BackgroundThe antioxidant and anti-inflammatory features of Formononetin, an isoflavone constituent extracted from traditional Chinese medicine, have been reported. The present study investigated that whether Formononetin plays a benefit on hyperoxic ALI.MethodsC57BL/6 mice were exposed to hyperoxia for 72 h to produce experimental hyperoxic ALI model. Formononetin or vehicle was administrated intraperitoneally. Samples from the lung were collected at 72 h post hyperoxia exposure for further study. Pulmonary microvascular endothelial cells isolated from the lung of C57BL/6 mice were used for in vitro study.ResultsFormononetin pretreatment notably attenuated hyperoxia-induced elevating pulmonary water content, upregulation of proinflammatory cytokine levels and increasing infiltration of neutrophil in the lung. Western blot analyses showed that Formononetin enhanced the expression of nuclear factor erythroid-2-related factor 2 (Nrf2) which is a key transcription factor regulating the expression of heme oxygenase-1 (HO-1). Formononetin increased HO-1 expression and activity compared with vehicle-treated animals. Moreover, Formononetin reversed hyperoxia-caused the reduction of M2 macrophage polarization. However, pretreatment of a HO-1 inhibitor reduced the protective effect of Formononetin on hyperoxic ALI. Cell study showed that the Formononetin-induced upregulation of HO-1 was abolished when the Nrf2 was silenced.ConclusionsFormononetin pretreatment reduces hyperoxia-induced ALI via Nrf2/HO-1-mediated antioxidant and anti-inflammatory effects.

Combined vacuum and surcharge preloading has gradually been widely used because of its advantages of low cost, green environmental protection, and good treatment effect. The conventional prefabricated vertical drain presents obvious defects in vacuum preloading treatment, such as obvious silting, serious bending of the drainage board, large attenuation of vacuum degree of drainage board along the depth, long construction period, and so on, which affect the final reinforcement effect. In this paper, the MIDAS finite element simulation of combined vacuum and surcharge preloading of prefabricated vertical drains (PVDs) and prefabricated horizontal drains (PHDs) is established through the comparative experiment of the engineering field. The comparative experimental study is carried out from the aspects of the vertical settlement, horizontal displacement, and pore water pressure. The results show that under combined vacuum and surcharge preloading, the consolidation effect of soft soil with PHDs is better than that with PVDs. When PHDs are used, the vertical settlement increases by 7.2% compared with PVDs; the horizontal displacement is larger; and the pore water pressure dissipates faster. This is because when the PHDs are adopted, the consolidation direction of the soil is consistent with the direction of the vacuum suction, which is mainly caused by vertical settlement. With the consolidation, the spacing between PHDs is gradually shortened, and the drainage distance is reduced, which can effectively reduce the consolidation time and improve the reinforcement effect of the soil. In addition, the PHDs can move downward uniformly with the soil during the consolidation process and have almost no bending deformation, which makes the vacuum transfer more uniform and effective.

Chlorella meal as feed ingredient in diets has been well evaluated in fish and shellfish, whereas limited information is available about Chlorella extract as feed supplement and their effects on growth performance and stress resistance of fish. This study was conducted to assess effect of Chlorella water extract (CWE) on growth performance, body composition, digestive enzyme activity, antioxidant and immune capacity, histomorphology, and sodium nitrite stress resistance of Oreochromis niloticus. In total of 1920 O. niloticus fingerlings with initial body weight of 0.9 ± 0.01 g were randomly distributed into six groups with four replicates (80 fish per replicate) and fed six diets containing 0 (C0), 2.7 (C2.7), 5.5 (C5.5), 11 (C11), 22 (C22), and 33 (C33) ml/kg dry matter of CWE for 56 d. Fish were suffered with a 24-h sodium nitrite stress test at the end of feeding trial. Results indicated that dietary CWE did not affect growth performance, body composition and histological appearance of fish, but quadratically increased (P < 0.05) protein efficiency ratio and trypsin activity as dietary CWE increasing. Inclusion of CWE in diets increased (P < 0.05) hepatic glutathione peroxidase and lysozyme activities. Fish fed CWE-containing diets had lower (P < 0.05) cumulative mortality rate but higher (P < 0.05) acid phosphatase and lysozyme activities as observed in the sodium nitrite stress test. In conclusion, dietary CWE did not affect growth performance, body composition and histological appearance, but improved protein utilization, trypsin activity, antioxidant and immune capacity, and sodium nitrite stress resistance of O. niloticus.

The tea aphid, Aphis aurantii, has become one of the destructive pests in tea plantations in the tropics and subtropics. Very few functional studies have so far focused on the developmental and reproductive biology at a molecular level, because of the lack of comprehensive genetic information. Full-length transcriptomes represent a very highly efficient approach to obtain reference gene sequences in non-model insects. In the present study, the transcriptome of A. aurantii was comprehensively sequenced using PacBio Iso-Seq technology. A total of 46.8 Gb nucleotides and 15,938 non-redundant full-length transcripts were obtained, 13,498 (84.69%) of which were annotated into seven databases. Of these transcripts, 2,029 alternative splicing events and 15,223 simple sequence repeats were detected. Among these transcripts, 4,571 (28.68%) and 11,367 (71.32%) were long non-coding RNAs (lncRNAs) and protein-coding genes, respectively. Five hundred and ninety transcription factors were detected. The first full-length transcriptome represents a significant increase in the known genetic information of A. aurantii. It will assist the future functional study of genes involved in its development and reproduction.

Background: Real-time reverse transcription-PCR (rRT-PCR) has been the most effective and widely implemented diagnostic technology since the beginning of the COVID-19 pandemic. However, fuzzy rRT-PCR readouts with high Ct values are frequently encountered, resulting in uncertainty in diagnosis. Methods: A Specific Enhancer for PCR-amplified Nucleic Acid (SENA) was developed based on the Cas12a trans-cleavage activity, which is specifically triggered by the rRT-PCR amplicons of the SARS-CoV-2 Orf1ab (O) and N fragments. SENA was first characterized to determine its sensitivity and specificity, using a systematic titration experiment with pure SARS-CoV-2 RNA standards, and was then verified in several hospitals, employing a couple of commercial rRT-PCR kits and testing various clinical specimens under different scenarios. Findings: The ratio (10 min/5 min) of fluorescence change (FC) with mixed SENA reaction (mix-FCratio) was defined for quantitative analysis of target O and N genes, and the Limit of Detection (LoD) of mix-FCratio with 95% confidence interval was 1.2≤1.6≤2.1. Totally, 295 clinical specimens were analyzed, among which 21 uncertain rRT-PCR cases as well as 4 false negative and 2 false positive samples were characterized by SENA and further verified by next-generation sequencing (NGS). The cut-off values for mix-FCratio were determined as 1.145 for positive and 1.068 for negative. Interpretation: SENA increases both the sensitivity and the specificity of rRT-PCR, solving the uncertainty problem in COVID-19 diagnosis and thus providing a simple and low-cost companion diagnosis for combating the pandemic. Fucoxanthin (Fx), a kind of primary carotenoids in brown seaweeds and diatoms, has attractive efficacy in human’s healthcare including loss weight, the prevention of diabetes and Alzheimer’s disease. Marine diatom Phaeodactylum tricornutum is now realized as a promising producer for commercial Fx production due to its higher content of Fx than brown seaweeds with easily artificial cultivation and Fx extraction. In the present study, to improve Fx production in P. tricornutum, the mixotrophic cultures were applied to optimize initial cell density, light intensity, light regime and nitrogen supplementation. The results showed that the higher initial cell density (1 × 107 cells mL–1) and lower light intensity (20 μmol m–2 s–1) were favorable for biomass production and Fx accumulation. The maximal Fx content [16.28 mg g–1 dry weight (DW)] could be achieved under blue light (BL), but the highest biomass concentration (5.53 g L–1) could be attained under red: blue light (R: B, 6:1) in the batch culture. A novel two-phase culture approach was developed to increase the biomass concentration to the highest value (6.52 g L–1) with the maximal productivity of Fx (8.22 mg L–1 d–1) through light shift from R:B ratio (6:1) in phase 1 to R:B ratio (5:1) by enhancing BL and tryptone addition in phase 2. The content and intracellular amount of Fx were also increased 8% and 12% in phase 2 compared to phase 1. The expression levels analysis revealed that genes encoding phytoene synthase (PSY), zeaxanthin epoxidase (ZEP), and fucoxanthin-chlorophyll-protein b (FCPb) were upregulated significantly, with downregulation of the gene encoding violaxanthin de-epoxidase (VDE), leading to the improvement of Fx in phase 2. The present study demonstrated the two-phase culture strategy could promote Fx productivity through enhancing biomass production and increasing Fx content, indicating that strengthening BL coupled with adding tryptone were effective to facilitate Fx production by mixotrophic cultivation of marine diatom P. tricornutum.

Carbon fiber/ceramic composite demonstrates excellent electro-thermal performance that can be applied in various heating applications. However high temperature ceramization inevitably causes oxidation of carbon fiber and negatively influences its electro-thermal property. Herein, a graphene coated carbon fiber/ceramic composite via one-step firing with rapid and stable temperature responsiveness is proposed. Crystal structure, morphological feature, electrical property and electrothermal behavior of the modified carbon fibers and carbon fiber/ceramic composite were investigated. The results showed effectively protective effect of coating and sintering of carbon fiber under N2 environment, resulting in greatly improved electro-thermal and mechanical properties. The composite based on 24 k carbon fibers and 1 cm thickness ceramic sintering reached about 65 °C at 6 V in only 10 min and the cooling process lasted 30 min from 82 °C to room temperature, presenting highly responsive heating and good temperature storage performance. The tensile strength and elongation at break of the single graphene coated carbon fiber increased 12% and 200%. The great electro-thermal features and strong mechanical properties of the modified carbon fibers make the carbon fiber/ceramic composite a good candidate for in-door temperature control and out-door deicing applications.

Summary: The failure of perovskite solar cells (PSCs) under ultraviolet (UV) irradiation is a serious barrier of commercial utilization. Here, a two-stage degradation process of TiO2-based PSCs is discovered under continuous UV irradiation in an inert atmosphere. In the first decay stage, oxygen vacancy-Ti3+ (Ti3+-VO) transform into active Ti4+-VO trap states under UV excitation and cause photocarrier loss. Furthermore, Ti4+-VO states can convert back into Ti3+-VO states through oxidizing I−, which result in the accumulation of I3−. Sequentially, the rapid decomposition of perovskite accelerated by increasing I3− replaces the photocarrier loss as the dominant mechanism leading to the second decay stage. Then, a universal method is proposed to improve the UV stability by blocking the transformation of Ti3+-VO states, which can be realized by polyethyleneimine ethoxylated (PEIE) modified layer. Yersinia pestis, Brucella spp., and Bacillus anthracis are pathogens that can cause infectious zoonotic diseases with high mortality rates. An upconverting phosphor-based quantitative immunochromatographic (UPT-LF) assay, a point-of-care testing method suitable for resource-limited areas, was calibrated to quantitatively detect pathogenic bacteria. The bacterial purity or activity were ensured via staining methods and growth curves, respectively. Growth assays showed that the classic plate-counting method underestimated bacterial numbers compared with the bacterial counting method recommended by the reference material of the National Institutes for Food and Drug Control, China. The detection results of the UPT-LF assay differed significantly between the bacterial cultures in liquid and solid media and between different strains. Accelerated stability assessments and freeze-thaw experiments showed that the stability of the corresponding antigens played an important role in calibrating the UPT-LF assay. In this study, a new calibration system was developed for quantitative immunochromatography for detecting pathogenic bacteria. The results demonstrated the necessity of calibration for standardizing point-of-care testing methods.

Summary: The Suzuki-Miyaura coupling is a fundamentally important transformation in modern organic synthesis. The development of new reaction modes for new chemical accessibility and higher synthetic efficiency is still the consistent pursuance in this field. An efficient Suzuki-Miyaura coupling enabled by a controllable 1,4-palladium migration was realized to afford stereodefined multisubstituted olefins and 1,3-dienes. The reaction exhibits remarkable broad substrate scope, excellent functional-group tolerance, versatile conversion with obtained products, and easy scalability. This paper considers the product service scheduling problem with service matching. This problem is a complex scheduling problem that integrates multiple traveling salesman problem (MTSP) and multiple service types. To solve the problem, a tabu search method was presented. With the objective of minimizing the total of engineers’ travel distances, total of the customer penalty values, and makespan, an optimization model of this problem is established. The historical search solutions are taken as tabu object, and limiting the quantity of search neighborhood solution is taken as the aspiration level. At last, the adaptability, validity, and stability of the model are verified by an example.

Mycotoxins are highly toxic metabolites produced by fungi that pose a huge threat to human and animal health. Contamination of food and feed with mycotoxins is a worldwide issue, which leads to huge financial losses, annually. Decades of research have developed various approaches to degrade mycotoxins, among which the biological methods have been proved to have great potential and advantages. This review provides an overview on the important advances in the biological removal of mycotoxins over the last decade. Here, we provided further insight into the chemical structures and the toxicity of the main mycotoxins. The innovative strategies including mycotoxin degradation by novel probiotics are summarized in an in-depth discussion on potentialities and limitations. We prospected the promising future for the development of multifunctional approaches using recombinant enzymes and microbial consortia for the simultaneous removal of multiple mycotoxins.

Fe-based metallic glass particle has an excellent application prospect in micro-forming field due to their special properties. Mono-sized Fe60Ni7.5Mo7.5P10C10B5 metallic glass particles with controllable particle diameters were prepared by pulsated orifice ejection method (POEM). The analysis of these obtained particles was carried out by optical microscope(OM), scanning electron microscope(SEM), X-ray diffraction (XRD), differential scanning calorimetry (DSC). The results show that the metallic glass particles prepared by POEM have the advantages of uniform particle diameter, high sphericity and consistent thermal history. The microstructure of particles is transformed from fully amorphous phase to crystal phase with increasing particle diameters. In the experimental conditions of this paper, the critical diameter for obtaining fully amorphous phase should be less than 285μm in Ar atmosphere. However, the particle with the maximum diameter of 383μm in He atmosphere also maintain amorphous phase. The critical cooling rate calculated by the theoretical model should be over 1300K/s.

Abstract Seasonal H3N2 influenza viruses are recognized as major epidemic viruses, exhibiting complex seasonal patterns in regions with temperate climates. To investigate the influence of viral evolution and mutations on the seasonality of influenza, we performed a genome-wide analysis of samples collected from 62 influenza A/H3N2-infected patients in Shanghai during 2016–2017. Phylogenetic analysis of all eight segments of the influenza A virus revealed that there were two epidemic influenza virus strains circulating in the 2016–2017 winter season (2016–2017win) and 2017 summer season (2017sum). Replication of the two epidemic viral strains at different temperatures (33, 35, 37, and 39 °C) was measured, and the correlation of the mutations in the two epidemic viral strains with temperature sensitivity and viral replication was analyzed. Analysis of the replication kinetics showed that replication of the 2016–2017win strains was significantly restricted at 39 °C compared with that of the 2017sum strains. A polymerase activity assay and mutational analysis demonstrated that the PA I668V mutation of the 2016–2017win viruses suppressed polymerase activity in vitro at high temperatures. Taken together, these data suggest that the I668V mutation in the PA subunit of the 2016–2017win strains may confer temperature sensitivity and attenuate viral replication and polymerase activity; meanwhile, the 2017sum strains maintained virulence at high temperatures. These findings highlight the importance of certain mutations in viral adaptation and persistence in subsequent seasons.

ObjectiveThe aim of this study was to further clarify clinical characteristics and predict mortality risk among patients with viral pneumonia.MethodsA total of 528 patients with viral pneumonia at RuiJin hospital in Shanghai from May 2015 to May 2019 were recruited. Multiplex real-time RT-PCR was used to detect respiratory viruses. Demographic information, comorbidities, routine laboratory examinations, immunological indexes, etiological detections, radiological images and treatment were collected on admission.Results76 (14.4%) patients died within 90 days in hospital. A predictive MuLBSTA score was calculated on the basis of a multivariate logistic regression model in order to predict mortality with a weighted score that included multilobular infiltrates (OR = 5.20, 95% CI 1.41–12.52, p = 0.010; 5 points), lymphocyte ≤ 0.8∗109/L (OR = 4.53, 95% CI 2.55–8.05, p < 0.001; 4 points), bacterial coinfection (OR = 3.71, 95% CI 2.11–6.51, p < 0.001; 4 points), acute-smoker (OR = 3.19, 95% CI 1.34–6.26, p = 0.001; 3 points), quit-smoker (OR = 2.18, 95% CI 0.99–4.82, p = 0.054; 2 points), hypertension (OR = 2.39, 95% CI 1.55–4.26, p = 0.003; 2 points) and age ≥60 years (OR = 2.14, 95% CI 1.04–4.39, p = 0.038; 2 points). 12 points was used as a cut-off value for mortality risk stratification. This model showed sensitivity of 0.776, specificity of 0.778 and a better predictive ability than CURB-65 (AUROC = 0.773 vs. 0.717, p < 0.001).ConclusionHere, we designed an easy-to-use clinically predictive tool for assessing 90-day mortality risk of viral pneumonia. It can accurately stratify hospitalized patients with viral pneumonia into relevant risk categories and could provide guidance to make further clinical decisions.

Abstract Background The accurate knowledge of the refractive index of air is very important for environmental compensation in length measurements. Methods In this study, we apply difference approximation methods to facilitate the calculation of the group refractive index of air (GRA) over all calculable wavelengths. Our approach involves the determination of a suitable combination of the step size Δλ and numbers of significant digits of calculations of the phase refractive index of air (PRA) by balancing the two main errors (round-off and truncation errors) over the entire calculable wavelength range. Results Based on our calculations, we find that the GRA computation over the range of all calculable wavelengths (301.5-1698.5 nm) can be easily approximated by the two-point central difference method with Δλ = 1.5 nm and 12-digit PRA accuracy. Conclusion The approximation accuracy is less than 5 × 10-9. Our approach can be used by non-expert users to obtain the GRA with sufficient accuracy.

Abstract Although perovskite solar cells with power conversion efficiencies (PCEs) more than 22% have been realized with expensive organic charge-transporting materials, their stability and high cost remain to be addressed. In this work, the perovskite configuration of MAPbX (MA = CH3NH3, X = I3, Br3, or I2Br) integrated with stable and low-cost Cu:NiO x hole-transporting material, ZnO electron-transporting material, and Al counter electrode was modeled as a planar PSC and studied theoretically. A solar cell simulation program (wxAMPS), which served as an update of the popular solar cell simulation tool (AMPS: Analysis of Microelectronic and Photonic Structures), was used. The study yielded a detailed understanding of the role of each component in the solar cell and its effect on the photovoltaic parameters as a whole. The bandgap of active materials and operating temperature of the modeled solar cell were shown to influence the solar cell performance in a significant way. Further, the simulation results reveal a strong dependence of photovoltaic parameters on the thickness and defect density of the light-absorbing layers. Under moderate simulation conditions, the MAPbBr3 and MAPbI2Br cells recorded the highest PCEs of 20.58 and 19.08%, respectively, while MAPbI3 cell gave a value of 16.14%.

Summary: A major obstacle for successful management of patients with colorectal carcinoma (CRC) is resistance to anti-cancer cytotoxic treatments. Here, we identified a mechanism of multidrug resistance in wild-type Kras CRCs based on the survival of a cell subpopulation characterized by Sirt5 expression. Sirt5+ cells in wild-type Kras CRCs are resistant to either chemotherapeutic agents or cetuximab and serve as a reservoir for recurrence. Sirt5 demalonylates and inactivates succinate dehydrogenase complex subunit A (SDHA), leading to an accumulation of the oncometabolite succinate. Succinate binds to and activates a reactive oxygen species-scavenging enzyme, thioredoxin reductase 2 (TrxR2), to confer chemotherapy resistance. In contrast, Sirt5+ cells exhibit an elevated succinate-to-aKG ratio that inhibits aKG-dependent dioxygenases to maintain cetuximab resistance. Our findings suggest that Sirt5 inhibitors in combination with chemotherapeutic agents and/or cetuximab may represent a therapeutic strategy for CRC patients harboring wild-type Kras. : Du et al. show that a Sirt5+ subpopulation represents a critical driving force behind the development of multidrug resistance in wild-type Kras colorectal carcinomas and that Sirt5-mediated SDHA inactivation has implications for this process. A novel technology called pulsated orifice ejection method(POEM) and used for preparing mono-sized and high-precision spherical micro particles was introduced in this article. The working principle of the technique was illustrated and it was in two modes:low-melting point diaphragm mode and high-melting point rod mode, depending on the different melting points of materials. The particles prepared by POEM have the advantages of mono-sized, uniform and controllable particle size, high sphericity, and consistent thermal history. By introducing the application of particles prepared by this method, showing the huge application prospects of this technology in electronic packaging, bioengineering, micro-fabrication, rapid solidification analysis of metal droplets, additive manufacturing and so on.With the development of POEM, this technology is predicted to have wider prospects due to its unique characteristics.

Abstract Phormicins belong to defensin family, which are important antimicrobial peptides (AMPs) in insects. These AMPs are inducible upon challenging by immune triggers. In the present study, we identified the cDNA of a phormicin gene (BdPho) in the oriental fruit fly, Bactrocera dorsalis (Hendel), a ruinous agricultural pest causing great economic losses to fruits and vegetables. The cDNA of BdPho contains a 282 bp open reading frame encoding 93 amino acid residues, and the predicted molecular weight and isoelectric point of BdPho peptide were 9.83 kDa and 7.54, respectively. Quantitative real-time PCR analyses showed that the transcription level of BdPho was the highest in adult during different developmental stages and was the highest in abdomen among adult tagmata. Moreover, BdPho was highly expressed in fat body among different tissues, both in female and male adult. The mRNA level of BdPho was significantly up-regulated to 7.46- and 14.53-fold at 3 and 6 h after the insects were challenged with peptidoglycans from Escherichia coli (PGN-EB), respectively, suggesting its antimicrobial activity against Gram-negative microorganisms. Furthermore, the expression level of BdPho was significantly up-regulated to 3.83-fold after mating, suggesting that female adults might enhance their immunity by up-regulating the expression level of BdPho during mating. These results firstly describe the basic properties of the phormicin gene from B. dorsalis, and lay the foundation for investigating functional properties of AMPs and exploring the molecular mechanisms in the immune system.

The shearer positioning method with an inertial measurement unit and the odometer is feasible in the longwall coal-mining process. However, the positioning accuracy will continue to decrease, especially for the micro-electromechanical inertial measurement unit (MIMU). In order to further improve the positioning accuracy of the shearer without adding other external sensors, the positioning method of the Rauch-Tung-Striebel (RTS) smoother-aided MIMU and odometer is proposed. A Kalman filter (KF) with the velocity and position measurements, which are provided by the odometer and closing path optimal estimation model (CPOEM), respectively, is established. The observability analysis is discussed to study the possible conditions under which the error states of KF can be estimated. A RTS smoother with the above-mentioned KF as the forward filter is built. Finally, the experiments of simulating the movement of the shearer through a mobile carrier were carried out, with a longitudinal movement distance of 44.6 m and a lateral advance distance of 1.2 m. The results show that the proposed method can effectively improve the positioning accuracy. In addition, the odometer scale factor and mounting angles can be estimated in real time.

Based on the importance of building energy saving, a college building in Chengdu is selected to analyze the effect of material type and thickness of thermal insulation layer of the exterior wall on building energy consumption by using DeST energy simulation software, and through the life cycle cost analysis model to study the economic thickness value and unit cost of different insulation materials. The results show that the effect of adding insulation materials on the building envelope has less effect on the cumulative annual cooling load, but the impact of cumulative annual heat load is greater, compared with no insulation material, it can save 21.52% heat load, 3.78% total load, and 25.34% total cost per unit area. Meanwhile, this change trend between the insulation thickness and total cost shows an anti-parabolic relationship, in which there is an optimal economic thickness, but different insulation materials also have different economic thickness. Considering the cost, polystyrene is preferred as insulation material in several common insulation materials with an economic thickness of 35 mm and a total cost of 76.28 RMB/m2. Based on this, the data and conclusions presented in this paper can provide some reference for building energy saving and existing building renovation in Chengdu. Recurrent tumors originate from cancer stem cells (CSCs) that survive conventional treatments. CSCs consist of heterogeneous subpopulations that display distinct sensitivity to anticancer drugs. Such a heterogeneity presents a significant challenge in preventing tumor recurrence. In the current study, we observed that quiescent CUB-domain-containing protein 1 (CDCP1)+ CSCs are enriched after chemotherapy in mutant Kirsten rat sarcoma viral oncogene homolog (Kras) colorectal carcinomas (CRCs) and serve as a reservoir for recurrence. Mechanistically, glucose catabolism in CDCP1+ CSCs is routed to the oxidative pentose phosphate pathway (PPP); multiple cycling of carbon backbones in the oxidative PPP potentially maximizes NADPH reduction to counteract chemotherapy-induced reactive oxygen species (ROS) formation, thereby allowing CDCP1+ CSCs to survive chemotherapeutic attack. This is dependent on silent mating type information regulation 2 homolog 5 (Sirt5)-mediated inhibition of the glycolytic enzyme triosephosphate isomerase (TPI) through demalonylation of Lys56. Blocking demalonylation of TPI at Lys56 increases chemosensitivity of CDCP1+ CSCSs and delays recurrence of mutant Kras CRCs in vivo. These findings pinpoint a new therapeutic approach for combating mutant Kras CRCs.

Abstract A novel ferroelectric coupling photovoltaic effect is reported to enhance the open‐circuit voltage (VOC) and the efficiency of CH3NH3PbI3 perovskite solar cells. A theoretical analysis demonstrates that this ferroelectric coupling effect can effectively promote charge extraction as well as suppress combination loss for an increased minority carrier lifetime. In this study, a ferroelectric polymer P(VDF‐TrFE) is introduced to the absorber layer in solar cells with a proper cocrystalline process. Piezoresponse force microscopy (PFM) is used to confirm that the P(VDF‐TrFE):CH3NH3PbI3 mixed thin films possess ferroelectricity, while the pure CH3NH3PbI3 films have no obvious PFM response. Additionally, with the applied external bias voltages on the ferroelectric films, the devices begin to show tunable photovoltaic performance, as expected for the polarization in the poling process. Furthermore, it is shown that through the ferroelectric coupled effect, the efficiency of the CH3NH3PbI3‐based perovskite photovoltaic devices is enhanced by about 30%, from 13.4% to 17.3%. And the open‐circuit voltages (VOC) reach 1.17 from 1.08 V, which is reported to be among the highest VOCs for CH3NH3PbI3‐based devices. It should be noted in particular that the thickness of the layer is less than 160 nm, which can be regarded as semi‐transparent.

Nanofocusing of incident light in the visible and infrared regions is achieved using tapered metallic nanostructures (TMNSs). Illumination under 633 nm excitation produces a very small (∼40 nm in the x-direction), very bright spot with a specific geometry, demonstrating near-field nanofocusing of the incident beam to the deep subwavelength scale. The key processes, including a theoretical understanding, numerical calculation, and a near-field optical measurement involving the metallic planar nanotip arrays, are discussed. As in the lightning-rod effect, there is a large number of surface states to accommodate free electrons, resulting in a very high surface density distribution of the free electrons over the nanoapexes. A theoretical model for calculating the free-electron distribution based on the surface energy state of the nanoapex is established. The spread of free electron oscillation, including the guiding of free electrons towards nearby planar nanoapexes to obtain near-field nanofocusing is examined. As the lightning-rod effect is a broadband phenomenon, the TMNSs are also examined in the far-infrared (far-IR) region, at the typical far-IR wavelength of 10.274 μm. It should be noted that the IR radiation can also be focused into a linear light-spot of ∼20 nm, far beyond the IR diffraction limit. A metasurface constructed by the orderly arrangement of TMNSs highlights their potential in applications such as surface enhanced Raman spectroscopy, ray absorbing materials, and low-cost nanolithography.

Objective: To explore the recovery pattern of the high-frequency acceleration vestibulo-ocular reflex (VOR) function in unilateral vestibular neuritis (UVN).Methods: Forty-seven consecutive patients with UVN were recruited within 10 days of symptom onset for this study. The high-frequency acceleration horizontal VOR function was assessed using the video head impulse test (vHIT). Patients returned for follow-up evaluation at ~6 months after the onset of symptoms. According to the dizziness handicap inventory questionnaire (DHI), the patients were classified into the normal to mild dizziness group (DHI score ≤30) and moderate to severe dizziness group (DHI score >30) at the follow-up. All the obtained horizontal vHIT gains and corrective saccades parameters were analyzed.Results: vHIT results showed a significantly horizontal VOR gain recovery in UVN patients at the follow-up on the lesion side (p < 0.01). A significantly reduction in the occurrence of corrective saccades (overt and covert) and velocity of corrective saccades (overt and covert) were observed at the follow-up (p < 0.05). At the follow-up, the normal to mild dizziness group (DHI score ≤30) had a significantly higher normal rate of VOR gain, the mean vHIT gains and occurrence of isolated covert saccades (P < 0.05). Furthermore, the occurrence of mixed saccades and the mean velocity of covert saccades were significantly lower in normal to mild dizziness group (P < 0.05).Conclusion: Apart from the recovery of the VOR gain, recovery pattern of corrective saccades can play a key role in vestibular compensate.

The shape of a free surface is an important factor that determines the effect of bench blasting. The structural dynamics theory was applied to establish a structural failure model of the layered rock considering the impact of a blasting gas intrusion. Combined with the continuous-discontinuous element method (CDEM), the influence of rock strata on the failure mechanism of back-break was analyzed. The results show that structural failure characteristics of stratum with different dip angles are different. The bending failure characteristics of dipping-in-face stratum are stronger than that in dipping-out-of-face stratum. With the increase of the dip angle and height of rock stratum, the bending failure length of dipping-in-face stratum increases and the maximum value reaches 5.24 m. The trend of failure along the stratum surface towards the bottom increases, which is an important reason for the formation of an unfavorable shape of free surface. However, the failure depth of the gently dipping stratum and dipping-out-of-face stratum is relatively uniform; the average value is about 0.5 m. Finally, combined with the results of the bench blasting field test of the Changjiu (Shenshan) limestone mine, which is the largest in the production of sand and gravel aggregates, we verify the correctness of the theoretical analysis results. Relevant research results can provide a theoretical basis and technical support for controlling the bench blasting effect.

The ATP-binding cassette (ABC) transporter is a protein superfamily that transports specific substrate molecules across lipid membranes in all living species. In insects, ABC transporter is one of the major transmembrane protein families involved in the development of xenobiotic resistance. Here, we report 49 ABC transporter genes divided into eight subfamilies (ABCA-ABCH), including seven ABCAs, seven ABCBs, 10 ABCCs, two ABCDs, one ABCE, three ABCFs, 16 ABCGs, and three ABCHs according to phylogenetic analysis in Zeugodacus cucurbitae, a highly destructive insect pest of cucurbitaceous and other related crops. The expressions level of 49 ABC transporters throughout various developmental stages and within different tissues were evaluated by quantitative transcriptomic analysis, and their expressions in response to three different insecticides were evaluated by quantitative real-time polymerase chain reaction (qRT-PCR). These ABC transporter genes were widely expressed at developmental stages but most highly expressed in tissues of the midgut, fat body and Malpighian tube. When challenged by exposure to three insecticides, abamectin, β-cypermethrin, and dinotefuran, the expressions of ZcABCB7 and ZcABCC2 were significantly up-regulated. ZcABCB1, ZcABCB6, ZcABCB7, ZcABCC2, ZcABCC3, ZcABCC4, ZcABCC5, and ZcABCC7 were significantly up-regulated in the fat body at 24 h after β-cypermethrin exposure. These data suggest that ZcABCB7 and ZcABCC2 might play key roles in xenobiotic metabolism in Z. cucurbitae. Collectively, these data provide a foundation for further analysis of ABCs in Z. cucurbitae.
