Now, the occurrence of pharmaceuticals in natural environment has been frequently reported around the world. As a kind of biologically active compounds specially designed to be effective even at very low concentration levels, pharmaceuticals in the environment could have adverse impacts to the health of human beings or other non-targeted organisms due to long-term exposures. To minimize the pharmaceutical pollution from the perspective of drug administration, a new concept called as eco-pharmacovigilance (EPV) has been proposed as a kind of pharmacovigilance(PV) for the environment. However, as a new and comprehensive science, EPV has not sophisticated methods in practice and formalized implementation model up to now. Since EPV is a special kind of PV, it could be feasible to draw on the experience of PV as a possible and reasonable starting point for EPV. In this paper, we discussed the common methods and activities used in PV including spontaneous reporting, intensive monitoring, database studies, and their potential applicability to the environment. And we concluded that these common methods in PV could be adapted and applied to EPV. Aims and Objectives: To explore the incidence of delirium in cerebrovascular patients admitted in our Neurosurgery Intensive Care Unit and analyse the risk factors leading to delirium., Background: Delirium is one of the most common mental disorders in general hospitals, but the incidence of different kinds of diseases and studies varies. Cerebrovascular patients in our Neurosurgery Intensive Care Unit are high-risk groups for delirium; identifying risk factors for delirium and taking early interventions are crucial for patient prognosis., Design: A prospective study., Methods: A convenience sampling method was used to collect data from 128 patients in the Neurosurgery Intensive Care Unit of Xuanwu Hospital, Capital Medical University, Beijing, China, between May 2016-January 2017. Researchers used Confusion Assessment Method for the Intensive Care Unit (Chinese version) to assess each patient's delirium statement twice a day at regular times. We also collected other independent data variables and followed up the short-term clinical outcomes daily., Results: On the basis of Confusion Assessment Method for the Intensive Care Unit evaluation, patients were divided into a delirium group and a nondelirium group. The prevalence of delirium among the 128 patients was 42.2%. Multivariate analysis showed that severity of illness, fever, the use of physical restraints and sleep deprivation were independent predictors of delirium in cerebrovascular patients in the Neurosurgery Intensive Care Unit., Conclusions: Cerebrovascular patients in the Neurosurgery Intensive Care Unit with a critical condition, fever or use of physical restraints or experiencing sleep deprivation were more prone to delirium., Relevance to Clinical Practice: Cerebrovascular patients in the Neurosurgery Intensive Care Unit showed a high incidence of delirium. There are many risk factors leading to delirium, some of which are independent predictors of intensive care delirium. Patients with delirium will suffer various adverse effects upon their short-term clinical outcomes. Therefore, nurses should pay close attention to changes in a patient's mental state and learn about the risk factors associated with delirium, in order to be able to take early measures to prevent delirium., (© 2017 John Wiley & Sons Ltd.)

This research was aimed at establishing the pilot-scale purification technology of lipopeptide from marine-derived Bacillus marinus. We studied lipopeptide surfactivity interferences on scale-up unit technologies including acid precipitation, methanol extraction, solvent precipitation, salting out, extraction, silica gel column chromatography and HZ806 macroporous absorption resin column chromatography. Then, the unit technologies were combined in a certain order, to remove the impurities gradually, and to gain purified lipopeptide finally, with high recovery rate throughout the whole process. The novel pilot-scale purification technology could effectively isolate and purify lipopeptide with 87.51% to 100% purity in hectograms from 1 ton of Bacillus marinus B-9987 fermentation broth with more than 81.73% recovery rate. The first practical hectogram production of highly purified lipopeptide derived from Bacillus marinus was achieved. With this new purification method, using complex media became possible in fermentation process to reduce the fermentation cost and scale-up the purification for lipopeptide production. For practicability and economy, foaming problem resulting from massive water evaporation was avoided in this technology.

A novel cultivation strategy called "Sequential Heterotrophy-Dilution-Photoinduction" was successfully applied in the cultivation of Galdieria sulphuraria for efficient production of phycocyanin (PC). Algae cells were firstly cultivated heterotrophically to achieve high cell density. The maximal dry cell weight of 17.8gL(-1) and maximal biomass productivity of 103.1mgL(-1)h(-1) were obtained. Then, a dilution step was applied to obtain a suitable cell concentration and finally these cells were transferred to high light condition for phycocyanin accumulation. During the photoinduction step, cells could accumulate phycocyanin up to 13.88% of dry cell weight in a cultivation period of 8days. By this technology, total PC productivity far surpassed those reported in all literatures of Galdieria sulphuraria, and was 147-fold and 12-fold of those in photoautotrophic and heterotrophic technologies, respectively. Therefore, this strategy provides a promising approach for high-efficient phycocyanin production from Galdieria sulphuraria to meet its mass cultivation and commercialization application., (Copyright © 2016 Elsevier Ltd. Objective: To investigate the changes of Wnt7b/β-catenin signaling pathway molecules in the differentiation of fetal rat alveolar epithelial type II cells (fAEC2)., Methods: Fetal rat lung tissues were extracted from pregnant SD rats (19 days), and then fAEC2 were isolated and purified. After fAEC2 were cultured for 48, 72, 96 hours, the morphological changes of fAEC2 and the expression of β-catenin were respectively observed by inverted microscope and immunofluorescence. The mRNA expressions of Wnt7b, cyclin D1, pulmonary surfactant C (SP-C) and aquaporin 5 (AQP5) were detected by real-time quantitative PCR. The protein expressions of cyclin D1, nucleus β-catenin, SP-C and AQP5 were measured by Western blotting., Results: β-catenin was expressed in cell membrane when fAEC2 were cultured for 48 hours; the expression of β-catenin decreased in cell membrane while enhanced in cytoplasm and nucleus at 72 hours; whole-cell β-catenin expression was lowered at 96 hours. The expressions of Wnt7b and cyclin D1 mRNAs were significantly raised at 72 hours and reduced obviously at 96 hours compared with 48 hours. SP-C mRNA expression level went down gradually with the extended culture time, and AQP5 mRNA level went up gradually. The protein expressions of cyclin D1 and nucleus β-catenin were significantly enhanced at 72 hours and weakened at 96 hours compared with 48 hours. SP-C protein expression was down-regulated with the prolonged culture time, and AQP5 protein expression was up-regulated., Conclusion: Wnt7b/β-catenin signaling pathway may play an important role in fAEC2 transdifferentiation.

Adequate cardiopulmonary resuscitation (CPR) skill is essential in improving survival rate of sudden cardiac arrest (SCA). However, the skill deteriorates rapidly following CPR training. We developed a computer game by using 3-Dimensional virtual technology (3-D CPR game) for laypersons in the purpose to improve skill retention. As the testing phase, a randomized control trial, in which we recruited 97 freshman medical students who had no prior CPR training experience, was used to test its effect on 3-month CPR Skill retention. The usability of the game was also tested using a 33 item questionnaire rated with 5-point Likert scale. Three months after the initial CPR training, the retention rate of CPR skill in the game group was significantly higher compared with the control (p<0.05) and the average score on 4 dimensions of usability were 3.99-4.05. Overall, using 3-D CPR game in improving CPR skill retention is feasible and effective.

Photoautotrophic cultivation with heterotrophic cells as seeds (heterotrophic cells/photoautotrophic cultivation) is an effective way for the development of microalgal biofuel, but its development potential from the point of process optimization has not been investigated in literatures. To evaluate this, the optimizations of medium and culture conditions for Chlorella ellipsoidea were studied. In the heterotrophic stage, the biomass concentration reached 11.04 g/L with the optimized medium in flask, which were 28.0% higher than that with the original medium, and the biomass concentration reached 73.89 g/L in 5-L fermenter. In the photoautotrophic stage, the culture medium and conditions were studied in a 2-L column photobioreactor. The maximum biomass concentration, lipid content and lipid productivity reached 1.62 g/L, 36.34% and 6.1 mg/(L·h) under the optimal photoautotrophic conditions. The lipids were mainly composed of C16-C18 fatty acids, which were raw material suitable for biodiesel. After optimization, heterotrophic cells/photoautotrophic cultivation can significantly improve the capacity of biofuel production by Chlorella ellipsoidea, this method is also expected to be an efficient way for the cultivation of other microalgae that can grow heterotrophically.

As the optimal source of astaxanthin, Haematococcus pluvialis was cultured for commercial production of astaxanthin through two continuous phases: cell growth and astaxanthin induction. In this study, the efficiency of an attached system for producing astaxanthin from H. pluvialis was investigated and compared to that of the suspended system (bubble column bioreactor) under various conditions. Results showed that this attached system is more suitable for photoinduction of H. pluvialis than the suspended bioreactor. Under the optimal conditions, the astaxanthin productivity of the attached system was 65.8 mg m(-2)d(-1) and 2.4-fold of that in the suspended system. This attached approach also offers other advantages over suspended systems, such as, producing astaxanthin under a wide range of light intensities and temperatures, saving water, ease to harvest cells, resisting contamination. Therefore, the attached approach can be considered an economical, environmentally friendly and highly-efficient technology for producing astaxanthin from H. pluvialis., (Copyright © 2014 Elsevier Ltd. The multistage model of nonmelanoma skin carcinogenesis has contributed significantly to our understanding of epithelial cancer in general. We used the Krt1-15CrePR1;R26R transgenic mouse to determine the contribution of keratin 15+ cells from the hair follicle to skin tumor development by following the labeled progeny of the keratin 15 expressing cells into papillomas. We present three novel observations. First, we found that keratin 15 expressing cells contribute to most of the papillomas by 20 weeks of promotion. Second, in contrast to the transient behavior of labeled keratin 15-derived progeny in skin wound healing, keratin 15 progeny persist in papillomas, and some malignancies for many months following transient induction of the reporter gene. Third, papillomas have surprising heterogeneity not only in their cellular composition, but also in their expression of the codon 61 signature Ha-ras mutation with approximately 30% of keratin 15-derived regions expressing the mutation. Together, these results demonstrate that keratin 15 expressing cells of the hair follicle contribute to cutaneous papillomas with long term persistence and a subset of which express the Ha-ras signature mutation characteristic of initiated cells., (© 2012 Wiley Periodicals, Inc.)

A novel cultivation strategy called "sequential heterotrophy-dilution-photoinduction" was developed for efficient algal biomass and lipid production. Three Chlorella species were first cultivated heterotrophically to achieve high cell density, then the broth was diluted to suitable concentration (2-5 g/L) and transferred to light environment for photoinduction. With this strategy, the Chlorella intracellular protein and chlorophyll increased rapidly to 50.87% and 32.97 mg/g by a 12-h illumination, which were close to the level of cells cultivated photoautotrophically. Moreover, the lipid contents were increased by 84.57%, 70.65% and 121.59% within 24-h photoinduction for C. vulgaris, C. pyrenoidosa and C. ellipsoidea, respectively. Maximum lipid content as 26.11% of biomass and maximum lipid productivity of 89.89 mg/L/d was both accomplished by C. pyrenoidosa. Further outdoor experiments showed consistent patterns. Therefore, the proposed strategy provided an effective approach for microalgal biomass production to meet the urgent need for both health food and biodiesel., (Copyright © 2012 Elsevier Ltd. The new, single-isomer, sulfated alpha-cyclodextrin, the sodium salt of hexakis(2,3-diacetyl-6-O-sulfo)-alpha-cyclodextrin (HxDAS), was used for the first time in acidic methanol background electrolytes (BGEs) to separate the enantiomers of weak base analytes by nonaqueous capillary electrophoresis (NACE). The concentration dependence of the effective mobilities and separation selectivities followed trends similar to those observed earlier in acidic methanol background electrolytes with heptakis(2,3-diacetyl-6-O-sulfo)-beta-cyclodextrin (HDAS) and octakis(2,3-diacetyl-6-O-sulfo)-gamma-cyclodextrin (ODAS). In general, interactions between the weak base analytes and HxDAS were weaker than with HDAS and ODAS. For some of the weak base analytes, separation selectivities observed in acidic aqueous and acidic methanol background electrolytes were complementary to each other, permitting the eventual separation of enantiomers that could not be achieved otherwise.

The third, concluding member of the family of single-isomer, fully sulfated alpha-cyclodextrins, the sodium salt of hexakis(2,3-di-O-methyl-6-O-sulfo)-alpha-cyclodextrin (HxDMS), has been synthesized on the kilogram scale, completing the nine-member array of the single-isomer, 6-O-sulfo CDs now available. HxDMS was tested for the capillary electrophoretic (CE) resolution of the enantiomers of nonelectrolyte, weak acid and weak base analytes contained in our CD screening kit. HxDMS complexed differently with many of the analytes tested than either its larger-ring analogs, heptakis(2,3-di-O-methyl-6-O-sulfo)-beta-CD (HDMS) and octakis(2,3-di-O-methyl-6-O-sulfo)-gamma-CD (ODMS) or its same-ring, but differently substituted analogs, hexakis(6-O-sulfo)-alpha-CD (HxS) and hexakis(2,3-di-O-acetyl-6-O-sulfo)-alpha-CD (HxDAS). For all analytes, the effective mobilities and separation selectivities as a function of the background electrolyte concentration of HxDMS followed the trends that were found for the other single-isomer, 6-O-sulfo CDs.

The second member of the family of single-isomer sulfated alpha-cyclodextrins, the sodium salt of hexakis(6-O-sulfo)-alpha-cyclodextrin (HxS), has been synthesized, analytically characterized, and used as the resolving agent for the capillary electrophoretic separation of the enantiomers of nonionic, weak-acid and weak-base analytes present in our initial screening kit. HxS interacted less strongly with many of the analytes tested than the larger-ring analogs, heptakis(6-O-sulfo)-beta-cyclodextrin (HS) and octakis(6-O-sulfo)-gamma-cyclodextrin (OS). For some of the analytes, the separation selectivities obtained with HxS were complementary to those observed with hexakis(2,3-di-O-acetyl-6-O-sulfo)-alpha-cyclodextrin (HxDAS), HS, and OS. For all analytes, the effective mobilities and separation selectivities as a function of the background electrolyte concentration of HxS followed the trends that were found for HxDAS, HS, and OS.

The hair follicle bulge possesses putative epithelial stem cells. Characterization of these cells has been hampered by the inability to target bulge cells genetically. Here, we use a Keratin1-15 (Krt1-15, also known as K15) promoter to target mouse bulge cells with an inducible Cre recombinase construct or with the gene encoding enhanced green fluorescent protein (EGFP), which allow for lineage analysis and for isolation of the cells. We show that bulge cells in adult mice generate all epithelial cell types within the intact follicle and hair during normal hair follicle cycling. After isolation, adult Krt1-15-EGFP-positive cells reconstituted all components of the cutaneous epithelium and had a higher proliferative potential than Krt1-15-EGFP-negative cells. Genetic profiling of hair follicle stem cells revealed several known and unknown receptors and signaling pathways important for maintaining the stem cell phenotype. Ultimately, these findings provide potential targets for the treatment of hair loss and other disorders of skin and hair.

Human neocentromeres are fully functional centromeres that provide mitotic stability to rearranged chromosomes that have separated from endogenous centromeres. A disproportionate number of neocentromeres has been observed in certain regions such as chromosome 3q (n=6), 15q (n=9) and 13q32 (n=7), suggesting that these regions contain DNA sequences with a high propensity for neocentromere formation. Therefore, we have addressed the role of primary DNA sequence in neocentromere formation by asking whether multiple independent neocentromeres that were cytologically localized to chromosome 13q32 are in fact localized to the same underlying genomic DNA. Analysis of four independent 13q32 neocentromeres using simultaneous FISH with ordered YAC probes and immunofluorescence with antibodies to CENP-C have localized three neocentromeres to a distal approximately 7 Mb domain in chromosome 13q32, and one to an overlapping proximal domain of approximately 7 Mb. DNA was obtained from three of these neocentromeres by CENP-A chromatin immunoprecipitation (ChIP) and used to screen ordered BACs using both a slot-blotted BAC pool approach and a genomic microarray that contiguously spans 13q31.3-13q33.1. The CENP-A binding domains from each of these neocentromeres was identified to distinct genomic locations of approximately 130, 215 and 275 kb within an approximately 6.5 Mb region. Thus, the lack of coincidence of these neocentromeres to the same underlying DNA sequence refutes the idea of a DNA sequence based neocentromere 'hotspot' in 13q32 and further supports the sequence-independent epigenetic formation of human neocentromeres. The screening of genomic microarrays with ChIP DNA provides a powerful method to identify mammalian DNA sequences associated with particular functional chromatin states.

The sodium salt of hexakis(2,3-di-O-acetyl-6-O-sulfo)-alpha-CD (HxDAS), the first member of the family of single-isomer, fully sulfated alpha-CDs, has been synthesized and used for the initial capillary electrophoretic separation of the enantiomers of nonionic, weak acid, weak base, and ampholytic analytes. HxDAS complexes less strongly with many of the analytes tested than the analogous beta- and gamma-cyclodextrin derivatives, namely, heptakis(2,3-di-O-acetyl-6-O-sulfo)-beta-cyclodextrin (HDAS) and octakis(2,3-di-O-acetyl-6-O-sulfo)-gamma-cyclodextrin (ODAS). Nevertheless, it facilitated the separation of the enantiomers of a large number of weak electrolyte and nonelectrolyte analytes in acidic aqueous background electrolytes. For all analytes, the effective mobilities and separation selectivities as a function of the background electrolyte concentration of HxDAS followed the trends that were found for HDAS and ODAS.

Objective: To explore the clinical feature of benign symmetric lipomatosis (BSL) so as to improve its diagnosis and treatment., Method: 28 patients of BSL treated in our hospital were analyzed and literature was reviewed. Surgical management is used in all patients., Result: There were no patients died. All patients were satisfactory with their appearance and function improved. 28 patients were followed up for 2-15 years (average 6 years and 5 months), only 5 cases recurred., Conclusion: BSL usually occurs in middle age men. Patients have a striking appearance "hump back" because of a diffuse, symmetric distribution of the lipomatous tissue in the cervical region. The etiology is related to alcohol abuse. Operation is the most effective treatment. The radical surgical therapy should not be emphasized because the important anatomic structures may be damaged. Abstaining from alcohol may help to reduce the rate of recurrence.

Objective: To explore the expression of WT1 gene in leukemia patients and its clinical implications., Methods: Expression of WT1 mRNA was detected in two leukemia cell lines (K562 and HL-60), 49 acute leukemia (AL) patients, 33 chronic myeloid leukemia (CML) patients and 25 healthy subjects by reverse trans-criptase-nested polymerase chain reaction (RT-Nested PCR)., Results: WT1 gene was expressed in all subtype of AL including K562 and HL-60 cell lines, 21/29 newly diagnosed and relapsed AL patients, 1/20 complete remission (CR) AL patients, 15/18 CML blastic crisis patients, 1/5 CML patients in accelerated phase, and 1/10 CML patients in chronic phase. WT1 gene was undetectable in 25 healthy subjects. The expression level of WT1 gene was related to the prognosis of AL, patients with relative level >/= 1.0 had lower CR rates and disease-free survival. For CML patients, WT1 gene expression was associated with the clinical phase, it increased with disease progressed., Conclusion: WT1 gene expression is associated with pathogenesis of leukemia. It is a prognostic factor and a marker for the detection of minimal residual disease in AL and may used as an indicator for diagnosing CML blastic crisis.

A mutant of Candida tropicalis FYD-2 was obtained from its parental strain SFP-1186 by ultraviolet treatments. On shaking flask, the yield of mixed dicarboxylic acid(DCA) by the mutant was 21.4% higher than that by its ancestor. The amount of mixed DCA reached 156 g/L for 120 h incubation in a 10 L autoconrolled fermentor where the culture medium contained 25% n-paraffin. The process of induced and screening mutant was introduced and the time course of fermentation in 10 L fermentor was discussed.
