201. Quasar Microlensing Statistics and Flux-ratio Anomalies in Lens Models

Abstract

Precise lens modeling is a critical step in time delay studies of multiply imaged quasars, which are key for measuring some important cosmological parameters (especially H _0 ). However, lens models (in particular those semi-automatically generated) often show discrepancies with the observed flux ratios between the different quasar images. These flux-ratio anomalies are usually explained through differential effects between images (mainly microlensing) that alter the intrinsic magnification ratios predicted by the models. To check this hypothesis, we collect direct measurements of microlensing to obtain the histogram of microlensing magnifications. We compare this histogram with recently published model flux-ratio anomalies and conclude that they cannot be statistically explained by microlensing. The average value of the model anomalies (0.74 mag) significantly exceeds the mean impact of microlensing (0.33 mag). Moreover, the histogram of model anomalies presents a significant tail with high anomalies (∣Δ m ∣ ≥ 0.7 mag), which is completely unexpected from the statistics of microlensing observations. Microlensing simulations neither predict the high mean nor the fat tail of the histogram of model anomalies. We perform several statistical tests which exclude that microlensing can explain the observed flux-ratio anomalies (although Kolmogorov–Smirnov, which is less sensitive to the tail of the distributions, is not always conclusive). Thus, microlensing cannot statistically explain the bulk of flux-ratio anomalies, and models may explore different alternatives to try to reduce them. In particular, we propose to complement photometric observations with accurate flux ratios of the broad emission lines obtained from integral field spectroscopy to check and, ideally, constrain lens models.

Abstract

Biomass storage is an essential requirement in the supply chain of bio-refineries and power plants. This research aims to evaluate the influence of long-term outdoor storage (1 year) of baled rockrose (Cistus laurifolius L.) shrub biomass on biofuel’s quality, pre-treatment processes and on combustion emissions in an industrial boiler. The raw material was obtained from different rockrose shrublands in north central Spain. A total of 233 tWM (tones of wet matter) of biomass were used to produce biofuels (30 mm of milled biomass and Ø 8 mm pellets) in the pre-treatment pilot plants at CEDER-CIEMAT. The combustion tests were conducted in an industrial moving grate boiler with a thermal power of 50 MWth, in a 17 MWe power plant. Outdoor storage improved some biofuel quality parameters, mainly the reduction in ash content, which allowed 30 mm of milled material to be classified as class I1 (ISO 17225-9:2022) and pellets as class I3 (ISO 17225-2-2021). No significant differences were observed in the total specific mass flow and energy consumption in the pre-treatment processes. The combustion tests had similar results, with the emissions being below the limits established in the directive (EU) 2015/2193. The results obtained indicated that the 1-year outdoor storage of rockrose-baled biomass, under Mediterranean conditions, was feasible for its subsequent use as biofuel.

Abstract

We find that the Fe III$\lambda\lambda$2039-2113 spectral feature in quasars appears systematically redshifted by amounts accountable under the hypothesis of gravitational redshift induced by the central supermassive black hole. Our analysis of 27 composite spectra from the BOSS survey indicates that the redshift and the broadening of the lines in the Fe III$\lambda\lambda$2039-2113 blend roughly follow the expected correlation in the weak limit of Schwarzschild geometry for virialized kinematics. Assuming that the Fe III UV redshift provides a measure of $M_{BH}\over R$ (${\Delta \lambda\over \lambda}\simeq{3\over2}{G\over c^2} {M_{BH}\over R}$) and using different estimates of the emitting region size, $R$ (either from gravitational microlensing, reverberation mapping or from the scaling of size with intrinsic quasar luminosity), we obtain masses for 10 objects which are in agreement within uncertainties with previous mass estimates based on the virial theorem. Reverberation mapping estimates of the size of the Fe III$\lambda\lambda$2039-2113 emitting region in a sample of objects would be needed to confirm the gravitational origin of the measured redshifts. Meanwhile, we present a tentative black hole mass scaling relationship based on the Fe III$\lambda\lambda$2039-2113 redshift useful to measure the black hole mass of one individual object from a single spectrum.

204. A Second-Order Moment of Microlensing Variability as a Novel Tool in Extragalactic Research

Abstract

We define a second-order moment of the observational differential microlensing curves that can be used to impose constraints on physical properties of lensed quasars. We show that this quantity is sensitive both to variations in the source size and the deflector mass. We formulize a methodology to recover the source size from the observational measurements when the mass spectrum is fixed. As a case study, we test it with a sample of four quadruple lenses, both in simulated scenarios and with real data from the $\textit{Chandra X-ray Observatory}$. In our simulations with a uniform stellar population the method works best to detect sources around $0.1$ Einstein radii, giving correct upper/lower limits for much smaller/bigger sizes without requiring a big leap in additional computational effort as compared to a single-epoch approach, yet taking advantage of multi-epoch information. We apply the method to a small sample of X-ray data from four objects assuming a range of star masses, and obtain a degeneracy relation between the source size and deflector mass. Combined with previous estimates for the size of the X-ray corona, the degeneracy relation suggests that X-ray microlensing is mainly induced by planetary mass objects.

Abstract

We study the BELs in a sample of 11 gravitationally lensed quasars with at least two epochs of observation to identify intrinsic variability and to disentangle it from microlensing. To improve our statistical significance we also include 15 systems with single-epoch spectra. MgII and CIII] emission lines are only weakly affected by microlensing, but CIV shows strong microlensing in some cases, even for regions of the line core, associated with small projected velocities. However, excluding the strongly microlensed cases, there is a strikingly good match between the red wings of the CIV and CIII] profiles. Analysis of these results supports the existence of two regions in the BLR, one that is insensitive to microlensing (of size $\gtrsim$ 50 light-days and kinematics not confined to a plane) and another that shows up only when it is magnified by microlensing (of size of a few light-days, comparable to the accretion disk). Both regions can contribute in different proportions to the emission lines of different species and, within each line profile, to different velocity bins, all of which complicates detailed studies of the BLR based on microlensing size estimates. The strength of the microlensing indicates that some spectral features that make up the pseudo-continuum, such as the shelf-like feature at {\lambda}1610 or several FeIII blends, may in part arise from an inner region of the accretion disk. In the case of FeII, microlensing is strong in some blends but not in others. This opens up interesting possibilities to study quasar accretion disk kinematics.

Abstract

QUIJOTE (Q-U-I JOint TEnerife) is an experiment designed to achieve CMB B-mode polarization detection and sensitive enough to detect a primordial gravitational-wave component if the B-mode amplitude is larger than r = 0.05. It consists in two telescopes and three instruments observing in the frequency range 10-42 GHz installed at the Teide Observatory in the Canary Islands, Spain. The observing strategy includes three raster scan deep integration fields for cosmology, a nominal wide survey covering the Northen Sky and specific raster scan deep integration observations in regions of specific interest. Abstract

BackgroundOvercoming β-lactam resistance in pathogens such as Pseudomonas aeruginosa is a major clinical challenge. Rapid molecular diagnostics (RMDs) have the potential to inform selection of empiric therapy in patients infected by P. aeruginosa.MethodsIn this study, we used a heterogeneous collection of 197 P. aeruginosa that included multidrug-resistant isolates to determine whether 2 representative RMDs (Acuitas Resistome test and VERIGENE gram-negative blood culture test) could identify susceptibility to 2 newer β-lactam/β-lactamase inhibitor (BL-BLI) combinations, ceftazidime/avibactam (CZA) and ceftolozane/tazobactam (TOL/TAZO).ResultsWe found that the studied RMD platforms were able to correctly identify BL-BLI susceptibility (susceptibility sensitivity, 100%; 95% confidence interval [CI], 97%, 100%) for both BLs-BLIs. However, their ability to detect resistance to these BLs-BLIs was lower (resistance sensitivity, 66%; 95% CI, 52%, 78% for TOL/TAZO and 33%; 95% CI, 20%, 49% for CZA).ConclusionsThe diagnostic platforms studied showed the most potential in scenarios where a resistance gene was detected or in scenarios where a resistance gene was not detected and the prevalence of resistance to TOL/TAZO or CZA is known to be low. Clinicians need to be mindful of the benefits and risks that result from empiric treatment decisions that are based on resistance gene detection in P. aeruginosa, acknowledging that such decisions are impacted by the prevalence of resistance, which varies temporally and geographically.

Abstract

Las cáscaras de naranja (Citrus sinensis) son residuos lignocelulósicos abundantes que pueden ser utilizados como fuente de carbono para obtener sólidos con potencial catalítico en la transformación de terpenos y sus óxidos en productos de valor agregado. Esta investigación buscó evaluar la isomerización de óxido de α-pineno a aldehído canfolénico, utilizando un catalizador obtenido de cáscaras de naranja. El material OAC-Zn se obtuvo por activación de las cáscaras con ZnSO4.7H2O, seguido de tratamiento térmico a 500 °C; también se obtuvo un sólido de la pirólisis de la cáscara a 500 °C (OC-500). Mediante DRX se identificó la presencia de ZnO y ZnS en OAC-Zn; los análisis por TGA indicaron estabilidad térmica en OAC-Zn y OC-500; las imágenes SEM mostraron superficies porosas de diferente morfología, así como la presencia de microporosidad en OC-500 y de mesoporosidad en OAC-Zn, que fueron confirmadas mediante adsorción física de nitrógeno. En los análisis EDX se identificaron los elementos C, O, Zn y S en OAC-Zn; los resultados de TPD-NH3 mostraron que los sólidos contenían acidez media y débil. Se sintetizó aldehído canfolénico con selectividad de 96 % sobre un material carbonoso mesoporoso, a partir de cáscaras de naranja utilizando ZnSO4.7H2O como agente activador

Abstract

Introduction Psychotic disorders have a huge impact on social functioning, which is the ability to stablish and maintain social activities such as interpersonal relationships and self-care activities of daily living. Research data support that the early intervention in people who have experienced a first episode of psychosis (FEP) -based on a multidisciplinary treatment including both psychopharmacological and psychosocial treatments-, has a relevant role in a favorable evolution. AGES-Mind study is based on manualized psychotherapeutic interventions for people with first-psychosis episodes. Objectives To describe the use of rehabilitation resources and social functioning in a group of people with FEP who were included in a psychotherapeutic group program versus a control group, at 12 and 24 months since the beginning of the intervention. Methods Longitudinal, analytical, observational, retrospective study on a cohort of 46 patients with first-episode psychosis within the last 5 years. 23 patients received group psychotherapy in the context of the AGES-Mind study and they were compared with 23 control patients who did not receive a group intervention (treatment as usual). Controls were matched by age, gender and time elapsed since the first episode of psychosis with those exposed to the intervention. Sociodemographic data, social functioning (self-care, social activities, social relationships, and behavior) and use of rehabilitation resources outcome variables were assessed. Results Significant differences were found regarding participation in social activities in the intervention group versus control group at 24 months. No significant differences were found in other dimensions of social functioning or in the use of rehabilitation resources. Abstract

Introduction Early intervention on a first psychotic episode is fundamental for a more favorable prognosis, and it usually combines pharmacological treatment, which mainly affects positive psychotic symptoms, with interventions that can improve the rest of the symptoms and associated problems such as deterioration in social functioning (Harvey & Penn, 2010; Fusar-Poli, McGorry & Kane, 2017). While Mindfulness is gaining more and more prominence in the field of psychotherapy (Chan et al., 2019; Cillesen et al., 2019), social cognition and social functioning are being researched as key targets on which to intervene after a first psychotic episode (Green, Horan & Lee, 2015). SocialMIND® is a mindfulness-based social cognition training tailor-made to improve social functioning in people who have suffered a first psychotic episode within the last five years. It is currently being compared with a group Psychoeducational Multicomponent Intervention (PMI) in a Randomized Controlled Trial (RCT) (Mediavilla et al., 2019). Both group psychotherapies include 17 sessions delivered over a 9 month period: 8 weekly sessions, 4 biweekly sessions and 5 monthly sessions. The results of SocialMIND® at 8 weeks showed improvements in social cognition and social functioning, specifically on affective social cognition and self-care (Mediavilla et al., 2021). Objectives To evaluate the efficacy of SocialMIND® in improving social functioning, measured by the Personal and Social Functioning (PSP) scale 16 weeks after starting the intervention, in people who have suffered a first psychotic episode in the last 5 years. Methods Randomized, controlled pilot trial (use of a psychoeducational multicomponent intervention or PMI as active comparator) of two parallel groups (SocialMIND® and PMI) with a 1:1 ratio using a blind evaluator. Results No statistically significant differences were found in the social functioning variable between the two treatment arms. Intragroup differences are observed in other secondary variables studied (social cognition) 16 weeks after starting the interventions. Conclusions SocialMIND® has not been shown to be more effective than a PMI in improving social functioning at 16 weeks after starting the intervention in people who have suffered a first psychotic episode in the five years prior to being included in the study. Conclusions After controlling for potentially confounding variables as sociodemographic, age and time since first-episode, participating in a group psychotherapeutic program does not seem to improve clinical variables or use of resources. Abstract

Previous research indicates that developmental contexts influence language development. These developmental contexts include bilingualism, which is the focus of the current paper. This study analyses the influence that the acquisition of a language, Catalan, has when the simultaneous acquisition of another similar language, Spanish, occurs. This influence is examined according to degree of exposure to the languages. The parents of 801 children completed the Catalan version of the MacArthur-Bates CDI-II questionnaire, which assesses the acquisition of vocabulary, morphology and grammatical complexity. Subjects were divided into three groups according to age and linguistic context (Catalan monolingual, medium bilingual and familiar bilingual). The results do not reveal substantial differences according to linguistic context. However, children aged 26--30 months with a medium bilingual profile show a higher level of vocabulary as compared to the other groups. The results are discussed in relation to the sociolinguistic context, caregiver diversity, typological distance between the languages and developmental stages of language acquisition. 227. Resolving the Innermost Region of the Accretion Disk of the Lensed Quasar Q 2237+0305 through Gravitational Microlensing

Abstract

We study three high magnification microlensing events, generally recognized as probable caustic crossings, in the optical light curves of the multiply imaged quasar Q 2237+0305. We model the light curve of each event as the convolution of a standard thin disk luminosity profile with a straight fold caustic. We also allow for a linear gradient that can account for an additional varying background effect of microlensing. This model not only matches noticeably well the global shape of each of the three independent microlensing events but also gives remarkably similar estimates for the disk size parameter. The measured average half-light radius, $R_{1/2}=(3.0\pm 1.5)\sqrt{M/0.3M\odot}$ light-days, agrees with previous estimates. In the three events, the core of the magnification profile exhibits "fine structure" related to the innermost region of the accretion disk (located at a radial distance of $2.7\pm 1.4$ Schwarzschild radii according to our measurement). Relativistic beaming at the internal rim of the accretion disk can explain the shape and size of the fine structure, although alternative explanations are also possible. This is the first direct measurement of the size of a structure, likely the innermost stable circular orbit, at $\sim 3$ Schwarzschild radii in a quasar accretion disk. The monitoring of thousands of lensed quasars with future telescopes will allow the study of the event horizon environment of black holes in hundreds of quasars in a wide range of redshifts $(0.5

RESUMEN La hipertensión arterial es el factor de riesgo cardiovascular más prevalente. A pesar del tratamiento farmacológico, un alto porcentaje de pacientes no consiguen un adecuado control. La denervación renal es una intervención mínimamente invasiva para el tratamiento de la hipertensión que implica la interrupción de los nervios simpáticos renales mediante un abordaje con catéter. Los estudios iniciales mostraron resultados prometedores, pero los controvertidos resultados del ensayo SYMPLICITY HTN-3 llevaron al abandono de la técnica. En los últimos 3 años han aparecido los resultados de nuevos ensayos clínicos, con nuevos dispositivos y en diferentes poblaciones, que demuestran definitivamente la eficacia de la denervación renal. En este documento de posicionamiento conjunto de la Sociedad Española de Hipertensión-Liga Española para la Lucha contra la Hipertensión Arterial (SEH-LELHA) y la Asociación de Cardiología Intervencionista de la Sociedad Española de Cardiología (ACI-SEC) se revisa la evidencia disponible sobre la eficacia y la seguridad de la denervación renal en el tratamiento de la hipertensión. A partir de los resultados de los ensayos clínicos, se generan recomendaciones sobre qué pacientes y en qué condiciones podrían ser candidatos a una denervación renal.

Abstract

ABSTRACT Hypertension is the most prevalent cardiovascular risk factor. Despite pharmacological treatment, a high percentage of patients do not achieve an adequate blood pressure control. Renal sympathetic denervation is a minimally invasive intervention for the management of hypertension involving the interruption of the renal artery sympathetic nervous system using a catheter-based approach. The early studies showed promising results, but the controversial results coming from the SYMPLICITY HTN-3 trial sent this technique into oblivion. Over the last 3 years, new clinical trials have appeared including new devices used in different populations, which definitively proves the effectiveness of renal sympathetic denervation. This joint position statement from the Spanish Society of Hypertension-Spanish League for Combating High Blood Pressure (SEH-LELHA), and the Interventional Cardiology Association of the Spanish Society of Cardiology (ACI-SEC) reviews the evidence available on the efficacy and safety profile of renal sympathetic denervation for the management of hypertension. Based on the results of clinical trials, recommendations have been established on what patients may be eligible for renal sympathetic denervation and under what circumstances.

239. Probing the Broad Line Region and the Accretion Disk in the Lensed Quasars HE0435-1223, WFI2033-4723, and HE2149-2745 using Gravitational Microlensing

Abstract

We use single-epoch spectroscopy of three gravitationally lensed quasars, HE0435-1223, WFI2033-4723, and HE2149-2745, to study their inner structure (BLR and continuum source). We detect microlensing-induced magnification in the wings of the broad emission lines of two of the systems (HE0435-1223 and WFI2033-4723). In the case of WFI2033-4723, microlensing affects two "bumps" in the spectra which are almost symmetrically arranged on the blue (coincident with an AlIII emission line) and red wings of CIII]. These match the typical double-peaked profile that follows from disk kinematics. The presence of microlensing in the wings of the emission lines indicates the existence of two different regions in the BLR: a relatively small one with kinematics possibly related to an accretion disk, and another one that is substantially more extended and insensitive to microlensing. There is good agreement between the estimated size of the region affected by microlensing in the emission lines, $r_s=10^{+15}_{-7} \sqrt{M/M_{\odot}}$ light-days (red wing of CIV in HE0435-1223) and $r_s=11^{+28}_{-7} \sqrt{M/M_{\odot}}$ light-days (CIII] bumps in WFI2033-4723) with the sizes inferred from the continuum emission, $r_s=13^{+5}_{-4} \sqrt{M/M_{\odot}}$ light-days (HE0435-1223) and $r_s=10^{+3}_{-2} \sqrt{M/M_{\odot}}$ light-days (WFI2033-4723). For HE2149-2745 we measure an accretion disk size $r_s=8^{+11}_{-5} \sqrt{M/M_{\odot}}$ light-days. The estimates of $p$, the exponent of the size vs. wavelength ($r_s\propto\lambda^p$), are $1.2\pm0.6$, $0.8\pm0.2$, and $0.4\pm0.3$ for HE0435-1223, WFI2033-4723, and HE2149-2745, respectively. In conclusion, the continuum microlensing amplitude in the three quasars and chromaticity in WFI2033-4723 and HE2149-2745 are below expectations for the thin disk model. Abstract

The idea that dark matter can be made of intermediate-mass primordial black holes in the $10M_\odot \lesssim M \lesssim 200M_\odot$ range has recently been reconsidered, particularly in the light of the detection of gravitational waves by the LIGO experiment. The existence of even a small fraction of dark matter in black holes should nevertheless result in noticeable quasar gravitational microlensing. Quasar microlensing is sensitive to any type of compact objects in the lens galaxy, to their abundance, and to their mass. We have analyzed optical and X-ray microlensing data from 24 gravitationally lensed quasars to estimate the abundance of compact objects in a very wide range of masses. We conclude that the fraction of mass in black holes or any type of compact objects is negligible outside of the $0.05 M_\odot \lesssim M \lesssim 0.45 M_\odot$ mass range and that it amounts to $20 \pm5$% of the total matter, in agreement with the expected masses and abundances of the stellar component. Consequently, the existence of a significant population of intermediate-mass primordial black holes appears to be inconsistent with current microlensing observations. 242. Effect of the Organizational Model of the Subject of Activities in the Natural Environment on Students' Satisfaction and Learning.

Abstract

The aim of this study was to identify the most suitable organizational model for teaching the university subject of Outdoor Activities in relation to learning acquisition and satisfaction. For this purpose, four models were designed, some of which were dominated by traditional education and others by innovative education. The data collection instruments were the Spanish version of the Physical Education of the Sport Satisfaction Instrument, adapted to the university context, and the questionnaire on the contents of the subject designed ad hoc. Data collection was carried out before and after the intervention with a total of 125 students. For data analysis, an ANCOVA was performed. The results showed that there were significant differences in satisfaction between groups (p = 0.029), but not in the boredom scale (p = 0.109). With regard to acquisition of learning between groups, there were significant differences (p = 0.005) in the overall grade of the subject. There were also significant differences in the scores for content taught differently depending on the group (p = 0.003), with a higher score in the most innovative group, the intensive-continuous group, compared to the fractioned (p = 0.005) and classic groups (p = 0.015). It is concluded that there is a positive effect between direct and continuous contact with nature, both in the acquisition of learning and in student satisfaction, with the most innovative model obtaining the best results of the study. Abstract

Transcription factors control genes to maintain normal hemopoiesis, and dysregulation of some factors can lead to acute lymphoblastic leukemia (ALL). Mycoviruses are known to alter the genetics of their fungal host. The present study evaluates the effects of the products of a mycovirus-containing Aspergillus flavus (MCAF), isolated from the home of a patient with ALL, on certain transcription factors of normal and ALL cell lines. Our published studies have shown that ALL patients have antibodies to MCAF, and that exposure of the mononuclear leukocytes of patients in complete remission to its products, unlike controls, results in the re-development of genetic and cell surface phenotypes characteristic of ALL. For the present study, normal, pre-B, and B-cell leukemia cell lines were exposed to the culture of MCAF. Pre- and post-exposure levels of PAX5, Ikaros, and NF-κB were assessed. Exposure to MCAF resulted in apoptosis, cell cycle changes, and complete downregulation of all transcription factors in normal cell lines. In acute leukemia cell lines, cellular apoptosis and alterations in the cell cycle were also noted; however, while there was downregulation of all tested transcription factors, residual levels were retained. The noted alterations in the transcription factors caused by MCAF are novel findings. The possible role of MCAF in leukemogenesis needs to be further investigated. Mycovirus-containing Aspergillus flavus was initially isolated from a leukemia patient's home. Our prior published studies have illuminated intriguing associations of this organism with leukemia. Unlike controls, patients diagnosed with acute lymphoblastic leukemia (ALL) harbor antibodies to this organism. Furthermore, the exposure of mononuclear cells from patients with ALL in complete remission to the products of this organism reproduced genetic and cell phenotypes characteristic of ALL. AbstractNon-traditional sport event has received much attention in recent years. The purpose of this study was to investigate the role of sport event image and attitudes toward event in predicting satisfaction with event and behavioral intentions. A sample of 182 obstacle course runners in Spain was surveyed to test the research model using structural equation modeling. The results showed that event characteristics, which is one of the dimensions of sport event image, and participants’ attitudes toward event positively influenced satisfaction, which in turn, together with attitudes, positively influenced behavioral intentions for repeat visits and recommendations. However, the destination characteristics dimension of sport event image had no impact on attitudes and satisfaction. Finally, satisfaction had a mediating effect on the relationships between event characteristics and behavioral intentions, and attitudes and behavioral intentions. Abstract

The current diagnostic criteria for the autism spectrum disorder (ASD) include the possibility to specify concomitant language difficulties. Purpose: Our main aim was to explore whether children with ASD-Level 1 (ASD-L1) present difficulties in the acquisition of structural language, as little work has been done in this regard so far. As a secondary aim we evaluated the degree to which the potential language impairment in ASD is directly associated with their social communication deficits or it represents a distinct deficit. Methods: To further clarify the nature and characteristics of putative language difficulties in ASD-L1, we evaluated language skills in 89 children and preadolescents diagnosed with ASD-L1, and a group of typically developing participants (TD). All of them were between 8 and 13 years old and had similar socioeconomic backgrounds. Results: Children with ASD-L1 obtained lower scores than those in TD group in repeating sentences, in finding the semantic relationships between words, and in applying word structure rules (morphology). Congruently, the core language standard score was lower in the ASD-L1 group, and the proportion of language delay was significantly higher in the ASD-L1 group than in the control group. Conclusion: Language scores were associated with autistic traits; thus, language performance in ASD-L1 is closely related to autistic symptoms. Abstract

Background: Urogenital schistosomiasis caused by Schistosoma haematobium is highly endemic in the municipality of Cubal in Angola. Currently, diagnosis is based on the observation of S. haematobium eggs in urine samples by microscopy but this method has low sensitivity. Few studies have been performed using molecular techniques in high-prevalence areas for the detection of S. haematobium. The objective of this study is to evaluate the usefulness of real-time PCR as a diagnostic technique for urogenital schistosomiasis among preschool-age children and its correlation with morbidity data. Methods: A cross-sectional study was conducted in Cubal, Angola, involving 97 urine samples from preschool-age children analyzed by the dipstick test, microscopic examination of filtered urine, and real-time PCR. The diagnosis of urogenital schistosomiasis was based on microscopy and/or real-time PCR results. Clinical and ultrasonography evaluation was performed to rule out complications of schistosomiasis. Results: We detected a total of 64.95% of samples positive by real-time PCR and 37.11% by microscopy. The sensitivity of parasitological diagnosis of urogenital schistosomiasis by real-time PCR and microscopy was 95.45% and 54.55%, respectively, and the sensitivity of real-time PCR compared with microscopy was 91.67%. A positive real-time PCR result was significantly related to older age (mean = 3.22 years), detection of eggs by microscopy, and abnormal urine dipstick results (18.56% with proteinuria, 31.96% with leukocyturia, and 31.96% with microhematuria) (p-value<0.05). Ultrasound analysis showed that 23.94% of children had urinary tract abnormalities, and it was significantly related to the real-time PCR diagnosis (p-value<0.05). Conclusions: Real-time PCR is a more sensitive technique than microscopy for urinary schistosomiasis diagnosis in preschool-age children in Cubal. This increase in sensitivity would allow earlier diagnosis and treatment, thus reducing the morbidity associated with schistosomiasis in its early stages. Author summary: Schistosomiasis is a neglected tropical disease that causes approximately 200,000 deaths worldwide each year. Urogenital schistosomiasis is caused by Schistosoma haematobium, which ultimately leads to lesions in the urinary tract due to egg deposition by the parasite. The World Health Organization recommends regular mass administration of praziquantel to prevent chronicity of the disease in high-endemicity areas. Preschool children are one of the most vulnerable groups, however, only children over two years of age are included in these strategies. The diagnostic techniques commonly used in endemic areas for egg detection by microscopy and urine hematuria have major limitations due to their low sensitivity. These limitations lead to misdiagnosis and undetected infections may eventually lead to the chronic form of the disease. Real-time PCR and microscopy were used to diagnose urogenital schistosomiasis in preschool children in Cubal, a high-transmission municipality in Angola. Urinary tract lesions were assessed by ultrasound. Of the samples tested, 64.95% were positive by real-time PCR, compared with 37.11% by microscopy, indicating sensitivities of 95.45% and 54.55%, respectively. Ultrasound analysis showed urinary tract alterations in 23.94% of the children. Abstract

Staphylococcus aureus infection and colonization in patients may be transmitted to healthcare providers and the environment and subsequently cause healthcare-associated infections in other patients. Pathogenic S. aureus strains produce virulence factors, such as Panton-Valentine Leukocidin (PVL), that contribute to the severity of infections and aid in their spread. The emergence of antimicrobial resistance (AMR) is additional concern with respect to S. aureus infection. In this study, the virulence genes and antibiotic resistance profiles of S. aureus were characterized from patients' clinical isolates, healthcare workers' (HCWs') nasal colonization screenings, and the environment at a tertiary healthcare hospital in Addis Ababa, Ethiopia. A total of 365 samples were collected from September 2021 to September 2022: 73 patients' clinical specimens, 202 colonization screenings from HCWs, and 90 hospital environment's swabs. Fifty-one (25.2%) HCW and 10/90 (11.1%) environment S. aureus isolates were identified. Among the 134 isolates, 10 (7.5%) were methicillin-resistant S. aureus (MRSA). Three (4.1%), five (9.8%), and two (20.0%) of the MRSA isolates were identified from the patients, HCWs, and the environment, respectively. Overall, 118 (88.1%) were ampicillin and penicillin resistant; 70 (52.2%) were trimethoprim sulfamethoxazole resistant; and 28 (20.9%) were erythromycin resistant. S. aureus isolates from patients were more resistant to antibiotics than isolates from HCWs or the hospital environment (p<0.05). A total of 92/134 (68.6%) isolates possessed the lukfF-PV gene, which was identified in 62 (85.0%), 26 (51.0%), and 4 (40.0%) of the patient, HCWs, and the environment, respectively. The proportion of lukfF-PV gene containing S. aureus isolated from patient samples was statistically significant. Four (40.0%) of the MRSA isolates also had the lukfF-PV gene. The identification of highly AMR and virulence factors from patients, HCWs and the environment is concerning. Abstract

This article discusses the prevalence of malaria in asymptomatic migrants from sub-Saharan Africa in Spain. The study used real-time PCR (qPCR) as a screening technique and found a prevalence of 5.0% among the screened migrants. The main species detected was Plasmodium falciparum, followed by P. malariae and P. ovale. The study also compared the sensitivity of qPCR with microscopy and found significant agreement between the two methods. However, the study has limitations, including its retrospective design and the inclusion of only self-referred patients. Overall, the findings highlight the importance of using nucleic acid amplification tests like qPCR for malaria diagnosis in this population. [Extracted from the article]
