204. Direct mass spectrometric analysis of ozonides: application to unsaturated glycerophosphocholine lipids

Abstract

The reaction of ozone with double bonds present in glycerophosphocholine lipids results in formation of ozonides that can be directly analyzed by mass spectrometry as either positive or negative molecular ion species generated by electrospray ionization. Polyunsaturated fatty acyl groups esterified to the phospholipid yielded a mixture of ozonide species with the maximum number of ozone molecules added equal to the total number of double bonds. Ozonide decomposition resulted in [Omega]-aldehyde and [Omega]-carboxylic acid products as revealed by ESIMS. Collisional activation of the ozone adducts for mono-and polyunsaturated phospholipids gave rise to fragment ions indicative of the position of the double bonds in these molecules. The major decomposition pathway for either positive or negative ozonide ion species involved charge remote fragmentation of the ozonide initiated by homolytic cleavage of the peroxide bridge followed by rearrangement to form the [Omega]-aldehyde and [Omega]-carboxylate acyl species. The reaction of ozone with phospholipids containing polyunsaturated fatty acyl groups is a useful method to probe the position of double bonds by electrospray ionization mass spectrometry.

Abstract

The very unstable biochemical intermediate, 5(S)-hydroxy-12-oxo-6(Z),8(E),10(E),14(Z)-eicosatetraenoic acid is synthesized by artificial methods. The synthesis is performed by the incubation of 12-oxo-LTB4 with human keratinocytes along with two new intriguing mediators. The synthesized LTB is compared with other similar compounds and its properties are determined.

Triply convergent strategy is used for the sequential union of propargylic arsonium salts with pyranosides and furanose to enable the total asymmetric synthesis of 3-hydroxyleukotriene B4(3-OH-LTB4). Palladium-mediated coupling is the vital step in the process. 3(S)-isomer is the major component in the enzyme produced product. Chromatography and mass spectra are employed for stereochemical assignment.

Abstract

The utilization of tandemmass spectrometry with mild acid hydrolysis is discussed by differentiating between three glycerophosphoethanolamine phospholids (GPE) subclasses and identifying the arachidonate-containing molecular species of GPE in human polymorphonuclear leukocytes. The need for derivatization and additional chromatography are circumvented by the applied rapid method of analysis. The application of fast atom bombardment ionization and tandem instruments has significantly improved the analysis of complex GPE.
