Abstract

Introduction: Diabetes mellitus is an ever-increasing medical condition that currently suffers 1 of 11 adults who may have lifelong commitment with insulin injections. Cell-laden hydrogels releasing insulin may provide the ultimate means of correcting diabetes. Here, we provide insights of this cell-based approach including latest preclinical and clinical progress both from academia and industry. Area covered: The present article focuses on reviewing latest advances in cell-laden hydrogels both from the technological and biological perspective. The most relevant clinical results including clinical trials are also discussed. Expert opinion: Current progress in technological issues (stem cells, devices, biomaterials) have contributed cell encapsulation science to have a very relevant progress in the field of diabetes treatment.

Abstract

Bioengineered functional muscle tissues are beneficial for regenerative medicine due to their treatment potential for various debilitating disorders, including myopathy and traumatic injuries. However, the contractile properties of engineered muscle constructs are lacking compared with their native counterparts. Here, we used microfluidic spinning to fabricate photocrosslinkable gelatin methacryloyl (GelMA) hydrogel fibres with well-defined surface morphologies for engineering muscle tissues. We examined whether the combination of topographical cues from surface micropatterning and biochemical stimulation with recombinant agrin can improve the generation of bioengineered muscle tissue. Topographical cues on micropatterned fibres promoted alignment of C2C12 myoblasts and augmented myotube formation during differentiation, as assessed by increased myotube length, aspect ratio, and the elevated mRNA expression of myogenic genes. Moreover, agrin treatment significantly increased acetylcholine receptor expression/clustering and myotube formation and upregulated dystrophin expression in differentiated C2C12 myotubes. Interestingly, the combination of topographical cues with agrin treatment further enhanced myotube maturation and functionality as shown by improved contractility under electrical stimulation. Thus, combining topographical cues and agrin treatment improved functions of engineered muscle tissue, which has potential in biorobotics, drug screening, tissue engineering, and regenerative medicine.

Abstract

We report on the measurement of the beam asymmetry Σ for the reactions → γ p → p η and → γ p → p η ′ from the GlueX experiment using an 8.2–8.8-GeV linearly polarized tagged photon beam incident on a liquid hydrogen target in Hall D at Jefferson Laboratory. These measurements are made as a function of momentum transfer − t with significantly higher statistical precision than our earlier η measurements and are the first measurements of η ′ in this energy range. We compare the results to theoretical predictions based on t -channel quasiparticle exchange. We also compare the ratio of Σ η to Σ η ′ to these models as this ratio is predicted to be sensitive to the amount of s ¯ s exchange in the production. We find that photoproduction of both η and η ′ is dominated by natural parity exchange with little dependence on − t .

Abstract

Abstract

We present precision measurements of the target and beam-target spin asymmetries from neutral pion electroproduction in deep-inelastic scattering (DIS) using the CEBAF Large Acceptance Spectrometer (CLAS) at Jefferson Lab. We scattered 6-GeV, longitudinally polarized electrons off longitudinally polarized protons in a cryogenic $^{14}$NH$_3$ target, and extracted double and single target spin asymmetries for $ep\rightarrow e^\prime\pi^0X$ in multidimensional bins in four-momentum transfer ($1.0, Comment: 18 preprint pages, 3 figures

Abstract

In this study, composite gelatin-polyaniline (PANI) nanofibers doped with camphorsulfonic acid (CSA) were fabricated by electrospinning and used as substrates to culture C2C12 myoblast cells. We observed enhanced myotube formation on composite gelatin-PANI nanofibers compared to gelatin nanofibers, concomitantly with enhanced myotube maturation. Thus, in myotubes, intracellular organization, colocalization of the dihydropyridine receptor (DHPR) and ryanodine receptor (RyR), expression of genes correlated to the excitation-contraction (E-C) coupling apparatus, calcium transients, and myotube contractibility were increased. Such composite material scaffolds combining topographical and electrically conductive cues may be useful to direct skeletal muscle cell organization and to improve cellular maturation, functionality, and tissue formation.

Abstract

High-momentum configurations of nucleon pairs at short-distance are probed using measurements of the $^{12}$C$(e,e'p)$ and $^{12}$C$(e,e'pN)$ reactions (where $N$ is either $n$ or $p$), at high-$Q^2$ and $x_B>1.1$. The data span a missing-momentum range of 300--1000 MeV/c and are predominantly sensitive to the transition region of the strong nuclear interaction from a Tensor to Scalar interaction. The data are well reproduced by theoretical calculations using the Generalized Contact Formalism with both chiral and phenomenological nucleon-nucleon ($NN$) interaction models. This agreement suggests that the measured high missing-momentum protons up to $1000$ MeV/c predominantly belong to short-ranged correlated (SRC) pairs. The measured $^{12}$C$(e,e'pN)$ / $^{12}$C$(e,e'p)$ and $^{12}$C$(e,e'pp)$ / $^{12}$C$(e,e'pn)$ cross-section ratios are consistent with a decrease in the fraction of proton-neutron SRC pairs and increase in the fraction of proton-proton SRC pairs with increasing missing momentum. This confirms the transition from an isospin-dependent tensor $NN$ interaction at $\sim 400$ MeV/c to an isospin-independent scalar interaction at high-momentum around $\sim 800$ MeV/c as predicted by theoretical calculation., Accepted for publication in Physics Letters B. 7 pages, 3 figures, and online supplementary materials

Abstract

Photon beam asymmetry Σ measurements for ω photoproduction in the reaction γ → p → ω p are reported for photon energies from 1.152 to 1.876 GeV. Data were taken using a linearly-polarized tagged photon beam, a cryogenic hydrogen target, and the CLAS spectrometer in Hall B at Jefferson Lab. The measurements obtained markedly increase the size of the database for this observable, extend coverage to higher energies, and resolve discrepancies in previously published data. Comparisons of these new results with predictions from a chiral-quark-based model and from a dynamical coupled-channels model indicate the importance of interferences between t-channel meson exchange and s- and u-channel contributions, underscoring sensitivity to the nucleon resonances included in those descriptions. Comparisons with the Bonn–Gatchina partial-wave analysis indicate the Σ data reported here help to fix the magnitudes of the interference terms between the leading amplitudes in that calculation (Pomeron exchange and the resonant portion of the J P = 3 / 2 + partial wave), as well as the resonant portions of the smaller partial waves with J P = 1 / 2 − , 3 / 2 − , and 5 / 2 + .

Abstract

Real-time monitoring of metabolically relevant biochemicals released in minuscule amounts is of utmost diagnostic importance. Superoxide anion as a primary member of reactive oxygen species, has physiological and pathological effects that depend on its concentration and release rate. Here we present fabrication and successfully testing of a highly sensitive electrochemical biosensor featuring a three-dimensional macroporous mesh of nanoporous gold tailored to measure the dynamics of extracellular superoxide concentration. Wide and accessible surface of the mesh combined with high porosity of the thin nanoporous gold coating enables capturing the analyte in pico- to nano-molar ranges. The mesh is functionalized with cytochrome- c (cyt- c ) and incorporated as a working electrode to measure the release rate of drug-induced superoxides from C2C12 cells through a porous membrane. The device displays a considerably improved superoxide sensitivity of 7.29 nA nM − 1 cm − 2 and a low level of detection of 70 pM. Such sensitivity is orders of magnitude higher than any similar enzyme-based electrochemical superoxide sensor and is attributed to the facile diffusion of the analyte through the well-spread nanofeatured gold skin. Superoxide generation rates captured from monolayer myoblast cultures containing about 4×10 4 cells, varied from 1.0 to 9.0 nM min − 1 in a quasi-linear fashion as a function of drug concentration. This work provides a platform for the development of highly sensitive molecular electrochemical biosensors.

Abstract

We report on the measurement of the γp→J/ψp cross section from Eγ=11.8 GeV down to the threshold at 8.2 GeV using a tagged photon beam with the GlueX experiment. We find that the total cross section falls toward the threshold less steeply than expected from two-gluon exchange models. The differential cross section dσ/dt has an exponential slope of 1.67±0.39 GeV-2 at 10.7 GeV average energy. The LHCb pentaquark candidates Pc+ can be produced in the s channel of this reaction. We see no evidence for them and set model-dependent upper limits on their branching fractions B(Pc+→J/ψp) and cross sections σ(γp→Pc+)×B(Pc+→J/ψp).

Abstract

First measurements of double-polarization observables in ω photoproduction off the proton are presented using transverse target polarization and data from the CEBAF Large Acceptance Spectrometer (CLAS) FROST experiment at Jefferson Lab. The beam-target asymmetry F has been measured using circularly polarized, tagged photons in the energy range 1200-2700 MeV, and the beam-target asymmetries H and P have been measured using linearly polarized, tagged photons in the energy range 1200-2000 MeV. These measurements significantly increase the database on polarization observables. The results are included in two partial-wave analyses and reveal significant contributions from several nucleon (N^{*}) resonances. In particular, contributions from new N^{*} resonances listed in the Review of Particle Properties are observed, which aid in reaching the goal of mapping out the nucleon resonance spectrum.

Abstract

Three-dimensional (3D) bioprinting is an emerging biofabrication technology, driving many innovations and opening new avenues in regenerative therapeutics. The aim of 3D bioprinting is to fabricate grafts in vitro, which can then be implanted in vivo. However, the tissue culture ex vivo carries safety risks and thereby complicated manufacturing equipment and practice are required for tissues to be implanted in the humans. The implantation of printed tissues also adds complexities due to the difficulty in maintaining the structural integrity of fabricated constructs. To tackle this challenge, the concept of in situ 3D bioprinting has been suggested in which tissues are directly printed at the site of injury or defect. Such approach could be combined with cells freshly isolated from patients to produce custom-made grafts that resemble target tissue and fit precisely to target defects. Moreover, the natural cellular microenvironment in the body can be harnessed for tissue maturation resulting in the tissue regeneration and repair. Here, we discuss literature reports on in situ 3D printing and we describe future directions and challenges for in situ 3D bioprinting. We expect that this novel technology would find great attention in different biomedical fields in near future.

Abstract

U radu se analizira tržište rada starijih radnika u kontekstu što dužeg ostanka na tržištu rada u Republici Hrvatskoj. Slijedom nepovoljne demografske situacije, dužeg životnog vijeka te ugroženosti sustava mirovinskog osiguranja, u proteklih nekoliko desetljeća u velikom broju zemalja provedene su inicijative produženja radnog vijeka. Analiza je pokazala da stariji radnici u Republici Hrvatskoj, unatoč nižoj radnoj aktivnosti u usporedbi s drugim zemljama, imaju dodatnog radnog kapaciteta za duži ostanak na tržištu rada koji pritom ne bi ugrozio zaposlenost mlađih skupina. Dokazano je i da tržište rada Republike Hrvatske nije prilagođeno starijim dobnim skupinama, te da najveće šanse za duži ostanak na tržištu rada, imaju visokoobrazovani i oni boljeg materijalnog statusa, odnosno oni koji općenito imaju najviše prilika, dok oni slabijeg obrazovanja, socioekonomskog statusa i zdravlja, iako možda ne žele napustiti tržište rada, imaju male mogućnosti, odnosno šanse za dužim ostankom na tržištu rada.

Abstract

The atomic nucleus is made of protons and neutrons (nucleons), which are themselves composed of quarks and gluons. Understanding how the quark–gluon structure of a nucleon bound in an atomic nucleus is modified by the surrounding nucleons is an outstanding challenge. Although evidence for such modification—known as the EMC effect—was first observed over 35 years ago, there is still no generally accepted explanation for its cause1,2,3. Recent observations suggest that the EMC effect is related to close-proximity short-range correlated (SRC) nucleon pairs in nuclei4,5. Here we report simultaneous, high-precision measurements of the EMC effect and SRC abundances. We show that EMC data can be explained by a universal modification of the structure of nucleons in neutron–proton SRC pairs and present a data-driven extraction of the corresponding universal modification function. This implies that in heavier nuclei with many more neutrons than protons, each proton is more likely than each neutron to belong to an SRC pair and hence to have distorted quark structure. This universal modification function will be useful for determining the structure of the free neutron and thereby testing quantum chromodynamics symmetry-breaking mechanisms and may help to discriminate between nuclear physics effects and beyond-the-standard-model effects in neutrino experiments.

Abstract

The beam spin asymmetry of the exclusive pseudoscalar channel e→p→e′p′η was measured for the first time in the deep-inelastic regime (W>2 GeV/c2 and Q2>1 GeV2/c2) using a longitudinally polarized 5.78 GeV electron beam at Jefferson Lab with the CEBAF Large Acceptance Spectrometer. The data were accumulated in 144 four-dimensional bins of Q2, xB, −t and ϕ over a wide kinematic range, where ϕ is the azimuthal angle between the lepton and hadron scattering planes, The measured azimuthal dependence with large amplitudes of the sin⁡ϕ moments is a clear indication of a substantial contribution to the polarized cross-section from transversely polarized virtual photons. In the framework of generalized parton distributions (GPDs) this contribution is expressed via longitudinal-transverse interference between chiral-even and chiral-odd GPDs. The experimental results are compared to the existing theoretical models demonstrating the sensitivity to the product of chiral-odd and chiral-even GPDs and provide new constraints to the existing GPD parameterizations. Abstract

We report the first experimental measurements of the nine 1-fold differential cross sections for the γp→π+π−p reaction, obtained with the CLAS detector at Jefferson Laboratory. The measurements cover the invariant mass range of the final state hadrons from 1.6 GeV Δ(1620)1/2−, Δ(1700)3/2−, N(1720)3/2+, and Δ(1905)5/2+ resonances, which have dominant decays into the ππN final states rather than the more extensively studied single meson decay channels.

Abstract

In recent years, many mechanical, physical, chemical, and biochemical features of biomatrices have emerged as important properties to dictate the fates of cells. To construct chemically defined biomaterials to recapitulate various biological niches for both cell biology research and therapeutic utilities, it has become increasingly clear that a simple hydrated polymer network would not be able to provide the complex cues and signaling required for many types of cells. The researchers are facing a growing list of mechanophysical and biochemical properties, while each of them could be an important cellular trigger. To include all these design parameters in screening and synthesis is practically difficult, if not impossible. Developing novel high throughput screening technology by combining assay miniaturization, computer simulations, and modeling can help researchers to tackle the challenge to identify the most relevant parameters to tailor materials for specific applications.

Abstract

We report on the measurement of the γp→J/ψp cross section from Eγ=11.8 GeV down to the threshold at 8.2 GeV using a tagged photon beam with the GlueX experiment. We find that the total cross section falls toward the threshold less steeply than expected from two-gluon exchange models. The differential cross section dσ/dt has an exponential slope of 1.67±0.39 GeV-2 at 10.7 GeV average energy. The LHCb pentaquark candidates Pc+ can be produced in the s channel of this reaction. We see no evidence for them and set model-dependent upper limits on their branching fractions B(Pc+→J/ψp) and cross sections σ(γp→Pc+)×B(Pc+→J/ψp). © 2019 American Physical Society.

The data were accumulated in 144 four-dimensional bins of Q2 , xB , − t and ϕ over a wide kinematic range, where ϕ is the azimuthal angle between the lepton and hadron scattering planes, The measured azimuthal dependence with large amplitudes of the sin⁡ϕ moments is a clear indication of a substantial contribution to the polarized cross-section from transversely polarized virtual photons. In the framework of generalized parton distributions (GPDs) this contribution is expressed via longitudinal-transverse interference between chiral-even and chiral-odd GPDs. The experimental results are compared to the existing theoretical models demonstrating the sensitivity to the product of chiral-odd and chiral-even GPDs and provide new constraints to the existing GPD parameterizations.

Abstract

In the past two decades, deeply virtual Compton scattering of electrons has been successfully used to advance our knowledge of the partonic structure of the free proton and investigate correlations between the transverse position and the longitudinal momentum of quarks inside the nucleon. Meanwhile, the structure of bound nucleons in nuclei has been studied in inclusive deep-inelastic lepton scattering experiments off nuclear targets, showing a significant difference in longitudinal momentum distribution of quarks inside the bound nucleon, known as the EMC effect. In this work, we report the first beam spin asymmetry (BSA) measurement of exclusive deeply virtual Compton scattering (DVCS) off a proton bound in $^4$He. The data used here were accumulated using a $6$ GeV longitudinally polarized electron beam incident on a pressurized $^4$He gaseous target placed within the CLAS spectrometer in Hall-B at the Thomas Jefferson National Accelerator Facility. The azimuthal angle ($\phi$) dependence of the BSA was studied in a wide range of virtual photon and scattered proton kinematics. The $Q^2$, $x_B$, and t dependencies of the BSA on the bound proton are compared with those on the free proton. In the whole kinematical region of our measurements, the BSA on the bound proton is smaller by 20\% to 40\%, indicating possible medium modification of its partonic structure.

Abstract

First measurements of double-polarization observables in $\omega$ photoproduction off the proton are presented using transverse target polarization and data from the CEBAF Large Acceptance Spectrometer (CLAS) FROST experiment at Jefferson Lab. The beam-target asymmetry $F$ has been measured using circularly polarized, tagged photons in the energy range 1200 - 2700 MeV, and the beam-target asymmetries $H$ and $P$ have been measured using linearly polarized tagged photons in the energy range 1200 - 2000 MeV. These measurements significantly increase the database on polarization observables. The results are included in two partial-wave analyses and reveal significant contributions from several nucleon ($N^\ast$) resonances. In particular, contributions from new $N^\ast$ resonances listed in the Review of Particle Properties are observed, which aid in reaching the goal of mapping out the nucleon resonance spectrum., Comment: Two authors added, figures updated

Abstract

We measured the triple coincidence A(e,e'np) and A(e,e'pp) reactions on carbon, aluminum, iron, and lead targets at Q2 > 1.5 (GeV/c)2, xB > 1.1 and missing momentum > 400 MeV/c. This was the first direct measurement of both proton-proton (pp) and neutron-proton (np) short-range correlated (SRC) pair knockout from heavy asymmetric nuclei. For all measured nuclei, the average proton-proton (pp) to neutron-proton (np) reduced cross-section ratio is about 6%, in agreement with previous indirect measurements. Correcting for Single-Charge Exchange effects decreased the SRC pairs ratio to ~ 3%, which is lower than previous results. Comparisons to theoretical Generalized Contact Formalism (GCF) cross-section calculations show good agreement using both phenomenological and chiral nucleon-nucleon potentials, favoring a lower pp to np pair ratio. The ability of the GCF calculation to describe the experimental data using either phenomenological or chiral potentials suggests possible reduction of scale- and scheme-dependence in cross section ratios. Abstract

We report on the measurement of the beam asymmetry ς for the reactions γ - p→pη and γ - p→pη′ from the GlueX experiment using an 8.2-8.8-GeV linearly polarized tagged photon beam incident on a liquid hydrogen target in Hall D at Jefferson Laboratory. These measurements are made as a function of momentum transfer -t with significantly higher statistical precision than our earlier η measurements and are the first measurements of η′ in this energy range. We compare the results to theoretical predictions based on t-channel quasiparticle exchange. We also compare the ratio of ςη to ςη′ to these models as this ratio is predicted to be sensitive to the amount of ss exchange in the production. We find that photoproduction of both η and η′ is dominated by natural parity exchange with little dependence on -t. © 2019 American Physical Society.

Abstract

The paper analyses the labour market for older workers from the point of view of prolonging their participation in the labour market in the Republic of Croatia. Due to unfavourable demographic situations, longer life spans and the threats to pension insurance systems, there have been many initiatives, in a number of countries, in the last couple of decades that have the goal of facilitating longer working lives. The current analysis has shown that older workers in the Republic of Croatia, despite their economic activity being lower than that of other countries, do have the additional working capacity that can permit longer labour market participation, without compromising the employment of younger cohorts. It has also shown that the labour market of the Republic of Croatia is not well suited to older age groups and that those with higher education and financial security, those who generally have the most opportunities, have the best chances for longer labour market participation ; while the odds are against longer labour market participation for those with lower education, socioeconomic status and health, even if they might sometimes be willing to participate in the labour market

The doubly strange Ξ baryons provide an effective way to study a puzzle called the missing-baryons problem where both quark models and lattice gauge theory predict more baryon excited states than are seen experimentally. However, few of these excited states have been observed with any certainty. Here, high-mass Ξ∗ states have been searched for in photoproduction with the detector of the CLAS Collaboration, and upper limits for the total cross sections have been established from threshold to W=3.3 GeV. In addition, the total cross sections of the ground-state Ξ−(1320) and first excited state Ξ−(1530) are presented, extending significantly the center-of-mass energy range of previous data.

Abstract

Skeletal muscle tissue engineering (SMTE) aims at repairing defective skeletal muscles. Until now, numerous developments have been done in SMTE, however it is still challenging to recapitulate the complexity of muscles with current methods of fabrication. In this review, after a brief description of the anatomy of skeletal muscle, and a short state-of-the-art on developments made in SMTE with “conventional methods”, we focus on the use of three dimensional (3D) bioprinting as a new tool for SMTE. We discuss on the current bioprinting methods, and provide an overview on the bioink formulations and properties used in 3D bioprinting. Finally, we highlight different advances made in SMTE by 3D bioprinting, and describe future needs and provide a short perspective.
