Abstract

Results show that nitrophorin 4 complexed with nitric oxide show ruffling of the heme in the presence or absence of nitric oxide. Data indicate that this protein-assisted heme ruffling may give rise to an autoreduction resistant nitrosyl-heme complex.

Research into the electronic structures of the bis-imidazole complexes of iron(III) tetraphenylporphyrin and iron(III) tetraphenylchlorin in frozen glassy solutions is presented. ESEEM spectra were used to establish the orientation of the imidazole ligand planes with regard to the g tensor axes.

Abstract

Evidence for stabilization of a nitric oxide-ferriheme complex via a ligand-induced conformational trap has been found. Kinetics and equilibria in ligand binding by nitrophorin (NP) 1-4 are discussed. Results of a study using kinetic and thermodynamic characterizations of NO and histamine binding to recombinant forms of all four NPs, suggest that tighter NO binding in the nitrophorins is the result of the trapping of the molecule in a nonpolar distal pocket, not of formation of especially strong hydrogen or hydrogen bonds. NPs 1-4 are ferriheme proteins from Rhodnius prolixus, a blood-sucking insect. They move NO to the victim, sequester histamine and inhibit coagulation of blood.

Abstract

The effect of axial ligand plane orientation on the isotropic shifts of the dichelated protohemin cyanide complexes of T.G. Traylor and A.P. Berzinis, who published the NMR spectrum of the mixture of two geometrical isomers in 1980. It was one of the first proofs of dependence of heme methyl shifts on the orientation of planar axial ligands. Now the (super.1)H NMR spectra have been investigated using 1- and 2D NMR techniques and the majority of the resonances have been assigned, with the exception of some of the meso-H resonances and those of the CH(sub.2) protons beyond the beta-CH(sub.2) of the 6- and 7-propionamides of each isomer. Each isomer has four widely separated methyl resonances, the order and spread of chemical shifts of which is consistent with predictions.

Abstract

The waterborne pathogen Cryptosporidium has been identified in surface drinking water supplies. Suspected sources of this pathogen include sewage and the feces of animals, particularly dairy calves. There are many dairy cattle and significant sewage effluent discharges in the Catskill-Delaware watershed that is part of the New York City water supply system. This water supply serves 8,000,000 customers with 5.8 x [10.sup.9] L (1.5 billion gal. of water daily). This paper is concerned with the movement and fate of pathogens from waste-water and dairy sources and the resulting raw water quality for New York City. Manure and Cryptosporidium oocysts are modeled as surface pollutants and assumed to move in response to runoff events in the six watershed-reservoir systems within the Catskill-Delaware watershed. Oocyst degradation in manure and in water is modeled with first-order kinetics. Rudimentary stream routing and reservoir modeling with a first-order decay function complete the fate and transport modeling of oocysts in the watercourse. Reported effluent discharge rates and oocyst concentrations in secondary treated sewage allow estimation of wastewater-derived oocyst contributions. This research highlights the importance of wastewater-derived oocysts, the need for expanded research into oocyst fate in streams and reservoirs, and the concentration of oocysts in sewage effluent.

Abstract

REGISTRY Investigators of the European Huntington's Disease Network and COHORT Investigators of the Huntington Study Group., Studying individuals with extreme phenotypes could facilitate the understanding of disease modification by genetic or environmental factors. Our aim was to identify Huntington's disease (HD) patients with extreme symbol digit modality test (SDMT) scores. We first examined in HD the contribution of cognitive measures of the Unified Huntington's Disease Rating Scale (UHDRS) in predicting clinical endpoints. The language-independent SDMT was used to identify patients performing very well or very poorly relative to their CAG and age cohort. We used data from REGISTRY and COHORT observational study participants (5,603 HD participants with CAG repeats above 39 with 13,868 visits) and of 1,006 healthy volunteers (with 2,241 visits), included to identify natural aging and education effects on cognitive measures. Separate Cox proportional hazards models with CAG, age at study entry, education, sex, UHDRS total motor score and cognitive (SDMT, verbal fluency, Stroop tests) scores as covariates were used to predict clinical endpoints. Quantile regression for longitudinal language-independent SDMT data was used for boundary (2.5% and 97.5% quantiles) estimation and extreme score analyses stratified by age, education, and CAG repeat length. Ten percent of HD participants had an extreme SDMT phenotype for at least one visit. In contrast, only about 3% of participants were consistent SDMT extremes at two or more visits. The thresholds for the one-visit and two-visit extremes can be used to classify existing and new individuals. Abstract

AIMS: The clinical effectiveness of primary prevention implantable cardioverter defibrillator (ICD) therapy is under debate. The EUropean Comparative Effectiveness Research to Assess the Use of Primary ProphylacTic Implantable Cardioverter Defibrillators (EU-CERT-ICD) aims to assess its current clinical value. METHODS AND RESULTS: The EU-CERT-ICD is a prospective investigator-initiated non-randomized, controlled, multicentre observational cohort study performed in 44 centres across 15 European Union countries. We will recruit 2250 patients with ischaemic or dilated cardiomyopathy and a guideline indication for primary prophylactic ICD implantation. This sample will include 1500 patients at their first ICD implantation and 750 patients who did not receive a primary prevention ICD despite having an indication for it (non-randomized control group). The primary endpoint is all-cause mortality; the co-primary endpoint in ICD patients is time to first appropriate shock. Secondary endpoints include sudden cardiac death, first inappropriate shock, any ICD shock, arrhythmogenic syncope, revision procedures, quality of life, and cost-effectiveness. At baseline (and prior to ICD implantation if applicable), all patients undergo 12-lead electrocardiogram (ECG) and Holter ECG analysis using multiple advanced methods for risk stratification as well as detailed documentation of clinical characteristics and laboratory values. Genetic biobanking is also organized. As of August 2018, baseline data of 2265 patients are complete. All subjects will be followed for up to 4.5 years. CONCLUSIONS: The EU-CERT-ICD study will provide a necessary update about clinical effectiveness of primary prophylactic ICD implantation. Abstract

Nuclear magnetic resonance measurements were taken of the rate of axial ligand rotation and rotational DeltaH++, DeltaS++ of the chemical exchange between pyrrole protons in tetramesitylporphyrinatoiron(III)bis(2-methylimidazole). The activation entropy in this particular tetraphenylporphyrin complex is close to the experimental error that can be estimated as roughly 6 J/K.mol, despite differences in the activation entropy between the Fe(III) and Co(III) complexes. Experimental methodology and results are discussed.

Abstract

Research was conducted to elucidate the anomalous properties exhibited by hyperfine shifts of carbon nuclei and protons of methyls of the heme of cytochrome c. The results provided strong evidence that the two methyl protons have anti-Curie temperature dependence. This was attributed to the opposite sign of the contact shift for the two nuclei and the metal-centered pseudocontact shifts for carbons. The findings were also consistent with theoretical predictions.

Abstract

A series of unsymmetrical derivatives of (tetraphenylporphyrinate)FeCl, where one phenyl ring has a 2-halogen, 2-CF3, or 2,6-dihalogen while the other three have p-OCH3 were studied by NMR and UV-visible spectroscopy and electrochemistry. Results showed that equilibrium constants for addition of 2 N-methylimidazole ligands, in chloroform, increase F < Cl < Br < I < F2 < CF3 < Cl2 < Br2, while those in dimethylformamide increase in the order Cl2 < CF3 < F. Halogens on ortho positions seemed to be electron-donating.

Abstract

ESEEM spectroscopy was utilized to determine the orientation of the g tensor in two model heme complexes, (OEPFe(imidazole)2)+ and (OEPFe-(4-(dimethylamino)pyridine)2)+ (OEP = octaethylporphyrinate). Frequency shifts and intensities were measured at several points across the EPR spectrum and then compared to the expected spectra. Results show that g(sub zz) is the largest g value n both cases. Findings also indicate that the orientations of g(sub xx) and g(sub xy) differ depending on the orientation of the parallel axial ligands.

Abstract

Research was conducted to examine the concept, origins and reasons of counterrotation of the g or % tensor and to delineate the conditions for co- and counterrotation. Results indicate that counterrotation is the only possibility for the (d(sub xz))4, (d(sub xy))1 configuration and that it is also more common for the (d(sub xy))2(d(sub yz))3 electron configuration. An investigation into the possibilities of nonlinear co- and counterrotation was also conducted.

Abstract

Investigations involving the synthesis, structure and thermodynamic characterizations of bispyridine and bis-imidazole complexes of iron (II) tetramesityleporphyrinate were conducted. The ferrocytochrome models including [Fe(TMP)(4-CNPy)2], [Fe(TMP)(3-CNPy)2] and [Fe(TMP)(4-MePy)2] were compared in their solid state with the same complexes in homogeneous solution. Results showed that the thermodynamics of legal binding were not affected by the electronic properties of the axial ligand while low-spin d6 metalloporphyrins preferred parallel orientation of planar axial ligands.

Abstract

Nuclear magnetic resonance (NMR) and electron paramagnetic resonance (EPR) spectroscopic analyses were done to characterize the oxomolybdenum(V)/iron(III) porphyrinate complexes, Fe(2,3-Mo-TTP)(2MeImH)2Cl (complex 1) and Fe(3,4-Mo-TTP)(2MeImH)2Cl (complex 2). The results showed that the axial ligand planes of both complexes are in perpendicular orientation. The NMR results also confirmed that the pyrolle protons of complex 1 are quite different from that of the NMeIm analogue which has parallel axial ligand planes and a rhombic Fe(III) EPR signal.

Abstract

Isocyanide complexes of (porphyrinato)iron(III) species, ((Porph)-Fe(t-BuNC)2)ClO4, Porph = OEP, TPP, were synthesized and characterized. Consistent with the predicted effect from the strong pi-acceptor character of the axial tert-butyl isocyanide ligands, the X-ray structure of the complex revealed that the porphyrinate ring is strongly ruffled.

Abstract

Cyclic voltammetry was utilized to determine the equilibrium constants for the complex formation of six 'hindered' iron porphyrinates with four pyridines and two imidazoles. Increases in the size of the phenyl 2,6 substituents also increase the equilibrium constants of the 'hindered' iron porphyrinates. Iron(III) complexes also exhibit linear free energy dependence due to electronic and steric effects that are enhanced by the charge of the central iron. The presence of bulky ortho substituents (Cl, Br, CH3) induces the formation of bis complexes between 2-methylimidazole and iron(II)tetraphenylporphyrinates.

Abstract

The low-spin complexes of three ferric tetramesitylporphyrin (FeIIITMP) are investigated by low-temperature magnetic circular dichroism (MCD) spectroscopy. Complexes 1 and 2 have ground state electronic configurations of (dxy)2(dxz,dyz)3 although they exhibit near-infrared porphyrin(pi)-ferric(d) charge transfer (NIR-CT) bands of completely varied MCD intensities. Complex 3 has vertical ligands and axial EPR spectrum of abnormal g-values and low MCD transitions in NIR, ultraviolet and visible wavelengths, which are attributed to a new (dxz,dyz)4(dxy)1 ground state.

Abstract

A correlation between data from Mossbauer quadrupole splittings and NMR is helpful in detecting the 57Fe NMR signals for valence electron cloud asymmetry. The 11 197 ppm chemical shift reported in literature for ferrocytochrome, c20, which is based on Mossbauer-NMR correlation, is acceptable. This correlation also indicates chemical shifts of 11 000 to 13 000 ppm for most bis-pyridine and nonhindered imidazole complexes of model heme and cytochrome b5.

Abstract

Aims: The clinical effectiveness of primary prevention implantable cardioverter defibrillator (ICD) therapy is under debate. The EUropean Comparative Effectiveness Research to Assess the Use of Primary ProphylacTic Implantable Cardioverter Defibrillators (EU‐CERT‐ICD) aims to assess its current clinical value. Methods and results: The EU‐CERT‐ICD is a prospective investigator‐initiated non‐randomized, controlled, multicentre observational cohort study performed in 44 centres across 15 European Union countries. We will recruit 2250 patients with ischaemic or dilated cardiomyopathy and a guideline indication for primary prophylactic ICD implantation. This sample will include 1500 patients at their first ICD implantation and 750 patients who did not receive a primary prevention ICD despite having an indication for it (non‐randomized control group). The primary endpoint is all‐cause mortality; the co‐primary endpoint in ICD patients is time to first appropriate shock. Secondary endpoints include sudden cardiac death, first inappropriate shock, any ICD shock, arrhythmogenic syncope, revision procedures, quality of life, and cost‐effectiveness. At baseline (and prior to ICD implantation if applicable), all patients undergo 12‐lead electrocardiogram (ECG) and Holter ECG analysis using multiple advanced methods for risk stratification as well as detailed documentation of clinical characteristics and laboratory values. Genetic biobanking is also organized. As of August 2018, baseline data of 2265 patients are complete. All subjects will be followed for up to 4.5 years. Conclusions: The EU‐CERT‐ICD study will provide a necessary update about clinical effectiveness of primary prophylactic ICD implantation. This study also aims for improved risk stratification and patient selection using clinical and ECG risk markers.
