This paper focuses on the boundary area between the Central and Southern Apennines thrust belt of the Italian peninsula, which separates two different patterns of distribution of active faults and strong earthquakes along the Apennines chain. In order to reduce the remarkable lack of data on this area, part of this boundary has been investigated. The study area corresponds to the surroundings of the town of Isernia, an important centre of the Molise region, and contains the Carpino and Le Piane (CLP) intermountain Basins of Quaternary age. The research was based on unpublished data, consisting of 65 well logs drilled in the CLP Basins, integrated with detailed field surveys (scale 1:10,000), mesoscale structural analysis and aerial photograph interpretation. This study allowed to define the geometry and kinematics of the CLP Basins. They result to be asymmetric halfgrabens partially filled with fluvio-lacustrine deposits, where the sedimentation is controlled by the activity of a set of normal faults striking N30jW and NE dipping. This set of faults, which developed mainly during the Late Pleistocene–Holocene, is about 10 km long and is located close to the northwestern end of the Boiano Basin extensional Fault System. The average slip rate calculated for part of the Holocene for one of the recognised faults ranges from 0.75 to 1.00 mm/year. The identification of the active CLP Basins Fault System partially fills the gap of data at the boundary between the Central and Southern Apennines. From a more general point of view, we hypothesise that this boundary might be regarded as a ‘‘persistent segment boundary’’, forming a long-term barrier to the propagation of rupture of active fault systems. The growing and widespread use of glyphosate-based herbicides (GBHs) has raised an intense public debate about the impact of environmental contamination on animal and human health, including male fertility. The aim of this study was to deepen the impact of glyphosate (Gly) and GBHs on mammalian sperm investigating the effect of in vitro exposure of stallion spermatozoa to Gly and to its commercial formulation Roundup® (R). Spermatozoa were incubated at 37 °C with different Gly or R concentrations (from 0.5 to 720 μg/mL Gly or R at the same Gly-equivalent concentrations). After 1 h of incubation motility, viability, acrosome integrity, mitochondrial activity and ROS production were assessed. Gly, at all the concentrations tested, did not induce any detrimental impact on the sperm quality parameters evaluated. Conversely, R starting from 360 μg/mL (Gly-equivalent dose) significantly (P < 0.05) decreased total and progressive motility, viability, acrosome integrity, mitochondrial activity and the percentage of live spermatozoa with intact mitochondria not producing ROS. Our results indicate that the commercial formulation R is more toxic than its active molecule Gly and that the negative impact on stallion sperm motility might be likely due to a detrimental effect mainly at membrane and mitochondrial level and, at least in part, to redox unbalance. Moreover, based on the data obtained, it can be hypothesized a species-specificity in sperm sensitivity to Gly and GBHs as horse spermatozoa were negatively influenced at higher concentrations of R compared to those reported in literature to be toxic for human and swine male germ cells.

Vitrified MII porcine oocytes are characterized by reduced developmental competence, associated with the activation of the apoptotic pathway. Resveratrol (R), a polyphenolic compound present in several vegetal sources, has been reported to exert, among all its other biological effects, an antiapoptotic one. The aim of this study was to determine the effects of R (2 µM) on the apoptotic status of porcine oocytes vitrified by Cryotop method, evaluating phosphatidylserine (PS) exteriorization and caspases activation. R was added during IVM (A); 2 h postwarming incubation (B); vitrification/warming and 2 h postwarming incubation (C); all previous phases (D). Data on PS exteriorization showed, in each treated group, a significantly higher (P<0.05) percentage of live nonapoptotic oocytes as compared with CTR; moreover, the percentage of live apoptotic oocytes was significantly (P<0.05) lower in all R-treated groups relative to CTR. The results on caspase activation showed a tendency to an increase of viable oocytes with inactive caspases in B, C, and D, while a significant (P<0.05) increase in A compared to CTR was recorded. These data demonstrate that R supplementation in various phases of IVM and vitrification/warming procedure can modulate the apoptotic process, improving the resistance of porcine oocytes to cryopreservation-induced damage.

The aim of this study was to verify the reliability of an open access CASA software (BGM) to evaluate the sperm motility of cattle and buffalo, comparing motility and kinematic parameters to those of a commercial one (HTM). Thirty frozen-thawed samples for each species were analyzed with both HTM and BGM, after 1 h of incubation at 37 °C. Sperm viability and mitochondrial membrane potential (MMP) were evaluated through flow cytometric analysis. Agreement of all motility variables between the two systems was assessed. Correlation analysis was performed to identify relationships between motion parameters and sperm viability and MMP. Bland Altman analysis showed good agreement between methods for all motility parameters except for curvilinear velocity (VCL) in cattle, and for average path (VAP), VCL and (amplitude of lateral head displacement) ALH in buffalo, that showed a proportional bias (P > 0.05). In both systems, positive correlation between both viability and high MMP and total and progressive motility of cattle spermatozoa were found; viability and the sperm with high MMP were positive correlated only with VAP, straight-line (VSL), VCL and ALH evaluated with HTM system. Different results were found for buffalo sperm motility parameters, since viability had positive correlations and mitochondrial activity negative ones. Results suggested that motility assessment performed by these two systems are comparable. The discrepancy of VCL, VAP, and ALH could be due to the difference in the algorithms between software. The open-access CASA plug-in is a reliable alternative to the expensive commercial CASA system for sperm motility assessment in cattle and buffalo.

The inception and growth of the active Carpino-Le Piane Basin Fault System (CLPBFS; central-southern Apennines, Italy) was analysed with respect to the neighbouring Isernia and Surrounding (ISFS) and Boiano Basin (BBFS) extensional Fault Systems. Ar-39-Ar-40 dating showed that the BBFS was already active 649 +/- 21 ka bp and that the ISFS was active at least 476 +/-10 ka bp, whereas the activity of the CLPBFS started certainly later than 253 +/- 22 ka bp, and very probably as recently as

Fold-thrust belts are commonly interpreted as "thin-skinned" structures, developed above a detachment, with the underlying basement remaining undeformed. However, in many areas, particularly where compressional tectonism was preceded by rifting, models of basement fault reactivation may be more appropriate. The contrasts between thin-skinned and deep-rooting, inversion-dominated deformation in building fold-thrust complexes are investigated using a case history from the Italian Apennines. Three sectors were chosen to represent the marked lateral variations in structural style evident in the thrust belt. The outer portion of the Marche (in the north) is contrasted with a section through the Lucanian Apennines in the south and with the Molise district of the Central Apennines. The Marche structures are readily explained in terms of inversion, a model that is consistent with new deep seismic data onshore and conventional seismic from the nearby Adriatic Sea. The displacements implicit for the inversion model are a factor of five less than for existing thin-skinned interpretations. However, these styles are not applicable throughout the Apennines. Well data in the Southern Apennines of Lucania demonstrate large-scale thin-skinned thrusting, with 57 km of horizontal displacement since earliest Pliocene time. This includes 14 km of shortening that ramps up through the buried Apulian Platform carbonates. These deeper structures may be restored using ramp-dominated thrust geometries. The Molise sector shows broadly the same structural style as for Lucania: allochthonous shallow-water carbonates and pelagic basin units overlie the carbonates of the Apulian Platform, with the major difference being that here, the pelagic basin units are detached at the level of the Oligocene-lower Miocene Argille Varicolori. In this setting, the Apulian carbonates may be restored using only 5 km of displacement. The overlying allochthon probably has accommodated about 45 km of displacement since the earliest Pliocene. Therefore, the Apennines show differing structural styles with differing displacements along their length. Thick-skinned thrusting models may be applied to the Marche and to structures in the buried Apulian units.

Equine spermatozoa highly rely on oxidative phosphorylation for their energy management. The present work aimed to characterize the role of mitochondria on horse sperm motility and ROS production by incubating spermatozoa with specific inhibitors of the different mitochondrial complexes. Equine spermatozoa were incubated 1 h and 3 h at 37 °C with: complex I inhibitor rotenone (5 μM, ROT), complex II inhibitor dimethyl-malonate (10 mM, DMM), complex III inhibitor antimycin A (1.8 μM, ANTI), the uncoupling agent carbonyl cyanide m-chlorophenyl hydrazine (5 μM, CCCP), ATP synthase inhibitor oligomycin (5 μM, OLIGO), and 2 μL vehicle DMSO (control, CTL). Samples were analyzed for sperm motility and for mitochondrial membrane potential (MMP), mitochondrial integrity, mitochondrial O2•– production, and cytoplasmic H2O2. A multivariate analysis was performed on the data. CCCP caused a pronounced MMP reduction at both time points while ROT and ANTI showed the same effect at 3 h. All treatments at 3 h incubation significantly reduced the percentage of sperm with early changes in membrane permeability with active mitochondria. The H2O2 production of live cells was low at 1 h incubation in all treatments; after 3 h a slight decrease in the percentage of low-H2O2 producing cells was recorded. All treatments, except DMM, induced a significant decline in sperm motility and kinematics and modified the pattern of sperm subpopulations. The effect of DMM was evident only after 3 h, increasing the percentage of slow sperm subpopulation. In conclusion, the disruption of mitochondrial integrity induces an increase of mitochondrial ROS production that could be detrimental for cell function and survivior.

Context While conventional semen analysis is a simple, time-saving, and economical means to evaluate sperm quality, it leaves biochemical and metabolic characteristics of spermatozoa aside. To address this issue, the use of fluorescent probes assessing functional sperm parameters, such as JC-1, DiOC6(3) and MitoTracker, has increased over the last decades. Apparently contradictory observations have nevertheless fostered an ongoing debate on their sensitivity and ability to evaluate the mitochondrial membrane potential (MMP) of sperm cells, thus warranting a re-examination of these probes. Aims The present study aims to elucidate the suitability and sensitivity of each probe to evaluate the MMP of bovine spermatozoa by flow cytometry. Methods Cryopreserved spermatozoa from ten bulls were thawed, stained with JC-1/SYTOXRed, DiOC6(3)/propidium iodide (PI) or MitoTracker Deep Red (MTDR)/PI, and evaluated with flow cytometry and fluorescence microscopy. Key results DiOC6(3), JC-1 and MTDR can be simultaneously co-stained with a viability marker. The results of the present study support the ability of DiOC6(3)/PI and JC-1/SYTOXRed, but not that of MTDR/PI, to monitor the MMP of spermatozoa. Conclusions JC-1/SYTOXRed assessed by flow cytometry was found to be the most sensitive and robust fluorescent probe to assess MMP. Moreover, DiOC6(3)/PI could be a suitable alternative when the flow cytometer is not equipped with a red laser and/or an adequate optical filter. Implications Both DiOC6(3) and JC-1, but not MTDR, could be used as probes to assess the mitochondrial membrane potential of bovine spermatozoa.

In situations where an excessive generation of reactive oxygen species overwhelms antioxidant capacity, a harmful effect on sperm function is exerted. Antioxidants are molecules capable of minimizing this detrimental effect, which is important in pig sperm due to the high content of polyunsaturated fatty acids in their plasma membrane. The present systematic review aims at evaluating whether supplementing semen extenders (for liquid storage at 17 °C) or freezing and/or thawing media (for cryopreservation) with antioxidants influences sperm quality and functionality parameters, and in vitro/in vivo fertility outcomes. We defined inclusion and exclusion criteria in a PICOS table according to PRISMA guidelines, and conducted a literature search through MEDLINE-PubMed in November 2020. After systematic selection, 75 studies were included: 47 focused on cryopreservation and 28 on liquid storage at 17 °C. More than 70% of the studies included in this review showed that adding semen extenders for liquid storage and/or freezing/thawing media for cryopreservation with antioxidants enhances sperm quality and functionality parameters. In addition, this supplementation improves in vivo/in vitro fertility outcomes, supporting the hypothesis that the beneficial effect observed upon sperm quality has a positive impact on reproduction outcomes.

The NEAM Tsunami Hazard Model 2018 (NEAMTHM18) is a probabilistic hazard model for tsunamis generated by earthquakes. It covers the coastlines of the North-eastern Atlantic, the Mediterranean, and connected seas (NEAM). NEAMTHM18 was designed as a three-phase project. The first two phases were dedicated to the model development and hazard calculations, following a formalized decision-making process based on a multiple-expert protocol. The third phase was dedicated to documentation and dissemination. The hazard assessment workflow was structured in Steps and Levels. There are four Steps: Step-1) probabilistic earthquake model; Step-2) tsunami generation and modeling in deep water; Step-3) shoaling and inundation; Step-4) hazard aggregation and uncertainty quantification. Each Step includes a different number of Levels. Level-0 always describes the input data; the other Levels describe the intermediate results needed to proceed from one Step to another. Alternative datasets and models were considered in the implementation. The epistemic hazard uncertainty was quantified through an ensemble modeling technique accounting for alternative models’ weights and yielding a distribution of hazard curves represented by the mean and various percentiles. Hazard curves were calculated at 2,343 Points of Interest (POI) distributed at an average spacing of ∼20 km. Precalculated probability maps for five maximum inundation heights (MIH) and hazard intensity maps for five average return periods (ARP) were produced from hazard curves. In the entire NEAM Region, MIHs of several meters are rare but not impossible. Considering a 2% probability of exceedance in 50 years (ARP≈2,475 years), the POIs with MIH >5 m are fewer than 1% and are all in the Mediterranean on Libya, Egypt, Cyprus, and Greece coasts. In the North-East Atlantic, POIs with MIH >3 m are on the coasts of Mauritania and Gulf of Cadiz. Overall, 30% of the POIs have MIH >1 m. NEAMTHM18 results and documentation are available through the TSUMAPS-NEAM project website (http://www.tsumaps-neam.eu/), featuring an interactive web mapper. Although the NEAMTHM18 cannot substitute in-depth analyses at local scales, it represents the first action to start local and more detailed hazard and risk assessments and contributes to designing evacuation maps for tsunami early warning.

Destructive tsunamis are most often generated by large earthquakes occurring at subduction interfaces, but also other “atypical” sources—defined as crustal earthquakes and non-seismic sources altogether—may cause significant tsunami threats. Tsunamis may indeed be generated by different sources, such as earthquakes, submarine or coastal landslides, volcano-related phenomena, and atmospheric perturbations. The consideration of atypical sources is important worldwide, but it is especially prominent in complex tectonic settings such as the Mediterranean, the Caribbean, or the Indonesian archipelago. The recent disasters in Indonesia in 2018, caused by the Palu-Sulawesi magnitude Mw 7.5 crustal earthquake and by the collapse of the Anak-Krakatau volcano, recall the importance of such sources. Dealing with atypical sources represents a scientific, technical, and computational challenge, which depends on the capability of quantifying and managing uncertainty efficiently and of reducing it with accurate physical modelling. Here, we first introduce the general framework in which tsunami threats are treated, and then we review the current status and the expected future development of tsunami hazard quantifications and of the tsunami warning systems in Italy, with a specific focus on the treatment of atypical sources. In Italy, where the memory of historical atypical events like the 1908 Messina earthquake or the relatively recent 2002 Stromboli tsunami is still vivid, specific attention has been indeed dedicated to the progressive development of innovative strategies to deal with such atypical sources. More specifically, we review the (national) hazard analyses and their application for coastal planning, as well as the two operating tsunami warning systems: the national warning system for seismically generated tsunamis (SiAM), whose upstream component—the CAT-INGV—is also a Tsunami Service Provider of the North-eastern Atlantic, the Mediterranean and connected seas Tsunami Warning System (NEAMTWS) coordinated by the Intergovernmental Coordination Group established by the Intergovernmental Oceanographic Commission (IOC) of UNESCO, and the local warning system for tsunamis generated by volcanic slides along the Sciara del Fuoco of Stromboli volcano. Finally, we review the state of knowledge about other potential tsunami sources that may generate significant tsunamis for the Italian coasts, but that are not presently considered in existing tsunami warning systems. This may be considered the first step towards their inclusion in the national tsunami hazard and warning programs.

BACKGROUND AND AIM: After the first cases of COVID-19 detected in Wuhan (China), the virus rapidly spread in the world, so much so that on February 20 the first autochthonous case was officially identified in Italy. However, this person had no apparent history of travel abroad or contact with people tested positive for the virus. For this reason, the aim of this literature review was to reconstruct the epidemiological dynamics of the first wave of SARS-CoV-2 infection in the Lombardy Region. METHODS: To this end, a systematic review was carried out on PubMed/MEDLINE and EMBASE, and on grey literature. All article assessing incidence, mortality and hospitalizations by Lombardy province and municipality, and the impact of the main containment and organizational measures were considered eligible. In addition, data on general mortality and mortality due to COVID-19, hospital admission, and serological and environmental were also retrieved. RESULTS: From the included studies, it emerged that Lombardy was the first European region in which the virus began to circulate as early as January 2020 (and probably even earlier). Despite the high number of cases and deaths recorded, the reproduction number observed in Lombardy Region was, at the beginning of March 2020, the same (or lower) than in other regions. CONCLUSIONS: In conclusion, data of the first epidemic wave in Lombardy, compared to other Italian and foreign regions, highlight the extreme criticality of having had the first autochthonous case (and the first substantial outbreaks) when knowledge was still scarce and individual prevention measures were not widespread.

Mammalian semen is a physiological fluid composed of a cellular fraction (spermatozoa), and a liquid fraction (seminal plasma). Once delivered to the female genital tract, spermatozoa should be able to capacitate; a process which involves a plethora of biochemical and physiological changes required to fertilize the oocyte. Sperm production (spermatogenesis) occurs in the testes, whereby pluripotent spermatogonia differentiate to form the most morphologically specialized cells in the body. Further maturation of spermatozoa occurs in the epididymis, where they are stored prior to ejaculation. During this whole process, spermatozoa are exposed to different environments and cellular processes which may expose them to substantial levels of oxidative stress. To avoid damage associated with the unchecked production of reactive oxygen species (ROS), both spermatozoa, and the parts of the male genital tract in which they reside, are furnished with a suite of antioxidant molecules which are able to provide protection to these cells, thereby increasing their chance of being able to fertilize the oocyte and deliver an intact paternal genome to the future offspring. However, there are a host of reasons why these antioxidant systems may fail, including nutritional deficiencies, genetics, and disease states, and in these situations, a reduction or abolition of fertilizing capacity may result. This review paper focuses on the endogenous antioxidant defences available to spermatozoa during spermatogenesis and sperm maturation, the site of their production and their physiological role. Furthermore, we revised the causes and effects of antioxidant deficiencies (congenital or acquired during the animal's adulthood) on reproductive function in different animal species.

Sperm are redox-regulated cells, and deregulation of their redox status is considered to affect male fertility and to reduce their fertilizing ability following biotechnological procedures, such as cryopreservation. Cystine (CysS), after incorporation in sperm via SLC7A11 antiporter, has been demonstrated to increase intracellular GSH content, the most important non enzymatic antioxidant. This study was aimed at investigating the role of SLC7A11 antiporter on frozen-thawed stallion sperm ability to respond to in vitro capacitating environment after post-thaw incubation with CysS and/or Sulfasalazine (SS), a specific inhibitor of SLC7A11 antiporter. Viability, motility, immunolocalization of tyrosine phosphorylated proteins and the ability to bind to heterologous zonae pellucidae were evaluated. Thawed sperm from seven stallions (2 ejaculates/stallion) was washed and resuspended in Tyrodes media; each thawed ejaculate was divided in Control (CTR) and 3 samples supplemented with: 0.5 mM Cystine (CysS), 500 mu M Sulfasalazine (SS) and 0.5 mM CysS thorn 500 mu M SS (CysS thorn SS). After 1 h of incubation at 37 degrees C, samples were washed twice, resuspended in capacitating BWW medium and incubated at 38 degrees C under 5% CO2. After 30 and 60 min, sperm motility, viability and tyrosine phosphorylated protein immunolocalization, used as capacitation status index, were evaluated. After 30 min of capacitation, 4 x 10(5) sperm were co-incubated with denuded pig oocytes in capacitation medium for 30 min for the heterologous binding assay. None of the sperm parameters studied (motility, viability and tyrosine phosphorylation) showed any difference respective to control. The number of sperm bound per oocyte (mean +/- SEM) tended to increase in CysS group (44.0 +/- 12.3) respect CTR (40.8 +/- 10.8) while decreased in SS group (32.4 +/- 7.8) (p < 0.01). Moreover, CysS thorn SS group showed a lower binding rate (32.0 +/- 10.0) compared to CysS (p < 0.001). Our results suggest that CysS supplementation of thawed stallion sperm can influence their ability to bind to heterologous zona pellucidae as the inhibition of CysS incorporation by SLC7A11 reduced the number of sperm bound per oocyte. Glyphosate, formulated as glyphosate-based herbicides (GBHs) including the best-known formulation Roundup, is the world's most widely used herbicide. During the last years, the growing and widespread use of GBHs has raised a great concern about the impact of environmental contamination on animal and human health including potential effect on reproductive systems. Using an in vitro model of pig oocyte maturation, we examined the biological impact of both glyphosate and Roundup on female gamete evaluating nuclear maturation, cytoplasmic maturation and developmental competence of oocytes, steroidogenic activity of cumulus cells as well as intracellular levels of glutathione (GSH) and ROS of oocytes. Our results indicate that although exposure to glyphosate and Roundup during in vitro maturation does not affect nuclear maturation and embryo cleavage, it does impair oocyte developmental competence in terms of blastocyst rate and cellularity. Moreover, Roundup at the same glyphosate-equivalent concentrations was shown to be more toxic than pure glyphosate, altering steroidogenesis and increasing oocyte ROS levels, thus confirming that Roundup adjuvants enhance glyphosate toxic effects and/or are biologically active in their side-effect and therefore should be considered and tested as active ingredients.

As the taste receptor for monosodium glutamate (umami) is expressed in both murine and human spermatozoa and the presence of α-gustducin and α-transducin, G proteins involved in the umami taste signaling, has been described in boar germ cells, the aim of this study was to evaluate if monosodium glutamate (MSG) would exert any effect on sperm-oocyte binding, invitro fertilization (IVF) and sperm parameters during invitro induced capacitation. The COVID-19 pandemic and response caused a worrying decline in vaccine uptake around the world. In Italy, the immunization coverage targets set in the 2017-19 National Immunization Prevention Plan (PNPV) have been met only partially. The current public health emergency is likely to have negatively impacted on immunization , with the risk of re-occurrence of Vaccine-Preventable Diseases (VPDs) outbreaks. As flu season approaches, both National Health Institutions and the scientific community in Italy have taken action. Well in advance as compared to previous years, the Ministry of Health released the Circular to launch the 2020-2021 influenza immunization campaign which this year is longer (starting on October 2020) and extends flu vaccine recommendations to more “at risk” subgroups, offered the vaccine free of charge. In addition, some Italian Regions have recently tried to make flu vaccination compulsory for all Healthcare Workers (HCWs). Since 2017, when the law on childhood vaccination in Italy was passed, compulsory vaccination has proved to be a successful strategy towards coverage increase. (www.actabiomedica.it).

Despite the great effort to raise awareness among health promotion, nowadays Public Health policies are not often recognized as important tools. For this reason, the Health in All Policies (HiAP) approach is instrumental in tackling this information gap. In 2018, the US Association of Schools and Programs of Public Health (ASPPH) launched an international campaign called "This is Public Health" (TIPH), whose aim was "to brand public health and raise awareness of how it affects individuals, communities and populations". The Association of Schools of Public Health in the European Region (ASPHER), in coordination with ASPPH, decided to create a European campaign to support and to reproduce the American one, by opening a challenge among the European Public Health Schools. The Schools and Programs of PH of Vita-Salute "San Raffaele" University, Milan, University of Parma, University of Pavia and Politecnico of Milan won this bid. In this "briefing on" we present a report on the Italian project for raising awareness of Public Health among general population and health care personell.

The wide use of glyphosate-based herbicides (GBHs) has become a matter of concern due to its potential harmful effects on human health, including men fertility. This study sought to investigate, using the pig as a model, the impact of pure glyphosate and its most known commercial formulation, Roundup, on sperm function and survival. With this purpose, fresh commercial semen doses were incubated with different concentrations (0-360 μg/mL) of glyphosate (GLY; exp. 1) or Roundup, at the equivalent GLY concentration (exp. 2), at 38 °C for 3 h. Glyphosate at 360 μg/mL significantly (P < 0.05) decreased sperm motility, viability, mitochondrial activity and acrosome integrity but had no detrimental effect at lower doses. On the other hand, Roundup did significantly (P < 0.05) reduce sperm motility at ≥ 5 μg/mL GLY-equivalent concentration; mitochondrial activity at ≥ 25 μg/mL GLYequivalent concentration; and sperm viability and acrosome integrity at ≥ 100 μg/mL GLY-equivalent concentration as early as 1 h of incubation. In a similar fashion, GLY and Roundup did not inflict any detrimental effect on sperm DNA integrity. Dog sperm cryopreservation is gaining importance both in breeding dogs for commercial purposes and for pet animals. Anyway, cryopreservation of mammalian spermatozoa, including dog ones, induces some negative effect on sperm fertility, leading to a lower use of this technique and limiting its widespread use. Therefore, studies to improve the quality of canine semen after cryopreservation could have a relevant impact on both the scientific advancement and the clinical practice. The aim of the present work was to investigate the putative ameliorative effect of Epigallochatechin-3-gallate (EGCG) addition to post thawing medium on dog sperm motility, mitochondrial activity, acrosome integrity and on zona-binding ability (zona binding assay). Spermatozoa were thawed in Tris-fructose-citrate medium supplemented with EGCG (0, 25 and 50 μM) and sperm motility, mitochondrial activity and acrosome integrity were assayed at 0.5, 1.5, 3 and 6 h after post thawing incubation at 37 °C. An aliquot of semen from each treatment group after 1.5 h post thawing incubation was washed and used to perform heterologous (using porcine oocytes) or homologous zona binding assay. The results obtained showed that no significant effect is exerted by EGCG on sperm parameters analysed neither at 0.5, 1.5, 3 or 6 h after thawing excepting for the reduction of the percentage of live cells with active mitochondria at the higher dose at 6 h; furthermore, both homologous or heterologous zona binding ability, was not influenced by EGCG. In conclusion, EGCG supplementation to thawing medium does not improve dog sperm quality or zona binding capacity.

Tannins have been demonstrated to have antioxidant and various health benefit properties. The aim of this study was to determine the effect of an ethanol extract (TRE) of a commercial oenological tannin (Quercus robur toasted oak wood, Tan'Activ R ® ) on female gamete using an in vitro model of pig oocyte maturation (IVM) and examining nuclear maturation, cytoplasmic maturation, intracellular GSH and ROS levels and cumulus cell steroidogenesis. To this aim, during IVM performed in medium either supplemented (IVM A) or not supplemented (IVM B) with cysteine and β-mercaptoethanol, TRE was added at different concentrations (0, 1, 5, 10, 20 μg/ml). The addition of TRE at all the concentration tested to either IVM A or IVM B, did not influence oocyte nuclear maturation. When IVM was performed in IVM A, no effect was induced on cytoplasmic maturation by TRE at the concentration of 1, 5 and 10 μg/ml, while TRE 20 μg/ml significantly reduced the penetration rate after IVF (p < 0.05) and the blastocyst rate after parthenogenetic activation (p < 0.01). Oocyte maturation in IVM B, compared to IVM A group, decreased GSH (p < 0.001) and increased ROS (p < 0.01) intracellular levels and in turn impaired oocyte cytoplasmic maturation reducing the ability to sustain male pronuclear formation after IVM (p < 0.001) and the developmental competence after parthenogenetic activation (p < 0.001). TRE supplementation to IVM B significantly reduced ROS production (5, 10, 20 μg/ml TRE) to levels similar to IVM A group, and increased GSH levels (10, 20 μg/ml TRE) compared to IVM B (p < 0.05) without reaching those of IVM A group. TRE supplementation to IVM B at the concentrations of 1, 5 and 10 μg/ml significantly improved (p < 0.001) oocyte cytoplasmic maturation enhancing the ability to sustain male pronuclear formation without reaching, however, IVM A group levels. TRE addition at all the concentration tested to both IVM A and IVM B, did not induce any effect on E2 and P4 secretion by cumulus cells suggesting that the biological effect of the ethanol extract is not exerted thought a modulation of cumulus cell steroidogenesis. In conclusion, TRE, thanks to its antioxidant activity, was partially able to reduce the negative effect of the absence of cysteine and β-mercaptoethanol in IVM B, while TRE at high concentration in IVM A was detrimental for oocyte cytoplasmic maturation underlying the importance of maintaining a balanced redox environment during oocyte maturation.

Rising interest in beneficial health properties of fruit and achievement of the best organoleptic qualities in relation to ripening times, led to focus the attention on peach fruits. Phenolic composition in peach fruit changes during ripening process and influences the final quality. A collection field has been set-up, where different peach genotypes of interest (melting flesh=MF and not melting flesh=NMF) have been grown. After pit hardening, fruits growth and ripening have been monitored, the latter using a non-destructive portable instrument (DA-meter) providing an index linked with the chlorophyll degradation thus related to ripening degree. Classical destructive quality analysis was also carried out during the sampling. The general aim of this research has been to investigate about the possible relation between phenolic compounds and ripening stages in peach fruit. Differences in the amount of phenolic compounds in flesh and peel during the maturation process have been evaluated. Quantification of the most important phenolic compounds was obtained using a LC-MS/MS system. Results reported that the DA index (I(DA)) decreased reaching the ripening stage, in both peach genotypes (MF and NMF) and it also showed a relationship with ethylene production of fruits, demonstrating a good reliability of the instrument. Preliminary results showed that from ‘Oro A’s phenolic analysis in pre-climacteric stage, the amount of quercetin 3-rutinoside and cyanidin 3-glucoside in the peel resulted substantially higher than in the flesh. Among the main flavan-3-ols, catechin resulted in higher content compared to epicatechin, in agreement with previous works. Among hydroxycinnamic acids, chlorogenic acid resulted in higher amount than neo-chlorogenic as reported by other studies.

On August 24th, 2016, a severe, very long seismic sequence started in Central Italy. It was characterized by nine major shocks M5+, two of which with moment magnitude Mw 6.0 (August 24th, 2016) and 6.5 (October 30th, 2016). A complex seismogenic fault system was activated, with the rupture of several segments. The affected area, which develops in NNW-SSE direction along the Apennines, was very large, due to both the large magnitude values and the distance among the epicenters of the nine major shocks. The maximum observed (cumulated) intensity was XI in both MCS and EMS scales. After 1 year, 78,500 seismic events had been recorded by the National Institute of Geophysics and Volcanology national seismic network. 299 people lost their life, all due to the first main shock. Devastating damage was experienced by buildings, cultural heritage, roads and other lifelines, resulting in huge economical direct losses. The emergency response was coordinated, according to the Law 225/1992, by the Italian National Department of Civil Protection. The main scientific features of the sequence and the main technical emergency activities are shown, discussed and, when possible, compared to the main recent Italian earthquakes, i.e., 1997 Umbria-Marche, the 2009 Abruzzo and 2012 Emilia earthquakes, pointing out analogies and differences.
