Tomato plants (Lycopersicon esculentum Mill.) inoculated with tomato white necrosis virus (VNBT) and treated with Virazole (1-beta-D-ribofuranosyl--1, 2, 4-triazole-3-carboxamide) at a concentration of 500 mg/l developed systemic virus symptoms in only 40 per cent of the plants in which a remarkable reduction in virus concentration was also observed. From inoculated and Virazole-treated plants which had produced no symptoms, no virus could be recovered. This result suggests that Virazole may inhibit replication of VNBT in tomato.

When tobacco protoplasts were inoculated with tobacco rattle virus, the proportion infected was inversely related to the protoplast concentration between 0.5 X 10(5) and 4 X 10(5) per milliliter when the inocula contained phosphate or Tris-HCL buffers, but was independent of protoplast concentration when using citrate buffer. The decrease in infection obtained at high protoplast concentrations in phosphate was diminished by increasing the poly-L-ornithine (PLO) concentration. At usual PLO concentrations, inocula in phosphate produced more infection that those in citrate, but at some suboptimal PLO concentrations the position was reversed. These effects may result from differences in the reaction between PLO and the different buffer anions.

The effect of several concentrations of nitrogen (0, 21, 70, 210, 420 and 630 ppm) on the growth and multiplication of cucumber mosaic virus in cucumber (Cucumis sativus L. Barshati) grown in sand culture was studied. The effect of nitrogen nutrition on the concentration of virus appeared to be directly related to vegetative growth of the host. 420 ppm level yielding the best growth also showed the highest virus concentration and total nitrogen percentage at 30th day of infection.

In a synchronous infection of cowpea chlorotic mottle virus in cowpea leaves, the three largest viral RNA components were synthesized throughout the infection at relatively constant ratios of two molecules of component 3 to one each of components 1 and 2. Component 4 (the coat protein message) was produced minimally during the initial stages of the infection, and as the infection progressed the proportion of component 4 continuously increased. The replicative forms of components 1, 2, and 3 were produced with kinetics similar to those of the corresponding single-stranded molecules.

The structure of turnip crinkle virus has been determined at 3.2 A resolution, using the electron density of tomato bushy stunt virus as a starting point for phase refinement by non-crystallographic symmetry. The structures are very closely related, especially in the subunit arm and S domain, where only small insertions and deletions and small co-ordinate shifts relate one chain to another. The P domains, although quite similar in fold, are oriented somewhat differently with respect to the S domains. Understanding of the structure of turnip crinkle virus has been important for analyzing its assembly, as described in an accompanying paper.

Ultraviolet light (UV) sensitivity and photoreactivation of algal virus (cyanophage) LPP-1 were studied after multiplication in host alga Plectonema boryanum in presence of 5-bromouracil (5-BU) alone and in conjunction with sulfanilamide. Virus particles containing 5-BU were more sensitive towards UV and also showed low photoreactivation. There was less incorporation of 5-BU in virus without pretreatment of host alga with sulfanilamide, an inhibitor of thymine synthesis. 5-BU-induced short trichome mutants of Plectonema boryanum were isolated. These mutants grew slowly in liquid medium as well as on agar plates and differed in other morphological characters. Reversion of short trichome mutants was observed with a frequency of about 10(-3), but revertants were different from parent alga. The short trichome mutants were sensitive to virus LPP-1 and resistant towards UV.

Highly infectious, purified AS-1 virus was obtained by treating virus-induced culture lysates with bentonite followed by rotary evaporation concentration and density gradient centrifugation.

In Anacystis nidulans, upon infection with cyanophage AS-1, after a lag period of 1 h the level of deoxyribonuclease (DNase) activity increaded rapidly up to 15- to 20-fold in 4 to 5 h in the light. In contrast, the ribonuclease and phosphomonoesterase activities increased significantly only 4 to 5 h after infection, i.e. as late as 1 h prior to lysis. In complete darkness, the nuclease levels remained unaltered. However, when the infected cells were exposed to light for 1 or 2 h after infection, the DNase level increased essentially to the same extent in the dark as in continuous light, although the complete replication cycle of the virus was impaired in the dark and cells lysed only in the continuously illuminated cultures. Inhibition of photosystem II with 3-(3,4-dichlorophenyl)-1-dimethylurea during the early illumination period strongly decreased the subsequent, infection-dependent increase in DNase activity in the dark. The virus-induced increase in DNase activity was also inhibited by chloramphenicol. The data suggest that, in spite of the obligate photoautotrophic nature of A. nidulans, dark metabolism is able to support fully the formation of some specific proteins if the triggering of their synthesis takes place in light.

Published data on the thermostability, longevity in vitro, and tolerance to dilution of viruses in sap from infected plants have been analyzed. It appears that none of these properties is a reliable criterion for virus classification or identification.

Several Nicotiana species and cultivars, with and without the N gene, were screened for the effect of polyacrylic acid (PAA) and salicylic acid (SA) on b-protein production and induced resistance. Whilst SA was effective in producing either b-proteins and resistance to a challenge viral infection in all the cultivars tested, the effectiveness of PAA appeared cultivar dependent. Only the cultivars Xanthi-nc (NN), Xanthi (nn) and hybrids between Xanthi-nc (NN) and other N. tabacum cultivars responded to PAA. This is the first time that the genetic determinant for the PAA response has been shown to be sexually transmitted as a dominant character, and also that it is distinct from the N gene.

Alfalfa mosaic virus (AMV) was found to be transmitted through seeds of Nicandra physaloids L. The average seed transmission rate of the AMV isolates T6, LMBG-4 and ST amounted to 23, 4 and 0 per cent, repectively. In the cytoplasm of parenchyma cells of embryo cotyledons of seeds from plants infected with the T6 isolate, electron microscopy revealed AMV aggregates of type 2A and aggregations of irregularly viral particles with a tendency to subparallel alignment, representing early stages of type 2A aggregates.

The adsorption rate of the cyanovirus N-1 infecting the nitrogen-fixing blue-green alga Nostocmuscorum decreased with aging of algal cultures and the virus failed to adsorb to the dead host cells. The adsorption rate declined in saline magnesium chloride solution compared to that in algal growth medium. The addition of amino acids like L-tryptophan and L-phenylalanine failed to enhance the adsorption rate of the virus. Optimal pH of adsorption was 7.6 to 8;1.

Cells of unicellular cyanobacteria of typological group Ia, containing approximately 50 mol% guanine + cytosine (G+C) in their DNA (R. Y. Stanier, R. Kunisawa, M. Mandel, and G. Cohen-Bazire, Bacteriol. Rev. 35:171-205, 1971), were susceptible to infection by the cyanophage AS-1. Cyanobacteria of the same typological group, containing approximately 65 mol% G+C in their DNA, did not adsorb the cyanophage AS-1 or adsorbed it at a low rate. AS-1 was not propagated by any of the investigated strains with a high G+C content in their DNA. However, cells of strains 6907 and 6911 were lysed by cyanophage AS-1. A comparison of the host range of this phage with the lipopolysaccharide composition of host and non-host cell walls suggests that lipopolysaccharides are involved in the adsorption process. About 8 microgram of lipopolysaccharide per ml from host strains inactivated 50% of the particles of a solution containing 100 PFU/ml after 60 min of incubation at 30 degrees C. Material with receptor activity was extracted from the host strain Anacystis nidulans KM. The extract was purified of glycolipids and pigments, and a fraction showing receptor activity was isolated. This fraction contained three polypeptides of molecular weights between 54,000 and 64,000. Heat and protease treatment of whole cells and of isolated receptor material decreased the receptor activity. The fluorescence intensity of A. nidulans cells labeled with 1-anilino-8-naphthalene sulfonate was increased when AS-1 was adsorbed to these cells. The participation of lipopolysaccharides and proteins in the formation of the receptor complex is discussed.

Inoculation of the leafhopper cell line AC-20 with wound tumor virus resulted in a productive noncytopathic infection with no detectable alteration of cellular protein synthesis. Virus-specific polypeptide synthesis, detectable by 8 h postinoculation, increased in a linear fashion, reaching a peak (approximately 10 to 15% of total protein synthesis) by 48 h postinoculation. The rate of viral protein synthesis continued at this level for several days but declined, relative to cellular protein synthesis, as infected cells were passaged. By passage 10, the synthesis of viral polypeptides was reduced to a level approximately 5% of that observed at 48 h postinoculation. Viral protein synthesis was not stimulated by superinfection. Viral antigens and infectious virus persisted in the majority (greater than 90%) of cells in an infected culture even after more than 100 passages. The synthesis of wound tumor virus polypeptides in infected insect vector cells appears to be regulated in a coordinated and selective manner.

A simple procedure for the isolation of double-stranded (ds) RNA from virus-infected plants is described. The method is based on grinding plant tissue in 4% p-aminosalicylic acid and recovery of ds RNA by phenol extraction and precipitation with 30% ethanol. The presence of both negative and positive virus RNA strands in RNA fractionated in agarose gels was verified by Northern blot hybridization with polynucleotide kinase labelled genomic RNA or complementary DNA (cDNA) probes. The procedure enabled detection of three major ds RNA species (MWs 4.2, 1.05 and 0.48 X 10(6)) and at least 4 minor bands with estimated MWs of 3.5, 2.5, 2.2 and 2.0 X 10(6) in Nicotiana tabacum plants systemically infected with tobacco mosaic virus (TMV). Cucumber mosaic virus (CMV)-infected Pachystachys coccinea plants contained 2 minor bands of MWs 0.49 and 0.35 X 10(6) in addition to the previously described 4 major ds RNAs and ds CARNA 5 (MW 0.22 X 10(6)). The patterns of ds RNA are useful for diagnosing natural infections of CMV and TMV in N. glauca plants and of citrus tristeza virus in Citrus spp.
