Background: The normal response to tissue injury involves a complex sequence of inflammation and repair of epithelial and mesenchymal structures through a coordinated process of chemotaxis and cellular proliferation directed by specific chemical mediators. One mediator that is probably important to these processes is basic fibroblast growth factor (bFGF), a polypeptide present in diverse tissues and a potent chemotactic, mitogenic, and angiogenic factor in vitro. Although many cells have receptors for bFGF, the precise role of this factor as a paracrine mediator in vivo remains disputable because a mechanism for its secretion has not been elucidated. bFGF is known to be capable of binding to heparin, and our data demonstrate bFGF in mast cells (MCs) that also produce many other mediators (including heparin) important for wound healing and angiogenesis., Experimental Design: We used nucleic acid in situ hybridization to detect bFGF-specific mRNA transcripts within MCs in formalin-fixed, paraffin-embedded specimens of nonlesional human skin and skin containing increased numbers of MCs, including granulation tissues, benign cutaneous mastocytosis, and solitary neurofibromas. Immunohistochemistry was used to confirm bFGF protein expression in each tissue studied., Results: All tissues examined contained MCs that expressed bFGF mRNA transcripts. bFGF protein was also detected in MCs, but the intensity of labeling was dependent on prior treatment of the tissues with heparinase, suggesting that epitopes recognized by the bFGF-specific antiserum were masked by endogenous heparin., Conclusions: The results further elucidate the mechanism of involvement of MCs in tissue repair and angiogenesis and suggest that bFGF produced by MCs is bound to intracellular heparin. Thus, it seems possible that release of heparin by degranulation from MCs also may result in the release of biologically active bFGF in vivo.

The identification of cell cultures contaminated with organisms from the class Mollicutes has led us to examine the effectiveness of polymerase chain reaction (PCR) for detecting these organisms in genomic DNA. We developed a previously identified nested PCR primer set and compared its ability to detect Mycoplasma with that of a commercially available PCR kit for detecting Mycoplasma. We found that although the commercial system detected and identified a few of the most common Mycoplasma species, the primer set (GPO-1, GPO-2, MGSO) detected the presence of all the common Mycoplasma species and many of the rare mycoplasma species previously encountered in tissue culture.

Mice lacking functional B cells because of a genetic deletion of the mu chain (IgM) gene were used to investigate the role of perinatal and postnatal transfer of maternal IgG in neonatal growth. Our results confirmed that immunoglobulin (Ig)-deficient mice successfully complete pregnancy and deliver healthy offspring. However, neonates nursed by Ig-deficient mothers showed growth retardation (runting) and high mortality during their first 10 days of life. This fatal course was seen whether or not the neonates were Ig-deficient. Cross-switching litters from phenotypically normal mothers to Ig-deficient mothers immediately after birth showed that perinatal Ig transfer normalized neonatal development for 10 days, but was not sufficient to sustain survival during the later part of the neonatal period. On the other hand, all Ig-deficient litters nursed by normal foster mothers showed normal development and 0% neonatal mortality. Administration of mouse IgG to an Ig-deficient mother or a neonate during the first critical week prevented runting. We assume that the growth- and health-promoting effects of IgG during early neonatal life are attributable mainly to the transfer of passive immunity to environmental pathogens. However, the finding that monoclonal IgG antibodies also enhanced neonatal growth and survival suggests that IgG-dependent growth-promoting mechanisms could be involved as well.

Type VII collagen, a major component of anchoring fibrils in the basement membrane zone, is now considered to be a primary genetic factor in the pathogenesis of dominant dystrophic epidermolysis bullosa (DDEB). In this study, we performed genetic linkage analysis in a Japanese family with DDEB using a PvuII polymorphism in the type VII collagen gene. The pedigree consisted of 10 affected and 13 unaffected living individuals and was diagnosed as having Cockayne-Touraine type of DDEB. Electron microscopic examination of the skin demonstrated a diminished number and rudimentary structure of anchoring fibrils. PCR-based detection of PvuII polymorphism resulted in 3 genotypes and co-segregated with DDEB phenotype in this pedigree. The maximum lod score was 2.10 at recombination fraction (theta) of 0. The absence of recombination between DDEB and type VII collagen gene locus, as well as the observation of altered anchoring fibrils, suggested that type VII collagen is a candidate gene for the Japanese family with DDEB, although the lod score was statistically not significant.

The L-type voltage-dependent calcium channel (L-VDCC) is assumed to be a critical component of excitation-contraction coupling in smooth muscle. Using pregnant rat myometrium, we examined the hypothesis that parturition is associated with significant changes in the expression of the alpha 1 subunit of the L-VDCC at the mRNA or protein level. The binding of radiolabeled dihydropyridine, which correlates with the total number of calcium channels in the membrane, was increased by 14 days' gestation, in comparison to that in nonpregnant controls. The elevation in binding capacity persisted through labor and fell postpartum. Northern and RNA dot-blot analysis demonstrated the highest level of expression on Days 20 and 21, with a 3- to 10-fold decrease during parturition. We believe these studies are most consistent with a one-day lag time between mRNA and protein expression, and generally support a modest increase in L-VDCC expression in pregnancy and labor. Reverse transcriptase polymerase chain reaction was used to examine changes in isoform expression in Motif IV, a region of the alpha 1 subunit known to be alternatively spliced. These studies revealed the presence of multiple isoforms in rat myometrium, with a predominance of IVS3B. Interestingly, a marked increase in the ratio of S3B:S3A was noted at parturition. In summary, these data demonstrate that the number of L-type calcium channels, although increased in pregnancy, do not change prior to, or with the onset of, myometrial contraction. Intriguingly, mRNA expression was markedly decreased at parturition. The change in isoform expression during labor is of unknown, but potential, physiologic significance.

A squamous cell carcinoma cell line Nakata proliferated in serum-free culture and was not responsive to exogenous fibroblast growth factor-1 (FGF-1). Immunostaining revealed that Nakata cells expressed FGF-1 in their cytoplasms and nuclei. Two molecular mass species of FGF-1 (16 and 18 kDa) were identified in cell extracts by Western blot. These cells also expressed high-affinity FGF-1 binding sites (Kd = 360 pM, 28,000 sites/cell). The results of cross-linking with [125I]FGF-1 demonstrated the presence of two bands with molecular masses of 160 and 140 kDa. The addition of FGF-1 specific antisense oligonucleotides at 25 microM to Nakata cells resulted in an 82% inhibition in cell growth and suppressed FGF-1 expression. This effect was dose-dependent and specific, because sense oligonucleotides were ineffective in inhibiting cell growth. In addition, Nakata cell growth was suppressed by an anti-FGF-1 neutralizing antibody, which resulted in a 52% inhibition at 8 micrograms/ml. These results demonstrate that Nakata cells produce FGF-1, and indicate that this growth factor acts in an autocrine manner by interacting with FGF-1 binding sites on Nakata cells.

Testin is an authentic Sertoli cell secretory protein consisting of two molecular variants designated testin I (M(r) 35 000) and testin II (M(r) 37 000). N-Terminal amino acid sequence analysis revealed that testin I is identical to testin II except that testin II has three extra N-terminal amino acids of threonine-alanine-proline (TAP). Earlier studies by immunoflorescence microscopy have shown that testin is detected in the seminiferous epithelium consistent with localization in the junctions between Sertoli cells as well as Sertoli-germ cells, and that it appears to be a component of junctional complexes in the testis. In the present study, we have examined the localization of testin in different stages of the spermatogenic cycle of the adult rat testis when germ cells migrate from the basal portion of the seminiferous epithelium to the tubular lumen. In stages I-IV, testin was localized mainly in the basement laminae in the junctional complexes between adjacent Sertoli cells as well as between Sertoli cells, spermatogonia, and pachytene spermatocytes. When elongated spermatids were embedded into the seminiferous epithelium in stage VII of the cycle, testin was detected predominantly on the concave side of the elongated spermatids, but relatively few testin reaction products were seen in the round spermatids. In the beginning of stage VIII of the spermatogenic cycle, intense testin immunoreactive substances were detected around the heads of the elongated spermatids; these substances were virtually undetectable in late stage VIII after the release of the mature sperm into the tubular lumen, suggesting that testin may be a novel marker to divide stage VIII into stages VIIIa and VIIIb.(ABSTRACT TRUNCATED AT 250 WORDS)

There have been no reports, to date, of successful introduction of foreign DNA into committed megakaryocyte precursor cells. We have successfully infected two megakaryocytic cell lines, one a committed cell line (CHRF-288-11) and the other a bipotential cell line (K562), with a retroviral vector containing the bacterial lacZ gene and a neomycin resistance marker. Modification of standard protocols was required for successful infection of the committed megakaryocyte cell line. Presence of the lacZ transgene was demonstrated at the molecular level by polymerase chain reaction (PCR), and its expression at the mRNA level by reverse transcriptase-PCR. Presence of the bacterial beta-galactosidase was demonstrated by both immunofluorescence and enzyme activity. Treatment of the CHRF-288-11 infected cells with phorbol esters, which induces megakaryocytic differentiation, increases expression of the lacZ transgene. The staining pattern of the lacZ reaction product was perinuclear and punctate in the CHRF-288-11 cells, whereas it was uniform throughout the cytoplasm of the K562 cells, suggesting different sorting mechanisms for bacterial beta-galactosidase in these two cell types. Overall, these results demonstrate the feasibility and provide a method for infecting cultured megakaryocytic cell lines with retroviral of vectors such that a molecular analysis of megakaryocyte differentiation can be accomplished.

An mRNA with a substantial similarity to the rat p62 mRNA that encodes a nucleoporin was cloned from the rat testis. A probe derived from a unique sequence in the nucleoporin-related (NPR) cDNA revealed a novel mRNA of 1.3 kb, different from the 2.7-kb transcript attributed to the p62 gene. This 1.3-kb transcript was not detected in Sertoli cells; it was found primarily in the haploid germ cells of the adult testis. The DNA sequencing revealed that the central region of the NPR cDNA sequence was identical to the 3' portion of the p62 cDNA containing heptad repeat sequences. However, the 5' region and the extreme 3' region of the NPR cDNA sequence were different from the p62 cDNA. Interestingly, the extreme 3' untranslated region (UTR) contained a 212-bp inverted repeat of a sequence located in the middle of the NPR cDNA that is identical to the p62 sequence. The inverted repeats of the NPR sequence could potentially hybridize, leading to the formation of circular transcripts. Using antibodies specific for the C-terminal regions of p62, a 26-kDa protein was detected from NPR cDNA hybrid-arrested translational products, and a 28-kDa protein was detected from the testis germ cell extracts but not from Sertoli cell extracts.

We have generated transgenic mice that express the simian virus 40 (SV40) large T antigen under the control of a 1109 bp 5'-flanking sequence of the human thyrotropin beta-subunit (TSH beta) gene. The hybrid gene, termed TTP-1, was microinjected into fertilized mouse eggs and 11 transgenic mice were obtained. One of the transgenic mice, a female, a phenotypical dwarf, developed a pituitary tumor and wasted away from 7 to 9 weeks after birth. To establish the transgenic mouse line, her ovaries were transferred to a normal female, whose ovaries were removed beforehand. To examine the tissue specificity of transgene expression, mRNA of SV40 large T antigen was monitored in various tissues from the transgenic mice by the reverse transcriptase-polymerase chain reaction analysis, and was detected only in the pituitary. Histological and immunohistochemical analyses showed that the pituitary tumors of the transgenic mice were composed of poorly differentiated pituitary cells expressing SV40 large T antigen. These results indicated that the 1109 bp sequence of the human TSH beta 5'-flanking region is essential for pituitary-specific expression of SV40 large T antigen in transgenic mice, which exhibited a dwarf phenotype and developed pituitary tumors. The tumors were composed of undifferentiated cells and did not produce thyrotropin. These transgenic mice should provide a valuable animal model for studying the pathogenesis of anterior pituitary tumors.

The performance of oligonucleotide primers containing deoxyinosine (dl) at all ambiguous positions for polymerase chain reaction, based on ambiguous sequence information derived either from compilations of consensus nucleotide sequences or from amino acid sequences, has been evaluated in two model systems represented respectively by amplification of conserved genomic regions from different types of human papillomavirus and by amplification of a region of the human lysozyme cDNA on the basis of the protein amino acid sequence. In both instances the dl-containing primers obtained the expected amplification products. When using short primers or primers with very high dl contents, however, peculiar reaction conditions had to be adopted to obtain successful amplification and, in the latter case, performance remained suboptimal. Comparison of results with those obtained using corresponding degenerate primers showed that the use of dl-containing primers can be advantageous in terms of both specificity and yield of the amplification product. Sequence analysis of amplification products showed that dG residues are always found at positions corresponding to the dl residues of the primers.
