Genetics ,QH426-470 - Abstract
A comparative cytogenetic analysis was carried out in five species of a monophyletic clade of neotropical Cichlasomatine cichlids, namely Cleithracara maronii Steindachner, 1881, Ivanacara adoketa (Kullander & Prada-Pedreros, 1993), Nannacara anomala Regan, 1905, N. aureocephalus Allgayer, 1983 and N. taenia Regan, 1912. Karyotypes and other chromosomal characteristics were revealed by CDD banding and mapped onto the phylogenetic hypothesis based on molecular analyses of four genes, namely cyt b, 16S rRNA, S7 and RAG1. The diploid numbers of chromosomes ranged from 44 to 50, karyotypes were composed predominantly of monoarmed chromosomes and one to three pairs of CMA3 signal were observed. The results showed evolutionary reduction in this monophyletic clade and the cytogenetic mechanisms (fissions/fusions) were hypothesized and discussed.

In the Czech Republic, major changes have happened in the field of infant institutional care in connection with amendment to the Act on Social and Legal Protection of Children and the Act on Family in 2013. Efforts to close down institutional facilities for children up to 3 years are in connection to deinstitutionalization, which has become societal philosophy of transformation of institutional care in the last decade. Students of helping professions are educated in modern trends, such as deinstitutionalization, because they should become “holders and implementers” of positive changes based on respect for human rights and freedoms. In connection with that, we focused on opinions on the closing down of infant homes and children’s homes for children up to 3 years among students of nursing fields and social pedagogy. Using the quantitative method of a questionnaire, we obtained responses from 416 respondents. One of the most interesting findings is that only 10 % of students agree with the idea of closing down these institutional facilities. There are also differences based on field of study. Students of social pedagogy on average agree more with this idea than students of nursing fields. This could be based on differences in curriculum.

Plaque-induced gingivitis is the most prevalent periodontal disease associated with pathogenic biofilms. The host immune system responds to pathogens through pattern recognition receptors (PRRs), such as Toll-like receptors (TLRs) and their co-receptor cluster of differentiation 14 (CD14).This study investigated the association between the functional polymorphism in the CD14 gene and the dental plaque microbiota in children with gingivitis.A total of 590 unrelated children (307 with plaque-induced gingivitis and 283 controls, aged 13-15 years) were enrolled in this case-control study. Dental plaque was processed using a ParoCheckGingivitis was detected in 64.2% of boys and 35.8% of girls (P .001). Children with gingivitis had a significantly higher occurrence of dental caries (P .001). No significant differences in the CD14 -260C/T allele and genotype distribution among individuals with or without gingivitis in the whole cohort were found. Children with gingivitis and P gingivalis, however, were significantly more frequent carriers of the CT and TT genotypes than children with gingivitis without P gingivalis or healthy controls (P .05).The CD14 -260C/T polymorphism acts in cooperation with P gingivalis to trigger plaque-induced gingivitis in Czech children.

A set of 92 genetic resources of spring barley split into groups according to the areas of origin was studied in terms of grain malting quality. The following malting parameters were monitored in this study: nitrogen content in barley grain, malt extract, relative extract at 45 °C, Kolbach index, diastatic power, apparent final attenuation, friability, β-glucans in wort, haze of wort measured at the angles of 15° and 90°. In the studied set, the largest variability was found in the haze of wort (90°), the smallest in the apparent final attenuation. The optimal values of the extract content in the malt dry matter according to the malting quality index were only detected in the genotypes originating from Europe. The group of the genetic resources from South America and Australia, together with the genotypes from the Middle East and Africa, differed statistically significantly in the values of the Kolbach index from other groups. The two-row genotypes differed statistically significantly in the values of the malt extract content, relative extract at 45 °C, Kolbach index, diastatic power, friability and β-glucan content in wort. Although the European and non-European groups differed in a number of traits, the non-European groups also included genetic resources that in their malting parameters approached or equaled the European malting varieties (such as Psaknon). In this study, several significant correlations were found; the strongest correlations were between friability and malt extract (r = 0.85), friability and Kolbach index (r = 0.84).

Nearby flanking objects degrade visual resolution. If the flankers are similar to the acuity target, this influence is called crowding (CW), whereas if the flanking stimuli are simple bars then the phenomenon is known as contour interaction (CI). The aim of this study was to compare the influence of the number and position of flankers on foveal CW and CI to investigate possible differences in mechanism of these two effects. Five normal observers viewed single, foveally presented Sloan letters surrounded by 1, 2 or 4 flankers (either a Sloan letter or one-stroke-width bars), presented at several edge-to-edge separations. Single flankers were presented in the right, left, top or bottom position, 2 flankers were placed equally to the right and left or top and bottom of the central target, and 4 flankers were equally spaced in all four directions. Percent correct letter identification was determined for each type, number, position and separation of flankers and confusion matrices were constructed for separations equal to 20% and 100% letter width. Increasing the number of flankers caused an increase in the magnitude of both phenomena. CW showed a greater magnitude than CI for higher numbers of flankers. Analysis of confusion matrices suggests that in addition to the edge-to-edge interaction that appears to mediate CI, letter substitution and feature pooling contribute significantly to CW when higher numbers of flankers are presented. Foveal CW is more strongly influenced by an increase in the number of flankers than CI, which can be explained by the presence of additional interaction effects.

Allium vegetables belong to the most widespread and commonly grown vegetables in Slovakia. In this study, we dealt with the content of selected heavy metals (Zn, Mn, Cr, Cu, Ni, Pb, and Cd) in cultivated garlic (Zahorský, Lukan, Makoi), onion (Amika, Red Matte, Diamant), and leek (Bavaria, Golem, Tango). We determined the total content of heavy metals (aqua regia), and available-mobile forms of heavy metals (1 M NH4NO3) in soils. Analytical final procedure was flame AAS. The results were compared with the limit values given by the Law no. 220/2004 (valid in the SR) as well as threshold values proposed by the European Commission (EC) (2006). Gained result showed that the total content of Cd (1.26 mg.kg-1) as well as the available mobile forms of Cd (0.16 mg.kg-1) and Pb (0.64 mg.kg-1) was exceeded on the soil on which Allium species were grown. Determined contents of heavy metals in garlic were in the range of 0.03–0.07 mg Cd.kg-1 FM (fresh matter), 0.02–0.11 mg Pb.kg-1 FM (fresh matter), in onion ranged from ND (not detected) to 0.03 mg Cd.kg-1 FM (fresh matter), 0.09–0.2 mg Pb.kg-1 FM (fresh matter), and in leek were in the range of, 0.07–0.13 mg Cd.kg-1 FM (fresh matter), and 0.3–0.5 mg Pb.kg-1 FM (fresh matter). Based on the obtained BAF values, we can conclude that the genus Allium does not belong to the accumulator of evaluated hazardous metals. Safe food production requires constant monitoring of hazardous metals in raw materials of plant origin, as these metals can have serious negative effects on human health.

The objective of this paper was to examine the total polyphenols, vitamin C and capsaicin contents of pepper (varieties: Garam F1, Alma, Poseidon, Promontor, Slovakia, Claudius, Bhut Jolokia, Serrano, and Candlelight) and their correlation with the antioxidant activity by using the high performance liquid chromatography and Folin-Ciocalteu method. The limit of detection for capsaicin was 0.09 µg.g−1.Vitamin C and capsaicin ranged from 95.76 to 2,139 and from 44.40 to 2,224 μg.g−1 dry matter (DM), respectively. The determined total polyphenol content (TPC) ranged from 4,230 to 9,939 mg GAE.kg−1 DM and the values of antioxidant activity (AA) determined by using DPPH •, FRAP and ABTS • + methods were in range 19.48–91.81, 19.08–61.46 and 15.72–65.70 μmol TE.g−1 DM, respectively. Based on the statistical evaluation, there was a high correlation between vitamin C content and antioxidant activity of peppers. The obtained results suggest that due to the composition of bioactive compounds, especially antioxidants, the consumption of pepper fruits is an ideal choice for the prevention of the cell damage.

This chapter introduces buckwheat as a possible raw material for the production of designed foods. It includes the description of common buckwheat as a source of basic nutrients for food production and gives specificities of buckwheat as a source of biologically active substances. Processed buckwheat seeds are important from the point of view of rational nutrition as a source of energy, carbohydrates, fibre, proteins, lipids, vitamins, and minerals. Buckwheat has also other nutritional advantages, especially the interesting content of polyphenolic compounds: phenolic acids, flavonoids, especially rutin, which are characterised by high antioxidant activity. This chapter describes how buckwheat can be processed into food products and discusses the results of the application of buckwheat to bread and pasta. Moreover, it includes the results of the clinical study. Based on the identified technological and sensory properties of bread products obtained during the baking experiment, the chapter summarises recommendations on the suitable added amount of buckwheat to get satisfactory results. Concerning pasta from buckwheat, it had very good technological, nutritional, and sensory qualities. The chapter concludes that, on the basis of findings, buckwheat is a raw material suitable for the production of designed foods.

We study turbulent Rayleigh-Bénard convection over four decades of Rayleigh numbers 4×10^{8}Ra2×10^{12}, while harmonically modulating the temperatures of the plates of our cylindrical cell. We probe the flow by temperature sensors placed in the cell interior and embedded in the highly conducting copper plates and detect thermal waves propagating at modulation frequency in the bulk of the convective flow. We confirm the recent numerical prediction [Yang et al., Phys. Rev. - Abstract
The aim of the work was to evaluate the safety of bee drone brood in terms of the risk of possible contamination with heavy metals, microbial contamination, and sensory acceptability. Bee drone brood dried at a temperature of 80 degrees C in a hot air oven (DBO) and lyophilized drone brood (DBL) were used. Sensory evaluation was performed on an oatmeal ball (O-B) and drone brood ball (DBO-B). For the determination of the minerals and other elements in DBO, spectrometric analysis was used. The content was as follows: Fe 60.87 mu g/g, Cu 54.83 mu g/g, Zn 257 mu g/g, Ag 82.7 mu g/g, Pb 2.1 mu g/g, Au 1.2 mu g/g, Ca 1336 mu g/g, Sr 3.13 mu g/g, Zr 1.67 mu g/g, Bi 3.87 mu g/g, Br 1.95 mu g/g. The detected content of lead in dry matter (2.1 mu g/g) was high above the limit set for crustaceans (0.4 mu g/g). By comparing microbial assay values, no statistically significant difference between freshly dried DBO and dried DBO after a month of storage was detected. Bee drone brood contains microbes far below the legal limits and it is a good source of calcium, iron, and zinc. Except for the juiciness, consumers rated O-B better than DBO-B. A statistically significant difference (p < 0.05) was found only in taste., Cílem práce bylo zhodnotit bezpečnost trubčího včelího plodu z hlediska rizika možné kontaminace těžkými kovy, mikrobiální kontaminace a senzorické přijatelnosti.

- Abstract
The pseudo 3D hierarchical structure mimicking in vivo microenvironment was prepared by phase separation on tissue culture plastic. For surface treatment, time-sequenced dosing of the solvent mixture with various concentrations of polymer component was used. The experiments showed that hierarchically structured surfaces with macro, meso and micro pores can be prepared with multi-step phase separation processes. Changes in polystyrene surface topography were characterized by atomic force microscopy, scanning electron microscopy and contact profilometry. The cell proliferation and changes in cell morphology were tested on the prepared structured surfaces. Four types of cell lines were used for the determination of impact of the 3D architecture on the cell behavior, namely the mouse embryonic fibroblast, human lung carcinoma, primary human keratinocyte and mouse embryonic stem cells. The increase of proliferation of embryonic stem cells and mouse fibroblasts was the most remarkable. Moreover, the embryonic stem cells express different morphology when cultured on the structured surface. The acquired findings expand the current state of knowledge in the field of cell behavior on structured surfaces and bring new technological procedures leading to their preparation without the use of problematic temporary templates or additives.

- Abstract
Essential features of well-designed materials intended for 3D bioprinting via microextrusion are the appropriate rheological behavior and cell-friendly environment. Despite the rapid development, few materials are utilizable as bioinks. The aim of our work was to design a novel cytocompatible material facilitating extrusion-based 3D printing while maintaining a relatively simple and straightforward preparation process without the need for harsh chemicals or radiation. Specifically, hydrogels were prepared from gelatines coming from three sources-bovine, rabbit, and chicken-cross-linked by dextran polyaldehyde. The influence of dextran concentration on the properties of hydrogels was studied. Rheological measurements not only confirmed the strong shear-thinning behavior of prepared inks but were also used for capturing cross-linking reaction kinetics and demonstrated quick achievement of gelation point (in most cases < 3 min). Their viscoelastic properties allowed satisfactory extrusion, forming a self-supported multi-layered uniformly porous structure. All gelatin-based hydrogels were non-cytototoxic. Homogeneous cells distribution within the printed scaffold was confirmed by fluorescence confocal microscopy. In addition, no disruption of cells structure was observed. The results demonstrate the great potential of the presented hydrogels for applications related to 3D bioprinting.

- Abstract
Present-day people from England and Wales have more ancestry derived from early European farmers (EEF) than did people of the Early Bronze Age1. To understand this, here we generated genome-wide data from 793 individuals, increasing data from the Middle to the Late Bronze Age and Iron Age in Britain by 12-fold, and western and central Europe by 3.5-fold. Between 1000 and 875 BC, EEF ancestry increased in southern Britain (England and Wales) but not northern Britain (Scotland) due to incorporation of migrants who arrived at this time and over previous centuries, and who were genetically most similar to ancient individuals from France. These migrants contributed about half the ancestry of people of England and Wales from the Iron Age, thereby creating a plausible vector for the spread of early Celtic languages into Britain. These patterns are part of a broader trend of EEF ancestry becoming more similar across central and western Europe in the Middle to the Late Bronze Age, coincident with archaeological evidence of intensified cultural exchange2-6. There was comparatively less gene flow from continental Europe during the Iron Age, and the independent genetic trajectory in Britain is also reflected in the rise of the allele conferring lactase persistence to approximately 50% by this time compared to approximately 7% in central Europe where it rose rapidly in frequency only a millennium later. This suggests that dairy products were used in qualitatively different ways in Britain and in central Europe over this period.

- Abstract
Smart hydrogels based on natural polymers present an opportunity to fabricate responsive scaffolds that provide an immediate and reversible reaction to a given stimulus. Modulation of mechanical characteristics is especially interesting in myocyte cultivation, and can be achieved by magnetically controlled stiffening. Here, hyaluronan hydrogels with carbonyl iron particles as a magnetic filler are prepared in a low-toxicity process. Desired mechanical behaviour is achieved using a combination of two cross-linking routes-dynamic Schiff base linkages and ionic cross-linking. We found that gelation time is greatly affected by polymer chain conformation. This factor can surpass the influence of the number of reactive sites, shortening gelation from 5 h to 20 min. Ionic cross-linking efficiency increased with the number of carboxyl groups and led to the storage modulus reaching 10(3) Pa compared to 10(1) Pa-10(2) Pa for gels cross-linked with only Schiff bases. Furthermore, the ability of magnetic particles to induce significant stiffening of the hydrogel through the magnetorheological effect is confirmed, as a 10(3)-times higher storage modulus is achieved in an external magnetic field of 842 kA.m(-1). Finally, cytotoxicity testing confirms the ability to produce hydrogels that provide over 75% relative cell viability. Therefore, dual cross-linked hyaluronan-based magneto-responsive hydrogels present a potential material for on-demand mechanically tunable scaffolds usable in myocyte cultivation.

In this first part of a two-paper set on the elastic properties of mullite and mullite ceramics the current literature is reviewed and, based on the available monocrystal data, a mutually consistent average set of elastic constants for polycrystalline mullite materials at room temperature is calculated and optimized via minimization of the total average relative root mean square (RMS) deviation. Based on the results of this paper, the recommended values of adiabatic elastic constants are 224.9 GPa for the Young’s modulus, 87.8 GPa for the shear modulus, 170.9 GPa for the bulk modulus and 0.281 for the Poisson ratio. The RMS deviation of this set is lower than for any other set, and thus these values exhibit a high degree of mutual consistency with respect to the elasticity standard relations. Micromechanical upper bounds and model relations are summarized for describing the porosity dependence of elastic moduli, and a simple relation is used to estimate the difference between adiabatic and isothermal elastic constants. The results show that this difference increases slightly with temperature, but is not larger than 4 % even at 1500°C. In particular, for the Young’s modulus the difference remains below 0.5 %. That means, the difference between adiabatic and isothermal Young’s moduli will usually be unmeasurably small and negligible in practice.

Cultivar is one of the most important internal factors affecting polyphenol concentration in the plants. However, influence of the grown locality, climate conditions and way of cultivation belong to important external factors. In our experiment the influence of different nitrogen doses (0 - 40 - 80 - 120 - 160 - 240 kg N.ha-1) applied in the form of Vermikompost on the total polyphenol content and derived total antioxidant activity in cv. Sorento were investigated. While in the 1st - 5th variants the determined polyphenol content in dry mater of potato tubers decreased from 399.2 to 70.40 mg.kg-1, in the 6th variant that was twice higher in comparison to the 5th variants (135.6 mg.kg-1). The statistically significant differences in values of total polyphenol content between variants (polynomial function of 2nd degree) were confirmed. The study also confirmed a strong statistical correlation between the content of polyphenols and the content of antioxidant activity has been confirmed (sign. F: 3.24E-10). The highest value of antioxidant activity was observed in the first variant. From the first to the fifth variant (7.62 - 4.84%), the value of antioxidant activity was decreasing and in the sixth variant this value increased to 6.31%.

There are many different types of datasets represented by maps in thematic cartography. It is possible to represent features and phenomenon referencing to area as well as to point. Furthermore, it is possible to represent qualitative data as well as quantitative data. There are many different ways to represent them. One of the most important and most used cartographic symbols of lettering is colour, resp. colour scales, which are chosen according to shown data. This article is focused on the perception of colour scales in relation to character of data. For experiment, a new questionnaire was created on the basis of colour scales classification. This questionnaire was posed to the students of years 1 and 2 of high school in order to find out how they perceive the colour scales. The study analyzes three main questions; whether students differentiate qualitative and quantitative datasets and corresponding colour scales, whether they prefer representing of features by context colours or by the colour they like more and whether they are familiar with principles of diverging colour scales. When processing it, the correlation between certain agents and the answers was established.

The aim of the study was to evaluate the effect of the season on incidence of footpad dermatitis and to determine the effect of footpad dermatitis on broilers performance expressed by EPEF (European Production Efficiency Factor). The incidence of footpad dermatitis was evaluated during the year. Daily mortality, weights and feed consumption were observed too. Scoring of the feet was done in slaughterhouse according to six-point scale (0–5) Ask (2010). There were classified between 1200 to 1500 shanks from each flock. To facilitate the evaluation of the feet damage the numbers in scoring groups were summarized as follows: negligible damage (0+1), intermediate damage (2+3) and severe damage (4+5). The highest incidence of negligible damage was found in the summer (34.0%, P < 0.05) followed by autumn (13.1%). The most severe damage of the feet was found in the spring (83.2%) followed by winter (72.4%). Only 12% of the feet were negligibly damaged. Almost 70% of the feet were severe damaged. The significant correlation between the feet damage and EPEF was not confirmed (P > 0.05). The study showed that footpad dermatitis is a severe problem. However chickens with severe feet damage were able to achieve excellent performance results.

This literature review aims to assess the evolution of performance monitoring articles in relation to the business strategy based on an overview of articles indexed in the WoS and Scopus databases. The initial dataset of articles was obtained based on carefully developed keywords. In the second phase, we restricted the selection to business and management journal articles and reviews written in English before 2019. The authors simultaneously reviewed the abstracts of all 571 articles in the third phase of the study. These were subsequently re-read and limited; thus, the final dataset for further analysis includes 157 articles published between 1983 and 2019. The most important result is that the field of business strategy and performance is not subject to trends. While in the field of business strategy the literature is still dominated by generic strategies according to Porter and the strategy typology according to Miles and Snow, in the field of performance measurement we identified the market position (sales volume, growth in sales volume, etc.) and profitability (profit margin, return on assets, etc.) as the dominant method. Implications for Central European audience: Despite a considerable amount of work focusing on bibliographic analysis, there remains a lack of literature on the link between strategy and performance. The paper brings potential value for the Central European audience by drawing attention to the systematisation of knowledge in the field of performance articles and its trends in relation to the field of business strategy. The overview of the theoretical frameworks contributes to a better orientation of the current state of the art and creates opportunities for future research. [ABSTRACT FROM AUTHOR]

In terms of the potential effects of the natural (geogenic) and anthropogenic agricultural environment on the crop quality, it must be remembered that it is necessary to maintain an uncontaminated environment with high soil production potential. The objective of this research was to analyse and evaluate soil under sugar beet, the compost produced and the soil affected by the addition of compost, in order to determine the Cd, NO
3 - -N, NH4 + -N contents in topsoil and soil with added compost (1:1 ratio), to evaluate the statistical significance of Cd in relation to the fertile forms of nitrogen, and to find out whether compost influences the monitored parameters. The analyses were carried out using the Atomic Absorption Spectrometry and Colorimetric methods. The obtained results were evaluated statistically using the SAS 9.4 software method and Spearman's correlation coefficient. The results showed that NO3 - -N contents in soil reached 14.73 mg·kg-1, NH4 + -N 9.50 mg·kg-1, Nin 23.05 mg·kg-1, Cd 2.96 mg·kg-1, soil + compost (1:1 ratio) NO3 - -N 23.93 mg·kg-1, NH4 + -N 26.42 mg·kg-1, Nin 42.63 mg·kg-1, Cd 2.71 mg·kg-1 dry matter. Very high statistical dependence (P < 0.001) was observed for all inorganic fertile forms of nitrogen. High statistical dependence (P < 0.01) was observed for soil Cd to soil NO3 - -N, soil + compost Cd to soil NO3 - -N, soil + compost Cd to soil Nin. Compost had a statistically highly significant effect on Cd in relation to forms of nitrogen, reducing Cd concentration levels and increasing the proportion of inorganic fertile forms of nitrogen.
