254. Performance-Based Teacher Education Curricula: Implications for Programs.

Abstract

The paper provides a brief survey of the rationale for and problems associated with performance-based teacher education (PBTE). Reviewing the research and activities of the Center for Vocational and Technical Education (Ohio State), the document identifies five elements that appear to distinguish PBTE from more traditional programs. These elements relate to: student competencies; criteria for assessing competencies; assessment of the student's competency; the student's rate of progress; and the intention of the instructional program. For the implementation of PBTE, six issues, and six strategies for dealing with the issues, are identified and discussed briefly. The issues and strategies relate to: identification of competencies; instructional materials; roles of faculty and students; interaction with various groups, institutions, and agencies; instructional support; and costs. 255. Youth with Special Needs in Non-Metropolitan Ohio High Schools.

Abstract

To determine characteristics of students and aspects of vocational education programs essential to the serving of youth with special needs in rural Ohio high schools, 154 ninth grade youths with special needs were compared with 169 other ninth grade students. Analysis of variance and coefficient of correlation "t" test and chi square were used to establish significant differences and to examine relationships. Some findings were: (1) One of seven ninth grade students in rural Ohio high schools was considered to have special educational needs, (2) Boys identified as disadvantaged outnumbered girls by a ratio of three to two, (3) The greatest numbers of students were considered to be intellectually handicapped, educationally deprived, economically deprived and socially disadvantaged, (4) Very few students were considered to be ethnically disadvantaged or physically handicapped, (5) Youth with special needs were found to have larger families, more broken homes, and lower education and occupational levels of parents, (6) Youth with special needs had lower grades, high rates of absence, lower reading levels, lower intelligence test scores, and lower educational and occupational aspirations, and (7) No significant differences were found in terms of race, place or origin, or whether or not the mother worked outside the home. Abstract

Apolipoprotein C-III (APOC3) inhibits triglyceride clearance by reducing lipoprotein lipase--mediated hydrolysis and hepatocyte uptake of triglyceride-rich lipoproteins. ARO-APOC3, a hepatocyte-targeting RNA interference therapeutic, inhibits APOC3 messenger ribonucleic acid expression, lowering triglyceride levels. The objective of this trial was to assess the safety, pharmacodynamic variables, and pharmacokinetic variables of ARO-APOC3 treatment. Healthy participants and adults with hypertriglyceridemia were randomly assigned to receive escalating single (day 1) or repeat (days 1 and 29) doses, respectively, of subcutaneous injections of ARO-APOC3 10, 25, 50, or 100 mg or placebo; they were followed up until day 113. Additional cohorts of healthy participants and adults with chylomicronemia received repeat doses of open-label ARO-APOC3. The primary objective was to evaluate the safety and side effect profile of ARO-APOC3. Key secondary and exploratory objectives included pharmacokinetic variables and changes in serum APOC3, triglyceride, and cholesterol levels. Eighty-eight participants received ARO-APOC3 and 24 participants received placebo across double-blind and open-label cohorts. Treatment-emergent adverse events (AEs) of transient, mild to moderate liver transaminase changes occurred in 10 participants: 1 patient receiving ARO-APOC3 25 mg, 5 patients receiving ARO-APOC3 50 mg, and 4 participants receiving ARO-APOC3 100 mg (1 healthy participant and 3 patients with hypertriglyceridemia). These events were asymptomatic, and transaminase levels returned to near baseline by the end of the trial. No AEs related to thrombocytopenia or platelet declines were reported. In the hypertriglyceridemia cohorts, the day 113 mean changes from baseline in APOC3 at the 10-, 25-, 50-, and 100-mg doses were -'62.0%, -'81.7%, -'90.1%, and -'94.4%, respectively, compared with -'1.6% with placebo. This corresponded to median changes in triglyceride levels of -'65.6%, -'69.9%, -'81.2%, and -'81.0% compared with -'2.8% with placebo. In this small trial of short duration, ARO-APOC3 was associated with few AEs and reduced serum levels of APOC3 and triglycerides in healthy participants and patients with hypertriglyceridemia. 266. A systematic review of solar driven waste to fuel pyrolysis technology for the Australian state of Victoria

Abstract

Australia’s waste management is heavily dependent on landfill. The Australian Bureau of Statistics predicts that as Australia’s population and economy grow, there will be a significant increase in the magnitude of waste output. Concentrated solar driven pyrolysis has been identified as a promising means in creating renewable liquid fuel and improving waste management. This technology is based on the upgrading of waste into a valuable commodity. This is of interest to city councils, communities and stakeholders, as both Australia’s annual waste generation and energy demands are growing rapidly. This paper provides a case for the implementation of solar-driven pyrolysis for biofuel production in the Australian state of Victoria as well as a comparative analysis of different Renewable Energy Sources and biomass reactions to justify the combination of Concentrated Solar Power with pyrolysis. This study is the first to assess the solar-driven pyrolysis under a Victorian setting. Victoria was chosen due to the comprehensive and readily available waste data kept by its government. The review concluded that a combination of Fresnel Reflector CSP and pyrolysis are best suited for regional Victorian environment. Fresnel reflector technology was found to complement slow pyrolysis well due to its 250–500◦C operating temperature, while regional Victoria was found to be a good trade-off between feedstock distance and DNI exposure. These requirements were important because feedstock transportation was found to cost $AUD 96 per 500km, and it was observed that there could be up to 1200 kWh/m2solar exposure difference between Victorian rural and metropolitan locations. This study provides a comprehensive framework of technical requirements pertaining to a Victorian solar-driven pyrolysis system, which will then act as a guidance for future designers.

267. SSO and other putative inhibitors of FA transport across membranes by CD36 disrupt intracellular metabolism, but do not affect FA translocation

Abstract

Membrane-bound proteins have been proposed to mediate the transport of long-chain FA (LCFA) transport through the plasma membrane (PM). These proposals are based largely on reports that PM transport of LCFAs can be blocked by a number of enzymes and purported inhibitors of LCFA transport. Here, using the ratiometric pH indicator (2′,7′-bis-(2-carboxyethyl)-5-(and-6-)-carboxyfluorescein and acrylodated intestinal FA-binding protein-based dual fluorescence assays, we investigated the effects of nine inhibitors of the putative FA transporter protein CD36 on the binding and transmembrane movement of LCFAs. We particularly focused on sulfosuccinimidyl oleate (SSO), reported to be a competitive inhibitor of CD36-mediated LCFA transport. Using these assays in adipocytes and inhibitor-treated protein-free lipid vesicles, we demonstrate that rapid LCFA transport across model and biological membranes remains unchanged in the presence of these purported inhibitors. We have previously shown in live cells that CD36 does not accelerate the transport of unesterified LCFAs across the PM. Our present experiments indicated disruption of LCFA metabolism inside the cell within minutes upon treatment with many of the “inhibitors” previously assumed to inhibit LCFA transport across the PM. Furthermore, using confocal microscopy and a specific anti-SSO antibody, we found that numerous intracellular and PM-bound proteins are SSO-modified in addition to CD36. Our results support the hypothesis that LCFAs diffuse rapidly across biological membranes and do not require an active protein transporter for their transmembrane movement.

Non-alcoholic steatohepatitis (NASH) is a common type of chronic liver disease that can lead to cirrhosis. Obeticholic acid, a farnesoid X receptor agonist, has been shown to improve the histological features of NASH. Here we report results from a planned interim analysis of an ongoing, phase 3 study of obeticholic acid for NASH.

Aberrant methylation of the promoter, P2, and the first exon, E1, regions of the tumor suppressor gene RASSF1A, have been associated with hepatocellular carcinoma (HCC), albeit with poor specificity. This study analyzed the methylation profiles of P1, P2 and E1 regions of the gene to identify the region of which methylation most specifically corresponds to HCC and to evaluate the potential of this methylated region as a biomarker in urine for HCC screening.Bisulfite DNA sequencing and quantitative methylation-specific polymerase chain reaction assays were performed to compare methylation of the 56 CpG sites in regions P1, P2 and E1 in DNA isolated from normal, hepatitic, cirrhotic, adjacent non-HCC, and HCC liver tissue and urine samples for the characterization of hypermethylation of the RASSF1A gene as a biomarker for HCC screening.In tissue, comparing HCC (n = 120) with cirrhosis and hepatitis together (n = 70), methylation of P1 had an area under the receiver operating characteristics curve (AUROC) of 0.90, whereas methylation of E1 and P2 had AUROC of 0.84 and 0.72, respectively. At 90% sensitivity, specificity for P1 methylation was 72.9% versus 38.6% for E1 and 27.1% for P2. Methylated P1 DNA was detected in urine in association with cirrhosis and HCC. It had a sensitivity of 81.8% for α-fetoprotein negative HCC.Among the three regions analyzed, methylation of P1 is the most specific for HCC and holds great promise as a DNA marker in urine for screening of cirrhosis and HCC.

270. The potential of variable speed diesel application in increasing renewable energy source penetration.

Abstract

Abstract Integration of renewable energy source (RES) generation to displace diesel generation can present clear economic, environmental and social benefit. While low level RES integration is relatively easy to achieve, both the cost and complexity escalate as systems target increasing RES penetration. A key barrier to greater RES penetrations remains the inefficiency of diesel generation to operate at low or partial loading. To achieve low or partial loading, conventional fixed speed diesel technologies must rely on prescribed purge routines, which serve to increase emissions intensity and fuel consumption. Fixed speed constraint remains the primary barrier to increased engine flexibility and improved partial load efficiency. This paper investigates redesign of the diesel generator to achieve variable speed operation. A suitable design basis is developed, with laboratory testing used to validate unit performance, ahead of economic evaluation. Economic modelling is presented to explore the improve engine flexibility, required of hybrid diesel applications. Variable speed application is shown to reduce diesel fuel consumption by up to 40 % in comparison to conventional hybrid diesel applications. [ABSTRACT FROM AUTHOR]

Abstract In the isolated power system, consumers are traditionally supplied with electricity produced by diesel generators. Conventional diesel generators demonstrate robust and efficient operation when connected to electrical grids with slowly varying loads. With the introduction of intermittent and stochastic renewable energy sources, such generators may not be responsive enough to retain the stability and reliability of the system. The problem becomes especially acute in cases when diesel generators are required to operate at a low load. Regulating devices (e.g. energy storage systems and/or dump load) used to improve the system reliability, increase system complexity and incur additional expenses or energy losses. This paper investigates low load diesel technology as a potential solution to high level renewable energy penetration. It mentions book of author "The British Museum: Storehouse of Civilizations" reflects on that museum's historic example of curatorship, and on what a museum is for, and writing it focused my mind on the dangers faced by local and regional museums and art galleries.

273. Resolvin E1 heals injured cardiomyocytes: Therapeutic implications and H-FABP as a readout for cardiovascular disease & systemic inflammation.

Abstract

• H-FABP leakage is a marker for damage to cardiomyocyte cells in culture. • Resolvin E (RvE1) is a therapeutic that decreases inflammation and leakage of H-FABP. • We propose a unifying mechanistic hypothesis of high systemic inflammation and therapy based on inflammation resolution for patients with CVD and multiple comorbidities. The purpose of this study is to investigate heart-fatty acid binding protein (H-FABP) leakage from cardiomyocytes as a quantitative measure of cell membrane damage and to test healing by Resolvin E1 (RVE1) as a potential therapeutic for patients with inflammatory diseases (cardiovascular disease and comorbidities) with high morbidity and mortality. Our quantitative ELISA assays demonstrated H-FABP as a sensitive and reliable biomarker for measuring cardiomyocyte damage induced by lipopolysaccharide (LPS) and healing by RvE1, a specialized pro-resolving mediator (SPM) derived from the Omega-3 fatty acid, eicosapentaenoic acid (EPA), a dietary nutrient that balances inflammation to restore homeostasis. RvE1 reduced leakage of H-FABP by up to 86%, which supports our hypothesis that inflammation as a mechanism of injury can be targeted for therapy. H-FABP as a blood biomarker was tested in 40 patients admitted to Boston Medical Center for respiratory distress, (20 patients with and 20 patients without COVID infection). High levels of H-FABP correlated with clinically diagnosed CVD, diabetes, and end-stage renal disease (ESRD) in both patient groups. The level of H-FABP indicates not only CVD damage but is a valuable measure for patients with increased inflammation disease comorbidities. [ABSTRACT FROM AUTHOR]

279. A Comparison of Domestic and International Students' Perceptions of the University Environment.

Domestic and international students' perceptions of the university environment were compared on 11 factors of the College Characteristics Index (CCI). Only five reached the level of significance, with the international students scoring significantly higher. None of the other comparisons approached the level of significance. (Author)
