Fungal secretomes contain a wide range of hydrolytic and oxidative enzymes, including cellulases, hemicellulases, pectinases, and lignin-degrading accessory enzymes, that synergistically drive litter decomposition in the environment. While secretome studies of model organisms such as Phanerochaete chrysosporium and Aspergillus species have greatly expanded our knowledge of these enzymes, few have extended secretome characterization to environmental isolates or conducted side-by-side comparisons of diverse species. Thus, the mechanisms of carbon degradation by many ubiquitous soil fungi remain poorly understood. Here we use a combination of LC-MS/MS, genomic, and bioinformatic analyses to characterize and compare the protein composition of the secretomes of four recently isolated, cosmopolitan, Mn(II)-oxidizing Ascomycetes (Alternaria alternata SRC1lrK2f, Stagonospora sp. SRC1lsM3a, Pyrenochaeta sp. DS3sAY3a, and Paraconiothyrium sporulosum AP3s5-JAC2a). We demonstrate that the organisms produce a rich yet functionally similar suite of extracellular enzymes, with species-specific differences in secretome composition arising from unique amino acid sequences rather than overall protein function. Furthermore, we identify not only a wide range of carbohydrate-active enzymes that can directly oxidize recalcitrant carbon, but also an impressive suite of redox-active accessory enzymes that suggests a role for Fenton-based hydroxyl radical formation in indirect, non-specific lignocellulose attack. Our findings highlight the diverse oxidative capacity of these environmental isolates and enhance our understanding of the role of filamentous Ascomycetes in carbon turnover in the environment.

As my colleagues have indicated, the thrill and value of qualitative assessment is that it let us loose to speak and dig into the questions that we had to keep silenced during rubric-based assessment. It allowed us to value our many questions about student writing and pedagogy. As we voiced our questions and discussed them vigorously we began to understand that the process could help each of us in refining course structure, pedagogy and assignments in the Transitional English classroom and in the college composition courses that follow.

Age and gender differences are prominent in the temperament literature, with the former particularly salient in infancy and the latter noted as early as the first year of life. This study represents a meta-analysis utilizing Infant Behavior Questionnaire-Revised (IBQ-R) data collected across multiple laboratories (N = 4438) to overcome limitations of smaller samples in elucidating links among temperament, age, and gender in early childhood. Algorithmic modeling techniques were leveraged to discern the extent to which the 14 IBQ-R subscale scores accurately classified participating children as boys (n = 2,298) and girls (n = 2,093), and into three age groups: youngest (< 24 weeks; n = 1,102), mid-range (24 to 48 weeks; n = 2,557), and oldest (> 48 weeks; n = 779). Additionally, simultaneous classification into age and gender categories was performed, providing an opportunity to consider the extent to which gender differences in temperament are informed by infant age. Results indicated that overall age group classification was more accurate than child gender models, suggesting that age-related changes are more salient than gender differences in early childhood with respect to temperament attributes. However, gender-based classification was superior in the oldest age group, suggesting temperament differences between boys and girls are accentuated with development. Fear emerged as the subscale contributing to accurate classifications most notably overall. This study leads infancy research and meta-analytic investigations more broadly in a new direction as a methodological demonstration, and also provides most optimal comparative data for the IBQ-R based on the largest and most representative dataset to date.

Background: Improvements in health behaviors and academic outcomes have been associated with school-based health centers (SBHCs). However, underlying mechanisms for these associations have been largely unexamined, particularly among lower-income youth. The current study examines the relationship between SBHCs and school connectedness and whether this relationship differs by youths' socioeconomic status (SES). Methods: Student-level cross-sectional data from 503 traditional high schools in California were analyzed using multilevel regression models. California Healthy Kids Survey 2013-2014 data included information on 3 dimensions of school connectedness and demographic characteristics including SES as measured by parental education. School-level demographic data was gathered from publicly available sources. Results: Although no significant relationship between SBHCs and any of the school connectedness dimensions emerged, there were significant cross-level interactions between SBHCs and parent education. SBHCs were more positively associated with school connectedness (adult caring, adult expectations, and meaningful participation) among lower SES students compared to students with higher SES. Conclusions: SBHCs may be particularly effective in affecting school connectedness among lower income youth populations. This has wide ranging implications with regards to planning (eg, careful selection of where SBHCs can be most effective), and future research (eg, examining the effectiveness of specific SBHC strategies that support connectedness).

School-based health centers (SBHCs) can take specific steps to provide culturally competent care for lesbian, gay, bisexual, transgender, and queer (LGBTQ) youth, potentially impacting well-being. A needs assessment survey was conducted among a convenience sample of SBHC administrators and medical directors to assess climates and actions supportive of LGBTQ quality medical care. Half (53%) of the SBHCs surveyed (N = 66) reviewed print materials for negative LGBTQ stereotypes, and 27.3% conducted exhaustive materials review. Regional differences were detected: 46.2% of Southern SBHCs conducted any materials review compared to 91.3% in the West and all in the East and Midwest (?[superscript 2], p < 0.001). In the last academic year, 45.5% conducted no medical provider trainings, and 54.5% conducted no general staff trainings on providing care for LGBTQ youth. On intake forms, 85.4% included preferred names, but only 23.5% included preferred pronoun. There are significant gaps in the extent to which SBHCs provide culturally competent care. These findings can guide future training and advocacy.

Gap junctions mediate communication between adjacent cells and are fundamental to the development and homeostasis in multicellular organisms. In invertebrates, gap junctions are formed by transmembrane proteins called innexins. Gap junctions allow the passage of small molecules through an intercellular channel, between a cell and another adjacent cell. The dipteran Rhynchosciara americana has contributed to studying the biology of invertebrates and the study of the interaction and regulation of genes during biological development. Therefore, this paper aimed to study the R. americana innexin-2 by molecular characterization, analysis of the expression profile and cellular localization. The molecular characterization results confirm that the message is from a gap junction protein and analysis of the expression and cellular localization profile shows that innexin-2 can participate in many physiological processes during the development of R. americana.

Sprinkler systems are one of the most popular methods of irrigation worldwide. One of their key parameters is the so-called level of uniformity, i.e. every portion of the soil should be irrigated with the same amount of water. Assessing the level of uniformity is crucial for optimal design of sprinkler systems. In this manuscript, a novel experimental benchmark is presented in order to test irrigation sprinklers, assess their performance, and define their acceptable working conditions. Different sprinklers have been tested, their water application depth curves have been determined, and their performance has been evaluated using a combination of metrics. Results show that the majority of sprinklers are characterized by very good performance in terms of operating pressures in the range 2.0-3.0 bar and tend to decrease their efficiency for operating pressures outside of that range.

Mercury (Hg) is a toxic heavy metal that poses significant environmental and human health risks. Soils and sediments, where Hg can exist as the Hg sulfide mineral metacinnabar (β-HgS), represent major Hg reservoirs in aquatic environments. Metacinnabar has historically been considered a sink for Hg in all but severely acidic environments, and thus disregarded as a potential source of Hg back to aqueous or gaseous pools. Here, we conducted a combination of field and laboratory incubations to identify the potential for metacinnabar as a source of dissolved Hg within near neutral pH environments and the underpinning (a)biotic mechanisms at play. We show that the abundant and widespread sulfur-oxidizing bacteria of the genus Thiobacillus extensively colonized metacinnabar chips incubated within aerobic, near neutral pH creek sediments. Laboratory incubations of axenic Thiobacillus thioparus cultures led to the release of metacinnabar-hosted Hg(II) and subsequent volatilization to Hg(0). This dissolution and volatilization was greatly enhanced in the presence of thiosulfate, which served a dual role by enhancing HgS dissolution through Hg complexation and providing an additional metabolic substrate for Thiobacillus. These findings reveal a new coupled abiotic-biotic pathway for the transformation of metacinnabar-bound Hg(II) to Hg(0), while expanding the sulfide substrates available for neutrophilic chemosynthetic bacteria to Hg-laden sulfides. They also point to mineral-hosted Hg as an underappreciated source of gaseous elemental Hg to the environment.

HIV prevalence varies throughout Africa, but the contribution of migration remains unclear. Using population-based data from ~22,000 persons, Grabowski et al. show that HIV-positive migrants selectively migrate to high prevalence areas and that out-migrants from these areas geographically disperse.

Plant-based milk has gained considerable attention; however, its high nutritional variation highlights the need for improved formulation designs to enhance its quality. This study aimed to nutritionally compare cow milk with plant-based milk produced from hazelnuts (H), Brazil nuts (BN), cashew nuts (CN), soybeans (S), and sunflower seeds (SS), and to perform physicochemical and technological characterization. The plant-based milk produced with isolated grains showed a nutritional composition inferior to that of cow milk in almost all evaluated parameters, protein content (up to 1.1 g 100 g−1), lipids (up to 2.7 g 100 g−1), color parameters, minerals, and especially calcium (up to 62.4 mg L−1), which were originally high in cow milk (up to 1030 mg L−1). However, the plant-based milk designed using a blend composition was able to promote nutritional enhancement in terms of minerals, especially iron (Fe) and magnesium (Mg), high-quality lipids (up to 3.6 g 100 g−1), and carbohydrates (3.4 g 100 g−1 using CN, BN, and S). The protein content was 1.3% compared to 5.7 in cow milk, and the caloric value of plant-based milk remained 32.8 at 52.1 kcal, similar to cow milk. Satisfactory aspects were observed regarding the shelf life, especially related to microbiological stability during the 11 d of storage at 4 °C. For the designed plant-based milk to be equivalent to cow milk, further exploration for optimizing the blends used to achieve better combinations is required. Furthermore, analyzing possible fortification and preservation methods to increase shelf life and meet the nutritional and sensory needs of the public would be interesting. [ABSTRACT FROM AUTHOR]

A new screen-printed electrode (SPE) modified with carbon black and ternary oxide (SiO2/TiO2/Nb2O5), denominated SPE/CB/SiTiNb and was developed for the direct detection of triclosan in tap water and mouthwash samples. The morphological characterization and electrochemical behavior of the sensors were performed by scanning electron microscopy, energy dispersive spectroscopy, and cyclic voltammetry. Differential pulse voltammetry was used to evaluate the proposed sensor in Britton–Robinson buffer 0.1 mol L−1 and showed a limit detection of 1.23 nmol L−1 in a linear range from 0.025 to 57.01 µmol L−1. Studies with possible interferents were performed demonstrating a significant selectivity of the sensor to the triclosan oxidation reaction. The sensor was applied in tap water and mouthwash real sample analyses, whose recoveries ranged from 93.4 to 98.8%, demonstrating that the sensor shows promise in the detection of triclosan in different matrices allied to high sensitivity, selectivity, low-cost, portability, disposability, and low sample volume, without pre-treatment, per analysis. Free energies G(X)n of 12 1,ω‐dihydrooligomers (X)n (n, number of the monomeric units) have been determined by quantum calculations. We note that these values are correlated by (a) an excellent linear relationship: G(X)n = An – 761.86 ± 10 (kcal mol–1) (TPSS‐TPSS/6.311 + + G(dp)); (b) for two oligomers (X)n and (Y)n, the difference of weighted free energies—G(X)n/n – G(Y)n/n—is a constant irrespective of n which results from the difference of free energies of the substituent. Consequently, from the Gn values of 1,ω‐dihydrooligoethenes (in fact n‐alkanes), a determination of the free energies of a 1,ω‐dihydrooligomers (X)n is obtained with a good accuracy by the calculation of the G value of its substituent; (c) the corresponding increments G(X)n/n – G(X)(n–1)/(n–1) are equaled and led to a single power law: [Gn/n – G(n–1)/(n–1) = 1282/n2.156 (kcal mol–1), R = 0.9992] or a linear relationship: [Gn/n – G(n–1)/(n–1) = 756.73/n(n–1) + 0.0211 (kcal mol–1), R = 1]. The introduction of mammals on oceanic islands currently threatens or has caused the extinction of many endemic species. Cats and rats represent the major threat for 40 % of currently endangered island bird species. Direct (predation) and/or indirect (exploitative competition for food resource) trophic interactions are key mechanisms by which invaders cause the decrease or extinction of native populations. Here, we investigated both direct and indirect trophic interactions amongst four predator species (i.e. animals that hunt, kill and feed on other animals), including three introduced mammals (Felis silvestris catus, Rattus rattus and Rattus exulans) and one critically endangered native bird, the Niau kingfisher (Todiramphus gertrudae). All four species’ diets and prey availability were assessed from sampling at the six main kingfisher habitats on Niau Island during the breeding season. Diet analyses were conducted on 578 cat scats, 295 rat digestive tracts (218 R. exulans and 77 R. rattus) and 186 kingfisher pellets. Despite simultaneous use of morphological and PCR-based methods, no bird remains in cat and rat diet samples could be assigned to the Niau kingfisher, weakening the hypothesis of current intense predation pressure. However, we determined that Niau kingfishers mainly feed on introduced and/or cryptogenic prey and highlighted the potential for exploitative competition between this bird and both introduced rat species (for Dictyoptera, Coleoptera and Scincidae). We recommend removing the cats and both rat species, at least within kingfisher breeding and foraging areas (e.g. mechanical or chemical control, cat sterilisation, biosecurity reinforcement), to simultaneously decrease predation risk, increase key prey availability and boost kingfisher population dynamics.

Soybean meal (SBM) is a co-product of the soybean oil industry that is rich in bioactive compounds, such as isoflavones. We aimed to study the effects of processing SBM by fermentation (Saccharomyces cerevisiae) (FSBM) and enzymatic hydrolysis (CelluMax C, a commercial cellulase) (ESBM) on its chemical composition, with emphasis on isoflavones. Fermentation increased protein content by 9%, ash content by 7%, dietary fiber by 11% and minerals by up to 38%, except for iron, which decreased by 26%. Fermentation completely removed oligosaccharides from SBM, while enzymatic processing decreased oligosaccharides by 45% in SBM. Both processes converted glycosylated isoflavones into the corresponding aglycones, the content of which increased by up to 7.7-fold. Biscuits containing SBM, FSBM and ESBM could be labeled as dietary sources of dietary fibers, potassium, phosphorous, calcium and zinc, as well as high in proteins, copper, iron, manganese and magnesium. While FSBM biscuits had lower sensory scores compared to SBM biscuits, ESBM biscuits had equivalent scores. During storage for 180 days at room temperature, the isoflavone profile of all biscuits remained stable. Moreover, storage did not impair microbiological and sensory qualities of any biscuits. Altogether, ESBM biscuits show great marketing potential.
