Objective The study was aimed to investigate the effect and its mechanism of cisplatin on the expression of programmed death ligand-1 (PD-L1) in TC-1 cell surface, in order to provide an experimental basis to improve the therapeutic efficacy for cervical cancer. Methods The concentration of cisplatin was optimized by MTT method through detecting the cells proliferation vitality. Then, the expressions of PD-L1 in TC-1 cells treated by cisplatin and PKD blocker GÖ6976 were measured by immunohistochemistry. Results (1) The proliferation of TC-1 cells were inhibited by cisplatin with the IC50 value of 25.5 µg/mL; (2) The expression of PD-L1 in TC-1 cell surface were (64.96 ± 11.55) % in cisplatin group, which was (39.59 ± 5.99) % in negative control group. There was a significant difference between the two groups (P < 0.05); (3) The expression of PD-L1 in TC-1 cell surface treated by cisplatin combined with PKD blocker GÖ6976 were (61.56 ± 3.43) % after 24 h, which were (79.54 ± 5.09) % in cisplatin group, and the difference was significant (P < 0.05). Conclusion Cisplatin can promote the expression of PD-L1 in TC-1 cell surface which could be significantly inhibited by the PKD pathway blocked. Asparaginase-associated pancreatitis (AAP) is a common and fatal complication after ASNase treatment in acute lymphoblastic leukemia(ALL). Here, a total of 1063 pediatric ALL patients treated with SCCLG-ALL-2016 regimen were collected since October 2016 to June 2020, including 35 patients with AAP. The clinical characteristics of AAP and non-AAP patients were compared. In AAP patients, the possible factors that affected the recurrence of AAP were analyzed, and the possible risk factors related to ALL-relapse were discussed. The results showed that age was a risk factor (P =.017) that affect the occurrence of AAP. In AAP patients, AAP tended to develop after the second use of PEG-ASNase (25.71%). In the follow-up chemotherapy, 17 patients re-exposed to ASNase and 7 cases developed AAP again with a percentage was 41.2%. There were no special factors that related with the recurrence of AAP. This study also found no association between the occurrence of AAP and prognosis of ALL, with the 4-year incidence of ALL relapse in AAP and non-AAP patients were 15.9% v.s.11.7% (HR: 1.009, 95% CI:0.370–2.752, P =.986), and there were no special factors that related with the ALL relapse among AAP patients. Based on the above results, the occurrence of AAP is related to age and should be vigilant after the second use of PEG-ASNase after use in pediatric ALL patients. The strong nonlinear behavior usually exists in rotor systems supported by oil-film journal bearings. In this paper, the partial derivative method is extended to the second-order approximate extent to predict the nonlinear dynamic stiffness and damping coefficients of finite-long journal bearings. And the nonlinear oil-film forces approximately represented by dynamic coefficients are used to analyze nonlinear dynamic performance of a symmetrical flexible rotor-bearing system via the journal orbit, phase portrait and Poincaré map. The effects of mass eccentricity on dynamic behaviors of rotor system are mainly investigated. Moreover, the computational method of nonlinear dynamic coefficients of infinite-short bearing is presented. The nonlinear oil-film forces model of finite-long bearing is validated by comparing the numerical results with those obtained by an infinite-short bearing-rotor system model. The results show that the representation method of nonlinear oil-film forces by dynamic coefficients has universal applicability and allows one easily to conduct the nonlinear dynamic analysis of rotor systems. A possible association of glutathione S-transferase T1 (GSTT1) null/presence gene polymorphism and an increased risk of developing gastric carcinoma is still unclear and hotly debated. This investigation was performed to assess the association of the GSTT1 null/presence gene polymorphism with the risk of gastric carcinoma via a meta-analysis to increase sample size and statistical significance. PubMed, Cochrane Library and CBM-disc (China Biological Medicine Database) were searched on March 1, 2013, association reports were identified, and eligible studies were recruited and synthesized. Fifty-two reports were found to be suitable for this meta-analysis for the association of the GSTT1 null genotype with gastric carcinoma risk. The results showed that there was a significantly increased gastric carcinoma risk when the GSTT1 null genotype was present in the overall population (OR 1.21, 95 % CI 1.11-1.32, P < 0.0001), Caucasians (OR 1.25, 95 % CI 1.05-1.48, P = 0.01), East-Asians (OR 1.18, 95 % CI 1.06-1.31, P = 0.003), and Chinese (OR 1.24, 95 % CI 1.07-1.44, P = 0.005). However, no statistically relevant association could be established for the Indian ethnic group (OR 1.33, 95 % CI 0.94-1.90, P = 0.11). Abstract: In the present study, we designed a peptide of the second outer loop of porcine β2-AR, and expressed the fusion protein in E. coli. Polyantibody was prepared in New Zealand White rabbit. The biological activity of the antibodies was measured both in vitro and in vivo. The antibody titer was 1:4,096,000. Western blots revealed that the prepared antibodies specifically reacted with correspondent antigens. Intracellular cAMP level of porcine skeletal muscle cells was significantly elevated after treatment of clenbuterol and anti-β2-AR/K31A relative to negative serum (P<0.01). ICI118551, blocker of β2-AR, decreased the elevation of intracellular cAMP (P<0.01). MAP kinase was phosphorylated after being treated with clenbuterol and anti-β2-AR/K31A. The backfat thickness decreased by 13% after immunization with the fusion protein. These results indicate that the polyantibody exerted a similar activity to its ligand in porcine model, and may be developed as a new agonist to β2-AR. [Copyright &y& Elsevier]

Many studies have demonstrated that apoptosis play an important role in cerebral ischemic pathogenesis and may represent a target for treatment. Neuroprotective effect of quercetin has been shown in a variety of brain injury models including ischemia/reperfusion. It is not clear whether BDNF-TrkB-PI3K/Akt signaling pathway mediates the neuroprotection of quercetin, though there has been some reports on the quercetin increased brain-derived neurotrophic factor (BDNF) level in brain injury models. We therefore first examined the neurological function, infarct volume and cell apoptosis in quercetin treated middle cerebral artery occlusion (MCAO) rats. Then the protein expression of BDNF, cleaved caspase-3 and p-Akt were evaluated in either the absence or presence of PI3K inhibitor (LY294002) or tropomyosin receptor kinase B (TrkB) receptor antagonist (K252a) by immunohistochemistry staining and western blotting. Quercetin significantly improved neurological function, while it decreased the infarct volume and the number of TdT mediated dUTP nick end labeling positive cells in MCAO rats. The protein expression of BDNF, TrkB and p-Akt also increased in the quercetin treated rats. However, treatment with LY294002 or K252a reversed the quercetin-induced increase of BDNF and p-Akt proteins and decrease of cleaved caspase-3 protein in focal cerebral ischemia rats. These results demonstrate that quercetin can decrease cell apoptosis in the focal cerebral ischemia rat brain and the mechanism may be related to the activation of BDNF-TrkB-PI3K/Akt signaling pathway. The V-system is a complete orthogonal system of functions defined on the interval [0, 1], generated by finite Legendre polynomials and the dilation and translation of a function generator, which consists of a finite number of continuous and discontinuous functions. The V-system has interesting properties, such as orthogonality, symmetry, completeness and short compact support. It is shown in this paper that the V-system is essentially a special multi-wavelet basis. As a result, some basic properties of the V-system are established through the well-developed theory of multi-wavelets. From this point of view, more other V-systems are constructed. Background: Matrix metalloproteinase-7 (MMP-7) may play an important role in the development of vulnerable carotid plaque. An A-to-G transition (-181A/G) in the promoter region of MMP7 is functional in vitro by altering the transcriptional activity of the gene. The aim of this study was to investigate the association between the MMP7 -181A/G polymorphism and vulnerable carotid plaque formation. Methods: The authors enrolled 641 patients and divided them into three groups according to the carotid ultrasound examination: vulnerable plaque group (n=118), stable plaque group (n=385) and no plaque group (n=138). Traditional atherosclerosis risk factors were recorded and the MMP7 -181A/G polymorphism was genotyped by polymerase chain reaction-restriction fragment length polymorphism. Results: In the multinomial logistic regression analysis, compared to the no plaque group, no relationship between MMP7 -181AG+GG genotypes and stable carotid plaque was observed [odds ratio (OR) 1.50; p=0.239]. However, the frequency of AG+GG genotypes was significantly higher in the vulnerable plaque group (OR 2.74; p=0.008). Age was a risk factor for plaque formation, while statin treatment can reduce the prevalence of atherosclerotic plaque. Additionally, using binary logistic regression analysis between the stable and vulnerable plaque groups, this MMP7 polymor phism was associated with vulnerable plaque independently of other factors [OR 1.83; 95% confidence interval 1.08- 3.11; p=0.026]. Conclusions: The MMP7 -181A/G polymorphism is associated with the development of vulnerable carotid plaques. Abstract: Polymorphisms in the Fc receptor–like 3 (FCRL3) gene have been reported to be associated with rheumatoid arthritis (RA) in Japanese populations. However subsequent studies have yielded conflicting results. Hence the aim of present study was to clarify whether these genetic variants in FCRL3 gene are associated with RA in a Chinese population. We conducted a case-control study of 234 RA patients and 260 controls by genotyping four polymorphisms in FCRL3. The genotype and allele distributions of four polymorphisms were significantly different in RA patients compared with controls (uncorrected p = 0.021 and p = 0.031; 0.027 and 0.008; 0.028 and 0.042; and 0.019 and 0.029, respectively). The FCRL3-169 C allele was significantly associated with rheumatoid factor (RF) and anti–cyclic citrullinated peptide (anti-CCP)–positive RA, but no association was detected for RF and anti-CCP–negative RA. Furthermore, the frequency of the –169C allele was increased disproportionately in female patients, and the resulting odds ratio for female homozygote was increased to 2.375 (uncorrected p = 0.006). Haplotype analysis showed that the most common haplotype TGGG was associated with decreased risk of RA (uncorrected p = 0.001, odds ratio = 0.656). However CACA appeared to be a risk haplotype for RA cases (uncorrected p = 0.031, odds ratio = 1.398). Taken together, these results suggest that FCRL3 polymorphisms and haplotypes may contribute to genetic susceptibility to RA in Chinese population. [Copyright &y& Elsevier]

The article presents an experiment directed toward the enhancement of the foreign growth hormone releasing factor (GRF) gene expression efficiency in skeletal muscle of mice through in vivo electroporation. GRF gene plasmids were injected ino skeletal muscle of mouse by electroporation in vivo. It resulted in the accumulative increase in body weights as the dosage increased. The experiment suggests that in vivo electroporation is able to enhance GRF expression in mouse skeletal muscle.

The article examines the tissue localization and development of B lymphocytes of chickens in Changchun, China. Researchers evaluated the caecal tonsil of chickens from embryos to 21-day-old and found that periellipsoid lymphatic sheath (PELS) and germinal center gradually matured when B lymphocytes increased They also discovered that humoral immune spleen function strengthen gradually from hatching to 21 day old.

Abstract: Triclosan is an antimicrobial agent widely used in many contemporary consumer and health care products. This study assayed phytotoxicity of triclosan using plant growth tests, soil microbial activity using soil respiration and phosphatase activity tests and soil microbial functional diversity using the Biolog ECO plates. The results showed that triclosan inhibited plant growth in soil, with rice seeds being more sensitive than cucumber seeds with EC50 values of 57 and 108mg/kg. Soil respiration was significantly inhibited in the treatments with triclosan at concentrations more than 10mg/kg (dry soil) during the first 4 days of incubation, but recovered later on after longer incubation. Phosphatase activity was also inhibited for all the soils treated with triclosan (from 0.1 to 50mg/kg dry soil), but a declining inhibition was observed after 2 days of incubation. Biolog analysis found that triclosan treatment increased the utilization of carbon sources and exerted no adverse effects on the functional diversity of soil microbial community. Objective To investigate the positive significance of standardized case in improving the teaching effect of "basic nursing techniques ". Methods We selected 64 health professionals in our school in grade 2012 as the research object. According to Student ID No. 1-64 odd and even numbers trainees were divided into experimental group (odd numbers) and control group (double number) . The control group students received traditional teaching. Experimental group students receivedtandardized case teaching. Results Experimental group students' average scores in theory, practice 1 and practice 2 examination were over 85 points, which were much higher than the control group (P < 0.05) . The satisfaction degree of students in the experimental group on standardized case pedagogy and teacher was higher than control group. Meanwhile, the students in the experimental group had more interest in basic nursing technical disciplines ( P < 0.05) . Conclusion Standardized case teaching can help students establish nursing norms awareness, strengthen the theoretical knowledge and practical operation, enhance students'interest in learning, so it has a positive meaning. Objective To study the expression of YB-1 in advanced cervical cancer tissues and the relationship between its expression and the clinicopathological characteristics, to investigate the role of YB-1 in cervical cancer invasion, metastasis and progress. Methods Immunohistochemistry was used to detect the expression of the YB-1 in 78 cases of middle and advanced stage cervical cancer tissue microarray chip and the relationship between its expression and the lymphatic metastasis, FIGO stage and the differentiated degree were analyzed. Results (1) The positive expression of YB-1 in cervical cancer was 24/78(30.8%); (2) The expression of YB-1 in cervical cancer was significantly related to FIGO stage and differentiation degree respectively. With the increase of FIGO stage and the decrease of differentiation degree, the expression of YB-1 increased gradually( P<0.05); The expression of YB-1 in cervical cancer was not significantly related to the lymph node metastasis. There was no significant difference between the lymph node metastasis positive and negative group(P > 0.05). In this study, seven laccase genes from different bacteria were linked with the signal peptides PelB, Lpp or Ompa for heterologous expression in E. coli. The recombinant strains were applied for the removal of sulfadiazine (SDZ), sulfamethazine (SMZ), and sulfamethoxazole (SMX). The results obtained for different signal peptides did not provide insights into the removal mechanism. The removal ratios of SDZ, SMZ, and SMX obtained with the recombinant strain 6#P at 60 h were around 92.0%, 89.0%, and 88.0%, respectively. The degradation pathways of sulfonamides have been proposed, including SO 2 elimination, hydroxylation, oxidation, pyrimidine ring cleavage, and N-S bond cleavage. Different mediators participate in the degradation of antibiotics through different mechanisms, and different antibiotics have different responses to the same mediator. The addition of 2,2'-azino-bis-(3-ethylbenzothiazoline-6-sulfonic acid) (ABTS) slightly promoted the removal of sulfonamides by most recombinant strains with different signal peptides, especially for the recombinant strain 2#O. The removal of sulfonamides by 1-hydroxybenzotriazole (HBT) varied with the recombinant strains. Syringaldehyde (SA) had a slight inhibitory effect on the removal of sulfonamides, with the most significant effect on strains 7#L and 7#O. An efficient and straightforward approach for the synthesis of carbamoylated chroman-4-ones has been well-developed. The reaction is triggered through the generation of carbamoyl radicals from oxamic acids under metal-free conditions, which subsequently undergoes decarboxylative radical cascade cyclization on 2-(allyloxy)arylaldehydes to afford various amide-containing chroman-4-one scaffolds with high functional group tolerance and a broad substrate scope. Primulina fangdingii, a species of Gesneriaceae new to science from Guangxi, China, is corroborated by both morphological and molecular evidence. The new species is similar to P. longzhouensis in leaf shape and size, but is easily distinguished from the latter by its adaxially glabrous leaf blades, yellow corolla with reddish inner surface and yellow filaments. It is also similar to P. repanda in corolla color and corolla shape, but can be diagnosed by its fleshy small leaves with glabrous adaxial surface, glandular and eglandular pubescent pedicels, yellow filaments and annular disc. In addition, Primulina fangdingii is somewhat similar to nine other species of Primulina, including P. cordifolia, P. danxiaensis, P. xiuningensis, P. chingipengii, P. cordistigma, P. diffusa, P. jingxiensis, P. petrocosmeoides and P. sinovietnamica, in either floral or vegetative morphology. However, all these species differ from P. fangdingii by a series of morphological characters. Phylogenetic analyses with ITS and two chloroplast DNA regions (rpL32‐trnL, trnL‐F) revealed that P. fangdingii is sister to P. luochengensis and isolated from its morphological relatives. Based on morphological observations, molecular phylogenetic analyses and literature consultations, a new species of Gesneriaceae, Petrocodon mirus, is described and illustrated here. This new species exhibits morphological similarity with P. pulchriflorus and P. hechiensis, but is easily distinguished from the latter two by its slightly leathery leaf blade, liner to ligulate‐oblong lip lobes, and shorter tube with constriction at middle. In addition, the new species differs from P. pulchriflorus by its reflexed adaxial lip lobes, and from P. hechiensis by its included stamen and pistil. However, the stigma of P. mirus is much similar to the chiritoid stigma diagnostic for ChiritaBuch.‐Ham. ex Don (now mostly included in Primulina); based on this observation a new hypothesis for the evolution of the chiritoid stigma is proposed. Auricular acupuncture has been used for pain management but a more recent technique which buries an intradermal needle into the skin may provide longer lasting stimulation and enhance analgesia. The purpose of this study was to evaluate the efficacy and feasibility of perioperative auricular acupuncture technique via intradermal needling buried on postoperative movement-evoked pain reduction in patients with advanced gastric cancer following open radical gastrectomy. This research is a protocol for a prospective, assessor- and analyst-blinded, randomized controlled pilot trial with two parallel groups. Forty patients who undergo elective open radical gastrectomy will be recruited from 210-bed oncology surgery ward at a 2500-bed medical center between 2019/8/23 and 2020/2/20. Patients will be randomized to either the control group receiving non-invasive sham auricular acupuncture or to the experimental group receiving auricular acupuncture technique via intradermal needling buried, delivered 24 h before surgery and retained & replaced every 3 days for 6 consecutive days. A standard general anesthetic technique will be used for both groups. Rescue analgesia will be given when the VAS score is≥4. Primary outcome measures are Functional Activity Score (FAS) and Numerical Rating Scale score (NRS) for movement-evoked pain. Secondary outcome measures are NRS score for pain at rest, use of rescues analgesic, total opioid consumption, postoperative nausea and vomiting, physiological recovery Outcomes, Quality of Recovery-9 (QoR-9) and patient's satisfaction. This protocol has been approved by the Institutional Review Board (IRB) of the Clinical Research Centre (Affiliated Hospital of Nanjing University of Chinese Medicine, IRB No. 2019NL-032-02). Background: MicroRNAs (miRNAs) have emerged as the critical modulators of the tumorigenesis and tumor progression. Methods: The levels of miR-663 in ovarian cancer cell lines and clinical tissues were detected using qRT-PCR assays. The Transwell invasion and wound healing assay were conducted to assess the roles of miR-663 in the migration and invasion of ovarian cancer cell in vitro. Rescue assays were carried out to confirm the contribution of tumor suppressor candidate 2 (TUSC2) in the aggressiveness of cancer cell which was regulated by miR-663. Results: The levels of miR-663 were up-regulated in ovarian cancer tissues in comparison with the corresponding normal tissues. Up-regulation of miR-663 increased the proliferation, colony formation, migration and invasion of ovarian cancer SKOV3 cell. Additional, over-expression of miR-663 increased the tumor growth of SKOV3 in xenograft model. Bioinformatics analysis and luciferase reporter assay identified that miR-663 decreased the level of TUSC2 via binding to the 3′-UTR of TUSC2 gene. Finally, the expression of TUSC2 was inversely associated with the level of miR-663 in ovarian carcinoma tissue and over-expression of TUSC2 inhibited the migration and invasion abilities of SKOV3 that was promoted by miR-663. Aryl radicals were generated for the first time from cheap and easily available aryl acyl peroxides in eco-friendly ethyl acetate under ambient conditions and visible-light illumination in the absence of any additive, metal catalyst, or external photosensitizer. The incorporation of amide groups into biologically active molecules has been proven to be an efficient strategy for drug design and discovery. In this study, we present a simple and practical method for the synthesis of amide-containing quinazolin-4(3H)-ones under transition-metal-free conditions. This is achieved through a carbamoyl-radical-triggered cascade cyclization of N3-alkenyl-tethered quinazolinones. Intracardiac thrombosis is a severe complication in patients with non-ischemic dilated cardiomyopathy. This study aims to develop and validate an individualized nomogram to evaluate the risk of intracardiac thrombosis in patients with non-ischemic dilated cardiomyopathy. This retrospective study included patients diagnosed with dilated cardiomyopathy at first admission. Clinical baseline characteristics were acquired from electronic medical record systems. Multiple methods were applied to screen the key variables and generate multiple different variable combinations. Multivariable logistic regression was used to build the models, and the optimal model was chosen by comparing the discrimination. Then we checked the performance of the model in different thrombus subgroups. Finally, the model was presented using a nomogram and evaluated from the perspectives of discrimination, calibration, and clinical usefulness. Internal validation was performed by extracting different proportions of data for Bootstrapping. Ultimately, 564 eligible patients were enrolled, 67 of whom developed an intracardiac thrombosis. Risk factors included d-dimer, white blood cell count, high-sensitivity C-reactive protein, pulse pressure, history of stroke, hematocrit, and NT-proBNP in the optimal model. The model had good discrimination and calibration, and the area under the curve (AUC) was 0.833 (0.782–0.884), and the model's performance in each subgroup was stable. Clinical decision curve analysis showed that the model had clinical application value when the high-risk threshold was between 2% and 78%. The AUC of interval validation (30% and 70% data resampling) was 0.844 (0.765–0.924) and 0.833 (0.775–0.891), respectively. This novel intracardiac thrombosis nomogram could be conveniently applied to facilitate the individual intracardiac thrombosis risk assessment in patients with non-ischemic dilated cardiomyopathy. Background: To date, the clinical management of advanced hepatocellular carcinoma (HCC) patients remains challenging and the mechanisms of E2F transcription factor 1 (E2F1) underlying HCC are obscure. Materials and Methods: Our study integrated datasets mined from several public databases to comprehensively understand the deregulated expression status of E2F1. Tissue microarrays and immunohistochemistry staining was used to validate E2F1 expression level. The prognostic value of E2F1 was assessed. In-depth subgroup analyses were implemented to compare the differentially expressed levels of E2F1 in HCC patients with various tumor stages. Functional enrichments were used to address the predominant targets of E2F1 and shedding light on their potential roles in HCC. Results: We confirmed the elevated expression of E2F1 in HCC. Subgroup analyses indicated that elevated E2F1 level was independent of various stages in HCC. E2F1 possessed moderate discriminatory capability in differentiating HCC patients from non-HCC controls. Elevated E2F1 correlated with Asian race, tumor classification, neoplasm histologic grade, eastern cancer oncology group, and plasma AFP levels. Furthermore, high E2F1 correlated with poor survival condition and pooled HR signified E2F1 as a risk factor for HCC. Enrichment analysis of differentially expressed genes, coexpressed genes, and putative targets of E2F1 emphasized the importance of cell cycle pathway, where CCNE1 and CCNA2 served as hub genes. Conclusions: We confirmed the upregulation of E2F1 and explored the prognostic value of E2F1 in HCC patients. Objective: The goal of this research was to review the literature from randomized controlled trials (RCTs) on the impacts of moxibustion on cancer-related fatigue (CRF) as well as provide credible evidence to guide clinical practice. Methods: Three English electronic medical databases (PubMed, Embase, and the Cochrane Library) and two Chinese databases (China National Knowledge Infrastructure and Wanfang) were searched. Only randomized controlled trials on the effect of moxibustion on CRF were included in this systematic review. Study selection, data extraction, and validation were all carried out independently by two reviewers. The revised Cochrane Risk of Bias tool was used to assess the quality of the RCTs (RoB 2.0). The Grading of Recommendations Assessment, Development and Evaluation (GRADE) system was applied to assess effect sizes in individual RCTs and pooled effect sizes in meta-analyses. Data were meta-analyzed using Stata (version 14.0). Results: In a random-effects meta-analysis of 24 RCTs with 1894 participants, the aggregated standardized mean difference (SMD) revealed a statistically significant association between moxibustion and alleviation from cancer-related fatigue (SMD = − 1.66, 95% CI = − 2.05, − 1.28, p = 0.000). Pooled results, however, show significant heterogeneity (I2 = 92.5%), and the evidence is insufficient to determine whether this association varies systematically by measuring tools and moxibustion modalities. Furthermore, evidence ranging from very low to low showed that moxibustion had an immediate positive effect on patients with CRF. Conclusion: Moxibustion may have a therapeutic effect on cancer-related fatigue. However, further large-scale, multicenter, high-quality RCTs on moxibustion for fatigue relief and safety are still needed because of the handful of studies included and the low methodological quality. Cyclospora spp., foodborne intestinal protozoa with potential zoonotic risk, are distributed across the globe. However, their presence in animals bred for fur has not been extensively studied, raising concerns about zoonotic transmission. To assess the prevalence of Cyclospora spp. in fur animals in China, we collected fresh fecal samples from 862 animals, including 352 blue foxes, 275 minks, and 235 raccoon dogs, across multiple provinces. The small subunit (SSU) rRNA gene of Cyclospora spp. was amplified using nested PCR, followed by restriction fragment length polymorphism (RFLP) analysis with the Kpn 2I enzyme. We also analyzed geographic location and clinical symptoms, such as diarrhea, as potential risk factors. Our findings showed an overall infection rate of 1.3 % for Cyclospora spp. in fur animals, with 2.3 % in blue foxes, 0.4 % in minks, and 0.9 % in raccoon dogs. Geographic location was a significant infection risk factor in blue foxes (P < 0.001), while diarrhea was a risk factor in all fur animals (P < 0.05). A molecular phylogenetic analysis based on SSU rRNA sequences revealed that two isolates clustered with human-derived Cyclospora cayetanensis , while the remaining isolates grouped with previously reported cattle-derived Cyclospora spp. This study is the first to report Cyclospora spp. infections in fur animals worldwide, underscoring the potential for zoonotic transmission. ThE present work focused on exploring Girdin expression within gastric cancer (GC), examining the effect of Girdin on the cell phenotype of GC, and clarifying the underlying mechanisms. Girdin expression in GC samples was identified by immunohistochemistry (IHC) and quantitative real-time PCR (qRT-PCR) assays. Girdin-targeting siRNAs were transfected into GC cells; later, we examined GC cell proliferation, migration, invasion, and apoptosis, respectively. Additionally, the protein expression was examined through Western blotting assay. Moreover, the tumor implantation experiment was conducted for examining Girdin knockdown in vivo. The results showed that Girdin expression elevated within GC samples, which was associated with the dismal prognostic outcome. Girdin knockdown suppressed GC cell proliferation, migration, and invasion, and enhanced apoptosis and cell cycle arrest. Girdin promoted the phosphorylation of AKT, GSK3β, and β-catenin. Moreover, Girdin inhibited the phosphorylation of β-catenin. Girdin suppressed cell apoptosis and stimulated cell migration and invasion, while AKT inhibitor (MK2206) treatment reversed the effect of Girdin overexpression, and GSK3β inhibitor (CHIR99021) treatment enhanced the effect of Girdin overexpression on GC cells. Besides, Girdin delayed tumor growth in vivo. In conclusion, Girdin was abnormally expressed in GC samples, which promoted the development of GC by regulating AKT/GSK3β/β-catenin signaling.
