Growth and maintenance of skeletal muscle is essential for athletic performance and a healthy life. Stimulating the proliferation and differentiation of muscle cells may help prevent loss of muscle mass. To discover effective natural substances enabling to mitigate muscle loss without side effects, we evaluated muscle growth with several compounds extracted from Catalpa bignonioides Walt. Among these compounds, pinoresinol and vanillic acid increased C2C12, a mouse myoblast cell line, proliferation being the most without cytotoxicity. These substances activated the Akt/mammalian target of the rapamycin (mTOR) pathway, which positively regulates the proliferation of muscle cells. In addition, the results of in silico molecular docking study showed that they may bind to the active site of insulin-like growth factor 1 receptor (IGF-1R), which is an upstream of the Akt/mTOR pathway, indicating that both pinoresinol and vanillic acid stimulate myoblast proliferation through direct interaction with IGF-1R. These results suggest that pinoresinol and vanillic acid may be a natural supplement to improve the proliferation of skeletal muscle via IGF-1R/Akt/mTOR signaling and thus strengthen muscles.

Alzheimer's disease (AD) is a neurodegenerative disease, in which the accumulation of β-amyloid (Aβ) peptide in the extracellular space causes a progressive reduction in cognitive performance. Aβ stimulates active oxygen species generation leading to oxidative stress and neural cell death. Vanillic Acid (VA) is the oxidant form of vanillin widely found in vanilla beans. VA has many properties, such as suppressing apoptosis and eliminating the harmful effects of oxidative stress in animal models. The VA effects on impaired learning and memory in Aβ rats were assessed. Forty adults male Wistar rats were assigned to the following five groups in random: the control, sham (received saline (vehicle) via intracerebroventricular (ICV) injection), Aβ (received Aβ1–40 via ICV injection), VA (50 mg/kg by oral gavage once a day through four weeks), and Aβ + VA (50 mg/kg) groups. Open field test, novel object recognition (NOR) test, Morris water maze (MWM) test, and passive avoidance learning (PAL) task were performed, and finally, we determined the malondialdehyde (MDA), total antioxidant capacity (TAC) and total oxidant status (TOS) levels. Aβ decreased the cognitive memory in NOR, spatial memory in MWM, and passive avoidance memory in PAL tests. In contrast, VA improved learning and memory in the treated group. Aβ significantly increased MDA and TOS and decreased TAC levels, whereas VA treatment significantly reversed TAC, TOS and MDA levels. In conclusion, VA decreased the Aβ effects on learning and memory by suppressing oxidative stress and can be regarded as a neuroprotective substance in AD. [ABSTRACT FROM AUTHOR]

The inhibition of a-glucosidase and a-amylase, enzymes which are implicated in the digestion of carbohydrates, can evidently decrease the post-prandial increase of blood glucose levels. It can be an significant approach in the management of blood glucose level in type 2 diabetic and borderline patients. At present, there is renewed attention in plant-based medicines and functional foods modulating physiological reactions in the prevention and treatment of diabetes and obesity. The plant kingdom is a extensive field to investigate for natural effective oral hyperglycemic agents that have insignificant or no side effects. Consequently, natural a-glucosidase and a-amylase inhibitors from plant sources offer an prominent strategy for the control of hyperglycemia. Nowadays majority of the people divert towards millets and taken the millets as a meal in the place of rice as because they show key results on who are suffering from hyperglycemia. Foxtail millet [Setariaitalica (L.) P. Beauv.] is a member of poaceae grass family found in both arid and semi-arid regions which is principally used as a fodder and cereal crop. All the food products prepared from foxtail millet are efficient in reducing the blood glucose levels both in normal as well as diabetic patients. To identify human a-glucosidase and a-amylase inhibitors phenolic compounds (Vanillic acid, Ferulic acid) and flavonoid (Kaempferol) of foxtail millets were screened using the structural based molecular docking approach. The findings of this study revealed that all the tested compounds exhibited inhibitory activity against both the enzymes. Comparatively among the three inhibitors tested kaempferol flavonoid was more effective with - 261.79Kcal/mol and -404.66Kcal/mol of binding free energy values against a-amylase and a-glucosidase correspondingly. Lignin bio-valorization is an emerging field of applied biotechnology and has not yet been studied at low temperatures. Paraburkholderia aromaticivorans AR20-38 was examined for its potential to degrade six selected lignin monomers (syringic acid, p-coumaric acid, 4-hydroxybenzoic acid, ferulic acid, vanillic acid, benzoic acid) from different upper funneling aromatic pathways. The strain degraded four of these compounds at 10°C, 20°C, and 30°C; syringic acid and vanillic acid were not utilized as sole carbon source. The degradation of 5 mM and 10 mM ferulic acid was accompanied by the stable accumulation of high amounts of the value-added product vanillic acid (85–89% molar yield; 760 and 1540 mg l−1, respectively) over the whole temperature range tested. The presence of essential genes required for reactions in the upper funneling pathways was confirmed in the genome. This is the first report on biodegradation of lignin monomers and the stable vanillic acid production at low and moderate temperatures by P. aromaticivorans. Alicyclobacillus acidoterrestris has been gaining attention due to its unique thermo-acidophilic properties and being associated with the deterioration of pasteurized beverages. The objective of this study was to evaluate the antibacterial activity of chitosan with various molecular weights (MWs) (164, 85, 29.2, and 7.1 kDa) and concentrations (0–100 μg/mL) against A. acidoterrestris and its effect on guaiacol production. Various chitosan MWs were co-incubated for 7 days, and the bacterial growth, guaiacol, and vanillic acid contents during storage were determined. The chitosans performed antibacterial effects against A. acidoterrestris. Further, 164 kDa chitosan showed excellent results in controlling the growth and guaiacol formation in A. acidoterrestris. These findings demonstrated the efficacy of chitosan antibacterial activity against A. acidoterrestris and mitigating the guaiacol formation. Chitosan's antibacterial properties are attributed to the elimination of cells and suppression of guaiacol production. Polycystic ovarian syndrome (PCOS) is one of the known causes of anovulatory fertility in the world. Previous research has linked oxidative stress could contribute to PCOS, and vanillic acid has shown antioxidant potential. Hence, the present study evaluated the effect of vanillic acid on letrozole-induced polycystic ovarian syndrome in female rats. PCOS was induced in Wistar female rats with letrozole (1 mg/kg, orally) in carboxymethoxycellulose (1 % w/v), administered for 21 days. After induction, the standard group received clomiphene citrate (1 mg/kg, orally) while other treatment groups were administered with vanillic acid at doses 25, 50, and 100 mg/kg, orally for 15 days, and without treatment was considered a negative control group. Different parameters studied were body weight, ovary weight, blood glucose, lipid profile, hormonal levels [luteinizing hormone (LH), follicle-stimulating hormone (FSH), and testosterone], markers for oxidative stress (superoxide dismutase, reduced glutathione, catalase, and malonaldehyde), and histopathology of the ovary. Statistical analysis was done for the results and p < 0.05 was considered to indicate the significance. Vanillic acid-treated animals showed a concentration-dependent activity on the tested parameters. The highest tested dose (100 mg/kg) produced a more prominent effect in significantly (P < 0.001) decreasing the body weight, and ovary weight and improving the hormonal imbalance. Also, vanillic acid significantly (P < 0.01) reduced elevated blood sugar and lipid levels. Additionally, vanillic acid reduced oxidative stress significantly (P < 0.001) in the ovaries of female rats. Histopathological reports showed a reduction in cystic follicles and appearance of normal healthy follicles at different stages of development after the administration of vanillic acid. Furthermore, these effects were observed to be comparable with those recorded for standard drug, clomiphene. The current study data suggests that vanillic acid has protected the letrozole-induced polycystic ovarian syndrome. In the event of several side effects associated with conventional treatments used for PCOS, the findings of this study suggest the promising role of vanillic acid. Till now, a wide range of chitosan (CHS)-based food packaging films have been developed. Yet, the role of molecular weight (MW), which is an important physical property of CHS, in determining the physicochemical and biochemical properties of vanillic acid (VA)-grafted CHS hydrogel films synthesized using CHS with different MWs has not been investigated until now. Three kinds of CHS including low, medium, and high MWs were grafted separately with VA through a carbodiimide mediated coupling reaction. No significant difference in water resistance properties was observed with increasing MW of CHS, in contrast to obvious decrease in light transmittance and opacity. The VA-g-CHS hydrogel films exhibited significantly improved light blocking capacity. A significant improvement in antioxidant (~6-fold) and antimicrobial (~1.2-fold) activity was observed after grafting with VA. In contrast, the free radical scavenging and antimicrobial activity decreased with increasing MW of CHS. Most importantly, VA-g-CHS hydrogel films could maintain the freshness of cherry tomatoes for up to 10 days at ~25 °C. However, no significant difference was observed depending on the MW value of CHS. This pioneering work is of great importance in guiding the selection of MW of CHS biomacromolecule to design hydrogel films with desired physicochemical and biochemical properties. An imbalance between soil microbial communities and phenolic acids is the major factor influencing soil sickness in Panax notoginseng crops. This study investigated how vanillic acid and p -hydroxybenzoic acid affect particular pathogens and soil microorganisms in P. notoginseng rhizosphere. Illumina MiSeq high-throughput sequencing revealed that exogenous phenolic acids led to alterations in the structures of microbial communities in the rhizospheric soil. Exogenous vanillic acid had a more pronounced effect on rhizospheric soil bacteria, as it caused a significant increase in the relative abundance of Pseudomonas , Enterobacterium , Pseudoarchaea , and Sphingomonas , while the relative abundance of RB41 , Bryobacter , and Haliangium decreased. Potential pathogen analysis revealed that exogenous vanillic acid significantly increased the abundance of potential pathogens, and the abundance of the Fusarium genus increased in response to the different vanillic acid and p -hydroxybenzoic acid treatments. Moreover, we discovered that vanillic acid promoted the mycelial growth of F. equiseti , while p -hydroxybenzoic acid strongly enhanced F. solani in culture experiments. Additionally, two phenolic acids were found to greatly enhance the spore germination of F. solani and F. equiseti. In summary, these two phenolic acids can alter the rhizospheric soil microbial community structure, especially the diversity of pathogens, and can significantly promote the mycelial growth and spore germination of pathogens. • Phenolic acids damaged microbial community in rhizospheric soil of P. notoginseng. • Phenolic acids could promote the spore germination of F. solani and F. equiseti. • Vanillic acid promoted the growth and reproduction of potential pathogens. Four vanillic acid-degrading bacterial strains, named LR5S13T, LR5S20, and M4R5S39T and LN1S58, were isolated from Kalidium cuspidatum rhizosphere and bulk soils, respectively. Phylogenetic analysis based on 16S rRNA gene as well as core genome revealed that LR5S13T and LR5S20 clustered closely with each other and with Halomonas ventosae Al12T, and that the two strains shared the highest similarities (both 99.3 %) with H. ventosae Al12T, in contrast, M4R5S39T and LN1S58 clustered together and with Halomonas heilongjiangensis 9–2T, and the two strains shared the highest similarities (99.4 and 99.2 %, respectively) with H. heilongjiangensis 9–2T. The average nucleotides identities based on BLAST (ANIb) and digital DNA-DNA hybridization (dDDH) values of strains LR5S13T to LR5S20, and M4R5S39T to LN1S58, were both higher than the threshold values for delineation of a species. The ANIb and dDDH values of the four strains to their closely relatives were lower than the threshold values. All four strains take phosphatidylethanolamine, phosphatidylglycerol, and diphosphatidylglycerol as the major polar lipids, Summed Feature 8, Summed Feature 3, and C 16:0 as the major fatty acids. Based on the phylogenetic and phenotypic results, the four strains should be classified as two novel Halomonas species. Therefore, Halomonas rhizosphaerae sp. nov. (type strain LR5S13T = KCTC 8016T = CGMCC 1.62049T) and Halomonas kalidii (type strain M4R5S39T = KCTC 8015T = CGMCC 1.62047T) are proposed. The geographical distribution analysis based on 16S rRNA gene revealed that the two novel species are widely distributed across the globe, specifically in highly saline habits, especially in Central and Eastern Asia. Abstract Approaching the end of the second decade of the 21st century, almost the whole demand of vanillin is met by the synthetic product obtained either via a petrochemical process starting from phenol and glyoxylic acid or from energy intensive alkaline oxidative depolymerization of lignin. Only a minor fraction is comprised of natural vanillin obtained from ferulic acid fermentation, and even less of highly valued Vanilla planifolia extracts. Are there alternative green production methods? And, if yes, are they suitable to find practical application?

Vanillic acid (VA), a dietary phenolic compound is generally studied for its anti-oxidative and anti-inflammatory effects. However, the effect of VA on insulin secretion and its mechanism of action has never been explored. In this study, we report that VA augments glucose-stimulated insulin secretion (GSIS) in both insulin-secreting cell-line INS-1 and isolated rat pancreatic islets. Potentiation of GSIS is accompanied by a concurrent increase in 3′,5′-cyclic adenosine monophosphate (cAMP) and activation of protein kinase A (PKA) in INS-1 and rat islets. The activated cAMP-PKA pathway, in turn, phosphorylates extracellular signal-regulated kinases 1/2 (ERK1/2) in INS-1 cells. Pharmacological intervention with PKA and ERK1/2 inhibitors revealed that VA potentiated GSIS is primarily dependent on PKA mediated ERK1/2 activity. These findings demonstrated that VA directly acts on insulin-secreting pancreatic β-cells to exert its insulinotropic effect thereby providing a novel role of VA in the regulation of insulin secretion.

Objectives: Synovitis plays an important role in knee osteoarthritis (KOA) pain. The activation of the NOD-like receptor protein 3 (NLRP3) inflammasome in fibroblast-like synoviocytes (FLSs) promotes KOA development. In this study, we aimed to investigate whether vanillic acid (VA), a monomer derived from Chinese herbal medicines, could target NLRP3 inflammasome-related synovitis to reduce pain.Methods: Rats in the KOA and KOA + VA groups were injected with monosodium iodoacetate (MIA) in the knee to induce KOA. From day 14, the KOA + VA group was given VA at 30 mg/kg every day via gastric intubation. FLSs were collected from the synovial tissues. We examined both the protein and gene expression of caspase-1, apoptosis-associated speck-like protein with a caspase recruitment domain (ASC), NLRP3, components of the NLRP3 inflammasome, and interleukin-1β (IL-1β) and IL-18 in vivo and in vitro.Results: The upregulation of caspase-1, ASC, and NLRP3 in the KOA model were reduced by VA. VA also lowered the level of IL-1β and IL-18 in the KOA model. In addition, VA relieved pain-related behavior of KOA model rats and downregulated the pain mediators CGRP, NGF, and TrkA in FLSs. Interestingly, we also observed reduced synovial fibrosis in the animal experiments.Conclusion: Our research showed that VA reduces synovitis and pain-related behaviors in a rat model of KOA, which provides the basis for further investigations into the potential therapeutic impact of VA in KOA.

The overproduction of reactive oxygen species (ROS) causes oxidative stress, such as Hydrogen peroxide (H 2 O 2). Acute oxidative stress is one of the main reasons for cell death. In this study, the antioxidant properties of vanillic acid- a polyphenolic compound was evaluated. Therefore, this study aims to check the effectiveness of vanillic acid in H 2 O 2 -induced oxidative stress in D. Mel-2 cell line. The efficacy was determined by biochemical tests to check the ROS production. The cytotoxicity of H 2 O 2 and vanillic acid was checked by MTT assay. The DNA fragmentation was visualized by gel electrophoresis. Protein biomarkers of oxidative stress were analyzed by western blotting. The results depict a promising antioxidant effect of vanillic acid. The IC 50 value of vanillic acid and H 2 O 2 was found 250 μg/ml and 125 μg/ml, respectively. The catalase activity, SOF, GPx, and PC was seen less in H 2 O 2 treated group compared with the control and vanillic acid treated group. However, the TBRAS activity was hight in H 2 O 2 treated group. The effect of H 2 O 2 on DNA fragmentation was high as compared with vanillic acid-treated cells. The protein expression of Hsp70, IL-6 and iNOS was seen significant in a vanillic acid-treated group as compared with H 2 O 2 treated group. These results reinforce that at low concentration, vanillic acid could be used as an antioxidant agent in the food and pharmaceutical industries. [ABSTRACT FROM AUTHOR]

The allelopathic potential of plant species and their related compounds has been increasingly reported to be biological tools for weed control. The allelopathic potential of Garcinia xanthochymus was assessed against several test plant species: lettuce, rapeseed, Italian ryegrass, and timothy. The extracts of G. xanthochymus leaves significantly inhibited all the test plants in a concentration- and species-specific manner. Therefore, to identify the specific compounds involved in the allelopathic activity of the G. xanthochymus extracts, assay-guided purification was carried out and two allelopathic compounds were isolated and identified as methyl phloretate {3-(4-hydroxyphenyl) propionic acid methyl ester} and vanillic acid (4-hydroxy-3-methoxybenzoic acid). Both of the substances significantly arrested the cress and timothy seedlings growth. I50 values (concentrations required for 50% inhibition) for shoots and roots growth of the cress and timothy were 113.6–104.6 and 53.3–40.5 μM, respectively, for methyl phloretate, and 331.6–314.7 and 118.8–107.4 μM, respectively, for vanillic acid, which implied that methyl phloretate was close to 3- and 2-fold more effective than vanillic acid against cress and timothy, respectively. This report is the first on the presence of methyl phloretate in a plant and its phytotoxic property. Nine yeast strains isolated from Latin American biodiversity were screened for ferulic acid (FA) consumption and conversion into aroma compounds such as vanillin, vanillic acid (VA), and 4‐vinylguaiacol (VG). Selected strains (Rhodotorula mucilaginosa UFMG‐CM‐Y3647, UFMG‐CM‐Y2190, UFMG‐CM‐Y665) were evaluated in flask experiments to investigate the influence of the pH media on bioconversion and a two‐step process was conducted to maximize the metabolites production. The effect of pH was found to be significantly important for FA bioconversion, as acidic conditions (pH < 6.0) improved VA accumulation, with highest production of 1.14 ± 0.02 and 1.25 ± 0.03 g/L shown by UFMG‐CM‐Y3647 and UFMG‐CM‐Y2190, respectively. The two‐step process favored 4‐VG production for most strains, being UFMG‐CM‐Y2190 the best producer, its cultures reaching 1.63 ± 0.09 g/L after 55 hr, showing a productivity of 29.59 ± 1.55 mg/(L·hr), as glucose affected the metabolites pool and redirected yeast metabolism. R mucilaginosa UFMG‐CM‐Y3647 was selected for scaled‐up cultivations in a 2‐L bioreactor, where pH‐controlled pH 5.5 and aeration of 2.5 vvm was found to be the best condition to improve VA productivity, attaining final concentrations of 1.20 ± 0.02 g/L−1 (78% molar yield) and a productivity of 40.82 ± 0.57 mg/(L·hr). This study aimed to investigate whether different doses of specific phenolic acids (chlorogenic acid, caffeic acid, ferulic acid, gallic acid, protocatechuic acid, p-hydroxybenzoic acid, syringic acid, vanillic acid, and p-coumaric acid), alone or in combination, can inhibit the early growth of the common ragweed (Ambrosia artemisiifolia L., Asterales: Asteraceae). A seed bioassay was performed in Petri dishes and placed in a climate chamber to assess the effects of five dose levels of phenolic acids to radicle and shoot length, as well seedling biomass of A. artemisiifolia. The lowest dose of phenolic acid corresponded to the natural phenolic acid concentration previously reported in dry plant tissue samples from Brassicaceae cover crop plants. Results show that the inhibition of the early growth of A. artemisiifolia depends strongly on phenolic acid. Across different treatments, high doses of phenolic acids significantly shortened shoots and radicles, as well as reduced seedling biomass. Treating seeds with ferulic acid alone, vanillic acid alone, p-hydroxybenzoic acid alone, or a mixture of all phenolic acids significantly reduced all early growth parameters. The estimated effective dose for the 50% inhibition (ED50) of radicle growth in A. artemisiifolia seedlings was 368.39 ± 59.85 × 10−8 mol with ferulic acid, 135.41 ± 17.65 × 10−8 mol with p-coumaric acid, 810.36 ± 134.15 × 10−8 mol with p-hydroxybenzoic acid, and 160.11 ± 12.30 × 10−8 mol with the combination of all phenolic acids.

Gout is an oxidative stress-related disease. Food-derived vanillic acid, a promising xanthine oxidase inhibitor, could potentially be used as a safe, supportive, and therapeutic product for gout. The extraction of vanillic acid from a classic Chinese herbal plant Amomum villosum with ethanol was investigated in the study. The optimum conditions were determined as extraction time of 74 min, extraction temperature of 48.36 °C, and a solid-to-liquid ratio of 1:35 g·mL−1 using the Box–Behnken design (BBD) of response surface methodology (RSM). The experimental extraction yield of 9.276 mg·g−1 matched with the theoretical value of 9.272 ± 0.011 mg·g−1 predicted by the model. The vanillic acid in Amomum villosum was determined to be 0.5450 mg·g−1 by high-performance liquid chromatography–diode array detection (HPLC–DAD) under the optimum extraction conditions and exhibited xanthine oxidase (XO) inhibitory activity, with the half-maximal inhibitory concentration (IC50) of 1.762 mg·mL−1. The nanoemulsion of Amomum villosum extract consists of 49.97% distilled water, 35.09% Smix (mixture of tween 80 and 95% ethanol with 2:1 ratio), and 14.94% n-octanol, with a particle size of 110.3 ± 1.9 nm. The nanoemulsion of Amomum villosum extract exhibited markable XO inhibitory activity, with an inhibition rate of 58.71%. The result demonstrated the potential benefit of Amomum villosum as an important dietary source of xanthine oxidase inhibitors for gout.

Vanillic acid (VA) exhibited antioxidant and neuroprotective properties in some neurodegenerative disorders. So, the current study examined the neuroprotective potential of VA as an antiepileptic agent in pentylenetetrazole (PTZ)-induced epileptic rats and the prospective role of Insulin like growth factor-1 (IGF-1) and nuclear factor-2 erythroid-related factor-2 (Nrf2)/heme oxygenase-1 (HO-1) pathway in this respect. Thirty male albino rats were equally subdivided into 3 groups; (1) normal control (NC) group, (2) PTZ-group: received PTZ (50 mg/Kg, i.p. every other day) for 14 days, and (3) PTZ + VA group: received PTZ and VA (50 mg/Kg daily for 2 weeks). The seizure score and latency were evaluated after PTZ injection. Also, the markers of oxidative stress (malondialdehyde (MDA), catalase, and reduced glutathione (GSH)), histopathological examination, the expression of glial fibrillary acidic protein (GFAP) (a marker of astrocytes) IGF-1, Nrf2, and HO-1 were assessed in the brain tissues by the end of the experiment. PTZ caused significant decrease in seizure latency and significant increase in seizure score by the end of the experiment (p < 0.01). This was associated with significant increase in MDA and GFAP with significant decrease in GSH, total antioxidant capacity (TAC) and IGF-1 in brain tissues compared to normal group (p < 0.01). On the other hand, treatment with VA caused significant attenuation in PTZ-induced seizures which was associated with significant improvement in oxidative stress markers and downregulation in GFAP and upregulation of Nrf2, HO-1 and IGF-1 in CA3 hippocampal region (p < 0.01). VA showed neuroprotective and anti-epileptic effects against PTZ-induced epilepsy which probably might be due to its antioxidant properties and upregulation of Nrf2/HO-1 pathway and IGF-1.

To investigate the quality differences between the seeds and husks of Amomum villosum and explore the rationality of using the seeds without husks, this study determined the content of protocatechuic acid, vanillic acid, epicatechin, quercitrin, volatile oil, water extract, and ethanol extract. The 2,2-diphenyl-1-picrylhydrazyl(DPPH), 2,2-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid)(ABTS), and hydroxyl radical scavenging activities were determined to evaluate the antioxidant activities of seeds and husks. The quality differences between the seeds and husks were assessed through orthogonal partial least squares-discriminant analysis(OPLS-DA) and analytic hierarchy process(AHP) combined with the entropy weight method(EWM). Significant differences(P&lt;0.05) were observed in all 10 indicators between the seeds and husks. The levels of epicatechin, quercetin, and volatile oil were higher in the seeds, whereas those of protocatechuic acid, vanillic acid, water extract, and ethanol extract were higher in the husks. The seeds showed stronger scavenging ability against DPPH and ABTS radicals, while the husks showed a stronger scavenging effect on hydroxyl radicals. OPLS-DA significantly discriminated between the seeds and husks. Furthermore, volatile oil, water extract, DPPH radical scavenging rate, quercitrin, ABTS radical scavenging rate, hydroxyl radical scavenging rate, and vanillic acid were selected as the main differential indicators by variable importance in projection(VIP). Comprehensive scores calculated by AHP combined with EWM indicated that the seeds were superior to husks in terms of overall quality. However, there are still some dominant components and a certain antioxidant effect in the husks. Therefore, it is suggested to using Amomi Fructus with a certain amount of husks or utilizing the husks for other purposes.

Vanillic acid (VA), as a plant-derived phenolic acid compound, has widespread applications and good market prospects. However, the traditional production process cannot meet market demand. In this study, Pseudomonas putida KT2440 was used for de novo biosynthesis of VA. Multiple metabolic engineering strategies were applied to construct these P. putida -based cell factories, including the introduction of a Hs-OMT opt , engineering the cofactor S -adenosylmethionine supply pathway through the overexpression of metX and metH , reforming solubility of Hs-OMT opt , increasing a second copy of Hs-OMT opt , and the optimization of the fermentation medium. The resulting strain, XCS17, de novo biosynthesized 5.4 g/L VA from glucose in a fed-batch fermentation system; this is the highest VA production titer reported up to recently. This study showed that P. putida KT2440 is a robust platform for achieving the effective production of phenolic acids.

White-rot fungi, such as Phanerochaete chrysosporium , are the most efficient degraders of lignin, a major component of plant biomass. Enzymes produced by these fungi, such as lignin peroxidases and manganese peroxidases, break down lignin polymers into various aromatic compounds based on guaiacyl, syringyl, and hydroxyphenyl units. These intermediates are further degraded, and the aromatic ring is cleaved by 1,2,4-trihydroxybenzene dioxygenases. This study aimed to characterize homogentisate dioxygenase (HGD)-like proteins from P. chrysosporium that are strongly induced by the G-unit fragment of vanillin. We overexpressed two homologous recombinant HGDs, PcHGD1 and PcHGD2, in Escherichia coli . Both PcHGD1 and PcHGD2 catalyzed the ring cleavage in methoxyhydroquinone (MHQ) and dimethoxyhydroquinone (DMHQ). The two enzymes had the highest catalytic efficiency ( k cat / K m ) for MHQ, and therefore, we named PcHGD1 and PcHGD2 as MHQ dioxygenases 1 and 2 (PcMHQD1 and PcMHQD2), respectively, from P. chrysosporium . This is the first study to identify and characterize MHQ and DMHQ dioxygenase activities in members of the HGD superfamily. These findings highlight the unique and broad substrate spectra of PcHGDs, rendering them attractive candidates for biotechnological applications.IMPORTANCEThis study aimed to elucidate the properties of enzymes responsible for degrading lignin, a dominant natural polymer in terrestrial lignocellulosic biomass. We focused on two homogentisate dioxygenase (HGD) homologs from the white-rot fungus, P. chrysosporium , and investigated their roles in the degradation of lignin-derived aromatic compounds. In the P. chrysosporium genome database, PcMHQD1 and PcMHQD2 were annotated as HGDs that could cleave the aromatic rings of methoxyhydroquinone (MHQ) and dimethoxyhydroquinone (DMHQ) with a preference for MHQ. These findings suggest that MHQD1 and/or MHQD2 play important roles in the degradation of lignin-derived aromatic compounds by P. chrysosporium . Ethnopharmacological Relevance Lagotis Brachystachya: Maxim is a traditional ethnic medicine commonly used in Tibet. In Tibetan medicine theory, Lagotis brachystachya is mainly used for the treatment of inflammatory related diseases. However, the active components and mechanism of the anti-inflammatory activity of Lagotis brachystachya are not clear., Aim of the Study: The putative anti-inflammatory active compounds from Lagotis brachystachya Maxim and its anti-inflammation related mechanism involving in the TLR4/MyD88/NF-κB and NLRP3 signaling pathways were investigated., Materials and Methods: In this study, we investigated the anti-inflammatory activity and mechanism of 32 compounds extracted from Lagotis brachystachya in HepG2 and THP-1 cells using the alcohol-induced HepG2 cell injury model and the monosodium urate (MSU) combined with lipopolysaccharide (LPS)-induced THP-1 cell inflammation model., Results: The results found that six compounds, including Echinacoside, Quercetin, Homoplantaginin, Tricin-7-O-glucoside, Apigenin and Luteolin-7-O-beta-d-glucopyranoside, were shown to exhibit significant anti-inflammatory effects in both cell models. Furthermore, these compounds were shown to inhibit the TLR4/MyD88/NF-κB and NLRP3 signaling pathways and reduce the release of pro-inflammatory cytokines IL-1β, TNF-α, and IL-6 in both cell models., Conclusion: These findings suggest that Echinacoside, Quercetin, Homoplantaginin, Tricin-7-O-glucoside, Apigenin and Luteolin-7-O-beta-d-glucopyranoside from Lagotis brachystachya have promising potential as natural anti-inflammatory agents for the treatment of inflammatory-related diseases. Maize ( Zea mays L.) is an important crop in Argentina. Aspergillus section Flavi can infect this crop at the pre-harvest stage, and the harvested grains can be contaminated with aflatoxins (AFs). During the production of bioethanol from maize, AF levels can increase up to three times in the final co-products, known as, dry and wet distiller's grain with solubles (DDGS and WDGS), intended for animal feed. Fungal enzymes like laccases can be a useful tool for reducing AF contamination in the co-products obtained from this process. The aim of the present study was to evaluate the ability of laccase enzymes included in enzymatic extracts (EE) produced by different species in the Basidiomycota phylum to reduce AF (AFB 1 and AFB 2 ) accumulation under the conditions of in vitro assays. Four laccase activities (5, 10, 15, and 20 U/mL) exerted by nine isolates were evaluated in the absence and presence of vanillic acid (VA), serving as a laccase redox mediator for the degradation of total AFs. The enzymatic stability in maize steep liquor (MSL) was confirmed after a 60 h incubation period. The most effective EE in terms of reducing AF content in the buffer was selected for an additional assay carried out under the same conditions using maize steep liquor obtained after the saccharification stage during the bioethanol production process. The highest degradation percentages were observed at 20 U/mL of laccase enzymatic activity and 1 mM of VA, corresponding to 26% for AFB 1 and 26.6% for AFB 2. The present study provides valuable data for the development of an efficient tool based on fungal laccases for preventing AF accumulation in the co-products of bioethanol produced from maize used for animal feed.

Characterizing uranium (U) mine water is necessary to understand and design an effective bioremediation strategy. In this study, water samples from two former U-mines in East Germany were analysed. The U and sulphate (SO 4 2- ) concentrations of Schlema-Alberoda mine water (U: 1 mg/L; SO 4 2- seemed to influence the microbial diversity of the two water samples. Microbial diversity analysis identified U(VI)-reducing bacteria (e.g. Desulfurivibrio) and wood-degrading fungi (e.g. Cadophora) providing as electron donors for the growth of U-reducers. U-bioreduction experiments were performed to screen electron donors (glycerol, vanillic acid, and gluconic acid) for Schlema-Alberoda U-mine water bioremediation purpose. Thermodynamic speciation calculations show that under experimental conditions, U(VI) is not coordinated to the amended electron donors. Glycerol was the best-studied electron donor as it effectively removed 99% of soluble U, 95% of Fe, and 58% of SO 4 from the mine water, probably by biostimulation of indigenous microbes. Using electronic spectroscopy and quantum chemistry methods, the electronic spectra of vanillic acid in various solvents are interpreted. In the ground electronic state, four protolytic forms of vanillic acid in water were detected: anionic, dianionic, cationic, and neutral. In water, vanillin and isovanillin in the ground state have the same spectral characteristics: the line position and intensity in their absorption spectra coincide. The absorption and fluorescence bands of vanillic acid shift toward shorter wavelengths compared to vanillin and isovanillin. The values of the minimum electrostatic potential indicate that vanillic acid has the deepest minimum in the region of the carbonyl oxygen atom. The study of the fluorescence spectra showed that the radiative properties of the ionic forms of vanillic acid are different. The data of quantum chemical calculations indicate that electronic transitions in the molecule are formed with the participation of charge transfer from the phenyl part of the molecule to the oxygen atoms of the methoxy and carbonyl groups. The presence of OH, OCH3, and carbonyl group in the structure of vanillic acid leads to the existence of a dianionic form, both in the ground and electronically excited states. Diabetes mellitus (DM) is a metabolic illness and it is a result of hyperglycemia ensuing since a deficiency in insulin production, insulin function. Based on WHO statistics, more than 347 million peoples are suffered from the DM globally and it may increase to 694 million in 2030. Vanillic acid (VA) is a phenolic derived compound from dietary vegetations and fruits with the many biological acitivities. This current research was planned to investigate the antidiabetic and anti-inflammatory potential of VA on streptozotocin-challenged DM in rats. Later than an overnight fast, the rats were induced diabetes via one intraperitoneal treatment of STZ (40 mg/kg b.wt) in a newly organized 0.1 M citrate buffer. We analyzed body weight, plasma glucose, liver marker enzymes, insulin, carbohydrate metabolic enzymes, glycosylated hemoglobin, protein profiles, oxidant/antioxidant levels, inflammatory mediators and also evaluate histopathology of pancreatic tissues in normal control and investigational animals. Diabetic animals were revealed an increased in HbA1c, blood glucose, food and fluid intake and relative weight of kidney and liver, whereas the reduced level of insulin and body weight. Furthermore, augmented levels of ALT, glucose-6-phosphatase, AST, TBARS, TNF-α, creatinine, LOOH, fructose 1,6-bisphosphatase, ALP, IL-6 and IL-1β whereas, reduced status of antioxidant enzymes were observed in diabetic rats. Histopathological analysis of pancreatic tissues was in support of the biochemical parameters. Diabetic rats treated by VA significantly alter all these parameters toward normal levels. Abstract Background Lignin is a major component of plant biomass and is recalcitrant to degradation due to its complex and heterogeneous aromatic structure. The biomass-based research mainly focuses on polysaccharides component of biomass and lignin is discarded as waste with very limited usage. The sustainability and success of plant polysaccharide-based biorefinery can be possible if lignin is utilized in improved ways and with minimal waste generation. Discovering new microbial strains and understanding their enzyme system for lignin degradation are necessary for its conversion into fuel and chemicals. The Pandoraea sp. ISTKB was previously characterized for lignin degradation and successfully applied for pretreatment of sugarcane bagasse and polyhydroxyalkanoate (PHA) production. In this study, genomic analysis and proteomics on aromatic polymer kraft lignin and vanillic acid are performed to find the important enzymes for polymer utilization. Results Genomic analysis of Pandoraea sp. ISTKB revealed the presence of strong lignin degradation machinery and identified various candidate genes responsible for lignin degradation and PHA production. We also applied label-free quantitative proteomic approach to identify the expression profile on monoaromatic compound vanillic acid (VA) and polyaromatic kraft lignin (KL). Genomic and proteomic analysis simultaneously discovered Dyp-type peroxidase, peroxidases, glycolate oxidase, aldehyde oxidase, GMC oxidoreductase, laccases, quinone oxidoreductase, dioxygenases, monooxygenases, glutathione-dependent etherases, dehydrogenases, reductases, and methyltransferases and various other recently reported enzyme systems such as superoxide dismutases or catalase–peroxidase for lignin degradation. A strong stress response and detoxification mechanism was discovered. The two important gene clusters for lignin degradation and three PHA polymerase spanning gene clusters were identified and all the clusters were functionally active on KL–VA. Conclusions The unusual aerobic ‘-CoA’-mediated degradation pathway of phenylacetate and benzoate (reported only in 16 and 4–5% of total sequenced bacterial genomes), peroxidase-accessory enzyme system, and fenton chemistry based are the major pathways observed for lignin degradation. Both ortho and meta ring cleavage pathways for aromatic compound degradation were observed in expression profile. Genomic and proteomic approaches provided validation to this strain’s robust machinery for the metabolism of recalcitrant compounds and PHA production and provide an opportunity to target important enzymes for lignin valorization in future.

Its extracts have been reported to show various pharmacological effects such as hepatoprotection, antitumor, antiatopic dermatitis, antilipid peroxidation, antisteatotic, anti-inflammatory, and antiallergic activities. However, there have been no reports on the effect of H. dulcis extracts in relieving constipation so far. The aim of this study was to investigate the effects of a hot-water extract of the branches of H. dulcis (WEHD) on low-fiber diet-induced constipation in Sprague Dawley rats. The in vivo laxative activity of WEHD was assessed by measuring the intestinal transit of charcoal meal and stool parameters. Furthermore, the in vitro spasmogenic activity of WEHD was evaluated by monitoring the temporal profiles of contraction of rat colon in the absence or presence of WEHD. In addition, constituent profiling was conducted using high-performance liquid chromatography analysis. Pretreatment with WEHD significantly enhanced the intestinal transit of charcoal meal and increased the frequency and weight of stools in rats. In addition, the frequency and amplitude of contractile responses of isolated rat colon were markedly enhanced by WEHD. Two organic phenolic acids, ferulic and vanillic acids, were identified in WEHD, of which vanillic acid exhibited spasmogenic activity. To the best of our knowledge, this is the first study to report the laxative and spasmogenic activities of H. dulcis and its constituents, suggesting that WEHD can serve as a complementary and/or alternative laxative in alleviating chronic constipation. Phenolic acids are a group of compounds that are resistant to а biological treatment, making them pollutants of а priority concern. Vanillic acid (VA) is a reference for compounds present in the polyphenolic fractions, typically found in olive processing and wine-distillery plants. Important degradation yields are achieved. The photocatalytic degradation of VA is investigated using a titanium dioxide/UV irradiation (TiO2/UV) photocatalytic technique. The effect of different parameters on the degradation of VA in aqueous media is also studied using the experimental design methodology. The influence of TiO2 concentration, the solution pH value and irradiation time on the removal rate of VA is demonstrated. An efficiency of 96% is reached at acidic pH and low concentrations of TiO2 and VA. Liquid samples are analyzed by high-performance liquid chromatography/mass spectrometry (HPLC/MS) and gas chromatography/mass spectrometry (GC-MS), which reveal a partial degradation of VA proceeding through a series of aromatic and ring cleavage intermediates. Sorghum halepense L (Poaceae), ordinarily it is known as Johnson grass and locally as baru. This study was designed to find the vascular mechanisms underlying the hypotensive activity of S. halepense. In this study, effect of S. halepense seed extract/fractions on various blood pressure parameters were evaluated in normal and fructose induced hypertensive rats by invasive technique. Possible underlying hypotensive mechanism of active fraction was determined by using various pharmacological inhibitors. S. halepense extract/fractions vasorelaxant effect were also evaluated on rat aorta rings in organ bath and various intracellular signaling pathway inhibitors were used for determination of underlying mechanisms. S. halepense extract/fractions produced blood pressure lowering effect with most significant effect by its aqueous soluble fraction at dose of 10mg/kg. This effect was attenuated by pretreatment of atropine. Aqueous soluble fraction produced endothelium dependent vasorelaxation in rat aortic rings that was inhibited by pretreatment of atropine after phenylephrine induced contraction. The vasorelaxant effect of aqueous soluble fraction was attenuated by potassium channel blockers and also produced inhibitory effect on calcium entry through calcium channels. It also suppressed phenylephrine induced contraction like verapamil. By HPLC analysis found vanillic acid and naringinin in it. In conclusion, aqueous soluble fraction of S.halepense possess phytoconstituents which may be responsible for hypotensive and vasorelaxant effect of Sorghum halepense. [ABSTRACT FROM AUTHOR]

Objectives: Vanillic acid (VA) is a flavoring agent, a phenolic acid, and an intermediary by-product formed during transformation of ferulic acid to vanillin. It has been investigated for diverse pharmacological actions and used in Chinese medicine for decades. However, there is no information in the literature about its mechanism of toxicity or safety with long-term use. The present study will not only supply information on its pharmacological profile but also encourage evidence-based pharmacotherapeutic use. Hence, we performed a subacute toxicity study. Materials and Methods: According to the Organisation for Economic Co-operation and Development Test Guideline 407 (2008), 3 groups of rats were formed consisting of 12 rats (6 male and 6 female) in each group. For the subacute toxicity, the dose was chosen after a limit test was conducted. VA (1000 mg/kg/day) was orally administered for 2 weeks to the treatment group, whereas the control group received an equivalent volume of the vehicle. To assess reversibility, VA (1000 mg/kg/day, p.o.) was administered to the satellite group for 2 weeks and animals were observed for an additional 2 weeks after treatment. The adverse signs, variation in body weight, and mortality were evaluated throughout the study period. On days 15 and 29, blood was collected to evaluate essential biochemical and hematological parameters. The animals were subsequently weighed and sacrificed. The weights of internal organs were recorded; gross necroscopy and histopathological studies were performed. Results: The hematological parameters of the satellite group increased and the serum sodium level decreased after the treatment. Satellite groups showed no other major change in biochemical parameters when compared to the control group. In addition, relative organ weights, gross necropsy examinations and histopathological structure of the internal organs showed no major alterations. Conclusion: VA showed no adverse effect on the process of leukopoiesis, erythropoiesis or on internal organs, as verified by hematological and biochemical evaluations, gross necropsy, and histopathological studies. Malaria is a vector borne disease responsible for high morbidity, mortality and poverty in many tropical and subtropical countries. The disease is transmitted through a bite from an infected female Anopheles mosquito, amongst which Anopheles gambiae s.s serves as the most prevalent vector. The control of An. gambiae s.s population can therefore lead to a reduction in malaria spreading. Previous studies have reported the crude extracts of Kotschya thymodora to be active against the larvae of An. gambiae s.s and Culex quinquefasciatus. In this report the phytochemical study on the crude aqueous ethanolic extract of K. thymodora leaves led to isolation of vanillic acid (1) and protocatechuic acid (2). The structures of these compounds and mosquitoes larvicidal activity against An. gambiae s.s were established by using spectroscopic techniques and WHO protocol of 1996 respectively. The two phenolic acids exhibited a moderate mosquito larvicidal activity with LC50 of 77.35 µg/mL (vanillic acid, 1) and 62.4 µg/mL (protocatechuic acid, 2) after 48 hrs exposure time. Rational metabolic-flow switching, which we proposed recently, is an effective strategy to produce an exogenous high-value natural product using transformed plant cells; the proof of this concept was demonstrated using bamboo (Phyllostachys nigra ; Pn) cells as a model system. Pn cells were transformed to express 4-hydroxycinnamoyl-CoA hydratase/lyase of Pseudomonas putida KT2440 (PpHCHL), which catalyzes the formation of 4-hydroxybenzaldehyde and vanillin from p -coumaroyl-CoA and feruloyl-CoA, respectively. The PpHCHL-transformed cells accumulated glucose conjugates of 4-hydroxybenzoic acid and vanillic acid, indicating that the PpHCHL products (aldehydes) were further metabolized by inherent enzymes in the Pn cells. The production titers of 4-hydroxybenzoic acid glucose ester, vanillic acid glucose ester, and 4-hydroxybenzoic acid glucoside reached 1.7, 0.17, and 0.14 g/L at the maximum, respectively. The therapeutic effect of phenolics on neurodegenerative diseases has been attributed to their potent antioxidant properties. In the present study, the neuroprotective activities of vanillin and vanillic acid were investigated in Fe2+- induced oxidative toxicity in brain tissues by investigating their therapeutic effects on oxidative imbalance, cholinergic and nucleotide-hydrolyzing enzymes activities, dysregulated metabolic pathways. Their cytotoxicity was investigated in hippocampal neuronal cell lines (HT22). The reduced glutathione level, SOD and catalase activities were ameliorated in tissues treated with the phenolics, with concomitant depletion of malondialdehyde and nitric oxide levels. They inhibited acetylcholinesterase and butyrylcholinesterase activities, while concomitantly elevated ATPase activity. Treatment with vanillin led to restoration of oxidative-depleted metabolites and reactivation of the pentose phosphate and purine metabolism pathways, with concomitant activation of pathways for histidine and selenoamino metabolisms. While vanillic acid restored and reactivated oxidative-depleted metabolites and pathways but did not activate any additional pathway. Both phenolics portrayed good binding affinity for catalase, with vanillic acid having the higher binding energy of −7.0 kcal/mol. Both phenolics were not cytotoxic on HT22 cells, and their toxicity class were predicted to be 4. Only vanillin was predicted to be permeable across the blood brain barrier (BBB). The crystal structures of the antimicrobial drug tinidazole [TNZ; systematic name: 1‐(2‐ethylsulfonylethyl)‐2‐methyl‐5‐nitroimidazole, C8H13N3O4S] and the 1:1 cocrystal of TNZ with the naturally occurring compound vanillic acid (VA; systematic name: 4‐hydroxy‐3‐methoxybenzoic acid, C8H8O4), namely, the TNZ–VA cocrystal, were determined by single‐crystal X‐ray analysis at 100 K. The supramolecular structure of the TNZ–VA cocrystal is composed of a carboxylic acid dimer and an O—H...N(heterocycle) synthon in the form of layers made up of O—H...N and O—H...O hydrogen bonds. The layers are joined via C—H...O hydrogen bonds, π–π stacking and C—H...π interactions. The energy framework analysis, together with interaction energy calculations using the DLPNO‐CCSD(T) method, indicates that the TNZ–VA cocrystal inherits strong interactions from the TNZ and VA crystals, which accounts for the enhanced thermal stability and reduced dissolution rate. To study on chemical constituents and bioactivity of Larix gmelinii needles, separation and purification of ethyl acetate extract from Larix gmelinii needles were carried out by silica gel column chromatography，semi-preparative high performance liquid chromatography，and re-crystallization. The structures of isolated compounds wrere identified by analysis of spectra of MS，NMR，and compared wdth the reported conferences. The antibacterial activity of the extract ■was also tested. The results 'were as follo'ws: Compounds 1-15 wrere isolated from the ethyl acetate extract and identified as Larixol ( 1)，( 2R)-5, 4,-dihydroxy-6-methyl-7-methoxy flavone ( 2 ), 2', 4,-dihydroxy-4, 6,-dimethoxydihydro- chalcone (3) , 2,,4-dihydroxy-4,,6,-dimethoxy-chalcone (4) , 2,,4,-dihydroxy-4,6,-dimethoxy-chalcone (5) , isorham- netin (6)，4'，5-dihydroxy-7-methoxy-8-methyl flavone (7)， kaempferol (8)， p-sitosterol (9)， stigmasterol (10)， daucosterol (11)，vanillic acid (12)，hydroxyl-benzoic acid (13)， dimethyl - matairesinol (14)，and 15-nonacosanol (15). Compounds 2，4，5，and 7 were isolated from the genus Pinus for the first time. The result of antibacterial activity indicated that the inhibition rates of ethyl acetate extract to Escherichia coli，Bacillus subtilis，B. cereus，and Staphylococcus aureus were 55%-70%，53%-72%，61%-71%，33%-65%，respectively，at the concentration range of 5-100 mg • mL 1. Objective: The pathological changes of urinary tract were observed after the treatment of infectious stone by vanillic acid. Methods: 40 male Wistar rats were randomly divided into blank control group, infectious stone group, vanillic acid group and acetohydroxamic acid group. Blank control group: 4 male Wistar rats were fed without special treatment. Infectious Stone Group: 12 male Wistar rats were implanted with 4 mm bacteria bearing foreign body in the front of the polyurethane vein indwelling needle infected with Proteus mirabilis into the bladder to induce the formation of infection stone. In the vanillic acid group, after the model were establishmented, 12 male Wistar rats were treated with vanillic acid at 50mg/ kg•d for 3 weeks. Acetohydroxamic acid group: after the model were establishmented, 12 male Wistar rats were treated with acetohydroxamic acid at 12.5mg/kg•d for 3 weeks. The rats were killed 21 days later.The kidney, ureter and bladder were fixed in 10% paraformaldehyde solution for 24 hours. The specimens were embedded in paraffin, and the pathological analysis was carried out after HE and immunohistochemical staining. Results: In addition to the blank control group, the other three groups of rats had calculus formation in the bladder. No obvious pathological changes were found in the kidney, ureter and bladder in the blank control group, and inflammatory reactions were found in the other three groups: severe urinary tract inflammation was found in the infectious stone group, followed by acetylhydroxamic acid group. In the vanillic acid group, the pathological changes of urinary tract were the lightest, with a small amount of inflammatory cell infiltration and mucosal hyperplasia, but no serious pathological changes such as mucosal loss. The expression of Tamm-Horsfall Protein (THP) in the ascending branch of loop of Henle and the proximal part of distal tubule in vanillic acid group was not significant compared with that in the infectious stone group and acetohydroxamic acid group. Ferulic acid is a fraction of the phenolics present in cereals such as rice and corn as a component of the bran. Substantial amounts of waste bran are generated by the grain processing industry and this can be valorized via extraction, purification and conversion of phenolics to value added chemical products. Alkaline alcohol based extracted and purified ferulic acid from corn bran was converted to vanillic acid using engineered Pseudomonas putida KT2440. The strain was engineered by rendering the vanAB gene nonfunctional and obtaining the mutant defective in vanillic acid metabolism. Biotransformation of ferulic acid using resting Pseudomonas putida KT2440 mutant cells resulted in more than 95 ± 1.4% molar yield from standard ferulic acid; while the corn bran derived ferulic acid gave 87 ± 0.38% molar yield. With fermentation time of less than 24 h the mutant becomes a promising candidate for the stable biosynthesis of vanillic acid at industrial scale. [ABSTRACT FROM AUTHOR]

Aim: The present investigation was aimed at isolating and identifying bacterial strains from cured vanilla beans. Additionally, the study focused on evaluating bacterial processes pertaining to the aromatic compounds production (ACP). Methods and Results: Three bacteria were isolated from Vanilla planifolia beans, previously subjected to the curing process. According to morphological, biochemical and 16S rRNA analysis, the strains were identified as Citrobacter sp., Enterobacter sp. and Pseudomonas sp. The polygalacturonase activity (PGA) was determined using the drop, cup‐plate and DNS methods. Aromatic compounds production was analysed by cup‐plate method using FA as substrate and quantified by high performance liquid chromatography (ppm), the functional groups of vanillic acid (VA) were identified by FT‐IR and the aromatic compounds (AC) resistance was determined and reported as minimum inhibitory concentration. Citrobacter sp., Enterobacter sp. and Pseudomonas showed PGA (70·31 ± 364, 76·07 ± 12·47 and 51 ± 10·92 U ml−1 respectively), were producers of VA (3·23 ± 0·49, 324 ± 41 and 265·99 ± 11·61 ppm respectively) and were resistant to AC. Conclusions: The Gram‐negative bacteria isolated from V. planifolia beans were responsible for ACP. Significance and Impact of the Study: This is the first evidence for the role of Gram‐negative bacterial isolates from cured Mexican V. planifolia beans in the process related to ACP. Many studies have evaluated the antimicrobial activity of natural products against various microorganisms, but to our knowledge there have been no studies of the possible use of natural products for their antimicrobial activity against Enterobacter hormaechei. In this study, we investigated vanillic acid (VA) for its antimicrobial activities and its modes of action against carbapenem-resistant E. hormaechei (CREH). The MIC of VA against CREH was determined by the agar diffusion method. The antibacterial action of VA against CREH was elucidated by measuring variations in intracellular ATP concentration, intracellular pH, membrane potential, and cell morphology. Moreover, the efficacy of VA against biofilm formation and VA damage to CREH cells embedded in biofilms were further explored. Our results show that VA was effective against CREH with a MIC of 0.8 mg/mL. VA could rupture the cell membrane integrity of CREH, as measured by a decrease of intracellular ATP, pH, and membrane potential, along with distinctive alternations in cell morphology. In addition, VA exerted a remarkable inhibitory effect on the biofilm formation of CREH and also killed CREH cells within biofilms. These findings show that VA has a potent antibacterial and antibiofilm activity against CREH and, hence, has the potential to be used clinically as a novel candidate agent to treat CREH infections and in the food industry as a food preservative and surface disinfectant. Main conclusion: During their domestication process, the species of the genus Opuntia lose their ability to survive in the wild. Presence and concentration of secondary metabolites which play a role in the interaction with their surroundings are modified but without an identifiable pattern. A domestication gradient based on morphological characteristics has been previously described for the species in the Opuntia genus. Secondary metabolites are a diverse group of bioactive compounds that relate to a species evolution, both in their natural and artificial (domestication process) selection environments. In addition, these compounds are associated with plant resistance to stress when growing in the wild. A comprehensive characterization of secondary metabolite profiles in the Opuntia genus that accounts for the genotypic differences related to the degree of domestication has not previously been conducted. This study evaluated the phytochemical composition of young cladodes from fifteen variants, of O. ficus-indica, O. albicarpa Sheinvar, and O. megacantha Salm-Dyck, identified as species with a highly advanced, advanced and intermediate degree of domestication, respectively, and O. hyptiacantha A. Web, and O. streptacantha Lem. identified as wild-intermediate and wild species. Analyses were carried out using a HPLC-diode array detection technique. Out of the 13 identified and quantified phenolic molecules and terpenoids, only the caffeic, ferulic and syringic acids, and the terpenoid β-amyrin were present in all variants. The flavonoid luteolin was absent in all five species. Gallic, vallinic, p-hydroxybenzoic, chlorogenic and p-coumaric acids were only present in 53–87% of variants; flavonoids quercetin, isorhamnetin, rutin and apigenin in 47–87% of the variants. Both, oleanolic acid and peniocerol, were present only in 60% of variants. Isorhamnetin was absent in O. hyptiacantha and quercetin in O. streptacntha. Differences and similarities in the secondary metabolites content showed no recognizable trend relating to the degree of domestication across the species in this genus. [ABSTRACT FROM AUTHOR]
