151. An endothelial cell-derived growth factor.

Abstract

Cell-free plasma-derived serum (PDS) is deficient in the platelet-derived growth factor and will not support the growth of 3T3 cells, fibroblasts, or smooth muscle cells. However, when PDS-containing medium is preincubated with endothelial cells, the medium becomes modified so that it will support growth. The activity produced by the endothelial cells results from a polypeptide of 10,000 to 30,000 daltons which has several features that differ from those of the platelet-derived growth factor, including heat instability and lack of adsorption to CM Sephadex.

152. The role of membrane-membrane interactions in the regulation of endothelial cell growth.

Abstract

A cell surface preparation from confluent endothelial cells can inhibit DNA synthesis of actively growing endothelial cells. The decrease in the rate of [3H]thymidine incorporation is concentration dependent and levels off at 47% of the control. The preparation has no affect on the growth of vascular smooth muscle cells. A similar preparation from smooth muscle cells does not show inhibitory activity with either endothelial or smooth muscle cells. The inhibition of growth can also be demonstrated by a decrease in thymidine index and growth rate. The inhibition is transient and after 48 h, the growth rate is similar to that of the control. In a wound edge assay, both migration and proliferation are inhibited. The inhibitory activity is partially labile to trypsin and abolished by pepsin, heating at 100 degrees C, or reduction. Cell surface iodination and analysis of the proteins removed by urea treatment by SDS polyacrylamide gel electrophoresis show at least 11 bands with apparent molecular weights from 250,000 to 18,000. These radiolabeled proteins, as well as the active component of the cell surface preparation, are sedimentable at 100,000 g for 1 h. They are both solubilized in 30 mM octyl glucoside but not by treatment with 0.1 M sodium carbonate, pH 11.5. These results suggest that the activity is due to a cell-surface membrane fraction and may provide a basis for studying the mechanism of density-dependent inhibition of growth in a normal cell of defined origin.

154. Expression and developmental control of platelet-derived growth factor A-chain and B-chain/Sis genes in rat aortic smooth muscle cells.

Abstract

Cultured arterial smooth muscle cells (SMC) can produce platelet-derived growth factor (PDGF)-like molecules. This property raises the possibility that SMC-derived PDGFs function as autocrine/paracrine regulators in the formation and maintenance of the artery wall. In this study we have asked if levels of mRNAs directing synthesis of PDGF are modulated in aortic SMC during postnatal development. We report here that genes encoding PDGF A- and B-chain precursors are expressed at similar low levels in intact aortas from newborn and adult rats. Marked differences in regulation of transcript abundance of these genes were revealed when aortic SMC were grown in cell culture. PDGF B-chain transcripts accumulated in passaged newborn rat SMC but not adult rat SMC, whereas PDGF A-chain RNA was found in comparable amounts in SMC from both age groups. Similarly, SMC from newborn rats secreted at least 60-fold more PDGF-like activity into conditioned medium than did adult rat SMC. PDGF B-chain transcripts in newborn rat aortic SMC are short-lived and increased 5-fold by 3 hr after treatment with cycloheximide. In contrast, PDGF A-chain transcripts are more stable, and their constitutive levels were generally unaffected by cycloheximide. These results show that PDGF A- and B-chain genes are transcribed in the normal rat aorta and provide evidence for age-related change in the control of PDGF B-chain gene expression in aortic SMC. Independent regulation of transcript levels in cultured SMC leaves open the possibility that PDGFs of different composition (AA, AB, BB) play different roles in normal function of the artery wall.

155. The platelet glycoprotein IIb/IIIa-like protein in human endothelial cells promotes adhesion but not initial attachment to extracellular matrix.

Abstract

On platelets the membrane glycoprotein IIb/IIIa complex (GPIIb/IIIa) functions in adhesive interactions with fibrinogen, von Willebrand factor, and fibronectin. However, the function of GPIIb/IIIa-like proteins on endothelial cells, as well as the ligand(s) the complex binds, is unknown. Using a highly specific polyclonal antibody we have explored the function of GPIIb/IIIa-like proteins on human umbilical vein endothelial cells (HUVE). Analysis by immunoblotting shows that this antiserum recognizes the endothelial GPIIIa-like protein of the complex. The IgG fraction of the polyclonal antiserum and its Fab' fragments detach confluent and subconfluent HUVE from extracellular substrata. The effect of the anti-GPIIb/IIIa IgG is not toxic as the detached cells maintain their viability after trypsinization and replating. Anti-GPIIb/IIIa IgG does not inhibit HUVE binding to extracellular matrix or purified fibronectin in an attachment assay despite the presence of intact GPIIb/IIIa on HUVE detached from substrate by various methods. Apparently, the GPIIb/IIIa-like protein on HUVE is important in normal HUVE adhesion to the extracellular matrix, but it is not required in the initial attachment of HUVE to extracellular matrix.

156. Localization of tissue factor in the normal vessel wall and in the atherosclerotic plaque.

Abstract

Tissue factor (TF)-producing cells were identified in normal human vessels and atherosclerotic plaques by in situ hybridization and immunohistochemistry using a specific riboprobe for TF mRNA and a polyclonal antibody directed against human TF protein. TF mRNA and protein were absent from endothelial cells lining normal internal mammary artery and saphenous vein samples. In normal vessels TF was found to be synthesized in scattered cells present in the tunica media as well as fibroblast-like adventitial cells surrounding vessels. Atherosclerotic plaques contained many cells synthesizing TF mRNA and protein. Macrophages present as foam cells and monocytes adjacent to the cholesterol clefts contained TF mRNA and protein, as did mesenchymal-appearing intimal cells. Significant TF protein staining was found deposited in the extracellular matrix surrounding mRNA-positive cells adjacent to the cholesterol clefts and within the necrotic cores. These results suggest that deposition of TF protein in the matrix of the necrotic core of the atherosclerotic plaque may contribute to the hyperthrombotic state of human atherosclerotic vessels.

157. Influence of Soil Organic Acids on Soluble Phosphorus in Miami and Wooster Silt Loam Soils

Abstract

Having previously determined the qualitative make‐up of the organic acids normally occurring in air‐dry samples of several Ohio soils (9), samples of Miami silt loam soil treated with alfalfa and wheat straw plus nitrogen were subjected to incubation in laboratory and greenhouse experiments. The purpose of these experiments was to determine whether or not there was a correlation between phosphorus solubility and organic acid occurrence. Field samples from 2‐year rotation plots were also periodically analyzed for soluble phosphorus and organic acids through a 10‐week period. The organic acids separated from soils treated with either alfalfa or wheat straw plus nitrogen were apparently identical with those in the control samples. Phosphate levels increased in the alfalfa‐treated and control samples after 4 weeks of aerobic incubation as did isocitric and possibly tartaric acids. However, correlation between these acids and phosphorus solubility in the wheat straw plus nitrogen‐treated samples was apparently lacking. A similar lack of correlation was apparent under anaerobic conditions. Further, it was found that the field samples contained no significant amounts of isocitric acid. The incorporation of sweet clover green manure appeared to have no influence either on the amounts or distribution of the several organic acids recovered from plots without green manure crop.

158. Separation of Organic Acids from Several Dormant and Incubated Ohio Soils

Abstract

The objectives of this study were to determine the organic acid content of several of the Grey‐Brown Podzolic soils in Ohio and to study changes in the acids occurring as a result of short period aerobic incubatious of the soil materials at constant temperature and moisture. The soil materials used in this investigation, Brookston silty clay loam, Crosby silt loam, Miami silt loam, and Wooster silt loam, were obtained from naturally wooded areas in the state. A chromatographic column containing silica gel was employed for the separation of the organic acids. This procedure permitted the separation and quantitative estimation of the organic acids extracted with ether from the soil material. The identities of the organic acids were indicated by a comparison of their positions on an elution calibration chart with those of known pure organic acids. The chromatographically separated acids were not chemically characterized. It was found that of the organic acids separated by this procedure, only two occurred in significant amounts, namely acetic and formic acids. In addition, very small quantities of other acids, including succinic and lactic acids and what are thought to be a group of high‐molecular weight fatty acids, were also separated. The latter probably resulted from alkali hydrolysis of the fats extracted from the soil materials. A 1% glucose treatment of the several soil materials apparently did not appreciably change the organic acid make‐up either qualitatively or quantitatively. Preliminary data are presented concerning the effect of incubation on phosphorus solubility.

159. Soil‐Conditioning Properties of Modified Agricultural Residues and Related Materials: II. Persistence of Soil‐Stabilizing Activity as a Function of Type and Extent of Modification

Abstract

Agricultural residues, cotton linters, several of their chemical derivatives, and other related products were tested in the laboratory for their in‐soil stability against microbial degradation for a period of 6 months in a controlled humidity cabinet. A modified Yoder wet‐sieving technique was used to test the stabilizing effectiveness of a given polymer incorporated into Miami silt loam soil. Forty‐ to sixty‐mesh wheat straw, incorporated at a concentration of 0.5%, imparted greater stability to soil aggregates, following the first month of incubation at a temperature of 28° C., than did comparable applications of soybean and cotton stalks. Corn stover applications produced intermediate results. Twenty‐ to sixty‐mesh cotton linters at 0.5% concentration yielded increasingly effective soil aggregation during the first 3 months of incubation, reaching a maximum aggregation value of 93%. Chemically‐oxidized cellulose and corn starch showed surprising in‐soil stability throughout the 6‐month incubation period. In carboxymethyl cellulose samples, a degree of substitution (DS) of 0.7 or less was apparently insufficient to impart resistance to degradation by the microflora of the soil. However, carboxymethyl cellulose with a DS of 1.2 retained approximately 70% of its initial soilstabilizing activity over the 6‐month test period. In the case of hydroxyethyl and methyl cellulose, data are presented which tend to show that a high degree of substitution is not an adequate criterion of resistance to microbial degradation. Rather, the susceptibility of such polymers to enzymatic hydrolysis is apparently attributable to a lack of uniformity of substitution, resulting in a relative abundance of unsubstituted units in the cellulose chain.

Abstract

The feasibility of producing soil‐conditioning agents from agricultural residues by chemical modification was investigated. Such reactions as xanthation, cyanoethylation, methylation, hydroxyethylation, sulfation, phosphorylation, acetylation, and oxidation were applied to wheat straw, corn stover, corncobs, bagasse, rice hulls, and their major components. From results of comparative reactions on cellulose, hemicellulose, and lignin, the major part of the soil‐conditioning activity of chemically‐modified agricultural residues was found attributable to the substituted cellulose fraction. The extent of substitution required to produce active products varies with the nature of the derivative but coincides in all cases with that required to give water‐soluble products. Within these limits the degree of polymerization has more influence on aggregate‐stabilizing activities than either the type or extent of substitution. Data on a number of microbial gums, plant polysaccharides, and starch derivatives are also presented.

Abstract

This study examined the effects of injury on the content of von Willebrand factor (vWF) in rat aortic endothelium. Endothelial cells from normal, endotoxin-treated, and balloon-injured rats were stained with vWF antibodies and visualized using a biotinylated secondary antibody and avidin-tagged peroxidase. Endotoxin treatment and balloon injury caused a threefold increase in intracellular vWF, and immunoelectron microscopy showed the endoplasmic reticulum to stain heavily by the vWF antibody. Weibel-Palade bodies were not observed in all the cell profiles examined. The basement membrane of the endothelialized vessels showed no vWF staining; however, after endothelial denudation this matrix was clearly stained by the antibody. These results suggest that endothelial injury leads to an increased intracellular content of vWF that is localized primarily in the endoplasmic reticulum.

Abstract

BACKGROUND: Global inequalities in access to health care are reflected in differences in cancer survival. The CONCORD programme was designed to assess worldwide differences and trends in population-based cancer survival. In this population-based study, we aimed to estimate survival inequalities globally for several subtypes of childhood leukaemia. METHODS: Cancer registries participating in CONCORD were asked to submit tumour registrations for all children aged 0-14 years who were diagnosed with leukaemia between Jan 1, 1995, and Dec 31, 2009, and followed up until Dec 31, 2009. Haematological malignancies were defined by morphology codes in the International Classification of Diseases for Oncology, third revision. We excluded data from registries from which the data were judged to be less reliable, or included only lymphomas, and data from countries in which data for fewer than ten children were available for analysis. We also excluded records because of a missing date of birth, diagnosis, or last known vital status. Abstract

Neovascularization in the walls of coronary arteries is associated with the presence of atherosclerotic plaque. The mechanisms responsible for the formation of these intraplaque microvessels are not understood. The purpose of this study is to examine the prevalence of endothelial cell replication in plaque microvessels. Two hundred and one primary and restenotic coronary atherectomy specimens were analyzed for the presence of microvessels and proliferation as reflected by positive immunolabeling for Ulex agglutinin and the proliferating cell nuclear antigen, respectively. In primary but not restenotic specimens, proliferation of any cell type was associated with the detection of microvessels on the same slide. However, intraplaque microvessels were more commonly found in restenotic compared to primary specimens (P = 0.004). Twelve highly vascularized specimens with evidence of replication were subjected to detailed histomorphological and quantitative image analyses. At 200 x, the most vascular optical field of each slide was identified and consistently included plaque macrophages. Total slide endothelial cell replication indices for these specimens varied, but in some instances were remarkably elevated (eg, 43.5%). The role of intraplaque angiogenesis may be analogous to that of tumor or wound angiogenesis and be important in development and progression of coronary artery lesions and restenosis.

191. Increased incidence of adenocarcinoma of the cervix in young women in the United States.

Abstract

This is the second report demonstrating an increase in the incidence of adenocarcinoma of the cervix in young white women since the early 1970s. The trend does not appear to be due to improved histologic classification over time. Case-control studies are clearly needed to investigate the reasons for the increase, including the hypothesized association with use of oral contraceptives before 20 years of age. However, the low incidence of the disease in young women (in whom an increased risk is most likely to be detectable, should it exist) suggests that collaborative efforts will be required to achieve even modest sample sizes.

Abstract

Background: Within the spectrum of congenital heart disease referred to as hypoplastic left heart syndrome (HLHS), there is variation in the morphology and function of the left ventricle which could influence outcomes after stage I Norwood palliation. Objective: To determine if left ventricular (LV) morphology is associated with outcome after stage I Norwood palliation for HLHS. Methods: Echocardiograms were reviewed from 108 patients who had undergone Norwood palliation at our institution over the past 11 years. Total cardiac diameter, thickness of the interventricular septum (IVS), LV area and LV myocardial area were calculated. Competing risk analysis was performed for survival to a stage II operation and to determine potential predictors. Results: From the Norwood operation up to stage II operation, mortality was predicted by IVS thickness, while the absence of right ventricular (RV) dysfunction was predictive of survival to stage II operation. For the complete pathway, from Norwood to the Fontan operation, mortality was predicted by IVS, a lower RV fractional area change and the presence of significant tricuspid regurgitation. Cardiac transplantation during this period was predicted by a lower RV fractional area change (p = 0.02) and a larger LV area in diastole. Conclusions: These results indicate that LV hypertrophy and decreased RV function adversely effect survival after the Norwood operation. They suggest that LV morphology, especially septal hypertrophy, can influence outcomes in HLHS and should be considered when evaluating treatment options. [ABSTRACT FROM AUTHOR]
