International audience; To evaluate the nanoparticle (NP) toxicity, much efforts have been devoted for developing methods to accurately disperse NPs into aqueous suspensions prior to in vitro toxicological studies. As NP toxicity is strongly dependent on their physicochemical properties, NP characterization is a key step for any in vitro toxicological study. This study demonstrates that the static multiple light scattering (SMLS) technique allows for the simultaneous screening of the NP size, agglomeration state, stability and dosimetry in biological media. Batch dispersions of TiO2 P25 NPs in water with various bovine serum albumin (BSA) mass fractions (from 0% to 0.5%) and dilutions of these dispersions into cell culture media were characterized with SMLS. In the batch dispersions, TiO2 NPs are stable and well dispersed for BSA mass fraction lower than 0.2% while agglomeration and rapid settling is observed for higher BSA mass fractions. Paradoxically, when diluted in cell culture media, TiO2 NPs are well dispersed and stable for BSA mass fractions higher than 0.2%. The TiO2 NP dosimetry of these dilutions was evaluated experimentally with SMLS and confronted with numerical approaches. The TiO2 NP bottom concentration evolves far more slowly in the case of the higher BSA mass fraction. Such measurements give valuable insights on the NP fate and transport in biological media to obtain in fine reliable size and dose-cytotoxicity responses.

International audience; Diarrheic shellfish poisoning (DSP) is caused by the consumption of shellfish contaminated with a group of phycotoxins that includes okadaic acid (OA), dinophysistoxin-1 (DTX-1), and dinophysistoxin-2 (DTX-2). These toxins are inhibitors of serine/threonine protein phosphatases 1 (PP1) and 2A (PP2A), but show distinct levels of toxicity. Aside from a difference in protein phosphatases (PP) inhibition potency that would explain these differences in toxicity, others mechanisms of action are thought to be involved. Therefore, we investigated and compared which mechanisms are involved in the toxicity of these three analogues. As the intestine is one of the target organs, we studied the transcriptomic profiles of human intestinal epithelial Caco-2 cells exposed to OA, DTX-1, and DTX-2. The pathways specifically affected by each toxin treatment were further confirmed through the expression of key genes and markers of toxicity. Our results did not identify any distinct biological mechanism for OA and DTX-2. However, only DTX-1 induced up-regulation of the MAPK transduction signalling pathway, and down-regulation of gene products involved in the regulation of DNA repair. As a consequence, based on transcriptomic results, we demonstrated that the higher toxicity of DTX-1 compared to OA and DTX-2 was consistent with certain specific pathways involved in intestinal cell response.

The increasing worldwide population and industrialization engender great pressure on available water resources. One of the solutions to the water scarcity is to no longer consider wastewater as mere waste, but rather as a potential alternative resource for various applications, after appropriate treatment. Nevertheless, urban wastewater is increasingly contaminated with organic micropollutants (OMP) such as biocides and pharmaceutical substances that are refractory to conventional treatment. Safe and sustainable reuse of this resource requires the use of appropriate tertiary treatment processes. With this in mind, membrane processes, especially nanofiltration and reverse osmosis, have recently attracted considerable interest, as they ensure good removal of micropollutants and the production of high-quality permeate. However, these separation processes still face the challenges of membrane fouling and concentrate management. In addition, advanced oxidation processes such as ozonation degrade micropollutants well but are likely to lead to the formation of by-products that are potentially more toxic than the parent compounds. The objective of this study was therefore to develop an integrated and innovative system coupling nanofiltration (NF) and ozonation (O3) applied to a membrane bioreactor (MBR) secondary effluent. The ozonation process degrades micropollutants and organic matter well. The nanofiltration process provides a satisfactory retention of micropollutants and ozonation by-products and produces a very good quality effluent. Pre-ozonation mitigates membrane foulinf in nanofiltration. This study demonstrates the synergistic operation of the hybrid process to efficiently treat secondary effluents containing priority substances targeted by legislation in order to provide safe and sustainable urban wastewater reuse.; L’accroissement de la population mondiale et de l’industrialisation exerce une forte pression sur les ressources en eau disponibles. Une des solutions à la conséquente raréfaction des ressources d’eau est de ne plus considérer les eaux usées comme déchets mais plutôt comme ressources potentielles alternatives, pour diverses application, après un traitement approprié. Néanmoins, les eaux usées urbaines sont de plus en plus contaminées par des micropolluants organiques (OMP) tels que des biocides et des substances pharmaceutiques réfractaires aux traitements conventionnels. Une réutilisation sure et durables de cette ressource requiert de recourir à des procédés de traitement tertiaire adaptés. Dans cette optique, les procédés membranaires, plus particulièrement la nanofiltration et l'osmose inverse, ont récemment suscité un intérêt considérable, car ils assurent une bonne élimination des micropolluants et la production de perméat de très bonne qualité. Cependant, ces procédés de séparation restent confrontés aux défis du colmatage de la membrane et de la gestion des concentrats. Par ailleurs les procédés d’oxydation avancée tels que l’ozonation dégradent bien les micropolluants mais sont susceptibles de conduire à la formation de sous-produits potentiellement plus toxiques que les composés-mères. L'objectif de cette étude était donc de développer un système intégré et innovant couplant la nanofiltration (NF) et l'ozonation (O3) appliqué à un effluent secondaire de bioréacteur à membrane (BaM). Le procédé d’ozonation dégrade bien les micropolluants et la matière organique. Le procédé de nanofiltration assure une rétention satisfaisante des micropolluants, des sous-produits d’ozonation et produit un effluent de très bonne qualité. La pré-ozonation permet d’atténuer le colmatage de la membrane de nanofiltration. Cette étude démontre le fonctionnement synergique du procédé hybride pour traiter efficacement les effluents secondaires contenant des substances prioritaires visées par la législation afin d'offrir une réutilisation sûre et durable des eaux usées municipales.

Quantum dots (QDs) are fluorescent semiconductor nanocrystals with exceptional optical properties, which make them particularly attractive in optoelectronic fields and for biomedical applications. However, during their life cycle, the aging of QDs can lead to the degradation of these compounds, inducing the release of toxic elements. Even if toxicity studies on indium-based QDs are still limited, they show a lower intrinsic toxicity in comparison to the heavy metal containing Cd-based QDs. In this context, our laboratory synthetizes different InP QDs with different shell designs, following a safer by design approach, with the aim of producing less toxic QDs with better optical properties. These QDs are composed of a InZnP/Zn(Se,S) core/shell structure which is covered or not by a thick or a thin additional ZnS layer. In this study, primary human keratinocytes which come from breast surgeries, were exposed to these QDs, either pristine or after simulating environmental weathering. First, the physico-chemical transformations of QDs during aging are characterized. Significant photophysical and structural modifications are highlighted and transformation products are identified. However, the results show that these physico-chemical transformations are slowed down by the presence of a double shell, especially when it is thick. Then, the evaluation of QDs toxicity are performed and new assays are developed via high content screening (HCS) on an automated microscope. While pristine QDs were relatively stable and not very toxic to cells, it was not true for their degradation products. Exposure of cells to aged QDs demonstrated high toxicity at low concentrations and modifyed the expression of some genes and proteins essential for cellular homeostasis. These results show that new generations of QDs are safer. However, it’s important to keep improving their photostability since their dissolution and the release of toxic elements at the end of their life are still inevitable., Les quantum dots (QD) sont des nanocristaux semi-conducteurs fluorescents aux propriétés optiques exceptionnelles, ce qui les rend particulièrement attractifs dans les domaines de l’optoélectronique et pour les applications biomédicales. Cependant, au cours de leur cycle de vie, le vieillissement des QDs peut conduire à la dégradation de ces composés, induisant la libération d'éléments toxiques. Même si les études de toxicité sur les QDs à base d'indium sont peu nombreuses, certaines révèlent une toxicité intrinsèque plus faible que les QDs contenant des métaux lourds comme le Cd. Dans ce contexte, notre laboratoire synthétise différents QDs d’InP recouverts de coquilles, conçus par une approche « safer by design », dans le but de produire des QDs moins toxiques avec de meilleures propriétés optiques. Ces QDs sont constitués d'une structure cœur/coquille de InZnP/Zn (Se,S) qui est recouverte, ou non, d'une couche additionnelle de ZnS, épaisse ou mince. Dans cette étude, des kératinocytes primaires humains, issus de chirurgies mammaires, sont exposés aux QDs, après synthèse ou après vieillissement environnemental simulé. Dans un premier temps, les transformations physico-chimiques des QDs au cours du vieillissement sont caractérisées et mettent en évidence d’importantes modifications photophysiques et structurales ainsi que la formation de produits de transformation. Néanmoins, les résultats montrent que les transformations physico-chimiques des QDs sont ralenties par la présence de la double coquille, notamment lorsqu’elle est épaisse. Dans un second temps, l’évaluation de la toxicité des QDs est effectuée et de nouveaux tests sont dévelopés en criblage à haut contenu (HCS) sur un microscope automatisé. Alors que les QDs non vieillis se sont révélés relativement stables et peu toxiques pour les cellules, il n’en fût pas de même pour leurs produits de dégradation. L’exposition des cellules aux QDs vieillis a mis en évidence une forte toxicité à faibles concentrations, modifiant l’expression de certains gènes et protéines essentiels à l’homéostasie cellulaire. Ces résultats montrent que les nouvelles générations de QDs sont plus sûres. Cependant, il est important de continuer à améliorer leur photostabilité puisque leur dissolution et le relargage d’éléments toxiques en fin de vie sont inévitables pour le moment.

The use of veterinary herbal medicine and aromatherapy is booming in the context of the fight against antibiotic resistance and the development of organic farming in France. Few herbal veterinary pharmaceutical specialties are currently available and few herbal substances have an MRL status. The toxicological profile of eleven essential oils and one component, methyl eugenol, was investigated from data available in the scientific literature. Very partial data was found for ten of these essential oils. Data are available on tea tree essential oil, however, there is still some uncertainty about the presence of methyl eugenol, a recognized carcinogen in rodents. Due to the lack of robust toxicological data, a classification of the plants studied could not be made., Le recours à la phytothérapie et l’aromathérapie vétérinaires est en plein essor dans le contexte de la luttecontre l’antibiorésistance et du développement de l’Agriculture Biologique en France. Peu de spécialités pharmaceutiques vétérinaires à base de plantes sont actuellement disponibles et peu de substances végétales ont un statut LMR. Le profil toxicologique de onze huiles essentielles et d’un composant, le méthyleugénol, a été étudié à partir des données disponibles dans la littérature scientifique. Des données très partielles ont été trouvées pour dix de ces huiles essentielles. Des données sont disponibles sur l’huile essentielle de Tea tree, il reste cependant une incertitude quant à la présence de méthyleugénol, reconnu cancérogène chez les rongeurs. Compte tenu de l’absence de données toxicologiques robustes, une classification des plantes étudiées n’a pas pu être réalisée.

Along the past thirty years, the minimal adhesive dentistry and biomimetism concepts have allowed the emergence of new less-invasive care solutions in restorative dentistry. Among them, adhesive ceramic restorations take an important place in the current therapeutic arsenal. The usual ceramic bonding, whose characteristics will be remind in the first part, is based on a well-defined protocol, requiring hydrofluoric acid to etch the prosthetic surface. However, this acid has various disadvantages which we will explain in a second part. The ammonium polyfluoride, which we will present in the last part, seem an alternative. The aim of this work is to analyze, with regard to the literature and clinical data, the advantages and drawbacks of this new ceramic primer.; Au cours des trente dernières années, les concepts de dentisterie adhésive a minima et de biomimétisme ont permis l'apparition de nouvelles solutions de soins, moins invasives, dans le domaine de l'odontologie restauratrice. Parmi elles, les restaurations adhésives en céramique occupent une place importante au sein de l'arsenal thérapeutique actuel. Le collage traditionnel de ces céramiques, dont les caractéristiques seront rappelées en première partie, repose sur un protocole bien défini, nécessitant le recours à l'acide fluorhydrique pour le mordançage de l'intrados prothétique. Cet acide présente cependant divers inconvénients que nous exposerons dans une seconde partie. Le polyfluorure d'ammonium, que nous présenterons dans la dernière partie, est présenté comme une alternative. Ce travail a pour objectif d'analyser, au regard de la littérature et des données cliniques, les avantages et inconvénients de ce nouveau primer pour céramique.

International audience; Genotoxicity is one of the key endpoints investigated as early as possible before marketing a product. Several assays can be performed to evaluate the genotoxicity and mutagenicity of compounds, covering the detection of DNA lesions and of DNA repair activities as well as the quantification of the three types of mutations at the gene, chromosome and genome levels. Most of these tests have an established or draft guideline from the Organization for Economic Co-operation and Development (OECD). Nevertheless, if these assays have been developed for chemicals, they are not fully suitable for testing the genotoxicity/mutagenicity of nanomaterials, and the adaptation of the experimental conditions for such compounds is still debated. This Special Issue “From Basic Research to New Tools and Challenges for the Genotoxicity Testing of Nanomaterials” aimed to provide new results on a large range of nanomaterials using a broad panel of genotoxicity assays and to solve some of the numerous issues faced on this topic.

Objectif : l’immunothérapie a révolutionné le traitement du cancer. Cependant, elles entraînent de nouvelles toxicités liées à leurs mécanismes d’action qui nécessitent une prise en charge particulièrement attentive. Dans ce contexte, il nous ait apparu intéressant d’analyser les toxicités graves des immunothérapies en vie réelle d’utilisation et de les comparer aux toxicités mentionnées dans les RCP. Matériels et Méthode : étude rétrospective de janvier 2017 à septembre 2019, incluant tous les patients traités par immunothérapie et pour toutes les indications, à l’Institut Paoli Calmette de Marseille. Une partie des résultats obtenus a été comparée aux données des RCP du Nivolumab et Pembrolizumab. Résultats : sur la période étudiée, 418 patients ont été traités par immunothérapie et 50 (soit 12%) ont développé des toxicités de grade 3-4. Les toxicités les plus représentés ont été : la toxicité digestive (3.6%), la toxicité pulmonaire (1.7%) et les toxicités hépatiques, cutanées et musculosquelettiques représentaient respectivement chacune 1.4%. 54% des patients (soit 27/50) ont été hospitalisés dont 3 patients passés en réanimation. Discussion-conclusion : la majorité des toxicités relevées dans notre étude bien que rares pour certaines sont des effets attendus des immunothérapies, puisque décrites dans les RCP. Le traitement pilier de ces toxicités repose sur la corticothérapie mais également sur une prise en charge pluridisciplinaire.

Objectif. Évaluer l’impact du statut nutritionnel sur la tolérance et la réponse à la chimiothérapie d’induction dans les cancers de stade avancé de la tête et du cou.Design de l’étude. Étude rétrospective monocentrique.Matériels et méthodes. Quatre-vingt-douze patients traités par une chimiothérapie d’induction associant docétaxel, cisplatine et 5-fluorouracile (TPF) pour un carcinome épidermoïde de stade avancé des voies aéro-digestives supérieures ont été inclus. La toxicité a été évaluée selon la Common terminology criteria for adverse events (CTCAE) version 4.0 et la réponse obtenue selon les critères Response evaluation criteria in solid tumors (RECIST) 1.1. L’état nutritionnel a été évalué par méthode anthropométrique (poids, perte de poids avant et durant le traitement, indice de masse corporelle (IMC)), biologique (albumine), mixte (Nutritional Index Risk) et scannographique (évaluation de la composition musculaire corporelle au niveau de la première vertèbre lombaire : indice de masse musculaire (IMM)).Résultats. En début de traitement, la valeur moyenne de l’IMC était de 22.7 ± 4.6 kg/m², l’albuminémie de 38.7 ± 5.8 g/L, le NRI de 97.6 ± 10.6 et l’IMM de 36.4 ± 7.9 cm²/m². Après le traitement, l’IMC était de 23 ± 4.5 kg/m², l’albuminémie de 30.2 ± 7.1 g/L et le NRI de 88.1 ± 9.2. Au cours du traitement par TPF, cinquante-sept patients ont présenté au moins une toxicité ≥ grade 3 liée à la chimiothérapie. L’IMM initial (p = 0.02), la perte de poids durant le traitement (p = 0.02), l’albuminémie (p = 0.006) et le NRI (p = 0.01) à la fin du traitement ont été associés à la toxicité du TPF. Le NRI au début du traitement était associé à la réponse au TPF (p = 0.04).Conclusion. La présence d’une sarcopénie avant le début de la chimiothérapie de type TPF est un facteur prédictif significatif de toxicité chez les patients atteints d’un cancer de la tête et du cou de stade avancé. L’analyse de la composition corporelle et en particulier l’évaluation de la perte de masse musculaire devrait donc faire partie du bilan avant ce type de chimiothérapie.

The eye is a complex and priceless key organ of human body. It can face attacks thanks to combined protections giving by its well-organized anatomy but it’s submitted to a delicate balance which making it sensible too. Through the analysis of the composition of eye drops, and especially preservatives, one has come to light: benzalkonium chloride. This component, widely used in many other area than ophthalmology, has in one hand many interesting properties but, in the other hand, more and more studies demonstrate side effects, mainly for long term use, affecting equally surface and deep layers of the eye. Many people around the world are concerned by eye drop chronic use in treatment for allergies, dry eye or glaucoma. Some alternatives have been found such as using new preservatives systems or preservative free solutions allowed by innovating primary containers. Both strategies have benefits and weaknesses and must consider many factors like the time-consuming research, financial costs, patient compliance or environmental impact., L’œil, organe majeur du corps humain est complexe et irremplaçable. Avec son anatomie structurée, il cumule les protections lui permettant de faire face aux agressions, mais il est également sensible et soumis à un équilibre fragile. Au travers de l’étude de la composition des collyres, et plus précisément des conservateurs, l’un d’entre eux est devenu majoritaire : le chlorure de benzalkonium. Ce dernier, largement utilisé dans de nombreux autres domaines que l’ophtalmologie, possède des propriétés intéressantes mais de plus en plus d’études démontrent des effets secondaires, souvent lors des utilisations à long terme, tant sur des structures superficielles que profondes de l’œil. L’utilisation chronique de collyres concerne de nombreuses personnes dans le monde que ce soit pour traiter les allergies, la sécheresse oculaire ou le glaucome. Des alternatives sont apparues, par une suppression complète de conservateur via un emballage primaire innovant ou par l’utilisation de systèmes de conservation avec un profil toxicologique plus favorable. Ces deux stratégies possèdent leurs avantages et inconvénients, tenant compte de critères aussi nombreux que variés, comme l’investissement financier, temporel, l’observance du traitement ou l’impact environnemental.

International audience; The production and use of plastics has constantly increased over the last 30 years. Over one third of the plastics is used in disposables, which are discarded within three years of their production. Despite efforts towards recycling, a substantial volume of debris has accumulated in the environment and is slowly degraded to micro- and nanoplastics by weathering and aging. It has recently been discovered that these small particles can enter the food chain, as for example demonstrated by the detection of microplastic particles in honey, beer, salt, sea food and recently in mineral water. Human exposure has further been documented by the detection of plastic microparticles in human feces. Potential toxic consequences of oral exposure to small plastic particles are discussed. Due to lacking data concerning exposure, biodistribution and related effects, the risk assessment of micro- and nanoplastics is still not possible. This review focuses on the oral uptake of plastic and polymer micro- and nanoparticles. Oral exposure, particle fate, changes of particle properties during ingestion and gastrointestinal digestion, and uptake and transport at the intestinal epithelium are reviewed in detail. Moreover, the interaction with intestinal and liver cells and possibly resulting toxicity are highlighted.

International audience; Most lipophilic phycotoxins have been involved in human intoxications but some of these toxins have never been proven to induce human gastro-intestinal symptoms, although intestinal damage in rodents has been documented. For investigating the in vitro toxicological profile of lipophilic phycotoxins on intestine, the epithelial Caco-2 cell line has been the most commonly used model. Nevertheless, considering the complexity of the intestinal epithelium, in vitro co-cultures integrating enterocyte-like and mucus-secreting cell types are expected to provide more relevant data. In this study, the toxic effects (viability, inflammation, cellular monolayer integrity, modulation of cell type proportion and production of mucus) of four lipophilic phycotoxins (PTX2, YTX, AZA1 and OA) were evaluated in Caco-2/HT29-MTX co-cultured cells. The four toxins induced a reduction of viability from 20% to 50% and affected the monolayer integrity. Our results showed that the HT29-MTX cells population were more sensitive to OA and PTX2 than Caco-2 cells. Among the four phycotoxins, OA induced inflammation (28-fold increase of IL-8 release) and also a slight increase of both mucus production (up-regulation of mucins mRNA expression) and mucus secretion (mucus area and density). For PTX2 we observed an increase of IL-8 release but weaker than OA. Intestinal cell models integrating several cell types can contribute to improve hazard characterization and to describe more accurately the modes of action of phycotoxins.

The major goal endodontic therapy has been achieved by condensing filling materials into the root canal. It’s not uncommon to find excess material into the periapical tissue. It therefore becomes obligatory to use fillings materials that have acceptable biocompatibility.The purpose of this investigation was to obtain a «toxicity profile» of some endodontics materials and to compare our observations to results present in literature.The gutta-percha and five endodontic filling cements were tested «in vivo» and «in vitro».The in vivo biocompatibility involved the placement of the test material in 10 mm. Teflon tubes with an outer diameter of 1.3 mm which were implanted subcutaneously into rats. The implants were left in situ for periods of 30 and 90 days. The «hemolysis test» is designed for «in vitro» evaluations.The istological examination showed different intensity and extent cellular responses. In some cases severe infiltration of inflammatory cell and areas with necrotis were seen.At conclusion, the endodontics material evaluated showed slight, moderate and severe reactions; therefore a different pattern in tissue response., Dans le domaine odontologique, ainsi que dans les autres branches de la médecine, l’exigence d’améliorer constamment la biocompatibilité des matériaux utilisés comme thérapeutique, surtout quand ces matériaux sont destinés à «cohabiter» longtemps avec l’organisme, est de plus en plus ressentie.Dans le cadre d’un vaste programme de recherche, les auteurs se sont proposé d’évaluer la biocompatibilité réelle de quelques matériaux endodontiques choisis parmi les plus courants en Italie: c’est dans ce but qu’ont été effectués des tests «in vitro» (test de l’émolyse d’hématies de lapin) et «in vivo» (selon la technique de Safavi et al. qui prévoit l’implantation du matériel à tester dans le tissu sous-cutané du rat).Les résultats des tests ont démontré la variabilité du grade de biocompatibilité des matériaux soumis à l’expérimentation. Certains matériaux n’ont provoqué en effet qu’une légère réaction des tissus, d’autres ont déterminé la formation d’importants infiltrats de cellules inflammatoires, de macrophages, de cellules géantes de corps étranger et de colliquation nécrotique tissulaire.

International audience; Aluminum (Al) can be taken up from food, packaging, or the environment and thus reaches the human gastrointestinal tract. Its toxic potential after oral uptake is still discussed. The fate of different solid and ionic Al species during the passage through the digestive tract is the focus of this research, as well as the cellular effects caused by these different Al species. The present study combines the physicochemical processing of three recently studied Al species (metallic Al0, mineral Al2O3, and soluble AlCl3) in artificial digestion fluids with in vitro cell systems for the human intestinal barrier. Inductively coupled plasma mass spectrometry (ICP-MS) and small-angle X-ray scattering (SAXS) methods were used to characterize the Al species in the artificial digestion fluids and in cell culture medium for proliferating and differentiated intestinal Caco-2 cells. Cytotoxicity testing and cellular impedance measurements were applied to address the effects of digested Al species on cell viability and cell proliferation. Microarray-based transcriptome analyses and quantitative real-time PCR were conducted to obtain a deeper insight into cellular mechanisms of action and generated indications for cellular oxidative stress and an influence on xenobiotic metabolism, connected with alterations in associated signaling pathways. These cellular responses, which were predominantly caused by formerly ionic Al species and only at very high concentrations, were not impacted by artificial digestion. A two-directional conversion of Al between ionic species and solid particles occurred throughout all segments of the gastrointestinal tract, as evidenced by the presence of nanoscaled particles. Nevertheless, this presence did not increase the toxicity of the respective Al species.

Bien que la cosmétique soit avant tout une question de bien-être et de plaisir, elle doit aujourd’hui s’inscrire dans une démarche de transparence au niveau de la composition du produit mais également de son impact écologique. C’est d’ailleurs pour cette raison que nous voyons se multiplier depuis quelques années les applications de décryptage sur smartphone, permettant de scanner le produit et d’en afficher par ce biais la composition. Cela a pour but de lever le voile sur la composition des produits et donc d’aider le consommateur dans ses choix. Nous nous sommes demandés si ces applications étaient bel et bien un outil scientifique et non pas qu’une question de tendance numérique. Par ailleurs, ces applications, bien qu’utiles dans la vie quotidienne du consommateur, ne valent pas un avis de scientifique fondé sur des études et validé par le Code de la Santé Publique et le règlement cosmétique. Elles ont tout de même permis de mettre le doigt sur les nouvelles exigences des consommateurs quant à la composition du produit et son impact écologique et éthique.

Food and feed can be naturally contaminated by several mycotoxins and concern about the hazard of exposure to mycotoxin mixtures is increasing. Among farm animals, pig is one of the most sensitive farm animal to mycotoxins and it can be exposed, through its rich cereal diet, to high concentrations of mycotoxins. In total, 524 finished pig feeds samples from worldwide were analyzed for more than 800 metabolites using, LC- MS/MS (liquid chromatography tandem-mass spectrometry) method. Eighty-eight percent of the samples were co-contaminated with DON and other regulated and emerging mycotoxins. The toxicity of DON and the 10 most common emerging mycotoxins was analyzed on the viability of porcine intestinal epithelial cells (IPEC-1) over a 48 h period. Among the emerging mycotoxins, 3 of them (brevianamide F, cyclo-(L-Pro-L-Tyr), and tryptophol) did not alter cells viability. The other mycotoxins were ranked in the following order of toxicity: apicidin> enniatin A1> DON> beauvericin> enniatin B> enniatin B1> emodin> aurofusarin. The combined toxicity of DON and the 10 emerging mycotoxins was assessed based on their actual ratios found in pig feed. We observed that, despite the very high frequency of co- contamination, there was a poor correlation between the concentrations of DON and emerging mycotoxins. Thus, we assessed the toxic effects of three mixtures corresponding to situations to which animals may be exposed. Ratio #1 was calculated using the P25 (1st quartile) concentration of the emerging mycotoxin and P75 (3rd quartile) concentration of DON. Ratio #2 was calculated using the median (2nd quartile) concentration of DON and each emerging mycotoxin. Ratio #3 was the reverse scenario of ratio #1. Cytotoxicity analyses showed that, in most of the mixtures, the combined toxicity was similar to the one of DON alone. This tool was used to assess the toxicity of DON (3 and 10 µM) at different incubation times (0 to 20 h), by studying gene expression, ATP and total protein contents. The incubation medium was used to assess the quality of PCLS by measuring liver damage markers (alkaline phosphatase, lactate dehydrogenase, alanine transaminase, aspartate transaminase and total proteins). We showed that these markers were not affected by either incubation time or treatment. PCLS treated with 10 µM DON for 4 h or more, showed an alteration in the expression of certain genes. International audience; The genotoxicity of nano-structured synthetic amorphous silica (SAS), a common food additive, was investigated in vivo in rats. A 90-day oral toxicity study was performed according to OECD test guideline 408 and the genotoxicity of pyrogenic SAS nanomaterial NM-203 was assessed in several organs, using complementary tests. Adult Sprague-Dawley rats of both sexes were treated orally for 90 days with 0, 2, 5, 10, 20, or 50 mg SAS/kg bw per day. Dose levels were selected to approximate expected human dietary exposures to SAS. DNA strand breaks were evaluated by the comet assay in blood, bone marrow, liver, and spleen according to OECD test guideline 489; mutations induced in bone marrow precursors of erythrocytes were assessed by the Pig-a assay and chromosome/ genome damage by the micronucleus assay in blood (OECD test guideline 474) and colon. No treatment-related increases of gene (Pig-a) or chromosome/genome (micronucleus) mutations were detected in the blood. The percentage of micronucleated cells was not increased in the colon of treated rats. Among the organs analyzed by the comet assay, the spleen was the only target showing a weak but biologically relevant genotoxic effect.

International audience; Dechlorane related compounds (DRCs), including Dechlorane Plus (syn-DP and anti-DP), Dechlorane-601,-602, -603 and Chlordene Plus (CP), constitute a group of polychlorinated flame retardants (FRs) that are still of industrial use. In particular, DRCs have been detected in various nvironmental matrices and in different aquatic and terrestrial biota, thus exhibiting bioaccumulation and biomagnification potentials. The present study aimed at producing first occurrence data of a range of DRCs in Silurus spp. samples from different rivers located in France. Determination was carried out by gas chromatography high-resolution mass spectrometry after a sample clean-up based on a multilayer silica column and gel permeation chromatography. The concentration of monitored SDRCs ranged from 1.58 to 408 pg.g-1 wet weight (54 - 11100 pg.g-1 lipid weight). The fractional abundance of syn- and anti-DP stereoisomers was similar to that reported by other studies with an average equal to 0.60. Dec-601 was not detected in any sample. Detection frequencies ranged between 34 and 100% for other DRCs. Investigated correlations between DRCs and polychlorobiphenyls (PCBs) suggest a link with lipid content but independent contamination sources.

International audience; Organophosphate triesters (PEFRs) are used increasingly as flame retardants and plasticizers in a variety of applications, such as building materials, textiles, and electric and electronic equipment. They have been proposed as alternatives to brominated flame retardants. This updated review shows that biomonitoring has gained incrementally greater importance in evaluating human exposure to PEFRs, and it holds the advantage of taking into account the multiple potential sources and various intake pathways of PEFRs. Simultaneous and extensive internal exposure to a broad range of PEFRs has been reported worldwide. Their metabolites, mainly dialkyl or diaryl diesters, have been used as biomarkers of exposure and have been ubiquitously detected in the urine of adults and children in the general population. Concentrations and profiles of PEFR urinary metabolites are seen to be variable and are highly dependent on individual and environmental factors, including age, country regulation of flame retardants, and types and quantities of emissions in microenvironments, as well as analytical procedures. Additional large biomonitoring studies, using a broad range of urinary diesters and hydroxylated metabolites, would be useful to improve the validity of the biomarkers and to refine assessments of human exposure to PEFRs.; Les triesters organophosphorés (RFOPs) sont de plus en plus utilisés comme retardateurs de flamme et comme plastifiants dans de nombreuses applications, tels que les matériaux de construction, les textiles et les équipements électriques et électroniques. Ils sont proposés comme alternatives aux retardateurs de flamme bromés. Cette synthèse actualisée des données bibliographiques montre que la surveillance biologique a progressivement acquis une plus grande importance dans l’évaluation de l’exposition humaine aux RFOPs. Elle a comme avantage de prendre en compte les multiples sources potentielles et les différentes voies d’absorption des RFOPs. Ces études montrent une exposition interne répandue et simultanée à un large éventail de RFOPs, dans le monde entier. Les métabolites des RFOPs, principalement des diesters dialkyl ou diaryl, ont été utilisés comme biomarqueurs d’exposition et ils ont été détectés de façon ubiquitaire dans l’urine des adultes et enfants de la population générale. Les concentrations et les profils des métabolites urinaires sont variables. Ils sont fortement influencés par des facteurs individuels et environnementaux, notamment l’âge, la réglementation du pays de retardateurs de flamme et les types et les quantités des émissions de RFOPs à partir des micro-environnements, ainsi que par les méthodes d’analyse. Quelques études de biométrologie urinaire ont mis en évidence des expositions professionnelles aux RFOPs. Des études de biosurveillance à grande échelle utilisant un large éventail de diesters et métabolites hydroxylés urinaires seraient utiles pour optimiser le choix des biomarqueurs et affiner l’évaluation de l’exposition humaine aux RFOPs.

Les patients glaucomateux ont souvent besoin d’un traitement local hypotonisant pendant de nombreuses années. Cette administration chronique des collyres a été impliquée dans la survenue des pathologies de la surface oculaire. L’objectif de cette étude est de déterminer la prévalence des pathologies de la surface oculaire chez les patients traités pour un glaucome ou une hypertonie oculaire. Il s’agit d’une étude observationnelle prospective lors de la consultation spécialisée des patients glaucomateux réalisée au service d’ophtalmologie du CHU HASSAN II de Fès. Tous les patients ont bénéficié d’un examen ophtalmologique complet comprenant une évaluation clinique de la surface oculaire. Un questionnaire dérivé de l’Ocular Surface Disease Index (OSDI) a permis d’évaluer le retentissement de cette atteinte et la qualité de vie des patients. Nous avons sélectionné 96 patients âgés entre 10 et 85 ans. L’acuité visuelle moyenne était de 0,32 LogMar, soit 5/10e.La moyenne de l’ancienneté du glaucome est de 5 ans, dans cette étude, 62 patients (64 %) présentaient des symptômes d’atteinte sévère de la surface oculaire. une atteinte légère chez 17 patients (18 %).Alors que 17 patients (18 %) ne présentent pas de symptôme. Ces patients ont été répartis en fonction de la classification clinique A, B et C. Le groupe A (pas d’atteinte), le groupe B (atteinte modérée) et le groupe C (atteinte sévère) comptaient respectivement 26 (27 %), 21 (22 %) et 49 (51 %) patients. Background Nanofibrillation of proteins by heating at extremely acidic condition for long durations (several hours to days) is studied enthusiastically in food science. The process progresses by the unidirectional self-assembly of peptides as building units of the fibrils. Scope and approach This communication provides a review on the underlying mechanism of protein fibrillation, and various technological properties of the fibrils, followed by discussing their biological and cellular effects. Key findings and conclusions Fibrillation of proteins and addition of the fibrils into liquid foods causes a significant increase of apparent viscosity. Nevertheless, certain post-fibrillation processes such as freeze-drying may result in viscosity reduction. Fibrils form cohesive viscoelastic interfaces, bringing about high foam and emulsion stability. The presence of non-fibrillated peptides and low molecular weight surfactants influence the foaming and emulsification properties of fibrillated protein solutions. Fibrillated protein can yield cold-set gels at extremely low concentrations, which is attributed to formation of space filling networks. Reinforcing polymeric films, conferring hydrophilic character to graphene, developing drug and nutraceutical delivery vehicles such as microcapsules, microgels and fibrillosome and utilization (mostly as scaffolds) in fabrication of biosensors and bio-sorbents are the other applications of protein fibrils. Fibrillation may increase the antioxidant activity of proteins. It can also influence protein digestibility. Fibrils do not exert any major toxicity towards human cell lines and can be exploited as biomimetic cell culture platforms and cellular transport shuttles.

Cet article présente une revue bibliographique sur la désinfection des eaux de piscines par le brome, sur les mécanismes de formation et le mode d'action du désinfectant, l'acide hypobromeux. L'examen des principaux sous-produits de bromation (bromamines, trihalométhanes, acides haloacétiques, haloacétonitriles, haloaldéhydes, bromate, nitrosamines, etc.) montre que certains de ces sous-produits sont présents dans l'eau de piscine à des concentrations de quelques µg∙L-1 à quelques dizaines de µg∙L-1. L'étude de leur toxicité aiguë et chronique confirme que ces composés doivent être surveillés (en particulier l'acide dibromoacétique et le bromate) pour assurer la sécurité des nageurs et du personnel technique. D'autre part, l'étude de l'efficacité biocide du brome comparé au chlore, vis-à-vis des bactéries pathogènes présentes dans l'eau de piscine, démontre que les espèces de bactéries Escherichia coli et Enterococcus faecalis sont plus sensibles au brome, à la différence de Pseudomonas aeruginosa qui présente une plus grande résistance. De plus, sur le plan médical, plusieurs cas de dermatites de contact irritantes sont rapportés lors de l'utilisation du brome, même si certaines études révèlent que les bromamines sont non irritantes, inodores et désinfectantes. Cependant, le brome présente une plus grande activité bactéricide que le chlore dans des eaux légèrement alcalines et relativement chaudes et peut être alors une alternative au chlore dans le traitement des piscines atypiques, même s'il demeure beaucoup plus onéreux que le chlore., This paper presents a bibliographical review on the disinfection of swimming pool water with bromine. The formation mechanisms and the mode of action of the disinfectant, hypobromous acid, are discussed. The review of the main bromination by-products (bromamines, trihalomethanes, haloacetic acids, haloacetonitriles, bromate, haloaldehydes, nitrosamines, etc.) shows that some of these by-products are present in pool water at concentrations of a few µg∙L-1 to a few tens of µg∙L-1. The study for acute and chronic toxicity confirms that these compounds should be monitored (especially dibromoacetic acid and bromate) to ensure the safety of swimmers and technical staff. The biocidal effectiveness of bromine versus chlorine, against pathogenic bacteria present in swimming water, is detailed. It has been shown that the bacterial species Escherichia coli, and Enterococcus faecalis are more sensitive to bromine, unlike Pseudomonas aeruginosa that is resistant to bromine. It has also been reported several cases of irritant contact dermatitis when using bromine though some studies have revealed bromamines to be disinfecting, odourless and non-irritating. Nevertheless, bromine exhibits greater bactericidal activity than chlorine in slightly alkaline and warm water and could replace chlorine in the treatment of atypical swimming pools, although chlorine is less costly than bromine.
