Let $C$ be a smooth curve embedded in a smooth quasi-projective threefold $Y$, and let $Q^n_C=\textrm{Quot}_n(\mathscr I_C)$ be the Quot scheme of length $n$ quotients of its ideal sheaf. We show the identity $\tilde\chi(Q^n_C)=(-1)^n\chi(Q^n_C)$, where $\tilde\chi$ is the Behrend weighted Euler characteristic. When $Y$ is a projective Calabi-Yau threefold, this shows that the DT contribution of a smooth rigid curve is the signed Euler characteristic of the moduli space. This can be rephrased as a DT/PT wall-crossing type formula, which can be formulated for arbitrary smooth curves. Abstract

Background: Health care workers (HCWs) during the COVID-19 pandemic report high levels of psychological distress. We examined whether concerns regarding transmission of COVID-19 to loved ones and social distancing from loved ones were associated with HCWs' distress. We tested whether living with others modified these associations. Method: HCWs at a New York City academic medical center (N = 767; 80.7% female, 58.5% White) enrolled in the COVID-19 Health Care Provider Study and completed a web-based survey between April 9, 2020 and May 11, 2020. Results: Controlling for demographics, distress regarding potential transmission to loved ones and social distancing from loved ones were each significantly associated with higher odds of a positive screen for acute stress, depression, and anxiety (ORs = 1.29-1.59, all ps < .01). Living with others was associated with lower odds of a positive screen for depression and anxiety, though the protective effect for anxiety was evident only for HCWs with no distress regarding transmission concerns. Conclusions: Transmission concerns and social distancing from loved ones were associated with greater odds of psychological distress, whereas living with others was associated with lower odds of distress. Interventions should consider ways to facilitate the ability of HCWs to receive social support from loved ones, while simultaneously protecting their family's health. 215. Retrospective Comparison of Targeted Anticancer Drugs Predicted by the CNS-TAP Tool Versus Those Selected by a Molecularly Driven Tumor Board in Children With DIPG

Abstract

The recent trial Pediatric Neuro-Oncology Consortium 003 (PNOC003) utilized a molecular tumor board to recommend personalized treatment regimens based on tumor sequencing results in children with DIPG. We separately developed the Central Nervous System Targeted Agent Prediction (CNS-TAP) tool, which numerically scores targeted anticancer agents using preclinical, clinical, and patient-specific data. We hypothesized that highly scored agents from CNS-TAP would overlap with the PNOC003 tumor board’s recommendations. For each of the 28 participants, actionable genetic alterations were derived from PNOC003 genomic reports and input to CNS-TAP to identify the highest scoring agents. These agents were then compared with PNOC003 recommendations, with a resultant concordance percentage calculated. Overall, 38% of the total agents recommended by the tumor board were also selected by CNS-TAP, with higher concordance (63%) in a subanalysis including only targeted anticancer agents. Furthermore, nearly all patients (93%) had at least 1 drug chosen by both methods. We demonstrate overlap between agents recommended by CNS-TAP and PNOC003 tumor board, though this does not appear to improve survival. We do observe some discordance, highlighting strengths and limitations of each method. We propose that a combination of expert opinion and data-driven tools may improve targeted treatment recommendations for children with DIPG.

Abstract

ObjectiveTo determine the impact of a cerebrospinal fluid enterovirus polymerase chain reaction (PCR) test performance on hospital length of stay (LOS) in a large multicenter cohort of infants undergoing evaluation for central nervous system infection.Study designWe performed a planned secondary analysis of a retrospective cohort of hospitalized infants ≤60 days of age who had a cerebrospinal fluid culture obtained at 1 of 18 participating centers (2005-2013). After adjustment for patient age and study year as well as clustering by hospital center, we compared LOS for infants who had an enterovirus PCR test performed vs not performed and among those tested, for infants with a positive vs negative test result.ResultsOf 19 953 hospitalized infants, 4444 (22.3%) had an enterovirus PCR test performed and 945 (21.3% of tested infants) had positive test results. Hospital LOS was similar for infants who had an enterovirus PCR test performed compared with infants who did not (incident rate ratio 0.98 hours; 95% CI 0.89-1.06). However, infants PCR positive for enterovirus had a 38% shorter LOS than infants PCR negative for enterovirus (incident rate ratio 0.62 hours; 95% CI 0.57-0.68). No infant with a positive enterovirus PCR test had bacterial meningitis (0%; 95% CI 0-0.4).ConclusionsAlthough enterovirus PCR testing was not associated with a reduction in LOS, infants with a positive enterovirus PCR test had a one-third shorter LOS compared with infants with a negative enterovirus PCR test. Focused enterovirus PCR test use could increase the impact on LOS for infants undergoing cerebrospinal fluid evaluation.

•This is the first report of Staphylococcus equorumcarrying two distinct cfr-bearing plasmids, both structurally different from previously identified cfr-containing plasmids.•Besides the cfrgene, these plasmids also genes for arsenic, copper and cadmium resistance, along with the lincosamide-pleuromutilin-streptogramin a resistance gene lsa(B). The co-occurrence of both antimicrobial and heavy metal resistance genes increases the potential for co-selection of these plasmids.•This study emphasizes the critical need for ongoing surveillance to prevent the spread of multi-drug resistance from food-producing animals, underlining its importance for food safety and public health.

228. Words Speak Louder than Action?: A Mixed-Methods Case Study

Abstract

The purpose of this mixed-methods case study was to examine the dynamic of Culturally Responsive Pedagogy (Gay, 2010) among nine secondary agriculture teachers in ethnically diverse schools. By reviewing the diversity of student enrollment in the agriculture programs, the participants were separated by two groups: diverse and non-diverse. A hybrid coding system of axial and magnitude allowed an unbiased panel to quantify the comments of each teacher. Results showed a difference in magnitude codes between comments provided by teachers with a diverse student enrollment and teachers with a non-diverse student enrollment. A large difference existed among four of the six characteristics of Culturally Relevant Pedagogy (CRP) and a medium difference existed in the latter two. Recommendations toward teacher home visits, implementation of curriculum modifications and diverse community engagement were directed toward the participants and practicing secondary teachers. Additional research and opportunities for CRP among teacher training institutions are also recommended.
