The goal of this study was to understand which factors, related to school science, can interfere with engagement of students-at-risk-of-dropping-out with school science and to know what kind of activities and teaching strategies are adequate to these students. This case-study involved a chemistry-teacher and ten male students. Data was based on teacher's interview, teacher's and students' notes regarding activities implementation and students' questionnaires. Results show that linking science with society by means of practical activities - that are student centred and require agency, autonomy, and the mobilization of complex competencies - facilitated the students' engagement with school science. The treatment of the scoliosis needs periodic radiologics evaluations, increasing the risks of the radiation. The use of methods no-invasivos it can be an alternative. The aim was to evaluate individuals with an instrument noninvasive for quantification of the hump deformity and to correlate these measured with the values of the Cobb angle. Forty one participants accomplished anthropometric evaluation and Adams test, being applied an instrument composed by level plane and a ruler, to measure the hump deformity. It was requested radiologics exam in the measures > 5 mm. The lineal coefficient of Pearson was used. The escoliose was diagnosed in 68,4% of the total of x-rays. There was better association among the measure of the thoracic deformity with the thoracic Cobb angle (right, r=0,91; left, r=0,83). The positioning adopted during Adams test could favor the best correlation of the thoracic values. It was concluded that the method no-invasivo it presented better results for thoracic curves. Salaria fluviatilis is one of the few freshwater members of the family Blenniidae and occurs around the Mediterranean Basin. This species is vulnerable or endangered in most countries where it occurs. Thus, information on its developmental biology and the establishment of methods for ex situ reproduction of highly endangered populations, to preserve them until natural habitats are restored, are much needed. A technique for the culture of this endangered species in controlled conditions is presented, together with the description of the full developmental sequence, from egg to adult. The use of the rotifer Brachionus calyciflorus at the onset of exogenous feeding proved to be an efficient way to allow larvae to reach the size when they can feed upon Artemia nauplii. Embryonic development lasted 12–14 d at 20–21 C. Newly hatched larvae measured 5.1 mm total length (TL). The mouth and anus were opened; the eyes were pigmented; there were almost no yolk; and the pectoral fins were small and unpigmented. Most larvae settled at 13.0–14.0 mm TL (27–31 d after hatching) and showed full juvenile pigmentation patterns at 27.0–28.0 mm TL (83 d after hatching). The larvae of this species showed agonistic behaviors once they began to settle. [ABSTRACT FROM AUTHOR]

Background: The expression of glucocorticoid-receptor (GR) seems to be a key mechanism in the regulation of glucocorticoid (GC) sensitivity and is potentially involved in cases of GC resistance or hypersensitivity. The aim of this study is to describe a method for quantitation of GR alpha isoform (GRá) expression using real-time PCR (qrt-PCR) with analytical capabilities to monitor patients, offering standard-curve reproducibility as well as intra- and inter-assay precision. Results: Standard-curves were constructed by employing standardized Jurkat cell culture procedures, both for GRá and BCR (breakpoint cluster region), as a normalizing gene. We evaluated standard-curves using five different sets of cell culture passages, RNA extraction, reverse transcription, and qrt-PCR quantification. Intra-assay precision was evaluated using 12 replicates of each gene, for 2 patients, in a single experiment. Inter-assay precision was evaluated on 8 experiments, using duplicate tests of each gene for two patients. Standard-curves were reproducible, with CV (coefficient of variation) of less than 11%, and Pearson correlation coefficients above 0,990 for most comparisons. Intra-assay and inter-assay were 2% and 7%, respectively. Conclusion: This is the first method for quantitation of GRá expression with technical characteristics that permit patient monitoring, in a fast, simple and robust way. [ABSTRACT FROM AUTHOR]

Telomerase reverse transcriptase (TERT) promoter mutations were recently shown to drive telomerase activity in various cancer types, including medulloblastoma. However, the clinical and biological implications of TERT mutations in medulloblastoma have not been described. Hence, we sought to describe these mutations and their impact in a subgroup-specific manner. We analyzed the TERT promoter by direct sequencing and genotyping in 466 medulloblastomas. The mutational distributions were determined according to subgroup affiliation, demographics, and clinical, prognostic, and molecular features. Integrated genomics approaches were used to identify specific somatic copy number alterations in TERT promoter-mutated and wild-type tumors. Overall, TERT promoter mutations were identified in 21 % of medulloblastomas. Strikingly, the highest frequencies of TERT mutations were observed in SHH (83 %; 55/66) and WNT (31 %; 4/13) medulloblastomas derived from adult patients. Group 3 and Group 4 harbored this alteration in <5 % of cases and showed no association with increased patient age. The prognostic implications of these mutations were highly subgroup-specific. TERT mutations identified a subset with good and poor prognosis in SHH and Group 4 tumors, respectively. Monosomy 6 was mostly restricted to WNT tumors without TERT mutations. Hallmark SHH focal copy number aberrations and chromosome 10q deletion were mutually exclusive with TERT mutations within SHH tumors. TERT promoter mutations are the most common recurrent somatic point mutation in medulloblastoma, and are very highly enriched in adult SHH and WNT tumors. TERT mutations define a subset of SHH medulloblastoma with distinct demographics, cytogenetics, and outcomes. Este livro surgiu como produto do trabalho de investigação realizado por uma equipa na Universidade de Lisboa, que sentiu necessidade de promover a discussão reflexiva sobre práticas de formação nos Mestrados em Ensino de Biologia e Geologia, Física e Química, Matemática e Educação Física da Universidade de Lisboa. Os dados recolhidos e a sua análise deram origem a uma grande quantidade de informação, muito rica, diversa, com particularidades mais acentuadas, por vezes, numa determinada área científica. A discussão subsequente permitiu perceber a existência de certas características importantes no funcionamento dos cursos de Mestrado em Ensino de Biologia e Geologia, Física e Química, Matemática e Educação Física, identificando-se aspetos relevantes que emergem do que é comum e do que se configura como exemplo nesses cursos de Mestrado em Ensino. Dá-se atenção, em primeiro lugar, a uma caracterização global, introdutória, dos próprios cursos de Mestrado em ensino e a aspetos teóricos relacionados com a formação inicial. Considerou-se, em seguida, definir como particularidades a serem exploradas os modos como cada área percebe algumas situações, mesmo que de um modo geral estejam presentes em todas elas.

The use of computational tools to identify biological targets of natural products with anticancer properties and unknown modes of action is gaining momentum. We employed self-organizing maps to deconvolute the phenotypic effects of piperlongumine (PL) and establish a link to modulation of the human transient receptor potential vanilloid 2 (hTRPV2) channel. The structure of the PL-bound full-length rat TRPV2 channel was determined by cryo-EM. PL binds to a transient allosteric pocket responsible for a new mode of anticancer activity against glioblastoma (GBM) in which hTRPV2 is overexpressed. Calcium imaging experiments revealed the importance of Arg539 and Thr522 residues on the antagonistic effect of PL and calcium influx modulation of the TRPV2 channel. Downregulation of hTRPV2 reduces sensitivity to PL and decreases ROS production. Analysis of GBM patient samples associates hTRPV2 overexpression with tumor grade, disease progression, and poor prognosis. Extensive tumor abrogation and long term survival was achieved in two murine models of orthotopic GBM by formulating PL in an implantable scaffold/hydrogel for sustained local therapy. Furthermore, in primary tumor samples derived from GBM patients, we observed a selective reduction of malignant cells in response to PL ex vivo. Our results establish a broadly applicable strategy, leveraging data-motivated research hypotheses for the discovery of novel means tackling cancer.

Esta publicação é financiada por fundos nacionais através da FCT - Fundação para a Ciência e a Tecnologia, no âmbito do projeto "Entre a terra e o mar: um projeto de integração de literacias" (iLit) (PTDC/CPE-CED/117923/2010)., O livro “Era uma vez…O mar. O mar como recurso educativo no 1º ciclo: O contributo do projeto iLit” foi desenvolvido no âmbito do projeto “Entre a terra e o mar: um projeto de integração de literacias”, que teve como principal objetivo o desenvolvimento de tarefas de investigação, focadas em situações reais do dia-a-dia, que promovem a integração de diferentes literacias. Estas investigações envolvem a exploração integrada de diferentes contextos de aprendizagem, dentro da sala de aula, em instituições de educação não formal, como museus de ciência e laboratórios de investigação, e o trabalho de campo. O livro encontra-se organizado em duas partes. A primeira parte inclui a fundamentação teórica, perspetivando recursos exteriores à escola como contextos de aprendizagem, e a integração das visitas de estudo no trabalho realizado em sala de aula, de acordo com ideias chave sobre a aprendizagem e as recomendações atuais na educação em ciência, assim como os resultados de investigação sobre a implementação das tarefas, tendo em conta o seu impacto nas aprendizagens dos alunos. 3. Good health - Abstract
The use of computational tools to identify biological targets of natural products with anticancer properties and unknown modes of action is gaining momentum. We employed self-organizing maps to deconvolute the phenotypic effects of piperlongumine (PL) and establish a link to modulation of the human transient receptor potential vanilloid 2 (hTRPV2) channel. The structure of the PL-bound full-length rat TRPV2 channel was determined by cryo-EM. PL binds to a transient allosteric pocket responsible for a new mode of anticancer activity against glioblastoma (GBM) in which hTRPV2 is overexpressed. Calcium imaging experiments revealed the importance of Arg539 and Thr522 residues on the antagonistic effect of PL and calcium influx modulation of the TRPV2 channel. Downregulation of hTRPV2 reduces sensitivity to PL and decreases ROS production. Analysis of GBM patient samples associates hTRPV2 overexpression with tumor grade, disease progression, and poor prognosis. Extensive tumor abrogation and long term survival was achieved in two murine models of orthotopic GBM by formulating PL in an implantable scaffold/hydrogel for sustained local therapy. Furthermore, in primary tumor samples derived from GBM patients, we observed a selective reduction of malignant cells in response to PL ex vivo. O livro decorre do Projeto Avaliação do Currículo das Ciências Físicas e Naturais do 3º Ciclo do Ensino Básico, financiado pela FCT (PTDC/CPE-CED/102789/2008) e inclui os resultados obtidos por vários estudos desenvolvidos. Trata-se de um estudo de avaliação, processo pouco corrente em Portugal. Passada mais de uma década do currículo das Ciências Físicas e Naturais ter sido introduzido no país, importa conhecer qual a interpretação do currículo pelos diferentes agentes educativos, como forma de dar um sentido aos resultados obtidos pelos alunos e às aprendizagens por eles realizadas. Qual o nível de penetração das ideias curriculares ao nível de professores e de autores dos manuais escolares? Como vivem os alunos esse currículo e que efeito teve o currículo na sua aprendizagem? Para tal, desenvolvemos um conjunto variado de estudos, com focos, objetivos e métodos diferentes. Os resultados permitem-nos perceber que o currículo das CFN não foi compreendido por grande parte dos professores nem por autores dos manuais escolares. As práticas mantiveram-se, na sua maioria, como eram antes, apesar da terminologia nova ter sido adotada e utilizada. Estes resultados permitem-nos elaborar algumas recomendações a editores de manuais escolares, instituições de formação de professores, decisores de política educativa e professores. Permitem-nos, além disso, contribuir para a compreensão da educação em Portugal, especialmente a de natureza curricular, podendo iluminar um caminho de melhor atuação na área das ciências.

UFU - Universidade Federal de Uberlândia A baixa estatura em crianças com doença renal crônica (DRC) tem etiologia multifatorial, altas prevalência e morbimortalidade e se torna mais frequente com a redução progressiva da taxa de filtração glomerular. O objetivo deste trabalho é analisar como os fatores de risco se associam à baixa estatura no início e no final do tratamento conservador. Este trabalho é uma coorte retrospectiva, na qual foram avaliados os prontuários dos pacientes com DRC, com idade no início do seguimento inferior a 13 anos, entre os anos de 1999 e 2015. Os pacientes (n=71) foram acompanhados durante 62,4 ± 49 meses, sendo 60,6% do sexo masculino. Foram avaliados idade, altura, presença de hipertensão arterial sistêmica (HAS), anemia, acidose metabólica, proteinúria e a TFG estimada (fórmula de Schwartz). Os pacientes foram divididos em grupos quanto à estatura inicial (normal ou baixa) e TFG inicial (menor ou maior que 30ml/min/1,73m2). Na análise estatística foram utilizados o teste do Qui-Quadrado, teste t pareado, curvas de Kaplan Meier e modelo de regressão de Cox multivariado. Vinte e cinco pacientes apresentavam baixa estatura no início do estudo. A acidose estava associada à baixa estatura no início do acompanhamento enquanto a TFG e a anemia estavam ao final. Houve redução significativa do número de pacientes hipertensos no grupo que apresentava estatura inicial normal e no grupo com TFG inicial ≥ 30ml/min/1,73m2, mas a HAS não esteve associada à baixa estatura. A dobra de creatinina, considerado marcador de progressão da doença renal, foi o fator independente mais significativo para a baixa estatura (r=10,3; P0,05). Conclui-se que, mesmo com o controle clínico das crianças com DRC no tratamento conservador, houve uma progressão do número de casos de baixa estatura durante o acompanhamento, provavelmente devido à referência tardia ao nosso serviço e à redução progressiva da função renal nestes pacientes. Os fatores de risco estudados estão diferentemente associados à baixa estatura no início e no final do acompanhamento das crianças com DRC em tratamento conservador. Short stature in children with chronic kidney disease (CKD) has a multifactorial etiology, high prevalence, and high morbidity and mortality, which becomes more frequent with the progressive reduction of the glomerular filtration rate (GFR). The objective of this study was to analyze risk factors associated with short stature at both the beginning and end of conservative treatment. This is a retrospective cohort study, in which the medical records of patients with CKD, aged less than 13 years at the beginning of follow-up, were evaluated between 1999 and 2015. Of the 71 patients examined, 60.6% were male and the average follow-up duration was 62.4 ± 49 months. Age, height, presence of systemic blood hypertension (SBH), anemia, metabolic acidosis, proteinuria, and estimated GFR were evaluated (Schwartz's formula). Patients were divided into groups based on initial stature (normal or low) and initial GFR (< or ≥30 mL/min/1.73m2). The Chi-square test, paired t-test, Kaplan-Meier curves, and multivariate Cox regression models were used in the statistical analysis. Twenty-five patients displayed short stature at the beginning of the study. Acidosis was associated with short stature at the start of follow-up, while GFR and anemia were associated with short stature at the end of follow-up. There was a significant reduction in the number of hypertensive patients in the GRF initial ≥ 30ml/min/1,73m2 and normal initial stature groups, but SBH was not associated with short stature. Vários investigadores fazem referência aos diversos riscos associados à maternidade precoce, tais como o elevado risco das mães adolescentes maltratarem os filhos, sendo até referenciadas como "capital de risco" para o seu desenvolvimento, e, no âmbito da literatura psicológica, têm sido destacadas as questões de natureza parental. Acresce a estas preocupações o efeito negativo da maternidade nos diversos níveis da trajetória desenvolvimental da adolescente, particularmente nos domínios educacional, socioeconómico, ocupacional, social e psicológico. Neste sentido, a presente investigação visa avaliar quais são os efeitos de um projeto educativo que visa capacitar mães adolescentes institucionalizadas num Centro de Apoio à Vida (CAV) para promover a saúde do/a filho/a e a sua própria saúde sexual. Foram envolvidas no projeto quatro mães adolescentes. Os dados recolhidos através de uma entrevista individual anónima no início e no fim do projeto, por observação participante e por análise dos documentos produzidos pelas adolescentes durante o projeto educativo, foram triangulados. Ao longo do projeto verificou-se que estas mães adolescentes melhoraram os seus conhecimentos sobre a amamentação, os cuidados com o/a filho/a e o evitamento da reincidência da gravidez e das infeções sexualmente transmissíveis. Também se observaram algumas evidências de melhoria a nível psicológico por exemplo, no nível de depressão, baixa autoestima e isolamento social. Este estudo mostra que será importante no futuro desenvolver projetos educativos orientados para a ação na promoção da maternidade saudável com adolescentes institucionalizadas., Several researchers refer to the various risks associated with early childbearing, such as high risk of teenage mothers mistreating children, even being referred to as "risk capital" for their development, and, within the psychological literature the issues of parental nature have been outstanding. In addition to these concerns the negative effect of motherhood on various levels of developmental trajectory of adolescent, particularly in socio-economic, occupational, social, educational and psychological domains exist. In this sense, this research aims to analyse what are the effects of an educational project that aims to empower adolescent mothers institutionalized in Life Support Center (Centro de Apoio à Vida - CAV) to promote the health of their child and their own sexual health. Were involved in the project four teenage mothers. The data collected through an anonymous individual semi-structured interview at the beginning and end of the project, participant observation and the analysis of documents produced by teenagers during the educational project, were triangulated. Throughout the project it was found that these teenage mothers improved their knowledge about breastfeeding, child-care and the avoidance of recurrence of pregnancy and sexually transmitted infections. Some evidences regarding the improvement of the psychological level, for example levels of depression, low self-esteem and social isolation were also observed. 237. Metopic craniosynostosis: Dynamic cranioplasty for trigonocephaly versus fronto-orbital remodeling and advancement - A retrospective study.

The aim of this study was to compare a traditional fronto-orbital remodeling and advancement (FORA) with the dynamic cranioplasty for trigonocephaly (DCT). The authors analyzed patients who underwent surgery for trigonocephaly. Perioperative data were compared. Parents were asked to use a visual analog scale to evaluate the pre- and postoperative distance between the eyes, the forehead shape, and the global appearance of the face. A panel of observers was asked to grade pre- and postoperative photographs using a similar visual analog scale. Pre- and postoperative anthropometric data were collected and analyzed in a subset of the study population aged 9 years or older. The total sample size was 51 patients (DCT n = 39; FORA n = 12). Durations of surgery and anesthesia were shorter in the DCT group (115 vs 194 min, p = 0.001; 226 vs 289 min, p = 0.001). Patients in the DCT group received similar preoperative ratings to those in the FORA group, but significantly higher postoperative ratings by parents for all three questions. There were no significant differences in postoperative ratings by the panel or postoperative anthropometric data. DCT is safe and effective. - Abstract
The aim of our study was to evaluate the effectiveness of intraoperative tranexamic acid (TXA) administration in decreasing transfusion needs in patients undergoing modified pi-plasty. A retrospective study was performed using a population of 59 patients who underwent modified pi-plasty, wherein patients who received TXA during surgery (study group, n = 26) were compared with patients who did not receive TXA (control group, n = 33). The primary study endpoints were intraoperative red blood cell substitution (%), postoperative red blood cell substitution (%), total red blood cell substitution (%), and hematocrit variation. Perioperative administration of TXA significantly decreased the total red blood cell substitution (62.90% vs 86.70%; p = 0.002) and resulted in a higher postoperative hematocrit (29.47% vs 27.53%; p = 0.060). In conclusion, perioperative administration of TXA significantly decreased blood transfusion requirements in patients with sagittal craniosynostosis undergoing modified pi-plasty., (Copyright © 2024 The Authors. 239. Abrogating Metastatic Properties of Triple-Negative Breast Cancer Cells by EGFR and PI3K Dual Inhibitors.

- Abstract
Triple-negative breast cancer (TNBC) is a devastating BC subtype. Its aggressiveness, allied to the lack of well-defined molecular targets, usually culminates in the appearance of metastases that account for poor prognosis, particularly when they develop in the brain. Nevertheless, TNBC has been associated with epidermal growth factor receptor (EGFR) overexpression, leading to downstream phosphoinositide 3-kinase (PI3K) signaling activation. We aimed to unravel novel drug candidates for TNBC treatment based on EGFR and/or PI3K inhibition. Using a highly metastatic TNBC cell line with brain tropism (MDA-MB-231 Br4) and a library of 27 drug candidates in silico predicted to inhibit EGFR, PI3K, or EGFR plus PI3K, and to cross the blood-brain barrier, we evaluated the effects on cell viability. The half maximal inhibitory concentration (IC
50 ) of the most cytotoxic ones was established, and cell cycle and death, as well as migration and EGFR pathway intervenient, were further evaluated. Two dual inhibitors emerged as the most promising drugs, with the ability to modulate cell cycle, death, migration and proliferation, morphology, and PI3K/AKT cascade players such as myocyte enhancer factor 2C (MEF2C) and forkhead box P1 (FOXP1). This work revealed EGFR/PI3K dual inhibitors as strong candidates to tackle brain metastatic TNBC cells.- Published

240. Usefulness of a clinicopathological classification in predicting treatment-related outcomes and multimodal therapeutic approaches in pituitary adenoma patients: retrospective analysis on a Portuguese cohort of 129 patients from a tertiary pituitary center.

- Abstract
Purpose: A clinicopathological classification has been designed to predict recurrence/progression in patients with pituitary adenomas (PAs). We aimed to study its usefulness in predicting PAs that will have a challenging disease course and may require more often complex multimodal and multiple therapeutic approaches., Methods: Retrospective analysis of 129 patients with PAs operated in our institution between 2001 and 2020 (84 non-clinically functioning PAs, 32 acromegaly, 9 Cushing's disease, 2 prolactinomas and 2 thyrotropinomas). Grading was based on invasion and proliferation: 1a (non-invasive, non-proliferative; n = 59), 1b (non-invasive, proliferative; n = 17), 2a (invasive, non-proliferative; n = 38), and 2b (invasive, proliferative; n = 15)., Results: Of the 129 patients, 68 (52.7%) were females, and the mean age at diagnosis was 53.7 ± 15.4 years. The mean follow-up duration was 93.1 ± 61.8 months. Grade 2b PAs when compared to other grades (2b-2a-1b-1a) had significantly higher rates of persistent tumor remnant within 1-year after operation (93-78-18-30%; p < 0.001), active disease at last follow-up (40-27-12-10%; p = 0.004), re-operation (27-16-0-5%; p = 0.023), irradiation (53-38-12-7%; p < 0.001), multimodal treatment (67-49-18-25%; p = 0.003), multiple treatment (33-27-6-9%; p = 0.017). Patients with grade 2b PAs also required a higher mean number of treatments (2.6-2.1-1.2-1.4; p < 0.001)., Conclusions: This clinicopathological classification appears to be a useful grading system to identify PAs that may be more refractory and more often require complex multimodal and multiple therapeutic approaches. Invasive PAs, especially grade 2b tumors, may be more likely to need complex treatment approach, including radiotherapy, and may display higher rates of active disease at last follow-up, despite receiving higher number of treatments., (© 2023. 241. Highlighted Advances in Therapies for Difficult-To-Treat Brain Tumours Such as Glioblastoma.

- Abstract
Glioblastoma multiforme (GBM) remains a challenging disease, as it is the most common and deadly brain tumour in adults and has no curative solution and an overall short survival time. This incurability and short survival time means that, despite its rarity (average incidence of 3.2 per 100,000 persons), there has been an increased effort to try to treat this disease. Standard of care in newly diagnosed glioblastoma is maximal tumour resection followed by initial concomitant radiotherapy and temozolomide (TMZ) and then further chemotherapy with TMZ. Imaging techniques are key not only to diagnose the extent of the affected tissue but also for surgery planning and even for intraoperative use. Eligible patients may combine TMZ with tumour treating fields (TTF) therapy, which delivers low-intensity and intermediate-frequency electric fields to arrest tumour growth. Nonetheless, the blood-brain barrier (BBB) and systemic side effects are obstacles to successful chemotherapy in GBM; thus, more targeted, custom therapies such as immunotherapy and nanotechnological drug delivery systems have been undergoing research with varying degrees of success. This review proposes an overview of the pathophysiology, possible treatments, and the most (not all) representative examples of the latest advancements.

242. The Influence of Real-Context Scientific Activities on Preservice Elementary Teachers' Thinking and Practice of Nature of Science and Scientific Inquiry.

- Abstract
Understanding how and why science works is a major goal of science education. The aim of this article is to analyze the influence of a research experience in real science contexts, in the thinking and practice of preservice elementary teachers regarding inquiry and nature of science teaching. An in-depth case study which highlights the affordances and shortcomings of the participants' immersion in real science contexts and in seminars and its impact on participants' thoughts and practices of nature of science and inquiry will be presented. Interviews, observations, diaries, and videotaped seminars were used for data collection. Our findings suggest that the research experience, as well as moments of reflection, contributed to enhance the relevance of an inquiry-based teaching and teaching about NOS in the participants' discourse. - Abstract
Glioblastoma (GBM) is the most common and malignant primary brain tumor in adults. The prognosis of patients is very poor, with a median overall survival of ~ 15 months after diagnosis. Cadherin-3 (also known as P-cadherin), a cell-cell adhesion molecule encoded by the CDH3 gene, is deregulated in several cancer types, but its relevance in GBM is unknown. In this study, we investigated the functional roles, the associated molecular signatures, and the prognostic value of CDH3/P-cadherin in this highly malignant brain tumor. CDH3/P-cadherin mRNA and protein levels were evaluated in human glioma samples. Knockdown and overexpression models of P-cadherin in GBM were used to evaluate its functional role in vitro and in vivo. CDH3-associated gene signatures were identified by enrichment analyses and correlations. The impact of CDH3 in the survival of GBM patients was assessed in independent cohorts using both univariable and multivariable models. We found that P-cadherin protein is expressed in a subset of gliomas, with an increased percentage of positive samples in grade IV tumors. Concordantly, CDH3 mRNA levels in glioma samples from The Cancer Genome Atlas (TCGA) database are increased in high-grade gliomas. P-cadherin displays oncogenic functions in multiple knockdown and overexpression GBM cell models by affecting cell viability, cell cycle, cell invasion, migration, and neurosphere formation capacity. Genes that were positively correlated with CDH3 are enriched for oncogenic pathways commonly activated in GBM. In vivo, GBM cells expressing high levels of P-cadherin generate larger subcutaneous tumors and cause shorter survival of mice in an orthotopic intracranial model. Concomitantly, high CDH3 expression is predictive of shorter overall survival of GBM patients in independent cohorts. Together, our results show that CDH3/P-cadherin expression is associated with aggressiveness features of GBM and poor patient prognosis, suggesting that it may be a novel therapeutic target for this deadly brain tumor., (© 2021 The Authors. - Abstract
The microenvironment of pituitary adenomas (PAs) includes a range of non-tumoral cells, such as immune and stromal cells, as well as cell signaling molecules such as cytokines, chemokines and growth factors, which surround pituitary tumor cells and may modulate tumor initiation, progression, invasion, angiogenesis and other tumorigenic processes. The microenvironment of PAs has been actively investigated over the last years, with several immune and stromal cell populations, as well as different cytokines, chemokines and growth factors being recently characterized in PAs. Moreover, key microenvironment-related genes as well as immune-related molecules and pathways have been investigated, with immune check point regulators emerging as promising targets for immunotherapy. Understanding the microenvironment of PAs will contribute to a deeper knowledge of the complex biology of PAs, as well as will provide developments in terms of diagnosis, clinical management and ultimately treatment of patients with aggressive and/or refractory PAs., (© 2022. - Abstract
Purpose: Glioblastoma is the most common primary malignant brain tumor in the adult, whose grim prognosis largely relates to the absence of effective treatment targets. Given its success in other cancers, immunotherapy has been trialed in glioblastoma and failed to demonstrate the expected benefit. Importantly, these disappointing results highlight the importance of understanding the unique and transforming biology of glioblastoma and its microenvironment. Our goal was to evaluate and characterize the expression of PD-L1 through immunohistochemistry in a large glioblastoma cohort. We further studied PD-L1 expression-associated prognosis and its correlation to systemic and neuropathological parameters., Methods: A series of 352 glioblastoma specimens (313 initial resection, 39 matched recurrences) was collected, with a detailed characterization of tumor neuropathological characteristics, including the presence, density and location of tumor infiltrating lymphocytes (TIL). Two hematological markers, absolute lymphocyte count and neutrophil-lymphocyte ratio (NLR), were used to analyze and correlate with systemic inflammation and immunosuppression. Immunohistochemistry was performed to evaluate PD-L1 expression., Results: Membranous PD-L1 expression was identified in 31% (98/313) of newly diagnosed and 46% (18/39) of matched recurrent tumors. TIL were found in 26% (82/313) of primary tumors and both density and location were found to be significantly associated with PD-L1 expression (p < 0.001). Interestingly, PD-L1 expressing tumors had more frequently areas with sarcomatous differentiation (p < 0.001) and were significantly associated with lower lymphocyte count (p = 0.018) and higher NLR ratio (p = 0.004) upon diagnosis. Importantly, PD-L1 expression was an independent poor prognostic marker in our cohort., Conclusion: Taken together, our data points to a putative role for PD-L1 expression in glioblastoma biology, which correlates to poor patient overall survival, as well as with a general systemic inflammatory status and immunosuppression., (© 2021. - Abstract
Cadherins are calcium-binding proteins with a pivotal role in cell adhesion and tissue homeostasis. The cadherin-dependent mechanisms of cell adhesion and migration are exploited by cancer cells, contributing to tumor invasiveness and dissemination. In particular, cadherin switch is a hallmark of epithelial to mesenchymal transition, a complex development process vastly described in the progression of most epithelial cancers. This is characterized by drastic changes in cell polarity, adhesion, and motility, which lead from an E-cadherin positive differentiated epithelial state into a dedifferentiated mesenchymal-like state, prone to metastization and defined by N-cadherin expression. Although vastly explored in epithelial cancers, how these mechanisms contribute to the pathogenesis of other non-epithelial tumor types is poorly understood. Herein, the current knowledge on cadherin expression in normal development in parallel to tumor pathogenesis is reviewed, focusing on epithelial to mesenchymal transition. Emphasis is taken in the unascertained cadherin expression in CNS tumors, particularly in gliomas, where the potential contribution of an epithelial-to-mesenchymal-like process to glioma genesis and how this may be associated with changes in cadherin expression is discussed.

- Abstract
The use of computational tools to identify biological targets of natural products with anticancer properties and unknown modes of action is gaining momentum. We employed self-organizing maps to deconvolute the phenotypic effects of piperlongumine (PL) and establish a link to modulation of the human transient receptor potential vanilloid 2 (hTRPV2) channel. The structure of the PL-bound full-length rat TRPV2 channel was determined by cryo-EM. PL binds to a transient allosteric pocket responsible for a new mode of anticancer activity against glioblastoma (GBM) in which hTRPV2 is overexpressed. Calcium imaging experiments revealed the importance of Arg539 and Thr522 residues on the antagonistic effect of PL and calcium influx modulation of the TRPV2 channel. Downregulation of hTRPV2 reduces sensitivity to PL and decreases ROS production. Analysis of GBM patient samples associates hTRPV2 overexpression with tumor grade, disease progression, and poor prognosis. Extensive tumor abrogation and long term survival was achieved in two murine models of orthotopic GBM by formulating PL in an implantable scaffold/hydrogel for sustained local therapy. Furthermore, in primary tumor samples derived from GBM patients, we observed a selective reduction of malignant cells in response to PL ex vivo . - Abstract
Aims: Though formalin remains to be the gold standard fixative in pathology departments, analytical challenges persist for nucleic acid evaluations. In our laboratory, formalin fixation of skin samples in particular impairs diagnostic accuracy and demands repetition of biopsies and analytical procedures. PAXgene Tissue Systems may be an alternative; however, according to manufacturer specifications it only allows fixation for 48 hours before having to add a stabiliser. This may be a challenge in laboratories, which are closed in weekends and bank holidays. Our aim was to validate this alternative fixative for dermatological samples with prolonged fixation times using standard laboratory protocols developed for formalin-fixed specimens. We compared the results with gold standard formalin fixation., Methods: Skin specimens were formalin or PAXgene fixed for either 2 hours, 24 hours, 3 days or 7 days, paraffin-embedded, analysed and scored by observers., Results: Generally, formalin outperformed PAXgene fixation in H&E stains and fluorescence in situ hybridisation (FISH), but both seem usable for diagnostics. Time of PAXgene fixation did not have an impact on alcian blue-Van Gieson (ABVG), H&E (p=0.48), nor immunohistochemistry (p=0.74). There was a tendency towards best PAXgene performance at 24 hours of fixation for FISH, and for DNA integrity analysis 24 hours or 3 days., Conclusions: Prolonging PAXgene fixation time to 3 days before adding stabiliser does not seem to have major impact on performance of general diagnostic analysis, but our preliminary results show optimisation of internal protocols are needed.
