153. Model Order Reduction of Aeroservoelastic Model of Flexible Aircraft

Abstract

This paper presents a holistic model order reduction (MOR) methodology and framework that integrates key technological elements of sequential model reduction, consistent model representation, and model interpolation for constructing high-quality linear parameter-varying (LPV) aeroservoelastic (ASE) reduced order models (ROMs) of flexible aircraft. The sequential MOR encapsulates a suite of reduction techniques, such as truncation and residualization, modal reduction, and balanced realization and truncation to achieve optimal ROMs at grid points across the flight envelope. The consistence in state representation among local ROMs is obtained by the novel method of common subspace reprojection. Model interpolation is then exploited to stitch ROMs at grid points to build a global LPV ASE ROM feasible to arbitrary flight condition. The MOR method is applied to the X-56A MUTT vehicle with flexible wing being tested at NASA/AFRC for flutter suppression and gust load alleviation. Our studies demonstrated that relative to the fullorder model, our X-56A ROM can accurately and reliably capture vehicles dynamics at various flight conditions in the target frequency regime while the number of states in ROM can be reduced by 10X (from 180 to 19), and hence, holds great promise for robust ASE controller synthesis and novel vehicle design.

Abstract

Brain tumors have become the leading cause of cancer-related death in children. An important hurdle to scientific and clinical progress in the field has been the limited availability of preclinical tumor models. Historically, few pediatric brain tumor cell lines have been established and these often poorly recapitulate the phenotypes of the original tumors. In recent years, the Children’s Brain Tumor Network (CBTN) has accelerated the development of patient-derived cell lines and xenografts, offering these resources to the community through open-source access. While these models are extremely valuable, their development process can be lengthy and result in clonally selected lines which presents a challenge for studying complex tumor biology. To address the need for three-dimensional tissue culture, our group in conjunction with CBTN, utilized organoid culture from fresh tissue specimens obtained directly from surgical resection of various pediatric brain tumor histologies. This resulted in the development and banking of over 30 organoid models, which included ependymoma, high-grade glioma, medulloblastoma, atypical teratoid-rhabdoid tumor, diffuse midline glioma, and low-grade glioma diagnoses. Tissue was processed within an hour post extraction and cultured with universal media composition for each diagnosis. Organoid growth was observed within 2-3 weeks of initiation and continued for up to three months before banking. Banked organoids established growth upon return to culture. Phenotypic analysis revealed organoid cell composition that represented clinical histology. Importantly, organoids returned to culture post-banking demonstrated similar cell composition to those in the original culture, indicating their utility for subsequent preclinical testing. Here we provide a simple and efficient workflow for the generation and characterization of three-dimensional tumor organoids generated from fresh surgical pediatric brain tumor tissue. The platform has the potential to accelerate investigations into tumor biology and empower a diverse array of translational studies for the pediatric brain tumor field.

Abstract

This paper presents a mathematically rigorous, subspace projection-based reduced order modeling (ROM) methodology and an integrated framework to automatically generate reduced order models for spacecraft thermal analysis. Two key steps in the reduced order modeling procedure are described: (1) the acquisition of a full-scale spacecraft model in the ordinary differential equation (ODE) and differential algebraic equation (DAE) form to resolve its dynamic thermal behavior; and (2) the ROM to markedly reduce the dimension of the full-scale model. Specifically, proper orthogonal decomposition (POD) in conjunction with discrete empirical interpolation method (DEIM) and trajectory piece-wise linear (TPWL) methods are developed to address the strong nonlinear thermal effects due to coupled conductive and radiative heat transfer in the spacecraft environment. Case studies using NASA-relevant satellite models are undertaken to verify the capability and to assess the computational performance of the ROM technique in terms of speed-up and error relative to the full-scale model. ROM exhibits excellent agreement in spatiotemporal thermal profiles (<0.5% relative error in pertinent time scales) along with salient computational acceleration (up to two orders of magnitude speed-up) over the full-scale analysis. These findings establish the feasibility of ROM to perform rational and computationally affordable thermal analysis, develop reliable thermal control strategies for spacecraft, and greatly reduce the development cycle times and costs.

Abstract

In this paper, we present an analytical method for solving the electric potential equation with the exact boundary condition. We analyze the dielectrophoresis (DEP) force with an n-phase ac electric field periodically applied on an interdigitated parallel electrode array. We compare our analytical solution with the numerical results obtained using the commercial software CFO-ACE. This software verifies that our analytical method is correct for solving the problem. In addition, we compare the analytical solutions obtained using the exact boundary conditions and the approximate boundary conditions. The comparison shows that the analytical solution with the exact boundary condition gives a more accurate analysis for DEP and traveling wave DEP forces. The DEP forces of latex beads are also investigated with different phase arrays for (n = 2, 3, 4, 5, 6). [DOI: 10.1115/1.2956610]

Abstract

Neural development requires the orchestration of dynamic changes in gene expression to regulate cell fate decisions. This regulation is heavily influenced by epigenetics, heritable changes in gene expression not directly explained by genomic information alone. An understanding of the complexity of epigenetic regulation is rapidly emerging through the development of novel technologies that can assay various features of epigenetics and gene regulation. Here, we provide a broad overview of several commonly investigated modes of epigenetic regulation, including DNA methylation, histone modifications, non-coding RNAs, as well as epitranscriptomics that describe modifications of RNA, in neurodevelopment and diseases. Rather than functioning in isolation, it is being increasingly appreciated that these various modes of gene regulation are dynamically interactive and coordinate the complex nature of neurodevelopment along multiple axes. Future work investigating these interactions will likely utilize “multi-omic” strategies that assay cell fate dynamics in a high-dimensional and high-throughput fashion. Novel human neurodevelopmental models including iPSC and cerebral organoid systems may provide further insight into human-specific features of neurodevelopment and diseases.

Abstract

GABA, a major inhibitory neurotransmitter in the adult brain, activates synaptic and extrasynaptic [GABA.sub.A] receptors, causing hyperpolarization of mature neurons. As in the embryonic nervous system, GABA depolarizes neural progenitors and immature neurons in the adult brain. Several recent studies have suggested that GABA has crucial roles in regulating different steps of adult neurogenesis, including proliferation of neural progenitors, migration and differentiation of neuroblasts, and synaptic integration of newborn neurons. Here, we review recent findings on how GABA regulates adult neurogenesis in the subventricular zone of the lateral ventricles and in the dentate gyrus of the hippocampus. We also discuss an emerging view that GABA serves as a key mediator of neuronal activity in setting the tempo of adult neurogenesis.
