Coastal retreat line is an important part of coastal land spatial planning, which balances the needs of resource utilization and ecological protection of coastal cities, and ensures the safety bottom line of healthy and sustainable development of coastal areas. At present, China's coastal retreat line lacks a systematic evaluation and management and control system. By combing the correlation mechanism between five impact dimensions such as social economy value, spatial development potential, landscape style risk, marine disaster risk, and ecological environment value and various elements of the coastal zone, this paper established an evaluation system for the importance of coastal retreat line in coastal areas with land and sea planning, summarized the hierarchical, classified, and differentiated management and control strategies of coastal retreat line in China, and took Dongying City as an example. The paper further discussed the application of the evaluation system in the demarcation and control of retreat line, to provide a reference for the control of coastal land spatial planning.

Tomato leaf mold is a common disease in tomato production and severely impacts the growth, fruit quality and yield of tomato plants. Research on tomato leaf mold has a long history and has focused mainly on the differentiation of pathogen races, the structure and function of pathogen Avr gene products, the cloning of Cf resistance genes and the analysis of disease resistance mechanisms. Interactions between Cf and Avr are in accordance with the ''gene-for-gene'' hypothesis and typical Cf/Avr interactions are part of effector-triggered immunity (ETI). However, an increasing number of studies have proven that pathogen-associated molecular pattern (PAMP)-triggered immunity (PTI) is involved in the disease resistance response system mediated by Cf genes. In addition, different Cf genes have both similar and different roles in the disease resistance response, indicating that the disease resistance mechanism of Cf genes is complex. In this paper, progress in tomato leaf mold research was reviewed, and the regulatory mechanism underlying the Cf-mediated disease resistance response was thoroughly explored. We hope this summary will lay a foundation for research on tomato leaf mold disease resistance mechanisms and is applicable to breeding for disease resistance.

Abstract Background: Sarcopenia is an age-related skeletal muscle disorder that involves a loss of muscle mass or strength and physiological function. Skeletal muscle deteriorates in both quantity and quality. The endocrine system is an important regulator of muscle metabolism. Therefore, we aimed to explore the relationship between biochemical markers and muscle mass in sarcopenia. Methods: We used the AWGS 2014 as the diagnostic criteria for sarcopenia, considering both the loss in muscle mass, muscle strength and physical performance. A total of 2837 elderly female participants over 50 years of age from the West China Health and Aging Trend (WCHAT) study were included. Insulin, glucose, 25(OH)VD, procalcitonin, alanine aminotransferase, aspartate aminotransferase, total protein, prealbumin, albumin, thyroid-stimulating hormone, free triiodothyronine, free tetraiodothyronine, triglycerides, cholesterol, high-density lipoprotein, very low-density lipoprotein, cortisol, and follicle-stimulating hormone were measured. Based on the findings of univariate analysis, multivariate regression and receiver operating characteristic (ROC) curves were established. Abstract Colorectal cancer (CRC) is one of the most commonly diagnosed cancer types worldwide. Despite significant advances in prevention and diagnosis, CRC is still one of the leading causes of cancer-related mortality globally. RAB27A, the member of RAB27 family of small GTPases, is the critical protein for intracellular secretion and has been reported to promote tumor progression. However, it is controversial for the role of RAB27A in CRC progression, so we explored the exact function of RAB27A in CRC development in this study. Based on the stable colon cancer cell lines of RAB27A knockdown and ectopic expression, we found that RAB27A knockdown inhibited proliferation and clone formation of SW480 colon cancer cells, whereas ectopic expression of RAB27A in RKO colon cancer cells facilitated cell proliferation and clone formation, indicating that RAB27A is critical for colon cancer cell growth. In addition, our data demonstrated that the migration and invasion of colon cancer cells were suppressed by RAB27A knockdown, but promoted by RAB27A ectopic expression. Therefore, RAB27A is identified as an onco-protein in mediating CRC development, which may be a valuable prognostic indicator and potential therapeutic target for CRC.

Abstract Background Angiotensin-converting enzyme 2 (ACE2) is a key enzyme of the renin-angiotensin system and a well-known functional receptor for the entry of severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) into host cells. The COVID-19 pandemic has brought ACE2 into the spotlight, and ACE2 expression in tumors and its relationship with SARS-COV-2 infection and prognosis of cancer patients have received extensive attention. However, the association between ACE2 expression and tumor therapy and prognosis, especially in breast cancer, remains ambiguous and requires further investigation. We have previously reported that ACE2 is elevated in drug-resistant breast cancer cells, but the exact function of ACE2 in drug resistance and progression of this malignant disease has not been explored. Methods The expression of ACE2 and HIF-1α in parental and drug-resistant breast cancer cells under normoxic and hypoxic conditions was analyzed by Western blot and qRT-PCR methods. The protein levels of ACE2 in plasma samples from breast cancer patients were examined by ELISA. The relationship between ACE2 expression and breast cancer treatment and prognosis was analyzed using clinical specimens and public databases. The reactive oxygen species (ROS) levels in breast cancer cells were measured by using a fluorescent probe. Small interfering RNAs (siRNAs) or lentivirus-mediated shRNA was used to silence ACE2 and HIF-1α expression in cellular models. The effect of ACE2 knockdown on drug resistance in breast cancer was determined by Cell Counting Kit 8 (CCK-8)-based assay, colony formation assay, apoptosis and EdU assay. Results ACE2 expression is relatively low in breast cancer cells, but increases rapidly and specifically after exposure to anticancer drugs, and remains high after resistance is acquired. Mechanistically, chemotherapeutic agents increase ACE2 expression in breast cancer cells by inducing intracellular ROS production, and increased ROS levels enhance AKT phosphorylation and subsequently increase HIF-1α expression, which in turn upregulates ACE2 expression. Although ACE2 levels in plasma and cancer tissues are lower in breast cancer patients compared with healthy controls, elevated ACE2 in patients after chemotherapy is a predictor of poor treatment response and an unfavorable prognostic factor for survival in breast cancer patients. Conclusion ACE2 is a gene in breast cancer cells that responds rapidly to chemotherapeutic agents through the ROS-AKT-HIF-1α axis. Elevated ACE2 modulates the sensitivity of breast cancer cells to anticancer drugs by optimizing the balance of intracellular ROS. Moreover, increased ACE2 is not only a predictor of poor response to chemotherapy, but is also associated with a worse prognosis in breast cancer patients. Thus, our findings provide novel insights into the spatiotemporal differences in the function of ACE2 in the initiation and progression of breast cancer.

Abstract Metastasis and chemical resistance are the most serious problems in the treatment of highly aggressive uveal melanoma (UM). The newly identified lncRNA OUM1 is overexpressed in UM, functions as a catalyst and regulates protein tyrosine phosphatase (PTP) activity by binding to PTP receptor type Z1 (PTPRZ1), which plays an important role in cell proliferation, metastasis and chemotherapy resistance in the UM microenvironment. Hence, siRNAs that selectively knocking down the lncRNA OUM1 (siOUM1) and its target gene PTPRZ1 (siPTPRZ1) were designed to inhibit the OUM1/PTPRZ1 pathway to reduce PTP activity, and this reduction in activity interrupts protein tyrosine phosphorylation, suppresses UM proliferation and metastasis and improves cisplatin sensitivity in UM cells. Then, to overcome the limitations of the difficulty of drug administration and traditional therapeutics, the indocyanine green (ICG)-labeled manganese metal–organic framework (MOF) nanoparticles (NPs) were fabricated and linked with arginine-glycine-aspartate (RGD) peptide to carry siOUM1/siPTPRZ1 and cisplatin to achieve targeted siRNA interference-mediated therapy, enhanced cisplatin therapy and chemodynamic therapy. This NP system also has a dual-modal imaging ability because ICG is a near-infrared region fluorescent dye and manganese has the potential to be used in magnetic resonance imaging. This study verifies the significance of the newly discovered lncRNA OUM1 as a new therapeutic target for aggressive UM and provides a drug delivery NP system for precise treatment of UM accompanied with a dual-modal imaging ability. In the software defined optical transport network (SDOTN) architecture, the manufacturer’s controller system is required to be connected to the optical transport network (OTN) orchestrator system through the northbound interface.However, there is currently no unified northbound interface standard, which brings many problems to the automatic provisioning of cross-domain and cross-vendor end-to-end services in SDOTN.The status and problems of OTN controller northbound open interface standards were expounded, and the current mainstream transport application programming interface (TAPI) and abstraction and control of traffic engineering network (ACTN) northbound interface standards from the perspective of network models were compared and analyzed.Finally, the research practice of China Unicom in the standardization of ACTN unified interface model and the construction of management and control system were introduced, and an outlook on the follow-up research and development direction was provided.

Abstract Background Drought stress has negative effects on plant growth and productivity. In this study, a comprehensive analysis of physiological responses and gene expression was performed. The responses and expressions were compared between drought-tolerant (DT) and drought-sensitive (DS) peanut varieties to investigate the regulatory mechanisms and hub genes involved in the impact of drought stress on culture. Results The drought-tolerant variety had robust antioxidative capacities with higher total antioxidant capacity and flavonoid contents, and it enhanced osmotic adjustment substance accumulation to adapt to drought conditions. KEGG analysis of differentially expressed genes demonstrated that photosynthesis was strongly affected by drought stress, especially in the drought-sensitive variety, which was consistent with the more severe suppression of photosynthesis. The hub genes in the key modules related to the drought response, including genes encoding protein kinase, E3 ubiquitin-protein ligase, potassium transporter, pentatricopeptide repeat-containing protein, and aspartic proteinase, were identified through a comprehensive combined analysis of genes and physiological traits using weighted gene co-expression network analysis. There were notably differentially expressed genes between the two varieties, suggesting the positive roles of these genes in peanut drought tolerance. Conclusion A comprehensive analysis of physiological traits and relevant genes was conducted on peanuts with different drought tolerances. The findings revealed diverse drought-response mechanisms and identified candidate genes for further research.

A CTCF mediated intrachromosomal looping of RB1 promoter and its downstream silencer served as a regulatory inducer to inactivate RB1, revealing a novel transcriptional mechanism underlying RB1 dysfunction independent of gene mutation.

INTRODUCTION: The incidence of type 1 diabetes mellitus (T1DM) is rapidly increasing worldwide. However, the incidence in Henan Province of China has been unknown for more than two decades. This study aimed to estimate the incidence of T1DM in the 0.5-14.9 years age group in Henan Province of China from 2017 to 2020. METHODS: A retrospective analysis of hospital registration data from 18 cities in Henan Province, China, identified 1726 patients (843 males, 883 females) between 0.5-14.9 years of age with newly diagnosed T1DM in Henan Province from January 1st, 2017, to December 31st, 2020, covering more than 19 million children years at risk. RESULTS: The crude incidence of T1DM per 100 000 person years for the 0.5-14.9 years age group in the Henan Province of China was 2.19 [95% confidence interval (CI): 1.99, 2.40], with a peak in the 10-14.9 years age group. Abstract Smart sensing fabrics are becoming increasingly attractive in the emerging wearable areas of medical and military so far. Here, for the first time, we present a smart humidity sensing fabric (SHSF) based on a moisture‐sensitive polyacrylamide hydrogel for respiratory monitoring and non‐contact sensing. Fabricated by in situ cross‐linking of the hydrogel precursors on the fibers of the fabric, the flexible SHSF shows excellent sensitivity, outstanding flame retardance, air permeability, water retention capacity, and stability after treatment with lithium bromide solution. Specifically, its conductance increases more than 311 times as humidity increased from 11% to 98%. Besides, the humidity sensor features good repeatability and the ability to work normally under folding due to its flexible nature. As a clothing material, hydrogel–fabric composite exhibits 4.3 times the burning time compared to cotton fabric, illustrating better flame retardance. The SHSF is used to monitor human breathing and non‐contact finger approaching in real time, demonstrating its flexibility in practical applications. This work provides strategies for preparing high‐performance, flame‐retardant SHSF for emerging wearable electronic devices.

IntroductionEmerging evidence demonstrates that the high-fructose and high-fat diet (HFHF) induced obesity and fatty liver disease has become one of the most common metabolic disorders worldwide. Therefore, innovative investigations on compounds targeting obesity and fatty liver diseases are urgently needed.MethodsThe high-throughput natural compounds screen was performed to screen the important compounds. A rat HFHF model was constructed, the regulatory function of Oxymatrine in HFHF-induced obesity was further explored.ResultsWe identified Oxymatrine, a natural compound extracted from Sophora flavescens, showed a potential compacity in high-fat diet-induced fatty liver disease. We found that oxymatrine significantly inhibited HFHF-induced obesity using a rat HFHF model. Additionally, we found that oxymatrine altered the enhancer landscape of subcutaneous adipose tissues by ChIP-seq analysis using antibodies against the H3K27ac histone modification. Motif enrichment analysis showed the Smad motif was significantly enriched in enhancers altered post-oxymatrine treatment. Further chromatin immunoprecipitation-quantitative PCR (ChIP-qPCR) analysis and luciferase reporter assays showed oxymatrine alters the binding of Smad3 on the enhancer regions of B-cell lymphoma 2 (Bcl2) and the enhancer activity of Bcl2.DiscussionTogether, our study highlighted oxymatrine could suppress high-fructose and high-fat diet-induced obesity by inhibiting the suppressor of mothers against decapentaplegic 3 (Smad3) binding on obesity-related enhancers.

The continuum manipulator improves the intraocular robot flexibility during retinal surgery. Herein, a micro‐3‐degree of freedom (DOF) force sensor with a length of only 3.8 mm that can adjust the puncture angle during retinal vein cannulation (RVC) is proposed. Taking advantage of 3D printing technology, the optical fiber and injection line are embedded in the nitinol tube with flexible hinges to achieve integration of the force sensing and injection functions. A new fiber Bragg grating (FBG) configuration method and linear decoupling algorithm are also proposed herein. Static calibration experiments demonstrate excellent resolution of the force sensor, of less than 0.072 mN for transverse force and 0.738 mN for the axial force in the range from −20 to 20 mN. The proposed sensor is installed on a newly developed actuator, and an ex vivo experiment is performed on pig cadaver eyes. The interaction forces at each stage of the RVC are recorded in detail for the first time, further confirming the feasibility and effectiveness of the developed sensor for monitoring the RVC interaction forces.

Studies have identified that mating induces a series of physiological changes in animals. In this period, males tending to invest more energy, immune peptides, and other substances to reduce the cost of living for females. This results in lower survival rates in later life than females. Meanwhile, both males and females shorten lifespans due to reproduction. However, the reasons why termites' queens and kings are both extremely long-lived and highly fecund are unclear. Therefore, this study aimed to examine the effects of mating on the expression of immune and DNA repair genes for lifespan extension in termite queens and kings. Here, we reported that mated queens show relatively higher expression of immune genes (phenoloxidase, denfensin, termicin, transferrin), antioxidant genes (CAT, SOD), detoxification genes (GST, CYP450) than virgin queens in the Reticulitermes chinensis. In addition, mated kings also highly expressed these genes, except for termicin, transferrin, GST, and CYP450. After mating, both queens and kings significantly upregulated the expression of DNA repair genes (MLH1, BRCA1, XRCC3, RAD54-like). Mismatch repair genes (MMR) MSH2, MSH4, MSH6 were considerably increased in mated queens, while MSH4, MSH5, MSH6 were upregulated in mated kings. Our results suggest that mating increases the expression of immune and DNA repair genes in the termite queens and kings, and thus possibly improving their survival during reproductive span due to the omnipresent pathogens.

Pediatric-type follicular lymphoma (PTFL) is a rare pediatric-type indolent B-cell lymphoma that clinicopathologically differs from adult lymphoma. Accurate diagnosis of PTFL, which is often challenging, is essential to avoid missed diagnosis, misdiagnosis, and overtreatment. To improve our understanding of PTFL, clinicopathological features, differential diagnosis, and molecular mutation characteristics of four patients of PTFL were analyzed using hematoxylin and eosin staining, immunohistochemistry, polymerase chain reaction, fluorescence in situ hybridization (FISH), and next-generation sequencing (NGS). A relevant literature review was also performed. All four PTFL patients were male, with ages of 6, 18, 13, and 15 years, and had St. Jude stage I or III. Microscopic results showed that the structure of the lymph nodes was destroyed; the tumor follicles were enlarged and irregular; medium–large blastoid cells with a consistent shape were visible in tumor follicles, and the nucleus was round or oval; and the “starry sky” pattern was easily observed. Tumor cells expressed CD20, PAX-5, BCL6, and CD10. None of the tumor cells expressed BCL2, CD3, CD5, MUM1, and CyclinD1. CD21 showed dilated growth of a follicular dendritic cell network in tumor follicles. EBER genes were negative in all cases. FISH testing also showed negative BCL2 gene breaks and IRF4 gene breaks in all cases. NGS detected 12 related mutant genes, including KMT2D, CD79B, GNA13, MYD88, PCLO, TCF3, IRF8, MAP2K1, FOXO1, POLE, INPP5D, and FAT4. Two of the four patients had an IRF8 gene mutation, and one patient had a dual mutation of the MAP2K1 gene. Our study revealed the unique clinicopathological features and molecular mutational characteristics of PTFL, consolidated our understanding of PTFL, and identified other rare mutant genes, which may further contribute to the study of the molecular mechanism and differential diagnosis of PTFL.

Changes in the canopy microclimate in intercropping systems, particularly in the light environment, have important effects on the physiological characteristics of photosynthesis and yield of crops. Although different row ratio configurations and strip widths of dwarf crops in intercropping systems have important effects on canopy microclimate, little information is available on the effects of intercropping on chlorophyll synthesis and photosynthetic physiological properties of dwarf crops. A 2-year field experiment was conducted in 2019 and 2020, with five treatments: sole maize (SM), sole peanut (SP), four rows of maize intercropping with eight rows of peanut (M4P8), four rows of maize intercropping with four rows of peanut (M4P4), and four rows of maize intercropping with two rows of peanut (M4P2). The results showed that the light transmittance [photosynthetically active radiation (PAR)], photosynthetic rate (Pn), transpiration rate (Tr), and stomatal conductance (Gs) of intercropped peanut canopy were reduced, while the intercellular carbon dioxide concentration (Ci) was increased, compared with SP. In particular, the M4P8 pattern Pn (2-year mean) was reduced by 5.68%, 5.33%, and 5.30%; Tr was reduced by 7.41%, 5.45%, and 5.95%; and Gs was reduced by 8.20%, 6.88%, and 6.46%; and Ci increased by 11.95%, 8.06%, and 9.61% compared to SP, at the flowering needle stage, pod stage, and maturity, respectively. M4P8 improves the content of chlorophyll synthesis precursor and conversion efficiency, which promotes the utilization efficiency of light energy. However, it was significantly reduced in M4P2 and M4P4 treatment. The dry matter accumulation and pod yield of peanut in M4P8 treatment decreased, but the proportion of dry matter distribution in the late growth period was more transferred to pods. The full pod number decreases as the peanut row ratio decreases and increases with year, but there is no significant difference between years. M4P8 has the highest yield and land use efficiency and can be used as a reference row ratio configuration for maize–peanut intercropping to obtain relatively high yield benefits.

Introduction: Collateral formation is insufficient in some patients with acute myocardial infarction (AMI). Peripheral blood CD14++CD16+ monocytes (intermediate monocytes; IM) and vascular endothelial growth factors (VEGFs) are associated with formation of collateral circulation. Methods: We enrolled 49 patients with AMI who underwent emergency percutaneous transluminal coronary intervention (PCI) (Group A) and 27 patients underwent delayed PCI 1 week after AMI (Group B). The percentage of circulating IM and levels of VEGFs in circulation were determined on day 8th. Left ventricular ejection fraction (LVEF) was measured 3 months after AMI. Results: The peripheral levels of IM and serum VEGF levels on day 8th were significantly higher in patients with well-developed collateral circulation in Group A than those in Group B. The levels of circulating VEGFs in the collateral circulation (+) subgroup in Group B were lower than those in the collateral circulation (−) subgroup. Moreover, the serum VEGF-B186 levels positively correlated with IM. Conclusions: Hyperacute collateral formation in patients with AMI correlated with a higher percentage of CD14++CD16+ monocytes and VEGF-B186 levels in the circulation, which was associated with milder left ventricular remodeling. The regulation of CD14++CD16+ monocytes and VEGF-B may be critical to the formation of collateral circulation and to healing AMI.

Abstract Meat is among the most consumed foods worldwide and has a unique flavor and high nutrient density in the human diet. However, the genetic and biochemical bases of meat nutrition and flavor are poorly understood. Here, 3431 metabolites and 702 volatiles in 423 skeletal muscle samples are profiled from a gradient consanguinity segregating population generated by Pekin duck × Liancheng duck crosses using metabolomic approaches. The authors identified 2862 metabolome‐based genome‐wide association studies (mGWAS) signals and 48 candidate genes potentially modulating metabolite and volatile levels, 79.2% of which are regulated by cis‐regulatory elements. The level of plasmalogen is significantly associated with TMEM189 encoding plasmanylethanolamine desaturase 1. The levels of 2‐pyrrolidone and glycerophospholipids are regulated by the gene expression of AOX1 and ACBD5, which further affects the levels of volatiles, 2‐pyrrolidone and decanal, respectively. Genetic variations in GADL1 and CARNMT2 determine the levels of 49 metabolites including L‐carnosine and anserine. This study provides novel insights into the genetic and biochemical basis of skeletal muscle metabolism and constitutes a valuable resource for the precise improvement of meat nutrition and flavor.

Bovine pasteurellosis, caused by serogroups A, B, and E of Pasteurella multocida (Pm), is mainly manifested as bovine respiratory disease (BRD) and hemorrhagic septicemia (HS). The disease has caused a great economic loss for the cattle industry globally. Therefore, identifying the Pm serogroups is critical for optimal diagnosis and subsequent clinical treatment and even epidemiological studies. In this study, a one-step multiplex real-time PCR assay was established. Three pairs of specific primers were prepared to detect the highly conserved genomic regions of serogroups A (HyaD), B (bcbD), and E (ecbJ) of Pm, respectively. The results depicted that the method had no cross-reaction with other bovine pathogens (Mannheimia hemolytica, Escherichia coli, Listeria monocytogenes, Staphylococcus aureus, Salmonella Dublin, Mycobacterium paratuberculosis, infectious bovine rhinotracheitis virus, and Mycoplasma bovis). The linear range (107 to 102 copies/μL) showed the R2 values for serogroups A, B, and E of Pm as 0.9975, 0.9964, and 0.996, respectively. The multiplex real-time PCR efficiency was 90.30%, 90.72%, and 90.57% for CartA, CartB, and CartE, respectively. The sensitivity result showed that the serogroups A, B, and E of Pm could be detected to be as low as 10 copies/μL. The repeatability result clarified that an intra-assay and an inter-assay coefficient of variation of serogroups A, B, and E of Pm was < 2%. For the clinical samples, the detection rate was higher than the OIE-recommended ordinary PCR. Overall, the established one-step multiplex real-time PCR assay may be a valuable tool for the rapid and early detection of the serogroups A, B, and E of Pm with high specificity and sensitivity.

Chinese cities contributes a large amount of CO2 emissions. Reducing CO2 emissions through urban governance is an important issue. Despite the increasing attention paid on the CO2 emission prediction, few studies consider the collective and complex influence of governance element system. To predict and regulate CO2 emissions through comprehensive urban governance elements, this paper use the random forest model through the data from 1903 Chinese county-level cities in 2010, 2012 and 2015, and establish a CO2 forecasting platform based on the effects of urban governance elements. The results are as follows: (1) The municipal utility facilities element, the economic development & industrial structure element, and the city size &structure and road traffic facilities elements are crucial for residential, industrial and transportation CO2 emissions, respectively; (2) Governance elements have nonlinear relationship with CO2 emissions and most of the relations present obvious threshold effects; (3) Random forest can be used to predict CO2 emissions more accurately than can other predictive models. These findings can be used to conducts the CO2 scenario simulation and help government formulate active governance measurements.

Breast cancer (BC) tissues have been proved to harbor microorganisms, which could potentially contribute to oncogenesis. Formalin-fixed paraffin-embedded (FFPE) tissues are the most widespread clinical samples in BC research. To verify the potential of FFPE tissues in microbiological analysis, we analyzed the microbial communities of FFPE and fresh frozen (FF) tumor samples from 30 participants diagnosed with BC deploying 16S rRNA sequencing. The operational taxonomic units (OTUs) analysis showed that 78.55% of OTUs in FFPE samples were consistent with FF samples. The composition of core bacteria did not change much, and there is also no difference in alpha diversity between FFPE and FF (without unclassified bacteria). Taxonomic variation results show that Firmicutes and Bacteroidota phyla, and their major classes, maintained the same proportion under two preservation methods. In addition, the major class Gammaproteobacteria, as well as its dominant orders Burkholderiales and Pseudomonadales all showed no significant difference in paired analysis. Moreover, the Proteobacteria and Actinobacteriota phyla showed no significant difference between FFPE and FF samples after subtracting unclassified bacteria. Therefore, premised with the intrinsic tumor heterogeneity and unclassified bacteria, there are potential values of FFPE tissues for intratumoral microbiome analysis in breast cancer.

Matrix stiffness plays an important role in determining cell differentiation. The expression of cell differentiation-associated genes can be regulated by chromatin remodeling-mediated DNA accessibility. However, the effect of matrix stiffness on DNA accessibility and its significance for cell differentiation have not been investigated. In this study, gelatin methacryloyl (GelMA) hydrogels with different degrees of substitution were used to simulate soft, medium, and stiff matrices, and it was found that a stiff matrix promoted osteogenic differentiation of MC3T3-E1 cells by activating the Wnt pathway. In the soft matrix, the acetylation level of histones in cells was decreased, and chromatin condensed into a closed conformation, affecting the activation of β-catenin target genes (Axin2, c-Myc). Histone deacetylase inhibitor (TSA) was used to decondense chromatin. However, there was no significant increase in the expression of β-catenin target genes and the osteogenic protein Runx2. Further studies revealed that β-catenin was restricted to the cytoplasm due to the downregulation of lamin A/C in the soft matrix. Overexpression of lamin A/C and concomitant treatment of cells with TSA successfully activated β-catenin/Wnt signaling in cells in the soft matrix. The results of this innovative study revealed that matrix stiffness regulates cell osteogenic differentiation through multiple pathways, which involve complex interactions between transcription factors, epigenetic modifications of histones, and the nucleoskeleton. This trio is critical for the future design of bionic extracellular matrix biomaterials.

In order to remove NOx from multiple sources with complex operating conditions, catalysts with excellent low-temperature activity and strong water and sulfur resistance are in high demand. Herein, we designed an efficient inter-connected porous CuOx-NbOx/kit-CeO2 catalyst for the selective catalytic reduction of NOx with NH3 (NH3-SCR). The catalyst exhibited relatively outstanding NH3-SCR activity in the low temperature range (100–300 °C) and showed remarkable stability and excellent resistance to SO2 and H2O. The inter-connected porous structure allowed the adsorption and diffusion of gaseous reactants, meanwhile, the interaction between Cu, Nb and Ce enhanced the surface acidity, redox properties and surface oxygen activity of the CuOx-NbOx/kit-CeO2 catalyst. On the basis of in situ DRIFTs studies, Eley-Rideal process was expected to be the dominant pathway with the coexistence of Langmuir–Hinshelwood mechanism at 250 °C.

Gliomas represent the most common and lethal category of primary brain tumors. Bisphenol A (BPA), a widely recognized endocrine disruptor, has been implicated in the progression of cancer. Despite its established links to various cancers, the association between BPA and glioma progression remains to be clearly defined. This study aimed to shed light on the impact of BPA on glioma cell proliferation and overall tumor progression. Our results demonstrate that BPA significantly accelerates glioma cell proliferation in a time- and dose-dependent manner. Furthermore, BPA has been found to enhance the invasive and migratory capabilities of glioma cells, potentially promoting epithelial–mesenchymal transition (EMT) characteristics within these tumors. Employing bioinformatics approaches, we devised a risk assessment model to gauge the potential glioma hazards associated with BPA exposure. Our comprehensive analysis revealed that BPA not only facilitates glioma invasion and migration but also inhibits apoptotic processes. In summary, our study offers valuable insights into the mechanisms by which BPA may promote tumorigenesis in gliomas, contributing to the understanding of its broader implications in oncology.

Caffeic acid O-methyltransferase (COMT) participates in various physiological activities in plants, such as positive responses to abiotic stresses and the signal transduction of phytohormones. In this study, 18 COMT genes were identified in the chromosome-level reference genome of mango, named MiCOMTs. A phylogenetic tree containing nine groups (I-IX) was constructed based on the amino acid sequences of the 71 COMT proteins from seven species. The phylogenetic tree indicated that the members of the MiCOMTs could be divided into four groups. Quantitative real-time PCR showed that all MiCOMT genes have particularly high expression levels during flowering. The expression levels of MiCOMTs were different under abiotic and biotic stresses, including salt and stimulated drought stresses, ABA and SA treatment, as well as Xanthomonas campestris pv. mangiferaeindicae and Colletotrichum gloeosporioides infection, respectively. Among them, the expression level of MiCOMT1 was significantly up-regulated at 6–72 h after salt and stimulated drought stresses. The results of gene function analysis via the transient overexpression of the MiCOMT1 gene in Nicotiana benthamiana showed that the MiCOMT1 gene can promote the accumulation of ABA and MeJA, and improve the salt tolerance of mango. These results are beneficial to future researchers aiming to understand the biological functions and molecular mechanisms of MiCOMT genes.

Flight parameters are crucial criteria for UAV control, playing a significant role in ensuring the safe and efficient completion of missions. Launch force and airspeed information are key parameters in the early and middle stages of flight, serving as important data for monitoring the UAV’s flight status. In response to challenges such as weak launch force, low identification rates, small airspeed, and low recognition accuracy in UAVs, a method for identifying UAV flight parameters based on launch force and airspeed is proposed. From the aspect of launch force identification, a recognition method based on a low-g value accelerometer information source is proposed, utilizing a ‘multi-level time window + threshold’ approach. For airspeed identification, an optimization method for airspeed measurement under the Kalman filter architecture is introduced. A device for airspeed measurement based on pressure sensors is designed, and the recommended installation position is determined through simulation. Furthermore, the feasibility and robustness of the proposed launch force identification and airspeed measurement optimization methods are validated through simulation. Finally, the effectiveness of the design is verified through centrifuge and wind tunnel experiments. This research provides technical support for the identification of the launch force and airspeed measurement in UAVs.

Water-based chloroprene latex is a solvent-free, environmentally friendly adhesive. Currently, its market demand is growing rapidly. However, there are problems such as a lack of heat resistance and poor mechanical properties, which limit its application. The introduction of vinyl-POSS (OVS) into the resin structure can effectively improve the thermal stability of chloroprene adhesives. In this paper, modified waterborne chloroprene latex was prepared by copolymerization of methyl methacrylate and OVS with chloroprene latex. The results showed that vinyl-POSS was successfully grafted onto the main chain of the waterborne chloroprene latex, and the modified waterborne chloroprene latex had good storage stability. With the increase in vinyl-POSS, the tensile strength of the chloroprene latex firstly increased and then decreased, the tensile property (peel strength of 20.2 kgf) was maintained well at a high temperature (100 °C), and the thermal stability of the chloroprene latex was improved. When the addition amount was 4%, the comprehensive mechanical properties were their best. This study provides a new idea for the construction of a new and efficient waterborne chloroprene latex system and provides more fields for the practical application of waterborne chloroprene latex. This newly developed vinyl-POSS modified chloroprene latex has great application potential for use in home furniture, bags, and seat cushions.

Salt stress is a limiting factor affecting the growth, development, and yield of peanuts. Breeding improvement is a possible solution to overcome salt stress. The salt tolerance of 57 peanut cultivars in Northeast China was identified using indoor simulation at the germination stage, the seedling stage, and field natural identification. At the germination stage, 75 mM NaCl was the most suitable screening concentration, and the seed vitality index of 57 cultivars was analyzed using the membership function and cluster analysis. Among these cultivars, 11 were identified as salt-tolerant and 19 were salt-sensitive during germination. In the seedling stage, six salt tolerance coefficients (STCs) showed significant correlation. A gray relational analysis was used in combination with evaluation grading, resulting in the identification of 14 salt-tolerant cultivars and 12 salt-sensitive cultivars. In the field screening, a comprehensive analysis was conducted using a principal component analysis of nine indices, including agronomic characteristics, yield characteristics, and SPAD. This analysis led to the determination of three comprehensive indices. The weighted membership function was used for comprehensive evaluation. Finally, three salt-tolerant cultivars and four salt-sensitive cultivars suitable for planting in Northeast China were screened out to provide an excellent germplasm for researching the salt-tolerant mechanism of peanuts.

The synchronous reluctance machine is well-known for its highly nonlinear magnetic saturation and cross-saturation characteristics. For high performance and high-efficiency control, the flux-linkage maps and maximum torque per ampere table are of paramount importance. This study proposes a novel automated online searching method for obtaining accurate flux-linkage and maximum torque per ampere Identification. A limited 6 × 2 dq-axis flux-linkage look-up table is acquired by applying symmetric triangle pulses during the self-commissioning stage. Then, three three-dimensional modified linear cubic spline interpolation methods are applied to extend the flux-linkage map. The proposed golden section searching method can be easily implemented to realize higher maximum torque per ampere accuracy after 11 iterations with a standard drive, which is a proven faster solution with reduced memory sources occupied. The proposed algorithm is verified and tested on a 15-kW SynRM drive. Furthermore, the iterative and execution times are evaluated.

Synchronous reluctance machines are considered a cost-effective solution for several industrial applications and present potential efficiency benefits compared to induction motors. In industrial applications, power supply oscillations can lead to short-term disturbances that can affect the drive operation; therefore, the control must be robust to guarantee high efficiency and service continuity. The focus of this study was to identify the speed boundaries considering different values of applied DC-link voltage, taking into account the highly nonlinear magnetic behavior of the machine and its cross-coupling characteristics. In addition, a comprehensive carrier-based implementation of a pulse width modulation strategy was proposed to achieve optimal efficiency in both the machine and converter, which is essential in the presence of “weak” grids. The proposed technique was demonstrated to meet the desired reference torque and rated speed, even during DC-link voltage drops (up to 7.4% of the rated voltage). The proposed methodology was experimentally validated on a 90 kW SynRM drive with a broader modulation range and higher efficiency. This work considered several different supply voltage levels to assess the stability and continuity of torque output and further proved the proposed method.

In order to use high alumina coal gangue as auxiliary cementitious admixture via a simple and convenient thermal activation technique, the thermal transformation, mineral phase transformation and structure changes of coal gangue at calcining temperatures of 500–1000 °C were analyzed using x-ray diffraction (XRD), differential thermal Analysis (DTA), thermogravimetry (TG), infrared analysis (IR) and scanning electron microscope (SEM). The mechanical properties of cement mortar with 30% coal gangue auxiliary cementitious admixture were also measured to determine the optimal calcining temperature. As calcining temperature was increased, the coal gangue experienced the following transformations: carbon combustion, dehydroxylation, metakaolin transformation and mullite transformation. The cement mortar with coal gangue auxiliary cementitious admixture calcined at 700 °C presented the highest 28-d flexural and compressive strength, increasing by 8.27% and 11.85% respectively as compared with the benchmark cement mortar. The maximum dosage of coal gangue auxiliary cementitious admixture in cement mortar was further identified to be less than 30% by mechanical properties testing. The activity of high alumina coal gangues at different calcining temperatures was explained from the view points of hydration degree and products. The present investigation can provide a useful reference to utilize high alumina coal gangue as auxiliary cementitious admixture by means of a simple thermal activation at 700 °C.

In this paper, a new preparation technology is developed to make high-alumina coal gangue (HACG) auxiliary cementitious admixture by calcining HACG–Ca(OH)2 (CH) mixture. HACG powders mixed with 20 wt.% CH were calcined within a temperature range of 600–900 °C, and the thermal transformation and mineral phase formation were analyzed. The hydration reaction between activated HACG–CH mixture and cement was also investigated. The results showed that HACG experienced a conventional transformation from kaolinite to metakaolin at 600 °C and finally to mullite at 900 °C, whereas CH underwent an unexpected transformation process from CH to CaO, then to CaCO3, and finally to CaO again. These substances’ states were associated with the dehydroxylation of CH, the chemical reaction between CaO and CO2 generating from the combustion of carbon in HACG, and the decomposition of CaCO3, respectively. It is the formation of a large amount of CaO above 800 °C that favors the formation of hydratable products containing Al2O3 in the calcining process and C-A-H gel in the hydration process. The mechanical properties of HACG–cement mortar specimens were measured, from which the optimal calcination temperature of 850 °C was determined. As compared with pure cement mortar specimens, the maximum 28-d flexural and compressive strengths of HACG–cement mortar specimens increased by 5.4% and 38.2%, respectively.

This study aims to address the current situation of the late start of mechanized harvesting technology for the pod pepper, the high damage rate of existing pod pepper harvesters, and the lack of theoretical support for key harvesting components. The Hertz theory is employed to investigate the damage mechanism of collisions between pod pepper and comb fingers. The study analyzes the maximum deformation of pod pepper and the critical speed at which damage occurs during the collision process. Furthermore, it explores the critical relative speed that leads to damage in pod pepper. Orthogonal tests are conducted to analyze the effects of rotational speed, hose thickness, and moisture content on the efficiency of pod pepper picking. The experimental results are then subjected to multifactorial ANOVA to identify the optimal test parameters. The structural and motion parameters of the picking device are optimized based on these conditions. It is determined that the critical relative velocity for damage to pod pepper during a collision with the comb finger is V0 = 11.487 m s−1. The collision velocities of pod pepper with different hose thicknesses are analyzed using the i-SPEED TR endoscopic high-speed dynamic analysis system to obtain the corresponding collision velocities for different hose thicknesses. The study finds that rotational speed, hose thickness, and the water content of pod pepper affect the damage rate and stem shedding rate. The optimal experimental parameters are determined to be a rotational speed of 705.04 rpm, hose thickness of 3 mm, and water content of the pepper of 71.27%.

Here, the structural, magnetic, and dielectric features of breathing pyrochlore LiInCr _4−x Co _x O _8 were investigated. The XRD analysis showed changes in Cr–Cr length and distorted CrO _6 octahedral as Co ion doping level rises. The x-ray photoemission spectroscopy measurements showed that there are both Co ^2+ and Co ^3+ ions in the LiInCr _4−x Co _x O _8 compounds. The difference between temperature-dependent magnetization curves during field cooling and zero-field cooling suggested that there is a spin-glass state, which was verified by the heat capacity measurement. Doping affects the bond length, which affects the magnetic properties and electronic properties. The dielectric characteristics were systematically studied, and the temperature dependence of the dielectric loss and dielectric constant was examined in the temperature range of 323−573 K at various frequencies. The dielectric behavior strongly changes with the change in temperature and/or frequency. The activation energies of grain boundary resistance of doped samples show a decreasing trend with increasing doping.

Deep-learning- and unmanned aerial vehicle (UAV)-based methods facilitate structural crack detection for tall structures. However, contemporary datasets are generally established using images taken with handheld or vehicle-mounted cameras. Thus, these images might be different from those taken by UAVs in terms of resolution and lighting conditions. Considering the difficulty and complexity of establishing a crack image dataset, making full use of the current datasets can help reduce the shortage of UAV-based crack image datasets. Therefore, the performance evaluation of existing crack image datasets in training deep neural networks (DNNs) for crack detection in UAV images is essential. In this study, four DNNs were trained with different architectures based on a publicly available dataset and tested using a small UAV-based crack image dataset with 648 +pixel-wise annotated images. These DNNs were first tested using the four indices of precision, recall, mIoU, and F1, and image tests were also conducted for intuitive comparison. Moreover, a field experiment was carried out to verify the performance of the trained DNNs in detecting cracks from raw UAV structural images. The results indicate that the existing dataset can be useful to train DNNs for crack detection from UAV images; the TransUNet achieved the best performance in detecting all kinds of structural cracks.

When a missile-borne pulsed laser forward detection system flies at supersonic speed, the laser beam will be distorted by the uneven outflow field, resulting in a significant reduction in ranging accuracy. In this paper, the impact of high flight speed on a pulsed laser detection system is studied. First, a new ray tracing method with adaptive step size adjustment is proposed, which greatly improves the computational efficiency. Second, the aerodynamic environment of a munition flying at high speed is simulated by an intermittent transonic and supersonic wind tunnel to obtain the schlieren data of the flow field at various Mach numbers. The schlieren data present a shock wave structure similar to that of the simulation. In addition, the variation patterns of the pulsed laser echo waveform of the model under different aerodynamic conditions are studied to evaluate the detectability and operational stability of the laser detection system under static conditions. The test results match the simulation results well, and the two offer relatively consistent shock wave structures, which verifies the correctness and effectiveness of the flow field simulation model. The test echo waveforms are in good agreement with the simulated echo waveforms; the relative errors between the peak values of test and simulated echo waveforms at various Mach numbers do not exceed 20%, and the correlation coefficients between the test and simulated echo waveforms all exceed 0.7, indicating high correlations between the two.

Infrared small target detection plays a crucial role in maritime security. However, detecting small targets within heavy sea clutter environments remains challenging. Existing methods often fail to deliver satisfactory performance in the presence of substantial clutter interference. This paper analyzes the spatial–temporal appearance characteristics of small targets and sea clutter. Based on this analysis, we propose a novel detection method based on the appearance stable isotropy measure (ASIM). First, the original images are processed using the Top-Hat transformation to obtain the salient regions. Next, a preliminary threshold operation is employed to extract the candidate targets from these salient regions, forming a candidate target array image. Third, to distinguish between small targets and sea clutter, we introduce two characteristics: the gradient histogram equalization measure (GHEM) and the local optical flow consistency measure (LOFCM). GHEM evaluates the isotropy of the candidate targets by examining their gradient histogram equalization, while LOFCM assesses their appearance stability based on local optical flow consistency. To effectively combine the complementary information provided by GHEM and LOFCM, we propose ASIM as a fusion characteristic, which can effectively enhance the real target. Finally, a threshold operation is applied to determine the final targets. Experimental results demonstrate that our proposed method exhibits superior comprehensive performance compared to baseline methods.

Vinyl-capped cationic waterborne polyurethane (CWPU) was prepared using isophorone diisocyanate (IPDI), polycarbonate diol (PCDL), N-methyldiethanolamine (MDEA), and trimethylolpropane (TMP) as raw materials and hydroxyethyl methacrylate (HEMA) as a capping agent. Then, a crosslinked FPUA composite emulsion with polyurethane (PU) as the shell and fluorinated acrylate (PA) as the core was prepared by core-shell emulsion polymerization with CWPU as the seed emulsion, together with dodecafluoroheptyl methacrylate (DFMA), diacetone acrylamide (DAAM), and methyl methacrylate (MMA). The effects of the core-shell ratio of PA/PU on the surface properties, mechanical properties, and heat resistance of FPUA emulsions and films were investigated. The results showed that when w(PA) = 30~50%, the stability of FPUA emulsion was the highest, and the particles showed a core-shell structure with bright and dark intersections under TEM. When w(PA) = 30%, the tensile strength reached 23.35 ± 0.08 MPa. When w(PA) = 50%, the fluorine content on the surface of the coating film was 14.75% and the contact angle was as high as 98.5°, which showed good hydrophobicity; the surface flatness of the film was observed under AFM. It is found that the tensile strength of the film increases and then decreases with the increase in the core-shell ratio and the heat resistance of the FPUA film is gradually increased. The FPUA film has excellent properties such as good impact resistance, high flexibility, high adhesion, and corrosion resistance.

The morphological characteristics of a crack serve as crucial indicators for rating the condition of the concrete bridge components. Previous studies have predominantly employed deep learning techniques for pixel-level crack detection, while occasionally incorporating monocular devices to quantify the crack dimensions. However, the practical implementation of such methods with the assistance of robots or unmanned aerial vehicles (UAVs) is severely hindered due to their restrictions in frontal image acquisition at known distances. To explore a non-contact inspection approach with enhanced flexibility, efficiency and accuracy, a binocular stereo vision-based method incorporating full convolutional network (FCN) is proposed for detecting and measuring cracks. Firstly, our FCN leverages the benefits of the encoder–decoder architecture to enable precise crack segmentation while simultaneously emphasizing edge details at a rate of approximately four pictures per second in a database that is dominated by complex background cracks. The training results demonstrate a precision of 83.85%, a recall of 85.74% and an F1 score of 84.14%. Secondly, the utilization of binocular stereo vision improves the shooting flexibility and streamlines the image acquisition process. Furthermore, the introduction of a central projection scheme achieves reliable three-dimensional (3D) reconstruction of the crack morphology, effectively avoiding mismatches between the two views and providing more comprehensive dimensional depiction for cracks. An experimental test is also conducted on cracked concrete specimens, where the relative measurement error in crack width ranges from −3.9% to 36.0%, indicating the practical feasibility of our proposed method.

The objective of this work is to systematically evaluate the performance of the hydrolysis-based kraft pulping process and associated pulp and black liquor characteristics. Acacia wood chips were auto-hydrolyzed under various severities, then the hydrolyzed wood chips were kraft pulping. The results indicated that the yield of pulp significantly dropped with intensifying the auto-hydrolysis severity. Meanwhile, the removal rate of pentosan reached 98.6% in the screened pulp at the P-factor of 1 000. The fiber length, fines and fiber crimp of the screened pulp were not affected by the auto-hydrolysis treatment. Auto-hydrolyzed pulps deteriorated fibrillation and beating response of the pulp in a refining process. However, fiber length and fines changed obviously after beating treatment. After auto-hydrolysis, the tensile index of the paper matrices decreased, some particle substances were found on the surface of the pulp fiber, and the solid and organic content of the black liquor were improved.

In order to promote the optimization of marine industry structure and the high-quality development of marine economy in Qingdao, this paper established the evaluation index system, analyzed the marine industry which had an important impact on the development of marine economy in Qingdao by using Lasso regression model, and put forward development suggestions. The results showed that the evaluation index system of Qingdao′s marine economic development level included 6 categories and 20 characteristic variables, which were marine fishery, marine chemical industry, total marine import and export, marine environment, marine transportation and coastal tourism. The optimal value of the adjustment coefficient was determined by 10 fold cross validation, and 10 characteristic variables were selected into the regression model by solving the Lasso regression coefficient. According to the estimated value of regression coefficient, the number of domestic passengers was the most important, the value of marine fishery output was important, and the value of cargo throughput was the least important. Lasso regression model had high prediction accuracy, model generalization ability and reliability, and belonged to robust sparse model. In the future, Qingdao should give full play to the industrial advantages of coastal tourism and marine fishery, focus on the development of marine transportation industry, protect the marine ecological environment and increase the investment in marine science and technology research and development.

Abstract Background Increasing studies have demonstrated potential disproportionate functional and ecological contributions of rare taxa in a microbial community. However, the study of the microbial rare biosphere is hampered by their inherent scarcity and the deficiency of currently available techniques. Sample-wise cross contaminations might be introduced by sample index misassignment in the most widely used metabarcoding amplicon sequencing approach. Although downstream bioinformatic quality control and clustering or denoising algorithms could remove sequencing errors and non-biological artifact reads, no algorithm could eliminate high quality reads from sample-wise cross contaminations introduced by index misassignment, making it difficult to distinguish between bona fide rare taxa and potential false positives in metabarcoding studies. Results We thoroughly evaluated the rate of index misassignment of the widely used NovaSeq 6000 and DNBSEQ-G400 sequencing platforms using both commercial and customized mock communities, and observed significant lower (0.08% vs. 5.68%) fraction of potential false positive reads for DNBSEQ-G400 as compared to NovaSeq 6000. Significant batch effects could be caused by stochastically introduced false positive or false negative rare taxa. These false detections could also lead to inflated alpha diversity of relatively simple microbial communities and underestimated that of complex ones. Further test using a set of cow rumen samples reported differential rare taxa by different sequencing platforms. Correlation analysis of the rare taxa detected by each sequencing platform demonstrated that the rare taxa identified by DNBSEQ-G400 platform had a much higher possibility to be correlated with the physiochemical properties of rumen fluid as compared to NovaSeq 6000 platform. Community assembly mechanism and microbial network correlation analysis indicated that false positive or negative rare taxa detection could lead to biased community assembly mechanism and identification of fake keystone species of the community. Conclusions We highly suggest proper positive/negative/blank controls, technical replicate settings, and proper sequencing platform selection in future amplicon studies, especially when the microbial rare biosphere would be focused.

Abstract A considerable number of glioblastoma (GBM) patients developed drug resistance to Temozolomide (TMZ) during chemotherapy, resulting in therapeutic failure and tumor recurrence. However, the exact mechanism of TMZ chemoresistance in GBM is still poorly clarified. As a novel identified lncRNA, LINC00520 was located on chromosome 14 and overexpressed in multiple human cancers. This study was designed and conducted to investigate the role and underlying mechanism of LINC00520 in GBM chemoresistance to TMZ. This paper presents the multi-physics fields research of a high-speed permanent magnet machine (HSPMM) for turbine application. At first, the design considerations for the HSPMM are introduced and presented. Then, the machine electromagnetic performances are comprehensively studied with machine stator slot numbers. Moreover, the influences from the PM magnetization methods and segmentation number on PM eddy current loss reduction are further investigated. Thermal analysis is also performed for the HSPMM with the cooling water fluid volume flow rate considered. In addition, rotor strength is studied for the HSPMM with several design factors on machine rotor strength performances further studied and researched.

Primary liver tumors (PLCs) and liver metastasis currently represent the leading cause of cancer‐related deaths worldwide due to poor outcomes, high incidence, and postsurgical recurrence. Hence, novel diagnostic markers and therapeutic strategies for PLCs are urgently needed. The human microbiome can directly or indirectly impact cancer initiation, progression, and response to therapy, including cancer immunotherapy; however, the roles of the microbiota in the tumor microenvironment are not clear and require more investigation. Here, we investigated intratumoral microbial community profiling on formalin‐fixed paraffin‐embedded tissue samples of patients with PLC by 16S ribosomal RNA using the MiSeq platform. We characterized the microbial communities in different histopathological subtypes and in the different prognoses of patients with PLC. The study revealed microbial population differences not only in carcinoma tissue and the matched adjacent nontumor tissue but in different histopathological subtypes, even in patients with PLC with different prognoses. Interestingly, the abundance of certain bacteria that have antitumor effects at family and genus level, such as Pseudomonadaceae, decreased in tumor tissue and was linearly associated with prognosis of patients with PLC. Conclusion: We provide a potential novel diagnostic biomarker and therapeutic strategy for early clinical diagnosis and treatment of PLC.

Abstract Background The importance of enteral nutrition (EN) in acute pancreatitis (AP) has been emphasised. Nasogastric (NG) feeding has been the preferred route for EN delivery in most AP patients intolerant to oral intake. However, gastric feeding intolerance (GFI) was frequently reported, especially in patients with more severe diseases. This study aimed to investigate the incidence and risk factors for GFI in moderately-severe to severe AP. Methods This is a single-centre, retrospective study. All the data were extracted from an electronic database from April 2020 to May 2021. Data were prospectively collected during hospitalisation. Patients diagnosed with moderately-severe to severe AP and admitted within seven days from the onset of abdominal pain were assessed for eligibility. Patients who showed signs of intolerance to gastric feeding and required switching to nasojejunal (NJ) feeding were deemed GFI. Multivariable logistic regression was performed to assess potential risk factors of GFI. Results A total of 93 patients were analysed, of whom 24 were deemed GFI (25.8%), and the rest tolerated NG feeding well (n = 69). In patients with GFI, the median time of switching to NJ feeding was five days (interquartile range: 4–7 days) after admission. The multivariable analysis showed that respiratory failure (odds ratio = 3.135, 95% CI: 1.111–8.848, P = 0.031) was an independent risk factor for GFI.The mean daily energy delivery in the following three days after switching to NJ feeding was significantly higher than the first three days after initiation of NG feeding in patients with GFI [920.83 (493.33–1326) vs. 465 (252.25–556.67) kcal, P

In response to the demand for short-range detection of anti-smoke environment interference by laser fuzes, this study proposes a smoke environment simulation of non-uniform continuous point source diffusion and investigates an experimental laboratory smoke environment using an ammonium chloride smoke agent. The particle size distribution, composition, and mass flow distribution of the smoke were studied. Based on a discrete phase model and a k-ε turbulence model, a numerical simulation was developed to model the smoke generation and diffusion processes of the smoke agent in a confined space. The temporal and spatial distribution characteristics of the smoke mass concentration, velocity, and temperature in the space after smoke generation were analyzed, and the motion law governing the smoke diffusion throughout the entire space was summarized. Combined with the experimental verification of the smoke environment laboratory, the results showed that the smoke plume changed from fan-shaped to umbrella-shaped during smoke generation, and then continued to spread around. Meanwhile, the mass concentration of smoke in the space decreased from the middle outward; the changes in temperature and velocity were small and stable. In the diffusion stage (after 900 s), the mass concentration of smoke above 0.8 m was relatively uniform across an area of smoke that was 12 m thick. The concentration decreased over time, following a consistent decreasing trend, and the attenuation was negligible in a very short time. Therefore, this system was suitable for conducting experimental research on laser fuzes in a smoke environment. Owing to the stability of the equipment and facilities, the setup could reproduce the same experimental smoke environment by artificially controlling the smoke emission of the smoke agent. Overall, this work provides a theoretical reference for subsequent research efforts regarding the construction of uniform smoke environments and evaluating laser transmission characteristics in smoky environments.

Abstract Background Postoperative pneumonia (PP) is one of the most common complications after cardiac surgery. This study was designed to access the diagnostic value of interleukin-6 (IL-6) for pneumonia within the first 5 days after cardiac surgery in adults. Method This prospective observational study enrolled 694 patients who admitted to our center from 10 October 2020 to 30 June 2021. Blood samples were collected after admission and on five consecutive days after surgery to measure IL-6, procalcitonin (PCT), C-reactive protein (CRP) and white blood cells (WBC) respectively. Combined with clinical data, we assessed the diagnostic performance of different biomarkers using univariate and multifactorial analyses as well as receiver operating characteristic curves (ROC) and the area under the curve (AUC). Tumor-associated macrophages (TAMs), one of the dominating constituents of tumor microenvironment, are important contributors to cancer progression and treatment resistance. Therefore, regulation of TAMs polarization from M2 phenotype towards M1 phenotype has emerged as a new strategy for tumor immunotherapy. Herein, we successfully initiated antitumor immunotherapy by inhibiting TAMs M2 polarization via autophagy intervention with polyethylene glycol-conjugated gold nanoparticles (PEG-AuNPs). PEG-AuNPs suppressed TAMs M2 polarization in both in vitro and in vivo models, elicited antitumor immunotherapy and inhibited subcutaneous tumor growth in mice. As demonstrated by the mRFP-GFP-LC3 assay and analyzing the autophagy-related proteins (LC3, beclin1 and P62), PEG-AuNPs induced autophagic flux inhibition in TAMs, which is attributed to the PEG-AuNPs induced lysosome alkalization and membrane permeabilization. Besides, TAMs were prone to polarize towards M2 phenotype following autophagy activation, whereas inhibition of autophagic flux could reduce the M2 polarization of TAMs. Our results revealed a mechanism underlying PEG-AuNPs induced antitumor immunotherapy, where PEG-AuNPs reduce TAMs M2 polarization via induction of lysosome dysfunction and autophagic flux inhibition. This study elucidated the biological effects of nanomaterials on TAMs polarization and provided insight into harnessing the intrinsic immunomodulation capacity of nanomaterials for effective cancer treatment.
