The fitness effects associated with Wolbachia infection have wide-ranging ecological and evolutionary consequences for host species. How these effects are modulated by the relative influence of host and Wolbachia genomes has been described as a balancing act of genomic cooperation and conflict. For vertically transmitted symbionts, like cytoplasmic Wolbachia, concordant host-symbiont fitness interests would seem to select for genomic cooperation. However, Wolbachia's ability to manipulate host reproductive systems and distort offspring sex ratios presents an evolutionary conflict of interest with infected hosts. In the parthenogenesis-inducing (PI) form of Wolbachia found in many haplodiploid insects, Wolbachia fitness is realized through females and is enhanced by their feminization of male embryos and subsequent parthenogenetic reproduction. In contrast, as long as Wolbachia is not fixed in a population and sexual reproduction persists, fitness for the host species is realized through both male and female offspring production. How these cooperating and competing interests interact and the relative influence of host and Wolbachia genomes were investigated in the egg parasitoid Trichogramma kaykai, where Wolbachia infection has remained at a low frequency in the field. A factorial design in which laboratory cultures of Wolbachia-infected T. kaykai were cured and re-infected with alternative Wolbachia strains was used to determine the relative influence of host and Wolbachia genomes on host fitness values. Our results suggest fitness variation is largely a function of host genetic background, except in the case of offspring sex ratio where a significant interaction between host and Wolbachia genomes was found. We also find a significant effect associated with the horizontal transfer of Wolbachia strains, which we discuss in terms of the potential for coadaptation in PI-Wolbachia symbioses.

Once-useful traits that no longer contribute to fitness tend to decay over time. Here, we address whether the expression of mating-related traits that increase the fitness of sexually reproducing individuals but are likely less useful or even costly to asexual counterparts seems to exhibit decay in the latter. Potamopyrgus antipodarum is a New Zealand freshwater snail characterized by repeated transitions from sexual to asexual reproduction. The frequent coexistence of sexual and asexual lineages makes P. antipodarum an excellent model for the study of mating-related trait loss. Under the presumption (inherent in the Biological Species Concept) that failure to discriminate between conspecific and heterospecific mating partners represents a poor mate choice, we used a mating choice assay including sexual and asexual P. antipodarum females and conspecific (presumed better choice) vs. heterospecific (presumed worse choice) males to evaluate the loss of behavioral traits related to sexual reproduction. We found that sexual females engaged in mating behaviors with conspecific mating partners more frequently and for a greater duration than with heterospecific mating partners. By contrast, asexual females mated at similar frequency and duration as sexual females, but did not mate more often or for longer duration with conspecific vs. heterospecific males. While further confirmation will require inclusion of a more diverse array of sexual and asexual lineages, these results are consistent with a scenario where selection acting to maintain effective mate discrimination in asexual P. antipodarum is weak or ineffective relative to sexual females and, thus, where asexual reproduction is associated with the evolutionary decay of mating-related traits in this system.

Wolbachia are the most successful intracellular bacteria in arthropods. They can manipulate host reproduction to favour infected females, which transmit Wolbachia to their progeny and increase the presence of Wolbachia in the population. The reproductive alterations caused by Wolbachia include feminization, parthenogenesis, male killing and cytoplasmic incompatibility (CI), among which CI is the most common. CI leads to embryonic lethality when Wolbachia‐infected males mate with uninfected females or those infected with an incompatible strain. This lethality can be rescued if females are infected with a compatible strain. Although CI was described in the 1960s and its connection to Wolbachia was made in the 1970s, the genes responsible for CI, called CI factors, were not identified until recently. Since then, significant progress has been made in understanding the molecular mechanism of CI using a combination of genetic, phylogenetic, biochemical and structural approaches. The detailed molecular mechanisms behind this fascinating endosymbiotic bacteria‐induced phenotype have begun to emerge. Here, we summarize recent progress in understanding the molecular mechanism of CI, especially focusing on the recently solved CI factor structures and discussing what these new structures brought in terms of CI mechanism. Major hypotheses on sex evolution predict that resource abundance and heterogeneity should either select for or against sexual reproduction. However, seldom have these predictions been explicitly tested in the field. Here, we investigated this question using soil oribatid mites, a diverse and abundant group of soil arthropods whose local communities can be dominated by either sexual or asexual species. First, we refined theoretical predictions by addressing how the effects of resource abundance, heterogeneity and abiotic conditions could modify each other. Then, we estimated the strength of selection for sexual species in local communities while controlling for phylogeny and neutral processes (ecological drift and dispersal), and tested its relation to resource and abiotic gradients. We show that sexual species tended to be favoured with increasing litter amount, a measure of basal resource abundance. Further, there was some evidence that this response occurred mainly under higher tree species richness, a measure of basal resource heterogeneity. This response to resources is unlikely to reflect niche partitioning between reproductive modes, as sexual and asexual species overlapped in trophic niche according to a comparative analysis using literature data on stable isotope ratios. Rather, these findings are consistent with the hypothesis that sex facilitates adaptation by breaking unfavourable genetic associations, an advantage that should increase with effective population size when many loci are under selection and, thus, with resource abundance. The genus Thyreophagus is distributed worldwide, containing species occurring in stored food, house dust, bark, subcortical habitats, scale insect associations, and nests of wasps and bees. As species of this genus prefer concealed habitats, the actual species diversity is currently underappreciated. In addition, some described taxa are poorly known and their species boundaries are not entirely clear. Only a few species are known from both adult and deutonymphal stages. Using high-resolution light microscope and scanning electron microscopy, we describe a new partenogenetic species, Thyreophagus calusorum sp. n. from Florida (USA) based on adult stage (female) and heteromorphic deutonymph. This species was reared in the lab and its morphological variation was also studied. We did not find any substantial variation in the shape of the female spermatheca, but in the heteromorphic deutonymph, we did find extensive variation in the shape of tibial setae hT I (a taxonomically important character). Furthermore, we summarized taxonomic data and compiled an annotated checklist of species of Thyreophagus, with 35 species-level taxa. We clarified the following nomenclatorial questions: type species designation in Monieziella Berlese, 1897 was done by A. Berlese in G. Leonardi 1900 (Tyroglyphus entomophagus Laboulbène) not by A. Jacot (1936) as it was though previously; Monieziella mali Berlese, 1897 is an unjustified emendation and junior objective synonym of the name Acarus malusShimer, 1868, currently Hemisacoptes malus (Shimer, 1868). Three subspecies of Thyreophagus were elevated to the species status due to their distinct morphology: Thyreophagus italicusVacante, 1989, stat. nov. (=Thyreophagus entomophagus italicusVacante, 1989); Thyreophagus mauritianus (Fain, 1982), stat. nov. Premise: Parthenogenesis is the capacity of organisms to develop embryos from unfertilized eggs. When parthenogenesis is coupled with unreduced gamete formation (apomeiosis), genetically maternal progeny result. Genetic elucidation of this form of reproduction in plants, apomixis, has important agronomic implications. However, genetic characterization of apomeiosis and parthenogenesis has been problematic in part because the traits usually co‐occur and are restricted to polyploids. In this work, the inheritance of parthenogenetic embryo development, by itself, was studied at the diploid level. Methods: Progeny resulting from a cross between a diploid (2n = 18), heterozygous, parthenogenetic pollen donor, and a diploid, wildtype, sexual seed parent were evaluated. Paternity was tested with conserved orthologous sequence (COS) markers, reproductive development of F1s was evaluated with microscopy of cleared ovules, and an amplified fragment length polymorphism (AFLP) marker (Eagc × Macg.615) co‐segregating with parthenogenesis was characterized at the sequence level. Results: Of 102 diploid biparental progeny, 47 exhibited parthenogenetic embryo and endosperm development, and 55 lacked development of the egg and central cell. This result is consistent with Mendelian inheritance for a single locus (P = 0.43). Isolation and sequencing of the AFLP marker indicates that it is likely a portion of a Ty‐Gypsy retrotransposon. Attempts to develop a sequence‐characterized amplified region marker from the AFLP were unsuccessful. Conclusions: This work shows that parthenogenesis can be transmitted simply at the diploid level. This advance is key in the development of a tractable system in Erigeron aimed at the identification of the parthenogenesis locus using genetic mapping strategies. [ABSTRACT FROM AUTHOR]

Summary: Despite previous research demonstrating the benefits of including growth factors and antioxidants to maturation medium to support embryo production, to date the effect of epidermal growth factor (EGF) and melatonin (Mel) on oocyte competency has not been studied. This study supplemented in vitro maturation (IVM) medium with EGF (10 ng/ml) and Mel (50 ng/ml) alone, or in combination, and evaluated cumulus cell (CC) gene expression and the development and quality of parthenogenetic blastocysts. No differences in CC gene expression levels indicative of developmental potential were found among the treatment groups. Antioxidant gene CuZnSOD was significantly (P < 0.05) decreased in CCs from the Mel group. Moreover, blastocyst rates on day 7 were significantly increased in EGF or Mel (P < 0.05), but not EGF+Mel. Significant decrease (P < 0.05) in GPX1, CuZnSOD, SLC2A1 and HSPA1A (P = 0.07) mRNA levels was observed in blastocysts from the Mel group. OCT4 gene expression was significantly increased (P < 0.05) in EGF+Mel and confirmed using immunofluorescence. Our results indicate that, despite the lack of changes of competence-related genes in CCs, IVM medium supplemented with Mel improved the culture environment sufficiently, resulting in improved blastocysts. Moreover, EGF and Mel combined during maturation increased OCT4 gene and protein expression in blastocysts, indicating its potential for stem cells. [ABSTRACT FROM AUTHOR]

Haploid plants with a doubled set of chromosomes (doubled haploid (DH)) significantly speed up the selection process by the fixation of genetic traits in each locus in the homozygous state within one generation. Doubled haploids are mainly attained by the formation of plants from the cultured gametophytic (haploid) tissues and cells in vitro, or by targeted reduction in the parent chromosome during intra- or interspecific hybridization. Since then, DH has become one of the most powerful tools to support various basic research studies, as well as applied research. This review is focused on the recent development of the production of doubled haploids in vivo and their fundamental bases. The various mechanisms and approaches responsible for the formation of haploids in vivo are discussed, particularly the induction of parthenogenesis by BBM-like proteins, the long constructed Salmon system of wheat, the usage of patatin-like phospholipases MTL/PLA1/NLD, the IG1 system, uniparental genome elimination during interspecific hybridization, and the perspective technology of centromeric histone 3 (CENH3) modification. Intersex is defined as sexually aberrant individuals composed of a single sexual genotype that simultaneously displays female and male characteristics. The occurrence of intersex individuals in egg parasitoids, Trichogramma spp., is often associated with the symbiotic bacteria, Wolbachia, which induce thelytokous parthenogenesis of Trichogramma. It is still unclear whether intersex was caused by the reduced Wolbachia titer, effects of abiotic factors, or interactions between abiotic factors and Wolbachia. To answer this question, the females of Wolbachia-infected (TDW) and uninfected (TD) Trichogramma dendrolimi were treated using different tetracycline or rifampicin concentrations. The results showed that the proportion of intersexes or males of TDW offspring significantly increased as the concentration of both antibiotics increased; however, they did not occur without antibiotics. Occurrence of intersex was not observed in TD offspring. Twelve types of intersex were found and categorized by masculinization levels. The masculinized level of offspring intersex increased with tetracycline but not with rifampicin. Wolbachia titers of maternal TDW females significantly decreased with higher antibiotics concentrations. Wolbachia titers of intersex offspring were higher than offspring males, lower than offspring females, and negatively correlated with masculinized level of offspring intersex. The results revealed that the frequency and masculinized level of offspring intersex is quantitatively dependent on Wolbachia titer. Male annihilation and mating disruption provide limited success in the control of Tuta absoluta (Meyrick) (Lepidoptera: Gelechiidae) populations. The ability of T. absoluta to reproduce parthenogenetically, due to Wolbachia (Hertig) (Rickettsiales: Ehrlichiaceae) infection, has been reported as a possible contributing factor to the limited success of management strategies aimed at male moths. The genetic diversity in T. absoluta from four localities in South Africa were investigated by employing a fragment of the mitochondrial cytochrome c oxidase subunit I and Wolbachia strains were identified with the multilocus sequence typing scheme. Fecundity and fertility of parthenogenetically produced eggs and female lifespan were also recorded. High rates (≥ 95%) of infection with a single Wolbachia strain (ST41) occurred, but genetic diversity was low and consisted of only four haplotypes, one of which was novel. None of the eggs laid by unmated females were viable. Wolbachia infection in South African T. absoluta populations does not lead to either parthenogenesis or failure of pheromone-based strategies. The extant reptiles are one of the most diverse clades among terrestrial vertebrates and one of a few groups with instances of parthenogenesis. Due to the hybrid origin of parthenogenetic species, reference genomes of the parental species as well as of the parthenogenetic progeny are indispensable to explore the genetic foundations of parthenogenetic reproduction. Here, we report on the first genome assembly of rock lizard Darevskia valentini, a paternal species for several parthenogenetic lineages. The novel genome was used in the reconstruction of the comprehensive phylogeny of Squamata inferred independently from 7369 trees of single-copy orthologs and a supermatrix of 378 conserved proteins. We also investigated Hox clusters, the loci that are often regarded as playing an important role in the speciation of animal groups with drastically diverse morphology. We demonstrated that Hox clusters of D. valentini are invaded with transposons and contain the HoxC1 gene that has been considered to be lost in the amniote ancestor. This study provides confirmation for previous works and releases new genomic data that will contribute to future discoveries on the mechanisms of parthenogenesis as well as support comparative studies among reptiles. Melanoides tuberculata (Müller, 1774) is a widely distributed aquatic gastropod, primarily found in tropical regions, which has also invaded geothermally heated waters in temperate regions. The spread of M. tuberculata globally has primarily been in association with the aquarium trade and aquatic plants. In New Zealand, M. tuberculata is known from both the aquarium trade and from one non-indigenous population in a geothermal stream. To date, the origins of M. tuberculata in the aquarium trade in New Zealand--or elsewhere--has not been systematically examined genetically, and it has not been confirmed whether New Zealand's nonindigenous population is genetically similar to, or originated from, individuals sold in aquarium stores. We examined the origins of New Zealand M. tuberculata, in the aquarium trade, and in the established non-indigenous population using COI marker sequences. Representatives of M. tuberculata in New Zealand have not originated from a single source; two distinct genetic clades were identified for individuals, with the origins of each inferred to be from Asia and Africa. The non-indigenous population and individuals sourced from one private aquarium owner were found to originate in Asia, and are genetically similar to those collected from Singapore, one of the most significant countries globally for the culturing and distribution of aquarium species. The remaining collections, from a display aquarium and three online suppliers in various New Zealand locations, were most similar genetically to populations native to Lake Malawi and Tanganyika, and non-indigenous populations from Australia. Overall, as M. tuberculata is an obligate parthenogen, each independently introduced clone may be considered a distinct non-indigenous taxon. We recommend further systematic genetic analyses of individuals in the aquarium trade elsewhere, to assess more fully the diversity and origins of Melanoides being transported in the aquarium trade. Determining how and how often asexual lineages emerge within sexual species is central to our understanding of sex-asex transitions and the long-term maintenance of sex. Asexuality can arise "by transmission" from an existing asexual lineage to a new one through different types of crosses. The occurrence of these crosses, cryptic sex, variations in ploidy, and recombination within asexuals greatly complicates the study of sex-asex transitions, as they preclude the use of standard phylogenetic methods and genetic distance metrics. In this study we show how to overcome these challenges by developing new approaches to investigate the origin of the various asexual lineages of the brine shrimp Artemia parthenogenetica. We use a large sample of asexuals, including all known polyploids, and their sexual relatives. We combine flow cytometry with mitochondrial and nuclear DNA data. We develop new genetic distance measures and methods to compare various scenarios describing the origin of the different lineages. We find that all diploid and polyploid A. parthenogenetica likely arose within the past 80,000 years through successive and nested hybridization events that involved backcrosses with different sexual species. All A. parthenogenetica have the same common ancestor and therefore likely carry the same asexuality gene(s) and reproduce by automixis. These findings radically change our view of sex-asex transitions in this group and show the importance of considering scenarios of asexuality by transmission. Aphids are hemimetabolous insects that undergo incomplete metamorphosis without pupation. The annual life cycle of most aphids includes both an asexual (viviparous) and a sexual (oviparous) phase. Sexual reproduction only occurs once per year and is followed by many generations of asexual reproduction, during which aphids propagate exponentially with telescopic development. Here, we discuss the potential links between viviparous embryogenesis and derived developmental features in the pea aphid Acyrthosiphon pisum, particularly focusing on germline specification and axis determination, both of which are key events of early development in insects. We also discuss potential evolutionary paths through which both viviparous and oviparous females might have come to utilize maternal germ plasm to drive germline specification. This developmental strategy, as defined by germline markers, has not been reported in other hemimetabolous insects. In viviparous females, furthermore, we discuss whether molecules that in other insects characterize germ plasm, like Vasa, also participate in posterior determination and how the anterior localization of the hunchback orthologue Ap-hb establishes the anterior-posterior axis. We propose that the linked chain of developing oocytes and embryos within each ovariole and the special morphology of early embryos might have driven the formation of evolutionary novelties in germline specification and axis determination in the viviparous aphids. Moreover, based upon the finding that the endosymbiont Buchnera aphidicola is closely associated with germ cells throughout embryogenesis, we propose presumptive roles for B. aphidicola in aphid development, discussing how it might regulate germline migration in both reproductive modes of pea aphids. In facultatively parthenogenetic populations, the prevalence of sexual reproduction depends on whether females mate and therefore produce sons and daughters or avoid mating and produce daughters only.The relative advantage of mating in such species may depend on a female's own reproductive origin (i.e. development from a fertilised or unfertilised egg) if parthenogenesis reduces heterozygosity similar to sexual inbreeding, or if it inhibits mating, sperm storage or fertilisation. But effects of reproductive origin on development and performance are poorly understood.Using the facultatively parthenogenetic stick insect, Extatosoma tiaratum, we quantified morphology, mating probability, and reproductive success in mated versus unmated females of sexual versus automictic (parthenogenetic) origin.We found strong evidence that increased homozygosity negatively impacted some traits in parthenogenetically produced females: compared to sexually produced females, parthenogenetically produced females were smaller and more prone to deformities in vestigial wings, but not more prone to fluctuating asymmetry in their legs.Parthenogenetically produced females received fewer mating attempts and avoided mating more often than sexually produced females. Yet, contrary to the expectation that sex should rescue parthenogenetic lineages from the detrimental effects of increased homozygosity, parthenogenetically produced females gained no net reproductive benefit from mating, suggesting that physiological constraints limit fitness returns of sexual reproduction for these females.Our findings indicate that advantages of mating in this species depend on female reproductive origin. These results could help to explain spatial distributions of sex in facultatively parthenogenetic animals and evolutionary transitions to obligate asexuality. In eukaryotes, gamete size difference between the two sexes (anisogamy) evolved from gametes of equal size in both mating types (isogamy). The gamete dynamics (GD) model for anisogamy evolution combines gamete limitation and competition and predicts that if gametes of both mating types can develop parthenogenetically (i.e. without fusing with the opposite mating type), large isogamy can evolve under gamete-limited conditions. Ulvophycean marine green algae that have been claimed to exhibit various gametic systems from isogamy to anisogamy are important models for testing such theories. However, in most previous papers, whether a species is isogamous or anisogamous has not been examined statistically. Caution is necessary regarding claims of slight anisogamy because of gamete size variation. We reveal (i) that the gametic system of Struvea okamurae is large isogamy using a generalized linear mixed model, which accounted for the variation of gamete size among individual gametophytes, and (ii) that gametes of this alga can actually develop parthenogenetically, contrary to a previous report. Its habitat environments and protracted duration of gamete release suggest that this alga might experience gamete-limited conditions. Struvea okamurae seems to produce large parthenogenetic isogametes following GD model predictions, as an adaptation to deep waters.

Assisted reproductive technologies (ART) play a crucial role in conserving threatened wildlife species such as Bos gaurus. ART requires a large number of mature oocytes, and small antral follicles (SAFs) in the ovary are often used to obtain abundant sources of bovine oocytes. However, oocytes from SAFs often experience difficulty completing maturation and obtaining high quality and quantity of blastocyst formation compared to fully grown oocytes. This study aimed to increase the number of high-quality mature oocytes and improve their potential for ART applications in cloned and interspecies intracytoplasmic sperm injection (ICSI) embryos by utilising L-ascorbic acid (LAA) in pre in vitro maturation (pre-IVM) culture. First, oocytes isolated from SAFs were cultured with the duration of pre-IVM 0, 6, 8, 10 h and different concentrations of LAA to determine good conditions for oocyte maturation. Then, mature oocytes were assessed for their developmental competence through parthenogenesis, cloned and interspecies ICSI embryos. The results showed that 8-h pre-IVM with 50 μg/mL LAA improved the maturation rate and developmental competence of parthenogenetic and clone embryos, especially, improving the high blastocyst quality by increasing cell number and expression of histone acetylation at lysine 9 (H3K9ac). In addition, the culture process improved the nuclear reprogramming of somatic cells after nuclear transfer into mature oocytes, resulting in an increased hatching rate of cloned embryos. It also enhanced the activation and the pronuclear formation rate of Gaurus-Taurus zygotes. Overall, the established pre-IVM culture method enhanced the meiotic and developmental competence of embryos. Among vertebrates, obligate parthenogenesis occurs exclusively in squamate reptiles. Premeiotic endoreplication in a small subset of developing oocytes has been documented as the mechanism of production of unreduced eggs in minutely explored obligate parthenogenetic lineages, namely in teiids and geckos. The situation in the lacertid genus Darevskia has been discussed for decades. Certain observations suggested that the ploidy level is restored during egg formation through a fusion of egg and polar body nuclei in Darevskia unisexualis and D. armeniaca . In this study, we re-evaluated the fusion hypothesis by studying diplotene chromosomes in adult females of sexual species D. raddei nairensis and obligate parthenogens D. armeniaca, D. dahli and D. unisexualis . We revealed 19 bivalents in the sexual species and 38 bivalents in the diploid obligate parthenogens, which uncovers premeiotic endoreplication as the mechanism of the production of non-reduced eggs in parthenogenetic females. The earlier contradicting reports can likely be attributed to the difficulty in identifying mispairing of chromosomes in pachytene, and the fact that in parthenogenetic reptiles relying on premeiotic endoreplication only a small subset of developing oocytes undergo genome doubling and overcome the pachytene checkpoint. This study highlights co-option of premeiotic endoreplication for escape from sexual reproduction in all independent hybrid origins of obligate parthenogenesis in vertebrates studied to date.

Autophagy is essential for oocyte maturation and preimplantation embryo development. ATG4C, a member of the ATG4 family, plays a crucial role in the autophagy process. The effect of ATG4C on the early embryonic development in pig has not been studied. In this study, the expression patterns of ATG4C were explored using qRT-PCR and immunofluorescence staining. Different concentrations of serum were added to in vitro maturation (IVM) medium to investigate its effects on oocyte maturation and embryonic development. Finally, the developmental potential of parthenogenetic embryos was detected by downregulating ATG4C in MII stage oocytes under 0 % serum condition. The results revealed that ATG4C was highly expressed in porcine oocytes matured in vitro and in parthenogenetic embryos. Compared with the 10 % serum group, the cumulus cell expansion, first polar body (PB1) extrusion rate, and subsequent developmental competence of embryos were reduced in the 0 % and 5 % serum groups. The mRNA levels of LC3, ATG5, BECLIN1, TFAM, PGC1α, and PINK1 were significantly increased (P < 0.05) in the 0 % serum group. ATG4C was significantly upregulated in the embryos at the 1-cell, 2-cell, 8-cell, and 16-cell stages in the 0 % serum group (P < 0.05). Compared with the negative control group, downregulation of ATG4C significantly decreased the 4-cell, 8-cell, and blastocyst rates (P < 0.05), and the expression of genes related to autophagy, mitochondria, and zygotic genome activation (ZGA) was significantly decreased (P < 0.05). The relative fluorescence intensity of LC3 and mitochondrial content in the ATG4C siRNA group was significantly reduced (P < 0.05). The in vitro maturation (IVM) quality of oocytes is directly related to the subsequent developmental potential of embryos and a fundamental of in vitro embryo production. However, conventional IVM methods fail to maintain the gap-junction intercellular communication (GJIC) between cumulus-oocyte complexes (COCs), which leads to insufficient oocyte maturation. Herein, we investigated the effects of three different three-dimensional (3D) culture methods on oocyte development in vitro, optimized of the alginate-hydrogel embedding method, and assessed the effects of the alginate-hydrogel embedding method on subsequent embryonic developmental potential of oocytes after IVM and parthenogenetic activation (PA). The results showed that Matrigel embedding and alginate-hydrogel embedding benefited the embryonic developmental potential of oocytes after IVM and PA. With the further optimization of alginate-hydrogel embedding, including crosslinking and decrosslinking of parameters, we established a 3D culture system that can significantly increase oocyte maturation and the blastocyst rate of embryos after PA (27.2 ± 1.5 vs 36.7 ± 2.8, P < 0.05). This 3D culture system produced oocytes with markedly increased mitochondrial intensity and membrane potential, which reduced the abnormalities of spindle formation and cortical granule distribution. The alginate-hydrogel embedding system can also remarkably enhance the GJIC between COCs. Many aphid species exhibit both cyclical parthenogenesis (CP) and the obligate parthenogenesis (OP) life history, which are genetically determined. In CP aphid lineages, the parthenogenetic individuals can switch from asexual to sexual reproduction quickly in response to environmental factors such as changes in photoperiod and temperature. However, the OP aphid lineages do not undergo sexual reproduction under any conditions. So far, mechanisms underlying the reproduction switch in CP aphids have not been fully elucidated. Rhopalosiphum padi, a serious worldwide insect pest of wheat, has both CP and OP lineages. Uridine diphosphate-glycosyltransferases (UGTs) are enzymes that participate in the metabolic detoxification of xenobiotics. Here, we identified 43 RpUGT genes from R. padi genome and transcriptome sequences, and found that: (1) the UGT content of the CP lineage was significantly higher than that in the OP lineage at the key time points when CP lineage mainly produce virginoparae, gynoparae, and males under inducing condition, while there were no significant difference under normal conditions; (2) RpUGT344J7 gene was highly expressed during the time points when CP lineages produce gynopara and males; (3) the critical time points for CP lineages to produce virginoparaee, gynoparae, and males were affected when the CP lineages were injected with dsRpUGT344J7; (4) the knockdown of RpUGT344J7 caused a significant reduction in the total number of virginoparae, gynoparae, and males in the offspring under inducing condition. Oak gall wasps typically exhibit a life cycle with one sexual and one asexual generation each year. These wasps can carry various endosymbionts, one of which is the maternally inherited bacterium Wolbachia that can induce several reproductive manipulations on its host. Cytoplasmic incompatibility (CI) has been described as the most prominent of these manipulations. CI leads to embryonic mortality in the hosts' offspring when infected males mate with either uninfected females or with females that harbour different Wolbachia strains. It has been hypothesized that Wolbachia can induce CI in oak gall wasps. To address this hypothesis, we derived a mathematical model to investigate the spread of a bacterial infection in naive populations and to determine the plausibility of CI occurrence. To validate our model, we used published data from Wolbachia-infected Belonocnema kinseyi populations in two approaches. Our first approach uses measurements of infection frequencies and maternal transmission in the sexual generation. For the second approach, we extended the model to compare predictions to estimates of mtDNA-haplotypes, which, like Wolbachia, are maternally inherited, and can therefore be associated with the infection. Both approaches indicate that CI is present in these populations. Trisomy 21 is the most prevalent aneuploidy disorder among live-born children worldwide. Itresults from the presence of an extra copy of chromosome 21 which leads to a wide spectrum ofpathophysiological abnormalities and intellectual disabilities. Nevertheless human chromosome21 (HSA21) possess protein non-coding regions where HAS-21 derived-microRNA genes aretranscribed from. In turn, these HSA21-derived miRNAs curb protein translation of severalgenes which are essential to meet memory and cognitive abilities. From the genetics andmolecular biology standpoints, dissecting the mechanistic relationship between DS pathology/symptoms and five chromosome 21-encoded miRNAs including miR-99a, let-7c, miR-125b-2,miR-155 and miR-802 seems pivotal for unraveling novel therapeutic targets. Recently,several studies have successfully carried out small molecule inhibition of miRNAs function,maturation, and biogenesis. One might assume in the case of DS trisomy, the pharmacologicalinhibition of these five overexpressed miRNAs might open new avenues for amelioration of theDS symptoms and complications. In this review, we primarily elucidated the role of HSA21-encoded miRNAs in the DS pathology which in turn introduced and addressed importanttherapeutic targets. Moreover, we reviewed relevant pharmaceutical efforts that based theirgoals on inhibition of these pathological miRNAs at their different biogenesis steps. We havealso discussed the challenges that undermine and question the reliability of miRNAs as noneinvasivebiomarkers in prenatal diagnosis.

Doubled haploids have been widely used worldwide in breeding programs and fundamental research as valuable homozygous material for about 100 years. The species Cucurbita pepo L. are represented by a huge variety of forms, include highly productive vegetable crops and have a wide distribution in the world. Despite the great economic importance, the creation of effective protocols to ensure stable production of doubled haploids in this species remains an urgent task. DH plants are of interest not only because of the acceleration of the breeding process, but also because of the realization of the huge potential of gametoclonal variability inherent in this highly polymorphic species. In this review, we analyzed the main technologies used for obtaining doubled haploids in vegetable crops of C. pepo: parthenogenesis in situ stimulated by treated/irradiated pollen, gynogenesis in vitro (unpollinated ovule culture in vitro) and androgenesis in vitro (anther/microspore culture in vitro). An analysis is presented of the research carried out from the beginning of the discovery of haploid plants to the current advances and evaluation of the prospects in the field of DH plant production. The main critical factors influencing the efficiency of each technology and its individual steps are considered. The developed technology of doubled haploids obtaining using non-pollinated ovary culture in vitro is presented. This technology allows to obtain up to 55 embryoids per one cultivated ovary (28 embryoids/ 100 cultivated ovules) To introduce haploid technologies into the breeding process it is necessary to evaluate the obtained plants for ploidy level. The use of direct counting of chromosomes in apical cells may present a certain difficulty in this species due to their large number (2n=40) and their small size. Depending on the level of laboratory equipment, ploidy determination using flow cytometry of cell nuclei and counting the number of chloroplasts in stomatal guard cells in the epidermis of the abaxial side of the leaf may be more convenient methods. The prospects for the use of molecular markers for assessment for homozygosity in DH technologies used, including C. pepo, are discussed in the review.

Aggregation of blastomeres is a promising method to improve the developmental competence of blastocysts and may be useful for the production of chimeric animals and the establishment of embryonic stem cell lines by increasing inner cell masses. Here, we determined the optimal conditions for blastomere aggregation using phytohemagglutinin-L (PHA-L) and examined PHA-L efficiency by comparing it with Well of the Well (WOW), a general blastomere aggregation method. As a result, we confirmed that treatment with 15 μg/ml PHA-L for 144 h was effective for blastomere aggregation and embryonic development of three zona-free 2-cell stage embryos (TZ2Es) after parthenogenetic activation (PA). The TZ2Es cultured with PHA-L showed a significantly (p < 0.05) higher blastomere aggregation rate than the WOW method (93.5 ± 1.9% vs. 78.0 ± 8.5%). In addition, our results demonstrated that TZ2Es aggregation through PHA-L improved the quality of PA-derived blastocysts and improved porcine embryonic stem-like cell (pESLCs) seeding efficiency and quality of colonies. It was also observed that PHA-L-derived pESLC could remain undifferentiated and exhibit typical embryonic stem cell pluripotency markers, embryoid body (EB)-forming ability, and differentiation into cell lineages of three germ layers. Pig blastomere aggregation technology is expected to improve embryo quality and the efficiency of embryonic stem cell establishment and embryoid-body formation. It can also be used in blastocyst complementation systems and in the production of chimeric animals.

Public aquaria showcase aquatic life while raising awareness and educating the public on biodiversity and the need for conservation. Recently, aquaria have followed in the footsteps of zoos by taking more directed approaches in species conservation as well as leveraging animals in their collection to fill biological knowledge gaps through research. Similar to zoos, aquaria are able to house animals that are not feasible to care for in traditional academic settings, allowing important life history information to be gained. In particular, our understanding of reproductive aspects of large migratory fishes such as elasmobranchs (sharks, skates and rays) have benefited from information gleaned in aquarium settings that would otherwise be difficult or impossible to study in the natural environment. For example, the ability of elasmobranchs to reproduce parthenogenetically was discovered through observation of sharks maintained in public aquaria. Since its discovery, parthenogenesis has been observed for many ex situ shark and ray species. Aquaria have made other important contributions to characterizing elasmobranch reproductive biology such as understanding reproductive cycling and trialing assisted reproductive techniques, among others. Here, we review the role aquaria have played in the field of elasmobranch reproductive biology and discuss how zoos and aquariums can continue to contribute to this field in the future.

Background: The reproductive cycle of the recently described sponge Tethya meloni was investigated for a period of 15 months (September 2018 – November 2019) in the Mar Piccolo of Taranto (Southern Italy) and was compared with data previously collected for the other two sympatric species of the same genus known for Mediterranean Sea, T. citrina and T. aurantium. Results: T. meloni is a gonochoric species with a sex ratio strongly shifted towards females. Asexual budding was a seasonal process, limited to few specimens. In a specimen collected in September 2018 both oocytes and buds occurred, suggesting that in T. meloni the sexual and asexual phases may coexist both at the population and individual levels. Conclusions: The data obtained from this research compared with the available literature confirm the high temporal variability of the reproductive cycles in the Mediterranean species of Tethya, but with common general characteristics. In sexual reproduction, the oocyte production period lasts several months, with a peak between summer and autumn while spermatogenesis, shorter but with greater reproductive effort, follows the onset of oogenesis. The combination of apomixis and hybrid production is hailed as the holy grail of agriculture for the ability of apomixis to fix heterosis of F1 hybrids in succeeding generations, thereby eliminating the need for repeated crosses to produce F1 hybrids. Apomixis, asexual reproduction through seed, achieves this feat by circumventing two processes that are fundamental to sexual reproduction (meiosis and fertilization) and replacing them with apomeiosis and parthenogenesis, resulting in seeds that are clonal to the maternal parent. Parthenogenesis, embryo development without fertilization, has been genetically engineered in rice, maize, and pearl millet using PsASGRBABY BOOM-like (PsASGR-BBML) transgenes and in rice using the OsBABY BOOM1 (OsBBM1) cDNA sequence when expressed under the control of egg cell-specific promoters. A phylogenetic analysis revealed that BABY BOOM (BBM)/BBML genes from monocots cluster within three different clades. The BBM/BBML genes shown to induce parthenogenesis cluster within clade 1 (the ASGR-BBML clade) along with orthologs from other monocot species, such as Setaria italica. For this study, we tested the parthenogenetic potential of three BBM transgenes from S. italica, each a member of a different phylogenetic BBM clade. All transgenes were genomic constructs under the control of the AtDD45 egg cell-specific promoter. All SiBBM transgenes induced various levels of parthenogenetic embryo development, resulting in viable haploid T1 seedlings. Poor seed set and lower haploid seed production were characteristics of multiple transgenic lines. Modern plant breeding programs rely heavily on the generation of homozygous lines, with the traditional process requiring the inbreeding of a heterozygous cross for five to six generations. Doubled haploid (DH) technology, a process of generating haploid plants from an initial heterozygote, followed by chromosome doubling, reduces the process to two generations. Currently established in vitro methods of haploid induction include androgenesis and gynogenesis, while in vivo methods are based on uniparental genome elimination. Parthenogenesis, embryogenesis from unfertilized egg cells, presents another potential method of haploid induction. PsASGR-BABY BOOMlike, an AP2 transcription factor, induces parthenogenesis in a natural apomictic species, Pennisetum squamulatum (Cenchrus squamulatus) and PsASGR-BBML transgenes promote parthenogenesis in several crop plants, including rice, maize, and pearl millet. The dominant nature of PsASGR-BBML transgenes impedes their use in DH technology. Using a glucocorticoid-based post-translational regulation system and watering with a 100 µM DEX solution before anthesis, PsASGR-BBML can be regulated at the flowering stage to promote parthenogenesis. Conditional expression presents a novel opportunity to use parthenogenetic genes in DH production technology and to elucidate the molecular mechanism underlying parthenogenetic embryogenesis. A cDNA microarray containing 43,661 differentially expressed genes was carried out on the blastoderm of fertilized and facultative parthenogenic turkey embryos at different hours of development. The total number of up-regulated (UR) and down-regulated (DR) genes at 0, 12, and 24 h of development were 725 and 1436, 942 and 599, and 589 and 1044, respectively. Common genes between 0 and 12 h, 12 and 24 h, and 0 and 24 h were 55, 67, and 110, respectively. The proportion of genes showing above 50-fold UR and DR at 0, 12, and 24 h of development were 2.0% and 1.5%, 0.5% and 1.2%, and 0.2% and 1.1%, respectively. Eight UR genes were validated (APOA1 , THRAP3 , ARL14EP , PSAP , MOG , MYBPC2 , MTIF3 and EDG4) and relative expression of six of them was significantly higher (P ≤ 0.05) in parthenogenic embryos, while two genes showed non-significant (P ≥ 0.05) variation. The expression of BCL11A , PRP4B , TCP1 , and TPI1 genes was significantly (P ≤ 0.05) DR in parthenotes in the micro-array study, while the TCP1 gene was up-regulated, and there was no variation in TPI1 gene expression in the PCR validation study. In conclusion, our findings demonstrate differential expression of a large number of genes in parthenotes at different stages of embryo development compared to fertilized embryos. Up-regulation of APOA1, MYBPC2, TCP1, and THRAP3 genes, suggest their crucial role in spontaneous facultative parthenogenic development in turkey birds. • This is the first microarray report for turkey parthenogenetic embryos. • The development of parthenogenetic embryo is comparatively slower than the normally fertilized embryos. • Five genes were found common among the three different stages of parthenogenesis. • Genes including APOA1, MYBPC2, TCP1, and THRAP3 were important for spontaneous facultative parthenogenic embryo development. Asexual reproduction is expected to have a twofold reproductive advantage over sexual reproduction, owing to the cost of producing males in sexual subpopulations. The persistence of sexual females, thus, requires an advantage to sexual reproduction, at least periodically. Here, we tested the hypothesis that asexual females are more sensitive to limited resources. Under this idea, fluctuations in the availability of resources (per capita) could periodically favour sexual females when resources become limited. We combined sexual and asexual freshwater snails (Potamopyrgus antipodarum) together in nylon mesh enclosures at three different densities in an outdoor mesocosm. After 1 month, we counted the brood size of fertile female snails. We found that fecundity declined significantly with increasing density. However, sexual females did not produce more offspring than asexual females at any of the experimental densities. Our results, thus, suggest that the cost of sexual reproduction in P. antipodarum is not ameliorated by periods of intense resource competition. [ABSTRACT FROM AUTHOR]

Sexual reproduction is ubiquitous in the natural world, suggesting that sex must have extensive benefits to overcome the cost of males compared to asexual reproduction. One hypothesized advantage of sex with strong theoretical support is that sex plays a role in removing deleterious mutations from the genome. Theory predicts that transitions to asexuality should lead to the suppression of recombination and segregation and, in turn, weakened natural selection, allowing for the accumulation of slightly deleterious mutations. We tested this prediction by estimating the dN/dS ratios in asexual vertebrate lineages in the genus Aspidoscelis using whole mitochondrial genomes from seven asexual and five sexual species. We found higher dN/dS ratios in asexual Aspidoscelis species, indicating that asexual whiptails accumulate nonsynonymous substitutions due to weaker purifying selection. Additionally, we estimated nucleotide diversity and found that asexuals harbor significantly less diversity. Thus, despite their recent origins, slightly deleterious mutations accumulated rapidly enough in asexual lineages to be detected. We provide empirical evidence to corroborate the connection between asexuality and increased amino acid substitutions in asexual vertebrate lineages. [ABSTRACT FROM AUTHOR]

The current article describes an interesting case of geographic parthenogenesis among European mites from the Labidostommidae family (Acari: Prostigmata). In earlier publications on Labidostomma luteum Kramer, 1879, the authors pointed out that most of the populations of this species in Europe consisted of parthenogenetic females. Until now, populations of both sexes were known only in southwestern France. In the north–west areas, the number of males in the populations has gradually decreased, and in the populations found west of Paris there are no males at all. During the research project carried out in 2018, Błoszyk found the presence of populations which consisted of both sexes of the species in question near the town of Bény (60 km NE of Lion). This is the easternmost site of L. luteum in France where males have been found. All other known sites located east and north of the Bény–Paris line have only parthenogenetic females. Thus, this line can be considered as the boundary of L. luteum populations reproducing sexually. However, establishing the exact course of this range requires further research. The structure of spined loaches diploid-poliploid complex of Cobitis genera representatives as well as Sabanjeewia aurata of the Eastern Ukraine river systems were investigated by allozyme analysis and cytometry. In total 39 samples and 1412 specimens from the upper left tributaries of the Middle Dnipro River, the Donets Basin, the coastal rivers of the Sea of Azov and the Lower Dnipro were studied. We identified the representatives of C. (superspecies taenia) (51.6 % of all researched specimens). C. melanoleuca (4.1 %), S. aurata (1.1 %), allodiploids C. (superspecies taenia) × C. melanoleuca (0.3 %) and two groups of polyploid hybrids (46.4 %). EET-group includes mainly C. elongatoides-2 taenia and C. elongatoides-taenia -tanaitica. EET-group (2.8 %) is represented only by C. elongatoides-2 tanaitica biotype. In the Lower Donets and the Lower Dnipro, there is a zone of introgressive hybridization between C. taenia and C. tanaitica. The structure of settlements differs accordingly to the river basins and has a tendency to decreasing of polyploids frequency respetivly to meridional direction. The analysis of loaches samples from the coastal rivers of the Sea of Azov collected in 1938 and samples from the Middle part of the Donets River in 1971 leads to the conclusion that polyploids in mentioned above regions appeared in 1960-1970s. The structure of diploid-polyploid settlements has an alternative character. More often, there is a predominance of diploids or polyploids. The reason of such bipolarity of settlements could be as reproductive displacement so a relatively strong tolerance of polyploids to anthropogenic climate change. [ABSTRACT FROM AUTHOR]

BACKGROUND: Thelytokous Wolbachia‐infected Trichogramma wasps are superior to bisexual uninfected wasps regarding biological control programs. However, continuous oviposition weakens the parthenogenesis‐inducing (PI) strength of Wolbachia. Whether this reduced PI strength relates to decreases in the titer of Wolbachia in the ovary and offspring eggs of Trichogramma remains unclear. Here, using fluorescence in situ hybridization (FISH) and reverse transcription quantitative polymerase chain reaction (RT‐qPCR) methods, we investigated how the penetrance of Wolbachia‐mediated parthenogenesis, Wolbachia density, and distributions of two Wolbachia‐infected Trichogramma species, T. pretiosum (TP) and T. dendrolimi (TD), were influenced by different host access treatments [newly‐emerged virgin females (NE), 7‐day‐old females without access to host eggs (NAH), and 7‐day‐old virgin females with access to host eggs (AH)]. RESULTS: Continuous oviposition decreased Wolbachia PI strength and titers in TP and TD. Continuous oviposition in AH decreased Wolbachia titers in abdomen and offspring eggs of TP and TD females, compared with NAH and NE; NAH had a lower thorax Wolbachia titer than NE. The numbers of parasitized host eggs and offspring wasps, and emergence rates of offspring deposited by AH were lower than those of NE and NAH, for either species. CONCLUSION: Weakened PI strength, driven by continuous oviposition in Trichogramma wasps, is associated with Wolbachia titer exhaustion in ovary and offspring eggs. Wolbachia density is dependent on PI strength in Trichogramma wasps, highlighting the side effects of continuous oviposition regarding thelytokous Wolbachia‐infected Trichogramma in biological control programs. © 2022 Society of Chemical Industry. The BABY BOOM (BBM) AINTEGUMENTA-LIKE (AIL) AP2/ERF domain transcription factor is a major regulator of plant cell totipotency, as it induces asexual embryo formation when ectopically expressed. Surprisingly, only limited information is available on the role of BBM during zygotic embryogenesis. Here we reexamined BBM expression and function in the model plant Arabidopsis thaliana (Arabidopsis) using reporter analysis and newly developed CRISPR mutants. BBM was expressed in the embryo from the zygote stage and also in the maternal (nucellus) and filial (endosperm) seed tissues. Analysis of CRISPR mutant alleles for BBM (bbm-cr) and the redundantly acting AIL gene PLETHORA2 (PLT2) (plt2-cr) uncovered individual roles for these genes in the timing of embryo progression. We also identified redundant roles for BBM and PLT2 in endosperm proliferation and cellularization and the maintenance of zygotic embryo development. Finally, we show that ectopic BBM expression in the egg cell of Arabidopsis and the dicot crops Brassica napus and Solanum lycopersicon is sufficient to bypass the fertilization requirement for embryo development. Together these results highlight roles for BBM and PLT2 in seed development and demonstrate the utility of BBM genes for engineering asexual embryo development in dicot species. Intracellular bacteria of the genus Wolbachia are the most abundant endosymbionts infecting many arthropods, with Coleoptera being the most diverse hosts in terms of taxonomy and ecology. There has been great progress in studies on the relations between Wolbachia and beetles, however, only some of the research details the consequences of infection. In this review, I summarise the knowledge on the evolutionary relations or ecological associations between Wolbachia and its beetle hosts. These bacteria often cause cytoplasmic incompatibility in the infected hosts and are responsible for a selective sweep of the mitochondrial genomes in some beetles. Wolbachia can manipulate the sex ratio or reproduction of some species of beetles, however, it does not induce parthenogenesis, with the possible rare exception of some Naupactini. Proof of the co-evolution of Wolbachia with beetles is missing, but some aquatic groups seem to be prone to co-speciation, unlike terrestrial taxa. On the other hand, there is a growing number of studies indicating or proving horizontal transmission of Wolbachia among beetle hosts, mostly via common host plants or the foraging substrate (such as dung). Wolbachia is not alone in infecting beetles as other endosymbiotic bacteria occur in beetles (Rickettsia, Spiroplasma, Cardinium, Arsenophorus), which have often been reported as interchangeable, suggesting the infection by various bacteria is dynamic. Nonetheless, there are still many issues associated with Wolbachia that are not yet been described in beetles (like the provision of nutrition or protection against pathogens) and high-throughput sequencing should be used to improve our understanding of Wolbachia-Coleoptera relations. Plasmodium parasites have a complex life cycle that includes development in the human host as well as the Anopheles vector. Successful transmission of the parasite between its host and vector therefore requires the parasite to balance its investments in asexual replication and sexual reproduction, varying the frequency of sexual commitment to persist within the human host and generate future opportunities for transmission. The transmission window is extended further by the ability of stage V gametocytes to circulate in peripheral blood for weeks, whereas immature stage I to IV gametocytes sequester in the bone marrow and spleen until final maturation. Due to the low gametocyte numbers in blood circulation and with the ease of targeting such life cycle bottlenecks, transmission represents an efficient target for therapeutic intervention. The biological process of Plasmodium transmission is a multistage, multifaceted process and the past decade has seen a much deeper understanding of the molecular mechanisms and regulators involved. Clearly, specific and divergent processes are used during transmission compared to asexual proliferation, which both poses challenges but also opportunities for discovery of transmission-blocking antimalarials. This review therefore presents an update of our molecular understanding of gametocyte and gamete biology as well as the status of transmission-blocking activities of current antimalarials and lead development compounds. By defining the biological components associated with transmission, considerations for the development of new transmission-blocking drugs to target such untapped but unique biology is suggested as an important, main driver for transmission-blocking drug discovery. The rarity of parthenogenetic species is typically attributed to the reduced genetic variability that accompanies the absence of sex, yet natural parthenogens can be surprisingly successful. Ecological success is often proposed to derive from hybridization through enhanced genetic diversity from repetitive origins or enhanced phenotypic breadth from heterosis. Here, we tested and rejected both hypotheses in a classic parthenogen, the diploid grasshopper Warramaba virgo. Genetic data revealed a single hybrid mating origin at least 0.25 million years ago, and comparative analyses of 14 physiological and life history traits showed no evidence for altered fitness relative to its sexual progenitors. Sex is evolutionarily more costly than parthenogenesis, evolutionary ecologists therefore wonder why sex is much more frequent than parthenogenesis in the majority of animal lineages. Intriguingly, parthenogenetic individuals and species are as common as or even more common than sexuals in some major and putative ancient animal lineages such as oribatid mites and rotifers. Here, we analyzed oribatid mites (Acari: Oribatida) as a model group because these mites are ancient (early Paleozoic), widely distributed around the globe, and include a high number of parthenogenetic species, which often co‐exist with sexual oribatid mite species. There is evidence that the reproductive mode is phylogenetically conserved in oribatid mites, which makes them an ideal model to test hypotheses on the relationship between reproductive mode and species' ecological strategies. We used oribatid mites to test the frozen niche variation hypothesis; we hypothesized that parthenogenetic oribatid mites occupy narrow specialized ecological niches. We used the geographic range of species as a proxy for specialization as specialized species typically do have narrower geographic ranges than generalistic species. After correcting for phylogenetic signal in reproductive mode and demonstrating that geographic range size has no phylogenetic signal, we found that parthenogenetic lineages have a higher probability to have broader geographic ranges than sexual species arguing against the frozen niche variation hypothesis. Rather, the results suggest that parthenogenetic oribatid mite species are more generalistic than sexual species supporting the general‐purpose genotype hypothesis. The reason why parthenogenetic oribatid mite species are generalists with wide geographic range sizes might be that they are of ancient origin reflecting that they adapted to varying environmental conditions during evolutionary history. Overall, our findings indicate that parthenogenetic oribatid mite species possess a widely adapted general‐purpose genotype and therefore might be viewed as "Jack‐of‐all‐trades." We genotyped multiple populations of all seven parthenogenetic species of rock lizards, genus Darevskia , as well as their putative sexual parental populations, using double digest RAD-sequencing genomic markers. Taking advantage of the conserved homology of the ZW/ZZ sex chromosomes among lacertid lizards, we aligned our short sequence reads to a reference Z chromosome assembly of the common wall lizard Podarcis muralis. This provided unique insight into the origin of all-female hybrid populations, which inherited their single Z chromosome exclusively from a paternal ancestor. The mapped Z-linked loci were used to construct a robust time-calibrated phylogeny. In each parent–offspring species pair, the geographically nearest population of the paternal species was identified as the most likely ancestor of the respective parthenogen, mirroring the trend observed previously on the maternal side in mitochondrial DNA. The estimated splits between the sampled paternal sexual ancestors and their daughter species occurred much earlier than suggested previously and during two narrow time periods: (1) the parthenogenetic Darevskia armeniaca , D. dahli , D. uzzelli and D. rostombekowi dated back to ~0.5 or ~0.9 Mya, depending on the calibration point used, while (2) D. bendimahiensis , D. sapphirina and D. unisexualis appear to have diverged ~1 or ~2 Mya. Simple Summary: The American cockroach is a widely distributed sanitary pest closely related to human life. The American cockroach is not only a hygienic pest that we all know but also beneficial to humans as its extract can be used medicinally and could be a model organism for physiology and neuroscience studies. In this study, we provide a life table of the American cockroach in a stable environment, including embryonic development, nymphal instars, and adult reproduction. Newly laid eggs hatch into nymphs after about 35 days of embryonic development. Under sufficient materials and space, gregarious nymphs undergo 14 molts before transforming into adults. Adult females can produce fertile offspring whether they have mated or not. On average, mated females produce an ootheca every 4 days, while unmated females produce an ootheca every 10 days. Each ootheca contains 12–16 eggs. Additionally, group living seems to improve the survival rate of offspring of unmated females. The American cockroach, Periplaneta americana (Insecta: Blattodea: Solumblattodea: Blattidae), is an urban hygiene pest but also a model organism for physiology and neuroscience study. However, the current description of the developmental process of the American cockroach is insufficient. In this study, we provide a life table of the American cockroach in a stable environment, including embryonic development, nymphal instars and adult reproduction. Our results show that there are 14 nymphal instars of the American cockroach in groups with sufficient living materials and space. The secondary sexual characteristics are evident in last-instar nymphs and adults, namely, the complete absence of the anal stylus in females. The entire embryonic development process was divided into 20 stages on the basis of lateral-view observations of the embryos. The formation of the embryo involves the fusion of paired blastoderm regions with higher cellular density, similar to that in other insects of Polyneoptera. With respect to reproduction, the gamogenetic females produced their first ootheca earlier than the parthenogenic females, and the frequency of oviposition was higher for the former throughout adulthood. Simple Summary: Sexual reproduction is dominant in animals, while asexual lineages are rare and evolutionarily short-lived. However, sexual reproduction has substantial costs, such as male production, inputs to courtship and mating, increased risk of predator exposure, and sexually transmitted diseases. A large body of theories has been proposed to explain the paradox of sex. One favored explanation is that asexuals are more likely to accumulate a greater number of deleterious mutations, known as Muller's ratchet. Trichogramma is a genus of egg parasitoid wasps and is widely used as a biological control agent for agricultural and forest pests. With asexual lineages in at least 16 species, Trichogramma provides an excellent model to investigate the causes and consequences of asexual reproduction. In this study, we sequenced and assembled two asexual Trichogramma mitogenomes, representing two divergent origins of asexual reproduction. The asexual T. pretiosum is induced by the endosymbiont Wolbachia, while T. cacoeciae presumably originates from interspecific hybridization. To test Muller's ratchet hypothesis, we compared these two asexual mitogenomes with their sexual relatives and found no association between asexual reproduction and mutation accumulation. This study provides a basis for further investigation into mitochondrial evolution and asexual reproduction in Trichogramma. Despite its substantial costs, sexual reproduction dominates in animals. One popular explanation for the paradox of sex is that asexual reproduction is more likely to accumulate deleterious mutations than sexual reproduction. To test this hypothesis, we compared the mitogenomes of two asexual wasp strains, Trichogramma cacoeciae and T. pretiosum, to their sexual relatives. These two asexual strains represent two different transition mechanisms in Trichogramma from sexual to asexual reproduction. Asexual T. pretiosum is induced by Wolbachia, while T. cacoeciae presumably originated from interspecific hybridization. We sequenced and assembled complete mitochondrial genomes of asexual T. cacoeciae and T. pretiosum. Compared to four sexual relatives, we found no evidence of higher mutation accumulation in asexual Trichogramma mitogenomes than in their sexual relatives. We also did not detect any relaxed selection in asexual Trichogramma mitogenomes. In contrast, the intensified selection was detected in Nad1 and Nad4 of the asexual T. pretiosum mitogenome, suggesting more purifying selection. In summary, no higher mitochondrial mutation accumulation was detected in these two asexual Trichogramma strains. The completion-of-tumor hypothesis involved in the dynamic interplay between the initiating oncogenic event and progression is essential to better recognize the foundational framework of tumors. Here we review and extend the gametogenesis-related hypothesis of tumors, because high embryonic/germ cell traits are common in tumors. The century-old gametogenesis-related hypothesis of tumors postulated that tumors arise from displaced/activated trophoblasts, displaced (lost) germ cells, and the reprogramming/reactivation of gametogenic program in somatic cells. Early primordial germ cells (PGCs), embryonic stem (ES) cells, embryonic germ cells (EGCs), and pre-implantation embryos at the stage from two-cell stage to blastocysts originating from fertilization or parthenogenesis have the potential to develop teratomas/teratocarcinomas. In addition, the teratomas/teratocarcinomas/germ cells occur in gonads and extra-gonads. Undoubtedly, the findings provide strong support for the hypothesis. However, it was thought that these tumor types were an exception rather than verification. In fact, there are extensive similarities between somatic tumor types and embryonic/germ cell development, such as antigens, migration, invasion, and immune escape. It was documented that embryonic/germ cell genes play crucial roles in tumor behaviors, e.g. tumor initiation and metastasis. Of note, embryonic/germ cell-like tumor cells at different developmental stages including PGC and oocyte to the early embryo-like stage were identified in diverse tumor types by our group. These embryonic/germ cell-like cancer cells resemble the natural embryonic/germ cells in morphology, gene expression, the capability of teratoma formation, and the ability to undergo the process of oocyte maturation and parthenogenesis. These embryonic/germ cell-like cancer cells are derived from somatic cells and contribute to tumor formation, metastasis, and drug resistance, establishing asexual meiotic embryonic life cycle. p53 inhibits the reactivation of embryonic/germ cell state in somatic cells and oocyte-like cell maturation. Based on earlier and our recent studies, we propose a novel model to complete the gametogenesis-related hypothesis of tumors, which can be applied to certain somatic tumors. That is, tumors tend to establish a somatic asexual meiotic embryonic cycle through the activation of somatic female gametogenesis and parthenogenesis in somatic tumor cells during the tumor progression, thus passing on corresponding embryonic/germ cell traits leading to the malignant behaviors and enhancing the cells' independence. This concept may be instrumental to better understand the nature and evolution of tumors. We rationalize that targeting the key events of somatic pregnancy is likely a better therapeutic strategy for cancer treatment than directly targeting cell mitotic proliferation, especially for those tumors with p53 inactivation. The scarcity of asexual reproduction in vertebrates alludes to an inherent cost. Several groups of asexual vertebrates exhibit lower endurance capacity (a trait predominantly sourced by mitochondrial respiration) compared with congeneric sexual species. Here we measure endurance capacity in five species of Aspidoscelis lizards and examine mitochondrial respiration between sexual and asexual species using mitochondrial respirometry. Our results show reduced endurance capacity, reduced mitochondrial respiration, and reduced phenotypic variability in asexual species compared with parental sexual species, along with a positive relationship between endurance capacity and mitochondrial respiration. Simple Summary: Cactus is an important drought-tolerant crop that has very various beneficial uses. In Morocco, cactus is a perfectly appropriate crop for land rehabilitation in the arid and semi-arid regions. With little investment, it can produce important resources for human and animal nutrition and generate adequate revenues for farmers. Unfortunately, the sustainability of this extremely resilient ecosystem has become seriously threatened by the appearance of Dactylopius opuntiae (Hemiptera: Dactylopiidae). This cochineal which is specific to cacti, was introduced in Morocco in 2014. The aim of this study was to evaluate the effect of temperature on life cycle parameters of D. opuntiae at 20, 23, 26, 32, and 40 ± 1 °C, 65 ± 5% RH, and a photoperiod of 12L:8D. Temperatures ranging from 26 °C to 32 °C were appropriate for the survival, development, and reproduction of the scale insect. Parthenogenesis in females was not observed during our study. This study also showed that D. opuntiae females required a higher thermal constant (769.23 D°) than males (357.14 D°) to grow to the mature adult stage. The effect of temperature on Dactylopius opuntiae (Cockerell) life cycle parameters was evaluated at 20, 23, 26, 32, and 40 ± 1 °C, 65 ± 5% RH, and a photoperiod of 12 L:8 D. Temperatures ranging from 26 °C to 32 °C were suitable for survival, development, and reproduction of D. opuntiae. The total developmental time of females ranged from 94.23 d (20 °C) to 43.55 d (40 °C). The average development time of males from egg to death ranged from 26.97 days at 32 °C to 50.75 days at 20 °C. The probability that a newly laid egg would survive to the adult stage was highest at 26 °C and 32 °C (44–60%). The parthenogenesis in females was not observed during our study. The longest oviposition period was observed when the cochineal was reared at 32 °C (17.97 days), and the highest fecundity was observed at 32 °C (355.29 egg/female). The highest proportion of females (0.80) was observed at 40 °C. According to the age-stage-two-sex life table, the highest value of the intrinsic rate of natural increase (rm) was recorded at 32 °C. The lower developmental thresholds for the total pre-adult female and male and adult female and male stages, were 10.15, 12.21, 10.54, and 21.04 °C, respectively. Dactylopius opuntiae females needed a higher thermal constant (769.23 D°) than males (357.14 D°) to achieve their development and reach the mature adult stage. Hybridization is a common evolutionary process with multiple possible outcomes. In vertebrates, interspecific hybridization has repeatedly generated parthenogenetic hybrid species. However, it is unknown whether the generation of parthenogenetic hybrids is a rare outcome of frequent hybridization between sexual species within a genus or the typical outcome of rare hybridization events. Darevskia is a genus of rock lizards with both hybrid parthenogenetic and sexual species. Using capture sequencing, we estimate phylogenetic relationships and gene flow among the sexual species, to determine how introgressive hybridization relates to the origins of parthenogenetic hybrids. We find evidence for widespread hybridization with gene flow, both between recently diverged species and deep branches. Surprisingly, we find no signal of gene flow between parental species of the parthenogenetic hybrids, suggesting that the parental pairs were either reproductively or geographically isolated early in their divergence. The generation of parthenogenetic hybrids in Darevskia is, then, a rare outcome of the total occurrence of hybridization within the genus, but the typical outcome when specific species pairs hybridize. Our results question the conventional view that parthenogenetic lineages are generated by hybridization in a window of divergence.
