of an eggshell specific protein to be successfully cloned and expressed in a prokaryotic system. The association of an antimicrobial protease inhibitor with the outer eggshell and cuticle of the table egg may enhance the food safety of this product.

Animal protection is now included in French and European legislation. It meets a major concern of western societies that stems from the acknowledgement that animals used by humans are sentient beings that should be guaranteed a minimum protection. The way animals perceive their environment determines their well-being: it will be good if the animal perceives that the environment meets its needs, while it will be low for an animal that perceives its environment as threatening or unpleasant. To assess the welfare state of an animal, many behavioural, physiological and biological indices need to be taken into account. The living conditions of farm animals are often chosen according to production objectives and may not always ensure a high level of animal welfare. At the same time, a low welfare state can have adverse effects on animal productivity. Hence remedial solutions that meet both production and welfare requirements should be developed. This special issue aims to refine the concept of animal well-being and will present a review of results of the research undertaken by the members of the AGRIBEA network. The findings can be used for drawing up regulations and advising farmers., La protection des animaux est inscrite dans la réglementation française et européenne. Elle répond à une attente sociétale majeure dans nos pays occidentaux, attente qui part du principe que les animaux que nous utilisons sont des êtres vivants sensibles à qui l’homme doit assurer un minimum de bien-être. La façon dont un animal appréhende son environnement va influencer son état de bien-être et ce, positivement si l’environnement satisfait ses besoins et ses motivations, mais négativement si l’environnement est perçu comme menaçant, désagréable. Le niveau de bien-être d’un animal est apprécié au travers de nombreuses observations portant sur son comportement, l’état d’activation des systèmes physiologiques, et son état général. Les conditions d’élevage, généralement choisies en fonction d’objectifs de production, peuvent également influencer le bien-être d’un animal. En retour, un bien-être dégradé peut entraîner une moindre productivité. Aussi des solutions d’élevage permettant de concilier production et bien-être animal doivent-elles être mises au point. Ce numéro spécial vise à mieux cerner la notion de bien-être animal et rassemble les principaux travaux entrepris par les membres du réseau Agri Bien-être Animal dans les différentes productions. Les résultats des recherches en bien-être animal sont utilisés lors de l’élaboration de réglementations. Ils doivent également servir au conseil en élevage.

La multiplicité des critères susceptibles d’être utilisés pour évaluer l’adaptation comportementale d’un animal à son environnement a motivé l’étude des relations entre réponses physiologiques et comportementales, dans l’espoir que l’identification de telles relations puisse simplifier l’évaluation du bien-être animal. La comparaison d’animaux placés dans des environnements différents et la comparaison d’animaux aux génotypes extrêmes suggèrent l’existence de relations entre certaines réponses physiologiques et comportementales. La possibilité de catégoriser certains animaux selon leur stratégie de réponse adaptative, caractérisée par des associations répétables entre réponses comportementales et physiologiques, conforte cette hypothèse. Cependant, les relations ainsi suggérées semblent difficilement généralisables : il n’existe pas de relation univoque entre les réponses physiologiques et comportementales d’un animal face à une situation perçue comme menaçante. En l’état actuel des connaissances, la prise en compte de manière concomitante de critères physiologiques et comportementaux est essentielle pour apprécier le bien-être d’un animal. A l’avenir, des investigations en génomique fonctionnelle et en neurobiologie pourraient améliorer notre compréhension de ces relations., Assessing the behavioural adaptation of an animal to its environment is complex, notably because numerous criteria can be taken into consideration. A better understanding of the relationships between criteria, particularly between behavioural and physiological responses, might help reduce the number of parameters required to assess animal welfare. The existence of relationships between behavioural and physiological responses of animals to potentially threatening situations has been suggested both by studies comparing animals reared in different environments and by those comparing animals with extreme genotypes. Moreover, the identification of coping styles in various species has strengthened the idea that physiological and behavioural responses could be related, although the precise laws governing such relationships are still difficult to establish and generalise. Thus, considering the complexity of these relationships, it appears reasonable to consider these two groups of responses as partially independent and as giving complementary information about animal welfare. In the future, the development of complex trait analyses as well as investigation at the level of the brain should improve understanding of the relationships between physiological and behavioural responses.

The need for semen preservation in domestic birds is a result of the reduction in genetic variability of domestic bird livestock and of the increasing risk of line extinction for health and safety reasons. Cryopreservation of embryos and primordial germ cells (PGC) is not routinely feasible in birds. The project therefore involved semen frozen in optimal safety and traceable conditions. Whole blood samples were also frozen to provide samples of analyses of genomes and health status. The feasibility of using ex situ conservation, i.e., collecting biological material to be stored outside the usual production area of the species (ex situ genetic stock), to preserve and manage rare breeds was tested with 4 subfertile populations: 3 rare experimental lines used for research into energy metabolism (R+), growth (Y33), and immunity (B4/B4), reared under known health status and the oldest endangered patrimonial French breed, the Gauloise doree with an unknown health status. A general infrastructure was set up for the health screening and remediation of diseases, collection and storage of frozen cells and 2 sites were created for the storage of frozen samples. The screening and remediation of diseases of the Gauloise doree, which was contaminated with various Salmonella and Mycoplasma strains, was achieved by successive treatment of parents, incubated eggs and young chicks with Baytril followed by Tiamulin. For each line, 474 to 994 semen straws have been frozen, thawed, and the semen evaluated. Insemination of frozen-thawed semen into females of the same genetic origin or of an egg-type commercial breed produced chicks in every case. For the most subfertile lines, insemination with egg-type females significantly increased the reproductive success. In conclusion, we report on the benefits of a semen and blood cryobanking complex for the management of endangered lines and strains of domestic birds. Current stocks made possible the restoration of more than 96% of the initial genome. This project also provided technical solutions to resolve some of the health problems frequently encountered for gene preservation in poultry.
