Conductivity detection is a universal detection technique often encountered in electrophoretic separation systems, especially in modern chip-electrophoresis based devices. On the other hand, it is sparsely combined with another contemporary trend of enhancing limits of detection by means of various preconcentration strategies. This can be attributed to the fact that a preconcentration experimental setup usually brings about disturbances in a conductivity baseline. Sweeping with a neutral sweeping agent seems a good candidate for overcoming this problem. A neutral sweeping agent does not hinder the conductivity detection while a charged analyte may preconcentrate on its boundary due to a decrease in its effective mobility. This study investigates such sweeping systems theoretically, by means of computer simulations, and experimentally. A formula is provided for the reliable estimation of the preconcentration factor. Additionally, it is demonstrated that the conductivity signal can significantly benefit from slowing down the analyte and thus the overall signal enhancement can easily overweight amplification caused solely by the sweeping process. The overall enhancement factor can be deduced a priori from the linearized theory of electrophoresis implemented in the PeakMaster freeware. Sweeping by neutral cyclodextrin is demonstrated on an amplification of a conductivity signal of flurbiprofen in a real drug sample. A new method based on micellar electrokinetic chromatography-tandem mass spectrometry (MEKC-MS/MS) has been developed for the identification and simultaneous quantification of thirteen benzimidazoles in animal urine. In order to obtain an appropriate separation with the highest sensitivity, different electrophoretic parameters were evaluated. Under optimum conditions, the separation was performed using ammonium perfluorooctanoate as volatile surfactant and electrophoretic buffer (50mM, pH 9). To increase the sensitivity, a stacking mode named sweeping was applied, using water as injection solvent at 50mbar for 75s, obtaining sensitivity enhancement factors from 50 to 181. The method was applied to different animal urine samples, including sheep, cow and goat. The sample treatment consisted of a 1:10 (v/v) dilution with water. Calibration using sheep urine samples can be used for both goat and cow urine samples with a relative bias below 25% and relative standard deviations lower than 8%. The limits of detection were below 70µgL -1 . As a result, the applicability of this rapid, simple, sensitive, and environmentally friendly method for therapeutic drug monitoring of benzimidazoles based on the analysis of animal urine has been demonstrated., (Copyright © 2017 Elsevier B.V. This review continues to update this series of biennial reviews, first published in Electrophoresis in 2007, on developments in the field of on-line/in-line concentration methods in capillaries and microchips, covering the period July 2014-June 2016. It includes developments in the field of stacking, covering all methods from field amplified sample stacking and large volume sample stacking, through to isotachophoresis, dynamic pH junction, and sweeping. A novel and simple method that combines an online concentration technique with an enantioseparation technique for capillary electrophoresis-namely, cation-selective exhaustive injection and sweeping cyclodextrin-modified micellar electrokinetic chromatography (CSEI-sweeping CD-modified MEKC)-realizes the effective enantioseparation of cationic analytes while keeping a significant increase of detection sensitivity. This technique consists of a slight modification of the basic CSEI-sweeping MEKC. The main idea is to simply add an anionic CD as a chiral selector into the micellar buffer including sodium dodecyl sulfate, but not to change any other buffers in order to preserve the online concentration mechanism. When applied to analysis of the street drug, methamphetamine, the method achieved not only a baseline enantioseparation but also limits of detection (LODs; S/N = 3) of 70-90 pg/mL (ppt) for each isomer. This translates to a more than 10 000-fold improvement compared to the LODs by the usual injection method. The present technique, which was made from a slight modification of CSEI-sweeping MEKC, would give an attractive approach that is applicable to almost any analytes for which CSEI-sweeping MEKC is applicable; all that is required is the selection of an appropriate anionic CD to be added to the micellar buffer., (© 2016 WILEY-VCH Verlag GmbH & Co. Simultaneous electrophoretic concentration and separation (SECS) was used as a simple and environmental friendly sample preparation strategy for herbicides in beer samples. An electric field was used to facilitate the separation and concentration of the analytes based on their charge from a 20 mL sample of diluted beer into two separate 20 μL aliquots of an acceptor electrolyte housed inside a micropipette. The anionic organophosphonate and cationic quaternary ammonium herbicides were concentrated in the anodic and cathodic pipette, respectively. Under optimized conditions, SECS was completed in 30 min at an applied voltage of 150 V, which provided analyte concentration factors of up to 90. After sample preparation, the SECS concentrate of cationic and anionic herbicides was analyzed by stacking CE with UV detection and also by LC-MS, respectively. The method detection limit for the diluted and undiluted sample was as low as 3 and 15 ng/mL, respectively. The method was linear over two orders of concentration with repeatability and intermediate precision of better than 5.8 and 7.0%RSD, respectively. A new method for determination of thiosulfate in human urine has been developed and validated. Analytical procedure is very simple and consists of only few steps: derivatization of thiosulfate with 2-chloro-1-methylquinolinium tetrafluoroborate, centrifugation of a mixture, separation of so-formed derivative by micellar electrokinetic chromatography with sweeping and UV detection at 375 nm. A fused-silica capillary with an inlet to detector length of 51.5 cm and a total length of 60 cm (75 μm id) was served as a separation column. The separation voltage of 20.5 kV (∼160 mA) and buffer solution consisting of 0.055 mol/L sodium phosphate (pH 8), 25% acetonitrile, and 0.035 mol/L sodium dodecyl sulfate were found to be the most suitable conditions for the effective separation. The limit of quantification for thiosulfate was 4 μmol/L urine. The method was validated and calibrated for thiosulfate in the range of 4-64 μmol/L (R(2) = 0.9997). A simple, rapid, and sensitive method using ultrasound-assisted dispersive liquid-liquid microextraction (UA-DLLME) combined with sweeping micellar electrokinetic chromatography (sweeping-MEKC) has been developed for the determination of nine organophosphorus pesticides (chlorfenvinphos, parathion, quinalphos, fenitrothion, azinphos-ethyl, parathion-methyl, fensulfothion, methidathion, and paraoxon). The important parameters that affect the UA-DLLME and sweeping efficiency were investigated. Under the optimized conditions, the proposed method provided 779.0-6203.5-fold enrichment of the nine pesticides compared to the normal MEKC method. The limits of detection ranged from 0.002 to 0.008 mg kg(-1). The relative standard deviations of the peak area ranged from 1.2 to 6.5%, indicating the good repeatability of the method. Finally, the developed UA-DLLME-sweeping-MEKC method has been successfully applied to the analysis of the investigated pesticides in several medicinal plants, including Lycium chinense, Dioscorea opposite, Codonopsis pilosula, and Panax ginseng, indicating that this method is suitable for the determination of trace pesticide residues in real samples with complex matrices.

A new method for the determination of hydrogen sulfide in hen tissues has been developed and validated. For estimation of hydrogen sulfide content, a sample (0.1 g) of hen tissue was treated according to the procedure consisted of some essential steps: simultaneous homogenization of a tissue and derivatization of hydrogen sulfide to its S-quinolinium derivative with 2-chloro-1-methylquinolinium tetrafluoroborate, separation of so-formed derivative by micellar electrokinetic chromatography with sweeping, and detection and quantitation with the use of UV detector set to measure analytical signals at 375 nm. Effective electrophoretic separation was achieved using fused silica capillary (effective length 41.5 cm, 75 μm id) and 0.05 mol/L, pH 8 phosphate buffer with the addition of 0.04 mol/L SDS and 26% ACN. The lower limit of quantification was 0.12 μmol hydrogen sulfide in 1 g of tissue. The calibration curve prepared in tissue homogenate for hydrogen sulfide showed linearity in the range from 0.15 to 2.0 μmol/g, with the coefficient of correlation 0.9978. Determination of tea catechins in human plasma might provide a means of better evaluation of their benefits. The main difficulty in their analysis is the low catechins concentrations in plasma and their susceptible to oxidation during sample pretreatment. In the current work, a sweeping-micellar electrokinetic chromatography (sweeping-MEKC) by long alkyl chain ionic liquid was investigated for the simultaneous determination of seven principal naturally-occurring tea catechins in human plasma under acidic conditions after the intake of green tea beverage. The effects of type and concentration of three 1-tetradecyl-3-methylimidazolium ionic liquids, namely bromide, acetate and hydrogen sulfate salts were studied. The seven catechins were successfully separated within 5min by micellar running buffer of 5mmolL(-1) 1-tetradecyl-3-methylimidazolium bromide and 15mmolL(-1) phosphate buffer at pH 4.5 under optimal parameters of 50mbar injection for 150s, 10kV, 25°C and 200nm. To prevent the possibility of IL adsorption, an appropriate rinsing protocol was established. The method has analytical ranges from 0.5, 1, 0.5, 1, 2, 1 and 1 to 500ngmL(-1) for GC, C, EC, EGCG, GCG, ECG and EGC, respectively (r ranged from 0.995 to 0.999). The intraday precision and accuracy were 0.1-0.9% RSD (n=10) and 97-106% recovery, respectively. Limits of detections of analytes were ranged from 0.2 to 1.2ngmL(-1). The current sweeping-MEKC achieved sensitivity enhancement factor (SEF) up to 183-fold of analytes concentrations compared to other hitherto published CE reports that is suitable to find out the trace amounts of catechins in plasma., (Copyright © 2014 Elsevier B.V. The current routes to couple dispersive liquid-liquid microextraction (DLLME) with capillary electrophoresis (CE) are evaporation of water immiscible extractants and backextraction of analytes. The former is not applicable to extractants with high boiling points, the latter being effective only for acidic or basic analytes, both of which limit the further application of DLLME-CE. In this study, with 1-octanol as a model DLLME extractant and six phenols as model analytes, a novel method based on acetonitrile stacking and sweeping is proposed to accomplish large-volume injection of 1-octanol diluted with a solvent-saline mixture before micellar electrokinetic chromatography. Brij-35 and β-cyclodextrin were employed as pseudostationary phases for sweeping and also for improving the compatibility of sample zone and aqueous running buffer. A short solvent-saline plug was used to offset the adverse effect of the water immiscible extractant on focusing efficiency. The key parameters affecting separation and concentration were systematically optimized; the effect of Brij-35 and 1-octanol on focusing mechanism was discussed. Under the optimized conditions, with ∼ 30-fold concentration enrichment by DLLME, the diluted extractant (8×) was then injected into the capillary with a length of 21 cm (42% of the total length), which yielded the overall improvements in sensitivity of 170-460. Limits of detection and qualification ranged from 0.2 to 1.0 ng/mL and 1.0 to 3.4 g/mL, respectively. Acceptable repeatability lower than 3.0% for migration time and 9.0% for peak areas were obtained. A simple method that consumes low organic solvent is proposed for the analysis of phthalic acid esters in Chinese white spirit using dispersive liquid-liquid microextraction coupled with sweeping-micellar electrokinetic chromatography. Tetrachloromethane and white-spirit-containing ethanol were used as the extraction and dispersing solvents, respectively. The electrophoresis separation buffer was composed of 5 mM β-cyclodextrin, 50 mM sodium dodecyl sulfate and 25 mM borate buffer (pH 9.2) with 9% acetonitrile, enabling the baseline resolution of the analytes within 13 min. Under the optimum conditions, satisfactory linearities (5-1000 ng/mL, r ≥ 0.9909), good reproducibility (RSD ≤ 6.7% for peak area, and RSD ≤ 2.8% for migration time), low detection limits (0.4-0.8 ng/mL) and acceptable recovery rates (89.6-105.7%) were obtained. The proposed method was successfully applied to 22 Chinese white spirits, and the content of dibutyl phthalate in 55% of the samples exceeded the Specific Migration Limit of 0.3 mg/kg established by the domestic and international regulations., (© 2014 WILEY-VCH Verlag GmbH & Co. Methotrexate (MTX) is widely used for the treatment of many types of cancer. Folinic acid (FNA) and folic acid (FA) were usually simultaneously supplemented with MTX to reduce the side effects of a folate deficiency. This study, for the first time, included on-line sample preconcentration by stacking and sweeping techniques under reduced or enhanced electric conductivity in the sample region using short chain alkyl imidazolium ionic liquids (ILs) as micelle forming agents for analyte focusing. Both analyte focusing by micelle collapse (AFMC) and sweeping-MEKC had been investigated for the comparison of their effectiveness to examine simultaneously MTX, FNA and FA in plasma and urine under physiological conditions. In sweeping-MEKC, the sample solution without micelles was hydrodynamically injected as a long plug into a fused-silica capillary pre-filled with phosphate buffer containing 3.0 mol/L of 1-butyl-3-methylimidazolium bromide (BMIMBr). Using AFMC, the analytes were prepared in BMIMBr micellar matrix and hydrodynamically injected into the phosphate buffer without IL micelles. The conductivity ratio between BGE and sample (γ, BGE/sample) was optimized to be 3.0 in sweeping-MEKC and 0.33 in AFMC resulting the adequate separation of analytes within 4.0 min. To reduce the possibility of BMIMBr adsorption, an appropriate rinsing protocol was used. The limits of detection were calculated as 0.1 ng/mL MTX, 0.05 ng/mL FNA and 0.05 ng/mL FA by sweeping-MEKC and 0.5 ng/mL MTX, 0.3 ng/mL FNA and 0.3 ng/mL FA by AFMC. The accuracy was tested by recovery in plasma and urine matrices giving values ranging between 90 and 110%. Both stacking and sweeping by BMIMBr could be successfully used for the rapid, selective and sensitive determination of pharmaceuticals in complex matrices due to its fascinating properties, including high conductivity, good thermal stability and ability to form different types of interactions by electrostatic, hydrophobic, hydrogen bonding and π-π interactions. In sweeping-MEKC, the using of BMIMBr enhanced the γ factor, k retention factor and the injected amount of sample. A novel method consisting of cation-selective exhaustive injection and sweeping (CSEI-sweeping) as on-line preconcentration followed by a micellar electrokinetic chromatography (MEKC) separation has been developed for the determination of 5-nitroimidazoles (5-NDZ) in environmental waters. Moreover, dispersive liquid-liquid microextraction (DLLME) has been proposed for first time as sample treatment technique prior to CSEI-sweeping-MEKC. DLLME was applied to 5mL of sample. Dibromomethane (1156μL) and 2-butanol (1363μL) were employed as extractant and dispersive solvents, respectively. Salting-out effect was achieved by the addition of 16% (w/v) NaCl to the samples. After DLLME and organic solvent evaporation, the residue was redissolved in a low conductivity solvent (5mM phosphoric acid with 5% of methanol) and electrokinetically injected at 9.8kV for 632s in a bare fused-silica capillary (57.2cm, 50μm I.D.). Prior to the injection, the capillary was rinsed with 50mM phosphate buffer pH 2.5, followed by a plug of a higher conductivity buffer (100mM phosphate pH 2.5, 50mbar, 264s) and a plug of water (50mbar, 2s). Separation was carried out applying -30kV at 20°C in 44mM phosphate buffer pH 2.5, containing 8% tetrahydrofuran and 123mM sodium dodecyl sulfate. Analytical signals were monitored at 276nm. Validation was performed in river and well waters, obtaining satisfactory results in terms of linearity, precision (% RSD generally lower than 10%) and trueness (recoveries higher than 70% in almost all cases). LODs ranged from 0.61 to 2.44ng/mL. The combination of this microextraction technique with the proposed capillary electrophoresis methodology supposes a simple, sensitive and cheap alternative for 5-NDZ analyses, in accordance with the aims of green chemistry., (Copyright © 2014 Elsevier B.V. This review highlights recent developments and applications of on-line sample preconcentration techniques in capillary electrophoresis (CE) from 2010 to April 2013. Various preconcentration techniques based on the analyte velocity change in two or three discontinuous solutions system including field-amplified stacking, transient isotachophoresis, pH-mediated stacking, sweeping, and their modified and combined techniques have been employed to enrich and separate biological, environmental, food, toxicological, forensic and nanoparticle samples in CE. More than 170 published research articles collected from Scopus databases from the year 2010 described the on-line sample preconcentration techniques. An on-line sample preconcentration method by two-step stacking i.e., sweeping and micelle to solvent stacking, in capillary zone electrophoresis (CZE) has been developed for the determination of strychnine and brucine in traditional Chinese herbal medicines. After experimental optimizations, the best separation was achieved by using 75 mM phosphate buffer (pH 2.5) with 30% methanol (v/v). Compared with normal CZE injection, 51- and 38-fold improvement in concentration sensitivity was achieved for strychnine and brucine, respectively. The calibration curve was linear in the range of 0.1-5.0 μg mL(-1) for both strychnine and brucine, with the correlation coefficients of 0.9998 and 0.9997, respectively. The limits of detection (S/N=3) for both alkaloids were 0.01 μg mL(-1). The inter-day (n=8) and intra-day (n=5) reproducibilities expressed as the relative standard deviations for corrected peak area were less than 9.5%. The method was applied to determine strychnine and brucine in two Chinese herbal medicines, with recoveries ranging from 94.2% to 105.4%. Sweeping is an enrichment method in MEKC, which includes following steps: stacking/destacking of the micelles, sweeping of analyte by the stacked/destacked micelles, destacking/stacking of the swept analyte zone and additional focusing/defocusing due to the retention factor gradient effect (RFGE). In this study, we investigate additional processes, regarding online focusing in cyclodextrin-modified MEKC (CD-MEKC) of hydrophobic basic analytes: dynamic pH junction (sample with pH different from that of BGE) and adsorption of analyte onto the capillary wall within the sample zone. It is demonstrated that the developed method for the assessment of the sweeping efficiency is also applicable to CD-MEKC taking ethylparaben as an example of acidic analytes and desloratadine as an example of basic analytes using different types of β-cyclodextrin. Our previous results regarding RFGE as an additional focusing/defocusing effect in sweeping-MEKC are confirmed for the case that the apparent distribution coefficient differs for the sample and the BGE due to a different content of the complex-forming agent cyclodextrin and due to a pH difference between the sample and the BGE. Despite being significantly more hydrophobic than ethylparaben, desloratadine shows an unexpectedly low enrichment factor. This enrichment factor is nearly unaffected by the addition of CD to the BGE. This unexpected behavior is attributed to wall adsorption of the protonated hydrophobic basic analyte within the sample zone, which significantly counteracts the sweeping process. A new method was developed for the determination of eight triazine herbicide residues in cereal and vegetable samples by on-line sweeping technique in micellar electrokinetic capillary chromatography (MEKC). Some factors affecting analyte enrichment and separation efficiency were examined. The optimum buffer was composed of 25 mM borate, 15 mM phosphate, 40 mM sodium dodecylsulfate (SDS) and 3% (v/v) of 1-propanol at pH 6.5. The separation voltage was 20 kV and the sample was injected at 0.5 psi for 240 s. The detection wavelength was set at 220 nm with the capillary temperature being at 25 °C. Under the optimized conditions, the enrichment factors were achieved from 479 to 610. The limits of detection (LODs, S/N = 3) ranged from 0.02 to 0.04 ng/g and the limits of quantification (LOQs) of eight triazine herbicides were all 0.1 ng/g. The average recoveries of spiked samples were 82.8-96.8%. This study reports a sweeping and micelle to solvent stacking (MSS) method for fast stacking of nitroimidazoles in capillary zone electrophoresis (CZE). The optimal experimental conditions are run buffer of 25mM sodium phosphate (pH 1.5) and 0.50% (v/v) methanol, micellar solution of 30mM sodium dodecyl sulfate, applied voltage of 28kV, and sample and micellar solution injection of 100s. By applying this new on-line preconcentration technique, the nitroimidazoles content can be determined within 9min with the limit of detection (S/N=3) ranging 2.3-3.0ng/mL, which is lower than that of conventional CZE analysis. The proposed MSS technique affords 20-, 12- and 38-fold improvements in sensitivity for the detection of dimetridazole, metronidazole and secnidazole, respectively. The relative standard deviations (RSDs) of intra-day and inter-day are 2.1-3.6% and 2.7-4.6% (n=6), respectively. The recoveries in pretreated rabbit plasma at spiked levels of 5.0-10.0μg/mL are 92.0-101.1% with RSDs lower than 3.4%. The analysis of hydrophobic basic analytes by micellar electrokinetic chromatography (MEKC) is usually challenging because of the tendency of these analytes to be adsorbed onto the inner capillary wall in addition to the difficulty to separate these compounds as they exhibit extremely high retention factors. A robust and reliable method for the simultaneous determination of loratadine (LOR) and its major metabolite desloratadine (DSL) is developed based on cyclodextrin-modified micellar electrokinetic chromatography (CD-MEKC) with acidic sample matrix and basic background electrolyte (BGE). The influence of the sample matrix on the reachable focusing efficiency is studied. It is shown that the application of a low pH sample solution mitigates problems associated with the low solubility of the hydrophobic basic analytes in aqueous solution while having advantages with regard to on-line focusing. Moreover, the use of a basic BGE reduces the adsorption of these analytes in the separation compartment. The separation of the studied analytes is achieved in less than 7min using a BGE consisting of 10mmolL(-1) disodium tetraborate buffer, pH 9.30 containing 40mmolL(-1) SDS and 20mmolL(-1) hydroxypropyl-β-CD while the sample solution is composed of 10mmolL(-1) phosphoric acid, pH 2.15. A full validation study of the developed method based on the pharmacopeial guidelines is performed. The method is successfully applied to the analysis of the studied drugs in tablets without interference of tablet additives as well as the analysis of spiked human urine without any sample pretreatment. Furthermore, DSL can be detected as an impurity in LOR bulk powder at the stated pharmacopeial limit (0.1%, w/w). The selectivity of the developed method allows the analysis of LOR and DSL in combination with the co-formulated drug pseudoephedrine. It is shown that in CD-MEKC with basic BGE, solute-wall interactions are effectively suppressed allowing the development of efficient and precise methods for the determination of hydrophobic basic analytes, whereas the use of a low pH sample solution has a positive impact on the attainable sweeping efficiency without compromising peak shape and resolution., (Copyright © 2013 Elsevier B.V.
