The biological effects of puerarin, the main endogenous component in Radix Puerariae, were explored to provide a scientific basis for the safe use of Radix Puerariae.Twenty Balb/C healthy mice were randomly divided into two groups according to body weight, namely the control group and the medication group,with 10 mice in each group,half male and half female.The mice in the medication group were intragastrically administrated with puerarin at a dose of 6000 mg• kg-1 once,and the mice in the control group were intragastrically administrated with PEG 400 in the same volume.After 7 days of continuous observation, the general activity status, body weight and death of mice were recorded daily. Seven days later, the mice were sacrificed after blood was taken from the orbital vein. The heart, liver and kidney of the mice were anatomically observed and HE staining was performed for histopathological study. The results showed that there was no significant difference in the body weight change and heart, liver and kidney indexes between the male and female puerarin mice and the control group CP>0.05).Serum urea( UREA) level in puerarin group was significantly higher than that in control group (P<0.05); The levels of lactate dehydrogenase(LDH) in serum of male and female mice and hydroxybutyrate dehydrogenase(HBDH) in serum of male rats in puerarin group were significantly lower than those in the control group (P<0.05). His topathological studies showed that puerarin had little effect on the liver of mice,while myocardial dissolution and focal renal tubulointerstitial hemorrhage were observed in mice. Sediment samples from the Gomti River basin were investigated to determine and evaluate trace metal concentrations, their biological effect, and potential ecological and human health risks for adults and children. The mean concentrations of trace metals were organized in the descending order of mercury (Hg) (0.08 mg/kg) > chromium (Cr) (0.06 mg/kg) > lead (Pb) (0.05 mg/kg) > arsenic (As) (0.02 mg/kg) > cadmium (Cd) (0.01 mg/kg). The current study illustrated that metals were attributed to the area from natural sources and different anthropogenic sources especially from industries. However, the concentration levels were lower than the sediment quality guidelines (SQGs) based on the effect-range classifications of threshold effect concentration (TEC) and probable effect concentration (PEC). Therefore, the concentration of the elements showed no adverse biological effects on aquatic organisms. The evaluated potential ecological risk index also revealed the low toxicity to the aquatic environment. Moreover, the applied sediment quality indices, geo-accumulation index (I geo), contamination factor (CF), and contamination severity index (CSI) suggested that the contamination levels of the elements were in the acceptable range, and the contamination had not notably impacted on the sediment quality. The risk index (HI) concerning age groups was significantly less than the threshold limit of 1 indicating that the contamination had no non-carcinogenic risk effect. The total carcinogenic risk (TCR) was less than a risk value of 1 × 10−4. Hence, the current study suggests that immediate remediation is not required due to an absence of alarming conditions in the study area. The application of terahertz (THz) technology has expanded gradually in this century. Studies on the usages of THz imaging and sensing in medical, military, and security areas are going deeper and deeper. In order to use THz technology safely and effectively, especially in the fields of diagnosis, live imaging, therapy, and security screening, it is of great urgency to identify its bioeffects and dose-effect relationship, and then injury threshold so as to study and evaluate the safety and bioeffects of THz radiation. Therefore, the research on THz bioeffects becomes a hotspot in our field, and is the precondition and prerequisite for the studies on the THz application and the knowledge of interaction between THz wave and bio-systems, as well as on founding a basis for its future application and development. Moreover, the researches on THz bioeffects are essential for evaluating health hazards, establishing safety standards, and using THz technology safely. This article reviews the progress and development of THz bioeffects in thermal effects and non-thermal effects, and then, summarizes and analyzes the enlightenment of the researches, and look forward to the future development.

Chronic kidney injury has gradually become a worldwide public health problem currently affecting approximately 10% of the population and can eventually progress to chronic end-stage renal disease characteristic by the result of epithelial atrophy. Interleukin-22 (IL-22) is a cytokine produced by activated immune cells, while acting mainly on epithelial cells ranging from innate immune response to tissue regeneration to maintain barrier integrity and promote wound healing. Accumulating data suggests that IL-22 has emerged as a fundamental mediator of epithelial homeostasis in the kidney through promoting tissue repair and regeneration, inhibiting oxidative stress, and producing antimicrobial peptides. Binding of IL-22 to its transmembrane receptor complex triggers janus kinase/tyrosine kinase 2 phosphorylation, which further activates a number of downstream cascades, including signal transducer and activator of transcription 3, MAP kinase, and protein kinase B, and initiates a wide array of downstream effects. However, the activation of the IL-22 signaling pathways promotes the activation of complement systems and enhances the infiltration of chemokines, which does harm to the kidney and may finally result in chronic renal failure of different autoimmune kidney diseases, including lupus nephritis, and IgA nephropathy. This review describes current knowledge of the basic features of IL-22, including structure, cellular origin and associated signaling pathways. Also, we summarize the latest progress in understanding the physiological and pathological effects of IL-22 in the kidney, suggesting the potential strategies for the specific application of this cytokine in the treatment of kidney disease.

The purpose of this study was to investigate the potential chiral recognition functions induced by two kinds of novel chiral microenvironment-responsive mesoporous silica nanoparticles (CMR-L-MSN and CMR-D-MSN) for the oral delivery of the poorly water-soluble achiral drug indomethacin (IMC). The as-synthesized CMR-L-MSN and CMR-D-MSN with molecular chirality and high specific surface area (SBET: 1141 and 1075 m2/g, respectively) were successfully prepared through grafting chiral molecular functional groups. The characterization results showed that CMR-L-MSN and CMR-D-MSN were spherical nanoparticles with opposite chiral features and clearly visible pore channels. With the rapid development of ultra-high-voltage (UHV) direct-current (DC) transmissions, the impact of static electric fields (SEF) in the vicinity of overhead UHV DC transmission lines on health has aroused much public concern. This study explored the effects of 56.3kV/m SEF on the spleen of mice. Results showed that SEF exposure of 21days significantly increased malonic dialdehyde content, superoxide dismutase activity, calcineurin activity, nitric oxide synthase (NOS) activity, and the mRNA expression levels of tumor necrosis factor-α (TNF-α) and nuclear factor-κB (NF-κB) in the spleen and caused the separation of nucleus and nuclear membrane, the disappearance of mitochondrial membrane, and the deficiency of mitochondrial cristae in splenic lymphocytes. By analysis and discussion, it was deduced that SEF could induce oxidative stress of the spleen by increasing the activity of NOS. Oxidative stress could further cause ultrastructural changes of splenic lymphocytes. Anxiety about potential health hazards of electromagnetic exposure has been growing in the past decades, with their widely application in many fields. The immune system plays pivotal role in maintaining body's homeostasis. Importantly, immune system is also a sensitive target for electromagnetic fields. In recent years, the biological effects of electromagnetic fields on immune cells have been attracting more and more attentions. Accumulated data suggested that electromagnetic exposure could affect the number and function of immune cells to some extent, including cell proportion, cell cycle, apoptosis, killing activity, cytokines contents and so on. The research objects basically covered all types of immune cells, mainly on PBMC, T lymphocytes, B lymphocytes, NK cells and macrophages. Meanwhile, there also are negative reports of electromagnetic fields on immune cells. This article reviews the results of epidemiological investigation, the progresses in animal studies and in vitro experiments, and the current attempts to explore potential mechanisms. Knowledge of the biological effects on immune cells associated with electromagnetic fields is critical for proper health hazard evaluation, development of safety standards, and safe exploitation of new electromagnetic devices and applications. This study investigates the role of age and sex on the cardiovascular effects of 3.5-MHz pulsed ultrasound (US) in a rat model. Ultrasonic bursts of 2.0-MPa peak rarefactional pressure amplitude (equivalent to an in vitro spatial-peak temporal-peak intensity of ~270 W/cm2 and a mechanical index of 1.1) were delivered in five consecutive 10-s intervals, one interval for each pulse repetition frequency (PRF) (6, 5, 4, 5, and 6 Hz; always the same order) for a total exposure duration of 50 consecutive seconds. Sixty F344 rats were split into 12 groups in a $3\times 2\times2$ factorial design (three ages, male versus female, and US application versus control). This study is the first study on US-induced cardiac effects that contains data across three age groups of rats (premenopause, fertile, and postmenopause) to mimic the fertile and nonfertile human window. US was applied transthoracically, while heart rate, stroke volume, ejection fraction, temperature, and other physiologic parameters were recorded at baseline and after exposure. Significant decreases in cardiac output compared to respective control groups were observed in multiple experimental groups, spanning both females and males. A negative chronotropic effect was observed in young male (~7%) and female (~16%) rats, in five-month-old male (~9%) and female (~15%) rats, and in old rats where the effect was not statistically significant. Younger groups and, to a lesser extent, lower weight groups generally had more significant effects. The aim of this study was to compare the local and systemic effects of current pulp capping materials containing resin with those of traditional materials in an animal study. A total of 48 rats were used: a control group (n=12) (sub-control and negative control), a resin-containing group (n=18) (Calcimol LC, Theracal LC, Activa--BioActive Base/Liner), and a traditional group (n=18) (Biodentine, ProRoot MTA, Dycal). The materials which had been placed in polyethylene tubes were implanted in subcutaneous pockets. The rats were sacrificed at 1, 2, or 4 weeks. Evaluations were made of subcutaneous connective tissue, the left kidney, liver, and blood samples. Of all the study groups, MTA demonstrated biocompatibility at a level close to that of the control groups. Inflammation was observed to be more severe in resin-containing materials, but Activa Base/Liner showed a more successful local and systemic tissue response. [ABSTRACT FROM AUTHOR]

Common fragranced consumer products, such as cleaning supplies and personal care products, emit chiral compounds such as limonene that have been associated with adverse effects on human health. However, those same compounds abound in nature, and at similar concentrations as in products, but without the same apparent adverse human health effects. We investigated whether different types of limonene may elicit different biological effects. In this study, we investigated the mortality rate of mosquito larvae in response to changes in their environment. Specifically, we tested different sources of naturally occurring R-limonene and chemically synthetized limonene, containing one of its enantiomeric forms (R-, S-) in mortality bioassays with Aedes aegypti mosquito larvae. We found that a natural source of limonene extracted from oranges induced lower mortality of mosquito larvae compared to synthetic sources at the same concentration. However, enantiomeric forms did not differ in their effects on mortality. Chitosan-based nanofibers (CS-NFs) are excellent artificial extracellular matrices (ECMs) due to the resemblance of CS with the glycosaminoglycans of the natural ECMs. Despite this excellent feature, the poor electrospinnability and mechanical properties of CS are responsible for important limitations in respect to its biomedical applications. To improve the CS's physico-chemical properties, new bioactive and biomimetic CS-NFs were formulated with polyethylene oxide (PEO), having incorporated different active components (ACs) with important beneficial effects for healing. Manuka honey (trophic and antimicrobial effects), propolis (antimicrobial effects), Calendula officinalis infusion (antioxidant effect, reepithelialization stimulating agent), insulin (trophic effect), and L-arginine (angiogenic effect) were selected as ACs. SEM morphology analysis revealed well-alignment, unidirectional arrays, with small diameters, no beads, and smooth surfaces for developed CS_PEO-ACs NFs. The developed NFs showed good biodegradability (NFs mats lost up to 60% of their initial weight in PBS), increased hemocompatibility (hemolytic index less than 4%), and a reduced cytotoxicity degree (cell viability degree more than 90%). In addition, significant antioxidant and antimicrobial effects were noted for the developed NFs which make them suitable for chronic wounds, due to the role of oxidative stress and infection risk in delaying normal wound healing. The most suitable for wound healing applications seems to be CS_PEO@P_C which showed an improved hemolysis index (2.92 ± 0.16%), is non-toxic (cell viability degree more than 97%), and has also significant radical scavenging effect (DPPH inhibition more than 65%). In addition, CS_PEO@P_C presents increased antimicrobial effects, more noticeably for Staphylococcus aureus strain, which is a key feature in preventing wound infection and delaying the healing process. Açaí (E. oleracea) and juçara (E. edulis) are berries considered a source of bioactive compounds, especially anthocyanins and unsaturated fatty acids, with recognized health-promoting activities. An integrative review was conducted to identify available clinical trials that evaluated the effects of açaí and juçara intake on the human organism. Science Direct and Medline databases were searched. Human studies that evaluated any biological activities after açaí and juçara intake were included in this review. Twenty-three clinical trials were identified up to April 12, 2020. Studies evaluated the biological effects of açaí (n = 17), juçara (n = 5), or both berries simultaneously (n = 1). The results of these trials suggest both types of berries may contribute to improved antioxidant defense and to attenuating metabolic stress and inflammation. However, considerable heterogeneity was observed among trials, and few studies explored the bioactive compounds of the food matrix provided in the interventions. The skin protects our body from various external factors, such as chemical and physical stimuli, microorganisms, and sunlight. Sunlight is a representative environmental factor that considerably influences the physiological activity of our bodies. The molecular mechanisms and detrimental effects of ultraviolet rays (UVR) on skin have been thoroughly investigated. Chronic exposure to UVR generally causes skin damage and eventually induces wrinkle formation and reduced elasticity of the skin. Several studies have shown that infrared rays (IR) also lead to the breakdown of collagen fibers in the skin. However, several reports have demonstrated that the appropriate use of UVR or IR can have beneficial effects on skin-related diseases. Additionally, it has been revealed that visible light of different wavelengths has various biological effects on the skin. Interestingly, several recent studies have reported that photoreceptors are also expressed in the skin, similar to those in the eyes. Based on these data, I discuss the various physiological effects of sunlight on the skin and provide insights on the use of phototherapy, which uses a specific wavelength of sunlight as a non-invasive method, to improve skin-related disorders.

Robust evidence confirms that occupational exposures to organic solvents are ototoxic and can aggravate the effects of noise on the auditory organ. Accordingly, Directive 2003/10/EC of the European Parliament and of the Council recommends that in assessing the risk of occupational hearing loss, the interaction of noise and organic solvents to hearing should be taken into account. Given that no prophylactic recommendations have been established to date, this study is intended to provide physicians with guidance in conducting audiometric tests in workers exposed to noise and organic solvents. The paper discusses the biological effects of noise and organic solvents on the auditory organ and current possibilities of estimating the risk of hearing loss due to isolated exposures to solvents, as well as combined exposures to these chemicals and noise. The scope of medical examination and the significance of individual hearing tests, such as pure-tone audiometry, impedance audiometry, auditory brainstem responses, speech in quiet and speech in noise audiometry, and dichotic digit test, in the diagnostics of solvent-induced hearing loss are presented. An algorithm for conducting hearing tests in workers exposed to organic solvents, and co-exposed to noise and organic solvents, has been proposed depending on the concentration of solvents and noise level. Shown also is the necessity to perform annual audiometric testing in all workers exposed to organic solvents whose concentration in the air exceeds values safe for hearing. In workers exposed to noise and organic solvents, audiometric examinations should be mandatory in all subjects in whom the level of noise exceeds the lower action level defined by Directive 2003/10/EC, which is L EX,8h = 80 dB. Taking into account the detrimental effects of organic solvents to central auditory pathways, in addition to pure-tone audiometry, speech audiometry (preferentially speech in noise) should be included. The introduction of preventive audiometric tests in workers exposed to organic solvents, along with an extension of indications and the scope of hearing tests in workers co-exposed to noise and solvents, is a prerequisite for effective prevention of occupational hearing loss in these professional groups. Amniotic fluid (AF) is essential for fetal development and maturation during pregnancy. The levels of proteins in AF have been determined in many studies to screen for potential biomarkers of pregnancy-associated abnormalities. Alpha-fetoprotein (AFP) is a major AF and plasma protein produced by the yolk sac and the liver during the fetal period. APF serum concentrations are commonly used for screening of many syndromes. Electromagnetic fields (EMFs) were reported to change gene expression in the embryo and in adults. The aim of this research was to assess the effects of 50hertz/1millitesla EMFs on AFP expression in the AF in the mouse embryo. AF samples were obtained from pregnant mice in gestational days 16 and 18. AFP relative expression was studied by western blotting. The results of this study showed that AFP relative expression increases in the EMF-treated AF as compared with either SHAM or control groups. It is concluded that EMF increases the AFP relative expression in the AF. It is also suggested that EMF may change the AFP expression in the AF by altering the expression of genes, including AFP, and/or by affecting the permeability of blood barriers.

Electromagnetic pollution of the environment has become a significant environmental factor. The intensity of technogenic electromagnetic fields far exceeds the background indicators to which organisms have adapted in the course of evolution, in particular by their biorhythms. Much data on the biological effects of low-intensity (non-thermal) extremely high-frequency electromagnetic fields (EHF-EMF) at different levels of the organization has been accumulated. There are many possible explanations for these impacts, but the mechanism of action is not established. The purpose of this work is to analyze the negative effects of the use of non-thermal coherent electromagnetic radiation of extremely high frequencies. In particular, the use of microwave therapy is considered. A review of reports on the harmful effects of non-ionizing electromagnetic and magnetic fields among the population and professional groups has been made; the presence of the problem of hypersensitivity to such fields was shown. Information on the development of the direction of study of the resonant biological influence of EHF-EMF by Ukrainian specialists was provided. Particular attention is paid to the use of non-thermal microwaves for physiotherapy procedures, pain relief, correction of psycho-emotional disorders, ulcer healing, in particular stomach and duodenum, etc. It is noted that the so-called "therapeutic" frequencies of EHF-EMF, even if controlled, can cause negative effects, although they are not perceived as damaging. Changes in objective vital signs in the case of targeted exposure by low-intensity millimeter-wave radiation, namely, hemodynamics, temperature increase in the exposure zone, muscle activity, and the speed of the passage of a nerve impulse, an encephalogram were established. Precautionary recommendations based on the scientific and practical experience of using microwave acupuncture methods in the conditions of medical institutions are formulated. The synergistic effect of various influences, in particular electromagnetic fields, may not be a direct trigger of the disease, but a factor of not compensated stress. Therefore, clinical studies conducted many years ago cannot simply be dismissed on the grounds that the millimeter radiation penetrates only the outer layer of the skin. Skin cells can also secrete mediators that cause changes in ionic calcium channels and the level of calcium in the cells. The interest in these works is justified by the fact that currently the sources of monochromatic millimeter radiation will be spread with new communication systems. The risks of their biological action should be taken into account when emitters are installed.

We chose diisononyl phthalate as our experimental research object; this compound is an isononyl ester of phthalic acid based on n-butene (CAS No. 28553-12-0). Our research goal was to establish peculiarities and regularities related to toxic effects produced by diisononyl phthalate on experimental animals’ bodies and give scientific substantiation for fixing its permissible quantities that could migrate from polymer materials and medical products. We accomplished our research applying a set of toxicological, physiological, hematologic, biochemical, immunologic, cy-togenetic, morphological, organoleptic, sanitary-chemical, and statistical techniques. We were the first to establish peculiarities of toxic impacts exerted by diisononyl phthalate after intragastric introduction into white rates. Those impacts were dose-dependent toxic effects produced both on a whole body and specific organs and systems with typical disorders in functional and morphological parameters, cell differentiation, and an increase in number of cells with changes in their genetic apparatus. We revealed that overall toxic effects as well as specific damaging ones produced by diisononyl phthalate on the endocrine and reproductive systems in experimental animals’ bodies became apparent via functional disorders in the thyroid gland and gonads, embryogenesis, and offspring post-natal development. A chronic experiment allowed us to determine quantitative regularities and how toxic effects produced by diisononyl phthalate manifested themselves; so we were able to determine limiting parameters and a biological marker as well as fix a threshold of its hazardous impacts on a body, and substantiate fixing permissible quantities that could migrate from polymer materials and medical products. Our experiment results gave grounds for creating a hygienic standard for diisononyl phthalate that provided hygienic assessment and safe distribution of materials and medical products made of polymers that contained the compound.

Nanoparticles of transition metal dichalcogenides, particularly of molybdenum (Mo), have gained a lot of focus due to their exceptional physicochemical properties and the growing number of technological applications. These nanoparticles are also considered as potential therapeutic tools, biosensors or drug carriers. It is crucial to thoroughly examine their biocompatibility and ensure safe usage. The aim of this review is to analyze the available data on the biological effects of different nanoforms of elemental Mo and its compounds. In the reviewed publications, different conditions were described, including different experimental models, examined nanoforms, and their used concentrations. Due to these differences, the results are rather difficult to compare. Various studies classify Mo related nanomaterials as very toxic, mildly toxic or non-toxic. Similarly, the mechanisms of toxicity proposed in some studies are different, including oxidative stress induction, physical membrane disruption or DNA damage. Quite promising, however, are the potential medical applications of MoS 2 nanoparticles in therapy of cancer and Alzheimer’s disease. Nowadays, environmental electromagnetic fields exist everywhere and the whole population is exposed. The most widespread technologies engendering exposure to electromagnetic fields for the public and workers are radiofrequency wireless communication systems. Such exposure can have direct effects on living tissue involving biological problems or personal symptoms, as well as indirect effects restricting healthcare appliances. This assessment aims to analyze, examine and categorize the consequences of non-ionizing electromagnetic fields emitted by radiofrequency devices. This concerns biological effects in living tissues and atypical personal symptoms for short and long-term exposures. The evaluation methodology in this field of investigation is also discussed. In the article, these consequences as well as their evaluation techniques, in the case of wireless digital communication tools, are analyzed and reviewed. The categorization of exposure sources, the corresponding effects and their assessment methodologies are analyzed. The concept of systematic reviews and meta-analyses and their applications in various assessments of these effects and symptoms are reviewed and discussed.

The global spread of 5th generation (5G) wireless systems causes some concern about health effects of millimeter waves (MMW). To investigate biological effects of local exposure to 5G-MMW on human body, a novel 60 GHz band exposure setup was developed, and its performance was validated. A spatial synthetic beam-type exposure setup using two dielectric lens antennas was proposed to achieve high intensity 60 GHz irradiation to the target area of human skin. Variety distributions and intensities of electromagnetic fields at the exposed area, which is modified by incident angles of the combined beams, were simulated using finite-difference time-domain methods. The exposure performance we estimated was verified by temperature elevations of surface in a physical arm-shaped silicone phantom during the MMW exposure. The interference fringes generated in the exposed area due to the combined two-directional beam radiations were observed both in the simulation and in the phantom experiment but eliminated by applying an orthogonalizing polarized feeding structure. Under these exposure conditions, the local temperature changes, which could evoke warmth sensations, were obtained at the target area of the human forearm skin, which means the achievement of exposure performance we intended.

Biophysical and clinical medical studies have confirmed that biological tissue lesions and trauma are related to the damage of an intrinsic electret (i.e., endogenous electric field), such as wound healing, embryonic development, the occurrence of various diseases, immune regulation, tissue regeneration, and cancer metastasis. As exogenous electrical signals, such as conductivity, piezoelectricity, ferroelectricity, and pyroelectricity, bioelectroactives can regulate the endogenous electric field, thus controlling the function of cells and promoting the repair and regeneration of tissues. Materials, once polarized, can harness their inherent polarized static electric fields to generate an electric field through direct stimulation or indirect interactions facilitated by physical signals, such as friction, ultrasound, or mechanical stimulation. The interaction with the biological microenvironment allows for the regulation and compensation of polarized electric signals in damaged tissue microenvironments, leading to tissue regeneration and repair. The technique shows great promise for applications in the field of tissue regeneration. In this paper, the generation and change of the endogenous electric field and the regulation of exogenous electroactive substances are expounded, and the latest research progress of the electret and its biological effects in the field of tissue repair include bone repair, nerve repair, drug penetration promotion, wound healing, etc. Finally, the opportunities and challenges of electret materials in tissue repair were summarized. Exploring the research and development of new polarized materials and the mechanism of regulating endogenous electric field changes may provide new insights and innovative methods for tissue repair and disease treatment in biological applications.

Background: The widespread use of radiofrequency (RF) sources, ranging from household appliances to telecommunications devices and military equipment, raises concerns among people and regulatory agencies about the potential health risks of RF exposure. Consequently, several in vitro and in vivo studies have been done to investigate the biological effects, in particular non-thermal, of this non-ionizing radiation. To date, this issue is still being debated due to the controversial results that have been reported. Furthermore, the impact of different RF signal modulations on biological systems remains poorly investigated. The present in vitro study aims to evaluate the cytotoxicity and genotoxicity of continuous or pulsed 1.6 GHz RF in human dermal fibroblasts (HDF)., Methods: HDF cultures were exposed to continuous and pulsed 1.6 GHz RF, for 2 h, with Specific Absorption Rate (SAR) of 0.4 W/kg. The potential biological effects of 1.6 GHz RF on HDF were assessed with a multi-methodological approach, analyzing the effects on cell cycle, ultrastructure, protein expression, mitotic spindle, CREST stained micronuclei, chromosome segregation and γ-H2AX/53BP1 foci., Results: 1.6 GHz RF exposure modified proteins expression and morphology of HDF. Specifically, the expression of different heat-shock proteins (HSP) (i.e., HSP-90, HSP-60, and HSP-25) and phospho-AKT were affected. In addition, both continuous and pulsed RF modified the cytoskeletal organization in HDF and increased the number of lysosomes, while the formation of autophagosomes was observed only after pulsed RF exposure. Mitotic spindle anomalies were also found after exposure. However, no significant effect was observed on cell cycle, chromosome segregation, CREST-stained micronuclei and γ-H2AX/53BP1 foci., Conclusion: The results of the present study show the absence of genotoxic damage in 1.6 GHz RF exposed HDF and, although mitotic spindle alterations were observed, they did not have an aneugenic effect. Purpose To investigate the feasibility of using quantitative MR elastography (MRE) to characterize the influence of aging and sex on left ventricular (LV) shear stiffness. Materials and Methods In this prospective study, LV myocardial shear stiffness was measured in 109 healthy volunteers (age range: 18-84 years; mean age, 40 years ± 18 [SD]; 57 women, 52 men) enrolled between November 2018 and September 2019, using a 5-minute MRE acquisition added to a clinical MRI protocol. Linear regression models were used to estimate the association of cardiac MRI and MRE characteristics with age and sex; models were also fit to assess potential age-sex interaction. Results Myocardial shear stiffness significantly increased with age in female (age slope = 0.03 kPa/year ± 0.01, P = .009) but not male (age slope = 0.008 kPa/year ± 0.009, P = .38) volunteers. LV ejection fraction (LVEF) increased significantly with age in female volunteers (0.23% ± 0.08 per year, P = .005). LV end-systolic volume (LVESV) decreased with age in female volunteers (-0.20 mL/m 2 ± 0.07, P = .003). MRI parameters, including T1, strain, and LV mass, did not demonstrate this interaction ( P > .05). Boron neutron capture therapy (BNCT) is a re-emerging technique for selectively killing tumor cells. Briefly, the mechanism can be described as follows: after the uptake of boron into cells, the thermal neutrons trigger the fission of the boron atoms, releasing the α-particles and recoiling lithium particles and high-energy photons that damage the cells. We performed a detailed study of the reactor dosimetry, cellular dose assessment, and radiobiological effects induced by BNCT in glioblastoma (GBM) cells. At maximum reactor power, neutron fluence rates were ϕ0 = 6.6 × 107 cm−2 s−1 (thermal) and θ = 2.4 × 104 cm−2 s−1 with a photon dose rate of 150 mGy·h−1. These values agreed with simulations to within 85% (thermal neutrons), 78% (epithermal neutrons), and 95% (photons), thereby validating the MCNPX model. The GEANT4 simulations, based on a realistic cell model and measured boron concentrations, showed that >95% of the dose in cells was due to the BNC reaction. Carboranylmethylbenzo[b]acridone (CMBA) is among the different proposed boron delivery agents that has shown promising properties due to its lower toxicity and important cellular uptake in U87 glioblastoma cells. In particular, the results obtained for CBMA reinforce radiobiological effects demonstrating that damage is mostly induced by the incorporated boron with negligible contribution from the culture medium and adjacent cells, evidencing extranuclear cell radiosensitivity.

Medicinal plants are rich sources of bioactive compounds widely used as medicaments, food additives, perfumes, and agrochemicals. These secondary compounds are produced under stress conditions to carry out physiological tasks in plants. Secondary metabolites have a complex chemical structure with pharmacological properties. The widespread use of these metabolites in a lot of industrial sectors has raised the need to increase the production of secondary metabolites. Biotechnological methods of cell culture allow the conservation of plants, as well as the improvement of metabolite biosynthesis and the possibility to modify the synthesis pathways. The objective of this review is to outline the applications of different in vitro culture systems with previously reported relevant examples for the optimal production of plant-derived secondary metabolites.

For the study of biological effects of direct current (DC) and extremely low frequency (ELF) electric fields, we have quantitatively analyzed red blood cell (RBC) movement in whole blood. Considering the inhomogeneous distribution of electric fields in vivo, five different electric field distributions were generated under a microscope. For theoretical analyses, we assumed electrophoresis and dielectrophoresis as basic motive forces and obtained the spatial distribution of blood cell velocity. The RBC velocity was measured using video image analysis. The spatial dependence of the velocity showed good agreement with that predicted by theoretical analysis. This result suggests the validity of the theoretical model based on electrophoresis and dielectrophoresis for the study of ELF electric field exposure to inhomogeneous animal and human bodies. Next, using the same measurement system, we attempted to find the electric field strength at which these effects occur. The threshold values were found to be 0.40 and 1.6 kV/m, respectively, for DC and AC electric field exposures. Furthermore, we investigated the reproducibility of the field effects in more realistic conditions of human exposure. The RBCs in microchannels were exposed to the electric field generated in capacitive coupling using electrodes separated by an air gap. Even in the new condition, similar effects were observed, which also verified the validity of the analysis described above. Marrubium vulgare L. (Lamiaceae) has a long history of use in traditional herbal medicine for the treatment of respiratory tract infections, inflammatory conditions, and pain. This study aimed to investigate the chemical composition, acute toxicity, and antinociceptive effects of the aqueous extract from M. vulgare leaves (AEMV). Antioxidant activity was evaluated using DPPH and reducing power assays. The chemical composition of AEMV was determined through LC-MS/MS, and the levels of total phenolics, flavonoids, and condensed tannins were quantified. Acute oral toxicity was assessed in male Swiss mice with a single oral dose of AEMV (1, 2, 5 g/kg). The analgesic impact was examined through writhing, hot plate, and formalin tests. Our findings not only confirmed the safety of the extract in animal models but also revealed significant antioxidant activity in AEMV. High-performance liquid chromatography (HPLC) analysis identified important bioactive compounds, with marrubiin being a major component. Furthermore, AEMV demonstrated robust antinociceptive properties in all conducted tests, highlighting its potential as a valuable natural source of bioactive compounds suitable for a wide range of therapeutic applications., (© 2024 Wiley-VHCA AG, Zurich, Switzerland.)

Background: 223Ra is currently used for treatment of metastatic castration resistant prostate cancer patients (mCRPC) bone metastases with fixed standard activity. Individualized treatments, based on adsorbed dose (AD) in target and non-target tissue, are absolutely needed to optimize efficacy while reducing toxicity of α-emitter targeted therapy. This is a pilot first in human clinical trial aimed to correlate dosimetry, clinical response and biological side effects to personalize 223Ra treatment. Methods: Out of 20 mCRPC patients who underwent standard 223Ra treatment and dosimetry, in a subset of 5 patients the AD to target and non-target tissues was correlated with clinical effects and radiation-induced chromosome damages. Before each 223Ra administrations, haematological parameters, PSA and ALP values were evaluated. Additional blood samples were obtained baseline (T0), at 7 days (T7), 30 days (T30) and 180 days (T180) to evaluate chromosome damage. After administration WB planar 223Ra images were obtained at 2–4 and 18–24 h. Treatment response and toxicity were monitored with clinical evaluation, bone scan, 18F-choline-PET/CT, PSA value and ALP while haematological parameters were evaluated weekly after 223Ra injection and 2 months after last cycle. Results: 1. a correlation between AD to target and clinical response was evidenced with threshold of 20 Gy as a cut-off to obtain tumor control; 2. the AD to red marrow was lower than 2 Gy in all the patients with no apparently correlation between dosimetry and clinical toxicity. 3. a high dose dependent increase of the number of dicentrics and micronuclei during the course of 223Ra therapy was observed and a linear correlation has been found between blood AD (BAD) and number of dicentrics. Conclusions: This study provides some interesting preliminary evidence to be further investigated: dosimetry may be useful to identify a more appropriate 223Ra administered activity predicting AD to target tissue; a dose dependent complex chromosome damage occurs during 223Ra administration and this injury is more evident in heavily pre-treated patients; dosimetry could be used for radioprotection purpose. The biological consequences of absorbed radiation doses are ill-defined for radiopharmaceuticals, unlike for external beam radiotherapy (EBRT). A reliable assay that assesses the biological consequences of any radionuclide is much needed. Here, we evaluated the cell-free plasmid DNA assay to determine the relative biological effects of radionuclides such as Auger electron-emitting [67Ga]GaCl 3 or [111In]InCl 3 compared to EBRT. Supercoiled pBR322 plasmid DNA (1.25 or 5 ng/μL) was incubated with 0.5 or 1 MBq [67Ga]GaCl 3 or [111In]InCl 3 for up to 73 h or was exposed to EBRT (137Cs; 5 Gy/min; 0–40 Gy). The induction of relaxed and linear plasmid DNA, representing single and double strand breaks, respectively, was assessed by gel electrophoresis. Chelated forms of 67Ga were also investigated using DOTA and THP. Topological conversion rates for supercoiled-to-relaxed (k sr x ) or relaxed-to-linear (k rl x ) DNA were obtained by fitting a kinetic model. DNA damage increased both with EBRT dose and incubation time for [67Ga]GaCl 3 and [111In]InCl 3. Damage caused by [67Ga]GaCl 3 decreased when chelated. [67Ga]GaCl 3 proved more damaging than [111In]InCl 3 ; 1.25 ng/μL DNA incubated with 0.5 MBq [67Ga]GaCl 3 for 2 h led to a 70% decrease of intact plasmid DNA as opposed to only a 19% decrease for [111In]InCl 3. For both EBRT and radionuclides, conversion rates were slower for 5 ng/μL than 1.25 ng/μL plasmid DNA. DNA damage caused by 1 Gy EBRT was the equivalent to damage caused by 0.5 MBq unchelated [67Ga]GaCl 3 and [111In]InCl 3 after 2.05 ± 0.36 and 9.3 ± 0.77 h of incubation, respectively. This work has highlighted the power of the plasmid DNA assay for a rapid determination of the relative biological effects of radionuclides compared to external beam radiotherapy. The biological effects of direct current (DC) and extremely low frequency (ELF) electric fields (EFs) have not been completely clarified. However, scholars have proposed that body hair movement on the body surface in the EFs triggers these effects. The electric forces in DC or alternating current (AC) EFs have been discussed, and a theoretical equation has been proposed, which considers either AC or DC EFs only. In contrast, superimposed EFs combining DC and AC EFs, inducing changes in the electric force, are widely used in the industry. In this regard, the biological effects of the EFs could be understood better by considering the superimposed conditions. This study generalizes a theoretical equation to include the superimposed EFs and calculate the electric force change acting on body hair with respect to the AC/DC ratio. The results show that the electric force that is acting on body hair changes with the AC/DC ratio of the superimposition, even when the total root mean square value is constant. Moreover, there is an optimal ratio for maximizing the electric force. This study could provide insights for an in‐depth understanding of the biological effects of EFs if the results of the developed equation are studied in conjunction with the actual effects. © 2021 Institute of Electrical Engineers of Japan. Background: Although the scientific community is extensively concerned with the effects of the EMF, the unambiguous explanation of its effects on living structures is still lacking. Goals: The goal of the study was to evaluate the effect of a low-frequency (LF) electromagnetic field (EMF) on the growth and multiplication of the yeast Saccharomyces cerevisiae. Methods: Yeast cells were exposed to a frequency of 900 Hz and a magnetic flux density of 2.3 mT. The duration of each experiment was 8 hours, in the beginning of the measurement the value of frequency, rms (root mean square) value of electric current (2 A), and magnetic flux density were fixed set on the exposure device. A paired experiment was performed, a sample exposed to EMF, and a sample shielded from the field. Subsequently, samples were taken every two hours, the number of cells was recorded, and then the concentration of the yeast cells was evaluated at time points. The time points reflected the exposure time of the samples exposed to EMF. Results: The results indicate that LF EMF at given parameters has an inhibitory effect on the growth and multiplication of yeast cells. Conclusion: Exposure to EMF can cause the differences in growth dynamics between cells exposed to the field and the unexposed ones. [ABSTRACT FROM AUTHOR]

The potential use of magnetic nanoparticles (MNPs) in biomedicine as magnetic resonance, drug delivery, imagenology, hyperthermia, biosensors, and biological separation has been studied in different laboratories. One of the challenges on MNP elaboration for biological applications is the size, biocompatibility, heat efficiency, stabilization in physiological conditions, and surface coating. Magnetoliposome (ML), a lipid bilayer of phospholipids encapsulating MNPs, is a system used to reduce toxicity. Encapsulated MNPs can be used as a potential drug and a gene delivery system, and in the presence of magnetic fields, MLs can be accumulated in a target tissue by a strong gradient magnetic field. Here, we present a study of the effects of DC magnetic fields on encapsulated MNPs inside liposomes. Despite their widespread applications in biotechnology and environmental, biomedical, and materials science, the effects of magnetic fields on MLs are unclear. We use a modified coprecipitation method to synthesize superparamagnetic nanoparticles (SNPs) in aqueous solutions. The SNPs are encapsulated inside phospholipid liposomes to study the interaction between phospholipids and SNPs. Material characterization of SNPs reveals round-shaped nanoparticles with an average size of 12 nm, mainly magnetite. MLs were prepared by the rehydration method. After formation, we found two types of MLs: one type is tense with SNPs encapsulated and the other is a floppy vesicle that does not show the presence of SNPs. To study the response of MLs to an applied DC magnetic field, we used a homemade chamber. Digitalized images show encapsulated SNPs assembled in chain formation when a DC magnetic field is applied. When the magnetic field is switched off, it completely disperses SNPs. Floppy MLs deform along the direction of the external applied magnetic field. Solving the relevant magnetostatic equations, we present a theoretical model to explain the ML deformations by analyzing the forces exerted by the magnetic field over the surface of the spheroidal liposome. Tangential magnetic forces acting on the ML surface result in a press force deforming MLs. The type of deformations will depend on the magnetic properties of the mediums inside and outside the MLs. The model predicts a coexistence region of oblate–prolate deformation in the zone where χ = 1. We can understand the chain formation in terms of a dipole–dipole interaction of SNP.

Ultrasound has the potential to be broadly applied in the field of agricultural food processing due to advantages such as environmental friendliness, low energy costs, no need for exogenous additives and ease of operation. High-frequency ultrasound is mainly used in medical diagnosis and in the food industry for the identification of ingredients and production line quality testing, while low-frequency ultrasounds is mainly used for extraction and separation, accelerating chemical reactions, auxiliary microbial fermentation and quality enhancement in food industry. Magnetic fields have many advantages of convenient use, such as non-toxic, nonpolluting and safe. High-intensity pulsed magnetic fields are widely used as a physical non-thermal sterilization technology in food processing, while weak magnetic fields are better at activating microorganisms and promoting their growth. Ultrasound and magnetic fields, due to their positive biological effects, have a wide range of applications in the food processing industry. This paper provides an overview of the research progress and applications of ultrasound and magnetic fields in food processing from the perspectives of their biological effects and mechanisms of action. Additionally, with the development and application of physical field technology, physical fields can now be used to provide significant technical advantages for assisting fermentation. Suitable physical fields can promote the growth of microbial cells, improve mycelial production and increase metabolic activity. Furthermore, the current status of research into the use of ultrasound and magnetic field technologies for assisting the fermentation of rare edible fungi, is discussed.

Exposure to ionizing radiation can occur during medical treatments, from naturally occurring sources in the environment, or as the result of a nuclear accident or thermonuclear war. The severity of cellular damage from ionizing radiation exposure is dependent upon a number of factors including the absorbed radiation dose of the exposure (energy absorbed per unit mass of the exposure), dose rate, area and volume of tissue exposed, type of radiation (e.g., X-rays, high-energy gamma rays, protons, or neutrons) and linear energy transfer. While the dose, the dose rate, and dose distribution in tissue are aspects of a radiation exposure that can be varied experimentally or in medical treatments, the LET and eV are inherent characteristics of the type of radiation. High-LET radiation deposits a higher concentration of energy in a shorter distance when traversing tissue compared with low-LET radiation. The different biological effects of high and low LET with similar energies have been documented in vivo in animal models and in cultured cells. High-LET results in intense macromolecular damage and more cell death. Findings indicate that while both low- and high-LET radiation activate non-homologous end-joining DNA repair activity, efficient repair of high-LET radiation requires the homologous recombination repair pathway. Low- and high-LET radiation activate p53 transcription factor activity in most cells, but high LET activates NF-kB transcription factor at lower radiation doses than low-LET radiation. Here we review the development, uses, and current understanding of the cellular effects of low- and high-LET radiation exposure.

Cellulose nanomaterials (CNMs) have emerged recently as an important group of sustainable bio-based nanomaterials (NMs) with potential applications in multiple sectors, including the food, food packaging, and biomedical fields. The widening of these applications leads to increased human oral exposure to these NMs and, potentially, to adverse health outcomes. Presently, the potential hazards regarding oral exposure to CNMs are insufficiently characterised. There is a need to understand and manage the potential adverse effects that might result from the ingestion of CNMs before products using CNMs reach commercialisation. This work reviews the potential applications of CNMs in the food and biomedical sectors along with the existing toxicological in vitro and in vivo studies, while also identifying current knowledge gaps. Relevant considerations when performing toxicological studies following oral exposure to CNMs are highlighted. An increasing number of studies have been published in the last years, overall showing that ingested CNMs are not toxic to the gastrointestinal tract (GIT), suggestive of the biocompatibility of the majority of the tested CNMs. However, in vitro and in vivo genotoxicity studies, as well as long-term carcinogenic or reproductive toxicity studies, are not yet available. These studies are needed to support a wider use of CNMs in applications that can lead to human oral ingestion, thereby promoting a safe and sustainable-by-design approach.

Competitive inhibitor and spectroscopic assays confirm the high affinity to GAGs heparin, heparan sulfate (HS), and its pentasaccharide mimetic Fondaparinux (FPX). Functional consequences of this affinity include inhibition of FPX cleavage by bacterial heparinase and mammalian heparanase enzymes with inhibition of cellular invasion and migration. The increasing use of nanoparticles and their many applications increases the likelihood of their presence in the environment. This possibility of presence necessitates the study of the effect of these substances on aquatic species. In this research, CoWO4 nanoparticles were synthesized by the ultrasonic method. Various conditions in the synthesis process were investigated to obtain the appropriate size of the nanoparticles. After selecting the optimum particles, these nanoparticles were used to investigate their effect on the growth of Dunaliella salina. For this purpose, the algal cells were subjected to three different concentrations of nanoparticles (15, 30, and 60 mg/L). The study results on algae growth parameters showed that these parameters depend on the value of nanoparticles. At 15 and 30 mg/L concentrations of the nanoparticles, numbers of cells, specific growth, biomass, and pigments showed a significant boost compared to the mentioned parameters of the control treatment. Measurement of malondialdehyde (MDA) showed that this parameter was directly related to the increase in the concentration of nanoparticles. At 60 mg/L of the nanoparticles, the MDA level was higher than the control and other treatments. This increase reflects the destructive effect of the nanoparticles on algal cells. Objective: Recent technological advances have exponentially expanded globally; harbouring upon Electromagnetic fields (EMF). The utilization of Electromagnetic field has become universal from everyday usage of electronic appliances such as micro wave ovens, tablets and portable computers to telecommunication systems mobile phone towers, radio-television broadcast systems and electronic power transmission systems resulting in electromagnetic field and associated radiations. EMF can have biological effects on cell at microlevel and have the potential ability to cause cell dysfunction manifesting in various biological effects. This review tried to gather evidence from the existing literature about the biological effects of EMF on human health. Materials and Methods: We did extensive literature search using PubMed and Cochrane database using key words, "electromagnetic fields", "Extremely low frequency electromagnetic fields (ELF-EMFs)", "biological effects", "health effects", "public health". We included 20 studies conducted from Dec 2009 to Dec 2019 in our systematic review. Data from each study was extracted by two independent researchers and discrepancies were resolved by consensus. Results: Significant biological effects of EMF exposure were reported on human health ranging from anxiety, depression, sleep disturbance, increased risk of Alzheimer's disease and ALS (Amyotrophic Lateral Sclerosis), hypersensitivity to infertility and increased risk of multiple carcinomas. Conclusion: Application of preventive measures in order to minimize the exposure becomes the need of the hour especially so in occupational settings. [ABSTRACT FROM AUTHOR]
