(J Autism Dev Disord 39: 693-705, 2009), was investigated in an independent sample of 1248 Dutch children with 1455 ADOS administrations (modules 1, 2 and 3). The greater comparability between ADOS administrations at different times, ages and in different modules, as reached by Gotham et al. with the calibrated severity measures, seems to be corroborated by the current study for module 1 and to a lesser extent for module 3. For module 2, the calibrated severity scores need to be further investigated within a sample that resembles Gotham's sample in age and level of verbal functioning.

The genomes of four Lactobacillus reuteri strains isolated from human breast milk and the gastrointestinal tract have been recently sequenced as part of the Human Microbiome Project. Preliminary genome comparisons suggested that these strains belong to two different clades, previously shown to differ with respect to antimicrobial production, biofilm formation, and immunomodulation. To explain possible mechanisms of survival in the host and probiosis, we completed a detailed genomic comparison of two breast milk-derived isolates representative of each group: an established probiotic strain (L. reuteri ATCC 55730) and a strain with promising probiotic features (L. reuteri ATCC PTA 6475). Transcriptomes of L. reuteri strains in different growth phases were monitored using strain-specific microarrays, and compared using a pan-metabolic model representing all known metabolic reactions present in these strains. Both strains contained candidate genes involved in the survival and persistence in the gut such as mucus-binding proteins and enzymes scavenging reactive oxygen species. A large operon predicted to encode the synthesis of an exopolysaccharide was identified in strain 55730. This section, multilevel policies, reviews the impact that has been and can be made by health policy changes at multiple levels, strategies and resources for increasing adherence to population prevention recommendations, and how changes at the microlevel and macrolevel of the environment can provide opportunities and rewards for healthy behaviors and disincentives for unhealthy behaviors. Policies that support primary prevention of cardiovascular disease require the participation of numerous stakeholders at multiple levels, such as governmental and regulatory agencies. Such policy changes support a healthy lifestyle, as in designated smoke-free areas; laws that mandate that food purveyors reduce sodium and fat content or, eliminate trans-fats; and availability of safe parks and bike and walking trails; and also provide a supportive environment that in turn reinforces adherence to primary prevention. Health-related policies have a major impact at the societal level in both developed and developing countries; thus, it is important to understand the role that policy plays in promoting a healthier lifestyle and the prevention of cardiovascular disease. This section discusses how health policies can impact primary prevention and adherence to healthful recommendations, with examples focused on physical activity and diet. Deze studie bevat onmisbare gegevens voor wie geïnteresseerd is in de toestand van de jeugd in Nederland.

The training focused on stimulating joint attention and language skills and was based on the intervention described by Drew et al. (Eur Child Adolesc Psychiatr 11:266-272, 2002). Seventy-five toddlers with autism spectrum disorder (65 autism, 10 PDD-NOS, mean age = 34.4 months, SD = 6.2) were enrolled. Analyses were conducted on a final sample of 67 children (lost to follow-up = 8). No significant intervention effects were found for any of the primary (language), secondary (global clinical improvement), or mediating (child engagement, early precursors of social communication, or parental skills) outcome variables, suggesting that the 'Focus parent training' was not of additional value to the more general care-as-usual.

(2007) proposed revised algorithms for the Autism Diagnostic Observation Schedule (ADOS) with improved diagnostic validity. The aim of the current study was to replicate predictive validity, factor structure, and correlations with age and verbal and nonverbal IQ of the ADOS revised algorithms for Modules 1 and 2 in a large independent Dutch sample (N = 532). Results showed that the improvement of diagnostic validity was most apparent for autism, except in very young or low functioning children. Results for other autism spectrum disorders were less consistent. Overall, these findings support the use of the more homogeneous revised algorithms, with the use of similar items across developmental cells making it easier to compare ADOS scores within and between individuals.

The objective of this study was to evaluate Fourier transform infrared (FTIR) spectrometry to measure milk ketone bodies to detect hyperketonemic cows and compare this method with milk fat to protein ratio to detect hyperketonemia. Plasma and milk samples were obtained weekly from calving to wk 9 postpartum from 69 high-producing dairy cows. The reference test for hyperketonemia was defined as plasma concentration of ß-hydroxybutyrate (BHBA) =1,200 µmol/L. The weekly prevalence of hyperketonemia during the first 9 wk of lactation was, on average, 7.1%. Both BHBA and acetone in milk, determined by FTIR, had a higher sensitivity (80%) to detect hyperketonemia compared with milk fat to protein ratio (66%). Specificity was similar for the 3 diagnostic tests (71, 70, and 71%). In conclusion, FTIR predictions of BHBA or acetone in milk can detect cows with hyperketonemia in early lactation with a higher accuracy compared with the use of milk fat to protein ratio. Because of the high proportion of false-positive tests, there are concerns about the practical applicability of FTIR predictions of acetone, BHBA, and fat to protein ratio in milk to detect hyperketonemic cows.

Recent findings indicate that dextran sodium sulfate (DSS)-induced colitis is associated with a prothrombogenic phenotype, with P-selectin playing a major role in platelet recruitment. It has been suggested that probiotics may ameliorate colonic inflammation. We therefore investigated how treatment with Lactobacillus reuteri influenced P-selectin expression, leukocyte and platelet endothelial cell interactions, and colitis severity in DSS-treated rats. Rats were divided into the following four groups: nontreated, DSS treated (5% in drinking water for 9 days), L. reuteri, and L. reuteri and DSS treated. The rats were anesthetized with Inactin (120 mg/kg ip), and the dual radiolabeled monoclonal antibody technique was used to quantify P-selectin expression. Leukocyte-endothelial and platelet-endothelial cell interactions were studied in colonic venules with intravital microscopy. Colitis severity was assessed using a disease activity index. Disease activity index increased, as did the expression of P-selectin in the entire colon after DSS treatment, but both were reduced to control levels with L. reuteri pretreatment. The increased platelet- and leukocyte-endothelial cell interactions after DSS treatment were abolished by pretreatment with L. reuteri. L. reuteri protects against DSS-induced colitis in rats. The protection is associated with reduced P-selectin expression and a decrease in leukocyte- and platelet-endothelial cell interactions.

RIVM rapport:Voedingsstatusonderzoek is een belangrijk onderdeel van het nieuwe Nederlandse voedingspeilingssysteem en heeft als doel de voorziening van een bepaalde micronutriënt te beoordelen voor de Nederlandse populatie of relevante subgroepen daarbinnen. Uit de resultaten van de voedselconsumptiepeiling 2005/2006 bleek dat de inneming van vitamine D en foliumzuur onvoldoende was bij jonge kinderen in de leeftijd van twee tot zes jaar. Voor ijzer was de inneming laag ten opzichte van de Nederlandse norm en voor selenium, zink, vitamine E en retinol activiteit equivalenten (RAE) bleek de inneming laag ten opzichte van de Amerikaanse normen. Om na te gaan of er daadwerkelijk sprake is van tekorten werd voor deze micronutriënten voedingsstatusonderzoek aanbevolen. In het huidige onderzoek is bij vier- en achtjarige kinderen uit verschillende delen van Nederland de status bepaald van magnesium, zink, ijzer, vitamine D en selenium. De resultaten bevestigen het beeld dat de voorziening van magnesium adequaat is bij vier- en achtjarige kinderen. De signalen voor een mogelijk inadequate inneming van selenium en vitamine D worden door dit voedingsstatusonderzoek niet bevestigd. De resultaten met betrekking tot zink en ijzer kunnen niet eenduidig geïnterpreteerd worden. In het ongunstigste geval is er bij een kwart van de achtjarige en de helft van de vierjarige kinderen sprake van milde zinkdeficiëntie en is volgens de in dit onderzoek gehanteerde definitie bij 4-5% van de vierjarigen en 1-3% van de achtjarigen sprake van een verhoogde kans op ijzerdeficiëntie. Om tot een betere vaststelling van de adequaatheid van de micronutriëntenvoorziening van kinderen te komen, wordt meer onderzoek aanbevolen naar de te hanteren parameters en grenswaarden van voedingsstatus en wordt tevens geadviseerd om geregeld voedingsstatusonderzoek bij kinderen uit te voeren, ook bij iets jongere en oudere kinderen.

Inhibition of mammalian target of rapamycin (mTOR) signaling in cultured human primary trophoblast cells reduces the activity of key placental amino acid transporters. However, the upstream regulators of placental mTOR are unknown. We hypothesized that glucose, insulin, and IGF-I regulate placental amino acid transporters by inducing changes in mTOR signaling. Primary human trophoblast cells were cultured for 24 h with media containing various glucose concentrations, insulin, or IGF-I, with or without the mTOR inhibitor rapamycin, and, subsequently, the activity of system A, system L, and taurine (TAUT) transporters was measured. Glucose deprivation (0.5 mM glucose) did not significantly affect Thr172-AMP-activated protein kinase phosphorylation or REDD1 expression but decreased S6 kinase 1 phosphorylation at Thr389. The activity of system L decreased in a dose-dependent manner in response to decreasing glucose concentrations. This effect was abolished in the presence of rapamycin. Glucose deprivation had two opposing effects on system A activity: 1) an “adaptive” upregulation mediated by an mTOR-independent mechanism and 2) downregulation by an mTOR-dependent mechanism. TAUT activity was increased after incubating cells with glucose-deprived media, and this effect was largely independent of mTOR signaling. Insulin and IGF-I increased system A activity and insulin stimulated system L activity, effects that were abolished by rapamycin. Milk, fat, and protein loss due to a new subclinical mastitis case may be economically important, and the objective of this study was to estimate this loss. The loss was estimated based on test-day (TD) cow records collected over a 1-yr period from 400 randomly selected Dutch dairy herds. After exclusion of records from cows with clinical mastitis, the data set comprised 251,647 TD records from 43,462 lactations of 39,512 cows. The analysis was carried out using a random regression test-day modeling approach that predicts the cow production at each TD based on the actual production at all previous TD. The definition of new subclinical mastitis was based on the literature and assumed a new subclinical case if somatic cell count (SCC) was >100,000 cells/mL after a TD with SCC 100,000 cells/mL. The production (milk, fat, or protein) losses were modeled as the difference between the actual and predicted production (milk, fat, or protein) at the TD of new subclinical mastitis, for 4,382 cow records, and 2,545 cow records after dilution correction. Primiparous cows were predicted to lose 0.31 (0.25¿0.37) and 0.28 (0.20¿0.35) kg of milk/d at an SCC of 200,000 cells/mL, for unadjusted and adjusted low SCC, respectively. For the same SCC increase, multiparous cows were predicted to lose 0.58 (0.54¿0.62) and 0.50 (0.44¿0.56) kg of milk/d, respectively. Moreover, it was found that the greater the SCC increase above 100,000 cells/mL, the greater the production losses. The estimated production losses were more precise than previously reported estimates.

Prediction of breeding values using whole-genome dense marker maps for genomic selection has become feasible with the advances in DNA chip technology and the discovery of thousands of single nucleotide polymorphisms in genome-sequencing projects. The objective of this study was to compare the accuracy of predicted breeding values from genomic selection (GS), selection without genetic marker information (BLUP), and gene-assisted selection (GEN) on real dairy cattle data for 1 chromosome. Estimated breeding values of 1,300 bulls for fat percentage, based on daughter performance records, were obtained from the national genetic evaluation and used as phenotypic data. All bulls were genotyped for 32 genetic markers on chromosome 14, of which 1 marker was the causative mutation in a gene with a large effect on fat percentage. In GS, the data were analyzed with a multiple quantitative trait loci (QTL) model with haplotype effects for each marker bracket and a polygenic effect. Identical-by-descent probabilities based on linkage and linkage disequilibrium information were used to model the covariances between haplotypes. A Bayesian method using Gibbs sampling was used to predict the presence of a putative QTL and the effects of the haplotypes in each marker bracket. In BLUP, the haplotype effects were removed from the model, whereas in GEN, the haplotype effects were replaced by the effect of the genotype at the known causative mutation. The breeding values from the national genetic evaluation were treated as true breeding values because of their high accuracy and were used to compute the accuracy of prediction for GS, BLUP, and GEN. The allele substitution effect for the causative mutation, obtained from GEN, was 0.35% fat. The accuracy of the predicted breeding values for GS (0.75) was as high as for GEN (0.75) and higher than for BLUP (0.51). When some markers close to the QTL were omitted from the model, the accuracy of prediction was only slightly lower, around 0.72. T
