Objetivo: Desenvolver e validar quanto ao conteúdo e à aparência uma interface virtual (jogo) destinada à conscientização e relaxamento da musculatura do assoalho pélvico e verificar seu nível de usabilidade e satisfação em mulheres saudáveis. Métodos: Essa pesquisa foi desenvolvida em 4 fases. Na fase 1, o conteúdo para estruturar a interface virtual foi definido por meio de consulta aos fisioterapeutas (especialistas em reabilitação do assoalho pélvico) e de pesquisa em base de dados para averiguar os protocolos clínicos, as características específicas da população alvo e o tratamento vigente baseado em evidência. Na Fase 2, o jogo virtual foi desenvolvido para interagir com sinais eletromiográficos captados a partir de eletrodos posicionados na região perianal. Na Fase 3, foi realizada a validação quanto ao conteúdo e aparência do jogo desenvolvido por meio de fisioterapeutas experientes. Foi adotado um nível de concordância mínimo de 80% para determinar a validade dos itens. Na fase 4, foi realizada a avaliação do nível de usabilidade e o grau de satisfação do jogo virtual. O nível de usabilidade foi avaliado através do MATcH (Checklist para Avaliação da Usabilidade de Aplicativos para Celular Touchscreen) e a satisfação foi avaliada através de uma escala visual analógica. Resultados: O jogo foi desenvolvido em duas dimensões, baseado em atividade de paraquedismo a partir de um salto de um avião. O paraquedas simulou a atividade dos músculos do assoalho pélvico: fechado correspondendo a uma contração e aberto ao relaxamento. O objetivo do jogo foi traçado para que o avatar chegasse a um alvo percorrendo o máximo de tempo possível com o paraquedas aberto (musculatura relaxada) após o salto (contração muscular). Dos 15 itens que representavam o conteúdo e aparência postos à validação, quatro precisaram de uma segunda ronda para alcançar o nível de concordância desejado. O jogo apresenta 3 níveis de dificuldade (fácil, médio e difícil) de acordo com a percentagem de relaxamento exigida em relação à contração voluntária máxima e como motivação o usuário pode coletar moedas denominadas “perinas” durante o jogo. O jogo apresentou um alto nível de usabilidade (Média: 63,04; 5,28 DP) e o nível de satisfação do jogo virtual mostrou uma média de 9,7 (DP=0,55) e não houve relato de efeito adverso durante o teste de usabilidade. Conclusão: Desenvolveu-se um jogo virtual para o treino de relaxamento da musculatura do assoalho pélvico que apresentou validade de conteúdo e aparência adequado e alto nível de usabilidade e satisfação entre as usuárias. É possível portanto, sugerir que o jogo desenvolvido possui uma promissora aplicabilidade na prática clínica, após futuros testes em ensaios clínicos. Contextualization: Virtual reality has been used as a treatment approach in several areas of physical therapy, including the muscular re-education of pelvic floor dysfunctions. Objective: Develop and validate regarding content and appearance a virtual interface (game) aimed at the awareness and relaxation of the pelvic floor musculature and verify the level of usability and satisfaction in healthy women. Methods: This research was developed in 4 phases. In stage 1, the content to structure the virtual interface was defined through consultation with physiotherapists experienced in pelvic floor rehabilitation and research based on data to ascertain the clinical protocols, the specific characteristics of the target population and the current evidence-based treatment. In Phase 2, the virtual game was developed to interact with electromyographic signals captured from electrodes positioned in the perianal region. In Phase 3, the validation was done regarding the content and appearance of the developed game. Through experienced physiotherapists who were asked to judge the content and appearance of the virtual game. A minimum level of agreement of 80% was adopted to determine the validity of the items. In phase 4, the level of usability and the degree of virtual game satisfaction were evaluated. The level of usability was evaluated through MATcH (Checklist for Evaluation of the Usability of Touchscreen Mobile Applications) and the satisfaction was evaluated through a visual analogue scale. Results: The game was developed in two dimensions, based on parachuting activity from a jump of an airplane. The parachute simulated the activity of the pelvic floor muscles: closed corresponding to a contraction and open to relaxation. The goal of the game was to have the avatar reach a target by running as long as possible with the open parachute (relaxed musculature) after the jump (muscle contraction). Of the 15 items that represented content and appearance put to validation, four required a second round to achieve the desired level of agreement. The game presents 3 levels of difficulty (easy, medium and difficult) according to the percentage of relaxation required in relation to maximum voluntary contraction and as a motivation the user can collect coins called "perinas" during the game. The game presented a high level of usability (Average: 63.04; 5.28 SD) and the level of virtual game satisfaction showed an average of 9.7 (0.55 SD) and there was no adverse effect during the game usability testing. Conclusion: A virtual game was developed for the relaxation training of the pelvic floor musculature that presented content validity and adequate appearance and a high level of usability and satisfaction among the users. It is therefore possible to suggest that the developed game has a promising applicability in clinical practice, after the future tests of clinical trials

Data loggers with sensors for salinity were installed in five sites in the North and South Bays, at 1.5 and at three meters depth. These sites were monitored during an event of intense rain when, in four days, almost twice the average rainfall expected for the whole month of September was registered. Salinities at 1.5 meter depth were lower than at three meters depth, in three of five monitored sites. The implications of such results for the shellfish aquaculture are discussed, based on the depths of the aquaculture areas established by the Brazilian government in the State of Santa Catarina. Resumo: Registradores de dados com sensores para salinidade foram instalados em cinco pontos de monitoramento nas Baías Norte e Sul, a 1,5 e a três metros de profundidade. Esses pontos foram monitorados durante um evento de chuva intensa, quando foi registrado em quatro dias quase o dobro da precipitação média esperada para o mês de setembro. Salinidades obtidas a 1,5 metros foram inferiores às registradas a três metros de profundidade, em três dos cinco pontos de monitoramento. Implicações desses resultados para os cultivos de moluscos são discutidas com base na profundidade dos parques aquícolas estabelecidos pelo Governo federal em Santa Catarina.

Tese (doutorado) - Universidade Federal de Santa Catarina, Centro de Ciências Agrárias, Programa de Pós-Graduação em Aqüicultura, Florianópolis, 2017. A presente tese é composta por quatro capítulos. No primeiro é feita a contextualização dos objetivos da pesquisa. O segundo apresenta uma revisão das práticas de monitoramento em vigor no Brasil, na União Europeia e nos Estados Unidos da América para a classificação de áreas de produção de moluscos. Os sistemas de classificação não são imediatamente comparáveis porque alguns são baseados no monitoramento de água e outros da carne dos moluscos. Para criar uma base comum de comparação, modelos de regressão foram desenvolvidos para correlacionar os níveis de organismos indicadores fecais (FIOs) na água e nos moluscos. Diferenças importantes nos níveis de segurança proporcionados pelas classificações estabelecidas pelas diferentes legislações foram identificadas. O terceiro capítulo descreve uma metodologia para o desenvolvimento de modelos de regressão para prever os níveis de FIOs em áreas costeiras influenciadas por fontes de poluição localizadas em múltiplas bacias hidrográficas. Concentrações de FIOs em mexilhões (Perna perna) e em água marinha medidas em 50 locais nas baías da Ilha de Santa Catarina (SC, Brasil) entre agosto de 2012 e outubro de 2013 foram utilizadas no estudo. Modelos de regressão foram utilizados para estudar as relações entre as variações espaciais dos níveis de FIOs e características geográficas de bacias hidrográficas próximas aos pontos de monitoramento. Ao considerar na análise bacias hidrográficas com exutórios localizados em até 3 km dos pontos de monitoramento foi possível aumentar em até 150% o coeficiente de determinação (R2) dos modelos. Variáveis meteorológicas foram testadas para prever a variação temporal nos níveis de FIOs. Para otimizar os modelos, o efeito cumulativo desses parâmetros foi avaliado considerando diferentes janelas de tempo antes das coletas. Essa abordagem permitiu aumentar o R2 dos modelos em até 160%. No quarto capítulo, um único modelo é desenvolvido para prever tanto variações espaciais quanto temporais dos níveis de FIOs. O referido modelo é validado e usado para prever a poluição fecal nas baías em diferentes cenários. Foram comparados 288 pares de resultados medidos/modelados, dados obtidos de março de 2009 a abril de 2010 em 15 pontos de monitoramento. Os resultados da validação indicaram que o modelo fornece uma representação justa dos níveis de FIOs no ambiente de estudo dentro das condições meteorológicas sob as quais o modelo foi treinado. Abstract : This thesis is composed of four chapters. The first one contextualizes the objectives of the research. The second one presents a review of the monitoring practices for classification of commercial shellfish production areas in force in Brazil, European Union and United States of America. The classification systems are not immediately comparable principally because some regulations are based on the monitoring of water and others of shellfish flesh. To create a common baseline to compare these regulations, regression models were developed based on monitoring data and used to correlate levels of faecal indicator bacteria in water and in shellfish. Important differences in the levels of shellfish safety provided by the classifications in the different legislations were identified. The third chapter describes a methodology for developing regression models to predict the FIO levels in coastal areas influenced by pollution sources located in multiple catchments. Concentrations of faecal indicator organisms (FIO) in mussels (Perna perna) and water sampled from 50 sites in the North and South bays of Santa Catarina (SC, Brazil) from August 2012 to October 2013 were used in the study. Regression models were used to study the relationships between the spatial variations of FIO levels and geographic characteristics of catchments located near the monitoring points. By considering in the analysis catchments with outlets within three kilometres from the monitoring points, it was possible to increase the coefficients of determination (R2) of the models up to 150%. Meteorological variables were tested to predict the temporal variation in the FIO levels. To optimize the models, the cumulative effect of these parameters was evaluated considering different time windows prior to samplings. This approach allowed increasing the models? R2 up to 160%. In the fourth chapter, a single model is developed to predict both the spatial and the temporal variations in the FIO levels. The referred model is validated and applied to predict the FIO levels in the bays under different scenarios. The analysis considered 288 pairs of measured/modelled results to validate the model, data obtained from March 2009 to April 2010 regarding 15 monitoring points. The validation results indicated that the model provides a fair representation of the FIO levels in the studied environment under the meteorological conditions under which the model was trained.

Instituto Brasileiro de Geografia e Estat??stica (Censo 2010), Brazil has 9.7 million Brazilians with some degree of hearing impairment or deafness, more than five per cent of the population. For the majority of those persons the main natural communication method is the Brazilian Sign Language (LIBRAS ??? L??ngua Brasileira de Sinais), instead of the spoken Portuguese. The computer-aided recognition of signs aims to expand the social and digital inclusion of the deaf community by means of those signs translation into audio or text format. This work presents one of the global LIBRAS parameters recognition, the hand gestures, using k-nearest neighbor???s classifier and 2D??PCA dimensionality reduction technique. A robust and representative data set of daily conditions, with 12.200 depth images from 61 hand gesture, captured by Kinect?? sensor, was built. Initially the images were segmented in a preprocessing step, which isolated the region of the right hand from the rest of the body. Seeking to eliminate the data set redundancy, the images were submitted to a dimensionality reduction by (2D) dimensional 2PCA technique determining the most representative forms of data from original pixels linear combination. The classifier k-nearest neighbors was the technique in the final stage in the hand gesture automatic recognition. This classifier could correctly categorize 96.31% of test samples (k = 1 and 10x10 feature matrix). The study was conducted with shoot tip explants of neem (Azadirachta indica A. Juss) to identify a viable regenerative process. Shoot tips were obtained from neem embryos cultured alternatingly in DKW medium supplemented with BAP and medium without hormones. Initial shoot development was influenced by cotyledon presence. Basal callus, excised from in vitro stem base, also presented organogenic potential. In some cases, plant lines, obtained from each seed, presented different characteristics. The most common characteristic observed in vitro was callus formation at the stem base. However, the rarest characteristics were stem callus formation and leaf senescence. The regenerated shoot tips were further subculture and rooted on a medium supplemented with IBA so that complete plants could be obtained. The rooted plants were transplanted to a greenhouse and successfully acclimatized. No significant differences in in vivo development were observed between neem plants from callus and from shoot tip propagation. RESUMOO estudo foi realizado com explantes de ápices caulinares de nim (Azadirachta indica A. Juss) para identificar um processo viável de regeneração. Brotos foram obtidos a partir de embriões de nim cultivados alternadamente em meio DKW suplementado com BAP e meio sem hormônios. O desenvolvimento inicial dos brotos foi influenciada pela presença dos cotilédones. Calos basais, isolados in vitro a partir da base dos caules, também apresentam potencial de organogênese. Em alguns casos, as linhagens de plantas, obtidas a partir de cada semente, apresentaram características diferentes. A característica mais comum observada in vitro foi a formação de calos na base do caule. Entretanto, as características mais raras foram a formação de calo ao longo de todo o caule e a ocorrência de senescência foliar. Os apices caulinares regenerados foram posteriormente micropropagadados e enraizados em um meio suplementado com IBA, permitindo a obtenção de plantas completas. As plantas enraizadas foram transferidas para casa de vegatação e aclimatizadas com sucesso. Nenhuma diferença no desenvolvimento in vivo foi observada entre plantas obtidas de calos e da propagação de ápices caulinares de nim.

Cerca de 6 milhões de brasileiros estão infectados e destes 5 a 10% podem apresentar as formas mais graves da doença. Entretanto, o desenvolvimento de resistência à infecção pelo Schistosoma mansoni representada por baixa carga parasitária em indivíduos residentes em áreas endêmicas, vem despertando bastante interesse e controvérsia. Através da citometria de fluxo, foram avaliados os perfis dos linfócitos T (CD4 e CD8) e linfócitos B (CD19), grau de ativação celular e produção de citocinas dos padrões de respostas Th1 (IFN-), Th2 (IL-5) e regulatório (IL-10) em indivíduos com diferentes cargas parasitárias e a possível associação no desenvolvimento da resistência à infecção. Além disso, neste estudo foi avaliado a resposta imune dos indivíduios resistentes à infecção participação dos antígenos candidatos vacinais, SWAP, rSm29 e rSm22.6. Foi observado que os indivíduos com baixa carga parasitária apresentam padrão de resposta misto do tipo Th1, Th2 e regulatório. Aliado a isto, os indivíduos resistentes apresentaram maior expressão de moléculas coestimulatórias (CD28 e CTLA-4). Na avaliação das células de memória, os indivíduos com baixas cargas parasitárias apresentam maiores proporções de células memória TCD4+. Por outro lado, sugere-se que a susceptibilidade à infecção e a esteja associada a uma possível exaustão do pool de células de memória central e aumento no compartimento de células de memória efetora. Contudo, não foram observadas diferenças significativas nos fenótipos das células T e B e também na produção de citocinas pelas CMSP após estímulo dos antígenos testados neste estudo. Schistosomiasis is an important parasite infection that impacts on socio-economic and public health aspects. About 6 million Brazilians are infected and 5 to 10% may experience the most severe forms of the disease. However, the development of resistance to infection by Schistosoma mansoni represented by low parasite burden, in individuals living in endemic areas, is of considerable interest and controversy. Using flow cytometry method were evaluated the profile of T lymphocytes (CD4 and CD8) and B lymphocytes (CD19) and the degree of cellular activation and cytokine production of Th1 (IFN-), Th2 (IL-5) and regulatory (IL-10) patterns in patients with different parasitic loads and possible association with the development of resistance to the infection. Moreover, in this study we evaluated the immune response of resistant individuals to the Schistosoma candidate vaccine antigens, SWAP, rSm29 and rSm22.6. It was observed that individuals with low parasite burden presented a mixed type of imune response, characterized by the Th1, Th2 and regulatory response. Added to this, the resistant individuals had a higher expression of costimulatory molecules (CD28 and CTLA-4). In relation to the memory cells, individuals with low parasitic loads had higher proportions of CD4+ T memory cell. Therefore, it is suggest that susceptibility to infection is associated with a possible exhaustion of the main memory cell pool and increased of effector memory cells compartment. There was, however, no significant differences in the phenotypes of B and T cells and cytokine production by PBMC stimulated by the S. mansoni antigens used in this study.

Our research has as its object of study aspects of mathematic formation at Polo Paracambi and the uses of the virtual platform as a learning environment. Data analysis was done in two stages and the following data collection instruments were used: the subjects registration forms, document analysis and monitoring in CEDERJ platform and the virtual environment of each discipline. At first stage we analyzed the profile of the entrants in the first half of 2012 in the course of Mathematics, research subjects. We observed that most of future teachers consists of young people aged between 22 and 26 years old, from public schools and residents on the metropolitan area from Rio de Janeiro city. We also show the use of the students in the courses Introduction to Computers, Basic Mathematics and Plane Geometry. In the second phase, based on the results obtained in the first stage, we selected seven undergraduates and captured their participation in the virtual environment. Particularly, we analyze the access in the courses Pre-Calculus, Discrete Mathematics and Didactics of Geometry. We found that the majority of the accesses occurred during the periods of assessment (distance or face-to-face) and, for the course Didactics of Geometry the number of access had kept constant for the role semester. Sabendo do crescimento da Educa??o a Dist?ncia e dos benef?cios trazidos pelos avan?os tecnol?gicos a esta modalidade, o CEDERJ tem investido no desenvolvimento profissional de professores. Nossa investiga??o tem como objeto de estudo aspectos da forma??o matem?tica no Polo Paracambi e tem como um dos focos o uso da Plataforma CEDERJ como ambiente virtual de aprendizagem na Licenciatura. A an?lise dos dados foi feita em dois momentos e foram usados os seguintes instrumentos de coleta: fichas cadastrais dos sujeitos, an?lise documental e acompanhamento na Plataforma CEDERJ e no ambiente virtual de tr?s Disciplinas. No primeiro momento, analisamos o perfil dos ingressantes no primeiro semestre de 2012 no curso de Matem?tica, sujeitos da pesquisa. Observamos que sua maioria ? composta por jovens na faixa et?ria entre 22 e 26 anos, provenientes das escolas p?blicas e moradores da regi?o metropolitana do Estado do Rio de Janeiro. Mostramos tamb?m o aproveitamento dos graduandos nas disciplinas Introdu??o ? Inform?tica, Matem?tica B?sica e Geometria Plana. No segundo momento, com base nos resultados obtidos na primeira fase, selecionamos sete graduandos e acompanhamos a sua participa??o no cen?rio virtual. Bibliografia: f. 268-290 Resumo: Esta tese propõe uma discussão sobre o processo de mudança na política nacional de segurança pública durante os governos Fernando Henrique Cardoso (1994 2002) e Luís Inácio Lula da Silva (2003 2010), a partir do Referencial de Coalizões de Defesa (RCD), modelo teórico proposto por Sabatier e Jenkins-Smith. Tendo como pressuposto que as políticas públicas são estruturadas em subsistemas formados por atores individuais e coletivos, organizações públicas e privadas, que lidam com uma determinada área ou problema de ordem pública, o RCD permite verificar a ocorrência de mudanças em subsistemas de política, a partir de uma investigação sobre alianças ou coalizões (formais e informais) e suas influências no modo como as políticas são formuladas. Assim, o estudo está centrado no papel desempenhado por empreendedores da política nacional de segurança pública (localizados na Secretaria Nacional de Segurança Pública do Ministério da Justiça), em articulação com setores do mundo acadêmico e da sociedade civil que, com novas ideias e crenças, formaram uma coalizão capaz de alterar o subsistema dessa política pública. A tese identifica ainda a coalizão conservadora que, ao longo de todo o período, defende a manutenção do status quo do subsistema de segurança pública brasileiro. O estudo acerca da política nacional de segurança pública está lastreado na crença segundo a qual a promoção da cidadania e a incontestabilidade dos direitos humanos, nas sociedades democráticas, estão relacionadas à efetividade da política de segurança pública, sendo que são poucas as produções acadêmicas brasileiras que tratam dessa questão. Ademais, o trabalho possibilitou a identificação não somente dos principais atores e ideias/crenças que definiram os rumos dessa política nas últimas duas décadas, mas, originalmente, as coalizões que disputaram espaços de poder e influência num subsistema que ainda carece de reformas substantivas numa sociedade que, mais democrática e plural, exige maior eficiência e legitimidade da política de segurança pública haja vista os péssimos indicadores de crime, responsáveis, em grande medida, pela violência quase generalizada que coloca em risco a cidadania e, por consequência, a própria ordem democrática em nosso país. Abstract: This thesis proposes a discussion on the process of change in national policy on public safety during the governments of Fernando Henrique Cardoso (1994 2002) and Luiz Inácio Lula da Silva (2003 - 2010), through the Advocacy Coalition Framework (ACF) model proposed by Sabatier and Jenkins-Smith. Based on the assumption that public policies are structured into subsystems formed by individual and collective actors, public and private organizations, that deal with a particular area or issue of public policy, the ACF allows us to verify the occurrence of changes in policy subsystems, from an investigation into alliances or coalitions (formal and informal) and their influence on how policies are formulated. Thus, the study is focused on the role played by entrepreneurs of the national policy for public safety (located on the National Secretariat of Public Safety of the Ministry of Justice), in conjunction with sectors of the academic world and the civil society, with new ideas and beliefs, formed a coalition able to change this policy subsystem. The thesis also identifies the conservative coalition that, throughout the period studied, supports maintaining the status quo of the Brazilian public safety subsystem. The study on the national safety policy is based on public belief that the promotion of citizenship and incontestability of human rights, in democratic societies, are related to the effectiveness of public safety policies and there are very few Brazilian academic researches dealing with this issue. Moreover, this thesis not only enabled the identification of the main actors and ideas/beliefs that defined the direction of this policy in the last two decades, but also allowed the identification of coalitions that originally disputed positions of power and influence in a subsystem which today still lacks substantive reforms in a more democratic and pluralistic society that requires greater efficiency and accountability of public safety policy considering the terrible crime indicators, largely responsible for the almost generalized violence that endangers citizenship and, consequently, the democratical order itself in our country. 5-Methylcytosine in DNA of eukaryotes, such as humans, is an important epigenetic mark. The recently characterized TET/JBP enzymes generate oxidized derivatives of methylcytosine, such as hydroxy-, formyl-, and carboxymethylcytosine in mammals, which serve as further epigenetic marks or intermediates for demethylation. Unlike animals, which contain one to three TET genes, fungi, such as mushrooms and rusts, display lineage-specific expansions with numerous TET/JBP genes, which are often associated with a unique class of transposable elements. We present evidence that expansion and turnover of these elements and associated TET/JBP genes play important roles in genomic organization, epigenetics, and speciation of fungal lineages, especially basidiomycetes (mushrooms, rusts, and smuts). Domesticated versions of these transposons might also participate in genome rearrangements or repair in humans.

The objective of this work was to compare the morphometry of hatchery-reared and wild-caught mangrove crab (Ucides cordatus) megalopae. Ten U. cordatus megalopae of each group (hatchery-reared and wild-caught) were individually analyzed using a stereoscopic microscope equipped with an ocular micrometer. Length, width, and height of all megalopae were measured, and the size of body appendices was determined. The results indicate that the hatchery-reared megalopae are more robust than the wild ones. Furthermore, some significant differences in the size of certain appendices can be cues of the kind of alterations that hatchery-reared individuals experience.

O objetivo deste trabalho foi comparar a morfometria de megalopas de caranguejo do mangue (Ucides cordatus) produzidas em laboratório e capturadas na natureza. Dez megalopas de U. cordatus de cada grupo (produzidas em laboratório e capturadas na natureza) foram analisadas individualmente utilizando microscópio estereoscópico equipado com ocular micrométrica. Comprimento, largura e altura de todas as megalopas foram medidos, e o tamanho dos apêndices corporais foi determinado. Os resultados são indicativos de que as megalopas produzidas em laboratório são mais robustas que as selvagens. Além disso, algumas diferenças significativas no tamanho de certos apêndices podem ser pistas do tipo de alterações que os indivíduos produzidos em laboratório experimentam.

Objective: To describe the frequency and seasonality of Trichostrongylus spp. infection in individuals attended at a public clinical laboratory. Materials and methods: Fecal samples of 9,283 individuals were evaluated by spontaneous sedimentation (Lutz) in the Parasitology Laboratory of the Pharmacy College, Federal University of Bahia, Brazil, from January of 2006 to May of 2008. The positive samples for either Trichostrongylus spp. or hookworms were further examined to evaluate the morphometry of nematode eggs. However, little is known about the phenotype and activation status of T cells in the process. Objective. To evaluate the profile of T cells in schistosomiasis patients with periportal fibrosis. Methods. It was a cross-sectional study, conducted in the village of Agua Preta, Bahia, Brazil, which included 37 subjects with periportal fibrosis determined by ultrasound. Peripheral blood mononuclear cells were obtained by the Ficcol-hypaque gradient and the frequency of T cells expressing the surface markers CD28, CD69, CD25, and CTLA-4 was determined by flow cytometry. Results. The frequency of CD4+CD28+ T lymphocytes was higher in individuals with moderate to severe fibrosis compared to patients with incipient fibrosis. We did not observe any significant difference in the frequency of CD4+ T cells expressing CD69 among groups of individuals. There was also no significant difference in the frequency of CD8+ T cells expressing CD28 or CD69 among the studied groups. Individuals with moderate to severe fibrosis presented a lower frequency of CD8+ T cells, CD4+CD25high T cells, and CD4+CTLA-4+ T cells when compared to patients without fibrosis or incipient fibrosis. The frequency of CD4+CD25low cells did not differ between groups. Conclusion. The high frequency of activated T cells coinciding with a low frequency of putative Treg cells may account for the development of periportal fibrosis in human schistosomiasis.

OBJECTIVE: To determine whether a multifaceted quality improvement intervention reduces the mortality of critically ill adults. DESIGN, SETTING, AND PARTICIPANTS: This study had 2 phases. Phase 1 was an observational study to assess baseline data on work climate, care processes, and clinical outcomes, conducted between August 2013 and March 2014 in 118 Brazilian ICUs. Phase 2 was a cluster randomized trial conducted between April and November 2014 with the same ICUs. The first 60 admissions of longer than 48 hours per ICU were enrolled in each phase. INTERVENTIONS: Intensive care units were randomized to a quality improvement intervention, including a daily checklist and goal setting during multidisciplinary rounds with follow-up clinician promptingfor 11 care processes, or to routine care. MAIN OUTCOMES AND MEASURES: In-hospital mortality truncated at 60 days (primary outcome) was analyzed using a random-effects logistic regression model, adjusted for patients' severity and the ICU's baseline standardized mortality ratio. Exploratory secondary outcomes included adherence to care processes, safety climate, and clinical events. RESULTS: A total of 6877 patients (mean age, 59.7 years; 3218 [46.8%] women) were enrolled in the baseline (observational) phase and 6761 (mean age, 59.6 years; 3098 [45.8%] women) in the randomized phase, with 3327 patients enrolled in ICUs (n = 59) assigned to the intervention group and 3434 patients in ICUs (n = 59) assigned to routine care. There was no significant difference in in-hospital mortality between the intervention group and the usual care group, with 1096 deaths (32.9%) and 1196 deaths (34.8%), respectively (odds ratio, 1.02; 95% CI, 0.82-1.26; P =.88). Among 20 prespecified secondary outcomes not adjusted for multiple comparisons, 6 were significantly improved in the intervention group (use of low tidal volumes, avoidance of heavy sedation, use of central venous catheters, use of urinary catheters, perception of team work, and perception of patient safety climate), whereas there were no significant differences between the intervention group and the control group for 14 outcomes (ICU mortality, central line-associated bloodstream infection, ventilator-associated pneumonia, urinary tract infection, mean ventilator-free days, mean ICU length of stay, mean hospital length of stay, bed elevation to ≥30°, venous thromboembolism prophylaxis, diet administration, job satisfaction, stress reduction, perception of management, and perception of working conditions). CONCLUSIONS AND RELEVANCE: Among critically ill patients treated in ICUs in Brazil, implementation of a multifaceted quality improvement intervention with daily checklists, goal setting, and clinician prompting did not reduce in-hospital mortality. O artigo discute a importância das ações de direitos humanos naeducação para a cidadania, argumentando que a construção deespaços urbanos mais humanizados se dá através da educaçãoem direitos humanos. Para tanto, apresenta ações de extensãouniversitária, assim como projetos que promovem os direitos desegmentos vulneráveis, coordenadas e desenvolvidas pelo Núcleode Direitos Humanos da Pró-reitoria de Extensão da PUCMinas. Conclui que a constituição de uma sociedade mais inclusiva,em cidades mais democráticas e pluralistas, na qual todosos cidadãos – sujeitos de direitos e deveres - são importantes naconstrução coletiva de novas relações de solidariedade e civismono seu espaço vivencial, faz parte da missão das Universidades.

Coordenação de Aperfeiçoamento de Pessoal de Nível Superior Three thermophilic lignocellulolytic microbial communities were selected by enriched method with continual subcultivation at 55 °C. One community was selected from cow manure compost, another from decaying sugar cane bagasse, and the last one was obtained by mixing aliquots of the culture suspension from the first two. Those communities were able to degrade a cellulosic strip in three cultivation days. Evaluating the maximal day of CMCase and xylanase activity, it was observed that the mixed consortium showed the best results, with CMCase activity of 0.09 U mg-1 at the second day of cultivation, and xylanase activity of 2.86 U mg-1 at the fourth day. These enzymes were partially characterized with relation to temperature and pH of optimal activity. It was observed that CMCase showed the highest activity at 60 °C and pH 5.4, and kept 80% of its activity in a pH range of 4.5-6.5. On the other hand, the best activity for xylanase was verified at 65 °C, and in that same pH range, the enzyme kept 97% of its residual activity. Cell-free extract was concentrated by ultrafiltration. The enzymatic cocktail obtained showed CMCase activity 25-fold higher and xylanase activity 55-fold higher than the crude enzymatic extract. The cocktail was conserved by adding 50% glycerol. After storage for 60 days at 4 °C, xylanase kept 80% of the initial activity and CMCase didn t show loss of activity when kept at 25 °C for the same period. The mixed community cellular mass constitutes an inoculant able to maintain the cellulolytic phenotype after rapid freezing and storage at - 80 °C for 60 days. Fed-batch essay suggested that this community has potential to be manipulated in order to continuously hold the cellulolytic enzymes expression over time. The results evidenced the acquisition of an enzymatic cocktail from an inoculant which cellulolytic activity supported pH changes and optimal activity around 60 °C. Três comunidades microbianas lignocelulolíticas e termofílicas foram selecionadas por subcultivo em meio de enriquecimento a 55 °C. Uma comunidade foi selecionada a partir de esterco de gado em compostagem, outra de bagaço de cana em decomposição e a terceira foi obtida a partir da mistura de alíquotas do meio de cultivo das duas anteriores. Tais comunidades apresentavam a característica de decompor uma fita de celulose em três dias de cultivo. Ao se avaliar o dia de máxima atividade CMCase e xilanase em cada comunidade, verificou-se que os melhores resultados foram encontrados no consórcio misto, com atividade de 0,09 U mg-1 para CMCase no segundo dia de cultivo e 2,86 U mg-1 para xilanase no quarto dia. Essas enzimas foram parcialmente caracterizadas em relação à temperatura e pH ótimos de atuação. Verificou-se que CMCase apresentou maior atividade a 60 °C e pH 5,4, e manteve 80% de sua atividade numa faixa de pH de 4,5 a 6,5. Já para a xilanase, a temperatura ótima foi de 65 °C e nessa mesma faixa de pH manteve uma atividade residual de 97%. O extrato livre de células foi concentrado por ultrafiltração e obteve-se um coquetel enzimático com atividade de CMCase e xilanase maior que no extrato bruto cerca de 25 e 55 vezes, respectivamente. O coquetel foi conservado pela adição de 50% de glicerol. Após 60 dias de armazenamento a 4 °C, a xilanase manteve 80% de sua atividade inicial e a CMCase não apresentou perda de atividade quando mantida a 25 °C pelo mesmo período. A massa celular da comunidade mista constitui um inoculante capaz de manter o fenótipo celulolítico após congelamento rápido e armazenamento a - 80 °C por 60 dias. Ensaio em batelada alimentada mostrou que essa comunidade apresenta potencial para ser manipulada a fim de se manter continuamente a expressão de enzimas celulolíticas ao longo do tempo. Os resultados mostraram que foi possível obter um coquetel de enzimas a partir do inoculante, cuja atividade celulolítica tolerou variações de pH e temperatura ótima em torno de 60 °C.

Conclusion: In the official game, the numbers of passes were more wrong, as possible factors such as place of departure, the structural organization of the team to arbitration, the fans, the collection of positive results, the pursuit of victory and the psychological pressure, influence resource fulness of the shares,which in turn reflected in the development of the passes, making bad passes. Objetivo: Hacer una comparación en dos situaciones: pases erróneos en el Juego/Entrenamiento y Partido Oficial, buscando observar si el Partido Oficial influye en el desempeño del equipo Sub 12 del Mairiporã. Materiales y métodos: Se utilizaron estáticas recolectadas manualmente de los cuatro partidos observados, dos Partidos/Entrenamiento y dos Partidos Oficiales. Las estadísticas se almacenaron en hojas de cálculo de control de exploración desarrolladas en el software Microsoft Office Excel 2010, para registrar el trabajo desarrollado se utilizó la videocámara Sony DCR-SR42 Handycam Digital. Resultados: Luego de las observaciones se concluyó que hubo 61 pases erróneos en el Juego/Entrenamiento y 94 pases erróneos en el Partido Oficial. Mostrando que el Partido Oficial influye en el desempeño del equipo Sub 12. Discusión: Los atletas del equipo Mairiporã Sub 12 no realizaron el pase correctamente y según Asin (1969), no se trata de lanzar el balón lo más lejos posible, es necesario y primordial en el fútbol moderno colocar el balón con precisión. Una de las características del fútbol es el toque de la pelota con el pie, aplicando el puntapié en el lugar correcto de la pelota y con la parte derecha del pie y dando la mayor velocidad y precisión al lanzamiento, Borsari (1989). Conclusión: En el partido oficial, los números de pases erróneos fueron mayores, ya que posibles factores como el lugar del partido, la organización estructural del equipo, el árbitro, la multitud, la recolección de resultados positivos, la búsqueda de la victoria y la presión psicológica, influye en el ingenio de las acciones, lo que en consecuencia se refleja en el desarrollo de los pases, haciendo que los pases sean incorrectos. Obiettivo: Fare un confronto in due situazioni: passaggi sbagliati in Partita/Allenamento e Partita Ufficiale, cercando di osservare se la Partita Ufficiale influisca sul rendimento dell'Under 12 di Mairiporã. Materiali e metodi: Sono state utilizzate le statistiche raccolte manualmente dalle quattro partite osservate, due partite/allenamento e due partite ufficiali. Le statistiche sono state archiviate in fogli di calcolo di controllo scout sviluppati nel software Microsoft Office Excel 2010, per registrare il lavoro svolto è stata utilizzata la videocamera Sony DCR-SR42 Handycam Digital. Risultati: Dopo che le osservazioni hanno concluso che ci sono stati 61 passaggi sbagliati nella partita/allenamento e 94 passaggi sbagliati nella partita ufficiale. Dimostrando che la partita ufficiale influenza le prestazioni della squadra Sub 12. Discussione: Gli atleti della squadra Mairiporã Sub 12 non hanno eseguito il passaggio correttamente e secondo Asin (1969), non si tratta di lanciare la palla il più lontano possibile, è necessario e fondamentale nel calcio moderno posizionare la palla con precisione. Una delle caratteristiche del calcio è il tocco della palla con il piede, applicando il calcio nel punto giusto del pallone e con la parte destra del piede e dando la massima velocità e precisione al tiro, Borsari (1989). Conclusione: Nella partita ufficiale, i numeri dei passaggi sbagliati erano più alti, poiché possibili fattori come il luogo della partita, l'organizzazione strutturale della squadra, l'arbitro, il pubblico, la raccolta di risultati positivi, la ricerca della vittoria e la pressione psicologica, influenza l'intraprendenza delle azioni, che di conseguenza si riflette sullo sviluppo dei passaggi, rendendo i passaggi errati. Conclusão: Na partida oficial, os números de passes errados foram maiores, visto que possíveis fatores como o local da partida, a organização estrutural do time, a arbitragem, a torcida, a cobrança de resultados positivos, a busca pela vitória e a pressão psicológica, influenciam na desenvoltura das ações, que consequentemente reflete no desenvolvimento dos passes, tornando os passes incorretos. We evaluated the immune response in S. mansoni infected individuals with different degrees of periportal fibrosis. Cytokine and chemokines were measured in serum and in supernatants of PBMC cultures stimulated with the soluble adult worm (SWAP) or egg (SEA) antigens, using a sandwich ELISA. The levels of IL-5 in response to SEA were higher in individuals with moderate to severe fibrosis (310.9 pg/mL) compared to individuals without fibrosis (36.8 pg/mL; P = 0.0418). There was also a higher production of TNF-α in cultures stimulated with SWAP in patients with insipient fibrosis (1446 pg/mL) compared to those without fibrosis (756.1 pg/mL; P = 0.0319). The serum levels of IL-13 and MIP-1α were higher in subjects without fibrosis than in those with moderate to severe fibrosis. However a positive association between serum levels of IL-13, TNF-α, MIP-1α, and RANTES and S. mansoni parasite burden was found. From these data we conclude that IL-5 and TNF-α may participate in liver pathology in schistosomiasis. The positive association between IL-13, TNF-α, MIP-1α, and RANTES with parasite burden, however, might predict the development of liver pathology.

We conducted a cross-sectional study of persons resistant to schistosomiasis and found no association between socioeconomic status and resistance to infection. Mononuclear cells of resistant subjects produced higher levels of interleukin-5 (IL-5), IL-13 and interferon-γ upon stimulation with soluble egg antigen (SEA) compared with infected persons. When stimulated with Sm21.6 or Sm22.6, levels of IL-10 were higher in cell culture of resistant persons. Levels of IgE against soluble adult worm antigen (SWAP) and against interleukin-4–inducing principle from S. mansoni eggs (IPSE) and levels of IgG4 against SWAP, SEA, and Sm22.6 were lower in the resistant group compared with the susceptible group. Our data suggest that socioeconomic status could not fully explain resistance to S. mansoni infection observed in the studied area. However, a mixture of Th1 and Th2 immune responses and low levels of specific IgG4 against parasite antigens could be mediating resistance to infection.

However, little epidemiological information was originated from schistosomiasis areas. This study aimed to investigate factors associated with asthma in a schistosomiasis endemic area. A questionnaire was used to obtain information on demographics, socioeconomic, and environmental features. The ISAAC questionnaire was used to identify individuals with asthma. Parasitological exam was done in all participants and skin prick test to aeroallergens in all asthmatics. Prevalence of Schistosoma mansoni infection was 57.4% and Ascaris lumbricoides, 30.8%. Asthma was found in 13.1% of the population, and 35.1% of them had a positive SPT. Active and passive smoking was positively associated with asthma, whereas A. lumbricoides was negatively associated. In a schistosomiasis hyperendemic region, current infection with A. lumbricoides is protective against asthma. However, we cannot rule out the involvement of S. mansoni infection in this process.

The mechanisms underlying this association may include the production of regulatory cells and cytokines. The aim of this study was to evaluate the immune response induced by the S. mansoni antigens, Sm22.6, PIII, and Sm29 and their ability to suppress allergen-specific IL-5 production by peripheral blood mononuclear cells (PBMC) from asthmatic individuals. PBMCs were stimulated in vitro with S. mansoni antigens in the presence or absence of antigen-1 of the mite Dermatophagoides pteronyssinus (Der p1). Cytokines were measured in PBMC supernatants by enzyme-linked immunosorbent assay (ELISA), and the phenotype of cells producing IL-10 was assessed using flow cytometry. High production of S. mansoni antigen-specific IL-10 was observed not only in cells of S. mansoni-infected individuals, but also in cells of noninfected asthmatic individuals. In the former group, the main cellular sources of IL-10 were CD4+ CD25+, and CD14+ cells. The levels of IFN-γ, IL-5, and IL-13 in the noninfected asthmatic group were ∼100 pg/ml in response to the antigens. Moreover, when S. mansoni antigens were added to cultures stimulated with Der p1, levels of IL-10 were increased (Der p1 = 234 ± 118; Der p1 + Sm22.6 = 1189 ± 595; Derp1 + PIII = 799 ± 331; Derp1 + Sm29 = 652 ± 288 pg/ml) with reduced levels of IL-5 (Der p1 = 286 ± 219; Der p1 + Sm22.6 = 93 ± 153; Derp1 + PIII = 132 ± 188; Derp1 + Sm29 = 96 ± 86 pg/ml). The S. mansoni antigens evaluated in the present study induced the production of the regulatory cytokine IL-10 and down-modulated the Th2 immune response that participates in the pathology of asthma.
